Full textHematopoiesis is a highly regulated process, initiated by hematopoietic stem cells (HSCs), which implies a dynamic regulation of the functions of HSC and its descent. The ERKs kinases are keys regulators of proliferation and differentiation of different cells types. Their role could be important in hematopoiesis and have not been studied in vivo yet. We have examined the role of ERK1 in adult hematopoiesis in ERK1 ̄/ ̄ mice. Loss of ERK1 resulted in an enhanced splenic erythropoiesis, characterized by an accumulation of erythroid progenitors and an enhanced number of immature erythroblasts in the spleen. Splenic stress erythropoiesis response has been shown to require BMP4-dependent signalling in vivo and to rely on the expansion of stress BFU-Es. A great expansion of stress BFU-Es and an increased level of BMP4 mRNA were found in ERK1 ̄/ ̄ spleens, suggesting that ERK1 controls a BMP4-dependent step, regulating thé steady state of splenic erythropoiesis. Study of medullar hematopoiesis had also show an enhanced numbers of HSCs and a defect in granulo-macrophagic differentiation in ERK1 ̄/ ̄ mice. X-Rays analysis of wild-type and ERK1 ̄/ ̄ mice allowed us to observe an enhanced bone density in ERK1 KO mice. So, loss of ERK1 induces osteopetrosis. Mechanisms involved will be caracterised by HSCs transplantation experiments, analysis of monocyte and megacaryocyte differentiation and by study of osteogenesis

Full textOsseointegration of titanium (Ti) implants depends on interaction between Ti surface and cells, which is modulated by several intracellular signaling pathways. Extracellular signal-regulated kinases (ERKs) are members of mitogen-activated protein kinases (MAPKs) family and act on both osteogenesis and adipogenesis and, therefore, may be involved in the process of Ti osseointegration. In this context, the aim of this study was to evaluate if the interaction between mesenchymal stem cells (MSCs) and Ti surfaces, either machined or with nanotopography, is modulated, at least in part, by ERK1/2 and the effect of ERK1/2 inhibition on osteoblast and adipocyte differentiation. Rat bone marrow MSCs were cultured on Ti discs either machined or with nanotopography under osteogenic and adipogenic conditions, in presence or not of the ERK1/2 inhibitor, PD98059, at a concentration previously determined (25 μM) and it was evaluated parameters related to osteoblast and adipocyte differentiation. The results showed that gene expression of the bone markers RUNX2, osterix (OSX), alkaline phosphatase (ALP) and osteocalcin (OC) was increased by ERK1/2 signaling inhibition in cells grown on machined Ti and only ALP and OC in cells grown on Ti with nanotopography. RUNX2 protein expression was slightly higher in cells grown on machined Ti, but not on Ti with nanotopography, when ERK1/2 signaling was inhibited and such inhibition did not affect extracellular matrix mineralization, irrespective of the evaluated Ti surface. Regarding adipocyte differentiation, ERK1/2 signaling inhibition increased gene expression of the adipose tissue markers PPARγ, adiponectin (ADIPOQ) and adipocyte fatty acid-binding protein (AP2) in cells grown on both Ti surfaces, with more prominet effect on machined one, without affecting lipid accumulation. In conclusion, our results showed that ERK1/2 signaling inhibition favored osteoblast differentiation of MSCs grown on machined Ti, but not on Ti with nanotopography. In addition, ERK1/2 signaling inhibition favored adipocyte differentiation of MSCs grown on both Ti surfaces, being more noticeable on machined one. Considering therapeutical applications, these results are relevant to drive the development of biomaterial surfaces to act on signaling pathways that regulate the process of osteogenesis.

Full textP42/p44 MAPKs (ERK) conduct extracellular signals from the cell surface to the nucleus to induce biological responses at the gene level. The nuclear transfer of ERKs is essential for mitogenic progression, since their forced cytoplasmic retention blocks the S-phase. Although the nucleo-cytoplasmic shuttling of ERK is permanent, the increased presence of ERK in a sub-cellular compartment is provided mainly anchoring proteins whose abundance, affinity and localization change during stimulation. Thus in quiescent cells, ERK is anchored in the cytoplasm by its activator MEK. The identity of ERK nuclear anchors of remains to be unveiled, however we showed that ERK accumulates in the nucleus in an inactive state during long-term stimulations. Phosphatases responsible for the nuclear inactivation of ERK neo-are synthesized, show a specificity towards phospho-tyrosine or double specificity and interact with specific docking sites. The probable candidates are the MKP1/2 phosphatases. These phosphatases are induced by activation of the ERK pathway and their half-life is increased upon ERK phosphorylation. In order to demonstrate if MKP1/2 participate in ERK nuclear anchoring, several techniques were tested to decrease MKP expression in cell cultures. With the new ARN interference technology, applied very effectively to decrease HIF1 alpha expression, the answer to this important question was to be provided

Full textLes protéines de la voie des ERK (extracellular signal-regulated kinases) ont entre autres un rôle dans la synchronisation des rhythmes circadiens. Deux membres atypiques de cette famille, ERK3 et ERK4, présentent une forte expression dans le cerveau, mais leur rôle reste inexploré. Dans cette étude, nous avons testé le rôle des protéines ERK3 et ERK4 à travers la charactérisation d'animaux déficients pour l'un de ces gènes. Nous avons démontré que les souris Erk4-/- présentaient un profil d'activité retardé, un plus faible niveau d'activité locomotrice, ainsi qu'une plus grande sensibilité aux effets masquants de la lumière, pour des animaux de fond genetique mixte. Nous révélons aussi que les souris Erk3+/- présentent une plus faible performance au test du Morris water maze, ainsi qu'un phenotype plus anxieux. Cette étude est la première a présenter une charactérization de souris mutantes pour ces protéines, et nous apportera une connaissance approfondie sur leurs rôles physiologiques.

Full textIn this study, we investigated in T-cells the role of eicosapentaenoic acid and docosahexaenoic acid in MAPK/ERK1/ERK2 regulation pathway. These PUFA alone failed to induce phosphorylation and enzymatic activity of MAPK. In order to ascertain their mechanism of action on MAPK activation we used anti-CD3 antibodies, phorbol 12-myristate 13-acetate (PMA) or okadaic acid (OA) to activate MAPK. PMA and anti-CD3 activate MAPK upstream of MEK as U0126, a MEK inhibitor, abolishes their action. On the other hand, OA seems to act upstream of MEK or via a MEK independent MAPK pathway. Furthermore, GF109203X, a PKC inhibitor, inhibited PMA-stimulated MAPK activity. Also, this pathway is coupled with PKC. EPA and DHA abolished PMA and anti-CD3-induced MAPK activation and the translocation of two PKC isoforms, PKC alpha and PKC epsilon. Also these PUFA regulate the ERK1/ERK2 pathway via these kinases. Their inhibitory effect are responsible for inhibition of NFkB translocation, IL-2 gene transcription and T cell proliferation. Together these observations suggest that EPA and DHA exert immunosuppressives effects in part via the PKC/MAPK pathway in T-cells

Following a loss of hepatic mass, the highly differentiated cells of the liver, the hepatocytes, leave their quiescence and proliferate in order to restore this loss. Many factors participate in this process, by allowing the activation of various signalling pathways. The MAPKs MEK/ERK cascade constitutes one of them. Our objective is to progress in the understanding of mechanisms controlled by the MEK/ERK pathway in the process of regeneration. This work precisely focused on the study of kinases ERK1 and ERK2, which could control specific functions in the healthy hepatocytes and hepatic cancerous cells, in which the pathway is often deregulated. The identification of mechanisms regulating the proliferation of hepatic cancer cells represents a major goal in the development of anti-tumoral treatments

Full textThe purpose of this study was to evaluate the effect of 21 days of treatment with aldosterone (Aldo) in the renal function and morphology of rats, correlating changes with the expression of EGFR and ERK1/2 phosphorylated. The Aldo did not alter physiological parameters, blood pressure and renal function of rats, but there was a tendency to increased K+ plasma concentrations. In renal cortex and medulla, Aldo increased the expression of EGFR and ERK1/2 phosphorylated and this effect was abolished by treatment Aldo plus spironolactone or RU 486 (Aldo receptor antagonists). Hormonal treatment also induced an increase in immunohistochemical staining for these proteins in the cortex and medulla; however, this effect was limited mainly to treatment Aldo plus spironolactone. There was an increase in glomerular area and the percentage of interstitial area after treatment with Aldo. Changes in renal morphology were partially reduced by treatment Aldo plus spironolactone or RU 486. These results indicate that Aldo may induce morphological changes in the kidney of dependent manner of MR and GR receptors and EGFR-ERK1/2 signaling, but without the impairment of renal function.

O presente estudo é uma investigação sobre a teoria estética de Herbert Marcuse. A estética em Marcuse está inerentemente atrelada ao estudo do desenvolvimento sócio-histórico da civilização científica e técnica. Segundo ele há uma administração científica das necessidades pulsionais dos seres humanos para a reprodução do status quo, que transforma mercadorias em objetos da libido. Para o autor, a tradição filosófica tratou de separar as faculdades mentais e sensuais dos seres humanos, inferiorizando o que compete aos sentidos em relação ao que compete à razão, criando uma visão bipartida de homem e de mundo. Marcuse busca reunir estas faculdades, que um dia já foram pensadas em conjunto. Desta forma, o autor resgata Eros para pensar uma dimensão estética da existência humana. Cria novos pressupostos para se pensar o retorno à unidade humana e vê na arte um caminho possível para isto se concretizar. Para tanto, vai se apoiar em Freud - pois para ele, Freud percebeu a capacidade dos indivíduos de internalizar e reproduzir inconscientemente a repressão dos impulsos humanos - utilizando conceitos desenvolvidos por ele, tais como princípio de prazer e princípio de realidade para contextualizar sua teoria estética. O filósofo ressalta ainda que o desenvolvimento da tecnologia criou uma racionalidade tecnológica que condiciona ao conformismo e a uma "consciência feliz" gerando uma falsa sensação de bem-estar corroborada pela dessublimação repressiva, característica da sociedade contemporânea, que enfraquece a revolta. O foco de Marcuse está na capacidade da arte de - por estar fundada na fantasia -c onciliar sensibilidade e razão e conduzir-nos para outra realidade, ou outro princípio de realidade, onde o mundo seria predominantemente erótico e prevaleceria a liberdade, a beleza e a felicidade.
This study consists of an investigation on Herbert Marcuse's aesthetic theory. In Marcuse, aesthetics is intrinsically related to the study of the socio-historical development of the scientific and technical civilization. According to him, there is a scientific management of human beings' instinctual needs to reproduce the status quo, which turns merchandise into objects of the libido. For this author, the philosophical tradition managed to divide man's mental and sensuous faculties, diminishing the features related to the senses as compared to those related to reason, thus creating a split notion of man and the world. Marcuse wants to bring these faculties back together, which were once thought of as a unity. And so the author relies on Eros to think of an aesthetic dimension of the human existence. He establishes new assumptions in order to be able to outline a return to human unity and finds in art a possible way for this to happen. He seeks Freud for support - to him, Freud realized the ability individuals have to internalize and unconsciously reproduce the repression of human impulses - and uses concepts developed by him, such as pleasure principle and reality principle, to contextualize his aesthetic theory. The philosopher further points out that the development of technology has created a technological rationality that conditions men to conformity and a "happy conscious", thus creating a false sense of well-being confirmed by repressive desublimation, a feature of contemporary society, which weakens rebellions. Marcuse's main idea lies in the ability of art as a fantasy-based matter - to conciliate sensibility and reason and to lead us to another reality, or another reality principle, in which the world would be essentially erotic and freedom, beauty and happiness would prevail.

Full textToday‘s society more often faces to technological innovation. Once technological miracles, as the radio, TV, computer, became a reality. Modern technologies surpass society‘s expectations. The society little by little enters the new digital era. The information and knowledge of these days reach the people momentarily. The traditional media‘s step to the digital era opened the new gates to sudden new information transmission, virtual communication, closer connection with all the world. In Lithuania it is more interested in the digital technologies, their transferred textual, audio and video information. The journalism of the digital era is the actual, new and little analyzed subject in Lithuania. It shows a very poor list of Lithuanian literature about the digital media. By doing first steps in the digital era we must familiarize more widely with it not only the digital era‘s users, but also those who hardly understand what it is. Familiarization with the medium of the digital era would increase the number of users and let for a big part of society to use these era‘s advantages. The aim of the master‘s study is to analyze the peculiarities of digital era‘s medium, practical benefit and perspectives. Full textIt is only in "erotic matters" that Plato's Socrates is wise, or so he claims at least on several occasions, and since his Socrates makes this claim, it is necessary for Plato's readers to investigate the content of Socrates' wisdom about eros. This dissertation undertakes such an investigation. Plato does not, however, make Socrates' view of eros easy to grasp. So diverse are Socrates' treatments of eros in different dialogues and even within the same dialogue that doubt may arise as to whether he has a consistent view of eros; Socrates subjects eros to relentless criticism throughout the Republic and his first speech in the Phaedrus, and then offers eros his highest praise in his second speech in the Phaedrus and a somewhat lesser praise in the Symposium. This dissertation takes the question of why Socrates treats eros in such divergent ways as its guiding thread and offers an account of the ambiguity in eros' character that renders it both blameworthy and praiseworthy in Socrates' estimation. The investigation is primarily of eros in its ordinary sense of romantic love for another human being, for Socrates' most extensive discussions of eros, those of the Phaedrus and Symposium, are primarily about romantic love. Furthermore, as this investigation makes clear, despite his references to other kinds of eros, Socrates distinguishes a precise meaning of eros, according to which eros is always love of another human being. Socrates' view of romantic love is then assessed through studies of the Republic, Phaedrus, and Symposium. These studies present a unified Socratic understanding of eros; despite their apparent differences, Socrates' treatment of eros in each dialogue confirms and supplements that of the others, each providing further insight into Socrates' complete view. In the Republic, Socrates' opposition to eros, as displayed in both his discussion of the communism of the family in book five and his account of the tyrannic soul in book nine, is traced to irrational religious beliefs to which he suggests eros is connected. Socrates then explains this connection by presenting romantic love as a source of such beliefs in the Phaedrus and Symposium. Because eros is such a source, this dissertation argues that philosophy is incompatible with eros in its precise sense, as Socrates subtly indicates even within his laudatory treatments of eros in the Phaedrus and Symposium. Thus, as a source of irrational beliefs, eros is blameworthy. Yet eros is also praiseworthy. Despite his indication that the philosopher would be free of eros in the precise sense, Socrates also argues that the experience of eros can be of great benefit in the education of a potential philosopher. Precisely as a source of irrational religious belief, the erotic experience includes a greater awareness of the longing for immortality and hence the concern with mortality that Socrates believes is characteristic of human beings, and by bringing lovers to a greater awareness of this concern, eros provides a first step towards the self-knowledge characteristic of the philosophic life
