Full textCDC25B is one of the three dual specificity phosphatases that control entry into mitosis through the dephosphorylation and subsequent activation of CDK1-CyclinB1 complexes. CDC25B has been reported to be overexpressed in a number of human tumours. In many cases, CDC25B upregulation is associated with more aggressive phenotypes thus making CDC25B an ideal target for cancer therapy. Regulation of CDC25B is still unclear. In particular, the specific functions of the three major splice variants of this phosphatase (CDC25B1, B2 and B3) remained to be clarified. In order to address this issue, we have constructed vectors that allows us to examine the expression and the localisation of the different isoforms in living cells by real time videomicroscopy. We have demonstrated that CDC25B isoforms have different mitotic stabilities. We have also observed a nucleo-cytoplasmic shuttling of the CDC25B phosphatases at the G2/M transition

Full textSince the pioneering work of Brown and Roshko on the effects of density variations within the mixed layer flow, several other theoretical, experimental and numerical studies harnessed to finely investigate this flow. The motivations are of practical order (chemical industry, aerodynamics, combustion. . .) or purely theoretical (the role of coherent structures,secondary instabilities). These studies have focused on, among others, the effects of compressibility and/or variable density. To our knowledge, the effects of viscosity variations in the mixing layer configuration are not discussed in the literature. The objective of this researchis the theoretical and numerical exploration of the variable viscosity temporal mixedlayer flow, especially during its initial phase of development. From a numerical viewpoint, the Navier-Stokes equations are solved in weakly compressible formulation, using the solver CHOC-WAVES, based on WENO scheme. The DNS approach is justified by the absence in the literature of subgrid models that account for the effects of variable viscosity. The transport equations of different mean and fluctuating quantities at a point and each scale (scale-by-scale energy budget) are rewritten in incompressible and variable-viscosity formulation. Additional terms, generated by the spatial and temporal variations of viscosity occur in these equations. These are used as a tool to explore the mixed layer flow and study the development of turbulence in a heterogeneous environment. The simulated viscosity ratios are Rv = [1 − 18]. The numerical results show that the mixing layer thickness δθ growsfaster when the viscosity ratio Rv is high. The vertical gradients of the longitudinal mean velocity are amplified by the viscosity gradients, a gain of almost 60 %, compared to initial values was observed. The production of turbulent kinetic energy is also amplified. The temporal evolution of the velocity fluctuations is accelerated, they are increased to nearly 120 % with respect to the constant viscosity flow. The self-similar regime of the Reynolds tensor is reached more quickly by the variable viscosity flow and the isotropy of the velocity fluctuations is improved

Full textAn accurate knowledge of rainfall is a preliminary condition of the optimisation of quantitative and qualitative control of urban rainwater. This study concerns the urban area of lille, and the data are provided by meteo-france, belgium's irm and cudl. The local rainfall is approached by analysis of data from one recording rain gauge at current times scales who are insufficient to answer to aims. It is thus necessary to study the rainy episode, delimited by a dry duration threshold from some minutes to some hours. A typology of rainy episodes is obtained in accordance with significant criterion for sewer network management. The spatial approach, based on a manual rain gauges network, includes a pca and hac of annuals rainfalls allowing to define homogeneous rainfalls areas, a definition of monthly organisation rainfalls of the lille area, and an analysis integrating the space dimension of rainy days and rainy sequences by a classification according to impacts on sewer network, who emphasises maladjustment of data's manuals rain gauges for the needs of urban hydrologists. News tools, more appropriates and complementary, are presented : a recording rain gauge network and a meteorological radar. The aim is to examine if using of imagery from the abbeville radar, joined to a recording rain gauge network, is possible. A qualitative analysis of radar data for some episodes is based on an independence statistical test (x2) which results are different : a wrong to a correct correlation between radar and recording rain gauge network. These different answers are detailed by two examples showing that the urban area of lille is on reach limit of the radar

Full textThe french-chinese mission SVOM (Space-based multi-band variable Astronomical Object Monitor), is dedicated to the study of ƴ-rays bursts, brief and intense X and ƴ photons flashes, associated with the catastrophic formation of black holes. SVOM will embark in 2021 four instruments observing from the visible to ƴ rays. ECLAIRs, the main telescope, is a coded mask camera able to detect and locate, about 200 bursts during the three years nominal life time of SVOM. Its detection plan counts 6400 Schottky CdTe detectors, grouped into a matrix of 32 pixels forming a module. The plan is divided into eight electronically independent sectors, each consisting of 25 modules. Each electronic is dedicated to read and encode, the time, the position, the multiplicity and the energies of the detected events on a sector. The validation of each functionality of the readout electronics allows to ensure the correct encoding of the events, to estimate the limits of the electronics and to build an analytical model of lost events efficient correction. An application to a lightcurve of a bright GRB permits to validate theoretically this model and measurements carried out on the prototype validates it experimentally. On the other hand, a complete model of the spectral response has been established to characterize each detector on the sector of the prototype and to estimate the response of the plan of detection of the mono-energetic photons. The physical processes of the radiation-matter interactions are simulated as well as a model of lost charges then a Gaussian convolution takes into account the electronic noise. This model, counting six free parameters, is fitted to spectra measured on the prototype. The study of the extracted parameters characterizes the performance of each detector in every voltage and t peaking configurations optimizing the performance of the instrument

Full textThe unstable dynamics of a swirling jet flow is studied, including density variations within the flow. A direct numerical simulation method was developed to solve variable density Navier-Stokes equations, using an accurate and efficient pressure solver. Analitically, two unstable bi-dimensionnal modes are highlighted, and are identified as Couette-Taylor and Rayleigh-Taylor modes. A three-dimensionnal mode is also highlighted, wich is created by the shear. Numerical simulations show that those modes are nonlinearly persistant. Then, the spatio-temporal instability behaviour is studied numerically, and show that the instability undergoes to a convective/absolute transition with density ratio s and rotation rate q. This dynamic is also studied experiementally through different methods, and Global selfsustained modes are highlighted wich are in ggod agreement with numerical results. Finally, the vortex breakdown phenomenon is studied, and show the crucial role of real viscosity. Indeed, the vorticity intensification is relaxed through the viscosity effect, while it is not treating the truncated Euler Equations

Full textThe current wireless communication systems, mobile phones, satellite services and wireless internet networks require a very high data rate and a highly reliable degree. These rates have increased rapidly in the new applications of data transmission of new generation. To take into account the spectrum limitations, the OFDM has been proposed thanks the orthogonality between sub-carriers and the data rate that approaches to the Nyquist-Shannon sampling rate. Furthermore, the antennas technic (MIMO) can provide significant various gains, a diversity gain that improves the link reliability and the spatial multiplexing gain where different data streams are transmitted over different antennas. The combination of these two systems (MIMO and OFDM) allows to exploit the robustness of the link on the frequency-selective channels and uncorrelated channels in space. One of the issue in the combination MIO-OFDM resides on the synchronization methods. The synchronization is divided into sub parts, timing synchronization and frequency synchronization. Timing synchronization is also divided into two parts, firstly, the coarse timing synchronization is used to estimate the beginning of each received frame, and secondly, the fine timing synchronization which detects the beginning of each OFDM symbol in the received frame. The principle of the frequency synchronization is to find the shifted phase between the transmitted frequency and the local frequency at the receiver. In a first part, we have proposed different methods for timing synchronization based on synchronization sequences known at the receiver. We did a study for various existing sequences to compare the efficiencies of each of these sequences in timing synchronization for MIMO-OFDM systems. In a second part, Matlab’s simulations were conducted to study the performance of our proposed methods in multi-paths frequency-selective channels. Simulations results show the acquisition timing synchronization probability in terms of SNR

Full textIntegration of microband electrodes in a microfluidic channel can be carried out in order to detect electrochemically, in situ and in real time, some electroactive species, On one hand, the aim of this work is to study, the influence of the mass transport, under laminar flow, of electroactive species on chronoamperometric responses of microelectrodes. On the other hand, this study deals with the perturbations of concentration generated downstream within the microchannel. Therefore, a theoretical approach has been performed by numerical simulations in order to solve the transport equation for different hydrodynamic and geometrical conditions. The analysis of the currents has led to specify the conditions which define the transient state and the steady state for one single electrode. The propagation of the concentration front has also been carried out. We have shown that a second electrode, used as a concentration probe, could detect this front. The validity of these datas has been tested by performing experimental measurements using microdevices integrating one or two working electrodes. All these results have led to determine some optimal conditions for analytical applications combining microfluidic and electrochemistry

Full textTwo-proton radioactivity is a decay mode of proton-rich nuclei located beyond the proton nuclear existence limit (drip line). Predicted in the 1960s, this process was observed for the first time in 2002 in the 45Fe decay study. Only four two-proton emitters were known so far: 45Fe, 48Ni, 54Zn and 19Mg. A search for new emitters was performed with the EURICA- WAS3ABi setup at the RIKEN Nishina center in 2015. The decay of 59Ge, 63Se, 67Kr and 68Kr was observed for the first time. Two-proton radioactivity of 67Kr together with the beta and delayed proton decay of exotic nuclei in the region were studied. A time projection chamber (TPC) developed at the CENBG (2004-2011) enabled the study of the correlations between the protons for 45Fe and 54Zn. A second generation of TPC is under construction within the ACTAR TPC (ACtive TARget for TPC) collaboration. This detector enables a reconstruction in three dimensions of the energy deposited in the active volume which allows a more efficient reconstruction of the tracks as compared to the previous TPC. The generic electronics GET (General Electronics for TPCs) manages the processing and acquisition of the signals. The characterisation of the GET electronics and the TPC demonstrator at CENBG is devoted to the second aspect of this PhD work

Full textThe research work depicted in this thesis concerns the multi-sensor data fusion or combination in distributed environments. The objective is to improve the data accuracy by taking into account the timestamping error. The target application considered in this thesis consists of a data acquisition and processing system, embedded in an instrumented vehicle. Firstly, a method of interval timestamping correspondence in distributed environment is proposed. Ln addition to the good synchronization quality, the method has the advantage of limiting the exchanged messages on the bus. Ln the second stage and in order to reuse existing algorithms based on exact dates, we propose a method to convert linearly interval dates into punctual dates. The timestamping error is thus reflected on data imprecision. Different experiments in the advanced driver assistance systems domain have validated this study

Full textThe microalgal biofilm, which is present on sediment surface, is the primary production unit on bare intertidal mudlfats. In order to improve our understanding of this primary production system, a study was undertaken at microscale to analyse the structural and functional properties of this biofilm. In the first part of this work, a detailed descriptive analysis lead to the characterisation of the spatio-temporal dynamics of the physical structure of the biofilm. It turns out that the biofilm exhibits its own dynamics at the sediment surface (as a discrete compartment in the top 200 µm), but still has a functional relationship with the total available biomass (PCB) distributed within the top 1 cm (linear relationship between the maximum size of the biofilm during an emersion and the total biomass). This descriptive survey also allowed to show the complexity of the “formation /dispersion” pattern of the biofilm during an emersion, and pointed out the fact that the biofilm tends to remain at the sediment surface all over the course of the emersion period. In the second part of this study, the aim was to assess the photosynthetic performance of in vitro biofilms. Pulse Amplitude Modulated (PAM) fluorometry proved to be a perfect tool to measure the photosynthetic activity of microalgae biofilms, especially by Rapid Light Curves (RLC) despite the complexity of the photobiological response which, in this instance, is a double function of light and time. RLC allowed to show that an increase in the size of the biofilm (the thickness) induces an under-estimation of the “true” physiological response of epipelic microalgae, but an increase of the resistance capacity of the biofilm as a whole to strong illuminations. These results clearly suggest a change in optical properties of the biofilm as a function of its size, and lead to an improvement of our representation of the biofilm during periods of photosynthetic activity

Full textT2K is a long baseline neutrino experiment currently running in Japan, which aims at measuring the mixing angle 013. The experiment uses a high intensity beam of muon neutrinos, and observes the oscillations into electron neutrinos, ve, at the Super-Kamiokande detector, 295 km away from the neutrino source. This thesis presents a measurement of the intrinsic beam contamination in electron neutrinos, which is the dominant background when looking for this kind of oscillations. As an introduction to the analysis, the T2K experimental setup is presented, as well as the first published T2K oscillation results. The beam contamination measurement is made from a data set taken in 2010 and 2011 by the near detector of the experiment, located at 280 m from the beam source. Selection criteria are defined to create two samples of charged current electron neutrino interactions and charged current vii interactions, in order to calculate the ratio of the number of interactions and estimate the beam contamination. The result is in agreement with the beam simulation and shows a contamination in ve of the order of 1%. This thesis also contains a study of the alignment of the MicroMegas modules on the near detector TPCs. It is shown that misalignments between modules can introduce a bias in the momentum mea-surement of the charged particles crossing the TPCs. Cosmic ray data is used to determine the relative misalignments between modules and correct the related effects in the event reconstruction

Full textThe study of the fundamental building blocks of matter necessitates always more powerful accelerators. New particles are produced in high energy collisions of protons or electrons. The by-Products of these collisions are detected in large apparatus surrounding the interaction point. The 125 GeV Higgs particle discovered at LHC will be studied in detail in the next e⁺e⁻ collider. The leading project for this is called ILC. The team that I joined is working on the R&D for a Time Projection Chamber (TPC) to detect the charged tracks by the ionization they leave in a gas volume, optimised for use at ILC. This primary ionization is amplified by the so-Called Micromegas device, with a charge-Sharing anode made of a resistive-Capacitive coating. After a presentation of the physics motivation for the ILC and ILD detector, I will review the principle of operation of a TPC (Chapter 2) and underline the advantages of the Micromegas readout with charge sharing. The main part of this PhD work concerns the detailed study of up to 12 prototypes of various kinds. The modules and their readout electronics are described in Chapter 3. A test-Bench setup has been assembled at CERN (Chapter 4) to study the response to a ⁵⁵Fe source, allowing an energy calibration and a uniformity study. In Chapter 5, the ion backflow is studied using a bulk Micromegas and the gas gain is measured using a calibrated electronics chain. With the same setup, the electron transparency is measured as a function of the field ratio (drift/amplification). Also, several beam tests have been carried out at DESY with a 5 GeV electron beam in a 1 T superconducting magnet. These beam tests allowed the detailed study of the spatial resolution. In the final test, the endplate was equipped with seven modules, bringing sensitivity to misalignment and distortions. Such a study required software developments (Chapter 6) to make optimal use of the charge sharing and to reconstruct multiple tracks through several modules with a Kalman filter algorithm. The results of these studies are given in Chapter 7. The TPC technique has been applied to neutron imaging in collaboration with the University of Lanzhou. A test using a neutron source has been carried out in China. The results are reported in Chapter 8

Full textSoil respiration (Rs) is a major flux in the carbon cycle. The aim of this work was to describe spatial and temporal variations of soil respiration in poorly drained forest soils, with contrasted depth and duration of soil saturation. Different methods of Rs measurements were tested in order to validate our values and to develop comparisons of flux between sites. We had measured soil respiration, soil temperature and soil water and different soil and plants parameters. Rs is showing a high spatial variability and a high interaction between spatial and temporal variability. Ln July, 2003, soil respiration was significantly related to soil pH values, microbial biomass and delta-pH. The measurements results allowed to parameter an empirical model of Rs (exponential equation of soil temperature and log-normal equation of soil water content). Two parameters were adjusted : optimal soil water content (W so), i. E. The value of soil water content which gives the higher soil respiration, and soil respiration at 10°C and optimal soil water content (Rs10). Wso is correlated to bulk density and to the weight of litter on the soil in summer. Rs10 is a good estimating factor of annual soil respiration. It is correlated to an indicator of microbial activities and to basal area. This relation gives especially good perspectives in order to spatialize soil respiration at the forest scale. AlI these results are showing high interactions between spatial and temporal variability. However, seasonal variations may be explained by soil temperature and soil water content. Spatial variations at the forest stand scale may be related to soil and vegetation parameters

Full textMany industrial applications require the deposition of thin films on substrates of different shapes and characteristics. The cathodic magnetron sputtering process relies on mechanical collisions between particles to perform the sputtering and is widely used. However, particles are sputtered mainly as neutrals and it is impossible to modify their trajectory or their deposition energy. This thesis deals with the study of two methods developed to ionize the sputtered vapor. The first method relies the use of an additional high density plasma created with a radio-frequency coil. It requires an optimization of the power transfer toward the plasma. This optimization being performed, I present spatially resolved measurements of the species temperatures and absolute densities. Finally, I show that these measurements allow to control the deposited layers contamination. In the second method, high power pulses are applied directly on the magnetron cathode and ensure an ionization ratio of approximately 60%. The study was focused on the transport of sputtered species. Densities and temperature measurements are explained with a simple one dimensional diffusion model for neutrals. Ambipolar diffusion was used for ions

Full textCupressus dupreziana A. Camus is a native tree of the Tassili n'Ajjer (Central Sahara). This conifer was regarded as a remnant, now lacking any regenerative capacity, of a forest that grew in wetter times. Investigations carried out between 1997 and 2001 show that there are 233 alive trees. A disappearance rate of 8% over the last thirty years has been recorded. The discovery of some very young trees testifies to this cypress' capacity for regeneration in situ. Determination of radial growth reveals a juvenile vigour in spite of arid climatic conditions. Pedonthracological analysis reveals the presence of Pistacia atlantica Desf. , its disappearance from the Tassili must be attributed to human over-exploitation ; this pistachio, and Laperrine's olive, must have formed quite widespread groves in the Tassili.
