Abstract:
Total Synthesis of 4'-ester Resveratrol Analogs and 8, 9-amido Geldanamycin Analog and toward the Total Synthesis of (-)-Englerin A Yong Wang Department of Chemistry and Biochemistry, BYU Doctor of Philosophy The phytoalexin resveratrol and its 4'-ester analogs have been prepared with a decarbonylative Heck reaction. The deprotecting step has been modified and improved to increase yield and avoid chromatography. A set of resveratrol analogs and resveratrol have been tested with melanoma and pancreatic cell assays. The 8, 9-amido Geldanamycin analog has been synthesized with a convergent route, involving 28 simplified steps in its longest linear sequence. Synthetic methodologies, such as Andrus auxiliary controlled asymmetric anti-glycolate Aldol and selective p-Quinone formation, were employed. The total synthesis of Englerin A starts from (R)-carvone, passed through the modified Farvoskii ring-contraction and ring closing metathesis to get the ring skeleton. Other routes involving isopropyl group installation before closure of the seven-member ring failed. Although there are still problems to build the isopropyl moiety and the bridged ether, several reasonable alternative routes to address the problems have been designed.

Abstract:
4’ -ester analogues of resveratrol were synthesized using reaction conditions developed to produce mono-ester products in the presence of two other unprotected phenols. Basic conditions were employed to deprotonate the most acidic 4’ phenol followed by addition of anhydrides or acid chlorides to give the ester product. The reaction favored 4’-ester formation in polar aprotic solvents with DMSO being the optimal solvent. (—)-Englerin A is a guaiane-type sesquiterpene containing two ester side chains. Mimics of (—)-englerin A were proposed that retained the ester side chains while replacing the non-polar core with less complicated structures. These proposed mimic cores would maintain the three-dimensional positioning of the esters which are responsible for the anti-cancer activity of (—)-englerin A. Three mimics were synthesized using the bicyclic terpenes borneol and fenchol. Installation of the second ester on the terpene core was accomplished throught the development and optimization of a unique methylene oxidation using chromium trioxide in glacial acetic acid. These mimics were screened against two kidney cancer cell lines. The compounds were shown to have IC50 (inhibitory concentration for 50 % of cells) values above 30 µM.

The growing awareness of society to ecological problems is providing a considerable increase in demand for seeds and seedlings of native species. The Balfourodendron riedelianum (Engler) Engler is a native woody species of economic importance and ecological landscape, popularly known as ivory wood. The purpose of this study was to identify the ideal temperature for germination and determine the seed physiological potential of this species collected from different sources. The objective also their behavior with respect to storage and predict the storage period seeds and determine the constants of the longevity equation. Seeds collected in Marechal Cândido Rondon, Entre Rios do Oeste and Diamonte D'Oeste / PR were studied. For germination evaluation, two temperatures, 25 oC and 20-30 oC constant alternating with a photoperiod of 16-8 hours, and as substrate germitest paper were used. It was stored for 360 days, fruit in two different packaging (bag Kraft paper and glass) and in two different environments (controlled and uncontrolled). The experimental design used was completely randomized. The evaluated variables were: fruit length, percentage of germination, first count of germination, germination velocity index, moisture content, weight of thousand fruit and seedling length. Storage periods were 0, 90, 180, 270 and 360 days after fruit processing. For the development of the equations of longevity it was determined constants that made the equation through regression lines. The germination seeds of Balfourodendron riedelianum (Engler) Engler, was not affected by constant temperature of 25 oC compared with temperatures of 20-30 oC. The seedling length ivory wood was not influenced by temperatures of 25 oC constant and alternating temperature of 20-30 oC, from 90 days of storage. Among provenances, Entre Rios do Oeste stood out from the others to 0 and 360 days when he presented the highest rate of germination. The fruit stored in glass containers in uncontrolled environment and environmental conditions, did not show variations, related to the water content, thus demonstrating the best way to store ivory wood fruit. The mass of a thousand fruits stick ivory were 329.5 g, 585.5 g and 450.0 g for the provenances of Marechal Cândido Rondon, Entre Rios do Oeste and Diamonte D'Oeste, respectively. To seed ivory wood, the constants found to predict longevity were: Ke = 2.8667; Cw = 9.7697 E-013; Ch = 1.3435 E-012 and Cq = 1.1651 E-013. Using the storage conditions with average temperatures of 23 oC or 15 oC, and the ivory wood seeds showing moisture content of 12%, can be stored for up to 2 years. The values of the Pearson correlation between the actual percent germination calculated and the percentage of germination were 73% and 76% for the provenances Entre Rios do Oeste and Diamonte D'Oeste, and 77% and 74% for uncontrolled and controlled environment, respectively
A crescente conscientização da sociedade para os problemas ecológicos vem proporcionando um considerável aumento na demanda por sementes e mudas de espécies nativas. O Balfourodendron riedelianum (Engler) Engler é uma espécie lenhosa nativa, de importância econômica, paisagística e ecológica, popularmente conhecida como pau marfim. Objetivou-se, neste estudo, identificar a temperatura ideal para germinação e determinar o potencial fisiológico de sementes desta espécie, colhidas de diferentes procedências. Objetivou-se também verificar o comportamento das sementes com relação ao armazenamento, predizer o período de armazenamento destas sementes e determinar as constantes da equação de longevidade. Foram estudadas sementes, colhidas em Marechal Cândido Rondon, Entre Rios do Oeste e Diamante D Oeste/PR. Para avaliação da germinação, foram utilizadas duas temperaturas, 25 oC constante e 20-30 oC alternada, com fotoperíodo de 16-8 horas, e como substrato o papel tipo germitest. Armazenou-se, por 360 dias, os frutos em duas diferentes embalagens (saco de papel Kraft e vidro) e em dois diferentes ambientes (controlado e não controlado). O delineamento, utilizado, foi inteiramente casualizado Foram avaliadas as variáveis: comprimento de frutos, massa de mil frutos, porcentagem de germinação, primeira contagem de germinação, índice de velocidade de germinação, grau de umidade e comprimento de plântulas. Os períodos de armazenagem foram: 0, 90, 180, 270 e 360 dias após o beneficiamento dos frutos. Para a elaboração das equações de longevidade determinou-se as constantes que compõem a equação através de curvas de regressão. A germinação das sementes de Balfourodendron riedelianum (Engler) Engler, não sofreu efeito da temperatura constante de 25 oC, quando comparadas com temperaturas alternadas de 20-30 oC. O comprimento das plântulas de pau marfim, não foi influenciado pelas temperaturas de 25 oC constante e temperatura alternada de 20-30 oC, a partir de 90 dias de armazenamento. Entre as procedências, a Entre Rios do Oeste destacou-se das demais aos 0 e 360 dias, quando apresentou os maiores valores de velocidade de germinação. Os frutos, armazenados em embalagens de vidro e em local sem controle das condições ambientais, não apresentaram variações relacionadas ao teor de água, evidenciando, assim, a melhor maneira de armazenar frutos de pau marfim. Os valores da massa de mil frutos de pau marfim foram 329,5 g, 585,5 g e 450,0 g para as procedências Marechal Cândido Rondon, Entre Rios do Oeste e Diamante D Oeste, respectivamente. Para sementes de pau marfim, as constantes encontradas a fim de predizer a longevidade foram: Ke= 2,8667; Cw=9,7697E-013; Ch=1,3435E-012 e Cq=1,1651E-013. Para as condições de armazenamento com temperaturas médias de 23 oC ou de 15 oC, e as sementes de pau marfim apresentando grau de umidade médio de 12%, podem ser armazenadas pelo período de até 2 anos. Os valores da Correlação de Pearson entre a porcentagem de germinação real e a porcentagem de germinação calculada foram 73% e 76%, para as procedências Entre Rios do Oeste e Diamante D Oeste, e 77% e 74%, para ambiente não controlado e ambiente controlado, respectivamente

Durante los últimos años, en nuestro grupo de investigación se han desarrollado reacciones de activación de alquinos catalizadas por metales de transición. Uno de los objetivos de esta tesis ha sido el desarrollo de nuevos catalizadores quirales para realizar una ciclación [4+2] intramolecular de aril eninos, obteniéndose un sistema catalítico de Au(I) capaz de formar el producto ciclado con un exceso enantiomerico del 88%. Los otros capítulos de este manuscrito tratan de la aplicación de reacciones catalizadas por Au(I) en la síntesis de productos naturales: las (-)-englerinas A y B, compuestos naturales que presentan una potente actividad biológica contra el cáncer de riñón y la schisanwilsonena A, un terpeno dotado de una actividad antiviral contra la hepatitis B.
Over the last years, our research group reported several methodologies dealing with the activation of alkynes using gold as a catalyst. One of the objectives of this Thesis was the development of a new catalytic system for the intramolecular [4+2] cycloaddition of aryl enynes. After screening of different ligands and several optimizations, up to 88% ee could be obtained by using a bulky chiral gold phosphite. The two other chapters of this Thesis are dealing with the synthesis of natural products involving a gold-catalyzed transformation as a key step in the synthesis. The total synthesis of (-)-englerins A and B is reported and a study towards the synthesis of Schisanwilsonene A is also described.

Resumo: No presente trabalho são abordados alguns aspectos da biologia reprodutiva de G. jasminiflora Engler, com relação à sua polinização e dispersão. O trabalho de campo foi realizado na Reserva Florestal da Mata de Santa Genebra (Campinas, SP), entre fevereiro de 1989 e dezembro de 1990. Além da descrição morfológica e funcional da flor, foi registrado o comportamento dos principais visitantes diurnos e noturnos, obtidos dados sobre o sistema de reprodução, e observado o desenvolvimento do fruto até sua deiscência. Considerações também foram feitas sobre o modo de dispersão das sementes e seu processo de germinação. A floração de G. jasminiflora inicia-se em meados de fevereiro e estende-se até abril. As flores são tubulosas, zigomorfas, hermafroditas, de cor branca e exalam odor agradável. A antese é crepuscular (em torno das 18:00 horas), e o tempo de vida da flor é de aproximadamente 48 horas. Embora a maturação dos órgãos reprodutivos seja simultânea, nas primeiras 24 horas a flor se apresenta em fase masculina, com anteras expostas e estigma e estilete inclusos no tubo da corola. Nas 24 horas restantes, a flor se apresenta ocupa a posição anteriormente ocupada pelas anteras já caídas.O néctar é produzido em quantidades pequenas, porém sua produção é continua em ambas as fases...Observação: O resumo, na íntegra, poderá ser visualizado no texto completo da tese digital
Abstract: The present study is concerned with some aspects of reproductive biology of Galipea jasminiflora Engler, particularly pollination and dispersal. Field work was carried out in the Santa Genebra Forest Reserve (Campinas, São Paulo State), from february 1989 to december 1990. Morphological and funcional aspects of the flower were studied, together with the behaviour of the most important diurnal and nocturnal visitors. The reproductive systems, the development of fruits and their maturation were also studied, as well as seeds dispersal and germination. The flowering period of G. Jasminiflora starts in february and extends until april. The flowers are tubular, zygomorphic, hermaphrodite, whitish and exhale and agreeable odour. Anthesis is crespucular (about 06:00 p.m.) and the life time of flower is approximately 48 hours. In the first 24 hours the flower exhibits the male phase, with the anthers exposed and the stigma and style included in the corola tube. Durign the remaing 24 hours the flower enters the female phase, the anthers detach down the filaments and style extends and exposes the stigma. The nectar exsudes in small quantities, but its production is continuous during both phases. The flowers of G. Jasminiflora are visited by several diurnal and noctural insects. Abstract:
A Genève, Ferdinand de Saussure a professé, en élaborant progressivement son exposé, trois cours de linguistique générale répartis entre 1907 et 1911. Après en avoir examiné les notions fondamentales en suivant, entre autres cotextes, des notes de lecture du maître, l’auteur revient sur les sources et l’élaboration du premier cours. Louis Caille et Albert Riedlinger sont parmi les élèves qui ont bénéficié de ce cours.L’auteur complète la transcription de la sténographie Caille, et effectue la transcription d’un manuscrit intermédiaire mis au jour par Daniele Gambarara en aout 2011, et peut ainsi comparer avec les cahiers Riedlinger déjà connus. La triple transcription - détaillée selon les indices des articles de la remarquable édition de Rudolf Engler – permet de préciser les conditions d’élaboration du texte du Cours I.Outre l’examen des manuscrits, du manuscrit Caille et de ses annotations marginales, des recherches supplémentaires sont faites, tant sur les conditions historiques - généalogiques et administratives concernant les élèves immatriculés (au cours), inscrits (à l’examen) ou examinés (à l’épreuve) -, qu’aux archives de l’université concernant cette période pour les vérifications indispensables. L’ensemble de ces informations permet de privilégier la conjecture selon laquelle Louis Caille tenait le rôle de secrétaire de séance pour ce cours neuf dont l’introduction modifiait par ailleurs plusieurs programmes de cette université.Il en résulte que, de facto, la seule source à proprement parler du Cours I est cette sténographie Caille, contrairement à ce que suggérait jusqu’ici la connaissance des textes considérés nécessairement dans l’ordre historique de leurs acquisitions ; les éléments de la synthèse des trois cours – le Cours de Linguistique Général (CLG), publié par Ch. Bally et A. Séchehaye – sont distingués les uns des autres, et son élaboration est analysable en détail
In Geneva, Ferdinand de Saussure professed, by developing gradually his presentation, three lectures of general linguistics between 1907 and 1911. Having examined the fundamental notions by following, among others texts, reading notes of the teacher, the author focused on sources and elaboration of the first lecture. Louis Caille and Albert Riedlinger are among the students who benefited from this lecture.The author completes the transcription of the stenography by Caille, and makes the transcription of a new manuscript releaved by Daniele Gambarara in august 2011, and therefore can compare with the Riedlinger’s already known notetakings. The triple transcription - detailed according to the ranking of the articles of the remarkable Engler’s edition - allows to specify the conditions of elaboration of the text of Cours I.Besides the examination of manuscripts, of the Caille manuscript, and of its marginal notes, additional researches are made, as well on the historical conditions - genealogical and administrative concerning the registered and examined candidates -, that on the archives of the university concerning this period for the essential checks. All these informations allow to favor the hypothesis that Louis Caille acted as a transcriber for this new lecture whose introduction also modified several programs of this university.As a result, de facto, the only source strictly speaking of Cours I is this Caille stenography, contrary to what suggested until now the knowledge of texts considered inevitably in the historic order of their acquisitions; the elements of the synthesis of the three lectures – the Cours de Linguistique Générale (CLG), published by Ch. Bally and A. Séchehaye - are distinguished from each other, and its elaboration is analyzable accurately
