Le facteur ef-1, intervenant dans la phase d'elongation de la synthese proteique, est constitue de deux elements : ef-1a, une proteine g et ef-1bgd, le complexe d'echange de gtp/gdp. La kinase cdc2, de la famille des proteines kinases cdk (dependantes des cyclines ; cdc2=cdk1) phosphoryle ef-1 pendant la transition g2/m. Nous avons etudie la structure et la phosphorylation de ef-1 dans l'ovocyte de xenope. Nous avons identifie deux nouvelles proteines ef-1 d1 et ef-1d2 appartenant au facteur ef-1. Ce sont deux nouvelles proteines d'echange de gtp/gdp, possedant un domaine c-terminal semblable a celui de la proteine ef-1b du meme complexe, et un motif d'interaction proteine-proteine de type leucine-zipper dans leur partie n-terminale. Elles sont toutes les deux presentes au sein des ovocytes, dans le complexe ef-1bgd. Nous avons determine que la valyl arnt synthetase de l'ovocyte est entierement presente dans le complexe d'echange de nucleotide, ef-1bgd. Il avait ete precedemment montre que ef-1g etait cible de la kinase cdc2/cyclineb. Nous avons determine que les deux proteines ef-1 d sont substrats in vivo de la kinase cdc2/ cycline b sur deux phospho-accepteurs une serine et une threonine. La threonine identifiee correspond a vt(92)paak dans ef-1d1 et vt(120)pak dans ef-1d2. Nous proposons en conclusion un modele de l'organisation de ef-1 a partir des observations chromatographiques, electrophoretiques et microscopiques du complexe purifie. Les etudes fonctionnelles de ef-1 et du role de ses phosphorylations devront tenir compte des progres realises dans la connaissance de la structure. Elles necessiteront de trouver de nouveaux modeles biologiques et de considerer non seulement l'activite multifonctionnelle proposee pour la proteine g, ef-1a, mais aussi la multitude des formes physiologiques du complexe ef-1bgd, et de ses sous-unites

The translational GTPases elongation factor Tu (EF-Tu) and LepA modulate the dynamics of tRNA on the ribosome. EF-Tu facilitates the delivery of aminoacyl-tRNA (aa-tRNA) to the translating ribosome and LepA catalyzes the retro-translocation of tRNA•mRNA from the E- and P-sites of the ribosome back to the P- and A-sites. Although an increasing body of structural and biochemical information is available, little is known about the functional cycle of LepA during retro-translocation, the kinetics of EF-Tu dissociation from the ribosome and the rate of EF-Tu conformational change during aa-tRNA delivery. This thesis reports the successful construction and biochemical characterisation of a mutant form of EF-Tu from Escherichia coli ideal for the specific incorporation of fluorescent labels, enabling measurements pivotal for uncovering the rate of EF-Tu conformational change and dissociation from the ribosome. Furthermore, to determine structural components critical for LepA’s function, mutant versions of the protein were constructed and biochemically characterised.<br>xii, 127 leaves : ill. Le complexe des facteurs de l'elongation polypeptidique ef-tu et ef-ts d'escherichia coli a ete cristallise afin d'etudier le mecanisme d'echange nucleotidique sur le plan structural, par la cristallographie aux rayons x. Les cristaux obtenus dans des solutions de peg 6000 peuvent se presenter sous deux formes morphologiquement similaires. L'une diffracte a basse resolution, l'autre a haute. La diffusion de vapeur d'eau peut convertir une forme en l'autre, et cette propriete faisait obstacle a la determination de la structure. La mise au point d'un protocole approprie de trempage et la collecte les donnees a temperature cryogenique, a permis d'obtenir des jeux de donnees complets de la forme diffractant a haute resolution, ce qui a conduit a la structure tridimensionnelle du complexe ef-tu:ef-ts a une resolution de 2. 5 angstroms. Le complexe est une molecule tetramerique contenant deux exemplaires de chaque facteur d'elongation. Les deux molecules de ef-ts forment un dimere et il n'y a que peu de contacts entre les deux molecules de ef-tu, ce qui est formule par 2ef-tu: ef-ts2. Ef-ts est en contact avec les domaines 1 et 3 de ef-tu. L'interaction de ef-ts avec ef-tu montre que le relargage du nucleotide guanidique est principalement du a l'alteration du site de fixation du magnesium par l'insertion de la chaine laterale d'une phenylalanine, appartenant a un motif conserve, dans la structure de ef-tu. Des sequences qui s'apparentent a ce motif peuvent etre identifiees dans d'autres facteurs d'echange nucleotidique

Nous avons etudie l'expression des genes specifiant plusieurs elements du systeme de synthese proteique pendant la gametogenese et l'embryogenese de xenopus laevis. Les genes etudies sont ceux qui codent pour le facteur d'elongation 1 alpha (ef-1 alpha), la thesaurine a et la thesaurine b. Ces deux dernieres proteines sont impliquees dans le stockage a long terme du rna 5s et du trna dans les oocytes. Elles ne sont pas detectables dans les cellules somatiques. Ef-1 alpha existe sous deux formes: une forme somatique (ef-1 alphas) et une forme oocytaire (ef-1 alphao). On demontre ici que le rna messager de ef-1 alphas est indetectable dans les oocytes et les spermatocytes, mais qu'il est present dans les cellules embryonnaires a partir du stade neurula. Le rna messager de ef-1 alphao est present non seulement dans les oogonies et les oocytes de tous les stades, mais aussi dans les spermatogonies et les spermatocytes primaires. Les rna messagers codant pour la thesaurine a et la thesaurine b sont egalement specifiques des cellules germinales. Ils sont traduits en proteines dans le testicule comme ils le sont dans l'ovaire. Cette etude revele une similitude frappante dans le programme d'expression des genes durant l'oogenese et la spermatogenese. Les produits des genes ef-1 alphao, thesaurine a et la thesaurine b peuvent etre consideres comme des marqueurs de la differenciation des cellules germinales

Les genes codant pour le facteur d'elongation de la tranduction ef-1 alpha d'arabidopsis thaliana ont ete clones et caracterises. Le facteur d'elongation ef-1 alpha est code par une petite famille multigenique de quatre membres chez cette espece. Les quatre genes, denommes a1, a2, a3 et a4, sont fonctionnels et codent pour une proteine identique, tres homologue aux divers facteurs d'elongation ef-1 alpha caracterises chez d'autres especes. L'analyse structurale a permis de montrer que les genes a1, a2 et a3 d'une part, et a4 d'autre part, doivent constituer deux sous-familles distinctes dans le genome d'arabidopsis thaliana. Les quatre genes sont interrompus par deux introns dont l'un est situe dans la region 5 non codante. Ils possedent tous dans leur promoteur plusieurs sequences susceptibles de jouer un role dans la regulation de leur expression. Des etudes d'analyse des transcrits ef-1 alpha effectuees dans differents organes de la plante et dans des conditions physiologiques variees (choc thermique, obscurite) indiquent que les quatre genes s'expriment de facon constitutive

La presente ricerca è stata realizzata con l’intento di approfondire alcuni temi cardine nello studio delle Funzioni Esecutive (FE). Il termine FE si riferisce a processi neurocognitivi, deliberati e top-down necessari per esercitare controllo volontario su pensieri ed emozioni e per la messa in atto di comportamenti finalizzati al raggiungimento di un obiettivo. Si tratta di abilità indispensabili in situazioni nuove ed ambigue. Al dominio esecutivo vengono ascritte non solo abilità puramente cognitive. Tra le FF figurano: controllo degli impulsi ed autoregolazione, attenzione, memoria di lavoro, iniziativa, pianificazione ed organizzazione, flessibilità cognitiva, problem solving, decision making affettivo, teoria della mente ed empatia. Ad oggi non esiste un punto di vista condiviso su alcuni temi che sono stati oggetto del nostro interesse: la valutazione, la struttura e lo sviluppo delle FE, i deficit esecutivi presenti in alcuni disturbi specifici dello sviluppo. Per quanto concerne la valutazione delle FE, uno dei nodi critici è la scarsa validità ecologica degli strumenti tradizionalmente utilizzati nella ricerca e nella clinica. Al fine di superare questo problema metodologico sono state proposte modalità alternative di valutazione basate sia sulla performance che su report di insegnanti e genitori. Nella presente ricerca abbiamo proposto una nuova batteria neuropsicologica volta a fornire una panoramica globale del dominio esecutivo mettendo il bambino davanti a problemi analoghi a quelli che la quotidianità elicita. La batteria include: Battersea Multitasking Paradigm Task, Test di Ragionamento visuo-spaziale, Test di Pianificazione Quotidiano, Junior Gambling Task e Test del Clacson. È stato inoltre elaborato un questionario, il Questionario per la valutazione delle Funzioni Esecutive, QuFE, del quale sono indagate struttura, attendibilità ed utilità clinica. A tale scopo il questionario è stato somministrato a genitori ed insegnanti di bambini con sviluppo tipico o atipico di età compresa tra gli 8 ed i 13 anni. I risultati suggeriscono una struttura a 5 fattori per la versione genitori ed una a 3 per la versione insegnanti. Le abilità che sembrerebbero essere indagate sono: abilità metacognitive, regolazione delle emozioni e del comportamento, organizzazione dei materiali, adattamento ed iniziativa. Entrambe le versioni del QuFE si caratterizzano per una elevata validità interna. Per quanto concerne l’utilità clinica, lo strumento sembrerebbe distinguere in modo efficace soggetti con sviluppo tipico da soggetti clinici, minore è la capacità di differenziare tra i profili di soggetti con differenti disturbi dello sviluppo. Lo sviluppo e la struttura delle FE sono stati esaminati somministrando ad un campione di 343 soggetti con sviluppo tipico di età compresa tra gli 8 ed i 13 anni la batteria presentata sopra. Sono stati documentati significativi cambiamenti nei profili di performance in tutti test somministrati. Dall’analisi fattoriale è emersa una struttura a 4 fattori distinti ma interdipendenti: vigilanza, comportamento strategico, inibizione/flessibilità cognitiva e memoria. La struttura è stabile nel tempo e non presenta differenze legate al genere. Infine sono stati indagati i profili esecutivi in bambini con Disturbo da Deficit dell’Attenzione ed Iperattività (ADHD) e Disturbi Specifici dell’Apprendimento (DSA). Sono state oggetto di valutazione le performance di bambini con sviluppo tipico (207) o atipico (65) di età compresa tra gli 8 ed i 13 anni. I nostri dati suggeriscono che i bambini con ADHD e DSA hanno competenze strategiche ed inibitorie sovrapponibili. In entrambi i gruppi si è osservato un deficit della vigilanza e della memoria, di estensione maggiore nel gruppo ADHD. Si conferma la presenza di un deficit esecutivo nelle due popolazioni oggetto di interesse, deficit però circoscritto e non generalizzato.

Production of mutant biological molecules for understanding biological principles or as therapeutic agents has gained considerable interest recently. Synthetic genes are today being widely used for production of such molecules due to the substantial decrease in the costs associated with gene synthesis technology. Along one such line, we have engineered tRNA genes in order to dissect the effects of G:U base-pairs on the accuracy of the protein translation machinery. Our results provide greater detail into the thermodynamic interactions between tRNA molecules and an Elongation Factor protein (termed EF-Tu in bacteria and eEF1A in eukaryotes) and how these interactions influence the delivery of aminoacylated tRNAs to the ribosome. We anticipate that our studies not only shed light on the basic mechanisms of molecular machines but may also help us to develop therapeutic or novel proteins that contain unnatural amino acids. Further, the manipulation of the translation machinery holds promise for the development of new methods to understand the origins of life.
Along another line, we have used the power of synthetic biology to experimentally validate an evolutionary model. We exploited the functional diversity contained within the EF-Tu/eEF1A gene family to experimentally validate the model of evolution termed ‘heterotachy’. Heterotachy refers to a switch in a site’s mutational rate class. For instance, a site in a protein sequence may be invariant across all bacterial homologs while that same site may be highly variable across eukaryotic homologs. Such patterns imply that the selective constraints acting on this site differs between bacteria and eukaryotes. Despite intense efforts and large interest in understanding these patterns, no studies have experimentally validated these concepts until now. In the present study, we analyzed EF-Tu/eEF1A gene family members between bacteria and eukaryotes to identify heterotachous patterns (also called Type-I functional divergence). We applied statistical tests to identify sites possibly responsible for biomolecular functional divergence between EF-Tu and eEF1A. We then synthesized protein variants in the laboratory to validate our computational predictions. The results demonstrate for the first time that the identification of heterotachous sites can be specifically implicated in functional divergence among homologous proteins.
In total, this work supports an evolutionary synthetic biology paradigm that in one direction uses synthetic molecules to better understand the mechanisms and constraints governing biomolecular behavior while in another direction uses principles of molecular sequence evolution to generate novel biomolecules that have utility for industry and/or biomedicine.

Le travail retranscrit dans cette thèse regroupe l'étude de différents processus biologiques impliqués dans la synthèse protéique bactérienne. Dans un premier chapitre, les origines de la synthèse protéique au temps du monde ARN sont traitées en guise d'introduction. Ce travail théorique se poursuit par la présentation d'une structure à haute résolution du facteur d'élongation G (EF-G) en complexe avec le ribosome par cryo-microscopie électronique à transmission (cryo-MET). Grâce aux avancées techniques de la cryo-MET, nous avons observé pour la première fois EF-G lié au ribosome en l'absence de tout inhibiteur. Cet état particulièr d'EF-G permet de visualiser une flexibilité de son doamine III. Cette étude permet aussi de rationaliser le fonctionnement de l'antibiotique acide fusidique. Nous nous sommes ensuite intéressés aux voies de sauvetage de la synthèse protéique et plus particulièrement de la trans-traduction. Ce mécanisme fascinant permet le recyclage des ribosomes bloqués sur un ARN messager défectueux. Cette voie de sauvetage est généralement vitale ou alors indispensable pour la virulence bactérienne. Nous avons réalisé une étude structurale préliminaire de la dégradation de l'ARNm défectueux durant ce processus. Après une revue traitant du sujet, nous présentons une étude de la trans-traduction comme cible pour le développement de nouveaux antibiotiques. Pour cela, nous avons mis au point un système rapporteur avec contrôle interne de l'activité trans-traductionnelle bactérienne. Après avoir mis au point ce système et validé son utilisation, nous l'avons exploité en testant des molécules ciblant la trans-traduction<br>The current PhD work brings together various studies linked to bacterial protein synthesis. The first chapter is about the origins of protein synthesis at the time of the RNA world. This theoretical work continues with the presentation of a high-resolution structure of the elongation factor G (EF-G) in complex with the ribosome by cryo-electron transmission microscopy (cryo-TEM). We describe for the first time EF-G bound to the ribosome in the absence of any inhibitor. This particular structure of EF-G displays a yet unseen positioning of its third domain, which becomes very flexible. This study helps to understand the way the antibiotic fusidic acid blocks translation. The work then switches to a study of trans-translation, the main rescuing system of stalled ribosomes in bacteria. Trans-translation is generally vital or at least necessary for bacterial virulence. We conducted a preliminary structural study on the way faulty mRNAs are degraded during this process. This is why we present a study of trans-translation as a target for the development of new antibiotics. For this we developed and validated a reporter system for trans-translation, which is used to screen molecules targeting trans-translation

La tenue résiduelle à l'impact basse vitesse est un critère dimensionnant des structures composites minces pour l'aéronautique. La majorité des travaux réalisés jusqu'ici porte sur l'analyse du comportement en zone courante. Ce travail élargit le domaine d'étude aux zones de variation d'épaisseur. L'objectif est d'étudier la tolérance aux dommages d'impact d'un stratifié comportant une zone de reprise de plis (ZRP) dans le cadre d'un dialogue essai/ calcul, en menant en parallèle une campagne d'essais expérimentaux et l'adaptation d'un modèle numérique de l'endommagement des composites.Le volet expérimental étudie successivement l'impact, la compression et la fatigue à R=-l après impact (CAI et FAI) d'éprouvettes comportant une ZRP. L'analyse des essais d'impact a permis d'identifier la résistance à l'impact et le mécanisme d'endommagement des éprouvettes. Ensuite, un outillage d'essai adapté à la géométrie des éprouvettes a été conçu pour les essais de CAI et de FAI. Les essais de compression montrent une tenue résiduelle statique après impact similaire à celle des éprouvettes sans perte de plis. Les essais de fatigue menés à 60 070 de la tenue résiduelle statique montrent une propagation des délaminages d'impact (en dessous du BVID) qui mène à la rupture des éprouvettes pour un nombre de cycles relativement faible. Alors que la tolérance aux dommages d'impact des ZRP sous chargement statique est comparable à celle des zones courantes lisses, on constate une forte vulnérabilité de ces zones sous chargement de fatigue.Le volet numérique a permis de tester l'approche DPM (Discrete Ply Model), développé lors de travaux précédents, sur une configuration particulière. Un maillage spécifique a été réalisé pour tenir compte des discontinuités de la ZRP. Au niveau de la loi matériau, une formulation unifiée de la rupture de fibres en traction/compression a été implémentée. Les résultats de la simulation d'impact sont en bonne corrélation avec les données expérimentales. Le modèle est capable de prédire la réponse globale de l'éprouvette ainsi que l'étendu des dommages internes. La modélisation de la CAI a permis de confirmer les mécanismes de rupture identifiés lors des essais. Ces résultats numériques sont remarquables puisqu' aucune modélisation locale particulière n'a été faite pour les arrêts de plis. L'approche DPM s'est révélée suffisamment robuste et bien adaptée à la modélisation de l'endommagement des stratifiés unidirectionnels<br>The residual strength after low velocity impact is a sizing criterion of thin composite structures in aeronautics. The majority of work on the subject is focused on the analysis of plain laminates. This study expands the field of interest to tapered area. The objective is to study the impact damage tolerance of a laminate with ply drop-off using an experiment/ modelling dialogue: on one hand carrying experimental test campaign and on other hand adapting numerical modelling of composite damage. The experimental part successively examines the impact, compression and fatigue R=-l after impact (CAI and FAI) of specimens with ply drop-off. Analysis of the impact results has enabled the identification of impact resistance and damage mechanism. Then, a testing tool for CAI and FAI was specially designed to suit the geometry of the specimens. Compression tests show a static residual strength after impact similar to plain laminates. Fatigue tests carried at 60% of CAI strength show a propagation of impact delamination (below B VID). Failure of specimens occurs after a relatively small number of cycles. While the impact damage tolerance of tapered laminates is comparable to plain laminates under static loading, high vulnerability is observed under fatigue loading. The numerical part allowed to test the Discrete Ply Model (DPM), developed in previous works, on a particular configuration. A specific meshing was realized to account for the discontinuities in the ply drop-off area. Regarding the material law, a unified formulation of the fiber breakage in tension/ compression is implemented. Impact simulation results correlated well with the experimental data. The model is able to predict the overall response of the specimen and internal damage. The modeling of the CAI enabled to confirm the failure mechanisms identified during tests. These numerical results are remarkable since no particular local modeling has been realized for the ply drop-off area. DPM approach has proven robust enough and well suited to damage modeling of unidirectional laminates

Cette recherche porte sur le développement d’un modèle de structure de géométrie complexe équipée de composants à comportement non linéaire viscoélastique dans le but de simuler sa réponse à des excitations définies par des densités spectrales de puissance (DSP). L’application industrielle concerne l’isolation vibratoire d’un module de refroidissement automobile monté sur plot de suspension en élastomère. Une revue du comportement des élastomères en fonction de leurs conditions d’environnement et de sollicitations identifie les paramètres des différents modèles analysés. Des essais préliminaires ont été menés pour définir les intervalles des niveaux de sollicitations et quantifier l’échauffement des plots. La caractérisation expérimentale de la suspension porte sur des plots en élastomère munis de leurs interfaces afin d’agréger dans un seul modèle les non linéarités du comportement viscoélastique, des glissements et des frottements. Les boucles effort-déflexion axiales et radiales mesurées sont traitées avec un système expert développé spécialement pour caler les paramètres du modèle retenu. Il s’agit du modèle de Dahl généralisé qu’il a fallu étendre aux aspects viscoélastiques. Ce processus de calage automatique a été codé avec un logiciel développé sous Octave/Matlab. Des méthodes d’interpolations et extrapolations rendent opérationnel ce modèle sur toute la gamme fonctionnement de l’application définie dans l’espace fréquence-déflexion. Ce processus a été codé dans le module UserSubroutine pour Abaqus. Soumettre le système mécanique non linéaire à des excitations définies par une DSP nécessite de s’appesantir sur le traitement des vibrations aléatoires. En effet il faut, pour calculer les réponses, considérer le passage fréquence-temps et inversement pour les comparer éventuellement aux exigences des normes. De plus, la taille et la complexité du modèle EF de la structure industrielle rendent impossible une résolution temporelle sur l’ensemble de ses degrés de liberté. Il s’agit alors de faire appel à des techniques d’homogénéisation et de condensation dynamique afin de prévoir la réponse aux excitations à large bande fréquentielle dans le but d’analyser les performances de l’isolation vibratoire<br>This research work regards the development of a complex structure model with non-linear viscoelastic components. The purpose of this study is to simulate the response of this structure submitted to a random vibration excitation based on a power spectral density definition (PSD). The industrial applicative case is the vibratory insulation of a automotive engine cooling module supported by elastomer mounts. A brief review of elastomers behavior depending on solicitations types enables to identify the parameters of the different investigated models. Preliminary tests have been conducted to define the range of amplitudes of excitations and evaluate the internal warming of rubbers during the full structure validation test. The experimental characterization of the suspension is based on rubbers mounts and their interfaces with the cooling module, in order to take into account in a unique model all nonlinearities due to the viscoelastic behavior, the slidings, and the friction. Measured force-deflection hysteretic cycles in axial and radial direction are post-processed with an expert system developed to obtain the parameters of the retained model: the modified Dahl’s model, generalized to viscoleastic aspect. This process has been developed with Octave/Matlab code. Interpolation and extrapolation methods enable to obtain a good model response on the global operating range. These methods have been coded in an Abaqus UserSubroutine. Imposing random vibration excitation of a non linear mechanical system based on PSD imposes to take into account signal processing aspects. To evaluate response levels versus norms requirements, it’s mandatory to consider the time-frequency transfer. In addition, the size and the complexity of the total finite element model of the industrial structure don’t allow a global resolution in the time domain for all the degrees of freedom. Bacteria are many, old and varied; different bacterial species have been evolving for millions of years and show many disparate life-styles and types of metabolism. Nevertheless, some of the characteristics regarding how bacteria organize their chromosomes are relatively conserved, suggesting that they might be both ancient and important, and that selective pressures inhibit their modification. This thesis aims to study some of these characteristics experimentally, assessing how changes affect bacterial growth, and how, after changing conserved features, bacteria might evolve. First, we experimentally tested what are the constraints on the horizontal transfer of a gene highly important for bacterial growth. Second, we investigated the significance of the location and orientation of a highly expressed and essential operon; and we experimentally evolved strains with suboptimal locations and orientations to assess how bacteria could adapt to these changes. Thirdly, we sought to understand the accessibility of different regions of the bacterial chromosome to engage in homologous recombination. And lastly, we constructed bacterial strains with chromosomal inversions to assess what effect the inversions had on growth rate, and how bacteria carrying costly inversions could evolve to reduce these costs. The results provide evidence for different selective forces acting to conserve these chromosome organizational traits. Accordingly, we found that evolutionary distance, functional conservation, suboptimal expression and impaired network connectivity of a gene can affect the successful transfer of genes between bacterial species. We determined that relative location of an essential and highly expressed operon is critical for supporting fast growth rate, and that its location seems to be more important than its orientation. We also found that both the location, and relative orientation of separated duplicate sequences can affect recombination rates between these sequences in different regions of the chromosome. Finally, the data suggest that the importance of having the two arms of a circular bacterial chromosome approximately equal in size is a strong selective force acting against certain type of chromosomal inversions.

Les entreprises aéronautiques ont un besoin continu de générer de grands et complexes modèles de simulation, en particulier pour simuler le comportement structurel de sous-systèmes de leurs produits. Actuellement, le pré-traitement des modèles de Conception Assistée par Ordinateur (CAO) issus des maquettes numériques de ces sous-systèmes en Modèles Eléments Finis (MEF), est une tâche qui demande de longues heures de travail de la part des ingénieurs de simulation, surtout lorsque des idéalisations géométriques sont nécessaires. L'objectif de ce travail de thèse consiste à définir les principes et les opérateurs constituant la chaîne numérique qui permettra, à partir de maquettes numériques complexes, de produire des géométries directement utilisables pour la génération de maillages éléments finis d'une simulation mécanique. A partir d'une maquette numérique enrichie d'information sur les interfaces géométriques entre composants et d'information sur les propriétés fonctionnelles de l'assemblage, l'approche proposée dans ce manuscrit est d'ajouter un niveau supplémentaire d'enrichissement en fournissant une représentation structurelle de haut niveau de la forme des composants CAO. Le principe de cet enrichissement est d'extraire un graphe de construction de modèles CAO B-Rep de sorte que les processus de génération de forme correspondants fournissent des primitives volumiques directement adaptées à un processus d'idéalisation. Ces primitives constituent la base d'une analyse morphologique qui identifie dans les formes des composants à la fois des sous-domaines candidats à l'idéalisation mais également les interfaces géométriques qui leurs sont associées. Ainsi, les modèles de composants et leurs représentations géométriques sont structurés. Ils sont intégrés dans la maquette numérique enrichie qui est ainsi contextualisée pour la simulation par EF. De cette maquette numérique enrichie, les objectifs de simulation peuvent être utilisés pour spécifier les opérateurs géométriques adaptant les composants et leurs interfaces lors de processus automatiques de préparation d'assemblages. Ainsi, un nouveau procédé d'idéalisation de composant unitaire est proposé. Il bénéficie de l'analyse morphologique faite sur le composant lui fournissant une décomposition en sous-domaines idéalisables et en interfaces. Cette décomposition est utilisée pour générer les modèles idéalisés de ces sous-domaines et les connecter à partir de l'analyse de leurs interfaces, ce qui conduit à l'idéalisation complète du composant. Enfin, le processus d'idéalisation est étendu au niveau de l'assemblage et évolue vers une méthodologie de pré-traitement automatique de maquettes numériques. Cette méthodologie vise à exploiter l'information fonctionnelle de l'assemblage et les informations morphologiques des composants afin de transformer à la fois des groupes de composants associés à une même fonction ainsi que de traiter les transformations d'idéalisation de l'assemblage. Pour démontrer la validité de la méthodologie, des opérateurs géométriques sont développés et testés sur des cas d'application industriels.

Diese Beobachtung steht im Kontrast zu nicht-serialisierenden Sprachen, wie dem Englischem, in welchen nur einer der beiden Bedeutungskomponenten durch das Hauptverb ausgedrückt wird. Im Zuge einer Untersuchung der mor-phosyntaktischen semantischen Eigenschaften zweier Typen von Resultativkonstruktionen, resultative Sekundärprädikation und die means-Konstruktion, entwickelt diese Arbeit einen neuen konfigurationel-len Ansatz innerhalb der Distributed Morphology, in welchem sprachübergreifende Variation als Inter-aktion von morphosyntaktischer und semantischer Komposition der jeweiligen Bedeutungskomponen-ten in Abhängigkeit von sprachspezifischen Restriktionen auf Wurzelbedeutung und Argumentstruktur beschrieben werden kann. Mit Hilfe eigener Feldforschung zeige ich, dass trotz der oberflächlichen Unterschiede zwischen serialisierenden und nicht-serialisierenden Sprachen Ozeanische Resultativkon-struktionen die zugrundeliegende Struktur der means-Konstruktionen aufweisen, in welchen das Man-nerverb an das kausative Hauptverb adjungiert wird und das darin enthaltende, unterspezifizierte kausa-tive Ereignis spezifiziert. Folglich unterscheiden sich beide Sprachtypen nicht signifikant in ihrer mor-phosyntaktischen und semantischen Komposition mit weitreichenden Implikationen für eine sprachübergreifende Typologie von Resultativkonstruktionen.<br>This dissertation approaches the event and argument structure of resultative construction (e.g., Peter wiped the table clean) from the perspective of two understudied and endangered Oceanic languages, Daakaka and Samoan, in which both the manner and result components are realized by verbal predicates, i.e. resultative serial verb constructions (RSVCs). This observation contrasts with non-serializing languages, such as English, in which only one of the two meaning components is expressed by the main verb. By examining the morphosyntactic and semantic properties of two types of resultative construc-tions, namely resultative secondary predication and means constructions, I develop a novel configurational analysis within the generative framework of Distributed Morphology that models cross-linguistic variation in terms of the morphosyntactic size and the semantic composition of the respective meaning components and their interaction with idiosyncratic requirements on roots and argument structure. Based on original fieldwork, I demonstrate that despite the superficial differences in categorial status, Oceanic RSVCs are an instance of the means construction, in which the manner verb directly adjoins to a causative verb modifying the underspecified causing event entailed in the event structure of the causa-tive predicate. Consequently, serializing and non-serializing languages do not vary significantly in their morphosyntactic and semantic composition with further implications for the typology of resultatives in the world’s languages.

Ziel dieser Arbeit ist die Rekonstruktion der phylogenetischen Verwandtschaftsverhältnisse mit Hilfe molekularer Datensätze für die Phyllopoda, die zusammen mit den Anostraca die Branchiopoda bilden und der Astacoidea (Astacida), einer Teilgruppe der Flusskrebse. Folgende molekulare Marker kamen zum Einsatz: 1) Für die Phyllopoda: Die 3. Domäne der mitochondrial codierten 12S rRNA, unter Berücksichtigung von Sekundärtrukturinformationen, das nukleare Gen EF-1 alpha und die Positionen von Introns im Gen EF-1 alpha. 2) Für die Astacoidea: Die 3. Domäne der 12S rRNA und das mitochondrial codierte Gen cox1. Durch die Wahl der molekularen Marker, die mit unterschiedlichen computerkladistischen Methoden ausgewertet wurden, konnten für die meisten Fragen eine eindeutige und im Fall der Astacoidea überraschende phylogenetische Aussage getroffen werden. Die gewonnenen Hypothesen werden ausführlich im Licht morphologischer Hypothesen diskutiert.<br>The phylogenetic relationships of the higher arthropod taxa are still of special interest. Especially the interrelationships of the different Crustacea taxa have long been debated. The focus of this investigation is to make a contribution to the phylogenies of two Crustacea taxa using molecular markers: The Phyllopoda which belong together with the Anostraca to the branchiopods, and of the Astacoidea, one of the two higher crayfish taxa (Astacida). The following molecular markers were used: 1) Phyllopoda: the 3rd domain of the mitochondrial encoded 12S rRNA taking into account informations of the secondary structure, the nuclear encoded proteingene EF-1 alpha and the positions of introns found in the coding region of EF-1 alpha. 2) Astacoidea: the 3rd domain of the 12S rRNA and the mitochondrial encoded proteingene cox1. The choice of the mentioned markers in combination with different computercladistical methods allowed to give a satisfying, and in the case of the Astacoidea a more surprising answer to most addressed phylogenetic questions. The gained hypotheses are then discussed in detail in the light of morphological features and hypotheses.

Cisplatin is a chemotherapy drug effective against several forms of cancer, but can also cause serious side-effects, including nephrotoxicity and ototoxicity. Curcumin, a natural plant compound, can increase cisplatin’s anti-cancer activity and counteract cisplatin’s deleterious effect on the auditory and renal systems. Unfortunately, curcumin exhibits poor bioavailability, which has promoted interest in the development of synthetic curcumin analogs (curcuminoids) that are soluble, target cancer, and do not cause side effects. This study investigated whether the curcuminoid (3E,5E)-3,5-bis[(2-fluorophenyl) methylene]-4-piperidinone (EF-24) increases the anti-cancer effects of cisplatin against a human ovarian cancer cell line (A2780) and its cisplatin-resistant counterpart (A2780cis), while preventing cisplatin-mediated side effects in a human kidney cell line (HEK-293T) and a mouse auditory hybridoma cell line (HEI-OC1). The effect of cisplatin and EF-24 on cellular viability was measured using the colorimetric 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide (MTT) assay. The expression and activity of signal transduction proteins in several apoptotic pathways was measured using caspase luminescence assays. Reactive oxygen species (ROS) production was also measured using flow cytometry. Our data suggest that cisplatin and EF-24 are effective against ovarian cancer cell lines, but both compounds may also have adverse effects on auditory and renal cells. This project provides relevant information that may improve our understanding of how these compounds function in different tissues, facilitating improved cancer treatment and circumvention of side effects commonly associated with cisplatin treatment.

Le gene codant pour le facteur d'elongation de la traduction ef-1alpha a1 chez arabidopsis thaliana a ete utilise comme systeme modele pour etudier les mecanismes de regulation de mise en place coordonnee des elements de la machinerie de traduction chez les angiospermes. Le role de deux cis-elements de ce promoteur, boite tef et boite telo, font l'objet de cette these. Des sequences homologues a la boite tef sont observees dans plusieurs promoteurs de genes codant pour des elements de la machinerie de traduction ou impliques dans le controle de l'etat redox de la cellule. La plupart de ces genes sont specifiquement exprimes ou sur-exprimes en reprise de croissance ou dans les tissus meristematiques. In vitro et in vivo nous avons montre que l'element tef est un cis-element ubiquitaire chez les angiospermes participant a l'activation transcriptionnelle lors de la reprise des divisions cellulaires. La sequence consensus de cet element est arggry annnnngtaa. L'importance de ce mecanisme de regulation nous a conduit a envisager la caracterisation des genes codant pour les proteines interagissant avec l'element tef. Nous rapportons ici les donnees preliminaires concernant la purification de ces proteines. La boite telo, element homologue a 1,5 unite de repetition des telomeres de plantes, est frequemment associee a l'element tef dans divers promoteurs. La deletion de cet element n'affecte pas l'expression. Toutefois nous montrons que des proteines nucleaires reconnaissent specifiquement ce motif. Par ailleurs une analyse statistique revele que la boite telo est sur-representee dans le genome d'arabidopsis et est preferentiellement localisee dans les regions 5' des genes. Cette localisation a ete confirmee par l'isolement et l'analyse de clones de cdna contenant des sequences telo. Nous proposons un modele permettant d'expliquer la sur-representation et la localisation de ces motifs a l'interieur du genome

Calmodulin is an essential EF-hand protein with a helix-loop-helix calcium binding motif. Understanding Ca(II) dependent activation of calmodulin and other EF-hand proteins is limited by Ca(II)-induced conformational change, multiple and cooperative binding of Ca(II) ions, and interactions between the paired EF-hand motifs. The goal of this research project is to probe key determinants for calcium binding properties and pairing interactions at the site specific level using a grafting approach and high resolution NMR. An individual Ca(II) binding site of the EF-hand motifs of calmodulin was grafted into a non-calcium dependent protein, CD2, to bypass limitations associated with natural EF-hand proteins and peptide fragments. Using high resolution NMR, we have shown that the grafted EF-loop III of calmodulin in the host protein retains its native conformation with a strong loop and β-conformation preference. Grafted ligand residues in the engineered protein are directly involved in binding of Ca(II) and La(III). The NMR studies support our hypothesis that both ligand arrangement and dynamic properties play essential role in tuning Ca(II) binding affinities. Using pulse-field diffusion NMR and protein engineering, we further demonstrated that grafted EF- loop remains as a monomer. Although the EF-loop with flanking helices dimerizes in the presence of Ca(II). Additionally, removal of conserved hydrophobic residues at the flanking helices of the EF-hand motif leads to be monomer in the absence and presence of metal ions. Our results suggest that conserved hydrophobic residues are essential for the pair-paired interaction in the coupled EF-hand protein. We have shown that our developed grafting approach can be applied to probe intrinsic Ca(II) binding affinities of different Ca(II) binding sites.

Ces dernières années, l’utilisation de la simulation numérique s’est généralisée dans le processus de développement des produits. Auparavant, les simulations étaient réalisées séquentiellement et n’intervenaient qu’en phase avale de l’activité de conception dans une optique de validation. Les approches actuelles tendent vers une simulation concourante avec l’intégration de paramètres multiples telles que les analyses stochastiques (statistiques), les simulations d’optimisation multi-physiques ou les analyses multi-scénarios. L’impact direct de ces approches et l’augmentation du nombre de simulations et de variantes de simulation, la croissance de la complexité des modèles de simulation, et l’augmentation du volume de données. Ceci a généré un fort besoin de gestion, de traçabilité, d’échange et de synchronisation des données de simulation pour garantir l’obtention des résultats indispensables aux prises de décision. Ces préoccupations sont à l’origine des travaux présentés dans ce mémoire, lequel spécifie un environnement de gestion de données de simulation dans une approche PLM. Ce dernier a été défini dans l’optique de favoriser l’intégration des données de CAO et d’analyse par éléments finis. Cet environnement, nommé EGDS, propose une modélisation novatrice des cycles de simulation à travers l’adaptation et l’enrichissement des méthodes de gestion des données issues des PLM. Afin de garantir son interopérabilité, un format neutre d’échange de données de gestion de simulation s’appuyant sur la norme STEP est également spécifié<br>Previously, simulations were sequentially carried out and only intervene downstream from design activity in a validation goal. The current approaches move towards a concurrent simulation with integration of multiple parameters such as stochastic analyses (statistical), multi-physics simulations of optimization of multi-scenarios analyses. The direct impact of these approaches is the increase in the number of simulation and alternatives of simulation, the growth of the complexity of the models of simulation, and the increase in the volume of data. These evolutions created a strong need for management, traceability, exchange and synchronization of the simulation data to guarantee obtaining of the decision-makings results. These concerns are at the origin of the work presented in this memory. This work specifies an environment of simulation data management in a PLM approach which was defined in optics to support integration of CAD data and finite element analysis. This environment, named EGDS, proposes an innovative modelling of simulation cycles through the adaptation and the enrichment of data management methods of the PLM approach. In order to guarantee its interoperability, EGDS also implements a neutral format dedicated to simulation data management exchanges based on STEP standard

Une approche de simulation numérique multi-échelles EF2 fondée sur la théorie de l'homogénéisation périodique a été développée pour prédire la réponse globale couplée mécanique et thermomécanique fortement non linéaire des structures composites 3D. La stratégie de calcul intègre les effets de la microstructure périodique en introduisant l'architecture des renforts et les lois constitutives locales. Les lois de comportement des constituants utilisées obéissent aux lois de matériaux standards généralisées et sont formulées dans un cadre de la thermodynamique des processus irréversibles (TPI). Les équations caractéristiques (équilibre et lois de la thermodynamique) sont formulées sous l'hypothèse des petites déformations et rotations, et résolues simultanément de façon incrémentale aux deux échelles (microscopique et macroscopique). Sur le plan numérique, une implémentation au moyen de routines UMAT imbriquées (Méta-UMAT) a été développée et combinée à une technique de parallélisation dans le code de calcul Abaqus/Standard. La stratégie de calcul multi-échelles est appliquée pour simuler la réponse globale de structures composites 3D soumises à des trajets de chargement thermomécaniques complexes. Les structures composites sont constituées d’une matrice polymère thermoplastique viscoélastique-viscoplastic avec endommagement ductile et renforcées par différents types de renforcements (fibres courtes ou tissus). L’endommagement anisotrope dans les torons de tissu est modélisé à travers une approche micromécanique permettant de suivre l’évolution de la densité de micro-fissures transverses. Cette stratégie de calcul peut être déployée sur les structures en matériaux composites ayant une microstructure périodique et dont les phases présentent différents types des lois de comportement non linéaires (rhéologie, mécanismes d'endommagement et couplage thermomécanique). Les capacités de l'approche multi-échelles sont démontrées en comparant les prédictions numériques aux résultats expérimentaux en termes de réponse globale et de champs de déformation macroscopiques et microscopiques. Les performances de l'approche sont également illustrées à travers l'accès aux répartitions spatio-temporelles des variables internes à l'échelle de la microstructure ainsi que la dissipation intrinsèque dans les phases constitutives<br>A multi-scale FE2 approach based on the periodic homogenization theory is developed to predict the overall response of nonlinear mechanical and fully coupled thermomechanical 3D composite structures. The computational strategy integrates the periodic microstructure effects by introducing the architecture of the reinforcement and the local constitutive laws.The considered constituents' constitutive laws obey generalized standard materials laws and are formulated within the framework of thermodynamics of irreversible processes. The characteristic equations (equilibrium and thermodynamics laws) are formulated under the assumption of small strains and rotations. They are solved simultaneously at both scales (microscopic and macroscopic) using an incremental scheme. For the numerical implementation, an advanced Meta-UMAT subroutine is developed and combined with a parallelization technique in the finite element commercial software Abaqus/Standard. The multi-scale computational strategy is applied to simulate the overall response of 3D composite structures under complex thermomechanical loading paths. The composite structures consist of thermoplastic polymer matrix with viscoelastic-viscoplastic behavior and ductile damage, reinforced by different types of reinforcements (short fibers or woven fabrics). The anisotropic damage within the yarns is modeled through a micromechanical approach to follow the transverse micro-cracks density evolution. This computational strategy is deployed on composite structures having periodic microstructure, whose phases exhibit different types of nonlinear behavior laws (rheology, damage mechanisms and thermomechanical coupling). The capabilities of the multi-scale approach are demonstrated (i) by comparing numerical predictions with experimental results in terms of global response, macroscopic and microscopic strain fields, and (ii) through the access to spatio-temporal distributions of internal variables at the microstructure scale as well as the intrinsic dissipation in the constitutive phases

L'objectif est d'apporter une réponse aux problèmes rencontrés lors de l'analyse non linéaire en grandes transformations des structures monolithiques et sandwichs minces, épaisses ou volumiques. Le point commun aux divers éléments provient de l'utilisation d'une FLAI, en considérant les effets de CT et la contrainte normale pour prendre correctement en compte les différents phénomènes à travers l'épaisseur. Des médications et des techniques efficaces ont été utilisés pour améliorer les performances de calcul et éviter les problèmes de blocages numériques. La robustesse et l'efficacité des différents modèles sont démontrées sur un grand nombre de tests de structures monolithiques ou stratifiées, en statique implicite ou dynamique explicite en 2D et 3D<br>This thesis deals with the development and validation of a family of FE solid-shell built from a 3D formulation using three degrees of freedom of displacement per node. The goal for these developments is to provide an answer to the problems encountered when analyzing large nonlinear transformations of monolithic and sandwich thin, thick and 3D structures. The common point to various elements is the use of the ULF, while considering the effects of TS and the normal stress to properly take into account the different phenomena across the thickness. Changes and effective techniques have been used to improve performance and avoid locking. Le coefficient de couplage électromécanique (CCEM) est un paramètre essentiel pour la description des matériaux piézoélectriques, il traduit la conversion d'énergie électrique en énergie mécanique et vice versa. Ce coefficient de couplage est étudié et déterminé dans le cadre de cette thèse pour des céramiques piézoélectriques. Ces dernières sont utilisées pour trois structures différentes ; la première structure étudiée est constituée d'une poutre longue et mince avec des petits patchs collés symétriquement sur les deux faces de la poutre en Aluminium, la seconde structure se compose d'une poutre courte et épaisse avec deux grands patchs. La dernière structure étudiée est une plaque composite multicouche du type aéronautique avec un seul grand patch. Ces trois structures ont été étudiées afin de déterminer le coefficient de couplage électromécanique effectif qui est considéré comme un indicateur de performance de l'amortissement passif shunté. Ce coefficient de couplage a été évalué de différentes manières en utilisant différents paramètres dont les conditions limites électriques, les propriétés élastiques des patchs, les propriétés modales de la poutre seule ainsi que les facteurs de couplages piézoélectriques. Une première étude expérimentale a été menée sur la poutre longue pour deux types de configurations en court circuit et circuit ouvert pour identifier ses propriétés modales. La poutre longue a été simulée pour deux types de polarisations, identiques et opposées, et simulée dans les deux codes Ansys® et Abaqus®. L'influence de la condition d'équipotentielle sur le coefficient de couplage a été étudiée. Une seconde campagne expérimentale et numérique sur une autre structure a été nécessaire pour valider les résultats obtenus. Pour pouvoir atteindre cet objectif, il était nécessaire de travailler sur une structure plus courte et plus rigide. Ainsi, la poutre courte a été simulée dans Ansys® et les résultats obtenus ont confirmé la nécessité de prendre en compte l'équipotentialité sur les faces des patchs. Cette condition a pour effet de réduire le couplage électromécanique et parfois de découpler certains modes. L'écart résultant de la corrélation expérimentale / numérique des deux poutres instrumentées a incité à recaler les modèles numériques. Ce recalage peut se présenter sous trois formes : mécanique en remplaçant l'encastrement par des ressorts linéaires, électrique en remplaçant les capacités fournies par le fabricant par les valeurs mesurées expérimentalement et électromécanique en utilisant les deux recalages précédents simultanément. Les deux poutres ont ensuite été simulées en déformations planes et contraintes planes et recalées afin d'approcher les résultats expérimentaux. L'étude de ces deux structures a permis de confronter les différentes méthodes d'évaluation du CCEM effectif, d'évaluer l'influence de l'équipotentialité sur les faces des électrodes et de comparer les simulations bidimensionnelles aux tridimensionnelles. Une plaque composite multicouche du type aéronautique a été ensuite étudiée pour généraliser la méthode d'évaluation du CCEM effectif pour les structures minces composites. La plaque seule a d'abord été simulée dans Ansys® pour valider le modèle numérique. Des tests sur la structure adaptative ont ensuite été menés pour l'évaluation du CCEM expérimental. La position choisie du patch a été déterminée par une analyse de l'énergie de déformation de la plaque seule pour les modes d'intérêt. Cette méthode de placement du patch s'est avérée efficace dans le sens où elle a conduit à des CCEM effectifs élevés pour certains modes de la bande de fréquence retenue.

La phosphorylase kinase (PhK), enzyme clé de la glycogénolyse, est un complexe hexadécamérique formé de quatre sous-unités différentes (alpha, beta, gamma, delta)4. L’activité catalytique portée par la PhK gamma est modifiée par l’action des sous-unités apparentées alpha et beta (4/5 de la masse de l’enzyme). Des mutations dans la PhK conduisent à une maladie de stockage du glycogène (GSD) de type IX, le désordre le plus répandu dans le métabolisme du glycogène. Peu de choses sont connues sur la structure de l’holoenzyme et sur celle des sous-unités de la PhK (exception faite du domaine catalytique de la PhK gamma et de la PhK delta (calmoduline)). Dans cette thèse, nous avons utilisé des méthodes d’analyse de séquences pour révéler des caractéristiques structurales et fonctionnelles des sous-unités alpha et beta. Nous avons ainsi confirmé l’appartenance du premier domaine (A) à la famille des glucoamylases (Glycosyl Hydrolase 15) et montré que les domaines C et D sont reliés aux protéines « calcineurin B-like », membres de la famille EF-hand impliqués dans la régulation calcium-dépendante de kinases. La plupart des mutations faux-sens affectant la PhK alpha et conduisant à une déficience en PhK sont situées dans les sites actifs de ces domaines, suggérant qu’elles pourraient avoir un impact direct sur leurs fonctions. Par ailleurs, nous avons recalé dans le volume obtenu par cryo-microscopie électronique (résolution 9. 9 Angstrom) les structures 3D des différents domaines de la PhK en nous aidant des contraintes décrites dans la littérature. L’ensemble de ces résultats ouvre de nouvelles perspectives pour comprendre comment les sous-unités de la PhK régulent son activité<br>Phosphorylase kinase (PhK) is a key enzyme in glycogenolysis. PhK is a hexadecameric complex, made of four different subunits (alpha, beta, gamma, delta)4. Catalytic activity is conferred by the gamma subunit and is modified by the two related regulatory subunits alpha and beta which together account for ~ 4/5 of the PhK mass. Mutations in PhK lead to Glycogen Storage Disease (GSD) type IX, which is the most frequently encountered disorder of glycogen metabolism. The structural features of the quaternary structure of the holoenzyme and the PhK subunits, except for the catalytic domain of the PhK gamma and the PhK delta subunit are poorly understood. Here we have used sensitive methods of sequence analysis to unravel hidden structural and functional features of the PhK alpha and beta subunits. We confirm that the first domain (A) belongs to the glucoamylase family (Glycosyl Hydrolase 15) and show domains C and D are related to calcineurin B-like proteins, which are EF-hand family members involved in the Ca2+-dependent regulation of kinases. Mutations leading PhK deficiency, mostly missense mutations in PhK alpha, are located within the predicted active sites of these domains, suggesting that they may have a direct impact on their predicted functions. Furthermore, we docked the 3D structures of the different PhK domains into the volume obtained by cryo-electron microscopy at 9. 9 Angstrom resolution constraining with various interaction data reported in the literature. Altogether, our findings open new perspectives to understand how the different PhK subunits may regulate the holoenzyme activity

Based on a conforming finite element method, a new triangular finite shell element is developped :-with an exact geometric description using an explicit map,-based on a refined model to satisfy interlayer continuity for both displacements and transverse stresses, and free edges conditions for the shell, avoiding the use of correction factors, and characterized by : its simplicity to be used, its robustness according to classical numerical pathologies like locking, spurious modes and so on, its capability to correctly predict both displacements, strains and stresses. The previous qualities are demonstrated through numerical tests issued from litterature and by comparing numerical and experimental results. Dans le cadre de l'étude par éléments finis de structures aéronautiques soumises à l'impact, la prédiction des modes de ruine engendrés au sein ou au voisinage des assemblages rivetés, telle que la déchirure d'une tôle le long d'une ligne de rivets, s'avère, encore aujourd'hui, délicate. En particulier, une difficulté majeure est liée à l'incompatibilité de l'échelle de l'aéronef et de celle de l'assemblage riveté. En effet, alors que la modélisation EF fine de l'assemblage riveté est requise pour localiser l'initiation et la propagation de la rupture, le nombre d'éléments nécessaire pour mailler la structure devient irréaliste. Il est donc proposé de formuler un super-élément fini perforé, permettant de localiser les champs mécaniques dans une structure soumise à un chargement de dynamique rapide. Le premier chapitre, de nature bibliographique, introduit les méthodes numériques permettant la représentation des discontinuités, et les notions relatives à la formulation de super-éléments finis. Il permet l'introduction et le choix pour la poursuite des développements de l'élément fini de plaque perforé à 8 noeuds de Piltner. Le deuxième chapitre met en évidence les hypothèses liées à la formulation des fonctions d'interpolation de cet élément, ainsi que leur lien avec les équations de la mécanique des solides. De plus, l'étude numérique de ces fonctions (tronquées à l'ordre 4) fournit des résultats jugés suffisamment satisfaisants pour la poursuite des travaux. Le chapitre III met alors en évidence les hypothèses liées à la formulation du principe variationnel de cet élément. Il est conclu qu'il est restreint aux calculs élastostatiques, et qu'il ne permet pas la modélisation d'un assemblage riveté. Les résultats numériques fournis par la formulation de Piltner à 8 noeuds (élastostatique), issus d'une implantation dans ZéBuLoN, étant jugés suffisamment satisfaisants pour la poursuite des travaux, les hypothèses limitant l'élément de Piltner à l'élastostatique sont levées, puis la nouvelle formulation est adaptée aux calculs de dynamique rapide (chapitre IV).

Downstream regulatory antagonist modulator (DREAM) is a calcium sensing protein that co-assembles with KV4 potassium channels to regulate ion currents as well as with DNA in the nucleus, where it regulates gene expression. The interaction of DREAM with A-type KV4 channels and DNA has been shown to regulate neuronal signaling, pain sensing, and memory retention. The role of DREAM in modulation of pain, onset of Alzheimer’s disease, and cardiac pacemaking has set this protein as a novel therapeutic target. Moreover, previous results have shown a Ca2+ dependent interaction between DREAM and KV4/DNA involving surface contacts at the N-terminus of DREAM. However, the mechanisms by which Ca2+ binding at the C-terminus of DREAM induces structural changes at the C- and N-terminus remain unknown. Here, we present the use of biophysics and biochemistry techniques in order to map the interactions of DREAM and numerous small synthetic ligands as well as KV channels. We further demonstrate that a highly conserved network of aromatic residues spanning the C- and N-terminus domains control protein dynamics and the pathways of signal transduction on DREAM. Using molecular dynamics simulations, site directed mutagenesis, and fluorescence spectroscopy we provide strong evidence in support of a highly dynamic mechanism of signal transduction and regulation. A set of aromatic amino acids including Trp169, Phe171, Tyr174, Phe218, Phe235, Phe219, and Phe252 are identified to form a dynamic network involved in propagation of Ca2+ induced structural changes. These amino acids form a hydrophobic network connecting the N- and C-terminus domains of DREAM and are well conserved in other neuronal calcium sensors. In addition, we show evidence in support of a mechanism in which Ca2+ signals are propagated towards the N-terminus and ultimately lead to the rearrangement of the inactive EF-hand 1. The observed structural motions provide a novel mechanism involved in control of the calcium dependent KV4 and DNA binding. Altogether, we provide the first mechanism of intramolecular and intermolecular signal transduction in a Ca2+ binding protein of the neuronal calcium sensor family.

La représentation mixte de forme qui en découle permet de transférer quelques hypothèses et objectifs du problème considéré en termes de contraintes qui doivent satisfaites pendant la génération du modèle intermédiaire, où, plus tard dans le processus, pendant la génération modèle de simulation où effectuer l'analyse mécanique. De plus, la relation qu'il y a entre l'application d'un ( mécanique et la suppression d'un sous-domaine de forme est mise en évidence. En particulier, la formulation critère mécanique a posteriori supportant les transformations de forme est fournie.

Store-operated calcium entry (SOCE) is the major Ca2+ entry pathway in most non-excitable cells maintaining prolonged elevation of cytosolic Ca2+ levels required for gene transcription. SOCE is activated by the loss of endoplasmic reticulum (ER) Ca2+ through stromal interaction molecules (STIM), ER-membrane associated Ca2+ sensors. In humans, STIM1 and STIM2 share 65% sequence similarity but differentially regulate SOCE. Biophysical studies on the luminal Ca2+-binding region suggests that STIM2 EF-SAM is more stable than STIM1. The NMR structure of Ca2+-loaded STIM2 EF-SAM determined in this work suggests a more stable SAM and a tighter EF-hand and SAM interaction in STIM2 may be account for its higher stability. Chimeric swapping of the EF-hand and SAM domains generates an unstable ES211. Introducing ES211 into cherryFP-STIM1 shows constitutive puncta which activate SOCE independent of ER depletion. The current work demonstrates that the instability of the EF-SAM plays an important role in regulating SOCE initiation.

The term executive functions is used as a general construct to characterize a set of cognitive functions that comprise several distinct subcomponents, and whose efficient performance is crucial for the control of thoughts, behaviors and emotions. Executive functions depend on the maturation of the frontal lobes of the brain, especially the prefrontal cortex, although they also involve the parietals, knowing that the front parietal network is strongly associated with it.This exploratory study had as main purpose to contribute to the validation of an executive functions inventory for the Portuguese population, namely, The Behavior Rating Inventory of Executive Function – Second Edition: Teacher Form (BRIEF-2; Gioia, Isquith, Guy, & Kenworthy, 2015).Precision analyzes were performed through the study of internal consistency (Cronbach's alpha) and construct validity (exploratory and confirmatory factor analysis), based on a sample of 114 children and adolescents.The results obtained revealed the good psychometric properties of this instrument in terms of internal consistency, presenting very good values. Regarding the structure of the instrument, the original study´s structure was not replicated, and it was found that the factorial model that is more adequate is constituted, not by three, but by two factors: Cognitive Regulation Index (Factor 1) and Behavior Regulation Index + Emotion Regulation Index (Factor 2), which explains 79.19% of the total variance.In conclusion, the results found in this exploratory study, especially in regards to internal consistency, reveal psychometric robustness and indicate that BRIEF-2, Teacher Form, may be a good option for the study of executive functions in Portuguese children and young people, between 6 and 18 years old.<br>Ao longo das últimas décadas, diversas investigações têm demonstrado interesse pela pesquisa e compreensão do sistema executivo. O termo funções executivas é utilizado como um construto geral para caraterizar um conjunto de funções cognitivas que comportam vários subcomponentes distintos, e cujo desempenho eficiente é crucial para o controlo de pensamentos, comportamentos e emoções. As funções executivas dependem da maturação dos lobos frontais do cérebro, sobretudo, do córtex pré-frontal, embora também envolvam os lobos parietais, sabendo-se que a rede frontoparietal lhe está fortemente associada.Este estudo exploratório teve como principal objetivo contribuir para a validação de um inventário de funções executivas para a população Portuguesa, designadamente, o Inventário de Avaliação Comportamental de Funções Executivas - 2ª Edição: versão para Professores (BRIEF-2, The Behavior Rating Inventory of Executive Function - Second Edition; Gioia, Isquith, Guy, & Kenworthy, 2015).Foram realizadas análises no âmbito da precisão através do estudo da consistência interna (alfa de Cronbach) e da validade de construto (análise fatorial exploratória e confirmatória), com base numa amostra composta por 114 crianças e adolescentes.Os resultados obtidos evidenciaram as boas propriedades psicométricas deste instrumento ao nível da consistência interna, observando-se valores muito bons. Relativamente à estrutura do instrumento, não foi replicada a do estudo original, tendo-se constatado que o modelo fatorial que melhor se adequa é constituído, não por três, mas sim por dois fatores: Índice de Regulação Cognitiva (Fator 1) e Índice de Regulação Comportamental + Índice de Regulação Emocional (Fator 2), que explicam 79.19% da variância total.Concluindo, os resultados observados neste estudo exploratório, principalmente no que se refere à consistência interna, revelam robustez psicométrica e indicam que o BRIEF-2, versão para Professores, poderá ser uma boa escolha para o estudo das funções executivas nas crianças e jovens portugueses, com idades compreendidas entre os 6 e os 18 anos.

O2 atmosphere) as well as reactive grown iron oxide films will be analyzed in detail by low energy electron diffraction (LEED) and scanning tunneling microscopy (STM). The stoichiometry at the surface of the iron oxide films will be determined by X-ray photoelectron spectroscopy (XPS) and Auger electron spectroscopy (AES). The main focus of this work is to shed light on the question whether the growth of iron oxide films on Ag(001) is accompanied by the formation of strain reducing dislocation
networks, or superstructures as found for other metal substrates in former studies. Here, we will distinguish between Fe films which were post deposition annealed in
a thin O2 atmosphere and reactively grown iron oxide films.

The high temperature adsorption of oxygen and the room temperature adsorption of CO on the Pt3Sn(111) and Pt3Sn(110) surfaces have been investigated by scanning tunneling microscopy (STM), low-energy electron diffraction (LEED), and Auger electron spectroscopy (AES). Beforehand the structure of the clean surfaces has been reviewed.
After exposure to several 1000 L O2 at sample temperatures of about 750 K on both Pt3Sn(111) and (110) an ultra-thin Sn-O surface layer is formed. For the (111) X-ray photoelectron spectroscopy (XPS) indicates that this layer does not yet exhibit oxide properties. STM topographs of the Sn-O phase show on both surfaces meshes of highly corrugated protrusions commensurate with the substrate. In the case of the (111), after additional thermal annealing with STM and LEED a (4 × 4) reconstruction is observed, that is due to a (2 × 2) supermesh of depressions in the p(2 × 2) mesh of protrusions. This structure is similar to findings reported for the oxidation of Sn/Pt(111) surface alloys. X-ray photoelectron diffraction (XPD) measurements in comparison with simulations yield a tentative model for the (111) Sn-O layer.
On the Pt3Sn(110) surface after oxygen exposure a c(2 × 2) hexagonal grid of protrusions with regard to the (2 × 1) substrate is observed with STM and LEED. STM reveals the existence of domains due to two equivalent positions of the Sn-O layer relative to the substrate. The domain boundaries zigzag around the [1-10] direction. The Sn-O layer can on both surfaces be removed by thermal annealing to more than 1050 K.
After CO adsorption at room temperature on both Pt3Sn(111) and (110) adsorbate structures are observable with the STM. On the (111) two different types of structures are found: ordered patches of protrusions and unordered clusters. These structures are seen only on (√3 × √3)R30° substrate regions, not on p(2 × 2) regions. Surprisingly on the (110) the CO molecules mostly arrange in dimers. For both (111) and (110) saturation coverage is already reached at about 30% of a closed monolayer. The CO can be desorbed by slightly heating the samples to about 400 K. STM topographs show that on both surfaces CO adsorbes in Pt sites, not on Sn.
It was possible to observe the CO adsorption on the (110) directly live with the STM. The observed adsorption processes hint to a dimer formation mechanism where a preadsorbed monomer and a CO molecule form the gas phase or a precursor phase stick together.
When on partially Sn-O phase covered Pt3Sn(111) and (110) surfaces CO is adsorbed at room temperature, the respective structures coexist. Neither is CO observed on the Sn-O phase nor does a reaction between CO and O occur.

Wir nutzen diese Methoden um die Adsorption von TMA auf der Rutil TiO2-Oberfläche zu untersuchen, optische Spektren von TiO2 zu berechnen und um die Adsorption von [7]-HCA auf der Calcit(10-14)-Oberfläche zu verstehen. Weiterhin beschäftigen wir uns intensiv mit PTCDA auf Ag(111), welches mit einer chemisch kontaktierten STM-Spitze manipuliert wird.

A fundamental change in societal values and economic structures is required to address increasing pressures on ecosystems and natural resources. Transition research has developed in the last decades to analyze the co-dynamics of technological, institutional, social and economic elements in the provision of key functions such as energy, water and food supply. This doctoral dissertation provides conceptual and methodological contributions to the pro-active governance of sustainability transitions. Three research gaps are identified that are addressed in this dissertation. First, a comprehensive conceptualization of learning in sustainability transitions is currently missing that comprises learning at multiple societal levels (ranging from individuals to policy-actors). Learning concepts are often not explicitly discussed in transition research even though learning is considered as fundamental for innovation processes, niche formation and development as well as breakthrough and diffusion of innovations. Second, methods for the analysis and design of transition governance processes are lacking that specify case-specific intervention points and roles of actors in the implementation of innovations. Third, participatory modeling approaches are only applied to a limited extent in transition research despite a high potential for supporting communication and learning.
The conceptualization of multi-level learning developed in this doctoral research conceptualizes learning at different societal levels as specific learning contexts ranging from individual and group contexts to organizational and policy contexts. The conceptual framework further differentiates between learning processes, intensity, objects, outcomes, subjects and factors, allowing for a more detailed analysis of learning within and across learning contexts. Thus, learning contexts can be linked by processes that involve actors from different learning contexts (e.g., community groups and policy-makers), as well as exchanges of physical aspects, institutions and knowledge (in the form of ‘learning factors’). This research has also provided a classification of model uses in transition research that supports a purposeful discussion of the opportunities of modeling and promising future research directions.
The methodology developed in this doctoral research aims at the analysis and design of transition governance processes by specifying the various opportunities to contribute to sustainability transitions through purposeful action at different societal levels, as well as related roles of stakeholders in implementing such processes of change. The methodology combines different streams of previous research: 1) a participatory modeling approach to identify problem perceptions, case-specific sustainability innovations as well as related implementation barriers, drivers and responsibilities; 2) a systematic review to identify supportive and impeding learning factors from the general literature that can complement case-specific factors; and 3) a method for the analysis and design of case-specific transition governance processes. Three case studies in Canada (topic: sustainable food systems), Cyprus (water-energy-food nexus) and Germany (sustainable heating supply) have been selected to test and iteratively develop the methodology described above.
The results for each case study reveal that there are learning objects (i.e., learning requirements) in all learning contexts, which underscores the importance of multi-level learning in sustainability transitions, ranging from the individual to the group, organizational and policy levels. Actors have various opportunities to actively facilitate societal transformations towards sustainable development either directly through actions at their particular societal levels (i.e., context-internal learning) or indirectly through actions that influence learning at other societal levels. In fact, most of the learning factors require cooperation across learning contexts during the implementation process. The comparing of learning factors across case studies underline the importance of several factor categories, such as ‘physical a ‘disturbance or crisis’, ‘information and knowledge’. Of the 206 factors identified by stakeholders, 40 factors are case-specific and not contained in the general, review-based factor list. This underscores the value of participatory research, as general, top-down analyses might have overlooked these case-specific factors.
The methodology presented in this dissertation allows for the identification and analysis of case-specific intervention points for sustainability transitions at multiple societal levels. The methodology furthermore permits the analysis of interplay between individual, group, organizational and policy actions, which is a first step towards their coordination. The focus on sustainability innovations links the broad topic of sustainability transitions to a set of opportunities for practical interventions and overcoming their implementation barriers. The methodology presented allows for the analysis and design of these interlinkages between learning contexts. While the methodology cannot provide any ‘silver bullets’ for inducing sustainability transitions, it is flexible enough to identify an appropriate abstraction level for analyzing and designing transition governance processes. The methodology developed in this doctoral research also provides several contributions for the development of participatory modeling methods in transition research. Thus, the participatory method supports an integrated analysis of barriers and drivers of sustainability innovations, and allows application in practice and education.
The concepts and methods developed in this research project allow for reflection on transition governance processes from a systemic viewpoint. Experiences in the case studies underline the applicability of the concepts and methods developed for the analysis of case-specific transition governance processes. Despite substantial differences in the geographic location, culture and topics addressed, all case studies include promising sustainability innovations and the engagement of multiple actors in their implementation. The diversity and multitude of initiatives in the case study regions provides an optimistic outlook on future opportunities for large-scale sustainability transitions.
