Abstract:
La sindrome di Pendred è, in ordine di frequenza, la seconda causa di ipoacusia su base genetica autosomica recessiva. Si manifesta con un ipoacusia accompagnata dalla presenza di un gozzo tiroideo con eventuale ipotiroidismo. Tali caratteristiche si accompagnano a malformazioni dell’orecchio interno, quali l’acquedotto vestibolare largo. Nel 50% dei casi vi è una mutazione del gene SLC26A4. Riportiamo nel presente lavoro il caso di una paziente portoghese di 47 anni affetta da ipoacusia di grado severo/profondo e ipotiroidismo. La madre e la sorella della paziente, entrambe decedute, erano a loro volta affette da ipoacusia associata a gozzo tiroideo. La risonanza magnetica e la TC hanno entrambe evidenziato un allargamento dell’acquedotto vestibolare e del sacco endolinfatico. La paziente è stata sottoposta a uno studio di GJB2 e GJB6 seguiti da uno screening di tutti gli esoni di SLC26A4 e delle regioni introniche 8 e 14. È stata rilevata, per la prima volta in omozigosi, una mutazione c.918 + 2T>C nella regione intronica 7 del gene SLC26A4. Sequenziando i campioni di controllo è stata rilevata una nuova mutazione c.821C>G presente in eterozigosi nell’estone 7 del gene SLC26A4, per la quale si è ipotizzato un ruolo dannoso. Il presente studio ha condotto alla scoperta di due nuovi genotipi di SLC26A4, e alla miglior definizione degli aspetti fenotipici associati alla sindrome di Pendred.

ESOI for the experimental condition was greater than for the control and within the experimental condition at 12, 16, and 20 min. No other differences were found for the IPT measures. There were no differences for ESOI and IPT between genders.Conclusions:Cryotherapy enables patients to tolerate greater output intensities but does not result in increased peak torque

Abstract:
Methanolic extracts from corpora cardiaca of three moth species, Hippoteon eson (Sphingidae), Imbrasia cytherea (Saturniidae) and Bombyx mori (Bombycidae) show adipokinetic activity in conspecific bioassays. Haemolymph carbohydrates in these moths are not affected. These extracts are also active in heterologous bioassays: haemolymph lipids are increased in Locusta migratoria, whereas a small effect on haemolymph carbohydrates was observed in Periplaneta americana. Therefore, locusts can be used to monitor adipokinetic activity in corpora cardiaca from moth extracts during isolation. The three moth species possess an adipokinetic peptide with the same retention time on reversed phase high performance liquid chromatography (RP -HPLC) as a peptide isolated previously from Manduca sexta, which was code-named Mas-AKH. H. eson contains a second active peak with a similar retention time on RP-HPLC as the hypertrehalosaemic peptide isolated previously from Helicoverpa zea , code-named Hez-HrTH. Both synthetic peptides, Mas-AKH and Hez-HrTH, produce an adipokinetic effect in the three experimental moth species. In H. eson, the haemolymph concentration of Mas-AKH or Hez-HrTH needed to elicit a maximum hyperlipaemic response is about 20 to 30 nᴍ.Flight behaviour in the three moth species is quite different: H. eson is a good hovering flyer, I. cytherea is a comparatively bad flyer and B. mori males show only degenerate flight movements during their mating dance. Haemolymph lipid levels in H. eson decrease drastically during 15 min of flight and return to pre-flight levels in a subsequent rest period. The amount of lipids metabolized during flight is 10.9 mg/gxhr. Haemolymph carbohydrate levels drop during flight, but remain low during the 45 min of recovery. Haemolymph lipids in “dancing” males of B. mori remain constant. In individuals, however, which have low initial lipid levels in the blood, lipid concentrations increase significantly in a subsequent 15 min rest period after “dancing”. Metabolic changes during flight in I. cytherea were not investigated due to this species’ poor flight performance.

Abstract:
P2X7R is a purinergic receptor with broad expression throughout the body, especially in immune system cells. P2X7R activation causes inflammatory mediators to release, including interleukin-1β (IL-1β), the processing and release of which are critically dependent on this ion channel activation. P2X7R’s therapeutic potential augments the discovery of new antagonistic compounds. Thus, we investigated whether the Eugenia sulcata essential oil could block P2X7R activity. The essential oil (ESO) dose-dependently inhibited ATP-promoted PI uptake and IL-1β release with an IC50 of 113.3 ± 3.7 ng/mL and 274 ± 91 ng/mL, respectively, and the essential oil nanoemulsion (ESON) improved the ESO inhibitory effect with an IC50 of 81.4 ± 7.2 ng/mL and 62 ± 2 ng/mL, respectively. ESO and ESON reversed the carrageenan-activated peritonitis in mice, and ESON exhibited an efficacy higher than ESO. The majority substance from essential oil, β-caryophyllene, impaired the ATP-evoked PI uptake and IL-1β release with an IC50 value of 26 ± 0.007 ng/mL and 97 ± 0.012 ng/mL, respectively. Additionally, β-caryophyllene reduced carrageenan-induced peritonitis, and the molecular modeling and computational simulation predicted the intermolecular interactions in the P2X7R situs. In silico, results indicated β-caryophyllene as a potent allosteric P2X7R antagonist, although this substance may present toxic effects for humans. These data confirm the nanoemulsion of essential oil from E. sulcata as a promisor biotechnology strategy for impaired P2X7R functions and the inflammatory response.

Abstract:
Metallothioneins (MTs) are low-molecular weight (6-18 kDa) and sulphur-rich proteins. These proteins have been identified in animals, plants and in both eukaryotic and prokaryotic microorganisms. MTs are involved in the homeostasis of essential metals as well as in the detoxification of the non essential ones. Furthermore they act as scavengers of reactive oxygen species (ROS). This work is part of wider studies on evolutionary aspects of MTs in teleosts. The molecular evolution of MTs in the Antarctic Notothenioid has been investigated. MT cDNA sequences of some members of Nototheniidae, Bathydraconidae, Artedidraconidae and Channichthyidae families have been characterised and evolutionary aspects have been inferred. Maximum likelihood (ML) and Bayesian methods were applied to the coding and UTR sequences. The results indicate the presence of two MT groups, each containing one of the two isoforms, MT-1 and MT-2. Likelihood tests (LH) results support the hypothesis that molecular evolution of notothenioid MTs is based on relaxed clock model. Several discrepancies (mismatches) were observed between MT gene genealogy and species phylogeny. On the basis of these mismatches analyses have been preformed in order to evaluate if positive selection could have been acted. The data indicate the absence of positive selection and suggest the presence of a strong purifying selection operating at the amino acidic level. In addition, phylogenetic analyses on UTRs indicate that 5' UTRs could have been evolved differently from 3' UTRs. Only the 3'UTR of both Notothenia coriiceps MT-1 and Cygnodraco mawsoni MT-2 could have been undergone to convergence events. Furthermore, some MT introns and promoter sequences were characterised. The phylogenetic analyses, based on these sequences, suggest they could be more useful than cDNAs in the study of molecular evolution of notothenioids MTs. These results showed that introns could be generally utilised in evolutionary analyses of nuclear genes.
Le metallotioniene (MT) sono proteine dal basso peso molecolare (6-18 kDa) caratterizzate da un elevato contenuto in cisteine. Esse sono state identificate in animali, piante e microorganismi eucarioti e procarioti. Le MT sono coinvolte nell’omeostasi dei metalli essenziali e nella detossificazione di quelli non essenziali, agiscono inoltre da scavengers delle specie reattive dell'ossigeno (ROS). Questo lavoro si inserisce nell’ambito degli studi sugli aspetti evolutivi di queste proteine nei teleostei. E’ stata quindi analizzata l’evoluzione molecolare delle MT nel subordine Notothenioidei. Sono state caratterizzati i cDNA delle MT di membri delle famiglie Nototheniidae, Bathydraconidae, Artedidraconidae e Channichthyidae. Le analisi filogenetiche sono state condotte sulle sequenze della regione codificante e delle UTR mediante metodi stocastici di maximum likelihood (ML) e baesiani. I risultati indicano che le MT clusterizzano in due gruppi, ognuno dei quali contiene una delle due isoforme, MT-1 e MT-2. I risultati del test di likelihood dimostrano che l'evoluzione molecolare delle MT dei Notothenioidei si baserebbe sul modello dell'orologio molecolare rilassato. Alcune discrepanze (mismatches) sono state osservate nel confronto tra la genealogia dei geni delle MT e la filogenesi delle specie. Sulla base di tali mismatches sono state altresì condotte analisi allo scopo di valutare se su queste MT abbia agito o meno la selezione positiva. Dai risultati ottenuti si può ipotizzare l'assenza di selezione positiva, ma la presenza di una forte selezione purificante che avrebbe operato a livello aminoacidico. Le analisi filogenetiche basate sulle UTR indicano che le 5' UTR si sarebbero evolute diversamente rispetto alle 3' UTR. Inoltre solo le 3' UTR di MT-1 di Nototthenia coriiceps e MT-2 di Cygnodraco mawsoni sarebbero andate incontro a fenomeni di convergenza. Successivamente sono state caratterizzate le sequenze di introni e di promotori di MT per alcuni dei Notothenioidei analizzati. Le analisi filogenetiche basate sulle sequenze introniche e dei promotori suggeriscono che, per lo studio dell'evoluzione molecolare delle MT nei Notothenioidei, tali sequenze sono più informative rispetto alle sequenze codificanti. Tali risultati fanno ipotizzare che le sequenze introniche possono essere proficuamente utilizzate negli studi sull’evoluzione di geni nucleari.

Abstract:
Diagnosis techniques and treatments for Escherichia coli mastitis in the dairy bovine were evaluated in two experiments. The first experiment evaluated eosin methylene blue agar as a method of distinguishing E.coli from other gram-negative mastitis pathogens. Escherichia coli will usually produce a green metallic sheen on eosin methylene blue agar. One hundred and twenty-nine milk samples or gram-negative isolates from milk samples were used to compare eosin methylene blue agar to a commercial biochemical test strip (the accepted standard). There was an intermethod agreement of 96.9% and a k-value of 93.7% indicating excellent agreement beyond chance between test methods. Eosin methylene blue agar is a reliable method for differentiation of E. coli from other gram-negative mastitis pathogens. The second experiment evaluated the efficacy of frequent milk-out as a treatment for E. coli mastitis. Sixteen Holstein dairy cows were divided into 2 blocks and randomly assigned to 1 of 4 treatment groups: 1) non-infected, not frequently milked-out, i.e. not treated (NI-NT), 2) experimentally infected with E. coli, not treated (EC-NT), 3) non-infected, frequently milked-out (NI-FMO), and 4) experimentally infected with E. coli, frequently milked-out (EC-FMO). Hours to bacterial, clinical and systemic cure were not different between the EC-NT and EC-FMO treatment groups. Serum a-lactalbumin concentrations were evaluated between treatment groups as a measure of udder health. Serum a-lactalbumin concentrations were higher in cows in the EC-NT treatment group than cows in the NI-NT, NI-FMO and EC-FMO treatment groups at 12 hours post-experimental challenge. Serum a-lactalbumin concentrations were higher in cows in the NI-FMO treatment group than in cows in the NI-NT, EC-NT and EC-FMO treatment groups at 36 hours post-experimental challenge. Results from this study do not support frequent milk-out as a treatment for E. coli mastitis.

Abstract:
Equatorial Guinea is the only former Spanish colony in Africa south of the Sahara. Consequently, the Spanish-language literature produced by its authors has been resistant to classification in both the fields of Hispanic and African literary studies. This thesis examines a selection of contemporary narratives written between 1994 and 2007 by the following authors: José Fernando Siale Djangany, Maximiliano Nkogo Esono, Juan Tomás Ávila Laurel and Joaquín Mbomio Bacheng. My main objective in this dissertation is to identify, explain and relate the ways in which post-independence authors express identity in their respective texts. In order to accomplish this task, this thesis posits situational interactions as the key sites for identity expression. Developed from the tenets of symbolic interactionism, the syncretic theoretical model of identity views it as telescopic. It is expected that, through the examination of the chosen texts, a contribution can be made to the understanding of the way in which each author expresses identity and can therefore feed into the larger discussion of identity in Equatorial Guinean narrative.

Abstract:
Postcolonial history of Equatorial Guinea is marked by dictatorships: Francisco Macías (1968–1979) first and then Teodoro Obiang (from 1979 r.). The first president murdered one third of the Guinea’s population, mostly those well educated: professors, doctors, engineers, lawyers, writers and artists. The now ruling president is responsible for torturing, intimidating and oppressing the next generations of the Guinea’s people. The state itself is facing a very painful political issues from the point of view of the citizens – traumatic experiences of the dictatorships, no freedom of speech and persecution of the opposition members. The economic issues are caused by corruption and demoralization of the ruling elite which is visible in the wealth’s redistribution of the reach Guinea’s soil and in the lack of the necessities. The social issues are caused by the messy educational and medical systems, as well as by the lack of the proper infrastructure. All of the above became the main topic of the rebellious works by Ramón Esono Ebalé (born in 1977), self-made artist, who in his illustrations, drawings and comic books tries to show the regime’s crimes. The works by Esono which we are about to analyze show how absurd and tragic the Guinea’s reality has become. We read them as an intentional, lucid and consequently projected struggle against the dictatorship.
