Full textSome steps of the ectomycorrhizae ontogenesis would require a subtle balance between active molecules such as IAA and regulating molecules such as hypaphorine, an indolic alkaloid synthesised and accumulated in large quantities by Pisolithus in contact with the host plant. Here the up-regulating activity of jasmonates on hypaphorine synthesis in Pisolithus hyphae was evidenced; a new class of IAA competitive-antagonists has been identified, the indole alkaloids. The activity of hypaphorine or/and IAA on gene expression in poplar roots, has been folowed. Amongst121 EST differentially expressed, only one coding for an extensinlike protein was regulated the opposite ways by both antagonist molecules. With hypaphorine, and some indole alkaloids, opens a new range of investigation of IAA activities

Full textThe first purpose of my work was to compare gene expression between the fungal compartment in close contact with the root and the extramatrical mycelium of the Paxillus involtus/Betula pendula symbiotic association. A gene coding a urea transporter, up-regulated in the extramatrical mycelium, and genes coding a phosphatidylserine decarboxylase and a protein of unknown function were cloned and characterized further. The second part of my work dealt with the mechanisms involved in the adaptation of ectomycorrhizal fungi to external nitrogen conditions. Studies concerned ammonium and peptide transport, and ammonium assimilating pathways. Moreover, we tried to determine the potential factors explaining the interspecific diversity observed among ectomycorrhizal fungi concerning nitrogen assimilation pathways, by coupling both biochemical and molecular approaches

Full textHypaphorine is an indole compound secreted by the fungus Pisolithus tinctorius to the host plant Eucalyptus globulus during the establishment of an ectomycorrhiza. Hypaphorine inhibits root hair tip growth and the aim of this work is to understand the signalisation pathway involved in this inhibition. Hypaphorine-induced inhibition is rapid and coincides with a transient depolarisation of the plasma membrane and a cytoplasmic calcium increase abolishing the calcium gradient in root hair. Microscopy dye images revealed that hypaphorine induces an aggregation of actin filaments and hinders vesicle accumulation in the apical growing region. Hypaphorine action could be the result of an indole-3-acetic acid (IAA) antagonism as it effects on growth, actin cytoskeleton organisation and vesicle accumulation are counteracted by IAA
