Abstract:
Mon travail de thèse porte sur les doubles EPW sextiques, une famille de variétés hyperkähleriennes qui, dans le cas général, sont équivalentes par déformation au schéma de Hilbert de deux points sur une surface K3. Notamment j'ai utilisé le lien que ces variétés ont avec les variétés de Gushel-Mukai, qui sont des variétés de Fano dans une Grassmannienne si leur dimension est plus grande que deux, des surface K3 si la dimension est deux.Le premier chapitre contient quelques rappels de théorie des équations de Pell et des réseaux, qui sont fondamentals pour l’étude des variétés hyperkähleriennes. Ensuite je rappelle la construction qui associe un revêtement double à un faisceau sur une variété normale.Dans le deuxième chapitre j’aborde les variétés hyperkähleriennes et je décris leurs premières propriétés ; j’introduis aussi le premier cas de variété hyperkählerienne qui a été étudiée, les surfaces K3. Cette famille de surfaces correspond aux variétés hyperkähleriennes en dimension deux.Je présente ensuite brièvement certains des derniers résultats dans ce domaine, notamment je définis différents espaces de modules de variétés hyperkähleriennes et je décris l’action d’un automorphisme sur le deuxième groupe de cohomologie d’une variété hyperkähleriennes.Les outils introduits dans le chapitre précédent ne fournissent pas de description géométrique de l'action de l'automorphisme sur la variété, dans le cas où la variété est un schéma de Hilbert de points sur une surface K3. Dans le troisième chapitre, j’introduis donc une description géométrique à une certaine déformation près. Cette déformation prend en compte la structure du schéma de la variété de Hilbert. Pour ce faire, j'introduis un isomorphisme entre une composante connexe de l'espace de modules des variétés de type K3[n] avec une polarization, et l'espace de modules des variétés de même type avec une involution dont le rang de l'invariant est un. Il s’agit d’une généralisation d’un résultat obtenu par Boissière, An. Cattaneo, Markushevich et Sarti en dimension deux. Les deux premières parties de ce chapitre sont un travail en collaboration avec Alberto Cattaneo.Dans le quatrième chapitre, je définis les EPW sextiques, en présentant l'argument de O'Grady, qui montre qu'un double revêtement d'un EPW sextique dans le cas général est une variété de type K3[2]. Ensuite, je présente les variétés Gushel-Mukai, en mettant l'accent sur leur lien avec les EPW sextiques ; cette approche a été introduite par O'Grady, poursuivie par Iliev et Manivel et systématisée par Kuznetsov et Debarre.Dans le cinquième chapitre, j’utilise les outils introduits dans le quatrième chapitre dans le cas où on peut associer une surface K3 à une EPW sextique X. Dans ce cas je donne des conditions explicites sur le groupe de Picard de la surface pour que X soit une variété hyperkählerienne. Cela permet d'utiliser le théorème de Torelli pour une surface K3 pour démontrer l'existence de quelques automorphismes sur X. Je donne des bornes sur la structure d'un sous-groupe d'automorphismes d'une EPW sextique sous conditions d'existence d'un point fixe pour l'action du groupe.Toujours dans le cas d'existence d'une surface K3 associée à une EPW sextique X, j’améliore la borne obtenue précédemment sur les automorphismes de X, en donnant un lien explicite avec le nombre de coniques sur la surface K3. Je montre que la symplecticité d'un automorphisme sur X dépend de la symplecticité d'un automorphisme correspondant sur la surface K3.Le sixième chapitre est un travail en collaboration avec Alberto Cattaneo. J'étudie le groupe d'automorphismes birationels sur le schéma de Hilbert des points sur une surface projective K3, dans le cas générique. Cela généralise le résultat obtenu en dimension deux par Debarre et Macrì. Ensuite j’étudie les cas où il existe un modèle birationel où ces automorphismes sont réguliers. Je décris de façon géométrique quelques involutions dont on avait prouvé l'existence auparavant<br>My thesis work focuses on double EPW sextics, a family of hyperkähler manifolds which, in the general case, are equivalent by deformation to Hilbert's scheme of two points on a K3 surface. In particular I used the link that these manifolds have with Gushel-Mukai varieties, which are Fano varieties in a Grassmannian if their dimension is greater than two, K3 surfaces if their dimension is two.The first chapter contains some reminders of the theory of Pell's equations and lattices, which are fundamental for the study of hyperkähler manifolds. Then I recall the construction which associates a double covering to a sheaf on a normal variety.In the second chapter I discuss hyperkähler manifolds and describe their first properties; I also introduce the first case of hyperkähler manifold that has been studied, the K3 surfaces. This family of surfaces corresponds to the hyperkähler manifolds in dimension two.Furthermore, I briefly present some of the latest results in this field, in particular I define different module spaces of hyperkähler manifolds, and I describe the action of automorphism on the second cohomology group of a hyperkähler manifold.The tools introduced in the previous chapter do not provide a geometrical description of the action of automorphism on the manifold for the case of the Hilbert scheme of points on a general K3 surface. In the third chapter, I therefore introduce a geometrical description up to a certain deformation. This deformation takes into account the structure of Hilbert scheme. To do so, I introduce an isomorphism between a connected component of the module space of manifolds of type K3[n] with a polarization, and the module space of manifolds of the same type with an involution of which the rank of the invariant is one. This is a generalization of a result obtained by Boissière, An. Cattaneo, Markushevich and Sarti in dimension two. The first two parts of this chapter are a joint work with Alberto Cattaneo.In the fourth chapter, I define EPW sextics, using O'Grady's argument, which shows that a double covering of a EPW sextic in the general case is deformation equivalent to the Hilbert square of a K3 surface. Next, I present the Gushel-Mukai varieties, with emphasis on their connection with EPW sextics; this approach was introduced by O'Grady, continued by Iliev and Manivel and systematized by Kuznetsov and Debarre.In the fifth chapter, I use the tools introduced in the fourth chapter in the case where a K3 surface can be associated to a EPW sextic X. In this case I give explicit conditions on the Picard group of the surface for X to be a hyperkähler manifold. This allows to use Torelli's theorem for a K3 surface to demonstrate the existence of some automorphisms on X. I give some bounds on the structure of a subgroup of automorphisms of a sextic EPW under conditions of existence of a fixed point for the action of the group.Still in the case of the existence of a K3 surface associated with a EPW sextic X, I improve the bound obtained previously on the automorphisms of X, by giving an explicit link with the number of conics on the K3 surface. I show that the symplecticity of an automorphism on X depends on the symplecticity of a corresponding automorphism on the surface K3.The sixth chapter is a work in collaboration with Alberto Cattaneo. I study the group of birational automorphisms on Hilbert's scheme of points on a projective surface K3, in the generic case. This generalizes the result obtained in dimension two by Debarre and Macrì. Then I study the cases where there is a birational model where these automorphisms are regular. I describe in a geometrical way some involutions, whose existence has been proved before

Class of 2009 Abstract<br>OBJECTIVES: To develop a novel characterization system utilizing immunofluorescent FLAG tagged EP2 and EP4 receptors to assist in the explanation of their unique cell signaling properties for exploitation in future drug development design. METHODS: Plasmids were obtained and isolated that contained cDNAs encoding FLAG-tagged EP2 and EP4 receptors for transient expression in HEK-293 cells. The sequences of these plasmids were confirmed by restriction enzyme analysis and DNA sequencing. Transfected cells were treated with vehicle, PGE2 or forskolin to assess appropriate receptor functionality based on cAMP induction. RESULTS: The two PGE2 receptor subtypes, EP2 and EP4, are similar in their activation of adenylyl cyclase (AC) and subsequent up regulation of cAMP production. These receptors differ, however, in that EP2 more efficiently stimulates cAMP production and EP4 signaling involves the activation of phosphatidylinositol 3-kinase (PI3K) and extracellular signal related kinases (ERKs). The PGE2- treated cells responded as predicted with intracellular production of cAMP, with the EP2 receptor responding more efficiently than the EP4 receptor. CONCLUSIONS: The intent is for these cells to be used as a novel assay system for the development of future selective EP2 and EP4 agonists. This research could potentially benefit in selectively targeting EP2 or EP4 pathways linked to prevalent ailments such as pain, fever, inflammation, possibly cancer or bone growth.

Abstract:
Chronic kidney disease is defined as the appearance of kidney functional or structural injury. Cyclooxygenase and prostaglandin E2 have been implicated in the pathogenesis of diabetic nephropathy, the leading cause of chronic kidney disease. Beneficial in certain settings, inhibition of the cyclooxygenase pathway can however be detrimental in patients with compromised cardiac or renal function. Moreover, the quest for new therapies to treat diabetic nephropathy is hampered by the lack of appropriate rodent models. This doctoral thesis is a culmination of three studies, the first to determine the role of the prostaglandin E2 EP1 receptor in diabetic nephropathy, the second to elucidate the vascular prostaglandin E2 EP4 receptor’s role in hypertension and lastly to establish and characterise a novel mouse model of diabetic nephropathy. The goal being to uncover new therapeutic avenues for the treatment of CKD, its causes and/or complications.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior<br>Epilepsy is one of the most common neurologic disorders. It has been suggested that seizures may be facilitaded by inflammation. PGE2 is one of the most important inflammatory mediators, and facilitates pentylenetetrazol (PTZ)-induced seizures by stimulating EP1 and EP3 receptors. However, up to the present moment, no study has investigated whether EP1 and EP3 receptors blocking attenuate seizures induced by convulsants other than PTZ. It is also unknown whether Na+,K+-ATPase activity alterations are involved in such an effect. Therefore, in the current study we investigated whether EP1 and EP3 ligands (agonists and antagonists) modulate PTZ- and kainic acid (KA)-induced seizures, and whether alterations in Na+,K+-ATPase activity mediate such a protective effect, in mice. EP1 and EP3 antagonists (ONO-8713 and ONO-AE3-240, respectively, 10 Og/kg, s.c.) attenuated PTZ (60 mg/kg, i.p.)- and KA (20 mg/kg, i.p.)-induced seizures. The respective agonists (ONO-DI-004 and ONO-AE-248, 10 Og/kg, s.c.) facilitated seizures in both acute models, and at noneffective doses, prevented the protective effects of the antagonists. Animals injected with PTZ presented decreased Na+,K+-ATPase activity in the cerebral cortex and hippocampus. On the other hand, animals injected with KA presented increased Na+,K+-ATPase activity in the same cerebral structures at the end of the experiment. These divergent findings suggest that alterations in Na+,K+-ATPase activity in both acute models depends on the convulsant agent used and make difficult to establish a relationship between Na+,K+-ATPase activity and seizure development. Moreover, EP1 and EP3 antagonists administration abolished Na+,K+- ATPase activity alterations induced by PTZ and KA, in such a way that these alterations seem to be related more to the presence of ictal phenomenon itself than to the seizure induction mechanisms. Notwithstanding, the currrent results clearly show that EP1 and EP3 receptors might constitute novel targets for anticonvulsants development, since EP1 and EP3 decreased seizures, regardless of the convulsant agent used.<br>A epilepsia é uma das disfunções neurológicas mais comuns. Tem sido sugerido que as crises epilépticas podem ser facilitadas pela ocorrência de inflamação. A PGE2 é um dos mediadores inflamatórios mais importantes que, agindo por meio dos receptores EP1 e EP3, facilita as convulsões induzidas por pentilenotetrazol (PTZ). Contudo, até a presente data, nenhum estudo investigou, de maneira sistêmica, se a ativação ou bloqueio de receptores EP1 e EP3 facilitam as convulsões induzidas por outros agentes; tampouco se alterações na atividade da Na+,K+-ATPase estão envolvidas nesse efeito. Assim, no presente estudo, investigamos se ligantes (agonistas e antagonistas) de receptores EP1 e EP3 modificam as crises induzidas por PTZ e ácido caínico (KA), e se tais efeitos estão associados a alterações na atividade da enzima Na+,K+-ATPase, em camundongos. Os antagonistas EP1 e EP3 (ONO-8713 e ONO-AE3-240, respectivamente, 10 Og/Kg, s.c.) atenuaram as convulsões induzidas por PTZ (60 mg/Kg, i.p.) e KA (20 mg/Kg). Os seus respectivos agonistas (ONO-DI-004 e ONO-AE-248 de 10 Og/Kg, s.c.) facilitaram as convulsões em ambos modelos agudos de crises epilépticas e, em doses não efetivas para gerar crises, preveniram os efeitos dos antagonistas. Os animais submetidos à administração de PTZ apresentaram, ao final do experimento, a atividade Na+,K+-ATPásica diminuída no córtex cerebral e hipocampo. Por outro lado, animais tratados com KA apresentaram um aumento na atividade Na+,K+-ATPásica nestas mesmas estruturas, que se correlacionou positivamente com a vigência de status epilepticus no momento do sacrifício. Os achados divergentes no que diz respeito à alteração da atividade da Na+,K+-ATPase nos dois modelos de crises agudas sugere que tais alterações estejam relacionadas ao tipo de agente convulsivante utilizado, e dificultam estabelecer, de forma inequívoca, uma relação entre atividade desta ATPase e sensibilidade à crises agudas. Ademais, a administração de antagonistas EP1 e EP3 aboliu as alterações da atividade da Na+,K+-ATPase induzidas tanto por PTZ como por KA, de tal forma que estas parecem estar mais associadas com o fenômeno ictal em si, do que com os mecanismos de indução da crise. Contudo, os resultados mostram de forma clara que os receptores EP1 e EP3 podem se constituir possíveis novos alvos para o desenvolvimento de drogas antiepilépticas, pois antagonistas EP1 e EP3 diminuíram as crises, independente do agente convulsivante utilizado.

Abstract:
Electron paramagnetic resonance (EPR) is a spectroscopic technique sensitive to unpaired electrons present in paramagnetic species such as free radicals and organometallic complexes. Electrochemistry (EC) is an interfacial science, where reduction and oxidation processes are studied. A single electron reduction or oxidation generates a paramagnetic species with an unpaired electron, thus making EPR a valuable tool in the study of electrochemical systems. In this work a novel electrochemical cell was designed and developed to be used with a specific type of EPR resonator, called loop gap resonator (LGR). After building and characterising the performance of the EC-EPR setup, it was adapted for quantitative measurements in electrochemical EPR (QEC-EPR). Thus, for the first time, the technique of EC-EPR has been fully characterised for analytical work, opening possibilities to study electrode reactions quantitatively with accuracy and precision not obtained before, as demonstrated in Chapter 8 of this thesis.

Creating reports with a huge amount of data can be a big help when it comes to decision making, Tele2 has collected data from a lot of di erent sources and are at the moment of writing developing a system that will easily be able to produce reports and comparisons. These reports are supposed to create the foundation for better decision making and to get a better view on how the company works today. The reporting environment Cognos is adapted to use data from various sources and make it easy to take out clear reports from this. To make their work easier we have received the task of transforming these reports from only being viewable on computers to also be viewable on tablets and other mobile devices, all to make it easier to look at these reports wherever you are and whenever you want. During our work we will install and congure the mobile package that Cognos has. We will also evaluate if this is the most e ective and best way to do this. The most important part of our work is to make these reports viewable on the mobile devices and also congured so that they are easy to view and read. To achieve this we have to adapt the reports so that they no longer only are developed to read on a big screen but also to a smaller one.

La firma del trattato internazionale più grande mai negoziato dall’Unione Europea avviene il 17 luglio 2018. L’obiettivo dell’Economic Partnership Agreement tra UE e Giappone è quello di creare una zona commerciale aperta che tocca oltre 600 milioni di persone, pari a un terzo dell’intero valore degli scambi commerciali a livello globale. Si tratta di un accordo win on win tra due mondi lontani che si avvicinano attraverso regole e obiettivi chiari e trasparenti. Per capire gli effetti di questo accordo, è necessario approfondire il contesto nel quale viene messo in atto ed è quindi necessario capire anche il punto di vista di Cina e USA che, al contrario delle due parti contraenti dell’EPA in questione, sono in lotta per contendersi la leadership globale. È importante capire anche quelle che sono le differenze tra iniziative come la BRI (Belt and Road Initiative), che rappresenta l’obiettivo di Pechino di unificare i mercati africani ed euroasiatici, e l’EPA. Verrà approfondita anche quella che è la nuova politica commerciale e geopolitica degli Stati Uniti, il principale alleato di UE e Giappone, in totale contrasto con i valori espressi dall’EPA. I negoziati dell’EPA, infatti, seppur inizialmente si concentrano sull'impatto economico, sulle barriere non-tariffarie classiche, come gli standard alimentari e sulla regolamentazione di nuova generazione, si focalizzano poi maggiormente sul progresso graduale di liberalizzazione commerciale sugli incrementi di efficienza e sulla maggiore integrazione normativa. L'impatto più significativo a lungo termine dell’accordo di partenariato tra Unione Europea e Giappone, deriva dall’attuazione di politiche a sostegno di un'ampia gamma di settori oltre al commercio. L’EPA è il perfetto esempio di come portare avanti trattative rispettose, caratterizzate dalla perseveranza e cooperazione tra alleati. È un accordo vantaggioso per tutti. Lo spirito di un accordo commerciale deve portare all’allineamento degli standard di livello inferiore con gli standard più elevati come le norme internazionali, anche se questo non significa semplificare il commercio reciproco, ma rafforza a sua volta gli standard per la comunità internazionale. In questo modo si crea una sorta di manuale commerciale globale, al quale i terzi che desiderano commerciare con l'Europa o il Giappone devono attenersi, dando luogo a una formulazione di comuni regole commerciali globali. L'EPA fornisce una base per la reciprocità che crea condizioni di parità tra Europa e Giappone.

Abstract:
The subject of the diploma thesis are procedures in EPS network related to the operation of mobile terminals. In the introduction of my work I described the EPS system, including the quality of services and bearers. The following section focuses on control procedures in the EPS system such as attach network procedure, detach procedure, service request, handover or tracking area procedure. Further, three technologies for Internet of Things were described, namely EC-GSM-IoT, LTE-M and NB-IoT. All the procedures mentioned above were also captured in the experimental VUT EPS network and analyzed by TravelHawk using software M5 and Wireshark. Theoretical and practical findings were subsequently used out for setting of laboratory task for subject MKPM.

El asma es una enfermedad crónica de las vías respiratorias cuyo abordaje terapéutico actual no frena su progresión, y en ocasiones no revierte la sintomatología. El análisis de mecanismos endógenos de protección puede resultar en la identificación de nuevas dianas farmacológicas efectivas. Los estudios preclínicos realizados hasta ahora en nuestro laboratorio junto con evidencia clínica y básica de otros grupos, sugirieron la hipótesis de que la activación selectiva del receptor prostanoide EP2 de le la PGE2 inhibe la actividad mastocitaria, y consecuentemente reduce el proceso patológico respiratorio inducido en ratones por exposición a aeroalérgenos, lo cual puede suponer una estrategia terapéutica antiasmática sinérgica a la del bloqueo mastocitario por otros mecanismos. Con el fin de verificarla, se planteó el primer objetivo: evaluar la función selectiva del receptor prostanoide EP2 en la modulación de la actividad mastocitaria, y en la reactividad respiratoria a aeroalérgenos. Se observó en poblaciones mastocitarias fenotípicamente diversas la inhibición de los mastocitos mediante dos agonistas EP2 químicamente distintos. Inhibición que también se demostró in vivo en un modelo de asma en ratón expuesto a aeroalérgenos de ácaros del polvo. El efecto inhibitorio sobre los mastocitos se acompañó de la prevención de la hiperreactividad bronquial, y apuntaba hacia un posible control del proceso inflamatorio broncovascular. La naturaleza protectora del agonismo EP2 se confirmó al administrar antagonistas en modelos in vitro e in vivo. Estos desencadenaron una sobreactividad de los mastocitos que agravó la alteración respiratoria. Todo ello sugiere que el efecto beneficioso de la PGE2 descrito en estudios clínicos experimentales, es consecuencia de la mediación del receptor EP2 mastocitario. El segundo objetivo planteado fue el desarrollo de un ratón transgénico que sobreexpresase el receptor EP2 específicamente en los mastocitos con el fin de disponer de una herramienta experimental para evaluar la relevancia relativa del receptor EP2 en la protección inducida por PGE2. Se logró con éxito insertar el gen de EP2 y sobreexpresarlo, demostrándose en algunos ensayos in vitro que los mastocitos aislados de los ratones transgénicos EP2 respondían con un incremento en el bloqueo. Sin embargo no se pudo confirmar in vivo el efecto beneficioso atribuible a la presencia de un mayor número de receptores EP2, probablemente debido a las fluctuaciones compensatorias en la expresión de otros genes relacionados. El desarrollo de la colonia transgénica de EP2 requiere de una caracterización más exhaustiva para investigar la importancia del EP2 mastocitario en la prevención de la alteración respiratoria. Finalmente, en un tercer objetivo se planteó evaluar comparativamente la efectividad preclínica de la inhibición mastocitaria mediante agonismo EP2 y bloqueo de la IgE con anticuerpos monoclonales, en modelos de alergia in vivo e in vitro. Se demostró en un modelo de anafilaxia cutánea in vivo y en poblaciones mastocitarias in vitro que el agonismo EP2 y la neutralización de la IgE ejercen un bloqueo de los mastocitos cuantitativamente equiparable. Para posteriores estudios de la potencial sinergia o complementariedad terapéutica entre ambos mecanismos en las vías respiratorias de ratones sensibilizados, se desarrolló con éxito un innovador modelo de anafilaxia respiratoria pasiva (PRA) en ratones transgénicos que expresaban el receptor humano de la IgE. Los resultados obtenidos en esta tesis permiten concluir que la activación del eje “PGE2 - EP2 mastocitario – asma” supone un mecanismo endógeno de protección frente a la disfunción de las vías respiratorias inducida por aeroalérgenos. Los nuevos modelos experimentales in vivo desarrollados en este proyecto permitirán caracterizar el mecanismo antiasmático, y confirmar la condición de diana terapéutica del receptor EP2, o de otras moléculas que formen parte de ese eje.<br>Asthma is a chronic disease of the airways whose therapeutic approach does stop progression, and occasionally does not revert the symptoms. Studying endogenous protective mechanisms may lead to the identification of newly effective pharmacological targets. Preclinical studies carried out in our laboratory together with clinical and basic evidence from other groups, suggested the following hypothesis: selective activation of the EP2 prostanoid receptor of PGE2 inhibits mast cell activity, and consequently reduces the pathological respiratory process induced in mice exposed to aeroallergens, and this may arise as an antiasthmatic therapeutic strategy synergic to mast cell blockade through other mechanisms. In order to check it, the first objective was to assess the selective function of the EP2 prostanoid receptor as a mast cell activity and aeroallergen-induced airways reactivity modulator. Phenotypically distinct mast cell populations were shown to be inhibited by means of chemically different agonists. Such inhibition was also seen in vivo in the asthma murine model induced through exposure to house dust mite aeroallergens. Mast cells inhibition was paralleled by a prevention of the airway hyperreactivity, and possibly a control of the bronchovascular inflammatory process. The protective nature of EP2 agonism was confirmed through the administration of antagonists in in vitro and in vivo models, where mast cells hyperreleasability and worsening of airways disorder were observed. All in all, this suggested that the described benefit of PGE2 in clinical experimental settings is mediated by the mast cells EP2 receptor. The second objective was to develop a transgenic mouse overexpressing the EP2 receptor specifically on the mast cells surface as an experimental tool to evaluate the relative relevance of EP2 in PGE2-driven protection. The EP2 gene was successfully inserted and overexpressed, owing to an increased blockade of the activity of mast cells isolated from transgenic mice. However, the beneficial effect attributable to a higher number of EP2 receptors could not be seen in vivo, possibly due to the compensatory fluctuation of related genes. The developed EP2 transgenic mice require a further characterization in order to investigate the role of mast cell EP2 in prevention of the respiratory dysfunction. Finally, the third objective was to compare the preclinical effectiveness of EP2 agonism versus anti-IgE using monoclonal antibodies, in allergy models in vivo and in vitro. Both, EP2 agonism and IgE neutralization exerted a quantitatively similar effect on mast cells population in vitro and in a model of cutaneous anaphylaxis in vivo. In order to further study the synergy or therapeutic complementarity between both mechanisms, an innovative model of passive respiratory anaphylaxis (PRA) was successfully established in transgenic mice expressing the human IgE receptor. The results of this thesis brought us to conclude that the “PGE2- mast cell EP2-asthma” axis is an endogenous protective mechanism against the airway dysfunction caused by aeroallergens. The new in vivo experimental models developed in this project, will allow to characterize the antiasthmatic mechanism, and confirm the EP2, or other molecules along the axis, as therapeutic targets.

Les accidents vasculaires cérébraux (AVC) représentent l’une des premières causes de morbi-mortalité dans les pays industrialisés et sont en constante augmentation. Pour l’heure, aucune autre thérapie que la thrombolyse intraveineuse n’est validée, or moins de 10% des patients y sont éligibles.Les premiers essais de thérapie régénérative par greffe de progéniteurs endothéliaux ont montré leur efficacité au niveau préclinique sur la récupération fonctionnelle, mais très peu de cellules parviennent jusqu’au site de la lésion pour y exercer leurs effets. De précédents travaux ont montré que l’érythropoïétine (EPO) augmentait la mobilisation et la prolifération de progéniteurs endothéliaux depuis la moelle osseuse, ainsi que leur adressage vers les sites ischémiés in vivo.Nous avons formulé l’hypothèse que l’EPO pouvait permettre aux progéniteurs endothéliaux vrais (ECFCs) d’atteindre le site ischémié en plus grand nombre pour y exercer un effet plus important, plus rapidement. Nous avons donc décidé d’évaluer trois stratégies d’optimisation : coadministration d’ECFCs et d’EPO, administration d’ECFCs prétraités à l’EPO, et enfin coadministration d’ECFCs et d’un dérivé de l’EPO dénué d’effet hématopoïétique. Pour nos derniers travaux, nous nous sommes aidés de l’imagerie isotopique µSPECT/CT pour quantifier l’adressage au niveau de la lésion des ECFCs avec ou sans optimisation pharmacologique à l’EPO, et évaluer longitudinalement leurs effets bénéfiques sur la rupture de la barrière hémato-encéphalique, l’apoptose cérébrale et le débit sanguin cérébral résiduel, en complément des techniques référentes et de l’évaluation clinique neurofonctionnelle<br>Stroke represents a leading cause of morbidity and mortality in industrialized countries and prevalence steadily increases. For now, no other therapy that intravenous thrombolysis is validated, still, less than 10% of patients are eligible.The first preclinical regenerative therapy trials using endothelial progenitor cells engraftment gave evidence of their efficacy on functional recovery, although very few cells reached the site of injury to exert their effects are. Previous works showed that erythropoietin (EPO) increased mobilization and proliferation of endothelial progenitor cells from bone marrow, as well as their homing to the ischemic sites in vivo.We hypothesized that EPO could enable true endothelial progenitor cells (ECFCs) to reach the ischemic site in larger quantity, faster, to exercise a greater effect. We decided to evaluate three optimization strategies: coadministration of ECFCs and EPO, administration of EPO-pretreated ECFCs, and finally coadministration of ECFCs and an EPO derivative devoid of hematopoietic effect.In this work, we used μSPECT/CT imaging to quantify the ECFCs homing to the ischemic site with or without EPO optimization after cerebral ischemia, and longitudinally assessed their beneficial effects on blood-brain barrier disruption, cerebral apoptosis and residual cerebral blood flow, complementary to the referent techniques and neurofunctional clinical evaluation

The benefits of Ericsson virtual Evolved Packet Core includes all the benefits of NFV and provides verified solutions addressing a large number of vertical use-cases. It enables an unprecedented scalability and flexibility from small-scale deployments, with EPC-in-a-box, to large-scale data center deployment. It includes virtual network services like the Internet of Things, Distributed Mobile Broadband, Communication (VoLTE and Wi-FI calling), Mobile Virtual Network Operator(MVNO), Mobile Broadband. Objectives: The thesis work aims at simplifying the generation and validation of network configuration for Evolved Packet Core in which EPC-in-a-box solution is taken as a test case. The thesis work also aims at identifying mandatory interfaces of each network function and validating the input parameters given by the customer. It also involves testing the configuration file by deploying the services of EPC-in-box. Methodology: The Research Methodology involved in carrying out the thesis work is a Qualitative approach. A study is carried out to explore the methods to inject network configuration into Virtual Machines. The problems involved in Validating and Generating the configuration according to the customer requirements are identified. A suitable method is developed to simplify the process. Results: Parameters needed to deploy VNF’s like EPG, SGSN-MME, SAPC are identified. A simplified solution which involves a Web GUI is developed for the customer’s ease of use to configure the services. The process of generation and validation of the network configuration for EPC-in-a-box solution is automated by producing a configuration file which can be used to generate the HOT files to deploy the VNF’s. Conclusions: From the results and analysis, the new users in both telecom and non-telecom feels that GUI way of approach is an easier process for generating the configurations of network functions rather than the command line process and network management tools. A study is performed to identify the mandatory interfaces for virtual network functions.

Svensk patenträtt har i stor omfattning harmoniserats med praxis utvecklad av det europeiska patentverket (EPO), vad avser patenterbarhetskriterier. Anledningen till harmoniseringen har motiverats av att det ska bli en mer enhetlig bedömning av patent i Europa. Harmoniseringen har i vart fall inte innefattat så kallad formell praxis, som innefattar regler om hand­läggandet hos det svenska patentverket, i förhållande till handläggandet av det vid EPO.   Vid Sveriges anslutning till European Patent Convention (EPC) år 1978, förespråkades att det svenska patentverket ansågs hålla en godtagbar standard i sin handläggning, men att det fram­tida förhållandet mellan svensk patenträtt och EPC kunde komma att innebära förändring om kvalitetsnivån inte kunde anses tillräcklig hög. Efter ett nyligen taget beslut av Patentbesvärsrätten (PBR) har Patent- och registreringsverkets (PRV) handläggning ifrågasatts, varför författarna valt att undersöka om en förändring av hand­läggningen vid PRV bör genomföras, i enlighet med det förslag som framställs i proposition 1977/78:1, med beaktande av praxis utvecklad av EPO.   I uppsatsen redogörs för hur EPO har tydligare regler avseende patent­ärenden än vad PRV har, varför en harmonisering av detta område bör diskuteras för säkerställandet av hög kvalitet vid handläggningen hos det svenska patentverket. Det är författarnas uppfattning att en ofullständig harmonisering även av formella regler resulterar i ett problem för säkerställandet av rättssäkerheten, eftersom lagregleringen i Sverige kan anses bristfällig. Således är det av intresse för svensk patenträtt att ta hänsyn till formell praxis utvecklad av EPO, alternativt, genom lagstiftningsförfarande, reglera bristen av tydliga regler.

An EPS is a complex and lively entity composed of intelligent modules that interact through bio-inspired mechanisms, to ensure high system availability and seamless reconfiguration. The actual economic situation together with the increasing demand of high quality and low priced customized products imposed a shift in the production policies of enterprises. Shop floors have to become more agile and flexible to accommodate the new production paradigms. Rather than selling products enterprises are establishing a trend of offering services to explore business opportunities. The new production paradigms, potentiated by the advances in Information Technologies (IT), especially in web related standards and technologies as well as the progressive acceptance of the multi-agent systems (MAS) concept and related technologies, envision collections of modules whose individual and collective function adapts and evolves ensuring the fitness and adequacy of the shop floor in tackling profitable but volatile business opportunities. Despite the richness of the interactions and the effort set in modelling them, their potential to favour fault propagation and interference, in these complex environments, has been ignored from a diagnostic point of view. With the increase of distributed and autonomous components that interact in the execution of processes current diagnostic approaches will soon be insufficient. While current system dynamics are complex and to a certain extent unpredictable the adoption of the next generation of approaches and technologies comes at the cost of a yet increased complexity.Whereas most of the research in such distributed industrial systems is focused in the study and establishment of control structures, the problem of diagnosis has been left relatively unattended. There are however significant open challenges in the diagnosis of such modular systems including: understanding fault propagation and ensuring scalability and co-evolution. This work provides an implementation of a state-of-the-art agent-based interaction-oriented architecture compliant with the EPS paradigm that supports the introduction of a new developed diagnostic algorithm that has the ability to cope with the modern manufacturing paradigm challenges and to provide diagnostic analysis that explores the network dimension of multi-agent systems.

No cenário televisivo, e possível agrupar todos os conteúdos, criando métodos (ou formas) de recomendação para o telespectador. Embora algumas ferramentas semânticas ainda não se encontrem suficientemente maduras, dificultando a confiança nestes serviços, este trabalho tentou utilizar os conceitos da web semântica para fazer recomendações em serviçós IPTV com o MEO.<br>Semantic Web is a concept that lets you get to knowledge instead of information through the network. Through the Semantic Web is possible to aggregate all contends in a more intuitive way. On television scenario, is possible group all contents, creating methods (or forms) of recommendation to the viewer. Although some semantic tools are not yet mature enough, making it difficult to trust on these services, this study attempted to use the concepts of the semantic web to make recommendations on IPTV services with MEO.

The issue of energy saving building is a current topic of today. The increasing costs of operating buildings reduce economic efficiency of the organization operations. A large part of the operating cost is energy cost. An effective tool for improving the energy performance of buildings is an EPC method. The implementation of energy projects through EPC is offered by Energy Services Company. This thesis evaluates the economic efficiency of the project implemented by the EPC and analyses its risk factors. The subject of EPC project is modernisation of an energy sector of a hospital building. The economic efficiency of the project is interpreted on the basis of economic indicators Net Present Value, Internal Rate of Return and Profitability Index. There was also performed Monte Carlo simulation supported by Oracle Crystal Ball software within the analysis of risk factors for the project.

The always increasing complexity of supramolecular systems requires not only new and efficient synthetic approaches but also the development of analytical methods capable of measuring the molecular scale phenomena exhibited by supramolecular assemblies. EPR spectroscopy, a magnetic resonance spectroscopic technique, has proven to be a useful tool for the investigation of supramolecular systems. My Ph.D. was devoted to the application EPR spectroscopy to the investigation of several supramolecular architectures. Among them, the investigation of an important non-covalent interaction, analogue of hydrogen bond (HB), called halogen bond (XB). The study of this interactions has been realized by the design and synthesis of a suitable spin probe. Exploiting these spin probes allowed us to measure the kinetic and thermodynamic parameters for this interaction in solution by performing complexation experiments with XB acceptors. A derivative of a dibenzo-24-crown-8 ether (DB24C8) containing the BTBN motif for the investigation of its host properties toward guests such as alkali metal cations and organic guests was then studied. The EPR analysis enabled the measurement of the degree of complexation and to discriminate the guest from the experimental EPR readout. The formation of a pseudorotaxane with a thread molecule containing both the dialkylammonium and bipyridinium recognition sites, and the switching of the ring between the two stations of the guest was detected directly in solution. The 4th chapter is devoted to the development and optimization of a mild and high yielding synthetic method for the synthesis of spin labelled [2]-rotaxanes by exploiting the active metal template copper catalysed azide alkyne cycloaddition (AT-CuAAC). The EPR investigation of these NN labelled rotaxanes provided many useful information on the conformational dynamics in solution. Their ability to form complexes with metal cations, and the effect it has on the EPR spectra of the spin labelled rotaxanes and threads was also investigated.

Wenn ein Chirurg heutzutage während der Operation Informationen aus der Patientenakte benötigt, ist er gezwungen, sich entweder unsteril zu machen oder Personal anzuweisen, ihm die entspre-chenden Informationen zugänglich zu machen. Aus technischer Sicht ist ein System zur intraoperati-ven Bedienung und Darstellung sehr einfach zu realisieren. Grundlage dafür ist eine elektronische Patientenakte (EPA), welche beispielsweise softwaregenerierten oder eingescannten Dokumenten verwaltet. Die vorliegende Arbeit widmet sich den folgenden Fragen: Wird ein solches System im Operationssaal sinnvoll genutzt? Welche Methoden zur sterilen Bedienung kommen infrage? Wie muss die grafische Darstellung auf den Operationssaal abgestimmt werden? Kann durch das Imple-mentieren aktueller Kommunikationsstandards auf alle verfügbaren Patientendaten zugegriffen werden? Dazu wurden in einer ambulanten HNO-Klinik zwei Pilotstudien durchgeführt. In der ersten Studie wurde das erste auf dem Markt befindliche kommerzielle Produkt „MI-Report“ der Firma Karl Storz evaluiert, welches per Gestenerkennung bedient wird. Für die zweite Studie wurde ein EPA-System entwickelt (Doc-O-R), welches eine Vorauswahl der angezeigten Dokumente in Abhängigkeit des Eingriffs traf und mit einem Fußschalter bedient werden konnte. Pro System wurden ca. 50 Eingriffe dokumentiert. Dabei wurde jedes angesehene Dokument und der Nutzungsgrund protokolliert. Die Systeme wurden durchschnittlich mehr als einmal pro Eingriff genutzt. Die automatische Vorauswahl der Dokumente zur Reduzierung der Interaktionen zeigte sehr gute Ergebnisse. Da das behandelte Thema noch in den Anfängen steckt, wird in der Arbeit am Ende auf die Vielzahl von Möglichkeiten eingegangen, welche bezüglich neuartiger Darstellungsmethoden, Bedienvorrich-tungen und aktueller Standardisierungsaktivitäten noch realisiert werden können. Dadurch werden zukünftig auch die Abläufe in der Chirurgie beeinflusst werden.

I detta examensarbete har åtgärder i ett energieffektiviseringsarbete av typen EPC analyserats. Umeå kommuns fastighetsavdelning startade sitt EPC-projekt 2008 syftet att energieffektivisera mer än 130 byggnader. Detta projekt sades då vara det största av sitt slag i Sverige. Bakgrunden till behovet av att utföra detta grundades i att en stor del av de tekniska systemens livslängd i fastigheterna började vara uppnådd samtidigt som ett energieffektiviseringskrav ålades fastighetsavdelningen. Siemens anlitades som entreprenör. De villkor som styrde projektet kom från beslutsfattande politiker och handlade om den viktiga besparingsgarantin. Denna garanti var en förutsättning för att få ekonomiska medel att finansiera ett projekt av den här storleken. EPC som modell valdes för att möjliggöra dessa krav. Den data som legat till grund för analysen av de energieffektiviseringsåtgärder som utförts har funnits samlad i eRoom, Siemens plattform för EPC-projekt. Den information som varit mest essentiell har varit de projektutvecklingsrapporter som för varje objekt beskrivit läge, problembild, åtgärdsförslag samt de energikartläggningar som innehållit alla energiberäkningar för energiläget i respektive fastighet samt potentiellt energiläge efter utförande av föreslagna åtgärder. De åtgärder som ansågs relevanta för att på det mest tydliga vis beskriva resultatet av Umeå EPC-projekt beslutades få ligga till grund för att reda ut hur besparingsfördelningen såg ut. Resultatet av detta arbete visar att de åtgärder som levererade störst del av den totala besparingen i ren energi var åtgärder kopplade till fläktar samt övriga åtgärder där styråtgärder av motorvärmare, injusteringar och pumpar rymdes. Efter dessa poster var tilläggsisolering samt belysningsåtgärder stora. Vidare vad gäller den ekonomiska besparingen var det konverteringar av uppvärmning som var absolut störst, därefter fläktarbeten samt kategorin övriga poster. Den ekonomiska investeringen var svår att på ett rättvist sätt jämföra mot hur stora besparingarna varit per åtgärd. En jämförelse av besparing mot antal åtgärder inom varje kategori visar däremot att konvertering till fjärrvärme samt värmeåtervinning var de mest effektiva åtgärderna.

Ermoluminescência (TL), Ressonância Paramagnética Eletrônica (EPR) e absorção Óptica (AO) de duas amostras de zirconita ou zircão (de cor marrom), de Minas Gerais, de origem exata e desconhecida. As duas curvas de emissão TL mostraram a presença de 4 picos TL visíveis em 140, 215, 270 e 350 ºC, bastante superpostos. Por deconvolução, outros picos foram revelados. A intensidade dos picos TL em 140, 215 ºC em função da dose apresenta um comportamento linear seguido de sublinear. O espectro de emissão TL é composto de duas bandas em 480 e 580 nm, que são devidas à presença do Dy na nossa amostra. Além de Dy muitos outros elementos são encontrados na amostra, porém, não têm influência na emissão TL. Os espectros de EPR da amostra em pó irradiada mostraram a presença de um sinal central em g=2,000, devido ao centro SiO45- que é isotrópico, o que é facilmente observado na amostra policristalina (em pó). Na amostra monocristalina pode-se observar que existem outros centros EPR não isotrópicos, produzidos durante a irradiação da amostra, e que são devidos à perda ou ganho de um elétron por parte do grupo SiO44-. Os estudos de decaimento isócrono do sinal SiO45 e dos picos TL mostraram que existe correlação entre o centro EPR e os centros TL, o que nos levou a um possível mecanismo TL, segundo o qual o Dy3+ perde um elétron passando para Dy4+ durante a irradiação. O elétron liberado é capturado por SiO44- formando o centro EPR, SiO45-. Quando se aquece a amostra para a leitura TL o processo inverso acontece, deixando Dy3+ em estados excitados, e na transição ao estado fundamental duas emissões de luz, uma em 480nm e outra em 580nm são observados. As bandas de AO são devido ao Urânio (1100), H2O (1900nm) e o radical OH (1400nm). Estas bandas não são afetadas pela irradiação gama utilizada neste trabalho.<br>In the present work the investigation of some properties of Thermoluminescence (TL), Electronic Paramagnetic Resonance (EPR) and Optical Absorption (OA) of two samples of zircon (of brown color) from Minas Gerais, but the unknown exact origin, were carried out. The glow curves of thermoluminescence emission have shown the presence of 4 TL peaks at 140, 215, 270 and 350 ºC although strongly overlaped. By deconvolution more peaks were revealed. The intensity of the TL peaks at 140 and 215 ºC as function of the radiation dose presents linear and sublinear behavior. The TL emission spectrum has shown two bands at 480 and 580 nm, which are due to the Dy content in our sample. Appart from Dy many other elements are found in the sample, however, they do not participate in the TL emission. The EPR spectra of the irradiated powder sample showed a central signal whith g=2,000 that is due to the SiO45- center. This signal is isotropic and easily observed in the polycrystalline sample (in powder). In the monocrystalline sample non-isotropic signals can be observed due to EPR centers produced by irradiation. They are due to the loss or gain of one electron from the SiO44- group. The isocronal decay curves of the SiO45- signal and that of TL peaks are similar indicating a correlation between the EPR and TL centers suggesting us a possible TL mechanism: the Dy3+ looses one electron and converts into Dy4+ during irradiation. The electron liberated is then captured by SiO44- forming the SiO45- EPR center. When the sample is heated in order to read out TL the inverse process takesplace leaving the Dy3+ in excited states. In the transition to the ground state two light emissions are observed, one in 480nm and other in 580nm. In the OA spectrum measuremment two bands due to the Uranium (1100 and 1503nm), one to H2O(1900nm) and one due to the radical OH (1400nm) are observed. These bands are not afected by the irradiation used in this work.

Electron paramagnetic resonance (EPR) dosimetry using the well established dosimeter material alanine is a generally accepted dosimetric method for measurements of high absorbed doses. Alanine EPR dosimetry is however not sensitive enough for high precision measurements of low (&lt; 5 Gy) absorbed doses using reasonably measurement times and small dosimeters. It has therefore not been possible to fully exploit the benefits of EPR dosimetry for applications in radiation therapy. The aim of this thesis was to show that sensitive EPR dosimetry is a competitive method for applications in radiation therapy fulfilling the requirements of measurement precision. Our strategy for reaching this goal was to search for new, more sensitive, EPR dosimeter materials fulfilling the criteria of being tissue equivalent, having a high radical yield and having a narrow EPR spectrum suitable for dosimetry. The best materials were found among formates and dithionates. Doping with small amounts of metal ions and recrystallisation in D2O were tested to further increase the sensitivity. Four promising candidate materials were tested regarding radical stability and dose response and among them lithium formate was chosen for dosimetry in radiation therapy applications. A high precision EPR dosimetry method was developed using lithium formate. The method included the development of a production method for EPR dosimeters with very homogenous shape, mass and composition. A read-out process was developed with maximal measurement precision for reasonably short measurement times. The method also included a dosimeter quality control before actual dose measurements. Measurement accuracy was controlled for every new dosimeter batch. This high precision lithium formate EPR dosimetry method was evaluated for pretreatment verifications of intensity modulated radiation therapy (IMRT) treatment plans. The precision and accuracy was shown to be sufficient (&lt; 5 %) for measurements of doses above 1.5 Gy using one single dosimeter and a measurement time of 15 minutes. The described evaluation is therefore a demonstration of the improved precision at low dose determinations that is available with our sensitive EPR dosimeter materials. While the EPR signal intensity is proportional to absorbed dose, the signal shape is in some cases dependent on the radiation quality. A new method is presented for simultaneous measurements of beam LET (linear energy transfer) and absorbed dose in heavy charged particle beams using potassium dithionate EPR dosimetry. The study shows that when irradiating a dosimeter with 35 MeV carbon ions, the ratio of the signal amplitudes from two radicals in potassium dithionate vary along the track indicating a dependence on linear energy transfer, LET. Potassium dithionate may therefore be a promising EPR dosimeter material for simultaneous measurements of absorbed dose and LET in heavy charged particle radiation fields.

Abstract:
Stroke is a leading cause of morbidity and long-term neurological disability in the U.S. Ischemic stroke, which accounts for approximately 90% of all strokes, is the result of an occlusion in the arteriole cerebrovascular network. No effective treatment options exist to provide neuroprotection from occlusion, and limited success has been seen clinically when attempting to restore blood flow to vulnerable neural tissue regions. Enhancement of pial collateral remodeling (Arteriogenesis) has recently been shown to improve blood flow and mitigate neural tissue damage following stroke (1-3). Arteriogenesis is the remodeling of pre-existing arteriole vessel which are able to re-route blood to blood-deprived regions of tissue. Arteriogenesis requires endothelial cell (EC) and smooth muscle cell proliferation, extracellular matrix degradation and recruitment of circulating bone marrow-derived cells (4-6). Unlike spouting angiogenesis, which requires weeks following occlusion to develop, arteriogenesis begins as early as 24-48hrs post-stroke (7, 8) and can expeditiously enhance blood flow to ischemic regions, making it an attractive target for therapeutic intervention. Our preliminary studies, in an EphA4 global knockout mouse model, indicated that EphA4 receptor tyrosine kinase severely limits pial arteriole collateral formation. The preliminary work also showed that activation of EC EphA4 receptor in vitro inhibited vascular formation. Additionally, ECs lining the collateral vessel have been shown to play a role in collateral remodeling (9). Taken together, the objective of this dissertation was to elucidate the cell autonomous role of the EphA4 receptor and given the central role of the EC in collateral remodeling, we postulated that EphA4 receptor on ECs the limits pial collateral formations. Using a cell-specific loss-of-function approach, we tested the hypothesis that EC-specific EphA4 plays an important role in pial collateral development and remodeling after induced stroke. The results from this dissertation show that (1) EphA4 expression on ECs suppress the formation of pial collaterals during development and limits EC growth via suppression of p-Akt in vitro (2) EC-specific EphA4 ablation leads to increased collateral remodeling, enhanced blood flow recovery, tissue protection and improved neurological behavioral outcomes after stroke and (3) Mechanistically, EphA4 limits pial collateral remodeling via attenuation of the Tie2/Angiopoietin-2 signaling pathway. The work presented in this dissertation demonstrate that EphA4 can be targeted therapeutically to increase pial collateral remodeling to alleviate neurological deficits after ischemic stroke.<br>Doctor of Philosophy<br>Stroke is the fifth leading cause of death in the United States. Ischemic stroke is the most common type of stroke and occurs when blood flow to part of the brain is impeded. Lack of blood results in cell death and tissue damage in the brain. In an effort to restore blood flow, specialized blood vessels in the brain called collaterals remodel and become larger to allow re-routed blood to the blood-deprived region of the brain. The duration it takes to remodel these remarkable blood vessels and re-route blood varies in humans, and sometimes is not able to prevent adequate tissue damage. The current work explores novel therapeutic targets to accelerate collateral remodeling in an effort to reduce tissue loss after stroke. We present studies which show that a protein called EphA4, found on endothelial cells restricts remodeling, and when inhibited in the brain can increase collateral remodeling and reduced adverse effects after ischemic stroke.

The possibility of using high-frequency electromagnetic radiation in the construction sector is a common knowledge. Various experiments have been carried out to dry construction materials, or even to destroy biotic pests. However, the majority of these experiments have been executed in laboratory conditions. It means they have been carried out under exact limiting conditions, often very far off those conditions in real building practice. Therefore, this dissertation aims at exploring the possibility of utilizing microwave technology in drying and heating of construction materials, in disposing of biotic pests and sterilizing building materials under real conditions in building practice. The thesis will explain that microwaves are a term given to a part of electromagnetic radiation. Electromagnetic radiation is based on the principle that the radiated energy is absorbed by water molecules present in moist building material. In the mass of the irradiated material, the radiated energy is transformed into thermal energy, where a so-called “motion” of water molecules occurs with the energy then being transformed into kinetic energy. The microwave technology itself offers a very wide range of use, which can be easily achieved, thus making the technology available to use in the future. Part of the experiments and the necessary measurements, too, has been carried out on selected types of building materials in the premises of the Faculty of Civil Engineering of Brno University of Technology. They have been executed under conditions which most closely correspond to the conditions in real buildings. Part of the experiments and the necessary measurements were carried out on particular constructions, by the so-called in situ method. As stated in the conclusion of the dissertation, laboratory measurements and the in situ measurements differ considerably in some cases.
