O efeito paradoxal (EP) é caracterizado pelo crescimento celular em concentrações de equinocandinas acima da concentração inibitória mínima (CIM). Apesar da resistência a esta classe de drogas ser um fenômeno pouco comum em isolados de Candida spp., a ocorrência do EP é freqüentemente observada entre isolados desta levedura durante os testes de susceptibilidade às equinocandinas. O objetivo deste estudo foi analisar a freqüência do EP da caspofungina (CAS) entre isolados de Candida spp. e avaliar os aspectos morfológicos e os mecanismos moleculares relacionados a este fenômeno. Para tanto, foram utilizados 77 isolados de Candida spp., incluindo 21 isolados de C. albicans, 22 de C. tropicalis, 23 de C. parapsilosis e 11 de C. orthopsilosis. Todos isolados foram submetidos ao teste de susceptibilidade da CAS, segundo recomendações do Clinical Laboratory Standards Institute (CLSI, documento M27-A3), para determinação da CIM e avaliar a presença do EP. Entre os isolados que apresentaram EP da CAS, foram selecionados dois isolados de cada espécie para aplicação dos demais testes propostos neste estudo. Os ensaios de concentração fungicida mínima (CFM), curva de morte e ensaio de viabilidade celular foram utilizados para avaliar o comportamento das células fúngicas durante o EP. Analisamos ainda as alterações ultraestruturais e bioquímicas da parede celular durante o EP e a expressão de genes relacionados à síntese da parede celular durante este fenômeno. Entre os 77 isolados de Candida spp. analisados, 42 (54,5%) apresentaram o EP da CAS em concentrações que variaram de 4,0 a 32,0 µg/mL de CAS. Os ensaios de viabilidade celular mostraram que as células do EP apresentaram-se viáveis, mesmo quando expostas a altas concentrações de CAS (16 µg/mL). Os ensaios de curva de morte provaram ser mais discriminatórios que MFC para caracterizar o EP, principalmente para isolados de C. parapsilosis e C. orthopsilosis. As quatro espécies de Candida analisadas neste estudo apresentaram um padrão semelhante de alterações ultraestruturais e bioquímicas da parede celular durante o EP. De forma geral, durante o EP houve uma diminuição de 2,7 a 7,8 vezes no conteúdo de β-1,3-glucana e um aumento de 4,0 a 6,6 vezes no conteúdo de quitina da parede celular. Sobre a análise ultraestrutural por microscopia eletrônica, as células do EP apresentaram aumento do tamanho celular, formação de aglomerados celulares, ausência de filamentação e anormalidades no septo de brotamento. Além disso, a parede celular das células do EP apresentou um predomínio da camada mais eletrodensa, indicando uma diminuição da camada de β-1,3-glucana. Por fim, os resultados da análise da expressão gênica sugerem que o aumento da síntese de quitina em células do EP de C. albicans é regulado pelas vias PKC, HOG e Calcineurina-Ca2+. Nossos resultados apresentaram detalhes sobre as alterações morfológicas e ultraestruturais durante o EP, mostrando que as diferentes espécies analisadas apresentaram resposta celular semelhante durante o EP da CAS. Podemos concluir ainda que as células do EP possuem mecanismos adaptativos responsáveis pelo aumento do conteúdo de quitina na parede celular, em resposta à inibição de β- 1,3-glucana. Esses mecanismos permitem que as células do EP sobrevivam a altas concentrações de CAS, sugerindo que a estimulação da síntese de quitina pode representar um mecanismo de escape à atividade desta droga.
The paradoxical growth effect (PG) has been described for echinocandins and is characterized by cellular growth at drug concentrations above the minimal inhibitory concentrations (MIC). Despite resistance to this class of drugs is an unusual phenomenon among Candida isolates, occurrence of PG is frequently observed during susceptibility tests for echinocandins. The aim of this study was to analyze the frequency of PG for caspofungin (CAS) among Candida strains and also to evaluate the morphological aspect, as well as the molecular mechanisms related to this phenomenon. Seventy-seven strains of Candida spp., including 21 isolates of C. albicans, 22 isolates of C. tropicalis, 23 isolates of C. parapsilosis and 11 isolates of C. orthopsilosis were studied. All strains were subjected to susceptibility tests of CAS, as described in the Clinical Laboratory Standards Institute (CLSI, document M27-A3), to determine the MIC and evaluate the presence of PG. Afterwards, two isolates each of Candida albicans, C. tropicalis, C. orthopsilosis and C. parapsilosis, all of which displayed PG in response to caspofungin, were subjected to MIC, minimal fungicidal concentration (MFC) and time-kill curve assays to evaluate the PG. We also analyzed the ultrastructural and biochemical changes in the cell wall of PG cells and the gene expression related to the synthesis of cell wall during this phenomenon. Of the 77 Candida isolates initially screened, 42 isolates (54.5%) displayed the phenomenon of PG in concentrations ranging from 4.0 to 32.0 µg/mL CAS. C. parapsilosis demonstrated the higher frequency of PG, followed by C. albicans, C. tropicalis and C. orthopsilosis. Cell viability assays showed that the PG cells were viable, even when exposed to high CAS concentrations (16 µg/mL). The time-kill curve assays were more discriminatory than MFC in detecting the PG effect, especially for C. parapsilosis and C. orthopsilosis isolates. The four different Candida species studied demonstrated similar alterations in the cell wall components and ultrastructure associated with PG. In PG cells, β-1,3-glucan content decreased 2.7- to 7.8-fold, whereas chitin content increased 4.0- to 6.6-fold. Electron microscopy study of the PG cells revealed morphological alterations, clumping and enlarged cells, absence of filamentation, abnormal septa and accumulation of chitin in the cell wall. Also, PG cells basically exhibited a single dark high density layer in the cell wall, indicating loss of the β-1,3-glucan layer. Finally, the analysis of gene expression suggests that the increase of chitin synthesis in PG cells of C. albicans is regulated by PKC, HOG and Calcineurin-Ca2+ pathways. Our results showed details about the morphological and ultrastructural alterations during PG, demonstrating that different Candida spp. analyzed presented similar cellular response during PG of CAS. Our findings also suggest that the PG cells have a compensatory mechanism responsible for the increase of chitin content in the cell wall, in response to β-1,3-glucan inhibition. These mechanisms allow the survival of PG cells to high concentrations of CAS, suggesting that stimulation of chitin synthesis may represent a rescue mechanism against caspofungin activity.

Com o objetivo de avaliar a interferência da espessura da parede das mangueiras gotejadoras na vazão oferecida pelo emissor foi desenvolvido um experimento no Laboratório de Hidráulica do Departamento de Engenharia Rural da Universidade Estadual Paulista “Júlio de Mesquita Filho” Campus de Botucatu. O trabalho conduzido em uma bancada de ensaio, avaliou quatro mangueiras de fabricação nacional (Tiquira, Manári, Amandi e Jurussu) da empresa Petroisa. As avaliações foram realizadas segundo a norma internacional ISO 9261:2004. Os parâmetros avaliados foram: coeficiente de variação de fabricação, relação vazão x pressão, determinação da equação característica, espessura de parede e diâmetro interno das mangueiras, espaçamento entre emissores e resistência à pressão hidráulica em temperatura ambiente e à temperatura de 40 oC. Como resultado os coeficientes de fabricação das mangueiras atenderam às especificações da norma ISO 9261:2004, com exceção da Manári. S egundo o catálogo fornecido pelo fabricante, as equações características das mangueiras apresentariam os mesmos valores, uma vez que os emissores são do mesmo modelo. National companies generally work with a single model of dripper that is inserted in the dripperlines with different thicknesses to suit different types of consumers. The current research evaluates the effect of wall thickness on the hydraulic performance of dripperlines. The experiment was conducted in a bench trial in the Laboratory Testing Equipment for Irrigation of Faculty of Agronomic Sciencies UNESP. Four Brazilian manufactured dripperlines (Tiquira, Manari, Amandi and Jurussu) of Petroisa Company were evaluated. The evaluations were done according to the ISO 9261:2004. The parameters determined were coefficient of variation of manufacture, relation pressure x flow, the characteristic equation, wall thickness and inside diameter, the spacing between drippers and hydraulic pressure resistance at room temperature and at 40 oC. As a result, the coefficients of variation of manufactore the specifications of the ISO 9261:2004 standard, except Manari. According to the catalog provided by the manufacturer, the characteristics of the dripperlines equations showed the same values, since the emitters are 2 the same type. Full textThe aim of this work is to study the stiffness effect of wall columns on structural behavior of reinforced concrete buildings subjected to seismic action. The premises of the Brazilian standard ABNT NBR14521:2006 were considered, which presents criteria for earthquake resistant design of structures. The linear analysis employed the methods of the Brazilian standard - equivalent static load method, response spectrum analysis and time history method - were applied to buildings with two types of structural systems, namely concrete frame and dual system concrete frame and wall columns. The results were analyzed in edge frames structures in the longitudinal and transverse directions, comparing the displacement of each floor and shear, bending moment and axial forces on the bases of the columns. A comparison was made between the three methods applied and the structural systems analyzed.

Full textRadiotherapy is widely used for the treatment of pelvic tumors, including malignancies of the bladder, intestine, and rectum. Radiotherapy has harmful effects, especially on tissues with intense cellular turnover, with the intestinal mucosa being highly susceptible. Within this context, dietary amino acid supplementation has proved to be a promising therapy for the minimization of this damage. The objective of the present study was to determine the effect of dietary supplementation with the amino acids L-arginine and glycine on the structure of the colon wall of rats submitted to abdominal irradiation. Forty male Wistar rats were randomly assigned to four groups of ten animals each: I non-irradiated control with no amino acid supplementation, II irradiated control with no amino acid supplementation, III irradiated group supplemented with L-arginine, and IV irradiated group supplemented with glycine. The animals received dietary supplementation for 14 days, with irradiation being applied on the 8th day of the experiment. Stereologic analysis showed that irradiation induced a reduction of the total volume of the colon wall of group II and III animals compared to healthy controls, but not of group IV. The mucosal layer of the irradiated animals of all groups was reduced compared to healthy non-irradiated animals. The irradiated animals that did not receive amino acid supplementation presented reduction of the muscle layer of the mucosa compared to groups I and IV, and the group of rats supplemented with glycine presented a significant increase of the submucosal layer compared to groups I and II. Group III animals showed a reduction of the muscularis propria layer compared to groups I and IV. Supplementation with L-arginine was effective in maintaining the partial volume of the epithelium of the mucosal layer. The present results suggest that glycine supplementation had a superior effect on the structure of the colon wall of rats submitted to abdominal irradiation compared to L-arginine supplementation since it was able to maintain the thickness of the wall and the epithelial surface of the mucosa, whereas L-arginine maintained the partial volume of the epithelium and the epithelial surface, but not the total volume of the intestinal wall.

Full textRadiotherapy is often used to treat prostate tumors, but the normal bladder is usually adversely affected. Using an animal model of pelvic radiation, we investigated whether glutamine nutritional supplementation can prevent radiation-induced damage to the bladder, especially in its more superficial layers. Male rats aged 3 to 4 months were divided into groups of 8 animals each: controls, which consisted intact animals; radiated-only rats, which were sacrificed 7 (R7) or 15 (R15) days after a radiation session (10 Gy aimed at the pelvico-abdominal region); and radiated rats receiving L-glutamine supplementation (0.65 g/kg body weight/day), which were sacrificed 7 (RG7) or 15 (RG15) days after the radiation session. Morphological and morphometric analysis of the urothelium were made. Nuclear density, lamina propria cell density and mast cells numbers per area were counted. The results showed that, in R7, epithelial thickness, epithelial cell density, and cell density in the lamina propria were not significantly affected. However, density of blood vessels in R7 was reduced by 48% (p < 0.05) and this alteration was mostly prevented by glutamine (p < 0.02). In R15, density of blood vessels in the lamina propria was not significantly modified. However, epithelial thickness was reduced by 25% (p < 0.05) in R15, and this effect was prevented by glutamine (p < 0.01). In R15, epithelial cell density was increased by 35% (p < 0.02), but glutamine did not protect against this radiation-induced increase. Cell density in the lamina propria was likewise unaffected in R15. Density of mast cells in the lamina propria was markedly reduced in R7 and R15. The density was still reduced in RG7, but a higher density in RG15 suggested a glutamine-mediated recovery. Alpha-actin positive cells in the lamina propria formed a suburothelial layer and were identified as myofibroblasts. Thickness of this layer was increased in R7, but was similar to controls in RG7, while changes in R15 and RG15 were less evident. In conclusion, pelvic radiation leads to significant acute and post-acute alterations in the composition and structural features of the vesical lamina propria and epithelium. Most of these changes, however, can be prevented by glutamine nutritional supplementation. These results emphasize, therefore, the potential use of this aminoacid as a radioprotective drug.

Full textThis work assessed the occurrence and features of hemicellulose in grasses as related to lignocellulosic biomass recalcitrance to enzymatic hydrolysis. The grasses materials included six sugarcane hybrids with varying lignin and hemicellulose content, besides a variety of forage sorghum. The mature sugarcane internodes were dissected into three different regions: pith, pith-rind interface and rind. The sorghum was dissected into pith and rind. Pith samples presented high glucan content (40- 55%) and low lignin content (14-22%). The xylan content increased from pith region (14-20%) to rind (20-23%). Hemicellulose composition was also assessed. Xylose (9- 23%), glucose (1-12%) and arabinose (1-4%) were the most abundant monomers at the end of the analyzes. Pith samples had the most arabinosilated xylans and high content of non-cellulosic glucans. The (1-->3),(1-->4)-ß-D-glucan content was high in piths (1,4-15%) and decreased towards rind (0,1-1%). The sugarcane arabinoxylans were submitted to a study of their structure, which confirmed the xylan backbone was substituted. Lignin was assessed by pyrolysis coupled with mass spectrometer, which showed an HGS polymer in sugarcane and a S/G ratio without a progressive pattern between internode fractions. The sugarcane samples were also evaluated as related to hemicellulose distribution between different tissues and cells by immunofluorescence microscopy. A preliminary analysis showed that antibodies specific to xylan and mixed-linkage glucan bound to hemicellulose extracts. Plant sections were treated with the antibodies CRCC-M140 and LM11 (for less and more substituted xylans) and another one against mixed-linkage glucans. Less decorated xylan epitopes were mainly present in vascular bundles and were not detected in pith parenchyma. Arabinoxylan epitopes were found in all fractions and cell types. The mixed-linkage glucan immunofluorescence showed they were mainly deposited in parenchyma cells from piths; they were only detected in phloem and protoxylem cells in vascular bundles. The crystallinity index was estimated for all samples. High crystallinity indexes were found in rind samples (0,44-0,56). The non-pretreated materials were digested with commercial cellulolytic enzymes. Piths were easily hydrolyzed (32-85% of glucan conversion after 72h), while rind samples were more recalcitrant (2-9%). Interface samples showed intermediary conversions (7-46%). Xylan hydrolysis occurred in a less extent for all fractions, possibly due to a high substitution level on xylans. The glucan digestibility was proportionally related to the substitution level of xylan (R2=0,86) and the mixed-linkage glucan content (R2 = 0,92). Among the grasses samples, those with lower recalcitrance had features such as low lignin content, high mixed-linkage glucan content and high substitution level of xylans. These features could serve as indicators in the process of generation and selection of plants for lignocellulosic biomass production with high digestibility degree.

Full textYeasts discarded as a byproduct from industrial processes are gaining more attention as a nutritional supplement for food and feed due to its high content of protein and vitamins. They can also be used as raw material for extraction of cellular components, such as β-Glucan and mannoproteins (MP). The MP is present in the outermost layer of the cell wall, which plays a fundamental role in the activation of host immune system and acts also as an effective bioemulsifier. The aim of this study was to characterize the MP molecule extracted from Saccharomyces uvarum cell wall (discarded after beer production), verify its bioemulsifier action, its healing capacity and the immune modulation response in mice. The yeast was autolyzed in order to obtain the cell wall from which the MP was extracted using water at 95 ºC during 9 hours and then precipitated with absolute ethanol. The MP was chemically analyzed (protein, lipids, carbohydrates and ashes content) and characterized the type of sugars present, molecular weight of the protein and there amino acid composition, as well. Were determined the emulsifying activity and physical and chemical stability of the emulsion at different pH and NaCl concentrations. The healing activity of MP was tested observing the wounds reduction size on pigs testicles after castration. It was analyzed the production of antibodies in mice immunized with intraperitoneal MP (in PBS) injection in different treatments (positive control, 1000 μg of MP, 100 μg the MP, 100 μg of MP applied 6 hours before the challenge, and 100 μg of MP applied to 24 hours before the challenge), which were challenged with sheep red blood cells, and nitric oxide (NO) quantification production in vitro by peritoneal macrophages and in vivo by macrophages and plasma obtained from mice treated with MP. Blood samples were also removed for hematological analyzes. The MP had higher values of emulsifying activity, and is stable at different pHs and NaCl concentrations. Also acted as an effective healing, reducing the size of the wounds of animals, although did not differ from control. Antibody production was higher in animals treated with 100 μg of MP during or 24 hours before the challenge, which showed high levels of IgT after the first challenge and specific IgG antibodies at the end of the experiment. As expected, there was a greater production of IgG antibodies to all treatments after the second challenge with sheep red blood. Regarding the innate immune response, the MP was able to stimulate NO production in vitro, differing from the control only when it was added at a concentration of 100 μg/mL, although it inhibited the production when it was added together with LPS. In the in vivo experiment, treatment with MP raised the level of NO only when added together with LPS in the assay with peritoneal macrophages, reduce platelet and red blood cells and increase the number of reticulocytes and segmented neutrophils of the blood.

Full textVisando avaliar a qualidade e a constituição da parede celular em frutos de tomate da variedade comercial H-9665, realizou-se um ensaio num campo de cultivo em Manique (39°00'33,5"N; 8°51'05,8"W, Vila Franca de Xira), conduzindo o sistema de rega em duas modalidades distintas, superficial (I) e subterrânea, a 30 cm de profundidade (SI). Efetuaram-se análises aos frutos colhidos em quatro fases de maturação: MG (verde maduro), B (pintado), LR (vermelho claro) e H (colheita) e, em paralelo, registou-se o desenvolvimento fenológico e parâmetros relacionados com o teor de humidade do solo. No geral, a modalidade de rega parece não ter afetado significativamente as características qualitativas do fruto (cor, pH, acidez cítrica, textura, consistência, teor de sólidos solúveis) e os parâmetros relacionados com a constituição da parede celular (açúcares totais (AT) e ácidos urónicos (AU)), à exceção da quantidade de celulose que foi superior, na maioria das fases de maturação, na modalidade SI. Por outro lado, a modalidade de rega parece ter afetado a produtividade (SI superior à I) e o desenvolvimento vegetativo (I superior à SI).

Full textThe incisional hernia (IH) formation is one complication after abdominal surgery, affecting up to 20% of the reoperated patients. The use of prosthesis increased the repair of IH, however there is not yet an ideal treatment to repair or prevent the IH. The latex membrane from rubber tree has demonstrated to be cicatrizing and angiogenic, both in in vitro and in vivo studies. The aim of this work was to evaluate the effect of the latex membrane on the repair of the injuried abdominal wall of rats. Male Wistar rats with 340 ± 20 g were used and it was induced an abdominal wall defect with 1 x 2 cm and the alba line at the centre. The animals were divided into two experimental groups: Control (without repair) and Latex (with membrane repair) and were euthanized at 3, 5, 7 e 28 days after the defect induction. The abdominal wall fragments were removed, divided into two parts and used for histological and biochemical analysis. The histological analysis revealed that the latex membrane induced blood vessels proliferation, new muscle fiber formation and tissue regeneration. It was also observed a fibroblast density and collagen deposition increasing that is important to increase the resistance to tension forces and prevent the incision dehisce and IH formation. There was also a gradual increasing on the deposition and organization of collagen fibers during the experiment. The latex membrane induced the expression of cellular growth factors, FGF-2 and TGFb; decreased the expression of IGF and PDGF and did not alter the TGFa expression. Tissue regeneration requires the influence of growth factors in a sequence of cellular events that results on the cell regulation, proliferation, surviving and differentiation. The modulation of growth factor by the latex membrane may have contributed positively to the abdominal wall defect regeneration. It can be concluded that the latex membrane from rubber tree, when used to reconstruct the rat abdominal wall, allows tissue regeneration, fiber connective tissue formation and may be used effectively in hernioplasty.
