Abstract:
IntroductionThe health burden due to depression is ever increasing in the world. Prevention is a key to reducing this burden. Guided internet cognitive–behavioural therapies (iCBT) appear promising but there is room for improvement because we do not yet know which of various iCBT skills are more efficacious than others, and for whom. In addition, there has been no platform for iCBT that can accommodate ongoing evolution of internet technologies.Methods and analysisBased on our decade-long experiences in developing smartphone CBT apps and examining them in randomised controlled trials, we have developed the Resilience Training App Version 2. This app now covers five CBT skills: cognitive restructuring, behavioural activation, problem-solving, assertion training and behaviour therapy for insomnia. The current study is designed as a master protocol including four 2×2 factorial trials using this app (1) to elucidate specific efficacies of each CBT skill, (2) to identify participants’ characteristics that enable matching between skills and individuals, and (3) to allow future inclusion of new skills. We will recruit 3520 participants with subthreshold depression and ca 1700 participants without subthreshold depression, to examine the short-term efficacies of CBT skills to reduce depressive symptoms in the former and to explore the long-term efficacies in preventing depression in the total sample. The primary outcome for the short-term efficacies is the change in depressive symptoms as measured with the Patient Health Questionnaire-9 at week 6, and that for the long-term efficacies is the incidence of major depressive episodes as assessed by the computerised Composite International Diagnostic Interview by week 50.Ethics and disseminationThe trial has been approved by the Ethics Committee of Kyoto University Graduate School of Medicine (C1556).Trial registration numberUMIN000047124.

Abstract:
This study investigated Iranian EFL teachers’ self-efficacy beliefs (TSEBs) about instructional strategies, student engagement and classroom management strategies they adopt for teaching. It also examined the relationship between TSEBs concerning the three sub-efficacies and teachers’ use of (non) communicative instructional practices. The participants of the study were 48 EFL teachers in Iranian language institutes. A survey questionnaire, semi-structured interviews, and classroom observations were administered to collect the data needed. The results demonstrated that teachers regarded themselves as high-efficacious, and there was a moderate positive relationship between TSEBs in terms of the three sub-efficacies and communicative instructional practices. TSEBs towards instructional strategies had the highest contribution to explaining communicative practice, which was also confirmed by the interview results. The data from observations indicated that TSEBs were not realized regarding instructional strategies, but student engagement and classroom management strategies were reflected in teachers’ instructions.

Abstract:
Business process improvement (BPI) methodologies play an important role in increasing a business’s performance and its competitiveness. Since World War II, a number of these methodologies have been developed. Each of these held to a different philosophy as to what drives process improvement. Therefore, each focused on a seemingly unique aspect or parameter of business processes. These leverage points included, amongst others, process cycle time, quality, process efficiency, on-time delivery and error rates. The methodologies were delineated from one another not only by these focal points but also the terminology, structure and emphasis used by their creators. The result was a spectrum of seemingly unique approaches. This spectrum was enlarged by practitioners (consultants and businesses) who tweaked the base methodology so as to make their product offerings stand out. This palette of business improvement approaches was further augmented by certain techniques, tools and methods being presented as complete methodologies. The result was that business managers are confronted with a seemingly confusing array of ostensibly unique options. The author, being a business process improvement consultant, felt that these options were not as unique as they might appear. He believed that beneath these individualistic exteriors lays a common set of factors that enable the methodologies to bring about sustainable improvements. Therein lays the theoretical framework of this thesis. The author further felt that identifying these common key factors and their respective importance to business process improvement would contribute significantly to both knowledge and practice. Upon completing the research, it became evident that the contribution was not just limited to the enlightenment regarding composition, commonality and efficacy. It also involved ways of delineating targets such as methodologies from a chaotic population. Most importantly, a significant contribution to knowledge was made by showing how to synthesize information out of a set of seemingly unique data points. This uniqueness having been created by the different terminologies structures and emphasises found in the evidence. In the thesis, four unique challenges were encountered. The first was to identify the current, core business process improvement methodologies out of the total field of offerings. The subsequent challenge was to analyse these methodologies for possible common constructs and components. The third challenge involved evaluating the efficacy of these common key factors. The final challenge was to characterize these key factors in the context of a theoretical, model-based methodology. The intended result was to create a holistic perspective of process improvement practices with justification at the constituent level. A lengthy literature review was required in order to identify the current methodologies. Surprisingly, the sources normally rich in research evidence such as journals and articles did not give a comprehensive overview of business process methodologies. The candidates had to then be further researched via books and other lengthy publications. The end result was that six unique and sustaining families of methodologies were identified during the systematic literature review. Each of these had been developed by a different business improvement innovator using a different orientation associated with the methodology’s core philosophical position. It appears that, for marketing reasons, each had strived to develop unique selling points and intellectual property that set their methodology apart from the others. As a result, the language, style and emphasis applied in each methodology were seemingly distinctive. These conditions made further analysis for commonality of key factors amongst the six methodologies difficult and time-consuming. Further research involved analysing the six BPI methodologies and synthesizing qualitative evidence in order to identify the key factors common amongst those methodologies. Again, journals and articles played only a minor role in this activity. For the most part, these sources focused more on application than on composition. In addition, those dealing with application of the methodologies tended to avoid justifying applicability in terms of critical composition factors. Other sources such as books provided a framework for the composition analysis but with significant voids and confusing entries. Therefore, in addition to the series of literature reviews, interviewing intellectual property owners and BPI practitioners from both business and consulting firms was necessary. Once the interviewing process had closed the gap and provided clarification of entries, a research method had to be found which could extract the common factors and make them apparent. The appropriateness of various research, analysis and synthesis approaches were reviewed and evaluated. As a result of this ancillary literature review, a form of reciprocal translations was viewed as the best means of dealing with the research evidence as well as the issues of languages, styles, cultures and subjectivity. The final results of the synthesis, after a follow-up round of literature reviews and interviewing, showed that 12 common key factors existed amongst the six BPI methodologies. Research into the efficacy of these 12 common key factors required yet another series of literature reviews and additional interviewing. Due to the nature of the topic, interview respondents outside of the business process improvement community also had to be included. Those respondents included individuals from human resource and industrial psychology disciplines. The result of this descriptive research enabled characterization of the key factors’ efficacies independently as well as holistically in the context of a theoretical, model-based methodology. These characterizations indicate that the 12 key factors appear to form the backbone of BPI methodologies in general. The research process and its findings have contributed to both knowledge and practice. It has provided insight into using a holistic approach in delineating targeted elements of a mixed population. It also contributed to the understanding of how to apply the reciprocal translation technique to qualitative evidence outside the field of ethnography. Additionally, it enables improved business practices by demystifying the various methodologies and furthering the understanding of their value adding components.

Abstract:
G protein coupled receptor agonists can be described by two parameters; affinity (ability to bind a receptor) and efficacy (ability to activate a receptor once bound). While affinity is now an accessible parameter through binding studies, agonist efficacies have historically been difficult to determine. This is due to the significant system dependence of agonist responses and is particularly true of traditional maximal response and potency measurements. The Black and Leff operational model determines agonist affinity and efficacy estimations directly from functional data. Its measure of efficacy (τ) offers improvement over maximal response determinations but remains a system dependent parameter. More recently, the Slack and Hall operational model has been described. This model uses receptor constitutive activity to estimate the system contribution to agonist responses to derive system independent measures of intrinsic agonist efficacy. This thesis explores the use of operational models in the analysis of agonist functional responses at the dopamine D2 receptor. This receptor is an important therapeutic target in the treatment of schizophrenia and Parkinson’s disease, and has well documented receptor constitutive activity. Maximal responses and potencies in two representative functional assays, CRE-SPAP reporter gene and receptor internalisation, were determined for a panel of ligands at the D2L receptor. In the receptor internalisation assay, agonist maximal responses and potencies were decreased, consistent with an assay of reduced receptor reserve. Applying the Black and Leff operational model to the responses of four representative agonists, in the absence and presence of the irreversible antagonist phenoxybenzamine, yielded affinity and efficacy estimations consistent with the known pharmacology of the compounds. However, experiments did not provide evidence for the receptor constitutive activity necessary to apply the Slack and Hall operational model. As such, site-directed mutagenesis was performed to generate a constitutively active D2L receptor. T6.34R mutation increased agonist potencies and relative maximal responses consistent with a receptor more likely to adopt the active state; however, potencies of the ergo-derived compounds were selectively decreased. Mutagenesis of three key binding site serine residues examined the mode of agonist binding in both wild type (WT) and T6.34R backgrounds. Each serine mutation had differential effects on the responses of structurally distinct agonists, suggesting agonists engage specific serine residues; while S5.42 and S5.46 were required for catechol agonist responses, S5.46 was essential for the responses of the ergo-derived compounds, bromocriptine and dihydroergocristine. It was unknown whether these specific modes of binding would generate functionally similar active conformations. Generally, the differences between the effects on CRE-SPAP reporter gene and receptor internalisation assay responses were sufficiently explained by differences in receptor reserve. However, there were differential effects of serine mutations on WT and T6.34R D2L receptors, particularly the opposing effect of S5.42A on quinpirole potencies in the CRE-SPAP reporter gene assay. This suggests that WT and T6.34R D2L receptors adopt different conformations. Finally, we have described a method to determine agonist affinity and efficacy from [35S]-GTPγS binding assays using operational models, which does not rely on irreversible antagonist treatments. Buffer Na+ substitution revealed consistent constitutive activation of the D2S receptor, but not of the D2L or D2L T6.34R receptors. Agonist responses at the D2S receptor, in the presence and absence of buffer Na+, were analysed with the Black and Leff and Slack and Hall operational models respectively, generating agonist affinity and efficacy estimations. A novel operational model, written specifically for the analysis of [35S]-GTPγS binding assays, was also examined. This model provided similar affinity derivations to the Black and Leff operational model but provided a τ value that may have reduced system dependency. Although there may be compromises in the associated experimental conditions and which receptors can be investigated, the Slack and Hall operational model can be applied to constitutively active systems to provide system independent measures of agonist intrinsic efficacy.

Recent work in anthropology has speculated on how developments in molecular biology and medicine might bring about new bodies, selves, and forms of sociality. This thesis explores how herbal and pharmaceutical antidepressants differently affect experiences of one's neurochemistry. It does so in two ways. First, it outlines the historical 'social life' of pharmaceutical antidepressants, including their co-production with depression, and the neurochemical body in a particular style of reasoning in biological psychiatry. Second, it presents and analyzes claims made for the efficacy of antidepressants made in vernacular North American books, advertisements, and pamphlets. Although the claims for both herbal and pharmaceutical antidepressants allude to the same realms of value---those of science, nature/history, and personal experience---their different social lives enable different access to the neurochemical body.

Abstract:
This thesis is a study of the contemporary forms of trance-possession rituals performed in Thai Buddhism. It explores the way in which the trance-possession rituals are conceptualised by Thai Buddhist people as having therapeutic potentiality, through the examination of the ritual efficacy that is established through participants’ lived experience. My main research question focuses on how trance-possession rituals operate within a contemporary Thai cultural context and what are the contributory factors to participants’ expressing a sense of efficacy in the ritual. This thesis proposes that applied drama can be used as a ‘lens’ to examine the participants’ embodied experiences, particularly in relation to the ritual’s potential efficacy. In addition, the thesis also draws on discourses from anthropology, to enable a clearer understanding of the Thai socio-cultural aspects. I proceed to examine the efficacy of trance-possession ritual by focusing on the Parn Yak chanting ritual and rituals in sak yant, the spiritual tattoo tradition, as the two examples. Through the interdisciplinary study as mentioned above, these rituals are investigated and interpreted through several aspects. This study uses interviews with monks, participants and people involved with rituals as well as documentary and archival research. As part of my research, I also critically reflect upon my ethnographic experiences, between 2006-2012, of a variety of these rituals that are performed in temples around central Thailand. My attendance at the Parn Yak rituals in and around Bangkok involved both complete participation as well as observation. For the rituals of sak yant tattooing, I observed a tattoo master’s practices at Wat Bang Phra temple in Nakhon Pathom province. This thesis intends to offer an alternative approach to examine participants’ experiences of efficacy during and after the rituals. The research examines the therapeutic transformation of participants through the embodied process during rituals, and suggests that participants’ embodiment during lived experience in ritual together with their historical and sociocultural context influence the ways that they articulate their sense of efficacy in the ritual. The thesis offers insights and ideas for further exploration of Thai Buddhist rituals as culturally therapeutic performances.

I 200 casi clinici riportati dal progetto RIARTE sono, senza dubbio, una fotografia fedele, reale e pratica della realtà clinica in Italia per quanto attiene alle due categorie osservate: pazienti con rischio cardio e cerebrovascolare superiore al 20% secondo le tabelle del rischio SCORE e pazienti con ipertensione di difficile controllo. In entrambi i casi clinici l’adozione, nelle misure farmacologiche, della terapia a base di calcio-antagonisti diidropiridinici è apparsa efficace, sicura, di facile associazione con altre terapie e caratterizzata da un rapporto costo-efficacia particolarmente favorevole.

Abstract Quaternary ammonium biocides are commonly used in cooling water systems to control organisms associated with microbiologically induced corrosion and microbial fouling. These compounds while being highly efficacious and low cost have, in general, high foaming tendencies which can limit their range of application. A new quaternary ammonium compound, decylisononyldimethylammonium chloride (DIDMAC), was developed to prevent foam problems while maintaining the advantages of standard quat treatments. Laboratory evaluations showed DIDMAC to produce significantly less foam than a standard alkyldimethylbenzylammonium chloride (ADBAC), approaching that of a polymeric quat. Bactericidal and algicidal evaluations showed DIDMAC to provide efficacy greater than the polymeric quat and similar to the ADBAC. Field evaluations confirmed the laboratory results showing DIDMAC to foam significantly less than the ADBAC and similar to the polymeric quat. In the field, DIDMAC bactericidal and algicidal efficacies were evaluated in combination with other nonoxidizing biocides. Optimized programs, consisting of weekly additions of 10-30 mg/L DIDMAC with semiweekly applications of 1.5 mg/L isothiazolone, exceeded the algicidal and matched the bactericidal performance of control thione/isothiazolone programs. The DIDMAC-containing programs, in addition to providing enhanced bactericidal and algicidal efficacy, reduced treatment costs by 27 to 43% compared to the control.

Calcium ion enhances the activity of the inorganic inhibitor and decreases the efficiency of the organic inhibitor. Magnesium has negligible influence on the efficacy of either inhibitor. Zinc, although not commonly found in oilfield brines, has a significant inhibitory influence on its own, but this influence is largely canceled by the presence of the organic or, especially, the inorganic halite inhibitor.

Abstract The corrosion inhibition of substituted pyridine derivatives and quaternized pyridines in the presence and absence of acetylenic alcohol synergists is reported. Electrochemical Frequency Modulation (EFM) was used as a method of monitoring corrosion rates of steel in acidic environments. Early stage pitting was evaluated via scanning electron microscope (SEM) for comparison of measured corrosion rates to observed pit formation. EFM was found to be a reliable and accurate method of monitoring general corrosion rates over time, and causality factor data was found to depend heavily on the type of corrosion occurring at the metal surface. In addition, the pyridine derivatives were found to have complex substituent effects on corrosion inhibition, with unsaturated substituents most significantly enhancing the performance relative to pyridine. Nearly all of the benzyl quaternized pyridine derivatives performed similarly to non-quaternized derivatives in the acid media, suggesting similar mechanisms of action, except for the N-benzyl quinolinium quaternary amine, which displayed significantly enhanced behavior relative to the amine. The alkynol synergists gave similar efficacies under the test conditions regardless of chain length or conformation. Substituent effects on the pyridine ring showed varying interactions with alkynols in pyridine-alkynol blends, with both synergistic and antagonistic effects being observed.

Abstract Eleven fluid samples, taken in two groups from pipeline pigging operations in the Texas Gulf Coast region were used in this study. All of the samples contained high amounts of Iron Sulfide (FeS), Sulfate Reducing Bacteria (SRB), and Acid Producing Bactera (APB) and were producing varying amounts of hydrogen sulfide (H2S). These pipelines had historically been treated with increasing amounts of glutaraldehyde and glut/quat blends, to no avail. Increasing problems associated with microbiological activity (biofilm, corrosion, FeS, H2S) required a revised treatment regime. The first part of this study was undertaken in order to determine a successful biocidal treatment program that would address both the planktonic and, if necessary, the sessile bacterial populations in the pipelines. Due to the perceived induced resistance to glutaraldehyde and the dirty water conditions in the pipelines, alternative dosing regimens and additional biocides were considered including tetrakis (hydroxymethyl) phosphonium sulfate (THPS), tributyl tetradecyl phosphonium chloride (TTPC), aldehydes, and cocodiamines. The second part of the study sought to determine how to link these findings to the general and localized corrosion caused by biological activity. A corrosion inhibitor package was developed that was compatible with the customer’s formulations and the recommended biocidal treatment program. Finally, the third part of this study combined these treatments and evaluated their biocidal and corrosion inhibition efficacies on carbon steel coupons in the presence of fluids and solids. A biocidal treatment and corrosion inhibition program are inherently intertwined and a successful treatment program must combine elements of both fields of study.

Abstract Volatile corrosion inhibitors (VCIs) compose a class of chemical compounds that have a suitable vapor pressure with which to diffuse throughout an enclosed volume, adsorb onto any exposed metallic surface within the volume, and inhibit corrosion in the adsorbed state. It has been demonstrated previously that the adsorption of volatile corrosion inhibitors (VCIs) depends upon environmental conditions, the variation of which in practical situations may dramatically affect the kinetics of VCI adsorption. Adsorption kinetics influence the overall performance of VCI as they are required to adsorb before corrosion inhibition may occur, although the extent of inhibition cannot necessarily be directly inferred from adsorption monitoring. This study attempts to evaluate VCI performance by utilizing surface enhanced Raman Spectroscopy to monitor the adsorption kinetics of VCI on exposed steel and copper surfaces while simultaneously performing real-time corrosion rate measurements with a multi-electrode array sensor in order to correlate VCI adsorption with corrosion inhibition. The relative efficacies of two VCIs to inhibit corrosion of carbon steel and copper were measured as the time dependent corrosion rate ratio of surfaces with VCI exposure to surfaces without VCI exposure in both humid and ambient environments. The results indicate that corrosion rate increases with initial VCI exposure compared to the same environment without VCI. A reduction in corrosion rate coincident with VCI adsorption was witnessed, but overall VCI containing environments were still found to yield elevated corrosion rates over non-VCI environments. Presumably, a critical concentration of adsorbed VCI is needed before inhibition starts to occur. The duration of this “incubation” period (the time before a critical inhibiting concentration is reached) was found to depend upon the specific VCI, but was on the order of a few days at room temperature. The results may have implications on the proper evaluation of VCI compounds and future development of VCI chemistry.

Abstract Microbiological monitoring of samples collected from oilfield operations is crucial for understanding microbial impacts on metal infrastructure. As many oilfields and related infrastructure are in remote locations, the time between sample collection, shipping to laboratory and sample processing for microbiological analysis can vary from days to weeks. Hence, robust sample handling and preservation methodology to arrest microbiological activity at the time of sample collection is needed. To identify the best preservation methodologies that can be adopted for solids, oily sludge samples were collected twice from the nose of the same pig trap during a routine pipeline pigging operation of a crude oil transmission pipeline over a span of two years. Samples were chemically preserved at the time of sample collection (with different alcohols or commercially available nucleic acid preservation reagents) or remained unpreserved, and all samples were stored either at 4°C or at room temperature following collection. Samples were assessed using 16S rRNA gene sequencing to determine microbial community composition at day 0 (time of sample collection) and at different intervals for up to 50 days of storage, and comparisons in microbial community composition were made to the day 0 results. A substantial shift in microbial community composition was observed in samples that were not chemically preserved or not kept cold at 4°C in as little as 7 days following sludge collection. In addition, greater microbial diversity and community shift was observed in samples kept at room temperature compared to cold storage. Chemical preservation reagents showed varying efficacies at preserving microbial community composition with increased lengths of storage. These results emphasize the need for careful sample preservation and handling of sludge samples for accurate monitoring of microbial communities, an analysis that can play a pivotal role in the early identification of potentially corrosive microorganisms during the routine inspection of pipelines.

In this report the potential effects of pulsed ocean alkalinity enhancement (OAE) on plankton growth dynamics, efficacy and carbon dioxide removal (CDR) are simulated at an oligotrophic site nearby the Canary Islands (Spain). Parameterizations of how phytoplankton and bacterial growth respond to variations in carbonate chemistry are introduced and applied. The model resolves vertical profiles within a 420 km2 area of OAE, and within a 2.05×105 km2 surrounding region. Both areas are connected and lateral dispersion causes a horizontal exchange of chemical and biological tracers. The analysis comprises 32 pulsed OAE scenarios of varying intensities and frequencies of alkalinity addition. The intensities of OAE are derived as fractions of total CaO available for OAE in Europe, from 0.05 % (low), 0.1 % (moderate), 0.2 % (high) and 0.4 % (extreme). These four intensities are co-varied with four frequencies of OAE: monthly, seasonal, annual-winter, and annual-summer deployments. The study shows how much the frequency of the OAE influences the efficacies determined locally and remotely. Many recent ocean modelling studies have demonstrated the added value of enhanced horizontal resolution, although it comes at a high computational cost. However, few modeling studies of ocean-based CDR have been done at high resolution. Here we assess the effects of model resolution on two simulated ocean-based CDR methods, unequilibrated ocean alkalinity enhancement (OAE) and the direct marine capture (DMC) of CO2 from seawater (with assumed permanent storage), in experiments with the FOCI Earth system model. To do this we utilized two FOCI configurations, one with a 1/2° ocean resolution and the other with a 1/10° ocean nest in the N. Atlantic. Both configurations were run in a series of “paired” experiments with identical climate forcing and CDR deployments. We show that model resolution does appear to matter when simulating OAE and DMC. For OAE, parameterization of physical processes in the coarse resolution version of the model appears to overestimate how long alkalized waters stay in contact with the atmosphere and where they are transported. This results in large differences in OAE efficacy with almost twice as much carbon sequestered when the model resolution is coarse. For the DMC simulations, at one site there were clear differences in the compensating CO2 flux induced by DIC removal, which was again higher with a coarse resolution, while at the other site variability was high and differences were difficult to determine. At both DMC sites there were clear differences in circulation with the two model resolutions, and thus on downstream biogeochemistry. We suggest that well resolving ocean physics may be necessary to best calculate unequilibrated OAE and DMC efficacies and side effects. These results should be confirmed using other models and with different resolutions.

L'utilisation de données probantes est cruciale pour la prise de décisions efficaces en matière de politiques publiques. Cela implique une meilleure organisation et coordination des acteurs et des données. Au Sénégal, des progrès significatifs ont été réalisés en matière de gestion des données probantes, grâce à divers acteurs tels que l’Agence Nationale de la Statistique et de la Démographie (ANSD) et la Direction Générale de la Planification et des Politiques Économiques (DGPPE). Cependant, malgré ces avancées, des défis persistent, notamment en matière de coordination et d’utilisation des données dans le processus décisionnel. En dépit de l’abondance de données disponibles, leur incohérence et le manque de synergie entre les acteurs créent des inefficacités. Les systèmes de suivi et d’évaluation existants, se concentrent davantage sur le suivi, que sur l'évaluation et l'apprentissage. Il est donc nécessaire de clarifier les cadres institutionnels, et d'améliorer la coordination entre les acteurs, pour maximiser l'impact des données probantes. Face à ces défis, plusieurs initiatives ont été mises en place pour illustrer des pratiques efficaces de gestion et d'utilisation des données probantes. Les concertations entre l’ANSD et les utilisateurs de données, le guide méthodologique de la DGPPE pour les Lettres de Politiques Sectorielles de Développement, et le projet Feed the Future en sont des exemples notables. Ces initiatives montrent l'importance de la participation des parties prenantes, de la méthodologie rigoureuse, et de l'amélioration des capacités des acteurs. Pour aller plus loin, le Sénégal doit renforcer l'efficacité de son écosystème de données probantes. Cela passe par l'amélioration de la coordination, la standardisation des processus de collecte et de partage des données, et le renforcement des capacités des utilisateurs. Il est crucial de promouvoir l'utilisation des données probantes et de clarifier les cadres institutionnels pour une meilleure gouvernance des politiques publiques. En somme, des efforts soutenus sont nécessaires pour optimiser l'impact des données probantes sur les politiques publiques, en assurant une coordination harmonieuse et une utilisation efficace des ressources disponibles.
