Abstract:
The article presents the results of an experimental study on rat poisoning with 1,2-dichloroethane (250 µl / kg, intragastric administration). The aim of this work was to find the relationship between biological markers and morphological changes in acute damage to rat kidneys with dichloroethane. The study of urine samples of experimental animals by gas-liquid chromatography (GLC) revealed 1,2-dichloroethane in the concentration range of 0,05 – 0,90 µg / ml (95% C.I. = 0,09 – 0,90 µg / ml). Daily excretion with urine was 0,60 – 4,50 µg / 16h (95% C.I. = 1,14 – 3,93 µg / ml). Levels of nephron-specific biomarkers in urine were 194,62 ± 9,02 IU / L (NAGase, p = 0,0022) and 2,93 ± 0,38 ng / ml (KIM-1, p = 0,0022), which exceeded those in the control group by 5 and 23 times, respectively. Edema, disorders of intrarenal hemodynamics, leukocyte migration in interstitium, uneven damage to various areas of the kidneys due to adipose dystrophy and necrobiotic changes in the nephrothelium of the proximal tubules mainly in the cortical layer of the kidneys have developed in the kidneys. Positive correlations between the level of 1,2-dichloroethane in the urine and the level of KIM-1 (Spearman r = 0,7427, 95% C.I. 0,2764 – 0,9260, p = 0,0083), -N-acetylglucosaminidase (Spearman r = 0,8248, 95% C.I. 0,4613 – 0,9512, p = 0,0019), the scores on the EGTI scale (Spearman r = 0,8064, 95% C.I. 0,7126 – 0,8719, p < 0.0001); the level of KIM-1 and the scores on the EGTI scale (Spearman r = 0,7427, 95% C.I. 0,2764 – 0,9260, p = 0.0083), -N-acetylglucosaminidase and the scores on the EGTI scale (Spearman R = 0,4684, 95% C.I. 0,2764 – 0,6244, p < 0,0001), the level of the low and medium molecular weight substances in the blood and the scores on the EGTI scale (Spearman R = 0,6909, 95% C.I. 0,5549 – 0,7909, p < 0,0001) were found.

In this paper, the concept of kinematic fractionation is introduced for epicyclic gear trains (EGTs) which contains structural fractionation as a degenerate case. With the concept of kinematic fractionation, kinematically independent group(s) embedded in an EGT can be identified. A composition list, which depicts the links and the link connection in the associated group, is used to determine the type of fractionation. It is found that most previously enumerated structurally non-fractionated EGTs in the literature are kinematically fractionated. It is shown that a structurally non-fractionated EGT may be kinematically fractionated while a structurally fractionated EGT must be kinematically fractionated. The concept of kinematic fractionation can lead to efficient topological analysis of EGTs with physical comprehension. [S1050-0472(00)01604-4]

Exhaust Gas Temperatue is an parameter where the hot gases’s temperature leave the gas turbine. EGTM is one of any factor to determine engine performance. A good perfomance of an engine when it has a big margin (EGTM), during operation of an engine the EGTM could decrease untill 0 (zero). So many factors could affect EGTM deteroration there are: distress hardware such as airfoil erosion, leak of an airseals, and increase of clearance between tip balde and shroud. Increase of clearance happens in high pressure compressor rotor clearance. In CFM56-7 have 9 stage(s) of high pressure compressor and each stage give the EGT Loses. The calculation of EGT Effect/Losses is actual celarance – minimum clearance x 1000 x EGT Effect °C, where actual clearance define by the substraction of outside diameter’s rotor with inside diameter’s shroud, minimum clearance define in the manual, 1000 is adjustment from mils/microinch to inch, and EGT Effect is temperature that define in the manual. The analysist had done with 6 (six) engine serial number and proceed by corelation that shown linkage between clearance and EGT Effect, the corelation is strong shown the result of corelation (r) is 0.994275999 or nearest 1.

Abstract:
ABSTRACT Trichoderma reesei strains were constructed for production of elevated amounts of endoglucanase II (EGII) with or without cellobiohydrolase I (CBHI). The endoglucanase activity produced by the EGII transformants correlated with the copy number of the egl2 expression cassette. One copy of the egl2 expression cassette in which the egl2 was under the cbh1 promoter increased production of endoglucanase activity 2.3-fold, and two copies increased production about 3-fold above that of the parent strain. When the enzyme with elevated EGII content was used, an improved stonewashing effect on denim fabric was achieved. A T. reesei strain producing high amounts of EGI and -II activities without CBHI and -II was constructed by replacing the cbh2 locus with the coding region of the egl2 gene in the EGI-overproducing CBHI-negative strain. Production of endoglucanase activity by the EG-transformant strain was increased fourfold above that of the host strain. The filter paper-degrading activity of the endoglucanase-overproducing strain was lowered to below detection, presumably because of the lack of cellobiohydrolases.

AbstractPurpose:The purpose of this study was to determine the complication rates associated with the use of the endotracheal tube (ET) a the use of the esophageal obturator airway/esophageal gastric tube airway (EOA/EGT during the treatment of patients with prehospital cardiac arrest.Methods:A descriptive, quasi-experimental study of 509 consecutive adults, cardiac arrest patients was conducted. Patients were examined prospectively for airway intervention type and complications. Some patients were examined at their final destinations (field, morgue, funeral home), while other patients were examined by EMS providers in the field when airway adjuncts were switched. Also, airways were evaluated for complications by emergency physicians at destination emergency departments.Results:The airway in use at the time of examination was the esophageal obturator airway (EOA) or esophageal gas lube airway (EGTA) in 208 patients (40.1%); the ET (endotracheal tube) in 232 patients (45.6%); and an oral or nasopha ryngeal airway in 47 patients (9.2%). Twenty-two patients (4.3%) had both an EOA/EGTA and an ET tube in place at the time of the examination. The survival rates were similar between the EOA/EGTA and the ET groups (28% and 32%, respectively). The complication rates overall also were similar, but the serious or potentially lethal complication rate was 3.3 times more common with the use of the EOA/EGTA than with the ET tube (8.7% versus 2.6%, respectively).Conclusions:The complication rate for the EOA/EGTA is unacceptably high, and careful thought must be given to its continued use. Serious questions also arise concerning the complication rates associated with the use of the ET: is the complication rate of 2.5% acceptable or should other airway alternative be considered for use in prehospital care?

For the widespread use of in vitro models of barrier tissues in practice, it is necessary to develop approaches to the automation of cultivation and monitoring of the state of cells. Impedance spectroscopy is a promising and informative method for assessing the state of cell barriers. In this work, the dynamics of electrical parameters in an in vitro model of the intestinal barrier with a high temporal resolution has been studied using impedance spectroscopy. For the first time, the detailed kinetics of changes in TEER, electrical capacitance and additional resistance of a monolayer of Caco-2 cells at various EGTA concentrations was recorded. It was demonstrated that spontaneous restoration of the integrity of the cell barrier is possible in the presence of 2 mM EGTA in the medium, which begins approximately 6 h after the compound addition. A strong increase in TEER was also found immediately after the addition of EGTA, which probably depended on the EGTA concentration and can be explained by a decrease in intracellular conductance due to the closure of transmembrane channels. impedance spectroscopy, intestine, TEER, electrical capacitance, EGTA The study was funded by the Russian Science Foundation (project 16-19-10597).

While there is compelling evidence for the impact of endosymbiotic gene transfer (EGT; transfer from either mitochondrion or chloroplast to the nucleus) on genome evolution in eukaryotes, the role of interdomain transfer from bacteria and/or archaea (i.e. prokaryotes) is less clear. Lateral gene transfers (LGTs) have been argued to be potential sources of phylogenetic information, particularly for reconstructing deep nodes that are difficult to recover with traditional phylogenetic methods. We sought to identify interdomain LGTs by using a phylogenomic pipeline that generated 13 465 single gene trees and included up to 487 eukaryotes, 303 bacteria and 118 archaea. Our goals include searching for LGTs that unite major eukaryotic clades, and describing the relative contributions of LGT and EGT across the eukaryotic tree of life. Given the difficulties in interpreting single gene trees that aim to capture the approximately 1.8 billion years of eukaryotic evolution, we focus on presence–absence data to identify interdomain transfer events. Specifically, we identify 1138 genes found only in prokaryotes and representatives of three or fewer major clades of eukaryotes (e.g. Amoebozoa, Archaeplastida, Excavata, Opisthokonta, SAR and orphan lineages). The majority of these genes have phylogenetic patterns that are consistent with recent interdomain LGTs and, with the notable exception of EGTs involving photosynthetic eukaryotes, we detect few ancient interdomain LGTs. These analyses suggest that LGTs have probably occurred throughout the history of eukaryotes, but that ancient events are not maintained unless they are associated with endosymbiotic gene transfer among photosynthetic lineages.

Abstract The purposes of this study was to investigate the efficacy of interpersonal relationships, cognition, instrumental activities of daily living, depression for elderly with cognitive impairment through intervention using the intergenerational somatosensory video game and company. The experimental design was used for this study. Eighty-nine elders with mild cognitive impairement and 180 adolescents were from nine junior and senior high school were recruited in this study. Eight day care centers were randomly assigned to the experimental group I (EGI) (5-week intergenerational somatosensory video game), the experimental group II (EGII) (8-week intergenerational somatosensory video game), the experimental group III (EGIII) (5-week intergenerational company), the experimental group IV (EGIV) (8-week intergenerational company) and control group for eight weeks of routine activities. Subjects were interviewed using structured instruments, including the Instrumental Activities of Daily Living (IADL), Token test, and Geriatric Depression Scale. Obtained data was analyzed using the Generalized Estimate Equation (GEE). The results revealed that there was no significant difference of eldsers’ characteristics among five groups. After intervention, instrumental activities of daily living scores in 8-week intergenerational somatosensory video game (EGII) and 8-week intergenerational company groups (EGIV) were significantly better than the control group. Token test score in 8-week intergenerational company (EGIV) were significant higher than that in the control group; while depression in 5-week intergenerational company (EGIII) was significantly lower thatn that in the control group. Based on the research findings, to arrange intergenerational action video game and intergenerational company to enhance elders’ attention, instrumental activities of daily living and decrease depression.

For many years text-immanent approaches to literature have dominated the scene of Afrikaans literary criticism. This article adds a voice to the ‘spontaneous' discourse in which ethical norms (especially socio-political guidelines), too, come into play when a literary text is studied. Since the context in which a text is written and read is of great importance in such an approach, speech act theory is used in order to determine the intentions (illocutions) of the writer in the texts, as well as the reactions (perlocutions) of readers to the text. The purpose of this is mainly to establish whether critique of ideology manifests itself in speech acts directed towards freedom and dignity for all people. On the other hand there may also be signs of unconscious ideological illocutions in the contradictions which occur within or between the different levels of communication (macro, meso and micro). These contradictions are related to socio-political contradictions which are repressed within the South African community. In order to illustrate this kind of approach, three novels of Etienne van Heerden are discussed, viz. Om te awol (1984), Toorberg (1986) and Casspirs en Campari's (1991).

The purpose of this study was to determine the incidence of anabolic steroid use among competitive male and female bodybuilders in Kansas and Missouri. A profile was established for users and non-users of anabolic steroids. The results of this study indicated that more than half of the male bodybuilders (54%) were using steroids on a regular basis compared to 10 percent of the female competitors. The types of steroid used were investigated and revealed that on average, four different types of anabolic steroid were used during the year, with individual use ranging from one to fifteen different types; including Dianabol, Deca Durabolin, Anavar, Testosterone, Androl 50, Winstrol, Primobolan, Equipoise, Finaject, Parabolin, HCG, Primacetate, Enanthate, Halotestin, and Maxibolin, in order of the most to least frequently used. The female bodybuilders reported that they had used an average of two different steroids including Deca Durabolin, Anavar, Testosterone, Dianabol, Equipoise, and Winstrol. The principal reason bodybuilders used steroids was related to their perception that these drugs were an important factor in winning competitions. Another important motivating factor for use was consistent with reports that significant gains in strength could be achieved by including anabolic steroids as part of the training regimen in spite of the reported adverse side-effects.

The suitability of the Ca2+ chelator ethylene glycol-bis(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA) to induce disruption of mammary tight junctions (TJ) and its effect on milk secretion were investigated in six goats. EGTA was administered via the teat of one gland as an isosmotic (300 mosmol/l) K-EGTA solution (68 mM EGTA), whereas the control gland received an isosmotic sucrose solution. Lactose, Na, K, and Cl in milk, blood lactose, and the presence of Evans blue (EB) in mammary lymph were used as indicators of TJ disruption. EGTA caused transient (approximately 60 h) changes (P < 0.05) in the concentration of lactose, K, Na, and Cl in milk, consistent with loss of TJ integrity. This was confirmed by a rapid (< 1 h) increase (P < 0.05) in blood lactose levels. Moreover, EB appeared in lymph < 1 h after EGTA+EB treatment. Milk secretion declined unilaterally by 15% (P < 0.05) after EGTA and did not return to baseline until approximately 60 h after EGTA. EGTA caused a unilateral, temporary (first 7 h) increase in mammary blood flow. This study shows that a rapid temporary disruption of mammary TJ can be successfully induced in vivo and that such disruption compromises milk secretion.

A patent survey is conducted to investigate the application status of epicyclic gear trains (EGTs) used in automatic transmissions (ATs). In this regard, 673 AT patents are investigated firstly, and 274 different EGTs are obtained after the patent processing. EGTs are presented by corresponding double bicolor graphs (DBGs) and they are sorted into 13 groups in accordance with the degree of freedom (DOF) of DBGs and the degree of hollow vertices. Then, structural characteristics of obtained EGTs, which are used to detect whether the EGTs obtained in the future can be used in ATs, are analyzed and summarized. Finally, a new approach is proposed to obtain new configurations of the EGTs with one main shaft based on the transformation relation between DBG and the basic graph. Several examples are presented to illustrate the validity of the proposed approach.

This in vivo study evaluated, through the physicochemical assay method for quantification of enhanced vascular permeability, the irritating potential of EDTA, EGTA, citric acid and saline. Thirty-two male Wister rats were anesthetized and four experimental sites were demarcated on their backs. Injections of 2% Evans blue (20 mg/kg) were administered intravenously into the lateral caudal vein. The test solutions were immediately injected intradermally (0.01 mL) into the experimental sites. The animals were killed 30 min, 1, 3 and 6 h after injection of the solutions and each piece of skin was submerged in formamide and incubated at 45ºC for 72 h. After filtration, the optical density was measured in a spectrophotometer and the total amount of dye extracted from the samples was calculated by means of a standard calibration curve. Data were analyzed statistically by two-way ANOVA and Tukey's HSD test. Compared to control, EDTA had the greatest volume of dye followed by EGTA and citric acid, for all time periods. There were statistically significant differences between all solutions (p<0.01). Considering the periods assessed, a significant difference was observed between the 3- and 6-h groups (p<0.05), but not between the 30-min and 1-h groups. Among the organic acids evaluated in this study, citric acid yielded the lowest amount of extracted dye. This indicates that the citric acid was the least irritating solution.

A régészeti megfigyelések alapján a vár legkorábbi előzményeként egy 11. század első feléből származó királyi udvarház épületeit azonosítottuk. Véleményünk szerint ez fogadta be az 1068 után először ideiglenesen, majd 1091 után véglegesen az egyházmegye központját, amely korábban Bihar várában lehetett. Rekonstruáltuk a román kori székesegyház építéstörténetét, és nagyrészt feltártuk a székesegyház északi oldalán a 12. század folyamán több lépésben kiépült káptalani kerengőt és püs pöki palotát, valamint több ponton azonosítottuk a 13. század első felében kiépülő, két részből álló első püspökvár falait (1. kép).Between 2016 and 2019, we conducted a series of excavations in Eger Castle, in the course of which we gained a number of new insights regarding the 11th–13th century history of the castle and the episcopal seat. On the testimony of the written sources and the archaeological record, the castle’s earliest antecedent was a royal manor house from the first half of the 11th century. In our view, this building functioned as the centre of the diocese – which had earlier probably been located in Bihar Castle – first temporarily after 1068, then, after 1091, permanently. We were able to reconstruct the architectural history of the Romanesque cathedral and we excavated the cathedral’s cloister and the episcopal palace on the cathedral’s northern side, built in a number of successive phases. We also succeeded in identifying the walls of the first episcopal castle built in the first half of the 13th century (Fig. 1).

The production and electrophoretic expression of endoglucanase(s) were compared in the wild-type and three developmental mutants of Aspergillus nidulans. In the wild type, the production of endoglucanase and its distribution in extracellular and intracellular fractions varied with the age of the culture and the yield was better in stable cultures (production of conidia and cleistothecia) as compared with shake cultures (vegetative hyphae only). Two developmental mutants, aco-T69 and aco-40, which lack the development of conidia and cleistothecia, produced low levels of endoglucanase enzymes as compared with the wild type grown under similar conditions. On the other hand, in aco-90, a mutant capable of producing cleistothecia but no conidia, endoglucanase production was better. The results indicate a correlation between cleistothecial development and endoglucanase level. The electrophoretic studies revealed the presence of three forms of endoglucanase, i.e., EGI, EGII, and EGIII. The first two were detectable in the wild type as well as in mutant strains when grown under various experimental conditions and at all the stages of development. However, the third form could be observed only during cleistothecial development, indicating that this isozyme is developmentally regulated.Key words: endoglucanases, development, Aspergillus nidulans.

The study of intracellular Ca2+ regulation usually requires using calcium chelators to adjust [Ca2+]. We examined the effects of these chelators on calcium accumulation in microsomes and saponin-permeabilized synaptosomes to assess their influence on apparent transport properties. At a fixed free Ca2+ of 0.6 microM, increasing ethylene glycol-bis(beta-aminoethyl ether)-N,N,N', N'-tetraacetic acid (EGTA) and total Ca2+ enhanced ATP-dependent 45Ca sequestration in synaptosomes and microsomes. The EGTA-Ca complex did not change the maximal initial calcium uptake rate or maximal steady-state accumulation. Rather, EGTA/Ca increased the apparent affinity of the microsomal transporter for Ca2+. The presence of the organic anion transport inhibitor probenicid (2.5 mM) had no effect on 45Ca accumulation in the presence of EGTA. Replacing part of the Ca2+ with Ni2+ but maintaining [Ca2+] approximately constant reduced 45Ca uptake, suggesting that the Ni-EGTA complex did not stimulate 45Ca transport. Our results imply that EGTA is not actively transported across the endoplasmic reticulum membrane, nor does the divalent ion-bound form of EGTA change the properties of the transporter. EGTA, and other mobile calcium chelators with similar structures, e.g., 1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid, indo 1, and fluo 3, may increase calcium uptake by delivering more Ca2+ to its transport site.

Trichoderma reesei is known to be one of the organisms capable for producing various types of cellulase in high concentrations. Among these cellulases, the highest catalytic efficiency of endoglucanases II (EGII, EC 3.2.1.4) are considered important for industrial application. The characterization of the EGII is necessary since it is widely used in high-temperature reactions in the industries. In this study, the recombinant EGII protein was expressed in Pichia pastoris and it has a molecular mass of approximately 52 kDa. Recombinant EGII was purified using Ni-NTA affinity chromatography and characterized by SDS-PAGE and western blot analyses. The enzyme activity of recombinant EGII was measured using the Nelson Somogyi method to determine its optimum pH and temperature. The result showed that the maximum EGII expression was achieved after 72 h of culture incubation. The crude enzyme has optimum activity at pH 5.0, resulting in 16.3 U/mL and 14.6 U/mL activity at 40 °C and 50 °C, respectively. While the purified enzyme gave the specific activity of 115.7 U/mg under the optimum condition. Finally, our study demonstrated that recombinant EGII could retain the endoglucanase activity for 89% and 80% at 40 °C and 50 °C, respectively.

In measurements of the intracellular free calcium concentration ([Ca2+]) using either microelectrodes or fluorescent probes, calibration is normally carried out in EGTA calcium buffer solutions. In the first part of the article the general properties of calcium buffer solutions are discussed, the equations used to calculate the apparent calcium binding constant (Kapp) are derived, and the difficulties in the calculation are discussed. The effects of the purity of EGTA as well as the influence of calcium contamination on the buffer solutions are explained. Because of the difficulties in calculating Kapp, and the importance of EGTA purity and calcium contamination, it is suggested that it is easier to measure all three under the appropriate experimental conditions using the method of Bers (1982). In the second part a do-it-yourself guide to the preparation of EGTA calcium buffer solutions is given. An experimental example is provided using the Bers method to measure purity, contamination, and Kapp. It is concluded that unless all three factors are known it is not possible to prepare accurate EGTA calcium buffer solutions.Key words: Ca buffers, EGTA, CaEGTA, Ca contamination, purity of EGTA.

The root growth angle defines how roots grow toward the gravity vector and is among the most important determinants of root system architecture. It controls water uptake capacity, nutrient use efficiency, stress resilience, and, as a consequence, yield of crop plants. We demonstrated that the egt2 (enhanced gravitropism 2) mutant of barley exhibits steeper root growth of seminal and lateral roots and an auxin-independent higher responsiveness to gravity compared to wild-type plants. We cloned the EGT2 gene by a combination of bulked-segregant analysis and whole genome sequencing. Subsequent validation experiments by an independent CRISPR/Cas9 mutant allele demonstrated that egt2 encodes a STERILE ALPHA MOTIF domain–containing protein. In situ hybridization experiments illustrated that EGT2 is expressed from the root cap to the elongation zone. We demonstrated the evolutionary conserved role of EGT2 in root growth angle control between barley and wheat by knocking out the EGT2 orthologs in the A and B genomes of tetraploid durum wheat. By combining laser capture microdissection with RNA sequencing, we observed that seven expansin genes were transcriptionally down-regulated in the elongation zone. This is consistent with a role of EGT2 in this region of the root where the effect of gravity sensing is executed by differential cell elongation. Our findings suggest that EGT2 is an evolutionary conserved regulator of root growth angle in barley and wheat that could be a valuable target for root-based crop improvement strategies in cereals.

The effect of EGTA, commonly present in Ca2+-free physiological saline solution, on the contractile responses induced by Ca2+ and phenylephrine was studied in dog mesenteric arteries and aortas of rats and rabbits. EGTA substantially enhanced the contractile responses of these vascular strips or rings to added Ca2+ after a prolonged preincubation period in the Ca2+-free medium. The maximal level of the enhanced contractile responses was independent of EGTA concentration, but the rate of the maximal responses was faster at higher EGTA concentration, presumably as a result of faster removal of intracellular Ca2+. Such a Ca2+-induced response was sensitive to the Ca2+ antagonist, nifedipine. EGTA present at low concentrations (50 and 400 μM) in Ca2+-free medium also inhibited the phenylephrine-induced contractile response more prominently for the longer preincubation periods of the vascular tissues in Ca2+-free medium. Our results suggest that EGTA, even when added at low concentrations to the vascular smooth muscle for a sufficiently long period in a Ca2+-free medium, may cause destabilization of the cell membranes leading to increased permeability to subsequently added Ca2+. EGTA may also remove the superficially bound Ca2+ and subsequently reduce the intracellular Ca2+ pool via extraction of the intracellular Ca2+ at the cell membrane surfaces.

Cells prepared from frog esophageal peptic glands by dispersal in low-Ca2+ medium (peptic acini) or 1 mM ethylene glycol-bis(-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA)-containing medium ("EGTA cells") were compared. EGTA cells were characterized by decreased secretory responses to agonists [bombesin (BB), acetylcholine, and isoproterenol] and intracellular messenger activators (forskolin, 12-O-tetradecanoyl-phorbol-13-acetate), decreased relative intrinsic efficacies of muscarinic agonists, and somatostatin insensitivity. Decreased BB and muscarinic receptor responses were not associated with changes in receptor number or characteristics. The time course of BB- and acetylcholine-stimulated pepsinogen secretion indicated that the marked reduction was confined largely to the late secretory phase (2–30 min), dependent on extracellular Ca2+, rather than early phase (2 min) secretion, which is related to release of intracellular Ca2+. The defect could be reversed by the Ca2+ ionophore A23187. BB-stimulated intracellular Ca2+ mobilization measured with fura-2/AM was similar in the two cell preparations, whereas BB-stimulated 45Ca2+ uptake was reduced threefold in EGTA cells, and this defect was also reversed by A23187. Somatostatin inhibited both BB-stimulated secretion and 45Ca uptake by peptic acini, but it had no significant effect on these parameters in EGTA cells. Cytochalasin B inhibited BB stimulation in peptic acini but not EGTA cells. These findings suggest that peptic cells isolated with EGTA exhibit decreased secretory responses that are due at least in part to impairment of a mechanism for uptake of extracellular Ca2+.

A pot experiment was made to study effects of ethylene glycol tetraacetic acid (EGTA, an inhibitor of Ca2+) and trifluoperazine (TFP, an inhibitor of calmodulin (CaM) on mycorrhizal colonization, growth performance, and chlorophyll, sucrose and glucose concentrations of four-month-old trifoliate orange (Poncirus trifoliata) seedlings under mycorrhization with Rhizophagus intraradices. Exogenous EGTA and TFP notably inhibited root mycorrhizal colonization, and the addition of EGTA also decreased soil hyphal length. In general, EGTA treatment decreased but TFP increased easily extractable glomalin-related soil protein (EE-GRSP) and total GRSP (T-GRSP) concentrations. In addition, EGTA and TFP applications generally significantly inhibited growth performance (height, stem diameter, leaf number, and shoot and root biomass), root traits (total length, surface area, volume, and number of 1st, 2nd and 3rd order lateral root), and chlorophyll a,b and a+b concentrations, the mycorrhizal inoculation generally reversed the negative effects and markedly increased these variables, irrespective of whether the seedlings were applied by inhibitors or not. EGTA and TFP treatments generally inhibited sucrose and glucose levels of leaf and root, except that TFP application notably increased root glucose in AM and non-AM seedlings. AMF inoculation resulted in carbohydrate modification: decrease in leaf sucrose, increase in root sucrose and leaf glucose, as well increase in root glucose under no-inhibitor and EGTA conditions and decrease in root glucose under TFP. It suggests that Ca2+ and CaM were involved in mycorrhizal and root development in trifoliate orange.
