Abstract This book contains a series of chapters reviewing the current scientific knowledge on RNAi, methods for developing RNAi systems in transgenic plants and a range of applications for crop improvement, crop production and crop protection. Some chapters examine both endogenous systems in transgenic plants and exogenous systems where interfering RNAs are applied to target plants, pests and pathogens. The biosafety of these different systems is examined and methods for risk assessment for food, feed and environmental safety are discussed. Finally, aspects of the regulation of technologies exploiting RNAi and the socioeconomic impacts of RNAi technologies are discussed.

Abstract:
There is a pressing need to develop novel therapies for myocardial infarction and heart failure, two conditions that affect over 20% of the world population. Despite important advances in achieving revascularization of the ischaemic myocardium and the usefulness of devices in assisting failing hearts, therapy for these conditions remains poor. The final extent of myocardial tissue loss after infarction is a major determinant of post-infarction mortality due to heart failure. In this chapter we review the current strategies aimed at counteracting injury due to acute myocardial ischaemia–reperfusion and the experimental approaches to achieve cardiac and vascular regeneration once damage has occurred. We critically discuss the possibility of inducing tissue restoration by gene transfer or exogenous cell implantation, and report on the exciting possibility of stimulating the endogenous capacity of cardiac regeneration using growth factors and small regulatory RNAs.

Abstract:
Reconstructing real-world system dynamics from time series data on a single variable is challenging because real-world data often exhibit a highly volatile and irregular appearance potentially driven by several diverse factors. NLTS methods help eliminate less likely drivers of dynamic irregularity. We set a benchmark for regular behavior by investigating how linear systems of ODEs are restricted to exponential and periodic dynamics, and illustrating how irregular behavior can arise if regular linear dynamics are corrupted with noise or shift over time (i.e., nonstationarity). We investigate how data can be pre-processed to control for the noise and nonstationarity potentially camouflaging nonlinear deterministic drivers of observed complexity. We can apply signal-detection methods, such as Singular Spectrum Analysis (SSA), to separate signal from noise in the data, and test the signal for nonstationarity potentially corrected with SSA. SSA measures signal strength which provides a useful initial indicator of whether we should continue searching for endogenous nonlinear drivers of complexity. We begin diagnosing deterministic structure in an isolated signal by attempting to reconstructed a shadow attractor. Finally, we use the classic Lorenz equations to illustrate how a deterministic nonlinear system of ODEs with at least three equations can generate observed irregular dynamics endogenously without aid of exogenous shocks or nonstationary dynamics.

Abstract:
Pregnant women experience a greater degree of sleep disturbance than their non-pregnant counterparts. Complaints range from sleep maintenance issues to excessive daytime sleepiness. Emerging evidence suggests that there is variability in sleep patterns and complaints which manifest differently among pregnant women. Moreover, it is well accepted that sleep disturbance can dysregulate normal immune and endocrine processes that are critically important to the health and progression of gestation. A possible consequence of sleep disturbance is an increased risk for adverse pregnancy outcomes. Then again, many endogenous and exogenous factors, including pregnancy-related physiological, hormonal, and anatomic changes, as well as lifestyle changes, can impact the degree and chronicity of sleep disturbance. Alas, there is still much to learn in terms of what women can/should expect with regard to the timing, degree, frequency, and/or severity of a specific pregnancy-related sleep disturbance(s), despite the number of published studies evaluating what sleep during pregnancy encompasses.

The immune response is a highly complex system that has developed to protect individuals from morbidity and mortality induced by exogenous exposures, including infections. As summarized in this chapter, alterations in the immune response, whether due to immunosuppressive or immune stimulatory effects, have important consequences with respect to cancer risk. Individuals with inherited immunological defects, acquired immunological deficiencies, chronic unresolved infections, and autoimmune conditions are at considerably increased risk for multiple cancers, suggesting an important role for the immune response in the development of cancer at various anatomical sites. Studies that have directly evaluated immunogenetic and immunological factors and cancer risk are beginning to identify specific immunological risk factors associated with individual cancers. Furthermore, technological advances have made it increasingly feasible to evaluate specific immunological factors and their relationship to cancer risk, suggesting that additional insights are likely in the coming years.
