Abstract:
Endothelin-1 (ET-1) is a powerful vasoconstrictor that contributes to blood pressure elevation. The biological effects of ETs are mediated by two receptors, namely, endothelin type A receptor (ETAR) and endothelin type B receptor (ETBR). Chinese herbal medicines (CHM) with antagonist activity for these two receptors were screened by establishing stable clones of CHO-K1 cells expressing high levels of human ETAR and ETBR, namely CHO-ETAR and CHO-ETBR.The aqueous extract of Prunellae Spica (P1) inhibited the binding of 125I-ET-1 to ETAR and ETBR in CHO-ETAR and CHO-ETBR cells, respectively. P1 suppressed the ET-1-induced mobilization of intracellular Ca2+ . Through the alcohol fractionation of P1, the antagonists of human ETAR and ETBR were found to belong to different, separable ingredients and the antagonist of ETAR is more soluble in alcohol. The two antagonists were also effective in the test on human primary cells, HASMC and HUVEC. P1 successfully prevented the development of ET-1-associated hypertension in rats without further purification. These results indicate the presence of anti-hypertensive ingredients in P. Spica extract, at least through the inactivation of ETAR and/or ETBR.

Abstract:
The aim of this study was to evaluate the role of endothelin-1 (ET-1) in the sensitivity of ovarian carcinoma to paclitaxel, one of the most common drugs used for the management of this tumour histotype. ET-1 is a powerful mitogenic peptide produced by ovarian carcinomas and it acts as an autocrine growth factor, selectively through ETA receptor (ETAR), which is predominantly expressed in this tumour. OVCA 433 and HEY, two ovarian carcinoma cell lines, which produce elevated amounts of ET-1 and express abundantly high-affinity ETARs, were used. As demonstrated by sub-G1 peak in DNA content histograms and terminal transferase deoxytidyl uridine end labelling assay, we found that paclitaxel induces cytotoxic effect through the activation of apoptosis in both cell lines. When the treatment with paclitaxel was performed in association with ET-1, paclitaxel-induced apoptosis was inhibited. In order to evaluate which ET-1 receptor mediated the effect of ET-1 on protection from paclitaxel-induced apoptosis, we performed experiments using two selective antagonists for ETAR (BQ-123) and for ETBR (BQ-788). We showed that ETAR blockade inhibits the ET-1-induced survival activity against paclitaxel-mediated apoptosis. However, no effect was observed on blocking ETBR with BQ-788. Our results establish a novel role for ET-1 in determining survival of ovarian carcinoma cells and suggest that pharmacological ETAR blockade using a specific ETAR antagonist may provide a novel approach to the treatment of ovarian carcinoma in combination therapy.

Abstract:
This study focuses on the impact of aliskiren and (or) glucagon-like peptide-1 analogue on the binding affinity/regulation of endothelin-1 (ET-1) to its receptor subtypes A (ETAR) and B (ETBR) at the level of the coronary endothelium and the cardiomyocytes in a type-1 diabetic rat model. Seven groups were used: (i) normal rats, (ii) rats with induced diabetes, (iii) rats with induced diabetes that were treated with insulin, (iv) rats with induced diabetes that were treated with exendin-4, (v) rats with induced diabetes that were treated with aliskiren, (vi) rats with induced diabetes that were co-treated with insulin plus aliskiren, and (vii) rats with induced diabetes that were co-treated with exendin-4 plus aliskiren. Heart perfusion with [125I]-ET-1 was employed to estimate ET-1 binding affinity (τ = 1/K–n) to ETAR and ETBR at the level of the coronary endothelium and the cardiomyocytes. Plasma ET-1 levels were measured using enzyme immunoassay, whereas densities of ETAR and ETBR were detected using Western blot. No significance differences were detected in the τ of ETAR and ETBR between normal and diabetic in cardiomyocytes and the coronary endothelium. Exendin-4 normalized the τ value for ETAR and ETBR on coronary endothelium, while aliskiren normalized it on cardiomyocytes. Furthermore, ETAR and ETBR densities were normalized with monotreatments of aliskiren and exendin-4, compared with up-regulated ETAR and down-regulated ETBR band densities in the diabetic animals. Our data indicate that aliskiren alleviates diabetes-associated hypertrophy in type 1 diabetes mellitus.

AbstractThe study aimed to explore the regulation of heat shock protein 47 (HSP47) on expressions of receptors associated with hepatic stellate cell (HSC) in liver fibrosis mouse models induced bySchistosoma japonicum(S. japonicum). Mouse fibroblasts (NIH/3T3) were transfected with HSP47 shRNA plasmid by lipofectamine transfection, and experimental fibrosis in HSCs was studied inS. japonicummouse models treated with HSP47 shRNAin vivo. HSP47 expression was assessed using Western blot and real-time PCR. Flow cytometry was adopted to determine the expression of cell membrane receptors. HSP47-shRNA could markedly down-regulate the expression of collagen (Col1a1 and Col3a1). The expressions of HSP47, endothelin receptor A (ETAR) and endothelin receptor B (ETBR) significantly increased in the liver tissue of infected mice. However, the expressions of ETAR and HSP47 and ETBR remarkably decreased after the administration of HSP47 shRNAin vitroandin vivo. ETAR and ETBR levels were found to be positively correlated with HSP47 expression. HSP47 might exert influence on liver fibrosis via the regulation of ETAR and ETBR.

ABSTRACT Eleven PhoP-PhoS homolog pairs were identified by searching the Enterococcus faecalis V583 genome sequence database at The Institute for Genomic Research with the Bacillus subtilis PhoP-PhoS sequences. Each pair appears to be a potential two-component system composed of a response regulator and a sensor kinase. Seven of the homologs were disrupted in E. faecalis strain OG1RF. TX10293, a mutant disrupted in one of these genes (etaR, the first gene of the gene pair designated etaRS), showed delayed killing and a higher 50% lethal dose in a mouse peritonitis model. The predicted EtaR protein sequence showed greatest similarity to LisR of Listeria monocytogenes (77%) and CsrR of Streptococcus pyogenes (70%); EtaS is 53% similar to LisK and 54% similar to CsrS. When grown in vitro, the TX10293 mutant was more sensitive to low pH (pH 3.4) and more resistant to high temperature (55°C) than wild-type OG1RF. In conclusion, many potential two-component systems are identified for E. faecalis, one of which, EtaRS, was shown to be involved in stress response and virulence.

Abstract:
It is unknown if endothelin-A and -B receptors (ETAR and ETBR) activate the production of superoxide via NAD(P)H oxidase and subsequently stimulate the formation of cyclic adenine diphosphate ribose (cADPR) in afferent arterioles. Vessels were isolated from rat kidney and loaded with fura 2. Endothelin-1 (ET-1) rapidly increased cytosolic Ca2+ concentration ([Ca2+]i) by 303 nM. The superoxide dismutase mimetic tempol, the NAD(P)H oxidase inhibitor apocynin, and nicotinamide, an inhibitor of ADPR cyclase, diminished the response by ∼60%. The ETBR agonist sarafotoxin 6c (S6c) increased peak [Ca2+]i by 117 nM. Subsequent addition of ET-1 in the continued presence of S6c caused an additional [Ca2+]i peak of 225 nM. Neither nicotinamide or 8-bromo- (8-Br) cADPR nor apocynin decreased the [Ca2+]i response to S6c, but inhibited the subsequent [Ca2+]i response to ET-1. The ETBR blockers BQ-788 and A-192621 prevented the S6c [Ca2+]i peak and reduced the ET-1 response by more than one-half, suggesting an ETBR/ETAR interaction. In contrast, the ETAR blocker BQ-123 had no effect on the S6c [Ca2+]i peak and obliterated the subsequent ET-1 response. ET-1 immediately stimulated superoxide formation (measured with TEMPO-9-AC, 68 arbitrary units) that was inhibited 95% by apocynin or diphenyl iodonium. S6c or IRL-1620 increased superoxide by 8% of that caused by subsequent ET-1 addition. We conclude that ETAR activation of afferent arterioles increases the formation of superoxide that accounts for ∼60% of subsequent Ca2+ signaling. ETBR activation appears to result in only minor increases in superoxide production. Nicotinamide and 8-Br-cADPR results suggest that ET-1 (and primarily ETAR) causes the activation of vascular smooth muscle cell-ADPR cyclase.

Human papillomaviruses (HPVs) are associated with cervical cancer and interact with growth factors that may enhance malignant transformation of cervical carcinoma cells. Endothelin-1 (ET-1) is released from HPV-transfected keratinocytes and induces increased growth response in these cell lines in comparison with normal cells. HPV-positive cancer cells secrete ET-1 and express mRNA for ET-1 and its receptors, whereas HPV-negative carcinoma cell lines express only the ETB receptor (ETBR) mRNA and do not secrete ET-1. In HPV-positive cancer cells, ETAR mediates the ET-1-induced mitogenic effect and sustains the basal growth rate of unstimulated cervical tumour cells. Therefore, ET-1 may be involved in the neoplastic growth of HPV-associated cervical carcinoma, where the increased ET-1 autocrine loop can be targeted for antitumour therapy. In the present work, the action of specific antagonists of ETAR (BQ-123 and ABT-627), was analysed in CaSki and C33A cells that are derived from human cervical carcinoma. CaSki cells are HPV-16-positive, produce ET-1 and posses ETAR and ETBR, whereas the C33A line is HPV-negative, does not secrete ET-1 and has no ETAR. In HPV-positive cancer cells ABT-627 strongly inhibited the proliferation induced by ET-1 and substantially reduced the basal growth rate of unstimulated cervical tumour cells, whereas the ETBR antagonist had no effect. These results demonstrate that ET-1 participates in the progression of neoplastic growth in HPV-associated carcinoma, in which ETAR expression is increased and could be targeted for antitumour therapy. In conclusion, an ET-1 autocrine loop is involved in tumour cell proliferation via ETAR, and ABT-627 is effective in controlling proliferation of cervical carcinoma cells.

Ovarian cancer is the leading cause of gynecologic cancer-related deaths. The endothelin (ET) axis, which includes ET-1, ET-2, ET-3, and the ET receptors, ETAR and ETBR, represents a novel target in tumor treatment. ET-1 may directly contribute to tumor growth and indirectly modulate tumor–host interactions in various tumors such as prostatic, ovarian, renal, pulmonary, colorectal, cervical, breast carcinoma, Kaposi’s sarcoma, brain tumors and melanoma. Extensive experimental evidence links ETAR overexpression with tumor progression in ovarian cancer. ETAR engagement can in fact activate multiple signal transduction pathways including protein kinase C, phosphati-dylinositol 3-kinase, mitogen-activated protein kinase and transactivate epidermal growth factor receptor, which play a role in ovarian tumor growth and invasion. The effects of ETAR signaling are wide ranging and involve both cancer cells and their surrounding stroma, including the vasculature. Upon being activated, the ETAR mediates multiple tumor-promoting activities, including enhanced cell proliferation, escape from apoptosis, angiogenesis, epithelial–mesenchymal transition and increased motility and invasiveness. These findings indicate that activation of ETAR by ET-1 is a key mechanism in the cellular signaling network promoting ovarian cancer growth and progression. The predominant role played by ETAR in cancer has led to the development of small molecules that antagonize the binding of ET-1 to ETAR. The emerging preclinical data presented here provide a rationale for the clinical evaluation of these molecules in which targeting the related signaling cascade via ETAR blockade may be advantageous in the treatment of advanced stage ovarian carcinoma.

Activation of renal mechanosensory nerves is enhanced by high and suppressed by low sodium dietary intake. Afferent renal denervation results in salt-sensitive hypertension, suggesting that activation of the afferent renal nerves contributes to water and sodium balance. Another model of salt-sensitive hypertension is the endothelin B receptor (ETBR)-deficient rat. ET and its receptors are present in sensory nerves. Therefore, we examined whether ET receptor blockade altered the responsiveness of the renal sensory nerves. In anesthetized rats fed high-sodium diet, renal pelvic administration of the ETBR antagonist BQ-788 reduced the afferent renal nerve activity (ARNA) response to increasing renal pelvic pressure 7.5 mmHg from 26 ± 3 to 9 ± 3% and the PGE2-mediated renal pelvic release of substance P from 9 ± 1 to 3 ± 1 pg/min. Conversely, in rats fed low-sodium diet, renal pelvic administration of the ETAR antagonist BQ-123 enhanced the ARNA response to increased renal pelvic pressure from 9 ± 2 to 23 ± 6% and the PGE2-mediated renal pelvic release of substance P from 0 ± 0 to 6 ± 1 pg/min. Adding the ETAR antagonist to ETBR-blocked renal pelvises restored the responsiveness of renal sensory nerves in rats fed a high-sodium diet. Adding the ETBR antagonist to ETAR-blocked pelvises suppressed the responsiveness of the renal sensory nerves in rats fed a low-sodium diet. In conclusion, activation of ETBR and ETAR contributes to the enhanced and suppressed responsiveness of renal sensory nerves in conditions of high- and low-sodium dietary intake, respectively. Impaired renorenal reflexes may contribute to the salt-sensitive hypertension in the ETBR-deficient rat.

Background/Aims: Studies have shown that a change in endothelin receptor expression in the artery is related to pregnancy-induced hypertension (PIH). However, the mechanism underlying this change remains unclear. Methods: To test whether the distribution of endothelin receptor type-A (ETAR) and type-B (ETBR) plays an important role in PIH, a reduction of uterine perfusion pressure (RUPP) rat model was used to mimic some of the features of PIH; the resulting variable endothelin receptor expression was investigated in the media and intima of the aorta. Single vascular smooth muscle cells (VSMCs) were isolated from RUPP and normal pregnant (NP) rats to study the effect of ETAR and ETBR in smooth muscle cells. Results: Compared with NP rats, RUPP rats had a significant redistribution of ETBR expression in the intima and media, while there was no significant difference in ETAR expression between the two groups. ETBR upregulation in VSMCs enhanced cellular contraction and contributed to PIH. The TNF-α plasma levels in RUPP rats were two-fold higher than those of NP rats, which upregulated the expression of ETBR in VSMCS through the NF-κB pathways in RUPP rats. Conclusion: Redistribution of ETBR between the media and intima played an important role in the pathogenesis of PIH.

A cogeração é a solução técnica disponível para produzir energia elétrica e térmica de forma combinada com a melhor eficiência, mediante a queima de um combustível, seja ele de origem fóssil ou de natureza renovável. Este trabalho tem como objetivo fazer o levantamento e a descrição das instalações de cogeração das ETAR - Sul e Norte, detidas pela empresa SIMRIA - Saneamento Integrado dos Municípios da Ria de Aveiro, S.A., bem como dos processos de tratamento inerentes para o seu correto funcionamento, a avaliação dos fluxos energéticos e a sua análise critica. Para tal realizou-se uma inventariação e consequente compilação dos dados referentes aos fluxos energéticos existentes em cada ETAR para o ano de 2013, ou seja, procedeu-se à análise da produção e do consumo de energia elétrica, produção e consumo de energia térmica, produção e consumo de biogás, consumo de gás natural e queima de biogás na tocha. Esta análise teve como intuito identificar e quantificar os fluxos energéticos existentes. Bem como conhecer as necessidades energéticas de cada ETAR e a sua atual gestão. Com este estudo foi possível realizar uma análise energética completa dos fluxos de energia para um dia típico de Verão e de Inverno e respetivos modos de aproveitamento, ou seja, das diferentes formas de gestão dos recursos disponíveis e das suas variações ao longo do ano. Para efeitos comparativos e de discussão foram também equacionados três cenários de produção de energia elétrica e térmica a partir da queima do biogás. Dois destes cenários contemplam a produção de energia elétrica e visam aproveitar a potência nominal de produção de cada motogerador. Contudo, por motivos legais a potência elétrica encontra-se limitada a 250 kW – regime de miniprodução. O terceiro cenário tem como intuito analisar o potencial de produção de energia elétrica e energia térmica de maneira a que todo o biogás produzido seja utilizado. Para validar os estudos efetuados aos diversos fluxos de energia, e tendo em consideração as necessidades energéticas de cada ETAR é feita uma análise crítica, onde são apresentadas várias propostas de melhoria que visam o melhor aproveitamento das diversas formas de energia geradas.
Cogeneration is the technical solution available to produce electrical and thermal energy with the best efficiency by burning a fossil or renewable fuel. This paper has the porpuse of doing the description of the cogeneration facilities of the WWTP - South and North, held by the company SIMRIA- Integrated Sanitation of Municipalities of the Ria de Aveiro, S.A., such as the processes of wastewater treatment for it to work correctly. In this work we carried out a survey and subsequent compilation data regarding to the existing energy flows in each WWTP for the year 2013, ie, the analysis of the production and consumption of electricity, production and consumption of thermal energy, biogas production and consumption, consumption of natural gas and the amount of biogas being burned in the torch. This analysis was aimed to quantify and identify the energy flows, as well as meet the energy needs of each WWTP and the current management of these resources. With this study it was also possible to establish a complete energy analysis of the energy flows for a typical day of summer and winter and respective modes of utilization, ie, the different ways of managing the available resources and their variations over the year. For comparison and discussion three scenarios were also equated for the production of electrical and thermal energy from the combustion of biogas. Two of these scenarios include the production of electricity and aim to take advantage of the rated power output of each power generator. However, for legal reasons the electrical power is limited to 250 kW – system of miniproduction. The third scenario is meant to examine the potential of production of electricity and thermal energy so that all biogas produced is used. To validate the studies performed to the different energy flows, and taking into account the energy needs of each WWTP, several proposals which aim improving the overall utilization of various forms of energy generated were presented.

O presente relatório reflete o estágio de natureza curricular realizado com o objetivo de constituir trabalho final de mestrado do curso Engenharia do Ambiente, da Universidade de Aveiro. O ponto de partida deste estágio foi o trabalho a desenvolver na área do abastecimento de água e recolha de águas residuais em baixa, com o objetivo de adquirir conhecimentos acerca de todas as atividades que envolvem a conceção e exploração de Estações de Tratamento de Águas Residuais. O estágio decorreu na AdRA – Águas da Região de Aveiro S.A. onde trabalhei no estudo e dimensionamento de uma nova ETAR a construir em Sever do Vouga. Este estágio tornou possível a absorção de um conjunto de conhecimentos multidisciplinares importantes para o desempenho de uma atividade profissional no futuro.
The present report reflects a curricular traineeship accomplished with the aim of constituting final work of the masters degree thesis in Engenharia do Ambiente, by Universidade de Aveiro. The bottom line of this traineeship was the work to develop in the area of water supply and wastewater collection, with the purpose of gather knowledge of all the activities surrounding the design and operation of wastewater treatment plants. The traineeship took place in AdRA – Águas da Região de Aveiro S.A. where I worked at the study and design of a new wastewater treatment plant to build in Sever do Vouga. This traineeship took possible the absorption of a relevant range of multidisciplinary knowledge advantageous to the performance of a professional activity in the future.

Dissertação apresentada na Faculdade de Ciências e Tecnologia da Universidade Nova de Lisboa para obtenção do grau de Mestre em Engenharia do Ambiente – Perfil Sanitária
Desde o final do século XIX até aos dias de hoje verificou-se uma forte evolução nos sistemas de tratamento de águas residuais. Esta evolução foi fundamental para dar resposta às maiores exigências de qualidade do efluente tratado, pela necessidade de minimização do espaço ocupado pelas ETAR, pela simplificação da operação e pela redução dos custos de investimento e de exploração. Assim, face às dificuldades de cumprimento da actual licença de descarga atribuída à ETAR de Frielas e aos muitos problemas operacionais relacionados com os tanques de arejamento, decantadores secundários e biofiltros, surgiu a possibilidade de estabelecimento de uma parceria entre a SIMTEJO e a empresa DHV que consistiu na conversão de um dos tanques de arejamento existentes na ETAR num reactor baseado numa nova tecnologia designada por NeredaTM, patenteada pela DHV. Esta tecnologia baseia-se no princípio SBR, com a vantagem de utilizar biomassa na forma de grânulos aeróbios o que permite que as sequências de enchimento e de descarga de efluente tratado sejam simultâneas no tempo, para além de garantir a remoção de carbono, azoto e fósforo num único reactor devido à elevada diversidade microbiana que constitui os grânulos, ao contrário das tecnologias clássicas de lamas activadas. Com recurso a um conjunto de adaptações físicas e operacionais comprovou-se ser possível a adaptação de um tanque de arejamento convencional num reactor NeredaTM. De modo a comprovar de forma mais concreta as vantagens associadas a esta nova tecnologia, realizou-se uma comparação técnico-económica entre a mesma e uma solução de lamas activadas do tipo A2O, ambas aplicadas à ETAR de Frielas. Este estudo teve em consideração objectivos de qualidade mais restritivos, como a necessidade de remoção de N e P, de acordo com o estabelecido no Decreto-Lei n.º 152/97, de 19 Junho. Assim, concluiu-se que uma solução baseada na tecnologia NeredaTM permite uma economia de cerca de 40 % nos custos de 1º investimento, 40 % nos custos de exploração actualizados e 30 % nos custos totais actualizados a 30 anos. Para além das claras vantagens económicas, esta nova tecnologia oferece também vantagens técnicas, que se relacionam com a maior estabilidade de operação, resultante da maior concentração da biomassa, menores SVI, que se traduzem na inexistência de problemas de sedimentabilidade das lamas, e ainda numa clara economia da área necessária para a ampliação do actual tratamento biológico, isto é, cerca de 25 % face a uma solução clássica de lamas activadas. Como principal ónus, salienta-se o maior período de arranque, relacionado com o processo de granulação, que, no entanto, poderá ser minimizado quando as primeiras instalações à escala industrial estiverem em funcionamento, uma vez que será possível proceder à inoculação dos novos reactores com grânulos existentes

O presente trabalho insere-se na temática da tecnologia associada à digestão anaeróbia. Esta apresenta-se como uma possível mais-valia para diversas situações de tratamento de águas residuais, integrada na linha de tratamento de lamas, bem como integrada na própria linha da fase líquida. Pretende-se estudar as potencialidades da integração da tecnologia UASB para a reabilitação de ETAR, de um modo geral, e aplicá-lo ao caso de estudo da ETAR de Porto Côvo, cujo funcionamento está baseado num sistema de lamas activadas com arejamento prolongado, em funcionamento desde Novembro de 1999, encontrandose preparada para receber um caudal médio diário de 1.190 m3/dia. A Câmara Municipal de Sines, proprietária da ETAR, pondera avançar com a reabilitação da mesma. Com o presente trabalho pretende-se estudar as vantagens e os inconvenientes da adopção de uma solução com reactores UASB, para o caso de estudo da reabilitação da ETAR de Porto Côvo.

Este trabalho tem como objectivo avaliar as condições de funcionamento da Estação de Tratamento de Águas Residuais (ETAR) de Fervença, Alcobaça e apresentar soluções que permitam viabilizar o seu correcto funcionamento nas actuais condições. Apesar de ter sido construída para o tratamento de águas residuais domésticas de uma população de 34 500 habitantes equivalente, e de a população actual ser ainda inferior, a ETAR encontra-se com problemas relativamente à eficiência de tratamento, sendo o seu efluente final de qualidade muito inferior ao permitido pela licença de descarga. Assim sendo, foi necessário conhecer as características do afluente a esta ETAR de maneira a conseguir compreender o que está a afectar negativamente esta estação para determinar as possíveis melhorias a aplicar. Com esta avaliação foi possível constatar que o afluente em causa não era apenas doméstico mas que continha uma forte componente industrial, que embora tenha sido considerada quando da construção da ETAR, não foi devidamente contabilizada, pelo que, para manter a linha de tratamento em causa com poucas alterações e a reduzidos custos, será necessário avaliar as soluções possíveis recorrendo, eventualmente, à implementação de um órgão de pré-tratamento. Devido às características desta água residual, conclui-se que a solução mais adequada, será a de optar por implementar um digestor anaeróbio. Para a sua concretização, foram avaliadas várias tipologias destes reactores com relação à sua viabilidade técnica/económica para se determinar aquela configuração que melhor se adaptava a este caso.

The purpose of this project was to conduct a survey for the established measureable parameters of Guia WWTP’s operations, focusing on the sludge treatment process and analyse the sewage sludge management and recycling model, aiming to an idealized sustainable one based upon viable economical and financial solutions. According to values contained in the most representative operational efficiency of sludge treatment and production parameters, alternative options to the currently used disposal site by the Guia WWTP for the disposal sewage sludge were presented and debated. Moreover, it is considered a perspective of optimization, namely, for the waste management process final phase (purified sludge), according to the possibility of the implementation of the treatment techniques presently recognized and efficiently applied, though, considering sizing and exploitation particularities of the concerning facilities. However, the used methodology including an extensive literature review, allowed a systemic qualitative screening of the produced sewage sludge at the WWTP, technical searching and relevant statistical data analysis allowed the systemic trace/pursue of the quantitative and qualitative markers on the generated sludge. The associated costs projected estimation culminated in an option ranking according to each technical-economical performance criteria, abiding to the legal requirements and suitable to the WWTP’s installed operating capacity.

Avaliar o efeito de actividades económicas próximas da ETAR na carateristica do elfuente e o efeito da precipitação no caudal que chega à ETAR. Procura ainda avaliar o funcionamento do reator biológico através de análises feitas em diversos pontos do reator bem como a medição da velocidade a que o elfuente passa pelo mesmo.
This work aims to monitor the operation of the WWTP at controlling the parameters of the organs of treatments, evaluating the features of the wastewater and leached with respect to BOD, COD and TSS. To evaluate the effect of economic activities near the Wastewater Treatment Plant, in the characteristic of the wastewater and the effect of precipitation in the flow that reaches the WWTP. It also seeks to assess the functioning of the biological reactor by testing done at various points of the reactor and measuring the rate at which passes through the same.

A análise do funcionamento de uma Estação de Tratamento de Águas Residuais (ETAR) é muito importante para conhecer a interação e gestão de todas as fases do processo de tratamento existentes nas ETAR e para perceber qual o tipo de gestão adequada. O estágio na ETAR Norte da SIMRIA ocorreu entre Março e Julho de 2013. O conhecimento dos principais órgãos da instalação e o modo como operam constituiu o primeiro objetivo do estágio, essencial para efetuar uma caraterização pormenorizada da ETAR ao qual se seguiu uma avaliação pormenorizada das condições em que a ETAR opera atualmente, através da compilação e análise dos dados respeitantes à operação no ano 2012. Através da comparação destes para o ano 2012 e 2013 foi possível compreender que a gestão de uma ETAR não se resume à manutenção de determinados parâmetros dentro de intervalos ideais sendo, por vezes e durante determinado período de tempo, impossível esta manutenção uma vez que o processo depende fortemente da qualidade do afluente. O desempenho da ETAR foi avaliado relativamente aos parâmetros constantes da Licença de Utilização dos Recursos Hídricos para a Rejeição de Águas Residuais e o funcionamento geral analisado face aos valores previstos no respetivo projeto de execução e face a gamas ideais presentes na bibliografia consultada. Para o dimensionamento do sistema para Nitrificação e Desnitrificação, o primeiro passo consistiu em validar um conjunto de frações de Matéria Orgânica e constantes cinéticas de forma a atingir um volume dimensionado igual, ou muito próximo, ao existente, no cenário atual de apenas remoção de Matéria Orgânica, para a partir daí realizar o dimensionamento do sistema para Nitrificação e Desnitrificação, de forma mais exata possível.
The analysis of the operation of a Wastewater Treatment Plant (WWTP) is very important to understand the interaction and management of all phases of the treatment process existent in the WWTP and to realize what kind of management are indicated. The internship in ETAR Norte of SIMRIA occurred between March and July 2013. The knowledge of the principal organs of the ETAR Norte and how they work constituted the first major goal of the internship and it is essential to perform a detailed characterization of the WWTP. The avaluation of the conditions under which it currently operates, through the compilation and analysis of the operative data of the year 2012 was the first step in the design of the treatment system for Nitrification and Denitrification. With the analysis of the operative data of 2012 and 2013 it was possible to understand that the management of a wastewater treatment plant is not just keep the values in the ideal ranges. Sometimes, in a period of time, is impossible to maintain the values in the wanted range because the process depends heavily on the quality of the influent. The performance of the WWTP was evaluated for the parameters listed in Reject Wastwatwe License and general functioning analyzed against the predicted values on the appropriate project execution and face to parameters considered ideal in the bibliography. For the sizing system for Nitrification and Denitrification the first step was to validate a set of fractions of organic matter and kinetic constants in order to achieve an equal sized volume, or very close to, existing in the current scenario only organic matter removal, to thereafter perform the sizing system for Nitrification and Denitrification, most accurate way possible.

O presente projeto teve como objetivo contribuir para a melhoria da qualidade do serviço prestado ao Cliente no contexto de uma Estação de Tratamento de Águas Residuais (ETAR). O trabalho foi desenvolvido na ETAR de Setúbal, sob gestão operacional da LUSÁGUA Serviços Ambientais, S.A. e contemplou o desenvolvimento de competências e procedimentos de trabalho relativos à proteção contra atmosferas potencialmente explosivas (ATEX). O projeto incluiu uma revisão preliminar da literatura sobre qualidade e segurança, em particular no contexto de atividades de serviços, assim como uma análise da legislação e normas no âmbito do trabalho em locais ATEX Os resultados do projeto permitiram promover procedimentos de trabalho, ações de prevenção e proteção, sinalização de aviso e regras de segurança nas operações em questão por forma a melhorar a qualidade do serviço realizado pelos trabalhadores, sempre de forma segura. Pretendeu-se também criar uma base de conhecimento que permita a disseminação futura desta temática a outras instalações sob responsabilidade da LUSÁGUA.
This project aimed for the improvement of the quality of the service provided to customers and stakeholders in the context of a wastewater treatment plant. The work was developed for the specific case of the wastewater plant of Setubal, managed by the company LUSÁGUA Serviços Ambientais, S.A. the project work included the development of competences and work procedures regarding the protection towards explosive atmospheres (ATEX). The project involved a preliminary literature review about quality and safety, notably in service operations, as well as a review of ATEX legislation and norms. The results of the project supported the development of work procedures, preventive protection actions, warning signs, and safety norms to improve the quality and the safety of the staff operations in the company. It also enabled the development of a knowledge base which can support the dissemination of such practices in other plants managed by the company LUSÁGUA.

No decurso deste trabalho serão descritas as atividades realizadas no trabalho de fim do Mestrado em Engenharia do Ambiente pela Universidade de Aveiro. A utilização de fontes de energia renováveis adquire hoje em dia uma importância elevada no contexto de desenvolvimento sustentável das nossas sociedades e no consumo racional dos recursos finitos que o planeta nos disponibiliza. No entanto é preciso avaliar se algumas apostas feitas têm o efeito desejável, ou seja, a produção de energia renovável numa perspetiva de desenvolvimento sustentável, ou se pelo contrário se mostram investimentos onerosos sem possibilidade de retorno adequado e neste caso, o porquê de algumas opções não produzirem os resultados esperados. O objeto deste trabalho e correspondente relatório é aferir se é vantajoso em termos técnicos e económicos a instalação de sistemas de cogeração para a produção de energia elétrica, reaproveitando o biogás produzido em sistemas de digestão anaeróbia de lamas, fazendo uma descrição exaustiva do processo de digestão de lamas e da sua gestão, dados os elevados custos de investimentos associados a estes sistemas e dadas as dificuldades processuais e operacionais da otimização e gestão da produção, utilizando como base de estudo a ETAR de Bragança no período decorrido ente 2008 a 2010. Desta forma pretende-se contribuir para uma maior ponderação numa possível escolha de reaproveitamento de biogás em ETAR´s de média dimensão dadas implicações inerentes as mesmas nomeadamente a nível de projeto, aspetos construtivos, custos de instalação e dificuldades operacionais.

Abstract:
Edge-to-edge repair (ETER) is a newly developed technique to correct such mitral valve (MV) malfunctions as regurgitation [1,2]. This technique changes MV geometric configuration by suturing the anterior and posterior leaflets at central or commissural edges, and consequently alters MV and left ventricle (LV) dynamics. For instance, stress in the MV elevated due to ETER may cause leaflets tearing near suture. Little has been known about shear stress on the MV and LV walls under MV ETER conditions, where high shear stress might cause platelet activation or hemolysis [3]. When ETER is done at the central leaflet edges, it generates two MV orifices, leads to two deflected jets, and completely changes vortices in the LV. ETER also reduces the orifice area, and increases jet velocity and transmitral pressure [1,2,4]. Flow patterns in the LV and ETER effects on the LV and MV functions have not been understood well.

Abstract:
The edge-to-edge repair (ETER) technique is a mitral valve (MV) repair procedure. It approximates the leading edges of the mitral leaflets by use of sutures, exhibits promising predictable repair results and offers the advantage of being performed pericutaneously without the need for open heart surgery. The technique is becoming a popular surgical procedure to correct MV prolapse caused by abnormal chordal elongation. MV prolapse leads to mitral regurgitation (MR). Generally the ETER technique is performed as a secondary procedure to ring annuloplasty. Although some groups have performed ETER without the ring annuloplasty, recent studies have shown that ETER technique alone leads to substandard results [1]. It is necessary to define the MV annulus mechanics in ETER condition during valve closure or mid-systole as it directly affects the annulus tension (AT) in the annulus plane. AT in the annulus plane is an important force component which balances the myocardium force and contributes to maintaining the annulus shape and size. This AT change caused by the ETER can lead to annulus dilation, reoccurrence of MR and impact reintervention. The objective of the current study is to understand this AT change in the MV annulus tension during the mid-systole in the ETER condition in order to improve the long term efficacy of the ETER procedure on MV annulus tension, after the technique has been applied with both anterior leaflet prolapse and posterior leaflet prolapse.
