Abstract:
One of the greatest problems facing researchers in the sub field of Artificial Intelligence known as Intelligent Tutoring Systems (ITS) is the selection of a knowledge base designs that will facilitate the modification of the knowledge base. The Class-Entity-Relationship-Attribute (CERA), proposed by R. P. Brazile, holds certain promise as a more generic knowledge base design framework upon which can be built robust and efficient ITS. This study has a twofold purpose. The first is to demonstrate that a CERA knowledge base can be constructed for an ITS on a subset of the domain of Cretaceous paleontology and function as the "expert module" of the ITS. The second is to test the validity of the ideas that students guided through a lesson learn more factual knowledge, while those who explore the knowledge base that underlies the lesson through query at their own pace will be able to formulate their own integrative knowledge from the knowledge gained in their explorations and spend more time on the system. This study concludes that a CERA-based system can be constructed as an effective teaching tool. However, while an ITS - treatment provides for statistically significant gains in achievement test scores, the type of treatment seems not to matter as much as time spent on task. This would seem to indicate that a query-based system which allows the user to progress at their own pace would be a better type of system for the presentation of material due to the greater amount of on-line computer time exhibited by the users.

The demands of software engineering increases with everyday passed and every organization shows interest to work on daily routine work in the computerized system to improve efficiency and accuracy of data. Most of organization demands different kinds of computerized software solutions that developed in modern technologies. There are different software development technologies that is getting popular with the passage of time and provide high quality product to their user. It is not an easy to decide which technology choose to develop computerized system where most software falls. The primary purpose of thesis is to compare two different modern software development technologies by develop an application with same functional requirements. This thesis report will help to user for selection of software development technology. Cash and bank financial application is choose to develop using .NET and Java Enterprise Edition. Microsoft Visual Studio development environment is used for .NET development and NetBeans IDE is used for Java Enterprise Edition application. .NET framework contains many language that are interrelated with each other like C#, VB, J#, C++ and F#. Cash and bank application is develop in C# and Java programming languages and persistence storage of data is design in Microsoft SQL Server. Cash and bank application is develop with three tier architecture and layer approach. The syntax of .NET and Java Enterprise Edition is quite similar but procedure to establish connection, data retrieval and handling the data in different way. NET platform provide built in libraries, components and efficient user controls that helps for fast development in short period of time.

Cette thèse explique et présente notre démarche de la réalisation d’un système à base de règles de reconnaissance et de classification automatique des EN en arabe. C’est un travail qui implique deux disciplines : la linguistique et l’informatique. L’outil informatique et les règles la linguistiques s’accouplent pour donner naissance à une nouvelle discipline ; celle de « traitement automatique des langues », qui opère sur des niveaux différents (morphosyntaxique, syntaxique, sémantique, syntactico-sémantique etc.). Nous avons donc, dans ce qui nous concerne, mis en œuvre des informations et règles linguistiques nécessaires au service du logiciel informatique, qui doit être en mesure de les appliquer, pour extraire et classifier, par des annotations syntaxiques et/ou sémantiques, les différentes classes d’entités nommées.Ce travail de thèse s’inscrit donc dans un cadre général de traitement automatique des langues, mais plus particulièrement dans la continuité des travaux réalisés au niveau de l’analyse morphosyntaxique par la conception et la réalisation des bases des données lexicales SAMIA et ensuite DIINAR avec l’ensemble de résultats de recherches qui en découlent. C’est une tâche qui vise à l’enrichissement lexical par des entités nommées simples et complexes, et qui veut établir la transition de l’analyse morphosyntaxique vers l’analyse syntaxique, et syntatico-sémantique dans une visée plus générale de l’analyse du contenu textuel. Pour comprendre de quoi il s’agit, il nous était important de commencer par la définition de l’entité nommée. Et pour mener à bien notre démarche, nous avons distingué entre deux types principaux : pur nom propre et EN descriptive. Nous avons aussi établi une classification référentielle en se basant sur diverses classes et sous-classes qui constituent la référence de nos annotations sémantiques. Cependant, nous avons dû faire face à deux difficultés majeures : l’ambiguïté lexicale et les frontières des entités nommées complexes. Notre système adopte une approche à base de règles syntactico-sémantiques. Il est constitué, après le Niveau 0 d’analyse morphosyntaxique, de cinq niveaux de construction de patrons syntaxiques et syntactico-sémantiques basés sur les informations linguistique nécessaires (morphosyntaxiques, syntaxiques, sémantique, et syntactico-sémantique). Ce travail, après évaluation en utilisant deux corpus, a abouti à de très bons résultats en précision, en rappel et en F–mesure. Les résultats de notre système ont un apport intéressant dans différents application du traitement automatique des langues notamment les deux tâches de recherche et d’extraction d’informations. En effet, on les a concrètement exploités dans les deux applications (recherche et extraction d’informations). En plus de cette expérience unique, nous envisageons par la suite étendre notre système à l’extraction et la classification des phrases dans lesquelles, les entités classifiées, principalement les entités nommées et les verbes, jouent respectivement le rôle d’arguments et de prédicats. Un deuxième objectif consiste à l’enrichissement des différents types de ressources lexicales à l’instar des ontologies
This thesis explains and presents our approach of rule-based system of arabic named entity recognition and classification. This work involves two disciplines : linguistics and computer science. Computer tools and linguistic rules are merged to give birth to a new discipline : Natural Languge Processsing, which operates in different levels (morphosyntactic, syntactic, semantic, syntactico-semantic…). So, in our particular case, we have put the necessary linguistic information and rules to software sevice. This later should be able to apply and implement them in order to recognise and classify, by syntactic and semantic annotations, the different named entity classes.This work of thesis is incorporated within the general domain of natural language processing, but it particularly falls within the scope of the continuity of the accomplished work in terms of morphosyntactic analysis and the realisation of lexical data bases of SAMIA and then DIINAR as well as the accompanying scientific recearch. This task aimes at lexical enrichement with simple and complex named entities and at establishing the transition from the morphological analysis into syntactic and syntactico-semantic analysis. The ultimate objective is text analysis. To understand what it is about, it was important to start with named entity definition. To carry out this task, we distinguished between two main named entity types : pur proper name and descriptive named entities. We have also established a referential classification on the basis of different classes and sub-classes which constitue the reference for our semantic annotations. Nevertheless, we are confronted with two major difficulties : lexical ambiguity and the frontiers of complex named entities. Our system adoptes a syntactico-semantic rule-based approach. After Level 0 of morpho-syntactic analysis, the system is made up of five levels of syntactic and syntactico-semantic patterns based on tne necessary linguisic information (i.e. morphosyntactic, syntactic, semantic and syntactico-semantic information).This work has obtained very good results in termes of precision, recall and F-measure. The output of our system has an interesting contribution in different applications of the natural language processing especially in both tasks of information retrieval and information extraction. In fact, we have concretely exploited our system output in both applications (information retrieval and information extraction). In addition to this unique experience, we envisage in the future work to extend our system into the sentence extraction and classification, in which classified entities, mainly named entities and verbs, play respectively the role of arguments and predicates. The second objective consists in the enrichment of different types of lexical resources such as ontologies

Abstract:
The picornaviruses are a family of positive sense, single-stranded RNA viruses that initiate translation of their genome using an internal ribosome entry site (IRES), which is typically around 450 nt in length. Currently, three classes of picornavirus IRES element exist, based on structural similarities and biological properties. A novel picornavirus IRES element was identified within the 5' untranslated regions of porcine teschovirus-1 (PTV-1), porcine enterovirus-8 (PEV-8) and simian virus-2 (SV2). These related elements are considerably shorter than other picornavirus IRES elements and surprisingly formation of 48S pre-initiation complexes on the PTV-1 IRES is achieved in the absence of the translation initiation complex eIF4F. Indeed, toeprinting analysis with the PTV-1 IRES indicated that it is able to directly interact with the 40S ribosomal subunit. These properties are unusual for picornavirus IRES elements but are remarkably similar to those of the Flaviviridae IRES elements. Secondary structure models for the PTV-1, PEV-8 and SV2 IRES elements have been derived and key structural features verified through mutagenesis and functional assays. These studies have suggested that a fourth type of IRES element exists within the Picornaviridae, which is structurally and functionally related to the hepatitis C virus (HCV) and pestivirus IRES elements. Picornavirus IRES elements require accessory proteins for their activity. The availability of these proteins may provide insight into the tissue tropism of the viruses. To assess the binding of accessory proteins to the FMDV IRES in cells, a system was designed that could allow the capture of RNA transcripts containing the FMDV IRES and attached proteins from cells. This system made use of a specific protein-RNA interaction between the iron response protein and iron response element, which was placed at the 3' end of the FMDV IRES. Functional assays indicated that key features of this system were operational as expected, but currently no proteins have been identified as FMDV IRES binding proteins using this system.

Abstract:
This small scale, qualitative study examines the apprenticeship experiences of a group of working class young men from the London area during the years 1959-1989, to determine the impact of those experiences on the formation of their adult, male, self-identities. The research contributes to current debates and literature on apprenticeships, youth transitions, masculinity, social class and identity. It records as social history the recollections of ten ex-apprentices from the telecommunications and construction industries (five from each sector) and includes evidence of how they chose their apprenticeships. Data consists of participants’ memories collected as oral histories during semi-structured interviews, and from written answers to pre-interview questionnaires. The research was designed to ascertain the impact of their apprenticeships on the construction of the participants’ adult identities as defined by the concept of ‘standard adulthood’ (Parsons, 1971). This I describe as a series of interconnecting communities of practice (Lave and Wenger, 1991; Wenger, 1998), into which the participants entered as legitimate peripheral participants. Drawing on situated learning theory (Lave and Wenger, 1991), I contend that their post-war apprenticeships were part of a social and cultural institution that continued to serve as an integrative vehicle for the attainment of the ‘classic markers of adulthood’ (Blatterer, 2007a; 2007b) as a ‘life apprenticeship’, and through participation in diverse adult social worlds enabled them to become full participants in society (Goodwin, 2007). This thesis also examines the influence of social factors such as families, education and class on the participants’ school to work transitions. The research reveals a noticeable difference in the ways that these factors determined the two participant groups’ choice of apprenticeship. Drawing on social reproduction theory and the concept of cultural capital (Bourdieu & Passeron, 1990 ), I reveal the extent that family and social class played in deciding the participants’ future life courses.

L'etude de l'oxydation d'oxynitrures appartenant a des types structuraux tres divers (perovskite, fluorine, spinelle, cubique nacl) a permis de definir une nouvelle classe de composes oxyazotes : les phases intermediaires. Cette denomination a ete choisie car ces produits d'oxydation apparaissent avant la transformation totale de l'oxynitrure en oxyde au cours du traitement thermique sous oxygene. Nous avons montre que toutes ces phases etaient caracterisees par une importante retention d'azote au sein d'une maille oxyde hautement lacunaire et isostructurale de l'oxynitrure precurseur. La formulation de ces nouveaux composes est voisine de m#n#+o#n#/#2n#x ou m designe l'ensemble des cations impliques (m = bata, law, lnti, y#2w, al, ti, cr ou nb). La quantite d'azote retenue dans ces phases est comprise entre 3,5 et 11% atomique. Il apparait de la simple lecture de cette composition que l'azote piege doit etre dans un degre d'oxydation proche de zero. Un premier modele de liaison de l'azote dans les phases intermediaires, resulte d'un parallele avec des structures existantes de la litterature. Dans ce contexte, on remarque qu'il existe un etat de liaison de l'azote couramment rencontre, aussi bien en chimie du solide qu'en chimie organique : il implique des paires nn en interaction avec un ou plusieurs elements metalliques. Des etudes spectroscopiques, notamment par xps et raman, sont en accord avec cette hypothese. En effet, une phase intermediaire presente, en spectroscopie de photoelectrons, un pic xps n1s voisin de 403 ev compatible avec l'existence d'entites n#2 dans un environnement symetrique. La signature raman de ces phases est bien un pic proche de 2330 cm#-#1, valeur caracteristique de la bande de vibration de n#2.

Abstract:
In this thesis, non-binary people’s experience of gendered embodiment as well as inhabiting and navigating society is analysed. This is primarily done by using the theoretical framework of institutional lines, specifically applied through a trans studies perspective. Other important influences have been theories about how regulatory norms impact queer and trans subjectivities. Furthermore, special notice has been made of how social categorisations such as sexuality, class, and (dis)ability impact the non-binary subject’s experience of gender and of inhabiting society. This study is based on interview material produced through in-depth interviews with eleven non-binary Swedish people, as well as autoethnographical narratives produced by the author. The study shows that several different institutional lines impact the non-binary subject’s navigation through social space, as well as their experience of gendered embodiment. It was also clear that it was difficult for non-binary people to be recognised as coherent subjects, because of them breaking the regulatory norms that dictate intelligibility. Non-binary people are therefore often seen as the abject. This could be exacerbated if one not only deviated from the straight line of gender, but also from other lines and thus did not follow the expected trans line. It was clear that one’s experiences as a non-binary person could not fully be reduced to one’s position in relation to any specific line, be it gender, sexuality, class, or body normativity, but was rather a result of all those lines.

Abstract:
This thesis describes a program of research designed to investigate longitudinally the role of debt in a cohort of 1232 final-year New Zealand secondary school students, their tertiary entry decisions and their attitudes towards tertiary education and student debt. It follows some of these students into their first year out of school into tertiary education or otherwise. Two surveys were conducted that employed the Attitude to Debt Scale (Davies and Lea, 1995) to address students’ debt and savings behaviour and estimates, tertiary education entry decisions, and attitudes to tertiary education and term-time working. Debt attitudes are found to be more complex than previously proposed, and this has significant ramifications for debt attitude theory and research. Longitudinal comparisons suggest students’ views regarding debt necessity does not change but their attitude to avoiding does. Students become more or less avoidant of debt depending on their circumstances. However, debt attitude results still support many of the findings of earlier research such as debt acquisition preceding a more tolerant attitude change. Debt and tertiary education attitudes are not well predicted. Students report engaging in term-time working to limit their student loans, but engaging in term-time working results in lower grades in their studies. Those from the middle and higher socio-economic classes are more likely to be positive towards tertiary education, and thus entrants, compared with the lower socio-economic classes. However, the results do not suggest this is due to debt attitudes or fear of debt.

Abstract:
The aim of the thesis is to explore the social-class origins and destinations of CNAA graduates. The thesis begins with a discussion of social class, its meaning and conceptualization, and social-class schemas are devised for analysing the origins and destinations of graduates. Social class, however, is defined in a broad sense to include the dimensions of gender and ethnicity. Polytechnics and colleges are shown to have a higher proportion of working-class and black students than the universities. But despite their commitment to expanding educational opportunities public sector institutions remain socially exclusive in so far as they draw the bulk of their students from more middle-class backgrounds. Likewise, although public sector institutions appear to have expanded opportunities for women, female students are found to be concentrated in a limited number of courses. The career destinations of graduates are examined next. Significant differences were found relating to social-class or1g1ns with a tendency for men from manual backgrounds, women and black graduates to enter lower-status occupations. These differences appear only partly to arise from differences in career aspirations. It is suggested that black and women graduates may be subject to some discrimination. Significant differences are found in the destinations of graduates according to their courses of study, and once allowance is made for this, the existing relationship between the social-class origins and destinations of graduates becomes much less marked. An attempt is made to explain the relationship between the social-class or1g1ns and destinations of graduates and their courses of study using the models of contest and sponsored mobility devised by Turner. Using a four-fold categorization of school curriculums, it was shown that those graduates who had undertaken a 'utilitarian' school curriculum were constrained as regards their choice of course, whilst those who had undertaken an 'academic' curriculum and had been sponsored through secondary education enjoyed a greater choice of degree subject. Graduates from working-class or1g1ns were found to be more likely than their middle-class peers to have undertaken a 'utilitarian' curriculum.

Estudamos tópicos relacionados a zeros de séries de Dirichlet e de funções inteiras. Boa parte da tese é voltada à localização de zeros de séries de Dirichlet via critérios de densidade. Estabelecemos o critério de Nyman-Beurling para uma ampla classe de séries de Dirichlet e o critério de Báez-Duarte para L-funções de Dirichlet em semi-planos R(s)>1/2, para p ∈ (1,2], bem como para polinômios de Dirichlet em qualquer semi-plano R(s)>r. Um análogo de uma cota inferior de Burnol relativa ao critério de Báez-Duarte foi estabelecido para polinômios de Dirichlet. Uma das ferramentas principais na prova deste último resultado é a solução de um problema extremo natural para polinômios de Dirichlet inspirado no resultado de Báez-Duarte. Provamos que os sinais dos coeficientes de Maclaurin de uma vasta subclasse de funções inteiras da classe de Laguerre-Pólya possuem um comportamento regular.
We study topics related to zeros of Dirichlet series and entire functions. A large part of the thesis is devoted to the location of zeros of Dirichlet series via density criteria. We establish the Nyman-Beruling criterion for a wide class of Dirichlet series and the Báez-Duarte criterion for Dirichlet L-functions in the semi-plane R(s)>1/p, for p ∈ (1,2], as well as for zeros of Dirichlet polynomials in any semi-plane R(s)>r. An analog for the case of Dirichlet polynomials of a result of Burnol which is closely related to Báez-Duarte’s one is also established. A principal tool in the proof of the latter result is the solution of a natural extremal problem for Dirichlet polynomials inspired by Báez-Duarte’s result. We prove that the signs of the Maclaurin coefficients of a wide class of entire functions that belong to the Laguerre-Pólya class posses a regular behavior.

Abstract:
The aim of this thesis is to investigate how politics was done at the level of the parliamentary constituencies at the time of the treaty of Amiens 1802-1803. This is achieved through two case studies of the elections in Middlesex and Nottingham, which are investigated as social practices. This thesis argues that understandings of masculinity and national identity, as well as questions about the nature of the constitution and citizen rights were central to participants in the extraparliamentary political process. Collective emotions were also highly important in the process of mobilising political support, and this thesis emphasises that participation in these elections was a collective effort; men and women from all levels of society were significant political actors. Moreover, this thesis demonstrates the importance of competences such as knowledge about the organisation of crowds and political violence in the performance of the election.

This documentary takes the government-held course, which is originally aimed at adult education, for example. With the population of foreign spouses increasing, the government regulates that foreign spouses can have their Taiwanese nationality only by passing the language examination or attending language courses. Therefore, the entry-level language courses for foreign spouses are established around Taiwan. Teacher Hwang and volunteer Wang are two of those who devoted themselves to foreign spouses’ education. They take care of the students’ life and visit their places. From their experiences of teaching, we see an epitome of Taiwan’s adult education for foreign spouses.

The challenge of the Business Project was to develop an entry strategy for Claus Porto to the German Market. The analysis and recommendations are described in this paper. Finally, a brief academic discussion on firm internationalization and a scoring model to measure the attractiveness of international markets are presented.

This research adapts qualitative methods as the approach. It uses the cooperative halfway class established by Vocational Training Center East as the study field, and the dropouts and relevant people of accepting re-entry guidance are interviewed. In order to reach the research goal, it generalizes a conclusion. The results are as following. This study aims to investigate the causes of dropout junior high students, to deeply research the run a school idea of alternative education measures to cooperative halfway class at the present stage, and the effectiveness and dilemma of re-entering schools. This research adapts qualitative methods as the approach. It uses the cooperative halfway class established by Vocational Training Center East as the study field, and the dropouts and relevant people of accepting re-entry guidance are interviewed. In order to reach the research goal, it generalizes a conclusion. The results are as following. I. The complex causes of the dropout Dropout students with personal factor were the most, school factor was in the next place, and few were family and peer factors. Though the causes are divided into four categories, but the dropouts are usually the interactions among the four factors. II. The idea of running a school Combining with non-governmental resources, it offers various courses and learning ways to reduce the dropout rate; moreover, carry out the idea of “Adaptive learning with student's talent.” Through the diversify adaptive education offered by cooperative halfway class, students can recognize themselves, work out their potentialities, guide the correct values, guide them to complete the compulsory school, offer various multi-skills courses and the exploration of career development, learn the employment techniques, and give the opportunities of practical training. III. The effectiveness of the re-entry guidance Mainly expound and prove from three aspects: assessment methods, students’ progressive performance, teachers and counselors’ efforts to achieve education goals. The research comes to a conclusion that the re-entry guidance of cooperative halfway class is effective. IV. Faced with current difficulties The parent or guardian of a child provides only limited support to us; the students drop out again after they enter vocational schools, teachers and counselors lack for further training or workshops; subject supported teachers are not able to offer normalized teaching for some reason, and influence learning efficiency; teachers are not able to bring out students’ learning motivation; much of teaching hardware is outdated and equipment is inadequate. Solving strategies are holding parent-teacher meetings, increasing interaction in Parent-Teacher communication. For indifferent and negative parents, the research suggests intervene administrative force through government department; propel the parents to cooperate the cooperative halfway class. Hope the government budget subsidy center establish the senior department, make the dropouts’ re-entry unaccommodated regular school education be able to complete their senior high studies at Vocational Training Center East, acquire the professional skill, it would be beneficial for taking up an occupation and development. The counselors of cooperative halfway class can positively strive for the opportunity of further training or workshops, and expect education administration authority for highly attention and assistance. Government education department coordinate and invite subject teacher with education enthusiasm and innovational teaching methods, so that they can promote student’s motivation and willingness in learning. Proactively fight for funds from government and non-governmental group to improve the problem of old hardware facilities and inefficient teaching equipments.

Abstract:
Viral infection of cells often requires many host cell factors. Despite the identification of numerous viral receptor molecules relatively little is known about the roles of these molecules in viral entry, and intracellular targeting. Major histocompatibility complex (MHC) class I proteins are components of the Simian Virus 40 (SV40) cell surface receptor. This interaction between virus and host cell is of interest since MHC class I proteins have typically been studied for their importance in cellular immune responses. MHC class I proteins acquire antigenic peptides in the endoplasmic reticulum (ER). SV40 is unique in its ability to target the ER from the cell surface. Thus, the possibility exists that surface MHC class I proteins may recycle and target the ER. A major question is do MHC class I proteins internalize and target SV40 to the ER. The goal of this thesis was to better characterize SV40 entry, and determine if MHC class I proteins are directly involved in viral internalization. Basic studies were therefore conducted to determine the fate of MHC class I proteins on fibroblasts. SV40 entry into cells was found to occur rather slowly. The majority of preadsorbed virions remain at the cell surface for up to 2.5 hours. Furthermore, three hours were required for preadsorbed SV40 to internalize and escape antiserum neutralization. This suggests that SV40 infectious entry is relatively slow. Cytosol acidification greatly reduced clathrin-dependent endocytosis, but had no inhibitory effect on SV40 infectious entry. Therefore, SV40 infectious entry does not occur by receptor-mediated endocytosis. Treatment of cells with PMA, nystatin, or filipin prevents internalization via caveolae. All three treatments prevented SV40 infectious entry. Thus, caveolae may be the sites of SV40 penetration into cells. To evaluate the role of MHC class I proteins in SV40 endocytosis, cell surface proteins were labeled with $\sp{125}$I. Protease treatment of cells was used to distinguish proteins that have internalized from those remaining at the cell surface. Protease resistant $\sp{125}$I-MHC class I proteins were not detected in the absence, or presence of SV40. Furthermore, a $\sp{125}$I-MHC class I protein-specific monoclonal antibody did not internalize into cells. These results suggest that surface MNC class I proteins are not internalized into cells, and SV40 does not induce their internalization. Truncated $\sp{125}$I-MHC class I proteins were detected in the media. 1,10-phenanthroline prevented accumulation of $\sp{125}$I-MHC class I proteins in the media. Thus, metalloprotease activity is involved in shedding of surface MHC class I proteins from CV-1 cells. The loss of the B$\sb2$-microglobulin subunit from the MHC class I heavy chain was found to precede metalloprotease cleavage of the heavy chain. Collectively, these results suggest that the fate of MHC class I proteins on fibroblast is shedding into the media, and these molecules are probably not directly involved in SV40 entry.

Abstract:
Simian virus 40 (SV40) entry through caveolae represents a novel form of endocytosis. SV40 translocation to caveolae was mainly examined by cell fractionation and electron microscopy (EM) in CV-1 African green monkey kidney fibroblast cells. SV40 translocated to the caveolin-enriched low density Triton-insoluble complexes as identified by Western blotting. SV40 partitioning into this fraction was inhibited by nystatin. EM analysis indicated that SV40 translocation to caveolae was prevented by the tyrosine kinase inhibitor genistein (thin sections), but SV40 was not blocked from entering the Triton-insoluble low density fraction (negative staining). Northern analysis suggested that an SV40 signal that upregulates c-myc is transmitted through caveolae. The role of the major histocompatibility complex (MHC) class I proteins was not clear in SV40 translocation to caveolae. This is partly because of the high background level of class I molecules in the low density fraction isolated by sodium carbonate sucrose gradients (LDF-C). Class I proteins were enriched in the LDF-C relative to the whole cell extract or to the plasma membrane isolated by silica coating. The clustering of class I molecules was independent of any induction. Indirect immunofluorescence labeling of class I proteins and caveolin indicated that class I molecules colocalize with caveolae. MHC class I signaling and its physiological relevance were analyzed using a monoclonal antibody (mAb) W6/32. By immunoprecipitation and thin layer chromatography, ras was found to be activated by W6/32. W6/32 did not seem to increase the tyrosine-phosphorylation level in phospholipase C-$\gamma1.$ Engagement of class I molecules did not seem to induce apoptosis, but resulted in a distinct physiological response, the adhesion strengthening in CV-1 cells to Jurkat cells. The adhesion strengthening seemed to be sensitive to the protein kinase C inhibitor calphostin C, but enhanced by the tyrosine kinase inhibitor genistein. Furthermore, the mAb-induced adhesion strengthening was transient, antibody-concentration dependent, but independent of the Fc portion of the mAb. A serum factor was found to inhibit the basal level of adhesion in CV-1 cells to Jurkat cells. These findings represent one of the first investigations of MHC class I signaling in non-lymphoid cell lines.
