Full textRheumatoid Arthritis, one of the most common autoimmune diseases, is characterized by progressive articular damage leading to joint deformities and disability. The multifactorial nature of RA provides high disease heterogeneity with specific combinations of a genetic background and environmental factors that influence the susceptibility, severity and outcome of the disease. The aim of this thesis was the identification and characterization of candidate genes in RA. We have confirmed the association and linkage of TRAF1-C5 and 6q23 gene regions, and demonstrated a trend for the association and linkage of the 4q27 gene region with RA in European descent populations. Furthermore, we provided evidence against the involvement of the PRKCH, CASP7, RANK and RANKL genes and the PTPN22–1123G allele in RA genetic susceptibility in the European population. We performed a large-scale gene expression profiling study using 48.701 cDNAs in PBMCs of RA patients and healthy controls. A differentially expression of 339 genes (238 down-regulated and 101 up-regulated) between the two groups was observed. We identified a remarkably elevated expression of a spectrum of new genes involved in immunity and defense mechanisms. Finally, we identified an association between HLA SE alleles and tobacco smoking for anti-CCP positivity in French population with familial and sporadic RA. All these complementary approaches that allowed the identification of new genes and gene-autoantibodies-environment interactions contribute to a better understanding of RA disease mechanisms and could lead to the identification of innovative clinical biomarkers for diagnostic procedures and therapeutic interventions

Full textThe aim of this work is to understand the mechanisms of intra- or intermolecular association which causes the formation of the Physical gels, during the thickening process of unsaturated of esters in sheet molding compounds by isocyanate. A model reactive s stem was chosen: a saturated hydoxy-terminated polyadipate with a diisocyanate in styrene. The phenomenon of physical gels was observed in large range of concentrations. With the aid of Fourier Transform Infrared the study of the gels shows that hydrogen bonds exist not only between polyurethane chains but also between polyurethane chains and styrene molecules. These hydrogen bonds were also investigated quantitatively. The characterization of solvating of these gels indicates that a great number of styrene molecules are « intercalated » in the form of monomolecular layers in the three-dimensional network which is formed by the hydrogen bonding, the measure of the rheological properties of the gels demonstrates that at low frequencies these gels are viscoelastic liquids, therefore they are produced by entanglements and hydrogen bonding and are considered only weak gels

Full textOsteochondral lesions are commonly observed in young horses and may be responsible for reduced performances in racing. The purpose of the PhD thesis was to identify genome regions, called quantitative trait loci (QTL), associated with various traits measuring osteochondrosis (OC) and recorded in the GENEQUIN program in a population of French Trotters horses. Genotyping was performed using the EquineSNP50 Illumina high density chip, which allows to exploit the linkage disequilibrium with genome-wide association studies. These analyses are subject to several problems in presence of family structure. We hence first proposed a comparison of power and robustness of a limited choice of models for this type of analysis. The originality of this work lies in the algebraic derivation of the distribution moments of the test statistics compared, making the outcome of this comparison more general and allowing a better understanding of differences. The results can be used to establish an experimental design. The second part was devoted to the QTL fine mapping of traits that measure OC in different joint sites. This study highlighted several significant QTL with low and medium effects but none of them were highly significant. We showed that OC is a polygenic trait and we were not able to identify QTL affecting both OC on the hock and the fetlock, rejecting the hypothesis of a single genetic determinism of susceptibility to this desease accross anatomical sites. Further studies will now focus on the identification of candidate genes and screening for mutation in an attempt to clarify the molecular physiopathology of OC and develop efficient strategies for risk assessment. Meanwhile, markers could be used in a marker-assisted selection context to improve horse health and welfare

Full textThe aim of this work was a better understanding and prediction of interactions between iron and Natural Organic Matter. Iron adsorption was performed onto an insolubilised humic acid as a NOM analogue. Combining spectroscopic techniques (IR, EPR and Mössbauer spectroscopy of 57Fe enriched samples) and chemical analysis onto the solution ([Fe]total using GF-AAS and redox speciation with colorimetry) allowed to constrain the modelling of these phenomena. We’ve demonstrated that interactions with organic matter modify the solubility of the precipitated Hydrous Ferric Oxide and that it can impose its potential to the solution, determining the [Fe(II)]/[Fe(III)] ratio. Furthermore comparing the adjusted model speciation predictions and spectroscopic results allowed to identify HFO nanoparticles, intimately associated with organic matter

Full textMalaria and sepsis are widespread infectious diseases causing hundreds of thousands deaths each year. There is a growing body of evidence for a genetic control of malaria and sepsis resistance. Both diseases are considered close because they are characterized by a systemic inflammation, some common organ dysfunctions, and disorders of coagulation. In order to identify new genes potentially involved in disease resistance, we performed genomewide genetic studies in two populations living in endemic regions for Plasmodium falciparum malaria in Burkina-Faso and in a cohort of septic shock patients. Genetic linkage studies revealed significant genetic linkages on chromosome 6p21.3 and 17p12 with, respectively, mild malaria and asymptomatic parasitaemia. We also performed the first genetic linkage studies concerning immunoglobulin G and their sub-classes against Plasmodium falciparum antigens. We detected significant linkages of IgG3 sub-class with chromosomes 8p22-p21 and 20q13 and between IgG4 sub-class and chromosome 9q34. Finally we performed the first two genomewide association studies identifying significant associations with all-causes mortality after a septic shock. We identified 32 SNPs significantly associated with early mortality and 108 SNPs significantly associated with late mortality. We identified a protein-protein network containing proteins associated with both early and late mortality. Furthermore, the network of protein-protein interactions involving proteins encoded by associated genes allowed us to establish a list of some altered functions during septic shock, most of them being related to the immune responses or the renin-angiotensin-aldosterone system

Full textFor most crops, water availability is a major component of yield. We must improve crop plants tolerance to water deficit in order to guarantee food security. Sunflower, a species of economic importance is also concerned by this challenge. The aim of this thesis is to identify genomic regions involved in the variability of yield and its components under drought. To this purpose, an association mapping approach has been led on a core collection of 384 cultivated and elite lines evaluated as hybrids combinations in a multi environment trial of 17 environments. In a first step, the separated analysis per environment led to the identification of 157 markers associated with at least one trait by apply a model taking into account the structuration in two groups: male restorer lines and female lines. Among these significant markers, 34 were associated with productivity but only on a specific environment, underlying the importance of genotype by environment interaction. In a second step, a crop model simulating sunflower genotypes yield depending on environmental conditions, led to the characterization of drought stress inside the multi environment trial. By using varieties defined before in the model, a drought index has been estimated for each environment. Thus, the panel response to this index has been tested in association mapping leading to the identification of several new loci involved in drought stress response and yield stability. QTL mapping in a bi parental population confirmed some regions and led to the detection of new ones

Full textMaize is one of the most cultivated cereals worldwide. This economically important species is very sensitive to water stress. It is thus important to improve the tolerance of maize to this abiotic stress. Association mapping is one of the approaches that make it possible to map loci underlying the tolerance to water stress or loci in linkage disequilibrium with the causal ones. This PhD project was divided in two separate parts. The first part aimed at evaluating the effect of structure models on association mapping results. A real maize data set of 342 inbred lines, genotyped with more than 12 000 single nucleotide polymorphisms (SNPs), was used to carry out this study. These inbred lines were phenotyped for both male flowering dates and thousand kernel weight. The complex structure of this panel was estimated with both dimensional reduction methods and STRUCTURE software (Bayesian approach) for several group numbers. Our results show a significant effect of the structure model used on association mapping tests. This effect depends on both the method and the number of groups used to control the panel structure. In order to select the most robust loci, a meta-analysis approach was carried out, making use of the results of association mapping tests with all structure models. The second part of the project aimed at mapping loci and genomic regions involved in maize drought tolerance. Two panels of elite inbred lines, representative of two heterotic groups were used. These panels were phenotyped under well-watered and water-stressed conditions. All inbred lines were genotyped with about 68 000 SNPs. The genotypic data was used to estimate the genetic diversity, the structure and the linkage disequilibrium within each panel. At the same time, the phenotypic data (flowering dates and yield related traits) were used to estimate genetic values which made it possible to calculate drought tolerance indices. To finish, association mapping tests were carried out between all polymorphic SNPs and drought tolerance indices. They allowed for the identification of 47 genomic regions that are specifically involved in drought tolerance

In effect his philosophical system, which found the right to happiness onvengeance (revolution), by meansof the expression of the desire for the soul's immortality, is based on reason. The concept of reason is part of the following framaework:reason logic-language-rhetoric-support (bodybrain). Reflections on sensitive aspects, on the hiatus between sound and sense rendered necessary the use of neuropsychological tools. A kinetic model was able to account for radiscev's intuitions, thanks to a key notion-that of sensitivity. Indeed, prosody enables us to link the verbal and non verbal componenets of language which- when seen as an aptitude - corresponds to the need to understand and to affect the emo- tion, and which is actualized both as language code structure logic(so- cial memory) and as rhetoric (biographical memory). The "voice" of radi- scev bears witness to this mouvement through the use he makes of the russian language which was in the midst of great change during the en- lightenment. The speakingsubject thus took on bio-psychosocial dimen- sions which history tried, for a time, to reduce to the absttract "man"

Full textOdorant molecules, generally hydrophobic, activate the olfactory receptors (OR) embedded in the membrane of sensitive neurons which are covered with an aqueous layer of mucus. It was proposed that odorant-binding proteins, named OBP, transport odorants through olfactory mucus towards OR. In this work we studied a human OBP variant, hOBP-2A, which has a narrow specificity for aliphatic aldehydes and acids. Using site directed mutagenesis, we demonstrated the fundamental role, in this specificity, of a lysin located in the hydrophobic cavity of hOBP-2A. The functional expression of an olfactory receptor, OR1G1, revealed an activation of this receptor by some compounds that bind to hOBP-2A. Study of the activation of OR1G1 with and without hOBP-2A revealed that hOBP-2A did not have impact on OR1G1 activity. One cannot however exclude the possibility that the OBP is involved in other biological processes than odorants transport

Full textPlatelets play a crucial role in the pathogenesis and thrombotic complications of atherosclerotic disease. Variability in platelet response to several agonists has been described, but the underlying mechanisms are unknown. We studied platelet response in 100 healthy volunteers and evidenced different phenotypic groups in term of platelet aggregation to ADP and TRAP. We then looked for genetic polymorphisms of one of the ADP receptors (P2Y12) and evidenced 2 haplotypes (H1/H2). The H2 allele is associated with platelet response to ADP in our population. Moreover, an intronic genetic polymorphism of the thrombin receptor PAR-1 is associated with platelet response to TRAP. Finally, the H2 allele is associated with peripheral arterial disease in a case-control study. In conclusion, we evidenced genetic polymorphisms of 2 platelet receptors with a functional role which may be interesting in clinical practice

In the context of diabetes, proteins are altered by glycation which leads to structural modifications and potentially affect their interactions.The first objective of this work was to determine the impact of albumin glycation on its binding to liraglutide, a drug increasingly used in the treatment of type 2 diabetes. Then, the second part of this work involved the production of recombinant functional human ApoA1 in order to study its interaction properties, in its lipid-free form or associated with phospholipids. The NMR (nuclear magnetic resonance) technique has been used on proteins previously labeled with fluorine (for liraglutide) or stable 13C/15N isotopes (for ApoA1). In addition, isothermal titration microcalorimetry (ITC), has been applied to the study of interactions with the advantage of not requiring any labeling. Various cloning strategies have been explored for the overexpression of ApoA1 in Clearcoli bacteria.The results demonstrate an alteration of the affinity of albumin for liraglutide in vitro and in vivo, depending on the degree of glycation. These results, supported by a lipidomic and peptide analysis, explain clinical observations concerning the decrease of efficacy of albumin-binding drugs in patients with poorly controlled diabetes. Regarding ApoA1, the choice of the fusion tag remains to be optimized, but both labeled and unlabeled ApoA1 were successfully overexpressed at high yields in a soluble form

Full textVon Willebrand factor (VWF) is a multimeric glycoprotein of haemostasis, mainly acting in platelet adhesion by binding to glycoprotein Ib (GPIb) with the A1 domain of its sub-unit. The multimeric structure results from the association of the sub-units into dimers and then, of the dimers into multimers, with the involvement of D3 and CK domains. The high molecular weight (HMW) multimers are required in its regular function. Von Willebrand disease (VWD) is a constitutive hemorragic affection which comprises different types. In type 2A, HMW multimers are absent and GPIb-binding is decreased. In type 2M, the binding is also decreased but independently from the multimeric structure. The aim of this work is to study the funtionnal structure of VWF by finding amino-acids involved in the GPIb-binding and in the multimerization process. Our methodologic approach consisted in the characterization of recombinant VWF with mutations identified in patients with type 2A or 2M VWD. In the first chapter, we show that amino-acid Ser1285 plays a major role in the normal conformation of the Al loop required for GPIb-binding. In the second chapter, we show that mutation of Cys 1157 or 1234 modify the conformation of D3 domain; mutations C1157F and C1234W induce the absence of the HMWmulimers, an intracellular retention in the endoplasmic reticulum and a decreased secretion of VWF. In the last chapter, we show that amino-acid Ala2801 is required for normal multimerization of VWF. The A2801D mutation probably induces an abnormal conformation of the CK domain, that results in the formation of unstable dimers.
