Abstract:
Cette description grammaticale de l’afar s’appuie essentiellement sur des données recueillies sur le terrain auprès de très nombreux locuteurs afarophones, vivant à Djibouti, en Erythrée et en Ethiopie. La richesse des données a permis de prendre en compte les variantes dialectales tout au long de la description. Celle-ci s’organise autour de plusieurs parties : système phonologique et phonétique, système nominal, système verbal, et syntaxe de l’énoncé. Cette étude a permis de mieux évaluer l’originalité de certains traits connus mais aussi de mettre en valeur des traits peu connus voire ignorés jusqu’à présent. Citons entre autres : redéfinition du statut de la rétroflexe ; approfondissement des valeurs sémantiques et syntaxiques attachées aux différents procédés de dérivation nominale et verbale ; illustration du rôle primordial de la relative dans une langue où il n’y a pas de catégorie « adjectif » ; étude détaillée de la polarité de genre et de l’accord du verbe en genre non en nombre (accord au singulier avec un nom marqué comme pluriel) ; distinction entre noms adverbiaux et adverbes sur des critères syntaxiques; mise en valeur de la portée syntaxique et sémantique des quatre postpositions; meilleure définition de la catégorie des idéophones sur le plan morphophonologique, syntaxique et sémantique ; mise en lumière des procédés de topicalisation. En apportant un nouvel éclairage sur l’afar, à l’intérieur du couchitique et de la famille afro-asiatique, cette thèse se veut aussi une contribution à la typologie des langues
This grammatical description of Afar is mostly based on fieldwork data collected from a great number of Afar speakers living in Djibouti, Eritrea and Ethiopia. The wealth of data has allowed us to embrace dialectal variations in the description. This description has been built around several components: the phonological and phonetic system, the nominal system, the verbal system, and the syntax.This study leads us to better assess the originality of some features that are already known, and also to bring out other features that were not well known or unknown until now: redefinition of the status of retroflex consonants ; enhanced understanding of semantic and syntactic values of different processes of nominal and verbal derivation ; illustration of the crucial role of the relative clause in a language without adjectives ; detailed study of gender polarity and verb agreement in gender – and not in number – (agreement in the singular with a name marked in the plural) ; differentiation between adverbial names and adverbs on the basis of syntactic criteria ; highlight of the syntactic and semantic significance of four postpositions ; better definition of ideophones from the morphophonological, syntactic and semantic aspects ; highlight of topicalization processes. By shedding new light on Afar within the Cushitic and Afro-Asian family, this thesis aims also to contribute to language typolog*y

Abstract:
La présente étude explore les relations maritimes établies pour des raisons commerciales entre le monde méditerranéen, l’Inde du Sud et Sri Lanka, entre le IIIe siècle av. n.è. et le VIIe siècle de n.è. Il s’agit dans un premier temps d’élaborer une synthèse quantifiée des imports issus du monde gréco-romain d’après les vestiges archéologiques découverts en Asie du Sud, en les confrontant aux autres types de sources. Cette synthèse nous conduit à nous pencher sur les contacts culturels que les liens commerciaux ont favorisé dans leur sillage : transferts, métissages, imitations et appropriations. Dans un dernier temps, ce sont les flux inverses qui ont fait l’objet de notre attention, décelables à travers les objets exportés par l’Inde et Sri Lanka vers les côtes de l’Arabie, de l’Afrique, du golfe Persique et de la mer Rouge. Il apparaît que les témoignages du commerce n’impliquent pas de très grandes quantités échangées mais n’ont pas été dénués malgré tout d’un impact certain sur les sociétés. This dissertation deals with the maritime connections that took place between South Asia (South India and Sri Lanka) and the Mediterranean world between the 3rd c. BCE and the 7th c. CE. It first establishes a global account of the archaeological remains found in South Asia that show the importation of Mediterranean products into this area, by comparison with other types of sources (texts, inscriptions, coins). The study then proceeds towards the social and cultural impact that these imported goods may have had on local populations, with regard to their proper way of appropriating foreign sources of inspiration depending on the regional context. Lastly, attention has been drawn on the return flow of goods from East to West, through archaeological vestiges located on the coasts of Egypt, Africa, Arabia and in the Persian Gulf. This leads to a reassessment of the global quantity of commercial goods crossing this large area, which may have been inferior to what was previously considered, whereas the social and cultural impact is not to be denied. The full picture of these interactions gives an image of a very intricate and complex network, involving lots of intermediaries, middlemen and local networks, which would have created a strong background for the direct long-distance links

Abstract:
Amino acid and DNA sequence comparisons of modular PKSs have shown that ketosynthase (KS) domains are highly conserved. Degenerate primers were designed and used in polymerase chain reactions (PCRs) to amplify the DNA encoding these enzymatic domains from a range of streptomycete genomic templates. Analysis of the PCR products did not only identify a number of known KS domains but included a number of unknown KSs. Using one of these products as a probe to screen a genomic library, a new PKS gene cluster has been identified in Saccharopolyspora erythraea. This new PKS gene cluster, named BOA, has been sequenced. Sequence analysis and database comparisons have led to the elucidation of the modular arrangement of this new cluster and annotation of the flanking genes. Based upon the modular arrangement of this new PKS a putative polyketide product has been proposed. In an effort to overexpress this new PKS and isolate the putative polyketide product a number of compounds that demonstrate bioactivity towards Gram-positive organisms have been discovered. One of these compounds has been isolated and its structure determined via nuclear magnetic resonance (NMR). Results from NMR analysis suggest that this bioactive compound is a tetrapeptide that functions as an ion-chelating siderophore. To confirm the function of domains and modules from the new PKS a range of hybrid PKSs were engineered. These synthases were constructed by substituting modules or domains from the well-studied DEBS 1-TE system with components from the new PKS. In a number of cases expression of these synthases resulted in production of the expected polyketide products. The results confirm the assigned function of the respective domain or module from the BOA PKS and demonstrate the potential use of these components in future engineering of hybrid synthases. A newly discovered gene from S. erythraea that encodes for red-pigment production has been used to develop a novel series of promoter probe plasmids.

A partir de l'identification de plusieurs frontières de l'espace djiboutien (auparavant Côte française des Somalis, puis Territoire français des Afars et des Issas), nous revisitons l'histoire coloniale de Djibouti sous l'angle de la fabrication des territoires. Nous étudions particulièrement cinq frontières que l'on peut tracer sur une carte : les limites ethniques, les limites internes, les frontières internationales terrestres, le corridor ferrovaire et les frontières maritimes. Nous étudions comment ces différentes frontières ont été découvertes ou inventées à Djibouti, dans le cadre général de la formation des espaces dans la Corne de l'Afrique au XIXe siècle, puis comment elles ont été réalisées. Nous analysons les évolutions - dont l'éventuelle consolidation - de ces frontières jusqu'à l'indépendance. Enfin nous en étudions les conséquences et répercussions, en particulier la coercition qu'elles exercent, directement ou indirectement, sur les habitants.

Abstract:
The aim of this study was to identify and measure areas of physiology relevant to the initiation of secondary metabolism and spore formation. It was discovered that the growth kinetics of Saccharopolyspora erythraea in a liquid culture environment were dependent on the type of nutrient limitation the organism was exposed to. Sacc. erythraea was able to produce erythromycin and formed structures which had properties similar to those of spores (designated 'sonic resistant units' in this thesis) in a nitrogen limited medium but, would produce erythromycin but no 'sonic resistant units' in a carbon limited medium. This resulted in two liquid culture systems, one which supported the formation of 'sonic resistant units' and one which did not, and thus enabled the comparison of the conditions which resulted in sporulation or secondary metabolism. The measurement of the rate of DNA, RNA and protein synthesis showed distinct profiles of synthesis in both media. Measurements of these rates in Streptomyces hygroscopicus showed similar profiles in the same media suggesting that the behaviour of the macromolecular synthesis rates in different nutrient limiting conditions might be applicable to streptomycetes in general. A change in macromolecular synthesis rate was implicated in inducing secondary metabolism. Antibiotics were used to modulate these rates in an attempt to induce secondary metabolism. Results from these experiments suggested the possible role of the intracellular concentration of charged and uncharged tRNA in induction of antibiotic synthesis. This relationship implies an analogy between the stringent response and the physiology of the induction of secondary metabolism. The formation of spores ('sonic resistant units') did not appear to be induced by changes in macromolecular synthesis rate or changes in tRNA levels or ratio. However, it was evident that the rates of macromolecular synthesis increased during the production of spores and this implied that it is the action of some intracellular agent that is inducing macromolecular synthesis to fuel the spore forming process.

Abstract:
The objective of this thesis is to use metabolic modelling techniques to investigate primary and secondary metabolism in S. erythraea and from this to identify key factors controlling flux distribution during secondary metabolism. S. erythraea is a member of the actinomycetes a group of bacteria responsible for the production of a number of commercially important small molecules. Actinomycete physiology is considerably more complicated than that seen in "simple" bacteria such as E. coli. The conjecture investigated in this thesis is that metabolic modelling techniques that take into account this extra complexity should be more useful in designing strategies for overproduction of desired metabolites than simpler models. The thesis gives the first detailed description of the dynamic changes in biomass composition seen during the batch cultivation of S. erythraea. It further shows that incorporation of this information into a flux balance model of the organism's metabolism significantly improves the flux distributions generated especially in the stationary phase. Using this improved technique growth phase and stationary phase metabolism are investigated. Some of the unusual stationary phase behaviour is shown to be the result of glucose uptake being independent of demand. Rigid control of branch points in the metabolic network is not found suggesting that the organism's metabolism is flexible. A reverse metabolic engineering strategy is applied, two variants of the wild type organism are compared with an industrial strain. The industrial strain is found to have a considerably lower glucose uptake rate than the parental strain. The relationship between TCA cycle flux, oxidative phosphorylation and organic acid secretion is investigated using an uncoupler. This project demonstrates that applied correctly flux balance analysis is a powerful tool for investigating actinomycete physiology. The insights gained are of direct relevance to the commercial production of secondary metabolites in S. erythraea.

Class of 2005 Abstract
Objective: The goal of this study was to determine the presence of erythro-methylphenidate (erythro-MPH) isomers contained within methylphenidate HCl (Metadate CD®) capsules. Methods: This experiment was conducted at a pharmaceutical manufacturing facility located in Tucson, Arizona. Methylphenidate HCl (MPH) capsules by Celltech Pharmaceuticals, Inc. were analyzed and compared to a reference standard. Thin-layer chromatography (TLC) was the technique used to qualitate the samples. The main outcome measure was the Rf values which were used to determine whether or not the MPH capsules contained erythro-MPH. Results: The study included ten, 20 mg MPH capsules and a reference standard (50 mg strength USP MPH related compound). The Rf value of the reference standard was 0.073 while the Rf value of the MPH samples ranged from 0.42 - 0.85. Conclusion: In this qualitative analysis of MPH capsules, there was no erythro-MPH isomers present in the MPH capsules.

Abstract:
Microscale operations are rapidly emerging as a useful tool for reducing the time and cost involved in fermentation process development. For both microbial and mammalian fermentations pH is a vital process parameter since it has a marked affect on cell growth rate, viability and product synthesis. It is the objective of this thesis to investigate the influence of pH on growth and erythromycin synthesis by Saccharopolyspora erythraea CA340 and implement an automated pH control system at the microscale. The engineering properties of mixing and gas-liquid mass transfer in a 24-well microtitre plate system were characterised. k ,a values up to 0.014 s"1 could be obtained for a soluble complex medium (SCM) and generally increased with increasing shaking frequencies and decreasing fill volumes. Similar trends were identified for measured liquid phase mixing times which were around 5-2284 s under operational conditions. The influence of different fermentation media types oil based medium (OBM), starch based medium (SBM), and SCM and various pH control strategies on growth and erythromycin synthesis by S. erythraea at the 7 L and 5 L scales were next investigated. Faster rates of growth were achieved during fermentations carried out with the SCM and SBM, with the maximum specific growth rate (umax) reaching 0.12 h"1. Growth rates were slower in the more viscous OBM, but erythromycin A titres were double those obtained with the SCM and SBM, however, the long mixing times and high viscosities associated with growth on OBM made this medium unsuitable for microwell fermentations of S. erythraea CA340. At the 7 L scale the implementation of base only or full pH control (NaOH and H3PO4 additions) increased both the maximum growth rate and erythromycin concentrations attained compared to fermentations without pH control. The effects were accurately reproduced in manually pH-controlled microwell fermentations (1000-fold scale translation). The outcome of these studies led to the specification of an automated pH control system at the microscale using a LabView-based software controller integrated with a robotic liquid handling system to monitor and control the pH during microwell fermentations. A specially designed microwell plate was built that allowed the insertion of a micro-pH probe into each well (total well volume 7 mL) and was used during automated pH controlled microwell fermentations. The implementation of base pH control at the microwell scale led to almost one and a half fold increase in both biomass yield and erythromycin A production. It also enabled the operation of a fed-batch process at the microwell scale which further increased erythromycin A production by 60 %. The prototype system thus has the potential to support fermentation process development at the microwell scale.

Abstract:
S. erythraea is the main producing organism of erythromycin, an important broad spectrum antibiotic. Its yield is low because the carbon source utilisation, product kinetics and the regulatory pathways are not yet fully understood. This work provides a detailed characterisation of the carbon source metabolism of S. erythraea. The organism was cultured under various conditions and the growth and product kinetics investigated. In batch bioreactor fermentations, S. erythraea was cultured on both glucose and gluconate as sole carbon and energy sources. The culture was subsequently evaluated in bolus feed addition fermentations using phosphoenolpyruvate, oxaloacetate, propionate and methyl oleate with glucose as main carbon source. On both glucose and gluconate, erythromycin production depended on the nature of the carbon source and the growth rate. On glucose, growth was fast with erythromycin production commencing after the growth phase. On gluconate, growth was subdued and erythromycin production was growth-related. Two phases were distinguished for both carbon sources: 1) erythromycin synthesis and 2) precursor accumulation phase. Erythromycin production was enhanced by increased activity of the pentose phosphate and the anaplerotic pathway. Growth rate and the carbon source uptake rate were found to have a major effect on erythromycin production. The split in pathway activities at key branch points were found to be dependent on the growth rate and the rate of carbon source uptake. During bolus feed addition fermentation in shake flasks, erythromycin levels were increased by the addition of oxaloacetate, propionate and methyl oleate respectively. Then effect of metabolite supplementation on erythromycin levels depended on the phase during which feeding is done. In bioreactor culture, levels of erythromycin were enhanced following addition of PEP or methyl oleate. The pyruvate metabolite node was found to be flexible responding to supplementation and the nature of the sole carbon and energy source. Carbon allocation to pyruvate was 1.7 % and 5.4 % for growth on glucose and gluconate respectively. This metabolite node is, therefore, critical to improving the biosynthesis of erythromycin. However, the phosphoenolpyruvate node appears rigid and addition of PEP was excreted as 7-Orhamnosyl flaviolin (red pigment). The ratio of the rate of carbon source consumption and oxygen uptake is concluded to be a critical parameter in erythromycin biosynthesis.

Abstract:
Il a été mis au point une synthèse asymétrique d'aminosucres en series L-ribose et L-erythrose par réaction de Diels-Alder entre un N-diényl pyroglutamate chiral et un nitrosodiénophile. Cette cycloaddition est fortement sétéocontrôlée. L'influence de différents paramètres (nature des substituants, température, solvant) a été étudiée et d'excellents excès diastéréoisomériques sont obtenus. La suite de la synthèse consiste en une cis-glycolisation stéréospécifique de l'adduit chiral de Diels-Alder. Le diol obtenu est alors réduit en aminosucre, selon diverses methodes. Ce travail a également été effectué en série racémique dans le but de réaliser une mise au point expérimentale optimale. Différentes transformations chimiques des aminosucres ont été étudiées, notamment leur transformation en aminoacides basée sur le principe de la réaction de Kiliani-Fischer. Dans les régions tropicales. Il existe une multitude de systèmes alternatifs avec une grande diversité de pratiques. Ces systèmes sont souvent basés sur une utilisation optimale des ressources animales et végétales locales présentes sur l'exploitation. L'objectif de ce travail est de mieux connaître certaines de ces ressources tropicales disponibles et pouvant potentiellement être intéressantes à utiliser en alimentation porcine dans le cadre de ces systèmes alternatifs de type polyculture - élevage. Les résultats montrent que la capacité d'Ingestion des feuilles de manioc et de patate douce est la même quel que soit la forme de présentation (feuilles fraîches ou en farine). La capacité d'ingestion des feuilles de madère est plus élevée lorsqu'elles sont distribuées en farine (95 vs.40SgIj), inversement, les feuilles fraîches d'érythrine sont mieux ingérées (246 vs. 488 g/j). Les résultats sur la valeur nutritionnelle des feuillages tropicaux montrent qu'ils ont une faible densité énergétique comprise entre 6.5 et 8.2 MJ/kg de MS. La détermination du profil en acides aminés des feuilles montrent que les acides aminés des feuilles de madère et de patate douce sont plus digestibles que ceux des feuilles d'érythrine et de manioc.Au final les teneurs en lysine digestible sont de 5,3 g/kg MS pour la patate et 6,3 g/kg MS pour le madère. En conclusion, l'ingestion, ainsi que la teneur énergétique et protéique des feuilles d'une feuille à l'autre varie en fonction de sa teneur en fibres mais également en fonction de leur concentration en tanins. En pratique, les feuilles de patate douce et de madère sont les plus intéressantes pour la réalisation de ration
In the troples, there are a multitude of alternative systems with wide variety of practices. These systems are often based on an optimal use of local plantand animal on the farm. The objective of this work is to better understand some of these tropical resources avaibable and can potentially be interesting to use in pig feed in the context of these mixed farming system.The results show that the feed intake capacity of cassava and sweet potato leaves is the same whatever the processing form (fresh leaves or meal).The intake capacity of cocoyam leaves18 higher when distributed Into meal (95 vs.408g1d). In opposite the Erythrina fresh leaves are best Ingested (246 va, 488 g 1dl. Results on the nutrltlonal value of tropical foliage show they have low energy density between 6.5 and 8.2 MJ1kg DM. The determlnation of amino acid profile show that the amina acids of cocoyam and sweet potato leaves are more dlgestible than the erythrina and cassava leaves. Ultlmately digestible lysine contents were 5.3 g/kg DM for potato and 6.3 g / kg DM for the cocoyam. ln conclusion, ingestion, and energy and protein content of leaves from one sheet to another depends on its fiber content but aIso according to their tannin. Abstract:
In dieser Arbeit wird das in vivo-Verhalten der murinen Erythro- und Granulopoese, einschließlich der hämatopoetischer Stammzellen, unter dem Einfluß von exogen appliziertem G-CSF und Erythropoietin mit Hilfe eines mathematischen Kompartimentmodelles untersucht. Der Schwerpunkt liegt auf der Identifizierung von linienübergreifenden Wachstumsfaktoreffekten. Zu diesem Zweck werden experimentelle Daten mit den Modellsimulationen unter Berücksichtigung verschiedener Modellannahmen verglichen. Die experimentellen Daten für die Modellentwicklung stammen zum einem kleinen Teil aus der Literatur, hauptsächlich betrachtet wurden jedoch Daten einer kooperierenden niederländischen Arbeitsgruppe. Die beiden Wachstumsfaktoren wurden kontinuierlich und simultan mittels osmotischer Minipumpen subkutan über einen längeren Zeitraum appliziert. Die experimentellen Daten werden zunächst mit Hilfe eines nichtlinearen Regressionsmodelles analysiert und quantitativ beschrieben, wobei Interaktionseffekte zwischen den Wachstumsfaktoren besondere Berücksichtigung finden. Es wird dann ein umfassendes mathematischen Differentialgleichungsmodell der murinen Erythro- und Granulopoese unter Berücksichtigung der linienübergreifenden Wachstumsfaktoreffekte und Interaktionen aufgestellt. Es wird zunächst überprüft, ob sich die beobachteten Daten unter Simultanstimulation durch die einfache Zusammenschaltung zweier bereits existierender Einzelmodelle der Erythro- und Granulopoese ohne weitere Modellannahmen erklären lassen. Dazu werden Daten von normalen als auch splenektomierten Tieren berücksichtigt. Es zeigt sich nach Prüfung verschiedener Hypothesen, dass erst unter Annahme einer durch Erythropoietin potenzierten Amplifikation der primär G-CSF-abhängigen Zellstufe der lienalen CFU-GM die experimentell beobachteten Effekte erklärt werden können. Es wird außerdem gezeigt, daß sich mit demselben Modell und denselben Modellparametern die bei splenektomierten Tieren zu beobachtetende G-CSFabhängige Entwicklung einer EPO-resistenten Anämie gut erklärt wird. In einem zweiten Teil der Arbeit wird ein Modellkonzept erarbeitet, mit welchem sich die Effekte nach Langzeitgabe von G-CSF mittels rezeptorvermitteltem G-CSF-Abbau erklären lassen. In einem dritten Teil wird geprüft, ob sich die hämatopoetische Zellzahldynamik nach Absetzen der G-CSF-Gabe durch eine aktive Rückmigration von Progenitoren aus der Milz in das Knochenmark erklären läßt. Das in dieser Arbeit entwickelte kombinierte Modell der Erythro- und Granulopoese impliziert eine Reihe von weiteren Fragen und bedarf der Überprüfung und Weiterentwicklung anhand weiterer experimenteller Daten. Dafür werden entsprechende Vorschläge erarbeitet, die weitere Einblicke in das komplexe Systemverhalten der Hämatopoese liefern könnten.
