Strains of C. acetobutylicum have been reported to exhibit some activity towards cellulosic and hemicellulosic substrates. The aim of this thesis was to establish a genebank of C. acetobutylicum P262 DNA in Escherichia coli and to isolate and characterise genes encoding enzymes which show activity towards hemicellulose and cellulose.

Les arabinoxylanes ont ete extraits du son de ble afin de pouvoir realiser une selection de micro-organismes hemicellulolytiques thermophiles. Une souche proche de bacillus flavothermus secretant des xylanases thermostables a ete ainsi isolee d'une source chaude du portugal. Aspergillus niger DSM 26641 was exposed to 60Co g-radiation to enhance the b-1,4-endoxylanase activity, restrict colony growth and improve robustness of pellets. The first promising mutant obtained after g-radiation of the fungal spores at 50-2000 Gy showed a restricted colony growth and an 82% enhancement in b-1,4-endoxylanase activity. Palm oil empty fruit bunches (EFB) is an abundant and cheap lignocellulose material in South East Asia. Its use as the sole medium for producing lignocellulose-hydrolyzing enzymes would increase its commercial value. A newly isolated Aspergillus niger DSM 26641 was investigated for its capability of producing hemicellulases in EFB hydrolysate obtained by treatment with pressurized hot water (1-20%, w/v) at 120-180◦C in a 1 L Parr reactor for 10-60 min. The optimal hydrolysate for the fungal growth and endoxylanase production was obtained when 10% (w/v) of empty fruit bunch was treated at 120◦C
