ABSTRACT Many viruses inhibit or retard apoptosis, a strategy that subverts one of the most ancient antiviral mechanisms. M11L, a myxoma virus-encoded antiapoptotic protein, has been previously shown to localize to mitochondria and block apoptosis of virus-infected cells (H. Everett, M. Barry, S. F. Lee, X. J. Sun, K. Graham, J. Stone, R. C. Bleackley, and G. McFadden, J. Exp. Med. 191:1487-1498, 2000; H. Everett, M. Barry, X. Sun, S. F. Lee, C. Frantz, L. G. Berthiaume, G. McFadden, and R. C. Bleackley, J. Exp. Med. 196:1127-1139, 2002; and G. Wang, J. W. Barrett, S. H. Nazarian, H. Everett, X. Gao, C. Bleackley, K. Colwill, M. F. Moran, and G. McFadden, J. Virol. 78:7097-7111, 2004). This protection from apoptosis involves constitutive-forming inhibitory complexes with the peripheral benzodiazepine receptor and Bak on the outer mitochondrial membrane. Here, we extend the study to investigate the interference of M11L with Bax activation during the process of apoptosis. Myxoma virus infection triggers an early apoptotic signal that induces rapid Bax translocation from cytoplasm to mitochondria, despite the existence of various viral antiapoptotic proteins. However, in the presence of M11L, the structural activation of Bax at the mitochondrial membrane, which is characterized by the occurrence of a Bax conformational change, is blocked in both M11L-expressing myxoma-infected cells and M11L-transfected cells under apoptotic stimulation. In addition, inducible binding of M11L to the mitochondrially localized Bax is detected in myxoma virus-infected cells and in M11L/Bax-cotransfected cells as measured by immunoprecipitation and tandem affinity purification analysis, respectively. Importantly, this inducible Bax/M11L interaction is independent of Bak, demonstrated by the complete block of Bax-mediated apoptosis in myxoma-infected cells that lack Bak expression. Our findings reveal that myxoma M11L modulates apoptosis by multiple independent strategies which all contribute to the blockade of apoptosis at the mitochondrial checkpoint.

Abstract:
This dissertation evaluates the degree of "fit" in employing Emotionally Focused Couple Therapy [EFCT] within the context of congregational ministry and/or a Christian counselling center to counsel evangelical couples who have experienced an "attachment injury." The present study introduces and examines the possibility that, given an appropriate level of theological reflection, EFCT is an appropriate counselling methodology for use by evangelical clergy and counselors. Beginning with an examination of the values, assumptions and practices of EFCT this study explores three dimensions of the interface between EFCT and ecclesiastical practice: a) a theological reflection on the values and assumptions that inform the practice of EFCT; b) a comparison of EFCT with the marital counselling theories of Howard Clinebell, Jr., Larry Crabb, Jr., H. Norman Wright, and Everett L. Worthington, Jr., noting how each of these theories conceptualizes and treats both the marital dyad and emotional experience; and, c) an examination of Christians' perceptions of, and receptivity to, this model. The research demonstrates that the pastoral adaptation of EFCT highlighted in this study was rated favorably but not superior to the other four models. Specifically, it is noted that those who had previously experienced marital counselling, pastoral or otherwise, appeared to be attracted to the EFCT model as it was presented, even though the exemplar did not explicitly incorporate either the use of scripture, prayer, religious homework, or spiritual themes such as forgiveness and mutuality in marriage. The fact that even in the absence of an explicitly spiritual emphasis EFCT received high ratings suggests there is something within the model that speaks to the committed evangelical believer. The study concludes that even though EFCT may not be known within the evangelical community the perspective it offers "fits" with the values of this part of the Christian community and seems to appeal to those who have previously experienced marital difficulties. As a result, it may be stated that EFCT offers a mode of intervention that is suitable for use with evangelical Christians.

Abstract:
This article proposes a hypothetical model for determining rate of diffusion of an innovation in a system. The model modifies Everett Rogers’ S-curve using an index created from Gartner’s hype cycle phases. Rogers’ model for technology innovation adoption demonstrates that cumulative technology diffusion in a system from zero through the late majority adopters’ phase forms a curve resembling the letter “S”. Hype cycles analyze the five emotional stages technology adopters go through from over-enthusiasm (hype) though disappointment until it plateaus (beginning of mainstream adoption). When numbers assigned to the phases of adoption from the hype cycle are used as multipliers and applied to the cumulative adoption data of an innovation (Rogers’ S-curve), the “S” becomes a “J”. With the J-curve you can determine the rate of innovation diffusion in an organization.
