Abstract:
The paper discusses the personal and political relations between the French surrealist poet Paul Éluard and Serbian writers and intellectuals, first of all Marko Ristić, a theorist and one of the founders of the Serbian surrealist movement. We will show the reception of Éluard’s works in the Serbian cultural space, his visit to Yugoslavia, then Éluard’s activities in Paris related to Yugoslav politics. One significant part of the article represents, until now, unpublished archival documents that testify to the friendship between Éluard and Ristić. We will try to explain how this friendship ended after the Cominform Resolution in 1948 and what consequences it had on Serbian and French culture.

Esta tesis presenta el estudio de una recopilación de textos poéticos dePaul Eluard seleccionados según el criterio de su afinidad con España.La interacción entre el poeta y España se evidencia partiendo delpropio texto poético minuciosamente analizado, comprobando a posteriori laveracidad de vivencias y hechos históricos con los datos meramentebiográficos o históricos aludidos. El corpus textual integra poemas desde elaño 1924 con el libro 9, "Mourir de ne pas mourir", hasta los poemas políticosdel libro 86 "Espagne", "En Espagne", y "Vencer juntos".La aproximación al sentido de la poética de Eluard se ha realizado conel estudio desde la triple perspectiva de la retórica, la poética y de la semio -lingüística. Este estudio ha puesto de manifiesto la escritura forjada por unalto impulso expresivo que consiguió plasmar en su imagen poética unaquerencia por lo español.En la poesía de Eluard se han detectado rasgos específicos dereminiscencias barrocas combinados con una técnica imaginativa de unsurrealismo muy personal de fuerte corte picassiano.
This doctoral thesis presents the study of a collection of poetic texts by PaulEluard selected according to their relevance with respect to Spain.The interaction between the poet and Spain is illustrated by an in-depthanalysis of the poetic text and the veracity of the poet's experiences and of thehistorical events narrated is then further corroborated by biographical andhistorical data. The textual corpus consists of poems ranging from 1924'sbook 9 "Mourir de ne pas mourir", to political poems included in book 86 suchas "Espagne", "En Espagne", and "Vencer juntos".Eluard's poetic sense has been approached from the three-foldperspective of rethoric, poetics and semio-linguistics. This study revealswritings born out of a sharp expressive impulse that succeeded in capturinghis longing for Spain.Eluard's poetry reflects specific traits of baroque reminiscencescombined with an imaginative technique embodied by a personal surrealismwith Picassian influences.

Abstract:
L'etude quantitative et qualitative de l'humour dans l'oeuvre de paul eluard conduit a analyser ses rapports avec le langage poetique. Le terme humour designe a la fois l'humour freudien, l'esprit, le comique et l'ironie. L'hypothese de la source inconsciente de l'humour eclaire le role de l'effet d'automatisme dans l'ecriture surrealiste. Notre methode s'inspire de la stylistique de m. Riffaterre, elle se fonde egalement sur la description psychologique de freud du mecanisme de liberation de l'humour et sur sa classification des techniques de l'humour ; elle utilise le concept de connotation tel que l'a defini c. Kerbrat-orecchioni ; son originalite tient dans l'etude des effets produits par les textes : effets esthetique, humoristique et humoristhetique (h e). La nature de ces effets est controlee pour chaque exemple aupres d'informateurs. La partie theorique, outre la definition des objectifs, de la methode et du terme humour montre qu'il n'y a pas lieu de distinguer les genres esprit, comique, humour, ironie, de mature et de techniques voisines (seule la technique de l'ironie est originale), dans une etude des effets. La partie analytique montre l'existence de toutes les varietes des techniques de l'humour dans les oeuvres, dans les textes dadaistes aussi bien que dans les autres; elle identifie egalement les traits proprement ironiques. L'analyse de textes dus a l'effet d'automatisme montre la relation entre automatisme et production des effets humoristiques et h e, ces derniers se revelant meme dans des textes graves. La partie synthetique demontre la similitude des structures des metaboles et des techniques de l'humour. L'effet des figures de rhetorique n'est pas fixe d'avance : il depend de leur utilisation en contexte ainsi que des modalites de la lecture. L'etude de l'idiolecte eluardien decrit des techniques humoristiques familieres au poete et nos pastiches reprennent certains de ces procedes. La conclusion dresse la liste des facteurs neutralisateurs et des facteurs liberateurs de l'humour. Finalement, la part de l'ironie est reduite et l'humour franc existe surtout dans la periode dadaiste, tandis que l'effet h e domine dans presque tous les textes. Cet effet caracterise l'ecriture surrealiste ou fonctionne d'une facon privilegiee l'effet d'automatisme
Humour in the works of paul eluard. A qualitative and quantitative survey of humour in paul eluard's works, lead to the analysis of its connotations with poetic language. The term humour refers to freudian humour, to wit, to comedy and to irony as well. The assumption that the origin of humour lie in the unconscious, explains the role of the consequence of automatism in surealist writing. M. Rifaterre's stylistics has inspired us for our method; which is also based on freud's psychological description of the release of humour. The concept of connotations according to c. Kerbrat - orecchioni's definition is used in this method, its orginality results from the survey of the varions effects generated from the texts : esthetic effects, humouristic effects and humou- ristic effects (h e). The nature of these effects is supervised by other colleagues. The theoretical part, besides defining goals, methods, and the term humour, shows that characterizing styles such as wit, humour, comedy, or irony proves to be quite irelevant, although they are similar in essence and in approach in a survey of the effects. (the approach of irony being the only original one). The analytict part reveals all the various approaches of humour throughout the works ; in dadaist texts as well as in others. In this section, the specific ironical aspects are clearly defined. The analysis of texts deriving from the consequence of automatism shows the relations between automatism, humouristic effets and h e, the latter is percievable even in solemn texts. The synthetic part demonstrates the similarity between the structure of figures of style and the approach of humour. The effect of rethorical figures of style is not defined be- forehand. It depends on their use in a specific context and also on the mode of reading. The survey of eluard's idiolect describes humouristic approaches which are familiar to the poet and in our pastiches, some of these approaches an repeated. The conclusion establishes a register of neutralizing and liberating factors of hu- mour. Finally, the part of irony is reduced and undisguised humour exists mostly in the da- daist period, whereas the h e effect prevails in alinost every text. This clearly marks the surealist writing where the consequence of automatism is highly priviledged

Ouvert par une exegegese des poemes publies par paul eluard entre 1924 et 1938, ce travail se poursuit avec une etude linguistique des mecanismes de la metaphore et s'acheve sur une restitution des sources et modeles eluardiens incluant la pratique du collage et du cadavre exquis. Il apparait que les poemes eluardiens disent et miment leur propre effectuation en retrouvant des symboles et des scenarios habituellement reperes par l'histoire des religions dans l'initiation et la cosmogonie mais finalement presents dans la culture surrealiste. Un veritable fantasme de l'origine est identifie et suivi dans la propension d'eluard a choisir telle ou telle image, telle ou telle forme syntaxique et a se rattacher a telle ou telle autre experience : chez eluard, les tensions semiques executent ce que les images donnent a voir alors que ce que fomente et valorise le groupe surrealiste favorise et modelise le tout. Un apercu sur l'evolution d'eluard y trouve son compte dans la mesure ou les primes figures du mal social sont decouvertes dans les figures de l'origine les plus depreciees
Opening with an analysis of poems published by paul eluard between 1924 and 1938, the work then proceeds with a linguistic study of metaphor mechanisms to end with a reconstruction of eluard's sources and models including the practice of collage and cadavre exquis. It appears that eluard's poems act and say their own genesis by recapturing symbols and developments usually revealed in the history of religions through initiation and cosmogony but equally alive in the surrealists' culture. Real fantasms of origin are identifiable throughout eluard's tendencies to choise such and such images, such and such syntaxic forms or to connect himself to such and such experiences : with eluard, semantic conflicts are in keeping with what images suggest whereas what is fomented and exhauced by the surrealist group will favour and shape the whole of it. A survey of eluard's evolution is all the more rewarding as the first images of social evil are clearly manifest in the most underrated images of origin

Dans cette étude, nous faisons d'abord deux différentes distinctions d'ordre "langagière" : entre l'expression en images et l'expression en mots. Les mots dans l'oeuvre d'Eluard peuvent acquérir des sens symboliques, métaphoriques ou perceptifs. Les images font référence à l'espace psychologique et visent souvent l'espace intermédiaire entre l'intérieur et l'extérieur d'un corps. Ensuite nous élargissons vers les thèmes et les sujets de cette poétique. Nous trouvons d'abord les questions de la nature et de l'esprit et de leur interaction dans l'espace. Lié à cela, se trouve la question de l'imagination et de la nature ainsi que celle de la correspondance entre les arts (tradiction à partir de Baudelaire). .
In this work, our main aim is to clarify the relation between texte and space in the work of the French poet Paul Eluard (1895-1952). We are focalising the themes and topics by using the methode of the so called critical thematics (founded by Jean-Pierre Richard and Michel Collot). Therefore we mainly discuss thematical matters like the role of phenomenology in the work of Eluard : questions concerning the relation between nature and spirit, imagination and nature, the correspondance of the arts and the role of spatial components in our perception of the world, are frequently evoked in his work. .

Les objets dans les poemes d'eluard sont d'abord des choses reelles. Mais, apres avoir ete selectionnes et combines de facon particuliere, ils deviennent quelque chose de surreel. Dans cette these, nous etudions les differentes manieres de production de la surrealite chez eluard. Il y a quatre categories de traitement des objets : la metaphorisaion des objets, leur union, leur juxtaposition et leur destruction. La metaphore est une des plus importantes categories methodiques de traitement des objets dans les poemes eluardiens, parce qu'elle a principalement pour fonction de transferer. Parmi certaines formes metaphoriques chez eluard, surtout, les metaphores non motivees sans compare, rendent les objets obscurs et surreels. Nous avons la complicite de la structure metaphorique dans le processus d'obtention de l'effet de surrealite des objets. Et eluard metaphorise souvent les objets selon les principes de la relativite et de la difference : par consequent, les metaphores chez ce poete ont, pour fonction de devier et d'interrompre. C'est a partir de cela que l'on peut affirmer que les objets metaphorise chez eluard deviennent etranges. Pour unir les objets, eluard utilise plusieurs methodes delicates : la superposition, le rassemblement et la connexion. Eluard unit un objet a d'autre tres distincts de lui. Par consequent, les objets unis deviennent surreels. Eluard utilise quatre manieres de juxtaposition : l'affrontement, l'inversion, la substitution et l'opposition. La juxtaposition augmente la surrealite des objets et realise une forme esthetique. La destruction des objets enfin est aussi une procedure d'obtention de la surrealite. Eluard detruit les objets a partir de quatre niveaux : la deformation, la denaturation, le depaysement et le destitution. Avec ces methodes, eluard cree des objets detaches de leurs sens usuels
Objects in eluard's poems are first real things. But after being selected and combined particularly, they become something surreal. In this thesis, we study the different manners to produce the surreality in eluard's poems. There are four types of treatment of the objects : the metaphor of the objects, their linkage, their juxtaposition, their destruction. The metaphor is one of the most important methodical categories of treatment of the objects in eluard's poems, because its main function is to transfer. Among certain metaphorical forms with eluard, specially the motiveless without compare metaphors make the objects obscure and surreal. One gets the complicity of the metaphorical structure in the process of obtaining the surreal effect of the objects. Eluard often write the objects metaphorically according to the principles of relativity and difference : therefore, the metaphors of this poet are used to deviate and to interrupt. Thus one can assert objects which are said metaphorically by eluard become strange. So as to link the objects, eluard uses several delicate methods: the superposition, the assemblage and the connection. Eluard links an object with others wich are different from it. Therefore, the linked objects become surreal. Eluard uses four ways of juxtaposition : the confrontation, the inversion, the substitution and the opposition. With eluard, the juxtaposition heightens the surreality of the objects and makes an aesthetic form. At last, the destruction of the objects is also a process of obtaining the surreality. Eluard destroys the objects from four levels : the deformation, the denaturation, the removal and the dismissal. With these methods, eluard creates objects which are taken off their usual meanings

Abstract:
Nous montrons que la poésie moderne, comme le mythe, vient pallier, par son pouvoir créateur de réalisation des images, les vides de la pensée. Elle substitue ainsi aux impenses du réel, une réalité ordonnée par la pensée mythique. L'impense se trouve en effet à l'origine de l'invivable, car il génère l'incapacité à nommer les choses et confronte l'être a un état de vertige. Cet état invivable entraine la nécessite d'un passage dans lequel le poète nie les structures du monde invivable, constituées de l'espace et du temps, et procède a l'invention d'un autre espace-temps se formant dans une écriture capable de l'exprimer. Celle-ci met en scène le poète dans le mythe de ses origines qui l'arrache du monde invivable. Le passage constitue une période de gestation faisant naitre le poète avec de nouvelles racines, ancrées dans un nouvel espace-temps. C'est ainsi que l'écriture fonde l'existence, car le poète est à l'origine d'un lieu auquel son imaginaire a donné forme. Elle fixe en effet ce que l'imagination a construit dans la rêverie, se trouvant ainsi à l'origine de la réalisation de cette rêverie. C'est en cela que l'on apparente l'écriture au nominalisme, en tant que réalisation des images substituées aux éléments du réel invivable. Quand, en effet, la pensée discursive montre ses limites, elle se trouve relayée par une pensée symbolique qui substitue aux concepts du discours, les images issues de l'imagination. Ce travail contient une partie épistémologique, ou nous définissons le mythe et la poésie moderne, que nous rapprochons dans une axiomatique convergeant vers la problématique constituée du plan. Celui-ci considère les trois phases dans lesquelles se développe l'écriture: l'écriture, le passage, et l'existence par l'écriture. C'est ainsi qu'une partie de lectures fait évoluer les œuvres de Rimbaud, Éluard et Jabès selon ces trois périodes constitutives de la parole mythique. La conclusion expérimente ces lectures sur les œuvres de Michaux. .
We demonstrate that modern poetry, like myth, compensates for blanks in through its image making creative power. It thus replaces real non-thinking with a reality ordered by mythical thinking. No thought in fact is the origin of non-life, because it generates an inability to name things and confounds the individual with a state of vertigo. This state of non-life creates the need for a transition in which the poet denies the structures of the nonviable works, made up of space and time, and proceeds to invent another space-time taking form in writing capable of expressing it. This places the poet in the myth of his origins which removes him from the non-livable world. The transition is a gestation period in which the poet is born with new roots, anchored in a new space-time. In this way writing funds existence, as the poet creates a place which his imaginary has formed. It fixes what the imagination has built in musing, thus bringing about the realization of this reverie. That is how writing is compared to nominalism, as a realization of the images replacing the elements of the non-livable reality. So when discursive thought reaches its limits, it is relayed by symbolic thought replacing the concepts of speech, the imaging issuing from the imagination. This work contains an epistemological part, where we define myth and modern poetry, which we bring together axiomatically converging towards the problematic constituted in the plan. It considers the three phases in which writing develops: the non-livable, the transition, and the existence through writing. In this way certain interpretations show the work of Rimbaud, Eluard and Jabès evolving in accordance. .

Despite recent advances made in the field of cancer biology and the therapies that have been developed [1, 2], it is clear that more advances are necessary for us to classify cancer as curable. The logical question that arises is simple: Why, despite all the technologies and medical innovations of our time, has a complete cure eluded us? This chapter sheds light on one of cancer’s most impactful attributes: its heterogeneity and, more specifically, the intratumoral heterogeneity of cancer metabolism. Simply put, what makes cancer one of the deadliest diseases is its ability to change and adapt. Cancer cells’ rapid evolution, coupled with their irrepressible ability to divide, gives most of them the advantage over our immune systems. In this chapter, we delve into the complexities of this adaptability and the vital role that metabolism plays in the rise and progression of this heterogeneity.

AbstractScar assessment has long relied on a variety of instruments, most of them questionnaires or scales that are susceptible to bias and limit the level of evidence in research. Throughout recent years, a variety of objective assessment tools have been developed. They allow for the exact quantification of different scar parameters such as texture, pliability, color, and size, thus facilitating detailed progress analysis throughout treatment.The advent of new technology, however, has its pitfalls, too. The number of different technologies available is hard to review, and oftentimes, clinical validation is lacking, making it hard for researchers to gauge whether a certain instrument is adequate for their individual projects.The use of each individual instruments has different requirements regarding patient preparation and calibration, and the measurements themselves can often be adjusted through a variety of parameters. This requires intensive familiarization with the respective instruments. Additionally, interpretation of the data can be complicated, and distinguishing statistically significant differences from clinically important differences remains difficult.Modern technology, however, allows researchers to measure treatment effects that otherwise would have eluded them through the use of subjective evaluation only.Through objective scar assessment technology and its detailed abilities to document changes throughout treatment, the evidence in scar-related research can be greatly improved, and current as well as new treatment options can finally be adequately gauged for their efficacy.

AbstractBeing able to argue about the performance of self-adjusting data structures such as splay trees has been a main objective, when Sleator and Tarjan introduced the notion of amortised complexity.Analysing these data structures requires sophisticated potential functions, which typically contain logarithmic expressions. Possibly for these reasons, and despite the recent progress in automated resource analysis, they have so far eluded automation. In this paper, we report on the first fully-automated amortised complexity analysis of self-adjusting data structures. Following earlier work, our analysis is based on potential function templates with unknown coefficients.We make the following contributions: 1) We encode the search for concrete potential function coefficients as an optimisation problem over a suitable constraint system. Our target function steers the search towards coefficients that minimise the inferred amortised complexity. 2) Automation is achieved by using a linear constraint system in conjunction with suitable lemmata schemes that encapsulate the required non-linear facts about the logarithm. We discuss our choices that achieve a scalable analysis. 3) We present our tool $$\mathsf {ATLAS}$$ ATLAS and report on experimental results for splay trees, splay heaps and pairing heaps. We completely automatically infer complexity estimates that match previous results (obtained by sophisticated pen-and-paper proofs), and in some cases even infer better complexity estimates than previously published.

Factor VIII/vWF displays high affinity for matrix bound sulphate groups. Linear salt and pH gradient elution from Dextran Sulphate Sepharose at 4°C resulted in complete separation of the complex from major protein contaminants with typical yields of 70-90% vWF:Ag and vWF (RCoF). Elution in physiological calcium concentrations gave VIII and VIII:Ag yields of 35% and 65% respectively. Addition of L-Lysine (1.0M) to all buffers inhibited VIII/vWF binding, although lysine gradients (0-1.0M) gave comparable binding and elution profiles as C-1.0M NaCl, but with impaired resolution between protein moieties. However, in the presence of L-Lysine, yields of VIII and VIII:Ag were significantly improved to 80% and 100% respectively when assay standardizations with appropriate lysine concentrations were performed. Furthermore, the binding of fibrinogen could be inhibited by 0.15M L-Lysine, 0.075M NaCl in the equilibrating buffer.The presence of enzymic activity, as assessed by S-2222 and S-2238 in the eluting fractions could be abolished by the application of recryoprecipitated material pre-adsorbed with Al(OK)3 and celite. Incorporation of lysine to the buffers with the associated increase in yields further supports the potential viability of the separation principle for large scale purification of Factor VIII.

Abstract:
Treatment of bleeding in von Willebrand's disease usually consists of the infusion of cryoprecipitate or plasma. DDAVP is effective in some patients. Commercial concentrates for the treatment of Hemophilia A are ineffective as a source of von Willebrand Factor (vWF) replacement in von Willibrand's disease presumedly because of the absence of higher molecular weight forms of vWF protein. A vWF concentrate obtained during the course of preparation of an affinity purified Factor VIII may provide an alternative therapeutic agent without impacting the available Factor VIII supplies. The process used for the preparation of a highly purified Factor VIII concentrate (MonoclateTM, Armour Pharmaceutical Co.) from cryoprecipitate includes an affinity chromatography step which separates vWF/Factor VIII complex from other proteins in cryoprecipitate using an anti-vWF monoclonal antibody (C. A. Fulcher & T. S. Zimmerman, 1982). Factor VIII is then dissociated from the vWF remaining on the column and is eluted immediately by 3M sodium thiocyanante (NaSCN) as a step in the regeneration of the column. Unless the NaSCN is rapidly removed from the elutriant, the vWF activity as measured by platelet agglutination is destroyed. We have taken the NaSCN eluate and processed it immediately over an in-line G-10 or G-25 Sephadex column which removes the NaSCN while the vWF is eluted with a buffered isotonic solution. Alternatively, the vWF has been precipitated from the NaSCN by ammonium sulfate or polyethylene glycol. VWF prepared by any of these methods retains platelet agglutinating activity and has a distribution of vWF multimers similar to those of vWF in normal plasma.The potency of vWF prepared from cryoprecipitate by this process is 20 units/ml and the specific activity is 20 units/mg. In several fractionations of kilogram amounts of cryoprecipitate, vWF was isolated and subjected to lyophilization and heating at 68° for 30 hours without loss of bioactivity. Drug development in progress suggest that the combined effect of affinity chromatography and exposure to NaSCN may negate the need for a heating procedure to reduce the risk of viral transmittance.

Abstract:
The American Red Cross is developing a Coagu.lation Factor X (FX) concentrate to provide a safer alternative for replacement therapy in Factor X deficient patients, who can experience thromboembolic complications with current treatments. Based on a survey of hemophilia treatment centers, we estimate the frequency of the homozygous disorder to be approximately 1/150th that of hemophilia A, or about 65 patients in the USA. We have devised a method for producing FX as a by-product of our Coagulation Factor IX concentrate (FIX). The method starts with adsorption of cryoprecipitate supernatant plasma with DEAE-Sephadex resin followed by elution of Vitamin K-dependent coagulation factors. This material is adsorbed to sulfated dejctran resin and Factors II and X are eluted by increasing the salt concentration. At 0.45 M NaCl, FII elutes quickly while FX is retarded and can be recovered essentially free of FIX by collecting the slower eluting material. FIX is then recovered at still higher ionic strength. The pooled FX is concentrated, diafiltered and treated to inactivate viruses. Approximately 30% of plasma FX was recovered in pilot scale experiments (600 liters plasma). Specific activity was > 51 FX units / mg protein corresponding to a purity of around 50% and 3000-fold purification over plasma. The ratios of Factors "X : II : IX : Protein C were 1.0 : <0.03 : <0.03 : 0.2. The major contaminant, conprising nearly 50% of the protein, was found to be inter-alpha trypsin inhibitor (IaI), a serine protease inhibitor whose function in plasma has not yet been determined. This inhibitor is also present in the DEAE-Sephadex eluate and. in the FIX concentrate. However, Western blot and HPLC analyses have shown that IaI is present in two different forms.In FX it behaves as expected for the IaI monomer (Mr = 160 kDa), while in the DEAE-eluate and in FIX it exists also in r higher molecular weight form (≥400 kDa) corresponding either to aggregates, complexes or larger native species not previously described. The nature of the possible interaction of Ial w.vdi these coagulation factors is unknown and is currently boinn evaluated.

1MAC principles were used to investigate the binding of Factor VIII/vWF from either Al(OH)3 - absorbed citrated PPP or cryoprecipitate using citrate-saline buffers. Factor VIII/ vWF showed high affinity for both Zn2+ - or Cu2+ - immobilised on biscarboxymethylaminoSepharose 4B at ambient room temperature, but no interaction with Ca2+, Mg2+, Mn2+ or Cd2+ - immobilised matrices.Factor VIII/vWF could be re-eluted as a single component from Zn2+ - columns using either decreasing pK, or increasing competitor ligandconcentrations. In pK gradients comprising 14mM citrate, 2.14mM CaCl2, 0.15m NaCl Factor VIII eluted at pH 6.50, whereas in pH gradients comprising 0.1m Tris, 2.5mM CaCl2 pH 7.5 and 0.1M succinate, 2.5mM CaCl2 pH 5.5 Factor VIII eluted at pH 6.06. Citrate behaves as a competitor ligand; at 4° in citrate buffer Factor VIII did not bind.In citrate-saline buffers pH 7.2 Factor VIII could be reeluted in high yield (100% Factor VIII:Ag; 70% VIII:C; 90% vWF:Ag) through application of linear competitor ligand gradients (at 11.5mM imidazole, 15.4mM dl-Eistidine, 45mM L-Lysine, or 1.7% BSA) , whereas 1.0M NaCl or 1.0M NH4Cl were ineffective. The Factor VIII activities co-eluted ahead of, but incompletely resolved from the re-eluted Fg and Fn. If 5 mg/ml. BSA was incorporated into the buffers Factor VIII was quantitatively recovered in the column wash - through fractions leaving all the Fg and Fn bound to the matrix.

Abstract:
A functional anti thrombin III CAT III) deficiency has been identified in three generations of a family with a high incidence of deep venous thrombosis. The deficiency presented as ~50% reduction in its heparin cofactor activity compared to its antigen concentration. No abnormality was detected by crossed immunoelectrophoresis of plasma in the presence or absence of heparin. Plasma from the propositus was precipitated with dextran sulphate, applied to heparin-Sepharose and the AT III stepwise eluted with NaCl . The AT III had a reduced ability to inactivate thrombin, when this was monitored by substrate hydrolysis or by SDS PA gel e1ectrophoresis. Its mobility was normal when examined by the latter technique on 10-20X gels under reducing and non-reducing conditions. Patient AT III was reapplied to heparin-Sepharose and eluted with a NaCl gradient. A minor active pool eluted in the same concentration range, 0.5-1.2M, used to purify normal AT III, while predominantly inactive AT III eluted at higher NaCl concentrations, 1.2-2.0M. Further purification of variant AT III was achieved by passage of heparin-Sepharose eluates through a thrombin-Sepharose column and structural studies are being undertaken on this material. It is concluded that AT III Glasgow has increased affinity for heparin but reduced ability to inactivate thrombin. It can be differentiated from two other AT III variants that bind heparin with high affinity, Milano and Northwick Park, but has similar features to those of AT III Chicago.

The present report first deals with the isolation and characterization of a substance found to demonstrate protein C like activity(PCLA). When it was activated by thrombin, the PCLA substance prolonged the activated partial thromboplastin time(aPTT). The activated PCLA substance also hydrolyzed synthetic substrates such as S�2238, S�2366 and S�2266,while the PCLA substance was not cross reacted with anti-protein C serum(Behring Manheim). The PCLA substance was adsorbed by both aluminium hydroxide gel and barium sulfate. The level of Km, optimum pH and optimum I.S. in the amido-lytic reaction with synthetic substrate is shown in the table.In the chromatography of Sepharose CL-6B gel, the PCLA substance was eluted in the fraction of higher molecular weight, while protein C was eluted with a lower fraction.This prompts an estimate that the molecular weight of the PCLA substance is approximately 200,000. Through DEAE-Sepharose CL-6B gel ion exchange chromatography, the PCLA substance was eluted by a lower ionic strength than was protein C.The levels of a PCLA substance were low in the patients with a vitamin K deficiency and in newborn infants. It is expected that our findings regarding the PCLA substance will be as clinically significant as that which we know about protein C at present.

AT exists as two isoforms differing in affinity for heparin (Thromb Res 27:23,1982; JBC 260:610,1985). The binding of human AT isoforms to commercial heparin, or purified unbleached heparin was studied. These heterogeneous, mixed-affinity (MA) heparins were coupled to agarose. When columns of these resins were eluted with NaCl gradients, after mixtures of the higher (ATvh) and lower (ATh) affinity isoforms of AT had been applied, ATvh always eluted after ATh. To determine the nature of this difference, low-affinity (LA) heparin was prepared by repeated passage of MA heparin over an ATh-ConA-Sepharose column. Neither AT isoform bound during passage over a LA-heparin agarose column equilibrated with 0.15 M NaCl. However, the ATvh was significantly retarded, while ATh eluted at about the same position as BSA. To demonstrate that ATh also interacts with a LA-site on heparin, a column containing MA heparin was loaded with ATh until the effluent concentration reached a steady state. Under the conditions, 21 mg of ATh was associated with the column in the steady state. Of this 10 mg eluted during a wash with 0.15 M NaCl, and 11 mg with 1.5 M NaCl elution. When the same experiment was performed using ATvh in place of ATh, approximately 25% more ATvh bound in both the steady state and to the washed column. To demonstrate that a secondary interaction is responsible for the separation of ATvh and ATh during heparin-agarose chromatography, the isoforms were applied together to MA-heparin columns in less than saturating amounts. When NaCl gradients were used to elute the columns in a reverse direction (bottom to top of column), ATh and ATvh co-eluted at relatively low salt concentrations. Thus, separation results during filtration of ATh and ATvh through the columns, after neutralization of the HA interaction. Finally, to determine if ATh and ATvh compete for the same binding sites on heparin, a MA-heparin column was loaded with a saturating amount of ATh. When ATvh was applied to this column, ATh was quantitatively displaced. It is concluded that (1) both AT isoforms bind LA sites on heparin (2) heparin contains many more LA sites than the HA ones; (3) ATvh binds more strongly to LA sites than ATh; and (4) LA binding may enhance activation of AT at the HA site by increasing the rate at which AT binds to this site.
