Diplomová práce se zabývá experimentální studií vlastností spreje z několika malých tlakových vířivých trysek používaných ve spalovacích komorách proudových motorů. Po desetiletí byla snaha zlepšit jejich rozprašovací charakteristiky a stále jsou možnosti, které nebyly téměř prozkoumány. Výzkum byl proveden za použití různých geometrií jednotlivých dílů trysky za účelem zjištění jejich vlivu na sprej. Byl použit různý počet vstupních portů (2, 3 a 4), různé vířivé komůrky (kulová, kuželová, konvexní, nízká kuželová) a rozdílné návrhy obtokového otvoru (osové, mimosové). Důraz byl kladen na posouzení stability spreje, kde pulzující sprej negativně ovlivňuje provozní charakteristiky spalovací komory. Vlastnosti spreje byly proměřeny fázovým dopplerovským analyzátorem, cirkulární homogenita byla hodnocena mechanickým paternátorem a rozpad kapaliny byl vizualizován pomocí jednoduchého laserového stínografu.

The proposal is based on the tradition of architecture in Brno – clean lines and simple shapes. Another inspiration is green. The green place arises around the skyscraper, but also inside skyscraper. It becomes a natural part of its. In other words : the green grows through the building.

The subject of this diploma thesis is the architectural design of Skyscraper "EVO" (experimental high-rise building) located in Brno. The building integrates various functions (residential, restaurant, shops, office space and gallery) at relatively small built-up area. The aim of the thesis is to create the design of a high-rise building with appropriate scale and which creates space for interconnection of different functions and will become new dominant of the city. The proposed building is located in the part of the city called Veveří. The building site is defined by Šumavská and Veveří streets. The building is designed as a single rectangular prism shaped volume. The entrances to the building are at the level of surrounding terrain. The building has 31 aboveground floors and 9 underground floors. The simple shape of building is structured by horizontal bands of consoles which create shields against excessive sun exposure and vertical atriums embedded in the main volume. These atriums connect interior and exterior. This atrium create connection between interior and exterior and ease the penetration of natural daylight inside the building. The dominant material used is glass, which is applied both in exterior and interior. The glass used on glazing of atrium sis colored transparent photovoltaic glass. Bands are in white color.

The diploma thesis deals with a temperature field in a solid fuel during the combustion process. At the beginning of this thesis, research of available literature was conducted to obtain information about the composition of wood, properties affecting the combustion process, and the basics of heat transfer. Afterward, the temperature profiles of the samples were recorded during the measurement in the observation furnace. The temperature profiles of samples of six different sizes which were made of spruce and beech wood were subsequently evaluated and compared. Based on experimentally obtained data, a mathematical model was created in the OpenModellica software. Finally, the results of the mathematical model were compared with experimentally obtained data and appropriate conclusions were deduced.

The CST Microwave studio software has been used for draft simulations. The resulting tags have been subjected to parameter measuring – the reflection coefficient, the minimum capacity needed to switch on the tag, the maximum reading distance. The influence of the proximity of metal objects and other materials has also been examined.

This master´s thesis deals with the analysis of experimental telemetrical ECG records with intention to determine the long-term influence of anticancer drug sunitinib on the electrical activity of heart. A laboratory rat was chosen as a model organism for the experimental study carried out at the Department of Physiology at Faculty of Medicine, Masaryk university. The sunitinib was applied to the rats at an early age and the ECG was measured with a 20-week delay using the Stellar telemetry system. To measure the effect of sunitinib on the electrical activity of the heart chambers, an analysis of the duration of the RR and QT interval and the width of the QRS complex was chosen. These parameters were detected by the wavelet transform method. Statistical analysis was performed using nonparametric tests - the Wilcoxon signed rank test, the MannWhitney test and the Friedman Test. The obtained results suggest that the use of sunitinib has no long-term effect on the observed parameters for the chosen animal model. After extension of the study, the results obtained could contribute to assess the effect of drugs on electrical activity of the human heart several decades after sunitinib treatment termination.

Existuje mnoho různých způsobů jak analyzovat disperzní systémy a fyzikálně chemické processy ke kterým v takových systémech dochází. Tato práce byla zaměřena na charakterizaci těchto procesů pomocí metod harmonické fraktální analýzy. Obrazová data sledovaných systémů byly analyzovány pomocí waveletové analýzy. V průběhu práce byly navrženy různé optimalizace samotné analýzy, převážně zaměřené na odstranění manuálních operací během analýzy a tyto optimalizace byly také inkorporovány do softérového vybavení pro Harmonickou Fraktální Analýzu HarFA, který je vyvíjen na Fakultě chemické, VUT Brno.

The purpose of this diploma thesis was to find optimal operational parameters of Ranque–Hilsch vortex tube that would give the best results of temperature separation, and to create a numerical model of the device. Firstly, extensive research of current literature was done and analytical model of the tube was created. Then, the numerical model of the vortex tube was designed using Star-CCM+ software. Afterward, best fitting turbulence model was chosen to do the calculation and optimal geometrical parameters of the tube were obtained as a result of CFD simulation. Then, inlet nozzles for the tube were designed and manufactured. Experimental evaluation of the vortex tube and description of its optimal settings form a substantial part of the project. Finally, the data obtained from the experiment were compared to the results of numerical analysis and conclusions were deduced.

Severe cerebellar diseases include a wide group of cerebellar hereditary degenerations with different types of heredity, different pathogenesis and diverse phenotypic manifestation. Many types of mouse models can be used to investigate these diseases and their therapy. These mice carry spontaneous mutations or are genetically modified. The aim of this work was to analyse the dynamics of morphological changes of cerebellar degeneration in Lurcher mice using fluorescent double staining. We also performed transplantation of embryonic cerebellar tissue suspension in adult Lurcher B6CBA mice, B6.BR mice and wild-type mice of both strains with two-month survival after surgery and transplantation of identical suspension in adult Lurcher B6CBA mice and C3H mice and wild- type mice of both these strains with six-month survival after surgery. This project has shown that pelvic organ prolapse is linked with maternal age and delivery-related injuries. Up to every eight women may have a symptomatic prolapse already one year after their first delivery. Moreover, those with muscle injury have a higher short-term risk of pelvic organ prolapse development. To improve our knowledge we further explored the potential of an ovine model for prolapse and vaginal surgery. We showed that many anatomical and morphological features and vaginal wall changes induced by specific lifespan factors (first delivery, artificial menopause, and hormonal replacement) are similar to what is observed in women. We further used this model for testing novel implants and mesh visualization techniques. We believe that the ovine model can be used in future research on pelvic organ prolapse pathophysiology and novel treatment modalities.t

Throughout the scientific world the topic of cannabis usage and its link with psychosis seems to be discussed intensively. Considering the fact that the Czech Republic is a country with one of the highest prevalence of cannabis usage in the world it becomes a sensitive issue even in our circumstances. In the theoretical part of the work we attempted to review current knowledge of a link between cannabinoid system, canabis usage and psychosis and to point out possible future therapeutic potential of cannabinoids in the treatment of psychotic diseases. In the practical part of the work we focused on verification of propsychotic features of THC in animal model with particular attention to validation of acute subcutaneous admonistration of this drug as a novel cannabinoid model of psychosis. At the same time we tried to elucidate antipsychotic effect of CBD in this model. We tested these hypotheses in two behavioral tests (open field test, PPI ASR) and electrophysiologically (quantitative EEG). The aim of the project was to test female advantages of promiscuous and/or polyandrous mating. Although, previous studies reported improved reproductive success of females copulating with multiple males in many model species, including Eublepharis macularius, this study failed to demonstrate such a benefit of polyandry. Traits reflecting female reproductive success, i.e. the number of produced eggs, egg mass, hatchability, hatchling mass, growth and survival were not influenced by copulation with multiple partners, but some of them were dependent on female condition. Only initial stages of juvenile growth were influenced by female identity and/or investment.

1 ABSTRACT The success of cancer vaccines depends on factors associated with the vaccine, which define the main parameters of effective immune responses such as its size and quality, as well as on factors related with the host, represented by the immunosuppressive mechanisms that allow the tumor to escape recognition by the immune system or negatively influence the function of effector T-cells. Attenuated, non-replicating viruses are at present preferred as VACV for safety reasons. A problem may arise concerning their lack of immunogenicity. Through the deletions of non-essential genes, vaccination vectors are therefore developed based on attenuated rVACV capable of replication, which induce a strong immune response. Genes of various immunological adjuvants (e.g., genes for cytokines and costimulatory molecules) are inserted into the vectors for the purpose of eliminating the influence of the immunosuppressive mechanisms of tumors. The first part of the work describes our study of the influence of vCCI on biological properties of rVACV derived from the Prague strain. The present thesis is focused on studies of the importance of cholinergic, purinergic and nitrergic mechanisms in the healthy and inflamed rat urinary bladder, furthermore in which way specific pre-treatment affects the state of inflammation. Experiments were performed on prepared strips from normal healthy rats and cyclophosphamide-induced cystitis rats. In vitro contractile response of the strips to MeCh and ATP were investigated in the saline/CYP control group as well in the presence of five various antagonists and inhibitors. The rats were pre-treated by 4-DAMP (2mg/kg) as a M3, M5 muscarinic receptor antagonist; DPCPX (1mg/kg) as a P1A₁ purinoceptor antagonist; PSB1115 (1mg/kg) as a P1A2B purinoceptor antagonist; L-NAME (60mg/kg) as an inhibitor of eNOS and finally by suramin (10mg/kg) as a non-selective P2 purinergic receptor antagonist. Altered effects were observed via functional and morphological studies. The best results showed DPCPX and L-NAME pre- treated groups. It was found that DPCPX and L-NAME pre-treatments normalized contractile response to MeCh and ATP of the inflamed bladders and helped to reduce inflammatory signs. Cardiac atrophy, as a result of a long term mechanical unloading observed in patients treated with mechanical circulatory supports, represents a substantial obstacle in cardiac recovery. The first aim of this thesis was to evaluate the course of unloading-induced cardiac atrophy in healthy and failing heart. Mechanical unloading was studied in an experimental model of heterotopic heart transplantation in rats. Heart failure was experimentally induced by chronic volume overload achieved by creation of aorto-caval fistula. The second aim of this thesis was to discover if dietary induced increase in fatty acids concentration in heart tissue, specifically myristic, palmitic and palmitooleic, that are believed to promote physiological heart growth, could attenuate the course of cardiac atrophy. The third aim of this thesis was to examine if increased isovolumic loading obtained by intraventricular implantation of newly designed spring expander would attenuate the development of cardiac atrophy. Background: Psilocybin is today the most studied psychedelic substance in neuroscience for its psychotomimetic and therapeutic effects. One of the institutions in the Czech Republic that studies this substance is the National Institute of Mental Health, where a clinical evaluation of its effects and safety is underway. The action and safety of this substance is examined here in terms of acute intoxication, but also in terms of long-term effects. Aim: To determine how volunteers perceive the experience with psilocybin administered in a controlled experimental study environment and whether they have experienced any afterglow effects after the cessation of acute intoxication for 28 days after administration, as well as how these effects occur and how long they persist. Methods: Data was obtained using semi-structured interviews. The research group consists of 20 interviews with 10 healthy volunteers of the clinical study at NUDZ. Interviews were conducted on day 28 after administration of an active dose of psilocybin or placebo. A qualitative thematic analysis of these interviews was performed in order to identify topics within the framework set by the research questions. For school purposes seem suitable probeware (school experimental systems), that combine modern technology with concept of student-centered learning (eg inquiry based science education) that should successfully prepare students for lifelong learning. Title of diploma thesis: Haemonchosis as a model parasitosis in sheep - the results of its control during experimental study One of the current major issues with breeding domestic animals worldwide is anthelmintic resistance, which damages animals' health and reduces their production. Research workplaces want to identify the mechanisms of resistance development and find out the ways how to prevent it. The aim of this diploma thesis was to describe experimental infection by the model parasite Haemonchus contortus, which was established on Texel breed sheep. H. contortus is one of the important endoparasites; it lives in abomasum of ruminants and feeds with their blood. Faecal samples of all animals entering to the experimental studies were examined by the qualitative ovoscopic method and the animals were thoroughly dewormed by albendazole and monepantel. Then they were infected with larvae L3 of different H. contortus strains. Three strains had been used - ISE (all anthelmintic sensitive), IRE (benzimidazole resistant) and WR (multi-resistant strain). Translation initiation in the hepatitis C virus (HCV) occurs through a cap- independent mechanism that involves an internal ribosome entry site (IRES) capable of interaction with and utilization of the eukaryotic translational machinery. We focused on the structural configuration of the different HCV-IRES domains and the impact of IRES primary sequence variations on secondary structure conservation and function. For this purpose we introduced into our laboratory, methods such as denaturing gradient and temperature gradient gel electrophoresis for screening the degree of heterogeneity and total amount of HCV-IRES variability accumulated in HCV infected patients over a period of time. The selected samples showed variable migration pattern of the HCV-IRES (from all the patients) visualized in DGGE and TGGE, were sequenced and evaluated for translation efficiency using flow cytometry. In some cases, we discovered that multiple mutations, even those scattered across different domains of HCV-IRES, led to restoration of the HCV-IRES translational activity, although the individual occurrences of these mutations were found to be deleterious. Free radicals are engaged in many cellular actions. Recently, free radicals of versatile kinds reveal to be engaged in changes of the vascular wall composition i.e. in its remodelling. Reactive oxygen species (ROS), mostly of radical nature, are engaged it these actions actively. There are many fluorescent dyes detecting ROS intracellularly. We use mostly dichlorofluorescin, dihydrorhodamine, and dihydroethidium that differ in specificity to individual kinds of ROS. Preliminary studies in our laboratory revealed the opportunity to trace changes of the vascular wall composition during the neonatal developmental period of the laboratory rats heart using above mentioned substances. Cooperative, altruistic and fairness-exhibiting behavior is an important topic in evolutionary and behavioral biology and the mechanisms leading to its evolution, ultimate as well as proximate precursors, are subject of much research in biological as well as social sciences, theoretical as well as experimental work. In light of the life history theory, I focused on the connection of one's health state and cooperative behavior in humans and tested the hypothesis that more healthy individuals would manifest more cooperative tendencies (as they would have more opportunities of future interactions and long-term benefits), and conversely. The data, obtained from a sample of university students engaged in experimental games (Dictator Game, Ultimatum Game, Expanded Ultimatum Game, Trust Game, and Reversed Dictator Game) and a health and personality-focused questionnaire, did not corroborate this hypothesis. My other hypotheses - that better memory and lower temporal discounting would be related to more cooperative behavior (stemming from the conditions for reciprocal cooperation) - were supported by the data, albeit only partially in the case of memory. The aim of this diploma thesis is to make available and analyze the production called Revue from the Case, which premiered on the experimental stage of the State Theatrical Studio Prague on March 25, 1968. This premiere was preceded by the successes of individual parts of the production at EXPO 58 in Brussels and EXPO 67 in Montreal. Based on the preserved archive materials for this production, whose main authors were Ladislav Rychman, Jiří Procházka and Jaromír Staněk, I will also attempt to name the basic principles, benefits and limits of Laterna magika.
