Abstract:
L'hématopoïèse est un processus hautement régulé, initié au niveau des cellules souches hématopoïétiques (CSHs) et dont la régulation est assurée à travers une hiérarchie cellulaire. Ceci implique un mécanisme de régulation dynamique des fonctions des CSHs et de leur descendance. Les ERKS sont des acteurs clés de la régulation de la prolifération et de la différenciation dans de nombreux types cellulaires, suggérant un rôle important dans l'hématopoïèse qui n'a pas encore été étudié in vivo. L'étude des souris ERK1 ̄/ ̄ a mis en évidence une splénomégalie, caractérisée par une accumulation de progéniteurs érythroïdes et une augmentation du nombre d'érythroblastes immatures. L'activation de la voie de signalisation BMP4 permet la mise en place d'une érythropoïèse splénique et le développement de BFU-Es de stress. Dans les souris ERK1 ̄/ ̄ nous avons observé une forte augmentation du nombre de BFU-E de stress et de l'ARNm codant pour BMP4. Notre étude nous a donc permis de démontrer que ERK1 régule l'érythropoïèse splénique par son action sur l'expression de BMP4. De plus, l'étude de l'hématopoïèse médullaire a montré une augmentation du nombre de CSHs ainsi qu'un défaut de différenciation granulo-macrophagique dans les souris ERK1 ̄/ ̄. Des radiographies ont permis d'observer une augmentation de la densité osseuse dans des souris ERK1 ̄/ ̄, suggérant une ostéopétrose. Les mécanismes mis en jeu seront caractérisés par des expériences de transplantation de CSHs, par l'analyse de la différenciation monocytaire et mégacaryocytaire et par l'étude de l'ostéogénèse
Hematopoiesis is a highly regulated process, initiated by hematopoietic stem cells (HSCs), which implies a dynamic regulation of the functions of HSC and its descent. The ERKs kinases are keys regulators of proliferation and differentiation of different cells types. Their role could be important in hematopoiesis and have not been studied in vivo yet. We have examined the role of ERK1 in adult hematopoiesis in ERK1 ̄/ ̄ mice. Loss of ERK1 resulted in an enhanced splenic erythropoiesis, characterized by an accumulation of erythroid progenitors and an enhanced number of immature erythroblasts in the spleen. Splenic stress erythropoiesis response has been shown to require BMP4-dependent signalling in vivo and to rely on the expansion of stress BFU-Es. A great expansion of stress BFU-Es and an increased level of BMP4 mRNA were found in ERK1 ̄/ ̄ spleens, suggesting that ERK1 controls a BMP4-dependent step, regulating thé steady state of splenic erythropoiesis. Study of medullar hematopoiesis had also show an enhanced numbers of HSCs and a defect in granulo-macrophagic differentiation in ERK1 ̄/ ̄ mice. X-Rays analysis of wild-type and ERK1 ̄/ ̄ mice allowed us to observe an enhanced bone density in ERK1 KO mice. So, loss of ERK1 induces osteopetrosis. Mechanisms involved will be caracterised by HSCs transplantation experiments, analysis of monocyte and megacaryocyte differentiation and by study of osteogenesis

Abstract:
A osseointegração de implantes de titânio (Ti) é dependente da interação entre a superfície de Ti e células, a qual é modulada por diversas vias de sinalização intracelular. Sabe-se que as quinases reguladas por sinais extracelulares (ERKs), membros da família das proteínas quinases ativadas por mitógenos (MAPKs), atuam tanto na osteogênese, quanto na adipogênese e, portanto, podem estar envolvidas no processo de osseointegração de Ti. Nesse contexto, o objetivo do presente estudo foi avaliar se a interação entre células-tronco mesenquimais (CTMs) e superfícies de Ti usinada e com nanotopografia é modulada, ao menos em parte, por ERK1/2 e o consequente efeito da inibição dessas ERKs na diferenciação osteoblástica e adipocítica. Para isso, CTMs derivadas de medula óssea de ratos foram cultivadas sobre discos de Ti usinados e com nanotopografia em condições osteogênicas e adipogênicas, na presença ou não do inibidor de ERK1/2, PD98059, em concentração previamente determinada (25 μM) e foram avaliados parâmetros relacionados à diferenciação osteoblástica e adipocítica. Os resultados mostraram que a expressão gênica dos marcadores osteoblásticos RUNX2, osterix (OSX), fosfatase alcalina (ALP) e osteocalcina (OC) foi aumentada pela inibição da via de sinalização de ERK1/2 nas células crescidas sobre Ti usinado e apenas ALP e OC, naquelas crescidas sobre Ti com nanotopografia. A expressão proteica de RUNX2 foi discretamente maior nas células crescidas sobre Ti usinado, mas não sobre Ti com nanotopografia, quando ERK1/2 foram inibidas e essa inibição não afetou a formação de matriz extracelular mineralizada, independentemente da superfície de Ti avaliada. Com relação à diferenciação adipocítica, a inibição da via de sinalização de ERK1/2 aumentou a expressão gênica dos marcadores adipocíticos PPARγ, adiponectina (ADIPOQ) e proteína ligadora de ácido graxo do adipócito ( AP2) nas células crescidas sobre ambas as superfícies de Ti, com efeito mais acentuado na superfície usinada, sem afetar a formação de acúmulo lípidico. Em conclusão, os resultados mostraram que a inibição de ERK1/2 favoreceu a diferenciação osteoblástica de CTMs crescidas sobre a superfície de Ti usinada, mas não sobre Ti com nanotopografia. Além disso, a inibição de ERK1/2 favoreceu a diferenciação adipocítica de CTMs crescidas sobre as superfícies de Ti com nanotopografia e usinada, sendo o efeito mais acentuado na usinada. Considerando aplicações terapêuticas, esses resultados são relevantes para direcionar o desenvolvimento de superfícies de biomateriais que atuem em vias de sinalização que sabidamente modulam o processo de osteogênese.
Osseointegration of titanium (Ti) implants depends on interaction between Ti surface and cells, which is modulated by several intracellular signaling pathways. Extracellular signal-regulated kinases (ERKs) are members of mitogen-activated protein kinases (MAPKs) family and act on both osteogenesis and adipogenesis and, therefore, may be involved in the process of Ti osseointegration. In this context, the aim of this study was to evaluate if the interaction between mesenchymal stem cells (MSCs) and Ti surfaces, either machined or with nanotopography, is modulated, at least in part, by ERK1/2 and the effect of ERK1/2 inhibition on osteoblast and adipocyte differentiation. Rat bone marrow MSCs were cultured on Ti discs either machined or with nanotopography under osteogenic and adipogenic conditions, in presence or not of the ERK1/2 inhibitor, PD98059, at a concentration previously determined (25 μM) and it was evaluated parameters related to osteoblast and adipocyte differentiation. The results showed that gene expression of the bone markers RUNX2, osterix (OSX), alkaline phosphatase (ALP) and osteocalcin (OC) was increased by ERK1/2 signaling inhibition in cells grown on machined Ti and only ALP and OC in cells grown on Ti with nanotopography. RUNX2 protein expression was slightly higher in cells grown on machined Ti, but not on Ti with nanotopography, when ERK1/2 signaling was inhibited and such inhibition did not affect extracellular matrix mineralization, irrespective of the evaluated Ti surface. Regarding adipocyte differentiation, ERK1/2 signaling inhibition increased gene expression of the adipose tissue markers PPARγ, adiponectin (ADIPOQ) and adipocyte fatty acid-binding protein (AP2) in cells grown on both Ti surfaces, with more prominet effect on machined one, without affecting lipid accumulation. In conclusion, our results showed that ERK1/2 signaling inhibition favored osteoblast differentiation of MSCs grown on machined Ti, but not on Ti with nanotopography. In addition, ERK1/2 signaling inhibition favored adipocyte differentiation of MSCs grown on both Ti surfaces, being more noticeable on machined one. Considering therapeutical applications, these results are relevant to drive the development of biomaterial surfaces to act on signaling pathways that regulate the process of osteogenesis.

Abstract:
This thesis examines the lives and emotions of male and female ground personnel who served in Bomber Command during the Second World War. Histories of Bomber Command usually focus on the flyers, or on the strategy of the bombing campaign. The experiences of four fifths of the service, the ground personnel, are often neglected. They are frequently reduced to two dimensional stereotypes, as the ‘Erk’ who serviced the aircraft, or the WAAF ‘chop girl’ whose sexual promiscuity presaged death. The RAF assigned personnel to different trades ranging from the most technical to the most mundane. The central theme of this thesis is the gendered hierarchy of trades within Bomber Command. It was constructed in part by widespread beliefs about fear, heroism and stoicism, the interconnected discourses of class and gender, and specific quirks of RAF culture. These included its trade selection process, dialect, pay scale, and trades’ perceived importance to the raison d’être of Bomber Command. The hierarchy is important in explaining the experiences of ground personnel, as some personnel were overstretched while others felt that they were not ‘doing their bit.’ Bomber Command servicemen and women were part of a community that experienced high rates of air crew loss. This thesis also discusses their emotional responses to service life and the treatment by the RAF medical services of those who suffered breakdowns. In examining non-combatant military personnel who served on operational stations but were also part of the home front, my thesis will inform and bring together different areas of study. It gives a voice to an important but previously underrepresented group and in doing so, contributes to the histories of the RAF, emotions, military medicine and psychiatry, as well as understandings of the wartime experience, work and citizenship.

L'interleukine-6 (IL-6) est une cytokine ayant un rôle majeur dans le système immunitaire et dans l'inflammation. Son rôle dans le métabolisme du cartilage est mal connu. Toutefois, il semblerait qu'elle puisse être impliquée à la fois dans les maladies inflammatoires et dégénératives du cartilage (arthrite et arthrose). L'objectif de notre travail a été d'étudier, chez les chondrocytes, les voies de signalisation mises en jeu par l'IL-6 et leur rôle dans ses effets biologiques. Nos résultats montrent que l'IL-6 en présence de son récepteur soluble sIL-6R, active à la fois les voies de signalisation des JAKs/STATs et des MAPK/ERKs. De plus, l'IL-6/sIL-6R inhibe la prolifération des chondrocytes, diminue l'expression de constituants matriciels (collagène de type II, axe protéique de liaison de l'agrécane) et augmente celle de métalloprotéases (collagénase de type 1, agrécanasess de type 1 et 2). Grâce à l'utilisation de parthénolide, qui inhibe l'activation des STATs, nous avons montré que l'IL-6/sIL-6R inhibe la transcription des constituants matriciels par l'intermédiaire de la voie JAK/STAT [etc]

Les MAPKs p42/p44 (ERK) conduisent les signaux extracellulaires de la surface des cellules vers le noyau. Le transfert nucléaire de ERI est requis pour la conduction mitogénique. Nous avons montré que la navette nucléo-cytoplasmique de ERK est continuelle, même dans les cellules quiescentes. L'accumulation dans un compartiment sub-cellulaire dépend de protéines d'ancrage dont l'abondance, l'affinité et la localisation varient pendant la stimulation. Dans les cellules quiescentes, ERK est ancrée dans le cytoplasme par son activateur MEK. ERK s'accumule dans le noyau sous forme inactive au cours des stimulations prolongées. Ceci a été démontré avec des anticorps phospho-spécifiques, en corrélation avec l'état de phosphorylation d'une cible nucléaire de ERK. Les phosphatases responsables de l'inactivation nucléaire de ERK sont néo-synthétisées, spécifiques des tyrosines phosphorylées ou ont une double spécificité. Ces phosphatases se lient très spécifiquement à ERK puisque la micro-injection nucléaire de peptides possédant les sites d'appontage d'interacteurs de ERK, bloque la déphosphorylation nucléaire de ERK. Les phosphatases MKP1/2 sont candidats car induites par l'activation de la voie ERK et leur demie-vie est augmentée par phosphorylation via ERK. Afin de démontrer si MKP1/2 participent à l'ancrage nucléaire de ERK, plusieurs techniques ont été mises en œuvre pour diminuer l'expression des MKPs dans les cellules en culture. Méthodiquement nous avons participé à l'application au laboratoire de la nouvelle technique d'interférence ARN afin de diminue efficacement l'expression de protéines cibles telles que les isoformes de HIF-Proline-Hydroxylases et les MKPs
P42/p44 MAPKs (ERK) conduct extracellular signals from the cell surface to the nucleus to induce biological responses at the gene level. The nuclear transfer of ERKs is essential for mitogenic progression, since their forced cytoplasmic retention blocks the S-phase. Although the nucleo-cytoplasmic shuttling of ERK is permanent, the increased presence of ERK in a sub-cellular compartment is provided mainly anchoring proteins whose abundance, affinity and localization change during stimulation. Thus in quiescent cells, ERK is anchored in the cytoplasm by its activator MEK. The identity of ERK nuclear anchors of remains to be unveiled, however we showed that ERK accumulates in the nucleus in an inactive state during long-term stimulations. Phosphatases responsible for the nuclear inactivation of ERK neo-are synthesized, show a specificity towards phospho-tyrosine or double specificity and interact with specific docking sites. The probable candidates are the MKP1/2 phosphatases. These phosphatases are induced by activation of the ERK pathway and their half-life is increased upon ERK phosphorylation. In order to demonstrate if MKP1/2 participate in ERK nuclear anchoring, several techniques were tested to decrease MKP expression in cell cultures. With the new ARN interference technology, applied very effectively to decrease HIF1 alpha expression, the answer to this important question was to be provided

Abstract:
We worked on finding a new kinase regulator to develop basic data to be used for cancer prevention. Our work found a link between three previously unrelated proteins involved in cancer, ERK2 and ERK6 and Scribble. The MAP kinase cascade is involved in cell proliferation, which is highly deregulated in cancer. Through the screening of ERK6 associating molecules, we found the cell polarity, and cell cycle related molecule Scribble. Furthermore, we found that Scribble was a dual-specific kinase regulator. We clearly demonstrated that these ERKs interact with Scribble through the LRR and the PDZ domains of Scribble. We hypothesize that Scribble may function as a scaffolding protein for ERK2 and ERK6, since Scribble has been found to down regulate the kinase activity of these ERKs.

Abstract:
Proteins of the ERK (extracellular signal-regulated kinases) pathway are known to be involved in the synchronization of the circadian clock. Two atypical members of this family, ERK3 and ERK4, are highly expressed in the brain, yet their exact physiological function remains unknown. In this study, their role was tested via the characterization of animals deficient for either the Erk3 or Erk4 genes. It was discovered that mice lacking Erk4 presented a lower and delayed profile of locomotor activity and under a mixed genetic background, Erk4-/- mice were found to be more sensitive to the masking effects of light. We also discovered behavioural deficits in the Erk3+/- mice, which presented a lower performance in the Morris water maze test, and an increased anxiety phenotype. This study is the first to characterize the behaviour and brain function of mice mutant for these proteins, and will improve our understanding of their physiological role.
Les protéines de la voie des ERK (extracellular signal-regulated kinases) ont entre autres un rôle dans la synchronisation des rhythmes circadiens. Deux membres atypiques de cette famille, ERK3 et ERK4, présentent une forte expression dans le cerveau, mais leur rôle reste inexploré. Dans cette étude, nous avons testé le rôle des protéines ERK3 et ERK4 à travers la charactérisation d'animaux déficients pour l'un de ces gènes. Nous avons démontré que les souris Erk4-/- présentaient un profil d'activité retardé, un plus faible niveau d'activité locomotrice, ainsi qu'une plus grande sensibilité aux effets masquants de la lumière, pour des animaux de fond genetique mixte. Nous révélons aussi que les souris Erk3+/- présentent une plus faible performance au test du Morris water maze, ainsi qu'un phenotype plus anxieux. Cette étude est la première a présenter une charactérization de souris mutantes pour ces protéines, et nous apportera une connaissance approfondie sur leurs rôles physiologiques.

Abstract:
La signalisation intracellulaire permet aux cellules de s'adapter a leur environnement. Certains stimuli declenchent une modification de la concentration de calcium libre dans les differents compartiments de la cellule eucaryote. Ces variations de la concentration de calcium constituent un mode de signalisation qui s'integre au reseau de signalisation intracellulaire constitue par des proteines. Le signal calcique a une grande importance dans la cellule pancreatique beta puisqu'il regule la secretion d'insuline en reponse au glucose. Nous avons chercher a etudier les liens entre le signal calcique et la signalisation proteique dans ce systeme cellulaire. Nous avons montre que les map kinases de type erk, des serine/threonine kinases impliquees dans de tres nombreuses voies de signalisation, sont activees en reponse au glucose. Cette activation depend, comme la secretion d'insuline, de l'entree de calcium par des canaux calciques regules par le potentiel membranaire. L'activation des map kinases par le glucose est regulee par une ou plusieurs isoformes de pkc et par une ou plusieurs tyrosine kinases. Contrairement a ce qui a ete montre dans d'autres types cellulaires, ni l'activite de la tyrosine kinase src, ni celle du recepteur de l'egf ne regulent l'activation des map kinases par le calcium. Nous avons egalement etudie les roles potentiels de l'activation des map kinases en reponse au glucose. Nous avons constate que l'inhibition de leur activation ne modifie pas la secretion d'insuline apres une heure de stimulation. Par contre, les map kinases qui sont rapidement transloquees dans le noyau en reponse au glucose, sont impliquees dans l'activation de la transcription du gene de l'insuline.

Au cours de ce travail, nous avons étudié le rôle de l'acide eicosapentaenoi͏̈que (EPA) et de l'acide docosahexaenoi͏̈que (DHA) dans la régulation de l'activation des MAP kinases / ERK1 et ERK2 des lymphocytes T. Ces acides gras polyinsaturés (AGPI) seuls n'induisent ni la phosphorylation ni l'activité enzymatique des MAP kinases (MAPK). Nous avons stimulé les cellules avec l'anticorps anti-CD3, le phorbol 12-myristate 13-acetate (PMA), ou l'acide okadai͏̈que (AO). L'acide okadai͏̈que semble agir en aval de MEK ou via une voie d'activation indépendante de MEK car U0126 ne diminue pas l'état de phosphorylation des MAPK induit par l'AO. L'application de GF109203X, un inhibiteur de protéines kinases C (PKC) entraîne une inhibition de l'activité des MAPK stimulées par le PMA, ainsi, suggérant que la voie d'activation est couplée à la PKC. En ce qui concerne l'EPA et le DHA, ils diminuent l'activation des MAPK induite par le PMA et l'anti-CD3. D'autre part l'EPA et le DHA inhibent l'activation de deux isoformes de PKC, les PKC alpha et epsilon. Ainsi, ces AGPI régulent la voie de signalisation de ERK1/ERK2 via ces kinases. De plus, les AGPI exercent des effets inhibiteurs sur la translocation nucléaire de NF-kB, de la transcription du gène de l'IL-2 et de la prolifération des cellules T. L'ensemble de ces observations permet de soutenir l'hypothèse selon laquelle l'EPA et le DHA exercent des effets immunosuppresseurs en partie via l'inhibition de la voie de signalisation PKC/MAPK dans les cellules T
In this study, we investigated in T-cells the role of eicosapentaenoic acid and docosahexaenoic acid in MAPK/ERK1/ERK2 regulation pathway. These PUFA alone failed to induce phosphorylation and enzymatic activity of MAPK. In order to ascertain their mechanism of action on MAPK activation we used anti-CD3 antibodies, phorbol 12-myristate 13-acetate (PMA) or okadaic acid (OA) to activate MAPK. PMA and anti-CD3 activate MAPK upstream of MEK as U0126, a MEK inhibitor, abolishes their action. On the other hand, OA seems to act upstream of MEK or via a MEK independent MAPK pathway. Furthermore, GF109203X, a PKC inhibitor, inhibited PMA-stimulated MAPK activity. Also, this pathway is coupled with PKC. EPA and DHA abolished PMA and anti-CD3-induced MAPK activation and the translocation of two PKC isoforms, PKC alpha and PKC epsilon. Also these PUFA regulate the ERK1/ERK2 pathway via these kinases. Their inhibitory effect are responsible for inhibition of NFkB translocation, IL-2 gene transcription and T cell proliferation. Together these observations suggest that EPA and DHA exert immunosuppressives effects in part via the PKC/MAPK pathway in T-cells

Abstract:
Platelet-derived growth factor (PDGF) is a family of signaling molecules that stimulates cell growth, survival and migration. PDGF is recognized by specific transmembrane proteins, the PDGF receptors, which relay the signals to the cell activating the Mitogen-activated protein (MAP) kinases and other signaling pathways. Aberrant activation of these pathways is frequently detected in cancer. Hence, the study of these processes is essential for identifying potential drug targets or diagnostic markers. In paper I, we identified Receptor Subfamily 4 Group A Member 1 NR4A1 to be regulated by PDGF via MAP kinases, clarifying the role of Extracellular signal–regulated kinases (Erk) 1/2, Erk5 and Nuclear factor κ-light-chain-enhancer of activated B cells (NF-κB) in its regulation. NR4A1 was found to be important for the tumorigenic potential, measured as anchorage-independent growth, of glioblastoma cells. Since the cellular responses elicited by PDGF result from the balance between phosphorylation and dephosphorylation events, we investigated the role of the dual specificity phosphatases DUSP4/MKP-2 and DUSP6/MKP-3. In paper II, we describe the crucial role of Erk1/2 and p53 in the expression of DUSP4/MKP2. Moreover, we observed that DUSP4/MKP-2 downregulation decreases Erk5 activation and accelerates PDGFRβ internalization and downregulation resulting in a specific inhibition of Signal transducers and activators of transcription (Stat) 3, Src and protein kinase C (PKC), and partially of p38, Stat1/5 and Phoshoplipase Cγ (PLCγ). In paper III, we report that DUSP6/MKP-3 creates a negative cross-talk between Erk1/2 and Erk5 and an auto-inhibitory feedback loop on the PI3-kinase/Akt pathway. In paper IV, we identify a new regulative mechanism of the PDGF pathway. PDGF induces Erk5 expression and activation that modulates the PDGFRβ activity. After Erk5 downregulation, the receptor undergoes to a faster and stronger activation that results in a faster internalization and degradation. In conclusion, we present a mechanism through which the PDGF/MAP kinases support tumor growth, and elucidate different regulatory pathways involved in PDGF signaling.

Extracellular signal-regulated kinases (ERKs) modulate cellular activities in response to extracellular stimuli and play important biological roles. Thus, perturbed kinase pathways induce pathological conditions, such as tumor development. Rit, a novel member of the Ras family GTPases, activase ERK6, and its over-expression confers tumorigenicity. We hypothesized the presence of scaffolding molecules specific to ERK6, similar to other known MAP kinases. We performed yeast two-hybrid assays using ERK6 as bait, and Scribble was identified as a binding partner. Scribble contains 16 LRR domains and four PDZ domains. We performed immunoprecipitation (IP) assays and discovered ERK2 as another binding partner. Surprisingly, no interaction was observed with the highly homologous MAP kinase, ERK1. No other representative kinases showed binding capabilities with Scribble. IP data confirmed that both ERK2 and ERK6 bind to Scribble through its LRR and PDZ domains. Deletion of ten aminoi acids from the C-terminus of ERK2 and ERK6 abolished these interactions. In vitro kinase assays indicated the kinase suppressing ability of Scribble. Focus formation assays were performed with RitQ79L and H-RasV12 as constitutive activators of ERK6 and ERK2, respectively, in the presence of Scribble. Results confirmed the role of Scribble as a tumor suppressor.

The liver is an essential organ, as certified by its incredible ability of regeneration. Following a loss of hepatic mass, the highly differentiated cells of the liver, the hepatocytes, leave their quiescence and proliferate in order to restore this loss. Many factors participate in this process, by allowing the activation of various signalling pathways. The MAPKs MEK/ERK cascade constitutes one of them. Our objective is to progress in the understanding of mechanisms controlled by the MEK/ERK pathway in the process of regeneration. This work precisely focused on the study of kinases ERK1 and ERK2, which could control specific functions in the healthy hepatocytes and hepatic cancerous cells, in which the pathway is often deregulated. The identification of mechanisms regulating the proliferation of hepatic cancer cells represents a major goal in the development of anti-tumoral treatments

Dear All, Eros moves. In January of 2007 I decided to do research about Eros and his presence in architecture. I decided to do a thesis about LOVE. This thesis it is a story about me, since when you love architecture you give yourself completely to it. What you see in these pages, it's me: my life, my desires, my passion for architecture, my fears, my bad moments, my good moments, my joy--all of me. Desire in architecture seemed to me at that moment something with which I didn’t know how to start working. It was so abstract that, when considered, almost anything can be a desire, and maybe it is. But this thesis is a story of how desire opened for me an infinite world of imagination and wonder--how Eros made me love the drawing, the line, the color, the wall, the shadow, the material....the architecture. I have chosen to explore desire through the designing of a post office, theatre school, and retail shops. The site is in Washington DC, in between 7th and 8th streets SE, adjacent to Eastern Market.

O objetivo desse estudo foi avaliar o efeito do tratamento com a aldosterona (Aldo), por 21 dias, sobre a função e a morfologia renal de ratos, procurando correlacionar as alterações com a expressão do EGFR e da ERK1/2 fosforilados. A Aldo não alterou os parâmetros fisiológicos, a PA e a função renal dos ratos, mas verificou-se uma tendência para o aumento das concentrações plasmáticas de K+. No córtex e na medula renal, a Aldo aumentou a expressão do EGFR e da ERK1/2 fosforilados, sendo que este efeito foi abolido pelo tratamento concomitante com a espironolactona ou o RU 486 (antagonistas dos receptores da Aldo). O tratamento hormonal também induziu um aumento na marcação imuno-histoquímica para essas proteínas no córtex e medula; contudo, este efeito foi reduzido, principalmente, com o tratamento conjunto com a espironolactona. Observou-se um aumento na área glomerular e na porcentagem da área intersticial, após o tratamento com a Aldo. As alterações na morfometria renal foram parcialmente reduzidas pelo tratamento com a espironolactona ou o RU 486. Esses resultados indicam que a Aldo pode induzir alterações morfológicas no rim, aparentemente, de maneira dependente dos receptores MR e GR e da via do EGFR-ERK1/2, mas sem o comprometimento da função renal.
The purpose of this study was to evaluate the effect of 21 days of treatment with aldosterone (Aldo) in the renal function and morphology of rats, correlating changes with the expression of EGFR and ERK1/2 phosphorylated. The Aldo did not alter physiological parameters, blood pressure and renal function of rats, but there was a tendency to increased K+ plasma concentrations. In renal cortex and medulla, Aldo increased the expression of EGFR and ERK1/2 phosphorylated and this effect was abolished by treatment Aldo plus spironolactone or RU 486 (Aldo receptor antagonists). Hormonal treatment also induced an increase in immunohistochemical staining for these proteins in the cortex and medulla; however, this effect was limited mainly to treatment Aldo plus spironolactone. There was an increase in glomerular area and the percentage of interstitial area after treatment with Aldo. Changes in renal morphology were partially reduced by treatment Aldo plus spironolactone or RU 486. These results indicate that Aldo may induce morphological changes in the kidney of dependent manner of MR and GR receptors and EGFR-ERK1/2 signaling, but without the impairment of renal function.

O presente estudo é uma investigação sobre a teoria estética de Herbert Marcuse. A estética em Marcuse está inerentemente atrelada ao estudo do desenvolvimento sócio-histórico da civilização científica e técnica. Segundo ele há uma administração científica das necessidades pulsionais dos seres humanos para a reprodução do status quo, que transforma mercadorias em objetos da libido. Para o autor, a tradição filosófica tratou de separar as faculdades mentais e sensuais dos seres humanos, inferiorizando o que compete aos sentidos em relação ao que compete à razão, criando uma visão bipartida de homem e de mundo. Marcuse busca reunir estas faculdades, que um dia já foram pensadas em conjunto. Desta forma, o autor resgata Eros para pensar uma dimensão estética da existência humana. Cria novos pressupostos para se pensar o retorno à unidade humana e vê na arte um caminho possível para isto se concretizar. Para tanto, vai se apoiar em Freud - pois para ele, Freud percebeu a capacidade dos indivíduos de internalizar e reproduzir inconscientemente a repressão dos impulsos humanos - utilizando conceitos desenvolvidos por ele, tais como princípio de prazer e princípio de realidade para contextualizar sua teoria estética. O filósofo ressalta ainda que o desenvolvimento da tecnologia criou uma racionalidade tecnológica que condiciona ao conformismo e a uma "consciência feliz" gerando uma falsa sensação de bem-estar corroborada pela dessublimação repressiva, característica da sociedade contemporânea, que enfraquece a revolta. O foco de Marcuse está na capacidade da arte de - por estar fundada na fantasia -c onciliar sensibilidade e razão e conduzir-nos para outra realidade, ou outro princípio de realidade, onde o mundo seria predominantemente erótico e prevaleceria a liberdade, a beleza e a felicidade.
This study consists of an investigation on Herbert Marcuse's aesthetic theory. In Marcuse, aesthetics is intrinsically related to the study of the socio-historical development of the scientific and technical civilization. According to him, there is a scientific management of human beings' instinctual needs to reproduce the status quo, which turns merchandise into objects of the libido. For this author, the philosophical tradition managed to divide man's mental and sensuous faculties, diminishing the features related to the senses as compared to those related to reason, thus creating a split notion of man and the world. Marcuse wants to bring these faculties back together, which were once thought of as a unity. And so the author relies on Eros to think of an aesthetic dimension of the human existence. He establishes new assumptions in order to be able to outline a return to human unity and finds in art a possible way for this to happen. He seeks Freud for support - to him, Freud realized the ability individuals have to internalize and unconsciously reproduce the repression of human impulses - and uses concepts developed by him, such as pleasure principle and reality principle, to contextualize his aesthetic theory. The philosopher further points out that the development of technology has created a technological rationality that conditions men to conformity and a "happy conscious", thus creating a false sense of well-being confirmed by repressive desublimation, a feature of contemporary society, which weakens rebellions. Marcuse's main idea lies in the ability of art as a fantasy-based matter - to conciliate sensibility and reason and to lead us to another reality, or another reality principle, in which the world would be essentially erotic and freedom, beauty and happiness would prevail.

Šiuolaikinės technologijos pranoksta visuomenės lūkesčius. Pamažu visuomenė įeina į naują skaitmeninę erą. Šių dienų informacija ir žinios žmones pasiekia akimirksniu. Tradicinės žiniasklaidos žengimas į skaitmeninę erą atvėrė vartus į staigų naujos informacijos perdavimą, virtualų bendravimą, artimesnę sąsają su visu pasauliu. Lietuvoje skaitmeninėmis technologijomis, jomis perduodama tekstine, garso bei vaizdo informacija vis labiau domimasi. Skaitmeninės eros žurnalistika yra aktuali, nauja ir mažai nagrinėta tema Lietuvoje. Tai parodo ir itin skurdus lietuvių literatūros sąrašas apie skaitmeninę mediją. The traditional media‘s step to the digital era opened the new gates to sudden new information transmission, virtual communication, closer connection with all the world. In Lithuania it is more interested in the digital technologies, their transferred textual, audio and video information. The journalism of the digital era is the actual, new and little analyzed subject in Lithuania. It shows a very poor list of Lithuanian literature about the digital media. By doing first steps in the digital era we must familiarize more widely with it not only the digital era‘s users, but also those who hardly understand what it is. Familiarization with the medium of the digital era would increase the number of users and let for a big part of society to use these era‘s advantages. Abstract:
The area of approximate algebraic computations is a fast growing area in modern computer algebra which has attracted many researchers in recent years. Amongst the various algebraic computations, the computation of the Greatest Common Divisor (GCD) and the Least Common Multiple (LCM) of a set of polynomials are challenging problems that arise from several applications in applied mathematics and engineering. Several methods have been proposed for the computation of the GCD of polynomials using tools and notions either from linear algebra or linear systems theory. Amongst these, a matrix-based method which relies on the properties of the GCD as an invariant of the original set of polynomials under elementary row transformations and shifting elements in the rows of a matrix, shows interesting properties in relation to the problem of the GCD of sets of many polynomials. These transformations are referred to as Extended-Row-Equivalence and Shifting (ERES) operations and their iterative application to a basis matrix, which is formed directly from the coefficients of the given polynomials, formulates the ERES method for the computation of the GCD of polynomials and establishes the basic principles of the ERES methodology. The main objective of the present thesis concerns the improvement of the ERES methodology and its use for the efficient computation of the GCD and LCM of sets of several univariate polynomials with parameter uncertainty, as well as the extension of its application to other related algebraic problems. New theoretical and numerical properties of the ERES method are defined in this thesis by introducing the matrix representation of the Shifting operation, which is used to change the position of the elements in the rows of a matrix. This important theoretical result opens the way for a new algebraic representation of the GCD of a set polynomials, the remainder, and the quotient of Euclid's division for two polynomials based on ERES operations. The principles of the ERES methodology provide the means to develop numerical algorithms for the GCD and LCM of polynomials that inherently have the potential to efficiently work with sets of several polynomials with inexactly known coefficients. The present new implementation of the ERES method, referred to as the ``Hybrid ERES Algorithm", is based on the effective combination of symbolic-numeric arithmetic (hybrid arithmetic) and shows interesting computational properties concerning the approximate GCD and LCM problems. The evaluation of the quality, or ``strength", of an approximate GCD is equivalent to an evaluation of a distance problem in a projective space and it is thus reduced to an optimisation problem. An efficient implementation of an algorithm computing the strength bounds is introduced here by exploiting some of the special aspects of the respective distance problem. Furthermore, a new ERES-based method has been developed for the approximate LCM which involves a least-squares minimisation process, applied to a matrix which is formed from the remainders of Euclid's division by ERES operations. The residual from the least-squares process characterises the quality of the obtained approximate LCM. Finally, the developed framework of the ERES methodology is also applied to the representation of continued fractions to improve the stability criterion for linear systems based on the Routh-Hurwitz test.

Abstract:
This thesis revolves around product development in a medium-sized Swedish company. The company in question is called S:t Erik and manufactures a wide range of products made out of concrete and operates in several locations in Sweden. The study has been carried out during 20 weeks with a starting point in January, year 2015. The goal of the study is to deliver a directly implementable development process that support the company's vision and aim towards a more reliable and resource-saving product development process. The study has been conducted through interviews with people directly involved in the product development process. In addition to the interviews, an observational study carried out in the company's production unit in Uppsala and a benchmarking at S:t Eriks plant in Staffanstorp. Which though co-examination with theory revolving the Stage gate process formed the basis for the developed proposals for improvement. To achieve a more accurate product development S:t Erik is recommended to work with its product development on a project basis with clear guidance in the form of a dedicated project manager in combination with a standardized development procedure. Through a proposed model including the work guidelines and though structured development phases S:t Eriks is presented with the opportunity to drastically decrease the risk involved with the company’s product development. The result also includes a proposal to carry out the proposed working model through the online feature in Word, in response to discovered problems revolving communication, access to information and transfer of knowledge. To ensure that S:t Erik is able to interpret and meet the expressed customer need, and at the same time simplify the internal communication they are recommended to incorporate a 3D printer in the early stages of the product development. For additional ways to meet customer needs a concept collection folder available for quick and easy collection of ideas and suggestions online or alternatively on the company intranet is suggested. Finally a procedure for how the working model could be used in practise is presented in an effort to archive a successful implementation at S:t Eriks.

It is only in "erotic matters" that Plato's Socrates is wise, or so he claims at least on several occasions, and since his Socrates makes this claim, it is necessary for Plato's readers to investigate the content of Socrates' wisdom about eros. This dissertation undertakes such an investigation. Plato does not, however, make Socrates' view of eros easy to grasp. So diverse are Socrates' treatments of eros in different dialogues and even within the same dialogue that doubt may arise as to whether he has a consistent view of eros; Socrates subjects eros to relentless criticism throughout the Republic and his first speech in the Phaedrus, and then offers eros his highest praise in his second speech in the Phaedrus and a somewhat lesser praise in the Symposium. This dissertation takes the question of why Socrates treats eros in such divergent ways as its guiding thread and offers an account of the ambiguity in eros' character that renders it both blameworthy and praiseworthy in Socrates' estimation. The investigation is primarily of eros in its ordinary sense of romantic love for another human being, for Socrates' most extensive discussions of eros, those of the Phaedrus and Symposium, are primarily about romantic love. Furthermore, as this investigation makes clear, despite his references to other kinds of eros, Socrates distinguishes a precise meaning of eros, according to which eros is always love of another human being. Socrates' view of romantic love is then assessed through studies of the Republic, Phaedrus, and Symposium. These studies present a unified Socratic understanding of eros; despite their apparent differences, Socrates' treatment of eros in each dialogue confirms and supplements that of the others, each providing further insight into Socrates' complete view. In the Republic, Socrates' opposition to eros, as displayed in both his discussion of the communism of the family in book five and his account of the tyrannic soul in book nine, is traced to irrational religious beliefs to which he suggests eros is connected. Socrates then explains this connection by presenting romantic love as a source of such beliefs in the Phaedrus and Symposium. Because eros is such a source, this dissertation argues that philosophy is incompatible with eros in its precise sense, as Socrates subtly indicates even within his laudatory treatments of eros in the Phaedrus and Symposium. Thus, as a source of irrational beliefs, eros is blameworthy. Yet eros is also praiseworthy. Despite his indication that the philosopher would be free of eros in the precise sense, Socrates also argues that the experience of eros can be of great benefit in the education of a potential philosopher. Precisely as a source of irrational religious belief, the erotic experience includes a greater awareness of the longing for immortality and hence the concern with mortality that Socrates believes is characteristic of human beings, and by bringing lovers to a greater awareness of this concern, eros provides a first step towards the self-knowledge characteristic of the philosophic life
