The Saccharomyces cerevisiae genes ELM1, ELM2, and ELM3 were identified on the basis of the phenotype of constitutive cell elongation. Mutations in any of these genes cause a dimorphic transition to a pseudohyphal growth state characterized by formation of expanded, branched chains of elongated cells. Furthermore, elm1, elm2, and elm3 mutations cause cells to grow invasively under the surface of agar medium. S. cerevisiae is known to be a dimorphic organism that grows either as a unicellular yeast or as filamentous cells termed pseudohyphae; although the yeast-like form usually prevails, pseudohyphal growth may occur during conditions of nitrogen starvation. The morphologic and physiological properties caused by elm1, elm2, and elm3 mutations closely mimic pseudohyphal growth occurring in conditions of nitrogen starvation. Therefore, we propose that absence of ELM1, ELM2, or ELM3 function causes constitutive execution of the pseudohyphal differentiation pathway that occurs normally in conditions of nitrogen starvation. Supporting this hypothesis, heterozygosity at the ELM2 or ELM3 locus significantly stimulated the ability to form pseudohyphae in response to nitrogen starvation. ELM1 was isolated and shown to code for a novel protein kinase homolog. Gene dosage experiments also showed that pseudohyphal differentiation in response to nitrogen starvation is dependent on the product of CDC55, a putative B regulatory subunit of protein phosphatase 2A, and a synthetic phenotype was observed in elm1 cdc55 double mutants. Thus, protein phosphorylation is likely to regulate differentiation into the pseudohyphal state.

AbstractA well-optimized design of an ion cyclotron resonance heating (ICRH) antenna is very important for steady-state plasma heating with high radio frequency (RF) power of several tens of megawatts. However, a sharp decrease in the coupling RF power because of impedance mismatch of ICRH system is an issue that must be resolved for present-day fusion reactors and International Thermonuclear Experimental Reactor. This paper has theoretically analyzed the ICRH antenna's impedance matching for ELMy plasmas on experimental advanced superconducting tokamak (EAST) by the transmission line theory. The results indicate that judicious choice of the optimal feeder location is found useful for adjustable capacitors' tolerance to the variations of the antenna input impedance during edge-localized mode (ELM) discharge, which is expected to be good for the design of ICRH antenna system and for real-time feedback control during ELM discharge on EAST.

Abstract:
Extreme learning machine (ELM) is an efficient training algorithm for single-hidden layer feed-forward neural networks (SLFNs). Two pruned-ELM named P-ELM1 and P-ELM2 are proposed by Rong et al. P-ELM1 and P-ELM2 employ [Formula: see text] and information gain to measure the association between the class labels and individual hidden node respectively. But for the continuous value data sets, it is inevitable for P-ELM1 and P-ELM2 to evaluate the probability distributions of the data sets with discretization methods for calculating [Formula: see text] and information gain, while the discretization will lead to information loss. Furthermore, the discretization will result in high computational complexity. In order to deal with the problems, based on tolerance rough sets, this paper proposed an improved pruned-ELM algorithm, which can overcome the drawbacks mentioned above. Experimental results along with statistical analysis on 8 UCI data sets show that the improved algorithm outperforms the pruned-ELM in computational complexity and testing accuracy.

Abstract:
Integrated pest management calls for new biocontrol solutions in management of forest diseases. Endophytic fungi that are commonly found in tree tissue may have potential in biocontrol. However, the links between endophyte status and disease tolerance are still unclear, and we know little about the mechanisms by which the endophytes can influence tree pathogens. The first goal of the thesis was to compare the endophyte status in elm (Ulmus spp.) trees with low vs. high susceptibility to Dutch elm disease (DED), caused by Ophiostoma fungi, and to find correlations between endophytes and the susceptibility pattern of the trees. The second goal was to investigate the potential mechanisms of antagonism by the endophytes towards the pathogen. Thus, endophytes were isolated from leaves, bark and xylem of elms that differed in DED susceptibility. The isolates were screened for their potential to counteract the pathogen in dual cultures. Selected strains were investigated using Phenotype MicroArrays to obtain the substrate utilization profiles that reflect the endophytes’ ability to compete with the pathogen for a nutritional niche. To test for a protective effect against the disease, promising isolates were injected into young elms. Preliminary analyses were done to identify the extracellular chemicals that some of the endophytes released into the growth medium. The results showed that the frequency and diversity of endophytes was higher in xylem of elms with high susceptibility to DED. Some endophytes deadlocked the pathogen with extracellular chemicals in vitro, while others had a faster growth rate. Several endophytes were able to utilize substrates more effectively than the pathogen. A preventive treatment with endophytes protected elms against DED, but the effect was unstable across years. Bioactive fungal extracts had a complex chemical profile, and the individual compounds in the extracts remain to be identified. Because endophytes antagonized the pathogen through different mechanisms, I suggest that an endophytebased biocontrol of DED could be best achieved through a synergistic effect of several endophyte strains.

A lack in infrastructure and skills, in an inherited Apartheid spatial legacy, leaves a critical opening for much needed added spatial value. By investigating and supporting existing networks and contributing to existing typologies, this dissertation speculates on educational resource infrastructure provision to marginalized communities in Pretoria, South Africa. It is a rethinking of our South African city landscapes and civic/pedagogical architectural offerings, manifesting a critical stance to foster a prosperous community that has the potential to thrive. Focusing a speculative knowledge exchange infrastructure intervention on already existing networks; this dissertation aims to resolve the potential of current typologies in urban planning, and the ability to foster a new teacher, pupil and community education infrastructure to empower local stakeholders to improve provided services. Building a prosperous future on top of past spatial inheritance.

Jean-Claude Eloy est un compositeur passionné de musiques extra-européennes (et en particulier de musique japonaise) qui revendique un multiculturalisme militant et refuse farouchement que sa démarche puisse être associée à un quelconque exotisme. Pourtant, et sans que la sincérité de son positionnement idéologique puisse être remise en cause, certaines ambiguïtés apparaissent au fur et à mesure de l’étude de sa production. Ainsi, si l’exotisme de surface caractéristique du XIXe siècle a disparu au profit d’un intérêt largement documenté pour les cultures musicales non occidentales, on n’en décèle pas moins la permanence de thèmes caractéristiques de cet exotisme. Il s’agira d’analyser ce paradoxe et d’en proposer des explications. Le travail part de l’hypothèse que l’exotisme repose sur un imaginaire anthropologique à partir duquel prennent naissance des expressions différentes selon les lieux et les époques. L’exotisme répondrait ainsi à des réalités humaines dans lesquelles le contexte colonial aurait joué un rôle uniquement facilitateur (et non suscitateur). Cet exotisme ne pourrait donc pas avoir disparu avec la fin des empires. Il s’agira ainsi de lire les ambiguïtés d’Eloy à la lumière d'un « mythe exotique » toujours vivace dans l’imaginaire collectif contemporain. S’appuyant sur le courant des postcolonial studies, cette lecture interrogera les résurgences exotiques présentes dans l’œuvre d’Eloy pour aboutir à la définition d’un exotisme postcolonial différente de celles qui ont pu être développées dans le domaine littéraire
Jean-Claude Eloy is a French composer. He is passionately fond of extra-European music, especially Japanese music. He calls for muticulturalism and refuses his works to be associated with exoticism. Even so - and without questionning Eloy’s intellectual honesty - it is possible to find out some ambiguities in his works. For instance, even though superficial exoticism, typical of XIXth century, no longer appears in his music, several themes of this exoticism can be found in some of this works. We will analyze this paradox and suggest some explanations. Our thinking is based on the proposition that exoticism comes from anthropoligical imagination and takes a variety of forms depending on the era. According to this proposition, colonialism has made the development of exoticism easier but didn’t create it. As a result, exoticism could not completely disappear with the end of colonial empires and still exists as a human reality. Postcolonial studies, semiotics and hermeneutics will be used as a background approach to explain the existence of exoticism in Eloy’s works

Le développement des organismes multicellulaires, la morphologie de leurs organes, la formation de connections nerveuses ou même la réponse immunitaire conduisent à la formation de cellules dont la mort a été programmée, ces cellules sont en apoptose. Les corps apoptotiques générés sont éliminés par phagocytose, que l'on peut ainsi considérer comme l'étape ultime du programme apoptotique. Cette élimination, assurée soit par des phagocytes professionnels ou des cellules environnantes est très efficace et peu de cellules en apoptose sont observées dans les différents tissus et organes. Deux familles de protéines, ELMO et DOCK, sont connues pour être au centre d'une voie de signalisation impliquée dans l'activation de la petite GTPase Rac et le remodelage du cytosquelette d'actine permettant l'internalisation de corps apoptotiques, mais la manière dont ELMO est activée par la reconnaissance des corps apoptotique reste largement méconnue. Récemment, ELMO a été identifiée comme cible de la kinase hématopoïétique Hck. La phosphorylation d'ELMO constitue un signal d'activation de la voie de phagocytose. Nous avons pu montrer que les domaines N et C-terminaux d'ELMO sont impliqués dans l'interaction avec le domaine SH3 de Hck. Contrairement au SH3 de DOCK, l'interaction du domaine C-terminal avec le SH3 de Hck est strictement dépendante de la présence du polyproline. Les données obtenues sur les cellules transfectées suggère l'interaction in cellulo du motif polyproline avec le domaine SH3 de Hck. La différence dans le comportement du domaine C-terminal d'ELMO avec les deux SH3 nous a mené à identifier un complexe ternaire entre les trois protéines. Le motif polyproline d'ELMO est adjacent à l'une des 5 tyrosines phosphorylées par Hck, la tyrosine 720, nos résultats montrent que la phosphorylation de cette tyrosine défavorise l'interaction entre le domaine C-terminal et le SH3 de Hck. Cette phosphorylation pourrait être à l'origine d'un effet régulateur de Hck sur le complexe ELMO/DOCK
Multicellular organisms development, tissue organization and morphogenesis, nerve connection network or even immune response lead to programmed cell death or apoptosis. Phagocytosis is the mean for the engulfment of the apoptotic corpses and can thus be considered as the final step of the apoptotic pathway. Both non-professional and professional phagocytic cells are implicated to achieve an efficient clearance of the apoptotic cells and debris, and very little dead cells are actually present in tissues under normal circumstances, preventing secondary necrosis and further inflammation. Two family of proteins, ELMO and DOCK have been identified as major characters in the signaling pathway leading to the activation of the small GTPase Rac, and actin remodeling, that eventually leads to engulfment. Nevertheless, very little is yet known about the upstream events leading to ELMO activation. Recently, ELMO has been characterized as a target for the hematopoietic cell kinase Hck, and consequent phosphorylation participates in ELMO activation. In the course of the present work, we demonstrated that both N- and C-terminal domains of ELMO are binding the SH3 domain of Hck. In contrast with the SH3 domain of DOCK, the SH3 domain of Hck binding to the C-terminal of ELMO is strictly dependent upon the polyproline motif. Our data obtained using transfected cells, further suggest the polyproline dependent interaction also occurs in cells. Taking into account the differential behavior of both SH3 domains from DOCK and Hck we next demonstrated a ternary complex formation between ELMO and both partners in vitro. Finally, we investigated the possible role for the Hck phosphorylation of Y720 located at the edge of the polyproline motif of ELMO and demonstrated that this phosphorylation clearly increases the Kd of ELMO/Hck interaction suggesting this phosphorylation could be part of a regulation loop in Hck dependent ELMO activation

Abstract:
The design and R&D for ITER In-Vessel Coils (IVCs) is being deployed. The concerned issue of “Edge Localized Modes” (ELMs) and “Vertical Stabilization” (VS) of the ITER plasma can be addressed by the implemented IVCs. The ELM and VS coils will be installed in the vessel just behind the blanket shield modules to reach the requirement of keeping strong coupling with the plasma. The 59mm Stainless Steel Jacketed Mineral Insulated Conductor (SSMIC) using MgO as the insulation is being designed for the IVCs to resist the special challenges, including the nuclear radiation, high temperature, electromagnetic and thermal fatigue. It is necessary to take the mechanical performances of the SSMIC and the feasibility of fabrication techniques into consideration of the R&D program. The mechanical performances of the SSMIC close to the actual work conditions, including the three point bend modulus, three point bend cyclical performance and the cyclical performance with a U-bend sample of the SSMIC prototypes have been investigated and the results are presented in this paper.
