Abstract:
ABSTRACT The gram-positive bacterium Staphylococcus epidermidis is the most common cause of infections associated with catheters and other indwelling medical devices. S. epidermidis produces an extracellular slime that enables it to form adherent biofilms on plastic surfaces. We found that a biofilm-releasing enzyme produced by the gram-negative periodontal pathogen Actinobacillus actinomycetemcomitans rapidly and efficiently removed S. epidermidis biofilms from plastic surfaces. The enzyme worked by releasing extracellular slime from S. epidermidis cells. Precoating surfaces with the enzyme p

Cell therapies require the in vitro expansion of adherent cells such as mesenchymal stromal cells (hMSCs) in bioreactor systems or other culture environments, followed by cell harvest. As hMSCs are strictly adherent cells, cell harvest requires cell detachment. The use of hMSCs for cell therapy requires GMP production in accordance with the guidelines for advanced therapeutic medical products. Therefore, several GMP-conform available proteolytic enzymes were investigated for their ability to promote hMSC detachment. Abstract:
Vitreomacular traction occurs due to incomplete or anomalous posterior vitreous detachment. Over time, the vitreous pulls anteriorly and causes retinal distortion and eventually reduced vision. Traditionally, vitreomacular traction was treated with vitrectomy surgery. In the past few years, there is a paradigm shift towards pharmacologic vitreolysis, which involves the intravitreal injection of enzymatic and non-enzymatic agents that facilitate posterior vitreous detachment. Many agents have been investigated and trialled including plasmin, microplasmin (Ocriplasmin), hyaluronidase, nattokinas

Vitreoretinal relationships exert a significant influence on the course of retinal vein occlusions (RVO) and post-thrombotic retinopathy. A single intravitreal injection of Actilyse 50 mkg in the period from 1 till 21 days from the disease onset stimulates a posterior vitreous detachment in 30.4% patients with non-ischemic and 24.8% patients with the type of ischemic RVO. The process of transition from the partial posterior hyaloid detachment to the complete one, and complete posterior hyaloid detachment in patients who did not have it from the beginning, was accompanied by a visual acuity inc

Cell therapy usually accompanies cell detachment as an essential process in cell culture and cell collection for transplantation. However, conventional methods based on enzymatic cell detachment can cause cellular damage including cell death and senescence during the routine cell detaching step due to an inappropriate handing. The aim of the current study is to apply the pH-responsive degradation property of poly (amino ester) to the surface of a cell culture dish to provide a simple and easy alternative method for cell detachment that can substitute the conventional enzyme treatment. The thermal degradation related to stability in long-term storage of a carbon nanotube-based biosensor has been investigated. The effect of storage temperature on detachment and denaturation of glucose oxidase (GOx) biocatalyst has been proved. The carbon nanotubes (CNTs) coated with polyethyleneimine (PEI) as entrapping polymer to attract more GOx to form a durable and layered CNT/PEI/GOx structure is used for long-term storage to minimize GOx detachment from the structure and minimize the possibility of enzyme and protein denaturation. After 120 days, the glucose response of the CNT/PEI/GOx

Total bilateral limbal stem cell deficiency leading to loss of corneal clarity, potential vision loss, pain, photophobia, and keratoplasty failure cannot be treated by autologous limbal transplantation, and allogeneic limbal transplantation requires subsequent immunosuppressive treatment. Cultured autologous oral mucosal epithelial cells have been shown to be safe and effective alternatives. These cells can be transplanted on supports or without support after detachment from the culture dishes. Dispase, known for epidermal sheet detachment, is reported as not usable for oral mucosa.
