Abstract:

The Hengduan Mountains are one of the most species-rich mountainous areas in the world. The origin and evolution of such a remarkable biodiversity are likely to be associated with geological or climatic dynamics, as well as taxon-specific biotic processes (e.g., hybridization, polyploidization, etc.). Here, we investigate the mechanisms fostering the diversification of the endemic Himalopsyche martynovi complex, a poorly known group of aquatic insects. We used multiple allelic datasets generated from 691 AHE loci to reconstruct species and RaxML phylogenetic trees. We selected the most reliable phylogenetic tree to perform network and gene flow analyses. The phylogenetic reconstructions and network analysis identified three clades, including H. epikur, H. martynovi sensu stricto and H. cf. martynovi. Himalopsyche martynovi sensu stricto and H. cf. martynovi present an intermediate morphology between H. epikur and H. viteceki, the closest known relative to the H. martynovi-complex. The gene flow analysis revealed extensive gene flow among these lineages. Our results suggest that H. viteceki and H. epikur are likely to have contributed to the evolution of H. martynovi sensu stricto and H. cf. martynovi via gene flow, and thus, our study provides insights in the diversification process of a lesser-known ecological group, and hints at the potential role of gene flow in the emergence of biological novelty in the Hengduan Mountains.

Abstract:

Weisheit wird gesucht, weil in einer zunehmend komplexer und intransparenter werdenden Welt Vertrauenswürdigkeit gesucht wird. Vertrauenswürdigkeit ist ihrerseits abhängig von der der jeweiligen Person oder Institution zugeschriebenen Unabhängigkeit. Der Beitrag argumentiert, dass sich der Begriff der Weisheit einer expliziten Definition entzieht und als Clusterbegriff mit teils kognitiven, teils psychologischen, teils moralischen Elementen gefasst werden sollte. Durch die letzteren Elemente unterscheidet sich Weisheit wesentlich von Lebensweisheit, wie sie Epikur und in seiner Nachfolge Schopenhauer verstehen. Sie sind für den „weisen Richter” charakteristischer als für den „weisen Ratgeber”. Die Frage, wie weit Schopenhauer, einer der meistgelesenen modernen Weisheitslehrer, selbst als weise gelten kann, wird am Ende skeptisch beantwortet: Schopenhauer erfüllte viele, aber nicht alle Bedingungen, die an Weisheit zu stellen sind.

A tanulmány Arany János verses epikájának és lírájának kortárs recepcióját elemzi. Röviden bemutatja az életmű értékelésének hullámzásait, a verses epikus és a lírikus korszakok különféle értelmezési irányait, majd a kortárs magyar költészet Arany Jánoshoz való viszonyulását vizsgálja Nagy László, Juhász Ferenc, Orbán Ottó, Ágh István, Kányádi Sándor, Térey János és Lövétei Lázár László munkásságának középpontba állításával. A tanulmány alaptézise szerint fordulat következett be a hagyománytörténésben: a XX. században Arany János lírája látszott korszerűbbnek, mára viszont fölerősödött az epikus struktúrák recepciója. A szerző úgy véli, hogy a magyar költészet történetében akkor erősödnek föl az epikus struktúrák, amikor a költők az individualitás és a kollektivitás etikai problémáit nem ontikusan elválasztva, hanem szinoptikusan összenézve kezelik, s ennek révén tesznek kísérletet az olyan dichotómiák feloldására, mint klasszicizálás és romantika, látszat és valóság, egyéniesség és közösségiség, személyesség és tárgyiasság, affirmáció és alakváltoztatás, valóságreferencia és nyelvi játékosság, sajátszerűség és idegenség, eredetiség és mintakövetés, haza és haladás.

Lucanus epikus technikájának jellemző jegye az istenapparátus hiánya. A jelenség magyarázatára az utóbbi bő évszázadban számos magyarázat született. Egy részük nem annyira az istenapparátus hiányáról beszél, mint inkább arról, hogy a költő az epikus narratíva e transzcendens aspektusát a hagyományos istenek helyett a teljesen személytelen fatum mal s a hol filozófikusan (fortuna), hol mintegy istenségként felfogott Fortuná val helyettesíti. A modern elméletek már Lucanus költői látásmódjának újszerűségével magyarázzák a jelenséget. Az istenapparátus hiánya azonban csak egyike azon tüneteknek, melyeknek mindegyike jól magyarázható a szónoki munkamódszerrel, a retorikai narratio szabályainak követésével.

Abstract:

Abstract Epicurus is well known as the letter writer par excellence among ancient philosophers. This is shown by examples from Alciphron, Diogenes Laertios, Seneca, and Plutarch. Additionally, the long list of partially preserved letters in the collections of fragments by Usener and Arrighetti is analyzed. These quotes demonstrate the use of letter collections originating with Epicurus and his first students. A new valuable testimony is provided by the publication in 2011 of POxy 5077. Its three fragments are clearly taken from a collection of letters of Epicurus. We find his name in a typical prescript: “Epicurus (to NN), greetings”. Leonteus and Mithres of the founding generation are mentioned. The copying of letters and the exchange of documents within the Epicurean community are fully on display. The little known shipwreck suffered by Epicurus is alluded to. All this is not without interest for students of early Christian letter collections.

A két részlet ilyen szempontú elemzése a varietas esztétikai alapján szerkesztett Metamor-phoses szerkezeti egyensúlyára éppúgy lehetséges magyarázattal szolgál, mint ahogy közelebb visz a teljes mű értelmezéséhez is. A két részlet párhuzamai és újragondolása újabb szempontokat vet föl az Orpheus-történet vergiliusi és ovidiusi feldolgozásának kapcsolatáról is, a parodisztikus újramesélés helyett utóbbinál is a költő dalnok szerepének fontosságára és drámaiságára hívva föl a figyelmet.

A cikk azt a problémát vizsgálja konkrét szöveghely értelmezésével, hogyan teremthet lehetőséget gazdagító új olvasatok megfogalmazására régebbi irodalmi, esztétikai előfeltevéseink felülvizsgálata. Az Aeneis Nisus és Euryalus-epizódjában a szerző egy homérosi ihletésű katonai epizódban az epikus és heroikus világtól (a cikk szóhasználatában epikus kódtól) idegen elemekkel bővíti ki az elbeszélést. A szerelmi elégia nyelve és a görög fiúszerelem fogalma azonban feszültséget hoz a homérosi katonatörténetbe. Ennek eredményeképpen a részletet záró költői megszólalás és maga a történet között az interpretáció szempontjából termékeny feszültség alakul ki. A cikk amellett érvel, hogy ezt a feszültséget nem szabad eltüntetnie az inter pretációnak, helyette az egységesség követelményét érdemes újragondolni.

Abstract:

Aim: Our previous study demonstrated that the cystatin C-based chronic kidney disease (CKD)-EPI equation and combined by serum creatinine (CKD-EPIscr-cys) had better capability to accurately evaluate glomerular filtration rate in the CKD participants. Considering that the accuracy of estimated glomerular filtration rate (eGFR) remains less ideally, it is essential to modify the equation by including the Chinese eGFR racial factor in order to improve its performance. Methods: Two prospective cohorts were enrolled in 2 medical centers. New equations were developed in 529 participants and validated in 313 participants. Reference glomerular filtration rate (rGFR) was taken by 99mTc-DTPA renal dynamic imaging method (Gates method). The primary outcomes of this study were bias, precision (interquartile range of difference [IQR]), and accuracy (the proportion of eGFR within 30% of rGFR [P30] and root mean square error [RMSE]) of eGFR versus rGFR. Results: In a development data set, Chinese coefficients for CKD-EPIscr (C-CKD-EPIscr), CKD-EPIcys (C-CKD-EPIcys), and CKD-EPIscr-cys (C-CKD-EPIscr-cys) were 0.871, 0.879, and 0.891, respectively. In a validation data set, C-CKD-EPIcys was the most accurate with highest P30 value (62.3%), relative lowest IQR (15.45), and RMSE (0.80) among 6 equations, though the bias of C-CKD-EPIcys was not better than CKD-EPIcys. C-CKD-EPIscr and C-CKD-EPIscr-cys equations were improved in bias (p < 0.001), precision, and accuracy (p = 0.004 and <0.001 for P30) compared with CKD-EPIscr and CKD-EPIscr-cys. Conclusion: C-CKD-EPIcys was the most accurate with the highest P30 value, relative lowest IQR, and RMSE among 6 equations. C-CKD-EPIscr and C-CKD-EPIscr-cys equations were improved in bias, precision, and accuracy. Other external validation of these equations is needed.

Abstract:

ABSTRACT Stereoselective disposition of sulbenicillin (SBPC) epimers in healthy human volunteers was studied in order to clarify the differences in pharmacokinetic behavior between the epimers. Stereospecific high-performance liquid chromatography was used for the determination of SBPC epimers. Plasma protein binding was measured in vitro with an ultrafiltration method. The binding was stereoselective, with the unbound fraction (fu ) of theR-epimer being approximately 1.3-fold greater than that of the S-epimer. SBPC was administered intravenously to human volunteers, and concentrations of SBPC in plasma and urinary excretion rates were measured. Renal clearance (CLR) for the unbound drug (approximately 400 ml/min) was greater than the glomerular filtration rate (GFR) (approximately 109 ml/min) for both epimers, suggesting that both epimers are secreted at the renal tubules. Renal tubular secretion appeared to be greater for the S-epimer. When probenecid was coadministered, the CLR values of both epimers were significantly reduced and were approximately equal to the GFR values. CLR was greater for the S-epimer (37.5 and 49.8 ml/min for R-SBPC and S-SBPC, respectively), which was simply due to the greaterfu of the S-epimer in plasma. In contrast, total body clearance was greater for the R-epimer (67.8 and 56.3 ml/min for R-SBPC and S-SBPC, respectively) because of the stereoselective degradation of theR-epimer in plasma. It was revealed that stereoselective degradation in the body had significant influence on the disposition of SBPC epimers.

Abstract:

Abstract The cryoscopic measurements show that title compounds are strongly associated in CHCl3 solutions. The association of the 20 R epimer is distinctly less pronounced than that of the 20 S epipmer. Self-association of the 20 S epimer leads to the formation of very large complexes. The 20 R epimer forms associates via water molecules. The dissimilarity may be explained in terms of different accessibility of the lactam carbonyl groups in the two epimers for the association. It is proposed that the association process is controlled by the configuration at the carbon atom C(20) and conformation around the C(20)-C(22) bond. Populations of side chain conformations of both epimers were determined by means of proton nuclear magnetic resonance. It was found for the 20 R epimer that the t and the -g rotamers are almost equally populated, and the rotamer +g is excluded. For the 20 S epimer the +g rotamer predominates over the t one, and the -g rotamer is excluded. The NMR data are fully consistent with the results of the molecular modelling studies.

Abstract:

Recently, hepatic immaturity was cited as a possible reason for high levels of the C-3 epimer of 25-hydroxyvitamin (25(OH)D) in premature infants: however what role, if any, the liver plays in controlling epimer concentrations is unknown. This study assesses 3-epi-25-hydroxyvitamin D (3-epi-25(OH)D) levels during the course of cholecalciferol supplementation in adults with chronic liver diseases (CLD). Vitamin D metabolites were analyzed in 65 CLD patients with 25(OH)D <30 ng/mL who received 20 000 IU cholecalciferol/week for 6 months. The primary outcome assessed serum 25(OH)D and 3-epi-25(OH)D in response to supplementation. Corresponding values from 16 CLD patients with sufficient vitamin D levels receiving no supplementation were compared. The epimer was detected in all samples and at lower relative concentrations with lower vitamin D baseline status, i.e., severe vitamin D deficiency (<10 ng/mL) as compared with deficient (10–19.9 ng/mL), insufficient (20–29.9 ng/mL), or sufficient (≥30 ng/mL) vitamin D levels (2.4% vs. 4.8%, 5.2%, 5.8%, respectively; P < 0.001). Similar relative concentrations for 3-epi-25(OH)D, ranging from 4.3%–7.1% (absolute concentrations: 1.1–4.0 ng/mL; all P < 0.001), were obtained in response to cholecalciferol in all supplemented patients, regardless of inadequacy threshold. Epimer levels significantly decreased (P = 0.007) in unsupplemented patients, coinciding with decreasing serum 25(OH)D concentrations over time. No epimer differences between patients with (n = 17) or without (n = 48) cirrhosis were demonstrated. The 3-epi-25(OH)D was present in serum of all patients at comparable levels to those reported by others. Epimer levels increased linearly with increasing 25(OH)D levels after supplementation. However, no effect of cirrhosis on epimer concentrations was observed.

Abstract:

The reduced hardware design of narrow implants increases the risk of fracture not only of the implant itself but also of the prosthetic constituents. Hence, the current study is aimed at estimating the probability of survival of anterior crowns supported by different narrow implant systems. Three different narrow implant systems of internal conical connections were evaluated (Ø3.5×10 mm): (i) Active (Nobel Biocare), (ii) Epikut (S.I.N. Implant System), and (iii) BLX (Straumann). Abutments were torqued to the implants, and standardized maxillary incisor crowns were cemented. The assemblies were subjected to step-stress accelerated life testing (SSALT) in water through load application of 30 degrees off-axis lingually at the incisal edge of the crowns using a flat tungsten carbide indenter until fracture or suspension. The use level probability Weibull curves and reliability for completion of a mission of 100,000 cycles at 80 N and 120 N were calculated and plotted. Weibull modulus and characteristic strength were also calculated and plotted. Fractured samples were analyzed in a stereomicroscope. The beta (β) values were 1.6 (0.9-3.1) and 1.4 (0.9-2.2) for BLX and Active implants, respectively, and 0.5 (0.3-0.8) for the Epikut implant, indicating that failures were mainly associated with fatigue damage accumulation in the formers, but more likely associated with material strength in the latter. All narrow implant systems showed high probability of survival (≥95%, CI: 85-100%) at 80 and 120 N, without significant difference between them. Weibull modulus ranged from 6 to 14. The characteristic strength of Active, Epikut, and BLX was 271 (260-282) N, 216 (205-228) N, and 275 (264-285) N, respectively. The failure mode predominantly involved abutment and/or abutment screw fracture, whereas no narrow implant was fractured. Therefore, all narrow implant systems exhibited a high probability of survival for anterior physiologic masticatory forces, and failures were restricted to abutment and abutment screw.

Pretendemos, neste artigo, estabelecer, defronte as proposições centrais do movimento transhumanista – expresso aqui na filosofia de FM-2030 – e o discernimento helenístico de Epicuro, (I) pontos de tangência e (II) fundamentação/corroboração do segundo ao primeiro. A morte, asserta FM-2030, deve ser superada. Tomando esta proposição como ponto arquimediano do pensamento transhumanista, investigamos na sabedoria de Epicuro um possível fortalecimento deste alicerce proposicional no afã de, para além do caráter formal de sua consistência, somar ao movimento contemporâneo o comedimento do hedonismo epicúreo, uma vez que as potencialidades provenientes da tecnociência podem ampliar os efeitos da desmesura humana. Tentamos, portanto, mostrar que um movimento com olhos para o futuro pode ser fortalecido com o mestre Epicuro. Nos focando no transhumanismo de FM-2030 e, em contraste com Epicuro e sua teoria dos desejos, buscamos elucidar que o filósofo legitimaria uma vida longeva, uma vez que a longevidade ou mesmo a imortalidade não implicamdescomedimento.

Late antique social structures in the provinces of Epirus Vetus and Epirus Nova, as represented by urban centres, ecclesiastical authority and Roman material culture, largely disappeared during the first half of the 7th c. Around this time the practice of furnished burial was adopted by sections of the population. This transition has traditionally been interpreted as reflecting an expression of ethnic identity on the part of an indigenous population. This paper argues, however, that these post-Roman cemeteries are a reflection of the more localised and fluid social structures that emerged in post-Roman Epirus rather than an attempt to construct or maintain ethnic identities.

Abstract:

O desinteresse estético é um dos conceitos centrais da estética, e Jerome Stolnitz, o mais proeminente teórico do desinteresse no século XX, afirmou que (i) o envolvimento dos pensadores antigos com esta noção era superficial e pouco desenvolvido e, consequentemente, (ii) a emergência do desinteresse no século XVIII marca o nascimento da estética como disciplina. Neste artigo, pretendo usar os trabalhos existentes de Epicuro para mostrar que o filósofo antigo não apenas tinha conceitos similares, como também os usava de maneiras cuidadosas e complexas. Eu argumento que, no enquadramento teórico de Epicuro, as artes pertenciam a categoria dos desejos "meramente naturais", e esta classificação, combinada com aquilo que sabemos da rejeição de Epicuro da crítica da arte, mostra que ele tinha desenvolvido cuidadosamente razões que fundamentavam a ideia de que a arte deveria ser abordada terminalmente, ao invés de instrumentalmente. Por fim, eu comparo a noção de desinteresse estético com as visões de Epicuro acerca da arte e argumento que de muitas maneiras a última não é inferior a primeira, e que, portanto, deve pertencer a história da estética.

Abstract:

For the last couple of decades, there has been a significant growth in sequencing data, leading to an extraordinary increase in the number of gene variants. This places a challenge on the bioinformatics research community to develop and improve computational tools for functional annotation of new variants. Genes coding for epigenetic regulators have important roles in cancer pathogenesis and mutations in these genes show great potential as clinical biomarkers, especially in hematologic malignancies. Therefore, we developed a model that specifically focuses on these genes, with an assumption that it would outperform general models in predicting the functional effects of amino acid substitutions. EpiMut is a standalone software that implements a sequence based alignment-free method. We applied a two-step approach for generating sequence based features, relying on the biophysical and biochemical indices of amino acids and the Fourier Transform as a sequence transformation method. For each gene in the dataset, the machine learning algorithm–Naïve Bayes was used for building a model for prediction of the neutral or disease-related status of variants. EpiMut outperformed state-of-the-art tools used for comparison, PolyPhen-2, SIFT and SNAP2. Additionally, EpiMut showed the highest performance on the subset of variants positioned outside conserved functional domains of analysed proteins, which represents an important group of cancer-related variants. These results imply that EpiMut can be applied as a first choice tool in research of the impact of gene variants in epigenetic regulators, especially in the light of the biomarker role in hematologic malignancies. Abstract:

Este trabalho se trata de uma pesquisa aos textos principais de Epicuro, filosofo helenista que desenvolveu teses a respeito de como se viver com um prazer necessário, porém módico ou que não cause dor e sofrimento na sua busca se houver a não concretização. No mundo moderno ou na era helenística, o ser humano sempre esteve em busca de seus objetivos, metas, sonhos e realizações e isto envolve uma consciência da plenitude de viver, aprender, experenciar e saborear ou não seus prazeres. Desta forma traçou-se um paralelo entre o hedonismo de Epicuro, Onfray e Lipovetsky para entendermos como o ser humano se move para ir em busca de seus prazeres. Para isso, fizemos uma pesquisa bibliográfica apontando o hedonismo perceptivo de cada filosofo citado acima e estudamos a condição atual dos sujeitos perante o mundo de hoje. As considerações sobre este estudo é a de que Epicuro ainda precisa ser muito bem compreendido e trazido aos nossos dias para que possamos refletir filosoficamente sobre nossas escolhas que dão vazão ao prazer de cada um.
