Abstract:

Abstract The aim of this paper is to clarify some important features of a specific text genre called essay. There are discussed some strategies according to which we might successfully write and read the essays, instead of analysing a general concept of essay and traditional searching for the necessary and sufficient features. In this contribution the term of essay is divided into two categories named „academic essay” and „classic essay”. T he main part of this paper focuses on the classic essay that is explained by the triangle whose vertices are represented by „factuality”, „authenticity” and „literariness”. It is demonstrated that in this case we might criticise weakened coherence of the text, but just with the regard to the classic essay as a compromise between truth and elegance.

Abstract:

The author supports the claim that attempts to formulate a universal definition of the term “populism” are not worthwhile, because the sense of the term is usually determined by a specific social context. Understanding the utopian nature of populism provides a better understanding of the utopian nature of democracy and allows for a humble departure from dreams of a perfect social order, because, as has been shown in numerous survey studies, the contemporary shift of social mood, attitudes, and opinions toward some version of populism is a relatively simple consequence of the deficiencies of democracy in its neoliberal version.

Abstract:

Edwardsiella tardais an important Gram-negative pathogen that employs a type III secretion system (T3SS) to deliver effectors into host cells to facilitate bacterial survival and replication. These effectors are translocated into host cells through a translocon complex composed of three secreted proteins, namely, EseB, EseC, and EseD. The secretion of EseB and EseD requires a chaperone protein called EscC, whereas the secretion of EseC requires the chaperone EscA. In this study, we identified a novel protein (EseE) that also regulates the secretion of EseC. AneseEdeletion mutant secreted much less EseC into supernatants, accompanied by increased EseC levels within bacterial cells. We also demonstrated that EseE interacted directly with EseC in a pulldown assay. Interestingly, EseC, EseE, and EscA were able to form a ternary complex, as revealed by pulldown and gel filtration assays. Of particular importance, the deletion ofeseEresulted in decreased levels of EseB and EseD proteins in both the bacterial pellet and supernatant fraction. Furthermore, real-time PCR assays showed that EseE positively regulated the transcription of the translocon operonescC-eseE, comprisingescC,eseB,escA,eseC,eseD, andeseE. These effects of EseE on the translocon components/operon appeared to have a functional consequence, since the ΔeseEstrain was outcompeted by wild-typeE. tardain a mixed infection in blue gourami fish. Collectively, our results demonstrate that EseE not only functions as a chaperone for EseC but also acts as a positive regulator controlling the expression of the translocon operonescC-eseE, thus contributing to the pathogenesis ofE. tardain fish.

Abstract:

This is a metaphysical essay devoted to the phenomenon of parting. The author analyses two events that happened to the Russian poet Marina Tsvetaeva on 31August 1941: parting with her life and parting with her son. A thorough analysis of the poet’s goodbye letter to her son, and in particular the handwritten underlining that is absent from reprints, sheds new light on why the great poet committed suicide. The main subject of discussion, however, remains the phenomenon of separation. The article is dedicated to the memory of Professor Cezary Wodziński, the supervisor of my doctoral dissertation, my teacher and friend. The methodology takes a phenomenological approach and linguistic analysis.

Abstract:

The process of translating literary texts balances on the border of creativity and reproduction, which attracts the attention of researchers and extends the knowledge of translation with new perspectives of reflection on the source text and the target culture. A unique example of this phenomenon comes with essay translation, a genre considered to be hybrid in all its varieties. The borderline area is a changing and complex space where hostility coexists with kindness, enriching the individual also in cognitive, emotional, aesthetic and anthropological aspects. The geographic dimension of the borderline space has not been without influence on the Slovenian cultural identity, which was shaped at the crossroads of several European cultures. This is why the belief exists that translation of Slovenian essays may be a challenge for the translator, just because it focuses on the “in-between” thought mechanism defined by the borderline space. This doctoral dissertation is an attempt to present translation strategies in translation of Drago Jančar’s essayistic texts from Slovenian into Polish. The essays of the Slovenian writer clearly show that the analysed borderline space can be perceived in a thematic, genological and communication perspective, which specifically emphasises their ontological continued existence on the borderline of literature and non-literature and strong foundation in numerous historical, cultural, social and political contexts. The dissertation is also focused on presenting possible difficulties and traps for the translator, which limit the broad spectrum of references present in the original, thus resulting in a change in the perception perspective of the secondary readers. The work draws from the research material of the Polish translations of Drag Jančar’s essays published in the Terra incognita (1993) and Essays (1999) collections, translated by Joanna Pomorska. Majority of the analysed essays come from the first volume, as they touch the discussed problem deepest. The whole dissertation consists of five chapters, the first two of which are dedicated to theoretical problems. The first chapter deals with the definition and semantic problems of the borderline concept. It presents definition-related difficulties and characterises the discussed category, emphasising its proprietary system of values, liquidity, uniqueness of the centreperiphery system, dynamism and dialogism. It also focuses on the borderline experience based on building relationships and defining identity, as well as describes the borderline inhabitant who builds consciousness with the “transversal reason” (according to the Wolfgang Welsch’s concept). The Welsch’s theory not only enhances the life-giving tension, but also emphasises the importance of openness to the world around. The “transversal reason” abandons authority and domination for understanding and co-existence. The second chapter is an attempt to synthesise reflections on what the essay is and what structural rules govern it, and how they are transferred into the other language within the translation process in the perspective of the textual, linguistic and cultural borderline space. The statement that the translation, the borderline, and the essay are all interlinked by the mental mechanism of tension summarises the theoretical considerations. The next three chapters constitute the essential part of the reflection on the problem stated in the title of the work. Their basis is the hermeneutic theory of interpretation that uses the terms of self-consciousness, understanding and experience. Translation of essays has been analysed in the linguistic perspective on the lexical, syntactic and morphological levels; in the pragmatic perspective in view of the different cultural scripts and communication styles; and in the cultural perspective in terms of the transfer of the meaning of the text from one cultural area to another in the perspective of the intertextual relations with other cultural texts. The double number category, lexical, stylistic and stylistic-syntactic solutions, as well as the use of participles, often resulting in changes in sentence structures, are of special interest. The analysed examples confirm the thesis of the different Slovenian and Polish communication styles. The Slovenian style features meandrity and ellipticity, use of imagery, vagueness, digressivity and periphrastia, and it uses complicated arguments and vague reasoning. The Polish style features increased simplicity of expressing content, linearity, economics of meanings and precision in naming. Borderline space is also becoming an important element of intercultural communication, as it significantly affects the communication aspect of translation. It shapes new perspectives, thus strengthening the elements of cultural dialogue in the translation process. It is mostly apparent in intertextual relations, which allow texts to enter dialogues with other texts of culture. As a result, they reinforce the polyphony of the essay, even though they can simultaneously become a trap for the translator. One can notice in the translation that the “in-between” mentality is mostly shaped with the genre form, whereas borderline awareness is exhibited through linguistic and cultural texts. Jančar’s essays are a synthesis of numerous literary, cultural, and social references, which the translation strives to protect, keeping in mind the impossibility of the exact equivalence. The translation employs the hermeneutic “in-between” idea in this case, based on the principle of “linguistic hospitality” referred to by Ricoeur. Its ethical dimension is summarised in overcoming the fear of the unknown and in the acceptance of the fact that perfect translation does not exist.

Abstract:

Predmet istraživanja u ovom radu jeste identifikacija i osvetljavanje dominantnih obrazaca avangardnog i hrišćanskog identiteta u delu Rastka Petrovića. Rastko uvek nastupa sa pozicije nekog ko je blisko upoznat sa umetnošću, istorijom i njenim nepredvidivim i nepravednim političkim projektima. Pokušavajući da rastumači njihove uzroke i posledice, stalno ističe potrebu za integralnim čovekom, kulturom i društvom uopšte, kako po pripadnike sopstvenog naroda, tako i na regionalnom i globalnom nivou. Rastkovo opredeljenje da je svet u nastajanju povezan s ideologijom dečaštva (mladića), znači da je čovek po njemu u događaju Bogomladenca doživeo vrhunac, tj. u potpunosti ispunio cilj ka kome oduvek teži. Suštinsko, a ne tek sporedno, akcidentalno određenje čoveka jeste da nadiđe sebe i stremi ka Bogu. Centralnu ulogu u okviru ove transcedencije trebalo bi da ima analiza apriorne strukture čovekovog bića i u njima prepozna otvorenost i sposobnost da se avangardna/hrišćanska poruka čuje i adekvatno razume. Rastko u tradiciji vidi istinsko uporište transcendencije i afirmacije verodostojnog bića i odgovarajuće slobode gde apsolutna sloboda i apsolutni autoritet nisu suprotstavljena svojstva, već izraz istog bića. Rastkove ideološke evropske pretenzije mogu se okvalifikovati kao veliki intelektualni napor i pregnuće da se stvori, usvoji i inkorporira jedan novi (obnovljeni) kulturni i religiozni mit koji se kao takav projektuje u praistoriju, a potom samoreguliše mnoge savremene fenomene.
The main subject of this PhD thesis was to identify and shed the light on the dominant form of avant-garde and the Christian identity in the work of Rastko Petrovic. Rastko always acted from the perspective of someone who was closely familiar with the art, with history and its unpredictable and unjust political projects. Trying to understand its causes and consequences, Rastko Petrovic has repeatedly stressed the need for the integral and comprehensive man, culture and society in general, both in terms of his own people, and at the regional and global levels. Rastko’s commitment to the idea that the emerging World is connected with the ideology of boyhood, means that the man as an individual has culminated in Divine Child event, and only through that event has fully met the purpose towards which it has always aspired. The essential and not only secondary and accidental determination of man is to transcend himself and strive towards God. An a priori analysis should have the central role within this transcendence of the structure of human beings with its ability to recognize in them the openness and the ability to properly hear and understand the avant-garde / Christian message. Tradition is in the eyes of Rastko Petrovic the only true stronghold of such transcendence and affirmation of one true and credible human being and adequate freedom where absolute freedom and absolute authority are not contradictory properties, but an expression of the one same being. Rastko’s ideological European claims may be characterized as a great intellectual effort and endeavor to create, adopt and incorporate a new (renewed) cultural and religious myth which, as such, projects in prehistory, and then self-regulates many contemporary phenomena.

У овом раду представљен је целокупан књижевни опус Борислава МихајловићаМихиза, једног од најзначајнијих српских критичара друге половине двадесетог века.Михајловићево дело грана се у три главна тока (критика, драма, аутобиографско-мемоарска проза) и два споредна (поезија и антологичарски рад), што његов опус чиниспецифичним и јединственим у послератној књижевности.У првом поглављу дат је његов развојни стваралачки пут, почев од књижевнедружине „Стражилово” у Сремским Карловцима, преко Симине 9а и првих јавнихнаступа на Правном факултету, па до штампања књиге стихова Песме (1947) и Огледа(1951).Друго поглавље посвећено је Михизовим новинским рецензијама објављеним уНИН-у, Књижевним новинама и Политици, током педесетих и шездесетих годинадвадесетог века.У трећем поглављу говори се о његовом боравку у Новом Саду, где је радио каоуправник Библиотеке Матице српске и активно учествовао у културном животу града.Михизов антологичарски рад представљен је у четвртом поглављу, а у петом сеанализирају његови ставови о критици, као и текстови објављени у књигама Од истогчитаоца, Књижевни разговори и Портрети.У шестом поглављу разматрамо Михајловићеву аутобиографско-мемоарскупрозу и указујемо на његов значај као рецезента другим писцима.Седмо поглавље посвећено је његовом раду у театру, почев од првихдраматизација, до оригиналних драма. У овом поглављу дат је и осврт на Михизовепозоришне есеје и критике.У завршном, осмом поглављу, приложена је Михизова целокупна библиографија.
U ovom radu predstavljen je celokupan književni opus Borislava MihajlovićaMihiza, jednog od najznačajnijih srpskih kritičara druge polovine dvadesetog veka.Mihajlovićevo delo grana se u tri glavna toka (kritika, drama, autobiografsko-memoarska proza) i dva sporedna (poezija i antologičarski rad), što njegov opus činispecifičnim i jedinstvenim u posleratnoj književnosti.U prvom poglavlju dat je njegov razvojni stvaralački put, počev od književnedružine „Stražilovo” u Sremskim Karlovcima, preko Simine 9a i prvih javnihnastupa na Pravnom fakultetu, pa do štampanja knjige stihova Pesme (1947) i Ogleda(1951).Drugo poglavlje posvećeno je Mihizovim novinskim recenzijama objavljenim uNIN-u, Književnim novinama i Politici, tokom pedesetih i šezdesetih godinadvadesetog veka.U trećem poglavlju govori se o njegovom boravku u Novom Sadu, gde je radio kaoupravnik Biblioteke Matice srpske i aktivno učestvovao u kulturnom životu grada.Mihizov antologičarski rad predstavljen je u četvrtom poglavlju, a u petom seanaliziraju njegovi stavovi o kritici, kao i tekstovi objavljeni u knjigama Od istogčitaoca, Književni razgovori i Portreti.U šestom poglavlju razmatramo Mihajlovićevu autobiografsko-memoarskuprozu i ukazujemo na njegov značaj kao recezenta drugim piscima.Sedmo poglavlje posvećeno je njegovom radu u teatru, počev od prvihdramatizacija, do originalnih drama. This doctoral thesis is presentation of the whole literary work of Borislav MihajlovićMihiz, one of the most prominent literary critics from the second half of 20th century.Mihajlović’s work is separated in three central segments (criticism, drama, autobiographymemoirprose) and two lateral (poetry and anthology work), which is why it is consideredunique in the post-war literature.First chapter covers development of his creative work, starting from literary clique„Stražilovo” in Sremski Karlovci, via Simina 9a and the first public appearances at theBelgrade’s Faculty of Law, to publishing poetry books „Pesme” (1947) and „Ogledi” (1951).The second chapter is dedicated to Mihajlović’s newspaper reviews published in NIN,„Književne novine” and „Politika”, during 1950s and 1960s.Main topic of the third chapter is his life in Novi Sad, where he worked as a head ofLibrary of Matica Srpska and actively participated in the cultural life of the city.Mihiz’s anthology work is presented in the fourth chapter, whereas fifth chapterincludes analysis of his attitudes toward literary criticism, as well as texts published in hisbooks „Od istog čitaoca”, „Književni razgovori” and „Portreti”.In the focus of sixth chapter is Mihajlović’s autobiography-memoir prose and hisimportance of being critic for other writers.Seventh chapter is about his theatre work, from first dramatisations to original plays.This chapter also contains retrospective view on Mihiz’s theatre essays and critics.In the final, eighth chapter, the whole Mihajlovic’s bibliography is contributed.

Abstract:

This graduation theses in introduction deals with principle and problems of electron signal detection in scanning electron microscopy and charging of insulating specimens in SEM. The experimental part of the thesis describe the methods of qualification and quantification of insulating specimen charging effect observed in environmental scanning electron microscope through the use of ionisation and scintillation detector in dependence on water vapour pressure in specimen chamber. The goal of the thesis is formation of the methodics useful to evaluate charging effect at insulating specimens observing and on the basis of measuring, determine optimal conditions for insulating specimen observation with ionisation and scintillation detector.

Abstract:

This thesis is focused on the environmental scanning electron microscopes including a description of the basic physical mechanisms. It describes the design and realization of the electrode ionization detector system suitable for the detection of signal electrons with a greater proportion of secondary electrons. It also deals with simulations of trajectories of electrons in the electrostatic field of the detector, and with verification of the functionality of the detector in the environment of water vapors in the specimen chamber of the electron microscope.

Abstract:

Истраживање има фокус на хуманим ензимима из суперфамилије алдо-кето редуктаза, које имају велики метаболички значај за хомеостатско функционисање организма. Неки од чланова подфамилије 1С алдо-кето редуктаза (AKR1C) имају улогу у развоју одређених патолошких стања, као што су леукемија, тумори простате,дојке и ендометријума, као и у смањивању ефекта хемотерапија. До сада није регистрован лек који директно утиче на протеине ове групе и самим тим је акценат на изналажењу специфичних лиганада (супстрата, инхибитора), који би могли да имају фармаколошку примену, али и на утврђивању везе између структуре и функције испитиваних лиганада према ензиму. Теза је имала фокус на протеинуAKR1C3. У овој дисертацији је представљена оптимизација ензимског есеја и испитивање потенцијалних лиганада и њиховог ефекта на ензимску активност одређених хуманих изоформи протеина из подфамилије AKR1C. Тестирана су синтетисанa стероиднa jeдињења, комерцијална једињења и биљни екстракти. Стероидни лиганди (AKR-1, -2, -3, -7, -9, -19 и -22) који су показали добре инхибиторне карактеристике су детаљније описани одређеним добијенимкинетичким параметрима и затим су кокристализовани са протеином икофакторм. Од 7 различитих комплекса протеина са најбољиминхибитором, за два комплекса су добијене дифракције са инхибитором и решене кристалне структуре са лигандом у везном месту и врло добром резолуцијом, AKR-7: 1.7 Å, AKR -19: 1.6 Å. Ови резултати представљају прве протеинске кристале чију су структуру решили истраживачи из Србије, а у научном смислу и одличну основу за даљи дизајн и тестирање једињења и кокристализације.
Istraživanje ima fokus na humanim enzimima iz superfamilije aldo-keto reduktaza, koje imaju veliki metabolički značaj za homeostatsko funkcionisanje organizma. Neki od članova podfamilije 1S aldo-keto reduktaza (AKR1C) imaju ulogu u razvoju određenih patoloških stanja, kao što su leukemija, tumori prostate,dojke i endometrijuma, kao i u smanjivanju efekta hemoterapija. Do sada nije registrovan lek koji direktno utiče na proteine ove grupe i samim tim je akcenat na iznalaženju specifičnih liganada (supstrata, inhibitora), koji bi mogli da imaju farmakološku primenu, ali i na utvrđivanju veze između strukture i funkcije ispitivanih liganada prema enzimu. Teza je imala fokus na proteinuAKR1C3. U ovoj disertaciji je predstavljena optimizacija enzimskog eseja i ispitivanje potencijalnih liganada i njihovog efekta na enzimsku aktivnost određenih humanih izoformi proteina iz podfamilije AKR1C. Testirana su sintetisana steroidna jedinjenja, komercijalna jedinjenja i biljni ekstrakti. Steroidni ligandi (AKR-1, -2, -3, -7, -9, -19 i -22) koji su pokazali dobre inhibitorne karakteristike su detaljnije opisani određenim dobijenimkinetičkim parametrima i zatim su kokristalizovani sa proteinom ikofaktorm. Od 7 različitih kompleksa proteina sa najboljiminhibitorom, za dva kompleksa su dobijene difrakcije sa inhibitorom i rešene kristalne strukture sa ligandom u veznom mestu i vrlo dobrom rezolucijom, AKR-7: 1.7 Å, AKR -19: 1.6 Å. Ovi rezultati predstavljaju prve proteinske kristale čiju su strukturu rešili istraživači iz Srbije, a u naučnom smislu i odličnu osnovu za dalji dizajn i testiranje jedinjenja i kokristalizacije.
This research focuses on human enzymes of the aldo-keto reductase superfamily, whose functions have a significant metabolic impact on organism homeostasis. Some members of the 1C aldo-keto reductase (AKR1C) subfamily play role in the development of specific pathological conditions, such as leukaemia, prostate cancer, breast cancer and endometrial cancer, as well as reducing the effectivness of chemotherapy. However, currently there are no approved and registered drugs that directly affect proteins from this subfamily. Therefore our main aim was to screen for specific ligands (substrates, inhibitors) with potential pharmacological applications, and to establish structure-activity relationships for these ligands and enzymes. This thesis mainly focuses on isoform AKR1C3. In this dissertation, optimization of an enzymatic assay and testing of potential ligands and their effects on the enzymaticactivity of specific human isoforms of proteins from subfamily AKR1C are presented. Tested ligands include synthetic steroidal compounds, commercial compounds and plant extracts. Steroid compounds, AKR-1, -2, -3, -7, -9, -19 and -22, were found to be good inhibitors of AKR1C3, and further kinetic studies were conducted. Finally, cocrystalization of protein AKR1C3 with cofactor and these inhibitors was accomplished. From 7 different complexes of protein with inhibitors, two structures were solved to very high resolution, AKR-7: 1.7 Å, AKR -19: 1.6 Å. These results represent the first protein crystal structures solved by researchers from Serbia, and results provide an excellent basis for further design and testing of new inhibitors.

Abstract:

The elder and more experienced certainly know or at least have a vague idea that there used to be a Greek brandy named Botrys containing 40 % of alcohol. Its name was probably derived from the name of Botrytis cinerea (botrytis bunch rot, more commonly). The Greek term is Βότρυς and its transcription into Latin alphabet is Votrus or Votris. However, if a scholar attempts to verify in such an elementary finding, they can get entangled in very complex and tricky historical facts. After weeks of hard work it turned out that it is probably easier to write a chapter on the history of Greek economy of the second half of 19th century than a few lines on a distillery producing a brandy called Botrys. And so this contribution somehow by the way describes a solution to the „raisin problem“ in order to conclude with some basic information on the label Botrys.
