Abstract:
System identification finds nowadays application in various areas of biomedical research as a tool of empiric mathematical modeling and model individualization. Hormone regulation is a classical example of biological feedback where control theories, in general, and system identification, in particular, are indispensable in unraveling the regulation mechanisms and explicating the complex dynamical phenomena arising in endocrine systems. The main function of endocrine feedback regulation is to maintain the hormone levels within a particular physiological range as well as to sustain an appropriat

Endocrine Disrupting Chemicals (EDCs) are a diverse array of natural and manmade substances capable of interfering with hormonally mediated processes. Due to their epigenetic effects and effects on tissue growth and development, organisms at certain life stages are more vulnerable than others and effects may manifest a long time after exposure. As a robust system, cells can wisely choose and switch between different signaling programs according to their differentiation stages and external environments. Cancer cells can hijack this plasticity to develop drug resistance. For example, breast cancers that are initially responsive to endocrine therapy often develop resistance robustly. This process is dynamically controlled by interactions of genes, proteins, RNAs and environmental factors at multiple scales. Abstract:
Corticosteroid medicines have anti-inflammatory and immunosuppressant effects. Corticosteroids are prescribed for a wide range of conditions in children. The duration of treatment ranges from a few days (short course) to long term. They are usually given orally in the different dosage form of tablets, syrup or soluble tablets. There are a wide range of adverse drug reactions (ADRs) associated with corticosteroids. This thesis initially evaluates the incidence of each individual ADR in children using systematic reviews. Abstract:
Diabetic retinopathy (DR) is the leading cause of visual impairment in the working-age population in developed countries and thus is one of the major ocular health problems worldwide. Diabetes seems to have paradoxical effects on the microvasculature in different tissues. The concentration of glucose in the cellular microenvironment determines the cells ability to proliferate, as well as its permeability, and react to different cytokines. The main cause of visual loss in diabetic eye disease is caused by an increase in microvascular endothelial permeability which leads to vessel leakage and fl

Regular physical activity among children with Type 1 Diabetes Mellitus (T1DM) can help optimise long-term health outcomes. This thesis explores the experience of physical activity among children aged 9-11 years with T1DM and their parents, develops a physical activity intervention and evaluates its feasibility. Social cognitive theories have been drawn upon to develop our understanding and inform theoretically-driven behaviour change strategies. First, a systematic review with meta-analysis evaluates existing physical activity interventions for children with T1DM. The findings confirm the heal

Background: The practice of diabetes self-care plays an important role in glycaemic control. However, not all patients with insulin-treated diabetes engage in their self-care activities. Although there is evidence that self-care practices in patients with insulin-treated diabetes can be understood and predicted by health beliefs proposed by Health Belief Model (HBM), little is known about adult patients due to several methodological weaknesses of previous studies. Furthermore, knowledge is lacking about adults with insulin-treated diabetes in Malaysia. Aim: To examine whether health beliefs su

Approximately 3 million people throughout the UK suffer with Type 2 diabetes mellitus (T2DM), and are 32% more likely to die early. There remains a lack of evidence for the long-term effectiveness and safety of anti-diabetic drugs in preventing diabetes-related complications, making it unclear how to optimally manage diabetes. Work to date includes observational studies subject to bias, and randomised controlled trials (RCTs), which may not reflect the ‘real world’ situation. The aim of this thesis is to combine such findings via systematic reviews, meta-analyses and retrospective cohort studi

Introduction: The escalating public health problem represented by obesity has spurred multidisciplinary research into adipose tissue and importantly, the molecular biology of the adipocyte. The concept of adipose tissue as an endocrine organ, in addition to an energy storage compartment, is now pivotal in linking excess adiposity to disease states. Recent studies suggest that obesity related metabolic disorders are characterised by mild chronic inflammation as a result of adipocytokine production from fat tissue leading to dysregulation in the pro/anti-inflammatory systemic balance.
