Abstract:
The paper analyses the development of the Bratislava–Schwechat Pipeline, which is supposed to connect the Slovak and Austrian oil pipeline networks. The research question asks why the project is being developed without taking into account arguments concerning environmental security even though rather significant environmental opposition to the pipeline has arisen. While the proponents of the project stress mainly its importance for the fostering of Slovak energy security, its opponents claim that the pipeline will endanger the current level of environmental security, since it presents a risk to Slovak fresh water reservoirs. In line with the theoretical approach, the paper argues that the conflict between these two types of security is positively skewed towards energy security, because its proponents are able to support their argumentation with concrete evidence. Moreover, advantages from improvements in energy security are visible in the short-term. On the other hand, the possible future negative environmental impacts of the pipeline are not so easy to assess (or quantify) and are of a long-term nature. The analysis also shows that due to the lack of activity of the Slovak Ministry of the Environment, a non-governmental organization Nie ropovodu (No to the pipeline) has become the main proponent of environmental security through the preparation phase of the project.

Abstract:

Abstract:
The identification of the hydrolysis and photodegradation products of flexible polyurethane foams (PUFs) with addition of biooriginated and biodegradable additive was the first topic of this dissertation work. Separation of polyurethane foam hydrolysis degradation products, designed for ecotoxicological tests, was managed by high-performance liquid chromatography coupled with mass spectrometry (HPLC/MS). The degradations product structure was elucidated by tandem mass spectrometry (MSn). PUF photodegradation products were obtained by exposure of materials by irradiation at 254 nm. Semi-volatile degradation products were isolated from the exposed polyurethane by n-hexane extraction; volatile compounds were collected by solid phase microextraction (SPME). Gas chromatography with mass spectrometry (GC/MS) and complete orthogonal tandem gas chromatography with mass spectrometry (GCxGC/TOF MS) was used for separation and identification of photodegradation products. The influence of the bio-filler on the character of degradation products and the possible effect of PUF degradation products on the environment was discussed at the end of this section. The determination of isoprostanes – markers of oxidative stress in tissues of beadlet anemone (Actinia equina) was the subject of the second topic. F2-isoprostanes were synthesized from the arachidonic acid. With thereby prepared isoprostanes the method of determination by liquid chromatography with tandem mass spectrometry (HPLC/MS/MS) was developed and optimized. The isoprostane isolation process from the Actinia equina tissues was optimized with solid phase extraction (SPE). The resulting methodology was used to quantify isoprostanes in tissues of anemones, which were exposed to both moderate and high temperature changes. The temperature changes were used to initiate the oxidative stress in organisms. In addition, concentration levels of unknown compounds were also monitored. These unknown compounds were extracted from tissues together with F2-isoprostanes and their identity is discussed in this dissertation work too. The possibility of using isoprostane levels in the Anthozoa tissues for the oxidative stress monitoring is discussed in the conclusion of this work.

Abstract:
The main objective of this work was to design a suitable composition of a protective coatings, made of several different layers of specific materials - with respect to residual stress, induced due to a mismatch in thermal expansion coefficients of each layer. Protective coating in this work means both the thermal and the environmental barrier. These coatings protect components against high temperatures and harsh environment. In this work, necessary theoretical background in the field of the thermal and environmental barrier coatings is introduced. There are mentioned some basic design approaches, commonly used materials and processing methods for the coating structure. The literature review gives an overview of modeling of such coated structures, in particular it is devoted to the thermal barrier coatings deposited by air plasma spray process. The next chapter closely describes classical laminate theory used for calculation of residual stresses in the coating. One of the assumptions of this theory is homogenous temperature field through the coating’s thickness. However, in this work was revealed a way to extend the classical lamination theory of such cases, in which the temperatures vary along the thickness of the coating. In the practical part, the analytical model was used for designing suitable properties of some coatings, which were consists of two, three and four layers. The calculations were performed both for constant temperature and for the temperature gradient. All results obtained from analytical approach were verified by numerical calculations.

Abstract:
Yttria-stabilized zirconia-based thermal barrier coating systems are the most widely used commercial coatings in the industry, with practical applications in aircraft engines and land-based power turbines. The purpose of thermal barriers is primarily to protect the substrate from high temperatures and also to increase its oxidation resistance. Currently, concerning the relatively frequent volcanic eruptions and increasing air traffic intensity in desert areas, increased attention is being paid to the development of new thermal and environmental coatings that will withstand the so-called CMAS attack and still successfully meeting the strictest requirements of the aerospace industry. Two newly developed experimental coatings consisting of three successive layers have been developed for this work. The upper two layers are thermal insulating ceramic coatings, where two different uppermost coatings were deposited. The first uppermost layer of the coating is a mixture of mullite and hexacelsian in a ratio of 70/30 wt. %. The second upper most type of coating consists of Al6Si2O13 + MgAl2O4 + BaCO3 in a ratio of 6:3:1 wt. %. The interlayer is made of the commercially utilized yttria-stabilized zirconia. The metallic CoNiCrAlY coating, which is directly deposited on the nickel-based superalloy MAR-M247, fulfils a compensatory function between the mechanical properties of the nickel superalloy and the ceramic coating. The thermal and environmental barrier system was deposited using air plasma spraying (APS) technology. The main objective of this work was to evaluate the effect of the newly developed thermal and environmental barrier coating, which has a high potential for the protection of component surfaces in an aggressive environment, on isothermical and thermomechanical fatigue behaviour of nickel-based superalloy MAR-M247. Low cycle fatigue tests were performed in strain control mode with constant strain amplitude on both uncoated and TEBC coated superalloy. Fatigue hardening/softening curves, cyclic stress-strain curves and fatigue life curves in the representation of total strain amplitude, plastic strain amplitude and stress amplitude on the number of cycles to failure were obtained. Microstructural analysis of MAR-M247 superalloy and a newly developed experimental coating was performed in a scanning electron microscope. The fatigue crack initiation sites were identified and the process of fatigue crack propagation was described. The dislocation arrangement after fatigue loading of MAR-M247 was investigated in a transmission electron microscope. The findings of isothermical and thermomechanical low cycle fatigue behaviour of uncoated and TEBC coated MAR-M247 superalloy and identification of damage mechanisms presented in this dissertation will improve the estimation of safe-life that is particularly relevant to aircraft engines components.

Abstract:
Environment, its condition and environmental factors influence human wellbeing both directly because it has direct impact on human's physical health and indirectly because it can cause stress and therefore have negative effect on wellbeing and health. Aim of this work is to find out which environmental factors and problems can cause stress and what factors, how and why, have impact on emergence of environmental stress and coping strategies. Important factors influencing environmental stress and especially coping strategies are stressor characteristics, perceived impact, coping resources, and especially perceived capability to cope with the stressor which is strongly influenced by information and experience and by behavior of others. An individual chooses passive or active coping strategy depending on these factors. Pro-environmental behavior could be one of these active coping strategies.

Abstract:
ROLE OF THE YXKO GENE OF BACILLUS SUBTILIS IN RESPONCE TO ENVIRONMENTAL STRESS Abstract Mutation of the yxkO gene, which encodes a putative ribokinase and belongs to the σB general stress response regulon, leads to reduced salt tolerance under potassium limitation in Bacillus subtilis. The biological function of the yxkO gene has not been determined yet, but it may be involved in the high affinity potassium uptake system, which has been described in Escherichia coli in contrast to Bacillus subtilis. Our goal was to describe another features of a mutant in the yxkO gene and to try to propose the role of this gene. Using the integration vector pMutin4, we prepared a Bacillus subtilis strain MP2 with a yxkO gene inactivation. The MP2 strain displays limited growth in a rich medium and it is a sensitive strain to tetracycline. Furthermore, this strain is unable to form endospores and the cells are longer, which indicates a septum formation defect. We accomplished a 2-D protein gel analysis to compare expression profiles of the MP2 strain and the 1A680 standard strain after salt and ethanol stress. The MP2 strain shows changes in productions of some energy metabolism enzymes and flagellin protein. We conclude that yxkO is a regulatory gene, whose product has a pleiotropic effect on many of cell functions.

Abstract:
Thorium is an element belonging to heavy metals, which is characterized by its radioactivity similarly to uranium and radon. Thorium is not commonly used in industry, but becouse of its radioactive properties it has a great potentional for future use in nuclear energetics. Thus, increasing release of Th into the soil, water and atmosphere can be expected in near future and through plant biomass it could become a part of food chains and webs and, thus, to represent a considerable health risks to humans. Studies devoted to research on thorium and its effects on plants has not been published much yet. The majority of these studies focus mainly only on plant abilities to accumulate thorium and/or monitoring distribution of thorium in plant body. But till now, a study is still missing, which would monitor the effect of thorium on the physiological characteristics of plants. Plants of Nicotiana glutinosa (L.) medium Thorium accumulatin were hydroponically cultivated Hoagland nutrient media differedin the presence of thorium, tartaric acid, putrescine and phosphates. In first part of the present study I monitored accumulation of thorium by tobacco plants under the influence of the above-mentioned modifications of media. In the second part, I studied the Th effect on the photosynthetic apparatus (contents...
