Abstract:
This study examines the views that tertiary English teachers hold on the need for English for Specific Purposes (ESP) in Burundi higher education. To do so, it investigates the extent to which they are familiar with the theory and practice of ESP. For this study, 32 English teachers were contacted to participate in it. They were all requested to complete an online questionnaire, but only 17 proved willing to do so. The findings revealed that the majority of teachers are familiar with the practice of ESP. Although many of them reported to have high familiarity with the field of ESP, a few of them conduct a Needs Analysis (NA) before teaching ESP courses. Results also indicated that tertiary English teachers highly acknowledge the importance of teaching ESP in Burundi higher Education and, therefore, would encourage the ministry of education to fund an ESP project.

Abstract:
Message Queuing Telemetry Transport (MQTT) is a connectivity protocol between machines or now better known as the Internet of Things (IoT). This protocol recognizes two basic functions of M2M communication, namely publish and subscribe (pub/sub). The MQTT protocol is designed as a very simple and very lightweight message delivery protocol, designed for devices that are limited and with low bandwidth capacity, high latency or on an unreliable network. The design principles are to minimize bandwidth requirements and device resource requirements, and keep trying to ensure reliability and guaranteed delivery rates. In this paper, VerneMQ performance reliability is tested - one of the MQTT brokers, with several stressing levels using ESP-32 as a publisher and notebook with the python application as a subscriber.

Abstract:
This research aimed at developing a research framework for the emerging field of enterprise systems engineering (ESE). The framework consists of an ESE definition, an ESE classification scheme, and an ESE process. This study views an enterprise as a system that creates value for its customers. Thus, developing the framework made use of system theory and IDEF methodologies. This study defined ESE as an engineering discipline that develops and applies systems theory and engineering techniques to specification, analysis, design, and implementation of an enterprise for its life cycle. The proposed ESE classification scheme breaks down an enterprise system into four elements. They are work, resources, decision, and information. Each enterprise element is specified with four system facets: strategy, competency, capacity, and structure. Each element-facet combination is subject to the engineering process of specification, analysis, design, and implementation, to achieve its pre-specified performance with respect to cost, time, quality, and benefit to the enterprise. This framework is intended for identifying research voids in the ESE discipline. It also helps to apply engineering and systems tools to this emerging field. It harnesses the relationships among various enterprise aspects and bridges the gap between engineering and management practices in an enterprise. The proposed ESE process is generic. It consists of a hierarchy of engineering activities presented in an IDEF0 model. Each activity is defined with its input, output, constraints, and mechanisms. The output of an ESE effort can be a partial or whole enterprise system design for its physical, managerial, and/or informational layers. The proposed ESE process is applicable to a new enterprise system design or an engineering change in an existing system. The long-term goal of this study aims at development of a scientific foundation for ESE research and development.

Cepeda, Sarah Shealy. "Metal-Assisted Hydrolysis of Biological Molecules." Digital Archive @ GSU, 2009. http://digitalarchive.gsu.edu/chemistry_diss/32.

Abstract:
In Chapter I is a general description of novel metal complexes which hydrolytically cleave peptides, proteins, DNA, and other biological molecules. These reagents are becoming the more important as potential therapeutic agents. A panel of ligands was investigated for coordination to ZrIV and other metals in groups 4, 5, and 6 to effect the greatest degree of hydrolysis. Chapter II describes a ZrIV complex which is capable of hydrolyzing a 30 amino acid peptide, insulin chain B, with amino acid specificity. Oxidized insulin chain B peptide was hydrolyzed after only 4 h of treatment at pH 7.0 and 60 °C using ZrCl4 in the presence of 4,13-diaza-18-crown-6. MALDI-TOF and ESI LC-MS mass spectra indicated that insulin chain B was hydrolyzed by ZrIV at the Gly8-Ser9, Ser9-His10, and Gly20-Glu21 amide bonds within the oligopeptide. To our surprise, the cysteine sulfonic acid sequences Cys(SO3H)7-Gly8 and Cys(SO3H)19-Gly20 were also cleaved. To the best of our knowledge, this constitutes the first example of metal-assisted hydrolysis of a Cys(SO3H)-Xaa amide bond. This is significant in light of the fact that cysteine sulfonic acid formation in proteins is triggered by oxidative stress and has been associated with amyloid fibril formation, Parkinson’s disease, and other deleterious, physiological processes. Chapter III describes the metal-assisted hydrolysis of sphingomyelin which is a principle phospholipid component of animal cell membranes. The sphingomyelin assays showed evidence of metal-assisted hydrolysis after 20 h of treatment at lysosomal pH 4.8 and cytosolic pH 7.0 at both physiological temperature 37 °C and 60 °C. The metal ion CeIV was the most reactive, followed by ZrIV, and then HfIV. The goal of this work is to develop metal-based reagents to reverse the lethal build-up of sphingomyelin that occurs in lysosomes of patients suffering from Niemann-Pick disease.

Abstract:
The CCAAT enhancer binding protein beta (C/EBPβ) transcription factor regulates differentiation, proliferation, and functionality of many cell types, including various cells of the immune system. A detailed molecular understanding of how C/EBPβ directs alternative cell fates remains largely elusive. Ectopic expression of C/EBPβ has been previously shown to reprogram committed B cell progenitors into inflammatory macrophages. We took advantage of this reprogramming system in order to examine how C/EBPβ regulates (trans)differentiation. To determine which C/EBPβ protein modules are important for reprogramming, C/EBPβ wild type isoforms and mutants were ectopically expressed in primary mouse B cell progenitors. The data showed that the translationally regulated long isoforms LAP* and LAP, but not the N-terminally truncated isoform LIP can reprogram lymphoid cells into myeloid cells. Furthermore, we found that conserved regions 2,3 and 4 in the C/EBPβ protein transactivation domain are necessary and sufficient for B-to-myeloid cell conversion. Interestingly, the reprogrammed myeloid cells were found to represent a heterogeneous mixture of different myeloid cell types. Detailed analyses of the reprogrammed CD11b+ cells revealed that discrete conserved regions in C/EBPβ activated distinct pro- and anti-inflammatory genes and triggered divergent differentiation programs. Moreover, not only structural C/EBPβ mutants, but also post-translational modification (PTM) site mutations led to different reprogramming outcomes. These data suggest that C/EBPβ orchestrates myeloid diversification by integrating PTMs with structural plasticity as signal dependent adaptable modular properties to determine cell fate.

Full text
Abstract:
The proteasome system of the cell is responsible for the degradation of proteins and plays a central role in the generation of epitopes which are presented to cytotoxic T-lymphocytes (CTLs) on MHC-class-I molecules. The stimulation of cells by interferon-gamma (IFNgamma) leads to the formation of immunoproteasomes that show an improved generation of many MHC-class-I epitopes compared to constitutive proteasomes. In healthy mice, immunoproteasomes are mainly expressed in the lymphatic tissues, whereas the non-lymphatic organs predominantly contain constitutive proteasomes. In this project the effect of Listeria monocytogenes infection on murine 20S proteasomes derived from the liver, spleen, small intestine and colon were investigated. The structure of the isolated proteasomes was analyzed by two-dimensional gel electrophoresis and western blots while the function was studied by in vitro processing of three oligomeric peptide substrates. Identification and quantification of the processing products was performed by HPLC-ESI-ion trap mass spectrometry. The project showed for the first time, that after infection 20S proteasomes isolated from non-lymphatic organs as well as from non-lymphatic cells displayed structural and functional plasticity: immunoproteasomes were induced post infection which could be correlated with the enhanced generation of immuno-relevant fragments. This was independent of the direct presence of Listeria monocytogenes in the organs and solely controlled by the cytokine IFNgamma. In addition, an increased posttranslational modification with the monosaccharide N-acetylglucosamine could be detected in liver-derived proteasomes after infection. Furthermore, a detailed analysis of the mass spectrometry data was established according to the cleavage site usage of constitutive and immunoproteasomes. The result was that immunoproteasomes are involved in improved generation of the immuno-relevant fragments by the faster cleavage and the changed usage of existing cleavage sites after infection.

Full text
Abstract:
Lipopolysaccharide (LPS) binding protein (LBP) is an acute phase protein with the ability to bind and transfer LPS of Gram-negative bacteria. This soluble pattern recognition molecule represents an important defense principle of the host. Regulation of the hepatic acute phase response and its termination are important mechanisms for limiting systemic inflammatory activity of the host. Here were analyze the cooperation of Interleukin (IL)-1, IL-6, and Dexamethasone (Dex) at LBP expression in the hepatoma cell lines HuH-7 and Hep G2. The major inducer of LBP expression is IL-6. Within the LBP promoter numerously highly consensus binding sites such as AP-1, C/EBP-beta? and STAT3 are present, that confer transcriptional activity as shown by truncation and mutation experiments. Additionally, activate NF-kappaB sites activated by IL-1 were detected at the LBP promoter. By mutation experiments of the promoter furthermore were found differentially active glucocorticoid response elements (GREs). The promoter contains GREs enhancing the activity as well as inhibitory ones. The enhancing effect towards LBP expression by Dex was mediated by IL-6. Dex stimulated the expression of the IL-6 receptor and therefore upregulated the IL-6 pathway. Transforming Growth Factor (TGF)-beta is able to inhibit LBP expression in stimulated cells. An AP-1 binding site was identified mediating inhibitory TGF-beta effects towards LBP promoter activity. Furthermore it was shown that a growth factor independence (Gfi)-1 binding site localized near the AP-1 site is essential for mediating the TGF-beta inhibitory effect. The relevancy of the Gfi-1 site fore mediating TGF-beta effects indicates a novel mechanism for understanding inhibitory TGF-beta effects at the transcriptional level. In summary the complex regulation of LBP were elucidate which may help to eventually develop novel intervention strategies for acute phase, sepsis, and septic shock.

Abstract:
The growing demand for traditional bicycle and e-bikes is an opportunity for the market of locking and antitheft systems for these types of vehicles. This dissertation intends to study the smart padlocks market, understand its strengths and limitations; create an intelligent padlock concept that tries to fill the gaps in this market, considering the customer’s needs; and finally, perform the mechanical design and automation of the system. The benchmarking of smart padlocks allowed us to understand the main characteristics of these locking systems, advantages, limitations and extras. User’s main features were identified and, using the Mudge diagram, these needs were ranked in descending order of value to the client. The QFD diagram was used to identify critical features for the customer, specify the correlation between customer needs and project requirements, and order the project requirements in descending order of improvement priority. The generated padlock’s concept is easy to mount on a typical bike, with high compatibility between bicycles and without the need for an external object to lock it, with a worm drive locking mechanism. The communication between the lock and the user is made by Bluetooth and GSM/GPRS to allow a global reach. The bicycle can be unlocked remotely, unlocking the padlock with the user’s approach or manual, in case of lack of battery. The application makes it possible to lock and unlock the bike, allowing it to be shared with third parties; tracks statistics such as the distance travelled, average speed and calories burned and can track the bike by GPS.The mechanical dimensioning of the worm drive system allowed the determination of the parameters of the worm wheel and the worm. The worm wheel has a prime diameter of 133 mm, with an outer diameter of 136 mm and an inner diameter of 129 mm, with 76 teeth. The worm has a prime diameter of 10 mm, an outer diameter of 13 mm and an inner diameter of 6 mm. The maximum worm length is 34 mm and the minimum length is 18 mm.The research and development in this field allows the improvement of smart padlocks, increasing security and alternatives available on this market, providing a better and more personalized experience for the user.

Abstract:

Abstract:
Abstract The non-Newtonian behavior of oil/water emulsion in the ESP stage is still not well understood. The industry relies on existing empirical correlations, which only valid for production pipelines without considering the effect of shear force acting on the system. This paper will present the analytical modeling of emulsion rheology in the ESP stage and its effect on ESP boosting pressure. An extensive experimental data set validates the analytical model accuracy. The Brinkman empirical correlation is the baseline of the analytical model development. Emulsion rheology in the ESP stage depends on many factors. Dimensionless analysis by the Buckingham-Phi theorem indicates that at least three parameters play an essential role in the emulsion rheology at the pump stage. Those parameters are concatenated and applied as the modified function of Brinkman empirical correlation. In addition, the pump boosting pressure performance observed experimentally to study the emulsion rheology effect at the ESP. More than a thousand experiment data points employed to test the proposed model, and its comparison is studied statistically. The dimensional analysis prevails that the turbulence effect at the stage condition reflected by the Reynolds number, the droplet size effect represented by the Weber number and the Strouhal numbers relates to the shearing effect due to impeller rotation. The analytical model and experiment perform with two different oil viscosity, 45 cp and 70 cp. The results reveal that the higher oil viscosity reaches the inversion point at a lower water fraction since the turbulence decreases with higher oil viscosity. The emulsion rheology from the experiment result shows a significant increase of emulsion viscosity at water fraction close to the inversion point since the increase of hydrodynamic forces due to a higher number of water droplets. The emulsion rheology model aligns with the experiment results for the inversion point at around 35% and 32% water-fraction, respectively. The emulsion rheology model shows a good agreement with the experimental data with a 15% standard deviation of relative error. Increasing water fraction up to the inversion point deteriorates pump boosting pressure since the high friction loss occurs due to higher emulsion viscosity. Nevertheless, as the water fraction passes theinversion point, the boosting pressure starts to rebound as the water turns into the continuous phase. The formation of oil/water emulsion in the ESP is inevitable during production operation and consequently affects the pump boosting pressure. The inversion point phenomena occur at a different range of water fractions for different oil viscosity. A better understanding of emulsion rheology at the pump stage will lead to an accurate artificial lift design and eventually avoid operation failure during production well operation.

Abstract:
Abstract This paper describes a new electrical submersible pump (ESP) design concept to overcome the challenges of applications in slim well completions or thru-tubing deployment. The housing of the conventional pump is removed, allowing the pump impellers to have a larger diameter. The impact of this design change on pump hydraulic performance is assessed in this paper. Downhole ESPs operate in environments where space is limited radially. This is especially the case for slim completions or for thru-tubing rigless deployment. To provide the required rate and total dynamic head, the current approach is to use permanent magnetic motors and operate the slim systems at rotational speed over the conventional speed of 3500-4000 RPM. High-speed operations require new pump stage designs to minimize erosion and vibration. This paper provides an alternative pump design, which removes the pump housing with the benefit of increasing the impeller tip diameter, and hence potentially reducing pump length and operational speed. To ensure the pump retains the well fluids, the diffusers are designed to be externally threaded with an O-ring feature. The centrifugal pump affinity laws are applied to evaluate the impact of removing the pump housing and increasing the impeller outside diameter. A typical ESP housing wall thickness is about 0.18-0.25 inch. With the housing removed, the incremental space available for the impeller tip to occupy is increased by 0.36-0.5 inch. Analysis shows that, for the same pump speed as a conventional pump with a housing, a housingless pump will increase the head generated by 23-32%, and the rate capacity about 36-51%, depending on the pump series. In general, the smaller the pump outer diameter, the greater the flow and head capacity increase. This is because the available space due to removing the housing becomes a considerable size of the impeller tip diameter for the smaller series pumps. The elimination of pump housing enables impellers with a larger diameter to be used to generate more head per stage. In comparison to a conventional pump of the same outside diameter, and providing the same amount of total dynamic head, the housingless pump can have fewer stages and a shorter length or operate at a reduced speed. The reduced length can help mitigating pump-bending stress for installation in deviated or horizontal wells. The reduction in required operating speeds will reduce pump wears, heat generation and vibration. The housingless ESPs have applications for slim well completions or thru-tubing deployments.

Abstract:
Centrifugal pump performance is affected when pumping viscous liquids, requiring a larger power input than the same pump handling water. In applications of chemical, civil, environmental, and mechanical engineering that involve centrifugal pumps, it is a challenge to accurately estimate and even more of a challenge to improve their performance when handling viscous liquids. When accurate performance data is needed, difficult experiments must be conducted with the operating viscous flow. The extension of the applicability of numerical techniques for solving fluid dynamics (CFD) permits the consideration of these tools as a definite possibility for predicting the performance of centrifugal pumps with viscous flows. The purpose of this study is to perform a 3D-CFD steady-state simulation of three different configurations of centrifugal pumps. The first is an impeller-diffuser pump (ns = 19) taken from an ESP model. The second is a Francis Pump-Turbine (ns = 28). Finally, the third configuration possesses an impeller and volute (ns = 32). The objective is to characterize and evaluate their performances with four different fluids from 1 to 420 cSt. These are: water at 25°C, SAE10 and SAE30 oils, and Fuel Oil Medium (FOM). For water flow conditions, the numerical results were compared with experimental data, and found to be consistent with global performance parameters. With regard to the higher viscosity fluids, the CFD calculation was compared with those obtained through the standard empirical method (ANSI/HI9.6.7). This resulted in good agreement between the performance results. The commercial software ANSYS-CFX was used for the CFD calculations. The resulting pump performance curve (head, hydraulic efficiency and power output) is consistent with that expected by theory. In general, as the viscosity of fluids increases, the hydraulic energy losses increase. Of the three pumps, slip factor for SAE30 oil was larger for all volumetric flows since it features the best guidance of the flow in the impeller blade passage. For the ns32 pump and the pump-turbine ns28, the volute losses rose from water to FOM, just like the impeller hydraulic losses. For these two turbo machines, the impeller losses were larger than volute losses. For the pumps with volute, the effects of fluid viscosity on the radial forces were evaluated. It was found that the radial forces decrease when the viscosity increases. This paper attempts to contribute to a better understanding of fluid dynamics within centrifugal pump impellers handling viscous fluids, and intends to shed more light on the approaches that performance prediction models should follow in the future.

Abstract:
Abstract In this work, a 3D, one-eighth homogenous fuel CFD model of the TN-32 UNF cask is constructed, and simulations are performed as part of the Extended Storage Collaboration Program (ESCP). The simulations are carried out using ANSYS/FLUENT1 package with geometry and boundary conditions recommended by ESCP. Simulation results, including external and internal cask temperatures, are compared with experimental data from the High-Burnup (HBU) project. A preliminary sensitivity analysis is conducted to determine the effect the different assumptions/simplification proposed by ESCP on the cask temperatures. This sensitivity analysis indicates that the cask internal natural convection has a significant effect on the prediction of internal temperatures when the drain holes at the base of the basket are modeled. Additionally, the results indicate that a small variation in the thickness of the peripheral basket-rail gap has a significant effect on the internal temperatures. The best simulation model is shown to include modeling of the drain holes, natural convection, a peripheral basket-rail gap of 4.78 mm (0.188 inch), and omission of the base inner liner-gamma shield gap.

Abstract:
Invasive exotic plant (IEP) species are a significant threat to natural ecosystem integrity and biodiversity, and controlling them is a high priority for the National Park Service. The North-ern Colorado Plateau Network (NCPN) selected the early detection of IEPs as one of 11 monitoring protocols to be implemented as part of its long-term monitoring program. From May 30 to June 1, 2019, network staff conducted surveys for priority IEP species along the Scenic Drive and Cathedral Valley Road monitoring routes at Capitol Reef National Park. We detected 119 patches of six priority IEP species along 34 kilometers of the two monitor-ing routes. There were more patches of IEPs, and a higher percentage of large patches, than in previous years. This indicates that previously identified infestations have expanded and grown. The most common (47.1%) patch size among priority species was 1,000–2,000 m2 (0.25–0.5 acre). The vast majority (93.2%) of priority patches ranked either low (58.8%) or very low (34.4%) on the patch management index scale. Tamarisk (Tamarix sp., 72 patches) was the most prevalent priority IEP species. African mustard (Malcolmia africana, 32 patch-es), field bindweed (Convolvulus arvensis, 9 patches), and Russian olive (Elaeagnus angusti-folia, 3 patches) occurred less commonly. Together, these four species represented 97.5% of all patches recorded in 2019. Four IEP species were found on the monitored routes for the first time: Russian olive (Elaeagnus angustifolia), quackgrass (Elymus repens), Siberian elm (Ulmus pumila), and African mustard (Malcolmia africana, not on the priority species list before 2019). Cathedral Valley Road had higher IEP priority patches per kilometer (5.68) than the Scenic Drive (2.05). IEP species were found on 37.9% (25 of 66) of monitored transects. Almost all these detections were Russian thistle (Salsola sp.). Russian thistle was widespread, present in 33.3% of transects, with an estimated cover of 0.2% across all transects sampled. Across routes monitored in all three rotations (2012, 2015, and 2019), Russian thistle has increased in frequency. However, its frequency remained about the same from 2015 to 2019, and percent cover remains low. Tamarisk and field bindweed have both increased in preva-lence since monitoring began, with tamarisk showing a dramatic increase in the number and size of patches. Immediate control of tamarisk and these other species is recommended to reduce their numbers on these routes. The NCPN plans to Capitol Reef in 2020 to monitor Oak and Pleasant creeks, completing the third rotation of invasive plant monitoring.
