Chiral hydroxylated cyclopentane derivatives are important structural units in many biologically active compounds and are also important synthons for the asymmetric synthesis of natural products. Synthesis of these types of compounds in optically pure form found increased interest in pharmaceutical chemistry. For this purpose 5-acetoxy-3-methyl-2-methoxy-2-cyclopentene-1-one and 5-acetoxy-3-ethyl-2-methoxy-2-cyclopentene-1-one were acetoxylated using manganese (III) acetate at a&rsquo<br>positions. Enzyme catalyzed enantioselective hydrolysis of hydrolyzed acetoxy derivatives gives the corresponding hydroxylated diketones in optically pure form.

Depuis plusieurs années, une nouvelle génération de matériaux appelés “matériaux intelligents” et définis par leur capacité d’adaptation à leur environnement, est intensément développée. Des systèmes sensibles à différents stimuli tels que le pH, la lumière, ou encore une force mécanique, impliquée dans un grand nombre de processus naturels, comme l’adhésion et la prolifération cellulaire, ont été rapportés. Ce travail de thèse a ainsi été dédié au développement de matériaux mécano-sensibles. Plus précisément de matériaux transformant une contrainte mécanique en un signal chimique, en mimant le processus physique utilisé par la nature, à savoir des changements conformationnels de protéines. Nous avons donc cherché à atteindre ce but en greffant covalemment des protéines ou des enzymes sur un substrat élastomère. Etirer le substrat devant induire des modifications de structure des protéines, conduisant ainsi à des modulations de leurs propriétés<br>Since many years, a new generation of materials called « smart materials » and defined by their capacity to adapt to their environment is intensively developed. Systems sensitive to different stimuli such as pH, light or ionic strength have been reported. One of these stimuli can also be a mechanical force which is involved in many reactions in nature such as, cells adhesion and proliferation, tissues growing or even plants developments. The aim of my thesis was dedicated to the elaboration of mechano-responsive materials. More precisely, materials that transform a stretching constraint into a chemical signal by mimicking the physical processes used by nature, namely protein conformational changes. We planned to achieve this goal by covalently grafting proteins or enzymes onto a stretchable substrate or incorporating them into cross-linked polymer networks. Stretching these materials should induce protein conformational changes leading to modifications of their properties

L'extension de l'espace moléculaire chimique accessible à partir de la biomasse végétale par des méthodes douces et propres est un sujet d'actualité qui stimule la communauté scientifique afin de développer des produits biosourcés à faible impact environnemental et d'élargir le champ d'exploitation de la biomasse. La fonctionnalisation de la cellulose, le polysaccharide le plus abondant sur la planète, et/ou des cello-oligosaccharides telle que décrite dans cette thèse s'inscrit dans cette démarche. Notre objectif était de développer une méthode chimio-enzymatique impliquant l'action d'une laccase assistée par un médiateur pour oxyder des cello-oligosaccharides ou des fibres cellulosiques, suivie d'une amination réductrice pour greffer des composés aminés sur le matériau cellulosique. Dans ce but, nous avons d'abord démontré l'oxydation du cellobiose et du méthyl cellobiose en utilisant la laccase de Trametes versicolor et le TEMPO comme médiateur. Les conditions d'oxydation ont été optimisées avec le méthyl cellobiose et appliquées à un mélange de cello-oligosaccharides et au cellopentaose. En utilisant l'analyse LC/MS, nous avons montré qu'une large gamme de composés oxydés est obtenue et que la méthode est efficace pour produire des cello-oligosaccharides acides potentiellement intéressants pour les domaines biomédical et nutraceutique. Ensuite, nous avons montré que la réactivité du cellopentaose oxydé avec deux molécules aminées, la p-toluidine et la rhodamine 123 (un colorant aminé), permettait la liaison du composé aminé aux oligosaccharides. À l'aide des techniques LC/MS et MS/MS, nous avons mis en évidence la présence d'une liaison amine forte et covalente entre les colorants et le cellopentaose, élargissant ainsi l'espace chimique accessible par ce procédé hybride. Après avoir réalisé cette preuve de concept, nous avons tenté la teinture de fils de coton. Les fibres cellulosiques sont l'un des principaux matériaux textiles biosourcés et biodégradables. Cependant, le traitement chimique des textiles et notamment les méthodes chimiques utilisées pour fixer les colorants de manière covalente sont extrêmement polluants et nocifs pour la santé. Proposer des alternatives plus respectueuses de l'environnement est un défi mais d'un intérêt primordial pour une entreprise comme PILI, impliquée dans le projet de thèse, qui développe des colorants naturels en utilisant la biologie de synthèse. Ainsi, le potentiel du procédé hybride à deux étapes a été utilisé pour greffer avec succès la p-toluidine, la rhodamine 123 et le rouge acide 33 sur des fils de coton. La liaison covalente établie entre ces colorants et la fibre de coton a été prouvée pour la première fois. De plus, une bonne homogénéité et une bonne résistance au lavage ont été observées pour la teinture avec l'acid red 33, démontrant la robustesse et l'applicabilité de l'approche en situation réelle. Ces résultats originaux ont été brevetés. En testant d'autres colorants aminés, nous avons également montré que la solubilité, la réactivité et la structure du colorant aminé sont des paramètres importants à prendre en compte pour l'optimisation de la teinture, ce qui ouvre la voie à la synthèse à façon de nouveaux colorants aminés adaptés à ce procédé hybride prometteur<br>The extension of the chemical molecular space accessible from plant biomass by soft and clean methods is a timely topic that stimulates the scientific community in order to develop biobased products with low environmental impact and to widen the field of biomass exploitation. The functionalization of cellulose, the most abundant polysaccharide on the planet, and/or cello-oligosaccharides as described in this thesis is part of this approach. Our objective was to develop a chemo-enzymatic method involving the action of a mediator-assisted laccase to oxidize cello-oligosaccharides or cellulosic fibers, followed by reductive amination to graft amino compounds onto the cellulosic material. To this end, we first demonstrated the oxidation of cellobiose and methyl cellobiose using the laccase from Trametes versicolor and TEMPO as a mediator. Oxidation conditions were optimized with methyl cellobiose and applied to a cello-oligosaccharide mixture and cellopentaose. Using LC/MS analysis, we showed that a wide range of oxidized compounds is obtained and that the method is effective in producing acidic cello-oligosaccharides potentially of interest for the biomedical and nutraceutical fields. Then, we showed that the reactivity of oxidized cellopentaose with two aminated molecules, p-toluidine and rhodamine 123 (an aminated dye), allowed the binding of the amino compound to the oligosaccharides. Using LC/ MS and MS/MS techniques, we provided evidence for the presence of a strong, covalent amine bond between the dyes and cellopentaose, thus enlarging the chemical space accessible through this hybrid process. After completed this proof of concept, we attempted the dyeing of cotton threads. Cellulosic fibers are one of the main biosourced and biodegradable textile materials. However, chemical processing of textiles and especially the chemical methods used to covalently fix dyes are extremely polluting and harmful to health. Providing more eco-friendly alternatives is a challenge but of prime interest for a company like PILI, which was involved in the thesis project and is developing natural dyes using synthetic biology. Thus, the potential of the two-pot/two-step hybrid process was used to successfully graft p-Toluidine, rhodamine 123 and Acid Red 33 onto cotton thread. The covalent bond established between these dyes and the cotton fiber was proven for the first time. In addition, good homogeneity and wash-fastness were observed for acid Red 33 dyeing, demonstrating the robustness and applicability of the approach in real life. These original results have been patented. By testing other amino dyes, we also showed that the solubility, reactivity and structure of the aminated dye are important parameters to be addressed for dyeing optimization, which opens the way to the custom synthesis of new amino dyes suitable for this promising hybrid process

Les protéines extraites des végétaux sont des matériaux relativement peu coûteux, non toxiques, biocompatibles et biodégradables. Elles représentent une bonne alternative aux protéines d’origine animale et aux polymères dérivés du pétrole. Dans le cadre de cette étude, les protéines extraites de graines de soja et de tournesol ont été utilisées en tant que matériaux enrobants pour la microencapsulation de la matière active hydrophobe (α-tocophérol) ou hydrophile (acide ascorbique) par le procédé d’atomisation. Les protéines de soja sont largement utilisées dans les applications alimentaires et non-alimentaires, notamment en microencapsulation. Elles sont donc étudiées dans ce travail comme matériau enrobant de référence. Les protéines de tournesol n’ont quant à elles pas d’application industrielle concrète, si ce n’est sous la forme de tourteaux dans l’alimentation animale. C’est pourquoi il nous semble pertinent de trouver des nouvelles voies de valorisation pour ce coproduit d’origine agricole. Plusieurs modifications des protéines, telles que l’hydrolyse enzymatique, l’acylation, la réticulation enzymatique et la cationisation ont été étudiées dans le but d’améliorer les propriétés encapsulantes du matériau enrobant. Dans le contexte de la chimie verte, toutes les modifications ont été effectuées sans utilisation de solvants organiques ni de catalyseurs chimiques. L’influence des modifications chimiques et enzymatiques des protéines, et des paramètres du procédé (pression d’homogénéisation, ratio matériau enrobant/matière active et concentration en protéines) sur les différentes caractéristiques des préparations liquides et des microparticules (viscosité, taille des gouttelettes dans le cas des émulsions, morphologie et taille des microparticules), ainsi que sur les paramètres liés au procédé d’atomisation (rendement et efficacité de microencapsulation) a été particulièrement étudiée au cours de ce travail. Les résultats obtenus confirment que l’extrait protéique de tournesol est tout à fait pertinent comme matériau enrobant et permet d’obtenir des efficacités de microencapsulation significativement plus élevées par rapport à celles obtenues avec l’extrait protéique de soja<br>Proteins extracted from vegetables are relatively low-cost, non-toxic, biocompatible and biodegradable raw materials. They represent a good alternative to animal-based proteins and petroleum-extracted polymers. In this study, proteins derived from soybean and sunflower seeds were used as wall materials for microencapsulation of hydrophobic (-tocopherol) or hydrophilic (ascorbic acid) active material by spray-drying technique. Soybean proteins are widely used in food and non-food applications, especially in microencapsulation. They were studied in this work as wall material of reference. Sunflower proteins are not actually used in industrial application, but only in the form of oil-cake for animal feeding. That’s why new ways of valorization of this agricultural by-product should be investigated. Several proteins’ modifications such as enzymatic hydrolysis, acylation, cross-linking and cationization were studied in order to improve encapsulating properties of wall material. In the context of green chemistry, all the modifications and preparations were performed without use of organic solvents and chemical catalysts. The effect of protein chemical and enzymatic modifications, and process parameters (homogenization pressure, wall/core ratio and protein concentration) on different characteristics of liquid preparations and microparticles (viscosity, emulsion droplet size, microparticle size and morphology) and on parameters related to the spray-drying process (yield and efficiency of microencapsulation) was particularly investigated in this study. The obtained results confirmed that sunflower proteins are quite suitable as encapsulating agent and provide the microencapsulation efficiencies significantly higher compared to those obtained with soy proteins

Dans la nature, la réduction du dioxygène est catalysée par des enzymes de la famille des oxydoréductases. A l’heure actuelle, ces protéines spécifiques et efficaces sont envisagés comme biocatalyseurs au sein de biopile enzymatique. Dans ce contexte, l’optimisation de l’orientation et de la connexion d’oxydases multi-cuivre (MCOs) pour la réduction d’O2 sur des matrices de nanotubes carbone (CNTs) fonctionnalisées a été étudiée. Dans un premier temps, le transfert électronique direct de la laccase est optimisé par la fonctionnalisation non covalente de CNTs par divers dérivés hydrophobes. La dynamique moléculaire ainsi que la modélisation électrochimique ont permis la rationalisation des performances des différentes biocathodes développées. Dans une seconde approche, la modification spécifique par des groupements pyrène de la surface de laccases modifiées par mutagénèse a également été envisagée. La fonctionnalisation supramoléculaire de CNTs par des feuillets de graphène fonctionnalisés d’une part, et par des nanoparticules d’or d’autre part, a également permis de favoriser la connexion de laccases. La seconde partie présente l’élaboration d’autres types de biocathodes basées sur la connexion directe de bilirubines oxydases. Plusieurs stratégies de fonctionnalisation covalente et non covalente de CNTs ont été envisagées. Les différentes biocathodes élaborées par l’assemblage supramoléculaire de MCOs et de matériaux nanostructurés délivrent des densités de courant de réduction du dioxygène de plusieurs mA cm-2. Ces nouvelles bioélectrodes combinées à une bioanode qui catalyse l’oxydation du glucose ont permis le développement de biopiles enzymatiques glucose/O2 délivrant des densités maximales de puissances allant de 250 µW cm-2 à 750 µW cm-2 selon les conditions expérimentales. Enfin une bioanode à base d’une hydrogénase hyperthermophile a été développée et associée à une biocathode à base de bilirubine oxydase pour former un nouveau design de biopile H2/O2. Au sein de ce dispositif, la biocathode à diffusion de gaz réduit directement l’oxygène provenant de l’air, ce qui permet de s’affranchir de l’utilisation d’une membrane séparatrice tout en protégeant l’hydrogénase de sa désactivation en présence d’oxygène. Cette nouvelle biopile délivre une densité maximale de puissance de 750 µW cm-2<br>The reduction of oxygen is realized in nature by oxidoreductase enzymes. Currently, these highly specific and efficient proteins are considered as biocatalysts for the development of biofuel cells. In this context, optimizing the orientation and the connection of multicopper oxidase (MCOs) for the reduction of O2 on functionalized carbon nanotubes was studied. In the first part of this manuscript, direct electron transfer of laccase is assessed and optimized by the non-covalent functionalization of CNTs by various hydrophobic derivatives. Electrochemical modeling and molecular dynamics enabled the rationalization of the developed biocathodes efficiency. In a second approach, the specific modification by pyrene moieties of laccases surface modified by protein engineered has also been considered. Additionally, supramolecular functionalization of CNTs by modified graphene sheets and gold nanoparticles also helped to promote laccase connection. The second part presents the development of other types of biocathodes based on the direct connection of bilirubin oxidase. Several strategies of covalent and non-covalent CNTs functionalization have been considered. The different biocathodes developed by the supramolecular assembly of nanostructured materials and MCOs delivered current density of several mA cm-2 for oxygen reduction. These new bioelectrodes combined with a bioanode which catalyzes the glucose oxidation have enabled the development of glucose/O2 enzymatic biofuel cells; delivering maximum power densities from 250 µW cm-2 to 750 µW cm-2 depending on the experimental conditions. Finally a hyperthermophilic hydrogenase based bioanode was developed and associated with a bilirubin oxidase-based biocathode to form a new design of H2/O2 biofuel cell. Within this device, the gas diffusion biocathode directly reduces oxygen from the air, which eliminates the use of a separation membrane while protecting the hydrogenase from its deactivation in the presence oxygen. This new biofuel cell delivers a maximum power density of 750 µW cm-2

La tecnologia glicosintasa ha esdevingut una eina important per a la síntesi d’oligosacàrids, polisacàrids i glicoconjugats. Les glicosintases són glicosidases mutades desproveïdes d’activitat hidrolítica però capaces de catalitzar eficientment la formació d’enllaços glicosídics amb rendiments elevats en fer ús d’un donador glicosídic activat. A més a més, el donador activat i el seu producte de transglicosidació poden actuar com acceptors donant lloc a l’autocondensació del donador o a l’elongació del producte de transglicosidació, produint polisacàrids. En el present treball es pretén sintetitzar nous polisacàrids artificials funcionalitzats amb estructures definides mitjançant l’ús de la tecnologia glicosintasa. D’una banda, s’ha intentat augmentar la massa molecular dels polisacàrids fent ús d’un mòdul d’unió de carbohidrats (CBM). Aquest mòdul podria millorar la solubilitat dels nous polímers a mesura que es van sintetitzant durant la reacció de polimerització. L’efecte en el grau de polimerització ha estat estudiat tant pel mutant glicosintasa en presència de CBM com per la proteïna de fusió glicosintasa-CBM. D’altra banda, s’han sintetitzat polisacàrids artificials funcionalitzats a partir de donadors disacarídics activats on la funcionalització desitjada ha sigut prèviament introduïda a la posició C-6’. Aquests donadors funcionalitzats actuen com a substrat pel mutant glicosintasa que catalitzarà la reacció d’autocondensació. El grup azido va ser escollit com a un grup funcional adequat donada la seva versatilitat i mida, suficientment petit com per a ser acceptat per l’enzim. Els donadors fluorur de 6-azidolaminaribiosa i fluorur de 6-azidocel·lobiosa han estat sintetitzats i avaluats com a substrats per les reaccions glicosintasa amb el mutant E134S de la 1,3-1,4-β-glucanasa de Bacillus licheniformis i el mutant E197A de la cel·lulasa d’Humicola insolens, respectivament. S’han sintetitzat 6-azido i 6-amino-6-deoxicel·luloses artificials amb una seqüència de funcionalització alternada. De forma oposada a la modificació química de cel·luloses on la seqüència de substitució és intrínsecament aleatòria, la polimerització catalitzada per glicosintases de donadors glicosídics modificats apropiadament, permet accedir a noves cel·luloses funcionalitzades amb seqüències de substitució definides i regulars.<br>La tecnología glicosintasa se ha convertido en una herramienta importante para la síntesis de oligosacáridos, polisacáridos y glicoconjugados. Las glicosintasas son glicosidasas mutadas desprovistas de actividad hidroítica pero capaces de catalizar eficientemente la formación de enlaces glicosídicos con rendimientos elevados utilizando un dador glicosídico activado. Además, el dador activo y su producto de transglicosidación pueden actuar como aceptores dando lugar a la autocondensación del dador o a la elongación del producto de transglicosidación, produciendo polisacáridos. En el presente trabajo se pretenden sintetizar nuevos polisacáridos artificiales funcionalizados con estructuras definidas mediante el uso de la tecnología glicosintasa. Por un lado, se ha intentado aumentar la masa molecular de los polisacáridos utilizando un módulo de unión de carbohidratos (CBM). Este módulo podría mejorar la solubilidad de los nuevos polímeros a medida que se van sintetizando durante la reacción de polimerización. El efecto en el grado de polimerización ha sido estudiado tanto para el mutante glicosintasa en presencia de CBM como para la proteína de fusión glicosintasa-CBM. Por otro lado, se han sintetizado polisacáridos artificiales funcionalizados a partir de dadores disacarídicos activados donde la funcionalización deseada ha sido introducida previamente en la posición C-6’. Estos dadores funcionalizados actúan como sustrato para el mutante glicosintasa que catalizará la reacción de autocondensación. El grupo azido fue escogido por ser un grupo funcional adecuado dada su versatilidad y tamaño, suficientemente pequeño como para ser aceptado por el enzima. Los dadores fluoruro de 6-azidolaminaribiosa y fluoruro de 6-azidocelobiosa han sido sintetizados y evaluados como sustratos para las reacciones glicosintasa con el mutante E134S de la 1,3-1,4-β-glucanasa de Bacillus licheniformis y el mutante E197A de la celulasa de Humicola insolens, respectivamente. Se han sintetizado 6-azido y 6-amino-6-deoxicelulosas artificiales con una secuencia de funcionalización alternada. De forma opuesta a la modificación química de celulosas donde la secuencia de sustitución es intrínsicamente aleatoria, la polimerización catalizada por glicosintasas de dadores glicosídicos modificados apropiadamente, permite acceder a nuevas celulosas funcionalizadas con secuencias de sustitución definidas y regulares.<br>The glycosynthase technology has become a powerful enzymatic tool for the synthesis of oligosaccharides, polysaccharides and glycoconjugates. Glycosynthases are mutated glycosidases devoid of hydrolase activity but able to efficiently catalyze the formation of glycosidic bonds in high yields when using an activated glycosyl donor. In addition, the activated donor and its transglycosylation product, can both act as acceptors leading to the autocondensation of the donor or to the elongation of the transglycosylation product leading to polysaccharides. The present work aims to synthesize novel artificial functionalized polysaccharides with defined structures by the use of the glycosynthase technology. On the one hand, an increase in the molecular weight of the polysaccharides is attempted by the use of a carbohydrate binding module (CBM). This module might improve the solubility of the new polymers that are synthesized during the polymerization reaction. The effect on the polymerization degree has been studied for both the glycosynthase mutant in the presence of CBM and for the fusion protein glycosynthase-CBM. On the other hand, functionalized artificial polysaccharides have been synthesized from activated disaccharidyl donors where the desired functionalization has been introduced at position C-6’. These functionalized donors act as a substrate for the glycosynthase mutant that will catalyze the autocondensation reaction. The azido group was chosen as a suitable functional group given its versatility and its size, small enough to be accepted by the enzyme. Donors 6-azidolaminaribiosyl fluoride and 6-azidocellobiosyl fluoride have been synthesized and evaluated as substrates for the glycosynthase reactions by the E134S 1,3-1,4-β-glucanase from Bacillus licheniformis mutant and the E197A cellulase mutant from Humicola insolens, respectively. Artificial 6-azido- and 6-amino-6-deoxicelluloses with an alternating functionalization pattern have been produced. As opposed to chemically modified celluloses where the substitution pattern is intrinsically random, the glycosynthase-catalyzed polymerization of properly modified glycosyl donors gives access to novel functionalized celluloses with defined and regular substitution patterns.

Le remplacement des additifs controversés dans les matrices céréalières constitue en enjeu majeur pour répondre aux attentes des consommateurs. Les poudres à lever sont des ingrédients fonctionnels permettant l’obtention de produits biscuitiers alvéolés selon les modes de fabrication industriels. Leur incorporation dans la pâte à biscuits conditionne l’expansion des pâtons lors de l’étape de cuisson. Dans ces travaux, nous avons considéré deux pâtes céréalières avec des taux d’hydratation différents qui déterminent les voies d’incorporation de gaz dans chacune d’elles, avec l’objectif de totalement supprimer les poudres à lever. Dans une pâte à biscuits laminée faiblement hydratée, l’étude a considéré l’utilisation de levures boulangères pour substituer les poudres à lever. La configuration du réseau de gluten module les propriétés élastiques de la pâte et sa capacité à s’étirer pour permettre l’expansion des biscuits à la cuisson. Dans une pâte jaune foisonnée, l’incorporation d’air repose sur la formation d’une mousse stable en même temps que le dégagement gazeux induit par les poudres à lever. L’élimination des poudres à lever, dans cette matrice, a été possible grâce à l’utilisation de protéines végétales fonctionnalisées via différents traitements (physiques ou enzymatiques). Les propriétés interfaciales des protéines de la pâte déterminent leur capacité à stabiliser les bulles d’air qu’elle contient. Celles-ci ont été étudiées par tensiométrie et rhéologie interfaciale. Une approche par plan d’expériences a été implémentée pour optimiser les fonctionnalités et ainsi garantir une alvéolation régulière dans les biscuits<br>The replacement of controversial additives in cereal matrices represents a major challenge to meet consumers’ expectations. Leavening agents are functional ingredients that are required to obtain porous biscuit products according to industrial manufacturing methods. Their incorporation into biscuit dough determines the expansion of dough pieces during the baking stage. In this work, we considered two cereal doughs with different hydration levels that determine the gas incorporation pathways, aiming to completely suppress the need for leavening agents. In a low-hydration laminated biscuit dough, the study considered the use of baker's yeast as a substitute for leavening agents. The configuration of the gluten network conditions the dough elasticity and its ability to stretch to allow the biscuits to expand during baking. In a sponge drop (whipped) dough, air incorporation relies on the formation of a stable foam simultaneously with the gas release induced by the leavening agents. The removal of leavening agents from this matrix was enabled by using functionalized plant proteins through various treatments (physical or enzymatic). A design of experiments approach was implemented to optimize functionalities and thus, ensure the obtention of biscuits with a uniform crumb structure. During this process, the interfacial properties of the dough proteins determine their ability to stabilize the air bubbles in the matrix. These were studied using tensiometry and interfacial rheology

La racine forcée d’endive est un coproduit de la culture de l’endive, produit caractéristique du Nord de la France, de la Belgique et des Pays-Bas. Actuellement sous utilisé en méthanisation ou en alimentation animale, ce coproduit contient pourtant des molécules d’intérêts à haute valeur ajoutée : les acides caféoylquiniques. Ces molécules possèdent des activités antioxydantes, anti-inflammatoires et permettent de limiter les maladies du désordre métabolique. Ce travail de thèse vise à intensifier le prétraitement, l’extraction, la purification et la fonctionnalisation des acides caféoylquiniques à partir des racines forcées pour développer de nouvelles molécules bioactives biosourcées potentiellement intéressantes pour le secteur cosmétique et nutraceutique. Une dernière partie de la thèse porte sur l’étude technico-économique du procédé pour estimer sa rentabilité économique en fonction du secteur d’application visé. La première partie porte sur l’effet des prétraitements conventionnels (découpe et séchage) ainsi que l’effet d’un prétraitement électrique par champs électriques pulsés sur les teneurs en acides caféoylquiniques dans la biomasse. L’effet de l’ajout d’une solution antioxydante lors de l’extraction est également étudié. Dans un deuxième temps, une optimisation de l’extraction est réalisée à partir de biomasse sèche et fraiche. L’influence de facteurs tels que la température, le ratio solide/liquide, ainsi que la nature du solvant a été étudiée. De plus, des cinétiques d’extraction ont été tracées pour étudier les paramètres cinétiques à l’aide d’un modèle empirique. La pureté de l’extrait obtenu étant faible, des étapes de purification sont donc nécessaires. Par la suite, les travaux se sont portés sur la purification de l’extrait brut à l’aide de résines macroporeuses ainsi que par extraction liquide/liquide. Pour la purification par résine, un screening de résines est réalisé suivi d’une optimisation des conditions opératoires de purification avec la résine choisie. Des modélisations des phénomènes d’adsorption sont réalisées pour déterminer les étapes limitantes ainsi que la capacité maximale d’adsorption. Pour l’extraction liquide/liquide, un screening de solvants verts est effectué à partir d’un milieu aqueux et hydro-éthanolique puis une optimisation des conditions opératoires avec le meilleur solvant est réalisée. La pénultième partie de la thèse cherche à fonctionnaliser par estérification les acides caféoylquiniques à partir d’une solution modèle puis d’un extrait réel. Les conditions d’estérification sont optimisées pour augmenter la vitesse de réaction ainsi que le taux de conversion. Des esters avec différentes longueurs de chaine sont obtenus et l’activité antioxydante ainsi que les propriétés anti-UV sont étudiées. La fonctionnalisation est par la suite effectuée sur un extrait réel. Une étude technico-économique conclut la thèse permettant d’ouvrir sur des perspectives quant aux conditions nécessaires à l’industrialisation du procédé de valorisation des racines forcées d’endive<br>Forced chicory root is a by-product of Belgian endive culture, a typical crop of northern France, Belgium and the Netherlands. Currently under-utilized in methanation or animal feed, this by-product contains molecules of interest: caffeoylquinic acids. These molecules have antioxidant and anti-inflammatory properties, and a potential for reducing metabolic disorders. This thesis aims to intensify the pre-treatment, extraction, purification and functionalization of caffeoylquinic acids from forced chicory roots to develop new bioactive biosourced molecules of potential interest to the cosmetics and nutraceutical sectors. The final part of the thesis deals with a technico-economical study of the process to estimate its economic profitability in relation to the targeted application sector. The first part focuses on the effect of conventional pretreatments (cutting and drying) and the effect of pulsed electric field pretreatment on caffeoylquinic acid content in biomass. The effect of adding an antioxidant solution during extraction is also investigated. Secondly, extraction optimization is carried out using dry or fresh biomass. The influence of factors such as temperature, solid/liquid ratio and solvent type were studied. In addition, extraction kinetics were performed to study kinetic parameters using empirical models. As the purity of the extract obtained is low, purification steps are needed. The thesis then focused on purifying the crude extract obtained using macroporous resins and liquid/liquid extraction. For resin purification, resin screening was carried out, followed by optimization of the purification operating conditions with the chosen resin. Models of adsorption phenomena are carried out to identify the limiting stages and the maximum adsorption capacity. For liquid/liquid extraction, green solvent screening is carried out on aqueous and hydro-ethanolic media, followed by optimization of operating conditions with the best solvent. The penultimate part of the thesis seeks to functionalize caffeoylquinic acids by esterification, starting with a model solution and then a real extract. Esterification conditions are optimized to increase both reaction speed and conversion rate. Esters with different chain lengths were obtained, and biological activities such as antioxidant activity and anti-UV properties were studied. Functionalization is also performed with real extract. A technico-economic study concludes the thesis, opening up prospects for the industrialization of the forced chicory roots valorization process

Silk is a natural protein fibre spun by spiders, some insects and silkworms. Spider silk is of interest because of its mechanical strength, elasticity and biocompatibility, in fact, spider silk is considered to be the strongest natural biopolymer known. Recombinant production of spider silk proteins has begun to meet the needs of the growing biotechnological interests, and recombinant production in various hosts has been a widely used approach, with varying results. In search of mimicking the silk proteins, 4RepCT has been engineered with only 4 gly/ala repeats followed by a non-repetitive C-terminal. This protein has been successfully produced in a soluble form using Escherichia coli, yet forming macroscopic silk-like fibres. Studies on 4RepCT fibres have shown biocompatibility and a limited immunological response when subcutaneously implanted in rats. Functionalization is a process where additional peptide motifs, protein domains or organic compounds are coupled to 4RepCT to add new properties for use in medical and biotechnology disciplines. There is, however, currently, a need for new coupling methods, in addition to, for example, manipulation on the genetic level. Therefore, this study was aimed to investigate a new method for functionalization of 4RepCT silk using two model proteins (domain Z and fibroblast growth factor-2) with the help of Sortase A mediated covalent coupling. The results have shown that a Sortase A enzyme with 3 mutations, coupling at ambient temperature and a reaction time between 0.5 and 1.5 h are suitable conditions for efficient coupling of the model proteins to 4RepCT silk coatings. Coupling of the Z domain to silk using the Coupling during coating (CDC) method and FGF2 to silk using the Coupling after coating (CAC) method showed best binding of their target molecules (IgG and FGF2 receptor, respectively). Analysis by Surface Plasmon Resonance was efficient to distinguish these differences in binding rates.<br>A seda é uma fibra proteica de origem natural produzida por aranhas e alguns insectos, como, por exemplo, o bicho - da - seda. A seda produzida pelas aranhas reveste-se de particular interesse devido à sua resistência mecânica, elasticidade e biocompatibilidade. De facto, a seda produzida pelas aranhas é considerada como sendo o polímero mais resistente que existe. A produção recombinante de proteínas da seda de aranha iniciou-se como objectivo de satisfazer as necessidades crescentes da industria biotecnológica, e tem sido produzida em vários hospedeiros com resultados variáveis. Na demanda de imitar as proteínas da seda, 4RepCT foi modificada de forma a conter apenas 4 repetições gly/ala, seguidas por um C terminal não repetitivo. Esta proteína recombinante foi produzida com sucesso, na forma solúvel, em Escherichia coli, gerando, porém, fibras macroscópicas semelhantes à seda. Estudos em fibras 4 RepCT mostraram a existência de biocompatibilidade e uma resposta imunológica limitada quando implantadas subcutaneamente em ratos. A funcionalização é um processo em que péptidos adicionais, proteínas ou compostos orgânicos são acoplados á 4RepCT para adicionar novas propriedades a estes polímeros para que possam ser utilizados nas áreas médicas e biotecnológicas. No entanto, actualmente existe a necessidade de desenvolver novos métodos de acoplamento associados, por exemplo, a técnicas de manipulação genética. Assim, este estudo visou a investigação de um novo método de funcionalização da seda 4RepCT , utilizando como modelo duas proteínas (domínio Z e factor 2 de crescimento de fibroblastos) com o auxílio do acoplamento covalente mediado pela sortase A. Os resultados obtidos demostraram que a temperatura ambiente um tempo de reacção entre 0,5 e 1.5 h são as condições adequadas para o acoplamento eficiente da sortase A com 3 mutações . Tanto o acoplamento do domínio Z á proteína da seda durante o método de revestimento (CDC) como o acoplamento doFGF2 á proteína da seda após o método de revestimento (CAC) apresentou melhor ligação ás suas moléculas alvo (IgG e receptor de FGF2, respectivamente).A análise de ressonância de plasma de superfície, foi eficiente para distinguir essas diferenças nas taxas de ligação.
