Abstract:
The use of Curriculum-Based Measurement (CBM) in schools continues to increase, as it is a useful brief assessment of students’ basic academic skills. CBM measures are used for multiple tasks such as identifying students at-risk, creating local norms, monitoring students’ progress during interventions, and assisting with special education eligibility determinations. Much of the research has focused on CBM in the areas of math and reading. Relatively few studies have examined the area of CBMWritten Expression. Even fewer studies exist exploring the reliability among alternate writing forms. This study determined alternate form reliability coefficients for written expression probes at the second, fourth, sixth, and eighth grade levels using productiondependent, production-independent, and accurate-production scoring methods. When all grade levels are combined, alternate forms reliability coefficients are at a sufficiently high level. However, some scoring methods resulted in much higher correlations at younger grade levels than older grade levels. In general, the correlations were lower at the eighth grade level. Implications of the results for school personnel are discussed.

This thesis presents the hypothesis that aesthetic forms of expression - such as music, painting, video - can be used for direct information delivery. In contrast to text or verbal narrative techniques, which require a conscious act of transcoding, these aesthetic forms stimulate more direct, emotional response. Such a hypothesis could open a new channel for the delivery of various types of information, providing us, in situations of information overload, with a background information channel, leaving our foreground concentrated on the more thought-demanding tasks. To develop a viable system based on the notion of using aesthetic forms of expression for direct information delivery, we need to develop the elements from which the system would consist. This research defines the "emon", a small discrete unit of aesthetic expression, which generates an expected emotional response that can affect human behavior. The study is currently restricted to the domain of music, with candidate emons being 1-15 seconds long loops of audio that are currently assumed to be the only audio source perceived by the user. The emons are characterized as units of an independently describable value, without the necessity of connection / abstraction to / from other pattern units - i.e. if a specific emon is played we'll be able to relate to its qualities without accessing our knowledge about other emons. In this thesis I discuss the guidelines for emons' creation, describe the categorizations process, and report the results of emons' testing performed by a group of 14 users. Given the hypothesis that the musical emons (small musical patterns) can be used to provide cues that affect behavior, a need arises in a system that can provide a further validity to the usefulness of that approach. In the "Implementation" chapter I report the ongoing development of the GuideShoes wearable system, that assists user in navigating an open space, such as streets, by sequencing emons (musical patterns) as navigational cues. I also discuss the navigation tools written for this project.

This document describes and analyses the process of research and production for the experimental play Via Marina: A woman's journey. Based on a personal experience between the Mexican and the Canadian lifestyles, Via Marina presents a vision toward the benefits of transdisciplinarity for the stage. By definition, transdisciplinarity is a new approach marked by the rapid advance of technology that seeks to create new areas of knowledge working between, across and beyond disciplines. For my Master's project I expermented among New Media, Women's Studies and Theatre Arts to learn to intertwine these disciplines on stage by creating a theatrical production based on a transdisciplinary technique. As a result of two-years of practical and theoretical research, the creation of the production Via Marina tries to break the conventional theatre forms using a transdisciplinary mixture of video, dance and feminist exploration of gender roles. It is just beginning of the development of a new language transcending disciplines in my work as actress, director, artist - woman.

Les myopathies centro-nucléaires (CNM) sont un groupe de maladies musculaires sévères caractérisées par une faiblesse musculaire générale. La forme la plus sévère est la CNM liée à l’X (XLCNM), causée par des mutations de la Myotubularine (MTM1). D’autres formes autosomales existent et sont causées par des mutations de l’Amphiphysine2 (BIN1) et de la Dynamine2 (DNM2). Les mécanismes pathologiques menant aux CNMs restent à éclaircir et à ce jour aucune thérapie n’est disponible pour traiter les patients. Nous avons modulé les niveaux de protéines de MTM1, BIN1 et DNM2 dans des modèles murins de CNMs. Nous avons découvert que la sous-régulation de DNM2 sauvait le modèle murin de XLCNM et que la sur-expression de la protéine BIN1 humaine sauvait le modèle murin XLCNM ainsi que la forme autosomale causée par les mutations DNM2. Nous avons montré que MTM1 contrôlait l’adhésion cellulaire et le recyclage de l’intégrine dans les cellules musculaires. Nous avons observé que la sur-expression de BIN1 sauvait la dérégulation du recyclage de l’intégrine dans le modèle murin de XLCNM, ce qui suggère un lien fonctionnel entre BIN1 et MTM1 nécessaire pour l’adhésion focale au niveaux musculaire. Notre étude montre que MTM1, BIN1 et DNM2 participe à une voie de signalisation commune et que BIN1 et DNM2 représentent de nouvelles cibles thérapeutiques pour le traitement des CNM
Centronuclear myopathies (CNM) are a group of severe muscle disorder characterized by general muscle weakness. The most severe form is the X-linked CNM (XLCNM), caused by mutations in Myotubularin (MTM1). Others autosomal forms are caused by mutations in Amphiphysin 2 (BIN1) and Dynamin 2 (DNM2). The CNM pathomechanisms are still unclear and to date there are no therapies available to the disease. To investigate the pathways dysregulated in CNM and to identify new therapeutic strategies, we modulated MTM1, BIN1 and DNM2 protein levels in the CNM mouse models. We discovered that DNM2 downregulation rescued the XLCNM mouse model and that the overexpression of human BIN1 rescued the XLCNM and the autosomal dominant CNM form due to DNM2 mutations. We have also showed that MTM1 controls cell adhesion and integrin recycling in mammalian skeletal muscle and BIN1 overexpression rescued the integrin recycling alteration in XLCNM mouse model suggesting that MTM1 and BIN1 are functionally linked and necessary for focal adhesions in muscle. Therefore, our studies highlight that MTM1, BIN1 and DNM2 are in a common pathway and, BIN1 and DNM2 could be new therapeutic targets to treat the different CNM forms

The phenomenon pastorinha is one among the constituent expression forms of the Brazilian culture returned for the time of native and so that we obtained knowledge on him centered the focus of our research in Parintins, in Medium Amazon. In the systemization of the results of the research the proposal is to present, a priori, conceptual elements for the understanding of what come to be it referred phenomenon, of as it was built and which his/her function in the process of civilization of the human element that would be the Brazilian people, under the aegis of Jesus' Company. Done like this, through oral reports originating from of the social groups linked to the referred phenomenon in Parintins lifted his report in Medium Amazon and in consonance with the classic and other pertinent readings could build the present speech contemplating the form of as one plays the pastorinha for Brazil out and, particularly, in Parintins. Our analysis goes back to the different forms of as the Iberian woman it was seen by the glance of the Iberian cut societies, centered in the rules consuetudinárias and for the glance of the dramatic arts according to the pastoral Solemnities, between the Medium Age and Renaissance. Already in the New World, for the sexual abuses imputed the indigenous woman, for the defense of the environment, for the child's protection.
O fenômeno pastorinha é uma dentre as formas de expressão constituintes da cultura brasileira voltada para o tempo de natal e para queobtivéssemos conhecimento sobre ele centramos o foco da nossa pesquisa em Parintins, no Médio Amazonas. Na sistematização dos resultados da pesquisa a proposta é apresentar, a priori, elementos conceituais para o entendimento do que vem a ser o referido fenômeno, de como foi construído e qual a sua função no processo de civilização do elemento humano que seria o povo brasileiro, sob a égide da Companhia de Jesus. Feito assim, através de relatos orais oriundos dos grupos sociais vinculados ao referido fenômeno em Parintins levantamos o seu histórico no Médio Amazonas e em consonância com os clássicos e outras leituras pertinentes pudemos construir o presente discursocontemplando a forma de como se brinca a pastorinha pelo Brasil afora e, particularmente, em Parintins.Nossa análise se volta para as diferentes formas de como a mulher ibérica era vista pelo olhar das sociedades de corte ibéricas, centradas nas regras consuetudinárias e pelo olhar das artes dramáticas conforme os Autos pastoris, entre a Idade Média e o Renascimento. Já no Novo Mundo, para os abusos sexuais imputados à mulher indígena, para a defesa do meio ambiente, para a proteção da criança.

Abstract:
The strong association of the human leukocyte antigen HLA-B27 (B27), with spondyloarthropathies (SpA), was discovered more than four decades ago, yet the role B27 plays in disease pathogenesis remains unclear. It has been demonstrated that HLA-B27 can form non-classical FHC forms, including B27 FHC homodimers (B272) and misfolded FHCs. Two leading theories of SpA pathogenesis proposed that both cell surface NC-B27 (the B27 homodimer theory) and intracellular (the B27 misfolding and UPR theory) can drive immune responses in SpA. Several studies demonstrated that cell surface NC-B27 binds to immune receptors and suggested that these interactions can activate Th17 immunity. My DPhil project focuses on the investigation of the cell surface expression of NC-B27 molecules in cells and tissues obtained from AS patients and B27 TG2 rats. I used both existing reagents and a novel HD6 antibody, generated against B272, to examine the expression of HLA-B27 by flow cytometry, confocal microscopy and immunoprecipitation. This thesis provides evidence that the expression of HD6- and HC10-reactive molecules in AS patients is rather restricted to specific cell types (e.g. osteoclasts) and possibly particular environmental factors and tissue types (e.g. synovial fluid and joint tissue). My data using B27 TG2 rats demonstrated that resident joint cells express HD6- and HC10-reactive molecules before the onset of clinical manifestations. Furthermore, I showed increased expression of NC-B27 on antigen presenting cells (APCs), and CD161+ and CD8+ cell populations in blood and secondary lymphoid organs. Based on these findings I hypothesized that cell surface expression of NC-B27 may not only be crucial for activation of Th17-driven immune responses, but may also shift the balance between immune tolerance and activation. Importantly, the expression of potentially pathogenic HD6-reactive molecules is restricted to specific cell and tissue types hence HD6 antibody may be a potential future therapeutic agent in SpA.

Abstract:
Rotavirus, the major cause of life-threatening infantile gastroenteritis, is a member of the Reoviridae family and is considered to be the single most important cause of virus-based severe diarrheal illness in infants and young children particularly 6 months to 2 years of age in industrialized and developing countries. Infection in infants and young children is often accompanied by severe life threatening diarrhea, most commonly following primary infection. Diarrhea is the major cause of death among children around the world. Responsible for 4 to 6 million deaths per year according to the World Health Organization (WHO), diarrhea is especially dangerous for infants and young children. Globally, it is estimated that 1.4 billion episodes of diarrhea occur in children less than five years of age annually. In the United States alone, rotavirus causes more than 3 million cases of childhood diarrhea each year, leading to an estimated 55,000 to 100,000 hospitalizations and 20 to 100 deaths. And is a major cause of mortality for children in developing countries with approximately one million deaths annually. Rotaviruses belong to the family Reoviridae and are spherical 70-nm particles. The virus genome contains 11 segments of double-stranded RNA, each encoding a viral capsid or nonstructural protein. The identification of a rotavirus nonstructural protein gene (NSP4) encoding a peptide, which functions both as a viral enterotoxin and as a factor involved in the acquisition of host cell membrane during virus budding from cells, provides a new approach for mucosal immunization. Protein expression through chloroplast transformation system offers a number of advantages like high level of transgene expression, transgene containment via maternal inheritance, lack of gene silencing and position effect due to site specific gene integration and also the possibility of multi gene engineering in single transformation event. It is also an environmentally friendly approach due to effective gene containment and lack of transgene expression in pollen. To achieve an enhanced immune response to rotavirus infection, a fusion gene encoding the cholera toxin B subunit linked to rotavirus enterotoxin 90 aa protein (CTB-NSP490) was introduced into transgenic chloroplast and was transformed into chloroplast genome of Nicotiana tabacum by homologous recombination. The chloroplast integration of CTB-NSP4(90) fusion gene was confirmed in transgenic tobacco plants by PCR analysis. Southern blot analysis further confirmed site specific gene integration and homoplasmy. Immunoblot analysis of transformed chloroplast confirmed the expression of CTBNSP490 fusion protein both in monomeric and pentameric forms that retained the binding affinity to the enterocytes GM1 ganglioside receptor. Expression levels of CTB-NSP4 protein was quantified by GM1 ganglioside binding ELISA assay; mature leaves expressed CTB-NSP4 fusion protein to upto 2.45 % in total soluble protein, 100-400 fold higher than nuclear expression which was only 0.006%-0.026%. Antibody titration and virus challenge experiments will be performed in mice at Loma Linda University to evaluate the antigenic and protective properties of the chloroplast derived CTB-NSP4 fusion protein.

Nesta dissertação investigamos a relação entre a Filosofia e a prática de escrita de textos de maneira a tratar dos seguintes problemas: Existem formas alternativas de expressão e desenvolvimento da Filosofia além daquelas relacionadas aos recursos da linguagem verbal? Em caso afirmativo, através de que formas a Filosofia poderia se expressar? Como parece haver, na tradição filosófica Ocidental, uma vinculação necessária entre a Filosofia e a linguagem verbal, temos como objetivo repensar as práticas filosóficas dentro da universidade e analisar a potencialidade de pensamentos existente em diversos formatos de pensar. Inicialmente apresentamos as contribuições de uma abordagem inter/multidisciplinar para a Filosofia, de modo a apontar o paradigma da complexidade como um recurso apropriado para investigar problemas filosóficos da contemporaneidade. Entendemos que o paradigma da complexidade tem se delineado de forma a proporcionar uma virada na Filosofia que extrapola o domínio da linguagem verbal. Como um estudo de caso, discutimos características centrais da Filosofia brasileira no contexto da universidade pública. A partir da caracterização da Filosofia na universidade brasileira, analisamos os limites da linguagem verbal como forma de expressão de pensamentos. Por fim, discutimos o potencial de formas não verbais na reflexão filosófica, analisando suas contribuições e limites para o desenvolvimento de um filosofar genuíno.
This thesis investigates the relation between philosophy and the practice of writing texts, in order to examine the following issues: Are there alternative ways of expression and development in philosophy, beyond those related to the resources of verbal language? If so, through what forms could philosophy express itself? As there seems to be a necessary connection between philosophy and verbal language in the Western philosophical tradition, we aim to rethink philosophical practices within the university and to analyze the existing potential of various formats of thinking. Initially, we present the contributions of an inter/multidisciplinary approach to philosophy, pointing to the paradigm of complexity as an appropriate resource for investigating contemporary philosophical problems. We understand that the complexity paradigm has been designed to provide a shift in philosophy that takes it beyond the realm of verbal language. As a case study, we discuss the central features of Brazilian philosophy in the context of the public university. In this context, we analyze the limits of verbal language as an expression of thoughts. Finally, we discuss the potential of non-verbal forms in philosophical reflection, analyzing their contributions and limits for the development of genuine philosophizing.

The aim of the study was to evaluate the expression of immunosuppressive (FOXP3, NKG2A), and cytotoxic (perforin) or cytotoxic / immunomodulating (IFNγ) T-cell properties representing biomarkers in the peripheral blood CD8h T-cell population of patients with advanced renal cell carcinoma (RCC) or high risk cutaneous melanoma and healthy controls by multicolour flow cytometry. Determination of the percentage of functionally competing T-cell subsets (especially immunosuppressive) in the CD8hCD57+ T-cell subpopulation in future may serve as one of parameters enabling to assess the overall status of antitumor immune response and select cancer patients most suitable for antitumor immunotherapy while dismissing those to whom it would be ineffective or even harmful.

Abstract:
Women are subject to a number of societal recommendations about what it means to be an 'ideal' woman. These recommendations take the form of social scripts, constructing an idea of ideal femininity, which women must perform in order to be socially accepted and successful. 'Emphasized femininity', a white, Western, script of femininity is dominant and has been critiqued by feminists, social theorists, and individual women for the limits that it places on women's behaviour. As a result a number of alternative scripts of femininity have arisen. These scripts can provide alternatives to restrictive understandings of female sexuality and beauty - they can serve to challenge 'appropriate' feminine behaviour and hence allow women to live more freely. Raunch femininity is a contemporary alternative that uses sexually explicit public performance, and encourages specific body and dress norms, in an attempt to challenge the norms of emphasized femininity. This thesis looks at raunch femininity, specifically its norms of sexuality and beauty, in the hopes of understanding what the effects of such a script are on women's behaviour. Theoretical understandings and explanations of women's lives are often contradicted by reports that women provide of their lived experiences. For this reason, this thesis investigates the lived experiences of women who self-identify as subscribers to this script in order to assess to what extent superficial expressions of freedom have deeper effects on women's freedom. The tension between theory and empirical reports is evident. However, in many cases, the reports of research participants reveal that the script of raunch femininity, like other scripts of feminine behaviour, has its own limits that women must abide with in order to be accepted. This thesis argues that these limits outweigh the benefits of this script.

The purpose with this study is to analyze and interpret the curriculum and through that highlight what opportunities the curriculum sets for the teachers to teach in accordance with aesthetic learning processes and different forms of expressions that’s a part of the lecture and the learning of the social studies subject in F-3. There is a lack of research on the aesthetic learning process and forms of expression linked to F-3 core subjects and almost no research linked to social studies. This study analyzes curriculum documents from 2011, syllabuses from 2011 and comment material from 2011 in the subject’s history, geography, religious education and civics and the aesthetic subject’s art and music. I have then compared these with the curriculum, syllabuses, and comment material for the same subjects in the upcoming curriculum of 2022. The study shows there is plenty of opportunities for teachers to build on the aesthetic learning process and different forms of expressions in the social studies subjects for F-3 in their teaching. The result has been discussed in relation to former research of aesthetic learning processes and different expressions. This discussion shows that education through aesthetic learning processes and different forms of expressions promotes thinking and analyzing skills among individuals enhancing their ability to actively participate in society.
Syftet med studien är att analysera och tolka läroplanen samt därigenom synliggöra vilka möjligheter läroplanen ger lärare att arbeta med estetiska lärprocesser och olika uttrycksformer som en del av undervisningen och lärandet i de samhällsorienterande ämnena i grundskolan F-3. Det finns sparsamt med forskning gällande estetiska lärprocesser och uttrycksformer kopplat till kärnämnena i grundskolans F-3 och näst intill ingen forskning alls gällande koppling till de samhällsorienterande ämnena. Studien utgår ifrån en textanalys där de analyserade dokumenten består av läroplanen från 2011, kursplaner från 2011 och kommentarmaterial från 2011 i ämnena historia, geografi, religionskunskap och samhällskunskap samt de estetiska ämnena bild och musik. Jag har sedan jämfört dessa med läroplan, kursplaner och kommentarmaterial för samma ämnen i kommande läroplaner 2022.  Studien visar att det finns gott om möjligheter i styrdokumenten som uppmanar och lämnar plats för läraren att använda sig av estetiska lärprocesser och olika uttrycksformer i de samhällsorienterande ämnena för F-3 i undervisningen. Resultatet har sedan diskuterats gentemot tidigare forskning om estetiska lärprocesser och olika uttrycksformer. Diskussionen visar att undervisning genom estetiska lärprocesser och olika uttrycksformer skapar tänkande och analyserande individer som kan delta aktivt i samhällslivet.

Questo lavoro mira ad offrire una nuova prospettiva dalla quale leggere la dialettica fra cultura pagano-mitica e cultura cristiana che attraversa tutta la tradizione letteraria italiana. Tema comune alle due visioni del mondo, la trasformazione è figura guida dell’intera ricerca, figura che assume declinazioni differenti e prende il nome di metamorfosi nella cultura mitica e quello di conversione nella cultura cristiana. La novità della prospettiva consiste nel tentativo di una lettura di carattere retorico di questi due fenomeni, con lo scopo di formalizzare due tipi di poetica che corrispondano al tema della metamorfosi e a quello della conversione. Applicando al vasto materiale offerto da Divina Commedia, Gerusalemme liberata, Adone, Promessi sposi, tragedie manzoniane e da alcune poesie tratte dall’Alcyone di d’Annunzio gli strumenti derivati dagli studi antropologici sui sacrifici arcaici di Girard e dalle intuizioni di Bottiroli a proposito della traduzione letteraria di alcuni schemi logici propri alla cultura occidentale si è potuta constatare una corrispondenza costante tra le forme dei contenuti ‘metamorfosi’ e ‘conversione’ e le particolari forme dell’espressione con cui essi sono stati rappresentati. Una volta individuate, le particolari forme dell’espressione sono state codificate in figure retoriche, tanto che si è giunti ad indicare lo schema, in generale, dell’antitesi come marca propria alla poetica della metamorfosi e la figura della correctio come caratteristica della poetica della conversione.
This work aims at offering a new perspective from which it is possible to read the dialectic between the heathen-mythical culture and the Christian culture that runs through the whole literary Italian tradition. Transformation is the theme in common between the two visions of the world, it is the leading figure of the whole research and it assumes different forms. It takes the name of metamorphosis in the mythical culture and of conversion in the Christian culture. The novelty of this perspective consists in the attempt of a rhetorical reading of these two phenomena, in order to formalize two kinds of poetics., that could correspond to the theme of metamorphosis and conversion. Applying to the large materials offered by Divina Commedia, Gerusalemme liberata, Adone, Promessi sposi, tragedies by Manzoni and some poems from Alcyone by d’Annunzio the instruments deriving from the anthropological studies about archaic sacrifices by Girard and from the intuitions of Bottiroli about literary transposition of some logical schemes, which are typical of the Western culture, it was possible to notice a constant correspondence between forms of contents ‘metamorphosis’ and ‘conversion’ and the particular forms of the expression, with which they have been represented. Once they have been pointed out, the particular forms of expression have been codified in figures of speech, so that it was possible to indicate the overall scheme of the antithesis as the proper sing of the poetics of metamorphosis and the figure of correctio as the feature of the poetics of conversion.

Nesta dissertação, partimos da base teórica da semiótica francesa para abordar o sincretismo da linguagem cinematográfica, sistema semiótico no qual é possível obter a significação homogênea da forma códica sincrética a partir da sincretização de linguagens concorrentes. Procuramos estabelecer o lugar do cinema na teoria semiótica para que a análise dos objetos sincréticos possa ser elaborada de maneira ampla considerando todas as linguagens envolvidas em sua manifestação. Assim, partimos das colocações do conceito de sincretismo de Hjelmslev (1975), Greimas e Courtés (1979) e dos avanços teóricos de Beividas (1983) para aprofundar a discussão a respeito de como pode ocorrer o arranjo e a organização das linguagens participantes do sistema de sincretização na linguagem do cinema, tendo em vista que se colocariam como funções do sistema sincrético e também como funtivos de novas funções que seriam criadas. Pretendemos demonstrar com a proposta do modelo de sincretização da forma códica sincrética que um filme pode ser analisado em uma abordagem abrangente da sincretização para que se alcance a complexidade da linguagem que constrói sua significação por meio da organização de diversas formas códicas, as linguagens individuais que participam do sistema sincrético. Por fim, buscamos abarcar as peculiaridades formais e materiais da linguagem cinematográfica com a análise semiótica de seu sistema de significação.
In this dissertation, we depart from the theoretical basis of French semiotics to treat the syncretism of cinematic language, semiotic system in which it is possible to obtain the homogeneous signification of the syncretic code form from the syncretization of concurrent languages. We aim at establishing the place of cinema in semiotic theory so that the analysis of syncretic objects can be broadly elaborated considering all languages involved in its manifestation. Thus, we assume the conceptualization of syncretism by Hjelmslev (1975), Greimas and Courtés (1979) and the theoretical advances proposed by Beividas (1983) to deepen the discussion about how the arrangement and the organization of languages that participate in the system of syncretization in the language of cinema may occur, assuming that they would act as functions of the syncretic system, and also as functives of new functions which would be created. We intend to demonstrate with a model of syncretization of the syncretic code form that a movie can be analyzed in a broad approach of syncretization so that the complexity of the language that constructs its signification is achieved through the organization of several code forms, the individual languages that participate in the syncretic system. Finally, we aim at covering the formal and material peculiarities of cinematic language through the semiotic analysis of its system of signification.

When Jane Austen described her writing as ‘two inches of ivory’ she was being selfdeprecating, maybe even sarcastic. But as I began an investigation into short-short forms and feminine voices her two inches of ivory became poignant, creating an image of a woman writer inside a tiny white space or writing on a tiny white space. This thesis represents the paths I have chosen – the changes in direction and the twists and turns of my creative process during three years spent exploring ideas related to this image. This journey produced themes and questions which spill across three sections: Passing Through, A Middle Place; Short-short Turns; and Harvested Short-shorts. Each element explores the way short-short stories come to us as writers, readers and editors and also considers the responsibilities and processes involved with taking on these roles.

Durante o desenvolvimento de um neurônio, genes são ativados e desativados, a anatomia se forma e as funcionalidades emergem. Estes três componentes influenciam continuamente uns aos outros. O estudo da forma, função e expressão gênica nos neurônios e no cérebro permanece um tema desafiador e com potencial a ser explorado. Neste contexto, uma importante questão ainda a ser respondida é como quantificar o inter-relacionamento entre forma, função e genes. Para isso, foram realizadas atividades envolvendo caracterização e comparação da forma neuronal, o estudo de processos dinâmicos ocorrendo em redes de estruturas ramificadas, e a comparação entre expressões gênicas. Os dados da base pública NeuroMorpho, que possui quase 6.000 neurônios segmentados, foram caracterizados utilizando-se métodos estatísticos e foram analisados pelo conceito de morfoespaço proposto por McGhee. Outra base pública explorada foi o Mouse Allen Brain Atlas, com imagens de expressão gênica de cérebros de camundongo. Foi proposta a utilização de um método baseado em diagramas de Voronoi para a comparação da distribuição espacial de densidades de expressão gênica entre genes, com o propósito de encontrar correlações entre distribuições. Também foram gerados dados sobre raízes de feijão para o estudo da influência de sua estrutura ramificada na dinâmica de propagação de doenças, seguindo o modelo SIR (Suscetível-Infectado-Recuperado). Integrando os desenvolvimentos anteriores, foi proposto um arcabouço para mensurar a influência da expressão gênica ao longo da escala biológica. Este arcabouço permite mensurar a influência da expressão gênica (escala molecular) na morfologia dos neurônios (escala celular), avançando à escala topológica formada pelas conexões sinápticas, e alcançando o nível funcional das dinâmicas sobre essa rede. Nesse contexto, deve-se ressaltar que a influência da expressão gênica é direta sobre a morfologia e indireta sobre a topologia e a dinâmica. As informações obtidas a partir do arcabouço são relevantes na investigação de como a expressão gênica influencia todo o processo, desde o neurônio individual até o funcionamento cerebral. O arcabouço proposto fornece uma metodologia sistemática, com um conjunto de ferramentas para essas análises.
During the development of a neuron, genes are turned on and off, the anatomy is shaped and the functionality emerges. These three components influence each other continuously. The study of form, function and gene expression in neurons and brain is still challenging and has many issues yet to be explored. In this context, an important question yet to be answered is how to quantify the inter-relationship between form, function and gene expression. In this way, we developed activities involving characterization and comparison of the neuronal form, the study of dynamical processes occurring in networks of branching structures, and the comparison between gene expressions. The data of the public database NeuroMorpho, which comprise almost 6,000 segmented neurons, were characterized using statistical methods and were analyzed by the concept of McGhee\'s morphospace. Another public database that was explored was the Mouse Allen Brain Atlas, with images of gene expression of mouse brains. We proposed to use a method based on Voronoi diagrams to compare the spatial distribution of the gene expression densities between genes, in order to find correlations in the distribution. We also generated data on bean roots to study the influence of their branched structures in the dynamics of disease spread, following the SIR model (Susceptible-Infected-Recovered). Integrating the previous developments, we proposed a framework to measure the gene expression influence through the biological scale. This framework allows the measurement of the gene expression (molecular scale) influence in the morphology of the neurons (cellular scale), advancing towards the topological scale formed by the synaptic connections, and reaching the functional level of the dynamics over this network. In this context, it is worth to note that the gene expression influence is direct on the morphology and indirect on the topology and dynamics. The obtained information through the framework is important on the investigation of how the gene expression influences the whole process, since the individual neuron to the cerebral functioning. The proposed framework yields a systematic methodology with a toolbox to carry out these analyses.

Unstructured textual data is rapidly growing and practitioners from diverse disciplines are expe- riencing a need to structure this massive amount of data. Topic modeling is one of the most used techniques for analyzing and understanding the latent structure of large text collections. Probabilistic graphical models are the main building block behind topic modeling and they are used to express assumptions about the latent structure of complex data. This dissertation address four problems related to drawing structure from high dimensional data and improving the text mining process. Studying the ebb and flow of ideas during critical events, e.g. an epidemic, is very important to understanding the reporting or coverage around the event or the impact of the event on the society. This can be accomplished by capturing the dynamic evolution of topics underlying a text corpora. We propose an approach to this problem by identifying segment boundaries that detect significant shifts of topic coverage. In order to identify segment boundaries, we embed a temporal segmentation algorithm around a topic modeling algorithm to capture such significant shifts of coverage. A key advantage of our approach is that it integrates with existing topic modeling algorithms in a transparent manner; thus, more sophisticated algorithms can be readily plugged in as research in topic modeling evolves. We apply this algorithm to studying data from the iNeighbors system, and apply our algorithm to six neighborhoods (three economically advantaged and three economically disadvantaged) to evaluate differences in conversations for statistical significance. Our findings suggest that social technologies may afford opportunities for democratic engagement in contexts that are otherwise less likely to support opportunities for deliberation and participatory democracy. We also examine the progression in coverage of historical newspapers about the 1918 influenza epidemic by applying our algorithm on the Washington Times archives. The algorithm is successful in identifying important qualitative features of news coverage of the pandemic. Visually convincing results of data mining algorithms and models is crucial to analyzing and driving conclusions from the algorithms. We develop ThemeDelta, a visual analytics system for extracting and visualizing temporal trends, clustering, and reorganization in time-indexed textual datasets. ThemeDelta is supported by a dynamic temporal segmentation algorithm that integrates with topic modeling algorithms to identify change points where significant shifts in topics occur. This algorithm detects not only the clustering and associations of keywords in a time period, but also their convergence into topics (groups of keywords) that may later diverge into new groups. The visual representation of ThemeDelta uses sinuous, variable-width lines to show this evolution on a timeline, utilizing color for categories, and line width for keyword strength. We demonstrate how interaction with ThemeDelta helps capture the rise and fall of topics by analyzing archives of historical newspapers, of U.S. presidential campaign speeches, and of social messages collected through iNeighbors. ThemeDelta is evaluated using a qualitative expert user study involving three researchers from rhetoric and history using the historical newspapers corpus. Time and location are key parameters in any event; neglecting them while discovering topics from a collection of documents results in missing valuable information. We propose a dynamic spatial topic model (DSTM), a true spatio-temporal model that enables disaggregating a corpus's coverage into location-based reporting, and understanding how such coverage varies over time. DSTM naturally generalizes traditional spatial and temporal topic models so that many existing formalisms can be viewed as special cases of DSTM. We demonstrate a successful application of DSTM to multiple newspapers from the Chronicling America repository. We demonstrate how our approach helps uncover key differences in the coverage of the flu as it spread through the nation, and provide possible explanations for such differences. Major events that can change the flow of people's lives are important to predict, especially when we have powerful models and sufficient data available at our fingertips. The problem of embedding the DSTM in a predictive setting is the last part of this dissertation. To predict events and their locations across time, we present a predictive dynamic spatial topic model that can predict future topics and their locations from unseen documents. We showed the applicability of our proposed approach by applying it on streaming tweets from Latin America. The prediction approach was successful in identify major events and their locations.

Studien handlar om estetiska lärprocesser i ämnet engelska. Syftet med studien är att bidra till en ökad förståelse för hur pedagoger i årskurs 1 - 3 arbetar med estetiska uttrycksformer i engelskundervisning och hur de kombinerar det med icke-estetiskt lärande. Frågeställningarna undersöks mot bakgrund av tidigare forskning samt vad pedagoger på fältet har för åsikter, uppfattningar och erfarenheter om estetiska uttrycksformer och läromedel som kombination i engelskundervisningen på lågstadiet. Studien är baserad på två forskningsfrågor som undersöks och blir besvarade: Hur kombinerar pedagoger estetiska uttrycksformer och icke-estetiskt lärande i engelskundervisningen i årskurs 1 - 3? Hur ser pedagoger på relationen mellan estetiskt lärande och icke-estetiskt lärande i engelskundervisning i årskurs 1 - 3? Studien består av en teoretisk och en empirisk del där fokus ligger på estetiska lärprocesser med ingång i uttrycksformerna musik och drama samt ett icke-estetiskt lärande som lutar mot läromedel i engelskundervisning. Den teoretiska delen utgår från det sociokulturella perspektivet med Vygotskijs syn på estetiskt lärande där han vill ha in det estetiska perspektivet i skolans undervisning. I denna studien har det framkommit att med hjälp av estetiska lärprocesser som hjälpmedel förstärks elevernas möjligheter att nå kunskaper i engelska språket. Kroppsspråket och sinnena är också en viktig del som hjälper eleverna att på ett enklare sätt befästa engelskkunskaper. Kombineras det estetiska och icke-estetiska lärandet med varandra så blir det en relation mellan lärandeformerna som leder till ett varierat lärande i engelskundervisningen i grundskolan.
This study is about aesthetic learning processes in the English subject. The purpose of the study is to contribute with an increased comprehension of how teachers in grade 1 - 3 work with aesthetic learning processes and how they combine aesthetic learning with non-aesthetic learning in the English subject. The study draws on previous research as well as working teachers’ opinions, perception and experience of working with the combination of the aesthetic form of expression and the teaching media in the English subject in primary school. The foundation of the study lies on two questions which were examined and answered during the course of the study. The questions are as follows: How does the teachers combine aesthetic learning with non-aesthetic learning in the English subject in grade 1 - 3? What do teachers think about the relations between aesthetic learning and non-aesthetic learning in the English subject in grade 1 - 3? The study consists of a theoretical and an empirical section where the main focus lies on aesthetic learning processes drawing on music and drama, on the one hand, and non-aesthetic learning based on teaching media, on the other, in the English classroom. The theoretical framework of the study has its roots in the sociocultural perspective with Vygotsky’s view on aesthetic learning in education. It emerged in the study that with the aid of aesthetic learning processes the pupil’s knowledge of the English language has increased. The body language and senses have an important function in helping the pupil’s to acquire the English language in an easier way. If aesthetic learning is combined with non-aesthetic learning it creates a relation between the two learning processes which leads to a varied learning in the English subject in primary school.

A hiperplasia adrenal congênita (HAC) por deficiência da enzima 21-hidroxilase (21OH) é uma doença autossômica recessiva que compromete a síntese de cortisol e/ou aldosterona. É a causa mais frequente de distúrbio da diferenciação sexual 46,XX. Apresenta uma diversidade fenotípica, a qual é decorrente de mutações no gene CYP21A2. Observa-se forte correlação do comprometimento da atividade enzimática predita pelo genótipo com a forma de apresentação clínica e com os valores hormonais; entretanto, esta correlação não é observada com o grau de virilização pré-natal da genitália externa em mulheres com a forma clássica. Supomos que variações inter-individuais na ação, síntese e metabolismo dos andrógenos possam influenciar o fenótipo da virilização. Objetivos: avaliar se variantes alélicas em genes relacionados a ação, síntese ou metabolismo de andrógenos na vida fetal possuem um efeito modulatório na variabilidade fenotípica da virilização genital em mulheres com a forma clássica carreando genótipos 21OH semelhantes. Também serão avaliadas se diferenças na expressão tecidual local dos genes HSD17B5, SRD5A1, SRD5A2 e RA influenciariam esta variabilidade fenotípica da forma clássica. Casuística: foram selecionadas 187 mulheres com a forma clássica da HAC 21OH provenientes de 4 centros médicos. Dados clínicos, hormonais e o grau de virilização genital foram obtidos de forma retrospectiva da análise de prontuários. A intensidade de virilização genital foi classificada de acordo com a escala de Prader (P) e as pacientes foram divididas em 4 grupos: P I+II, P III, P IV e P V. As pacientes também foram agrupadas de acordo com o genótipo 21OH: grupo A carreadoras de mutações que predizem < 2% de atividade enzimática residual (n= 122) e grupo B carreadoras de mutações que predizem 3 a 7% de atividade residual (n= 58). Metodologias: foram amplificadas e re-sequenciadas as regiões que flanqueiam os exons dos genes CYP3A7, PXR e CAR. Para as variantes funcionais, foram re-sequenciados os exons 12-13 do POR e a região promotora do HSD17B5. As variantes V89L e A49T do SRD5A2 foram rastreadas por PCR-RFLP e o nCAG do RA por eletroforese capilar e análise pelo GeneScan. A determinação da expressão gênica em pele genital foi feita por PCR em tempo real utilizando os genes endógenos CYC, PGK1 e B2M. Os testes t-test, Mann-Whitney, Kruskal-Wallis, Fisher e regressão linear uni- e múltipla foram utilizados na análise estatística. Resultados: o Prader score no genótipo A variou de II a V (III: III IV) e no genótipo B de I a V (III: II - III) (P< 0,001). Foram encontradas em 2,5% dos alelos a variante CYP3A7*1C, em 24% CYP3A7*2, 31% rs2307424 CAR, 25% A503V POR, 33% -71G HSD17B5, 17% rs2518047 HSD17B5, 31% V89L SRD5A2 e em 1% dos alelos a variante A49T SRD5A2. Foi identificada associação das variantes rs2307424 CAR (P= 0,023 ;r2= 0,253) e rs2518047 HSD17B5 (P= 0,006; r2= 0,144) com o grau de virilização genital, tem sido encontradas em maior frequência no grupo de pacientes com virilização mais intensa. Todas as outras variantes não apresentaram associação com o Prader score (P> 0,05). As diferenças de expressão de todos os genes analisados em amostras de pele genital não foram estatisticamente significantes (P> 0,05), embora observou-se que em 4/7 amostras de pacientes com Prader score IV houve uma super-expressão do gene SRD5A2 em relação a 1/5 pacientes com Prader score III. Conclusão: neste estudo multicêntrico observamos que o genótipo 21OH se correlacionou com a intensidade de virilização genital em mulheres com a forma clássica. A variante rs2307424 do gene CAR, relacionada ao metabolismo pré-natal de andrógenos, e a variante rs2518047 do gene HSD17B5, relacionada à síntese de testosterona, associaram-se à fenótipos de virilização mais intensos. Não identificamos diferenças na expressão tecidual dos genes relacionados à síntese e/ou ação periférica de andrógenos em pacientes com os diferentes graus de virilização
Congenital Adrenal hyperplasia (CAH) due to 21-hydroxylase deficiency (21OH) is an autosomic recessive disorder characterized by an impairment in the cortisol and/or aldosterone synthesis, being the most frequent cause of 46,XX disorder of sex development. The disease presents a wide phenotypic variability resulting from different CYP21A2 gene mutations and a strong correlation has been observed among genotypes, clinical forms and basal hormone levels. However, this correlation is not observed regarding the degree of prenatal external genitalia virilization in females and an interindividual variability in the synthesis, metabolism and/or peripheral action of androgens could corroborate for these findings. Objectives: to evaluate if allelic variants in genes related to the androgen synthesis, metabolism and peripheral action could modulate the genital phenotype in CAH females bearing similar CYP21A2 mutations. Differences in the HSD17B5, SRD5A1, SRD5A2 and RA gene expression in genital skin were evaluated among patients with different degrees of external genital virilization. Patients: were selected 187 CAH females and clinical and hormonal data were retrospectively evaluated. The degree of external genitalia virilization was classified according to Prader (P) scores and patients were divided into 4 groups: P I+II, P III, P IV and P V. Patients were also grouped according to 21OH genotypes: group A bearing mutations predicting < 2% of residual enzymatic activity (n= 122) and group B between 3 to 7% (n= 58). Methodology: the exonic flanking regions of CYP3A7, PXR e CAR genes were PCR amplified and sequenced. The exons 12-13 of POR and the promotor region of HSD17B5 were sequenced to screen the functional polymorphisms. The V89L and A49T SRD5A2 alleles were screened by PCR-RFLP and the CAG polymorphic tract of AR gene by capillary electrophoresis and GeneScan analysis. The differential gene expression in genital skin was evaluated by real time PCR and the CYC, PGK1 e B2M housekeeping genes were used. The t-test, Mann-Whitney, Kruskal-Wallis, Fisher and uni- and multiple linear regression tests were used in statistical analysis. Results: Prader score in group A varied from II to V (III: III - IV) and in group B from I to V (III: II - III) (P< 0,001). The CYP3A7*1C allele was identified in 2.5% of alleles, CYP3A7*2 in 24%, rs2307424 CAR in 31%, A503V POR in 25%, -71G HSD17B5 in 33%, rs2518047 HSD17B5 in 17%, V89L SRD5A2 em 31% and A49T SRD5A2 in 1% of alleles. The rs2307424 CAR (P= 0.023; r2= 0.253) and rs2518047 HSD17B5 variants (P= 0.006; r2= 0.144) were associated with the degree of external genitalia virilization, and they were found in a higher frequency in more virilized patients. The remaining variants were not associated with Prader scores (P> 0.05). The HSD17B5, SRD5A1, SRD5A2 and RA gene expressions did not significantly differ between patients presenting Prader score III and IV (P> 0.05); however, 4/7 samples from patients with Prader IV and just 1/5 patients with Prader III presented an increased SRD5A2 expression. Conclusion: In this multicentric study the 21OH genotypes were correlated with the degree of external genitalia virilization in CAH females. The rs2307424 CAR and the rs2518047 HSD17B5 variants, related to the prenatal androgen metabolism and synthesis, respectively, explained some of the interindividual variability of genital phenotype in CAH females bearing similar CYP21A2 mutations. Differences in the expression of genes involved in the peripheral androgen action did not corroborate for the variability of genital phenotype in CAH

Abstract:
The intent of this essay is to shed light on the relevance and meaning of the concept of ”form” in Merleau-Ponty’s phenomenology. Drawing on his early texts as well as on the published notes of his first course held at the Collège de France, we argue that the notion of form essentially carries two meanings in the thought of the philosopher. One epistemological, stating that the perceived always assumes the figure-ground structure (signifying a figurative dimension of form). Another ontological according to which form designates the originary manifestation of the world in the event of expression (signifying a genetic dimension of form). Finally, we argue that the interplay of these two dimension of form lead Merleau-Ponty to an intuitive understanding of rhythm as the sensible manifestation of form, suggesting a potentially fruitful encounter, converging upon this notion, with the thought of Henri Maldiney.

Type is to be seen é uma plataforma ligada à rede que pretende fazer com que as pessoas, de uma forma geral, e os seus utilizadores, em particular, reflictam sobre as letras enquanto forma de expressão gráfica e não apenas como peças que em conjunto formam palavras. As letras são o menor elemento da tipografia, um conjunto de letras irá formar uma fonte, e os vários comportamentos de uma fonte constituirão um tipo de letra. Enquanto tema central do projecto, interessa-me estudar a tipografia, não tanto pelo lado do desenho de letras e seus pormenores técnicos, mas antes como forma (ou imagem), numa tentativa de diminuir o receio que existe no meio do design em relação a convencionalismos e tradições, nomeadamente no que diz respeito à funcionalidade e invisibilidade do design e legibilidade da tipografia, que por vezes não permitem que os trabalhos atinjam o seu máximo potencial expressivo. O projecto ganhou amplamente o título de “A tipografia como forma de expressão autónoma” devido à importância da componente teórica, onde se faz a contextualização da parte prática. Este site nunca poderia fazer sentido sem falarmos antes do modernismo e do advento dos meios digitais. Sendo assim, o primeiro capítulo corresponde ao estudo do pico do modernismo e aos "ismos" da sua arte. Eles constituíram uma das épocas mais marcantes e revolucionárias na arte e serviram para alterar a forma como as pessoas olhavam para os tipos de letra. Antes disto, a tipografia estava apenas ligada à imprensa mas, a partir das décadas de 1910/ 1920, as letras começaram a mover-se livremente nas páginas pela primeira vez. Foi adoptada uma atitude nova que iria marcar de forma profunda as décadas seguintes. O segundo capítulo concentra-se no advento do computador e da internet e no nascimento daquilo que pode ser chamado de era digital, tendo início na década de 1980. Com o aparecimento dos meios digitais e a invenção do Postscript, que veio tornar possível trabalhar a tipografia dentro dos computadores, a possibilidade de criar tipos de letras chegou progressivamente às mãos de todos os utilizadores. Por esta altura, o design gráfico tomava novos contornos e ganhava um destino mais amplo e um papel cada vez mais importante na sociedade. Vários foram os designers que contribuíram para uma espécie de anarquia organizada dentro da tipografia, que elevou o seu potencial expressivo ao mais alto nível. No terceiro capítulo é explicada a componente prática: o site propriamente dito. Aqui são descritas as várias actividades que o utilizador poderá realizar no site, apenas com recurso a letras, cor e formas geométricas básicas, numa experiência interactiva.

O fungo dimórfico Paracoccidioides brasiliensis, é o agente etiológico da paracoccidioidomicose, uma das micoses sistêmicas humanas mais prevalentes na América Latina. Componentes da cadeia respiratória mitocondrial são potenciais alvos quimioterapêuticos. Nesse sentido, em nosso trabalho demonstramos diferenças entre a cadeia respiratória do fungo P. brasiliensis e do hospedeiro mamífero. A respiração, potencial de membrana e fosforilação oxidativa mitocondrial de esferoplastos de P. brasiliensis foram avaliados in situ, os quais demonstraram a presença de uma cadeia respiratória funcional. A adição de ADP induziu a transição da respiração do estado de repouso para o fosforilativo, em esferoplastos energizados com succinato, NADH, NAD+ e substratos ligados ao complex I. A presença de uma NADH-ubiquinona oxidoredutase foi demonstrada pela capacidade das células oxidarem NADH exógeno, assim como, pela respiração insensível a rotenona e sensível a flavona. Além disso, a respiração mantida por NAD+ foi sensível a rotenona e flavona, sugerindo que vias metabólicas citosolicas contribuem para a produção de NADH e substratos ligados ao complexo I. Foi demonstrado também que os níveis de expressão do complexo I e da NADH desidrogenase alternativa alternam-se durante a curva de crescimento de leveduras de P. brasiliensis. A respiração induzida por NADH ou succinato foi parcialmente inibida por KCN ou antimicina A e sensível à inibição por BHAM, indicando a presença de uma oxidase alternativa. A fim de se caracterizar esta enzima oxidase alternativa, o seu gene foi clonado e expressado em xii S. cerevisiae e E. coli, o qual conferiu uma respiração resistente a cianeto e sensível a BHAM. S. cerevisiae expressando oxidase alternativa mostraram uma menor taxa de multiplicação celular e diminuição na geração de EROs. Nós também observamos que inibidores da cadeia transportadora de elétrons retardaram a transição de micélio para levedura em culturas de P. brasiliensis. Além disso, estes inibidores e outras drogas geradoras de EROs aumentaram a expressão do gene da oxidase alternativa, assim como a produção de EROs em leveduras de P. brasiliensis.
Paracoccidioides brasiliensis, a thermically dimorphic fungus, is the etiological agent of endemic paracoccidioidomycosis, one of the most prevalent human systemic mycosis in Latin America. Components of the respiratory chain constitute potential pharmacological targets, and here are reported differences between the respiratory chain of the mammalian host and the fungus P. brasiliensis. Respiration, membrane potential and oxidative phosphorylation of mitochondria from P. brasiliensis spheroplasts were evaluated in situ, which demonstrated the existence of a functional respiratory chain. Adenosine 5\'-diphosphate (ADP) induced a transition from resting to phosphorylating respiration in mitochondria energized by succinate, NADH, NAD+ and complex I linked substrates. The presence of an alternative NADH-ubiquinone oxidoreductase was indicated by: the ability of the fungus to oxidize exogenous NADH; the insensitivity substrate-supported respiration to rotenone and sensitivity of this respiration to flavone. In addition, the sensitivity of NAD+-supported respiration to rotenone and flavone suggest that citosolic pathways contribute to NADH and complex I linked substrates production. Moreover, it was demonstrated that expression levels of complex I and alternative NADH dehydrogenase change during P. brasiliensis growth curve. The partial sensitivity of NADH or succinate-supported respiration to antimycin A and cyanide, as well as the sensitivity to BHAM, indicates the presence of an alternative oxidase. Therefore, to characterize the alternative oxidase its gene was cloned and heterologously xii expressed in S. cerevisiae and E. coli, wich confered, a cyanide resistant respiration and BHAM sensitivity. Moreover, S. cerevisiae that expressed alternative oxidase showed slower growth rate and decreased ROS generation. We also observed that electron transport pathways inhibitors delayed the P. brasiliensis mycelium to yeast transition in cultures. Besides, these inhibitors and other ROS generated drugs increase the ROS production and altenative oxidase expression.

The aim of this research is to design and implement a program that will be able to manipulate multiple algebraic expressions and transform them to a 'normalised form' . The expression to be manipulated is user defined (the user will be able to input an expression or read it from a file). This expression will be read in by a scanner and checked with a parser using a set of grammar rules which are to be defined later on. The expression will be able to contain constants (both real and imaginary), identifiers (strings or individual characters) and operators which include +, -, *, /, ( ), A (power sign), E (exponential sign for constants). The algebraic manipulation process simplifies and sorts the expression into order. The simplification involves removing all bracketed expressions by various methods including multiplying them out. Like terms will then be collected together and sorted into a user defined order. The order can be alphabetical, terms in ascending or descending power etc. For rational expressions , the whole expression is converted into one fraction. The program will also be able to inform the user of the total degree of the expression or if, for example, the expression considered is a polynomial in x, then the program will inform the user of the degree of the polynomial with respect to x. The program is built on a PC platform and an Object Oriented language will be used. The language initially used was Borland Turbo Pascal version 7 which uses Turbo Vision (Object Oriented data structures and user interface), but with the advent of 32 bit programming Borland C++ 5 was considered a better choice. The STL (Standard Template Library) is used for the data structures and OWL (Object Windows Library) is used for the user interface.

In this dissertation, we address the problem of generative object categorization in computer vision. Then, we apply to the classification of facial expressions. For the first part, we are inspired by the method Hierarchical Dirichlet Processes to generate intermediate mixture components to improve recognition and categorization, as it shares with documents modelling topic two similar aspects: its nonparametric and its hierarchical nature. After we obtain the set of components, instead of boosting the features as Viola and Jones, we try to boost the components in the intermediate layer to find the most distinctive ones. We consider that these components are more important for object class recognition than others and use them to improve the classification. Our target is to understand the correct classification of objects, and also to discover the essential latent themes sharing across multiple categories of object and the particular distribution of the latent themes for a special category. In the second part, regarding the relation between basic expressions and corresponding facial deformation models, we propose two new textons, VTB and moments on spatiotemporal plane, to describe the transformation of human face during facial expressions. These descriptors aim to catch both general shape changes and motion texture details. The dynamic deformation of facial components is so captured by modelling the temporal behaviour of facial expression. Finally, SVM based system is used to efficiently recognize the expression for a single image in sequence, then, the weighted probabilities of all the frames are used to predict the class of the current sequence. My thesis includes finding the proper methods to describe the static and dynamic aspects during facial expression. I also aim to design new descriptors to denote characteristics of facial muscle movements, and furthermore, identify the category of emotion
Dans cette thèse, nous avons abordé la problématique de la classification d'objets puis nous l'avons appliqué à la classification et la reconnaissance des expressions faciales. D'abord, nous nous sommes inspirés des processus de Dirichlet, comme des distributions dans l'espace des distributions, qui génèrent des composantes intermédiaires permettant d'améliorer la catégorisation d'objets. Ce modèle, utilisé notamment dans la classification sémantique de documents, se caractérise par le fait d'être non paramétrique, et d'être hiérarchique. Dans une première phase, l'ensemble des composantes intermédiaires de base sont extraites en utilisant l'apprentissage bayésien par MCMC puis une sélection itérative des classifiers faibles les plus distinctifs parmi toutes les composantes est opéré par Adaboost. Notre objectif est de cerner les distributions des composantes latentes aussi bien celles partagées par les différentes classes que celles associées à une catégorie particulière. Nous avons cherché dans cette seconde partie à appliquer notre approche de classification aux expressions faciales. Ce travail a consisté à trouver les méthodes adéquates pour décrire les aspects statiques et dynamiques au cours de l'expression faciale, et donc à concevoir de nouveaux descripteurs capables de représenter les caractéristiques des mouvements des muscles faciaux, et par là même, identifier la catégorie de l'expression

ENGLISH ABSTRACT: This study is aimed at establishing what personal catharsis is, specifically defining what it is and under what conditions it may occur. During the creation and execution of an autobiographical solo, I had gone through a process of attaining what has since been labelled personal catharsis and recognition of the other, which gave arise to me finding confidence and discovering my creative voice of identity. However, it was not known when or how these processes took place. In addition I was not certain exactly what personal catharsis was in the context of me and my identity. An investigate was also needed into the possibility of generalising personal catharsis as a process that could guide people towards gaining confidence and finding their creative voice of identity. In this thesis, the process of the creation of the autobiographical solo is deconstructed. In doing so the journey of the discovery of the other is highlighted and the role of recognition between the inner expressive form and the external expressive form. Through the means of a source study the concepts of behaviour and the other in reference to identity construction and performance of everyday self are discussed. Then personal catharsis is explored in comparison to the traditional use of the term. Using performance-based research methodologies in conjunction with a questionnaire, a 12-week series of workshops was held to explore the generalisation of personal catharsis and the issues that arose during the study. The workshops were aimed at investigating the possibility of creating a process to attain personal catharsis. It was found that personal catharsis is a subjective process that takes place through the recognition of the external expressive form. It is through this recognition that the dislocation of self can be reconnected and the other can become more transparent to the internal expressive form. This process is readily attained through the exploration of the character of self. However, although new behaviour can result from personal catharsis, the individual needs to choose the internal expressive form as the dominant part in expressive behaviour.
AFRIKAANSE OPSOMMING: Die doel van hierdie studie is om te bepaal en te beskryf wat presies persoonlike katarsis is en hoe dit as proses verloop. Tydens die skepping en uitvoering van ʼn outobiografiese solo het ek die bereiking van persoonlike katarsis – soos ek dit sedertdien gedoop het – en herkenning van die ander beleef. Dit het daartoe gelei dat ek selfvertroue en die skeppende stem van my identiteit gevind het. Tog het ek nie besef wanneer of hoe hierdie prosesse plaasgevind het nie. Daarby was ek nie seker wat presies persoonlike katarsis in die konteks van my wese en identiteit was nie. Ek wou ook die moontlikheid ondersoek om persoonlike katarsis as proses te veralgemeen sodat dit mense daartoe kon lei om selfvertroue en die skeppende stem van hul identiteit te vind. In hierdie tesis dekonstrueer ek die skepping van die outobiografiese solo as proses. Daardeur werp ek lig op die ontdekkingsreis na die ander en die rol wat herkenning tussen die interne ekspressiewe vorm en die eksterne ekspressiewe vorm speel. Die begrippe gedrag en die ander in verhouding tot die konstruksie van identiteit en die uitvoering van die alledaagse self word deur middel van ʼn literatuurstudie bespreek. Hierdie proses word maklik deur verkenning van die karakter van die self behaal. Alhoewel persoonlike katarsis nuwe gedrag tot gevolg kan hê, moet die individu egter die interne ekspressiewe vorm as die dominante deel in ekspressiewe gedrag kies.

Abstract:
This research aimed to investigate form as a possible element of developing the stage performers consciousness and visual sensitivity. Form should be understood here as the inner and essential structure to construct space and materials in which will reflect the artists subjective impression. His/Her way of shaping space will reveal his/her thoughtsand sensation. In this work, both artistic and visual processes are based on the principles of visual perception and the complex semiotics of a theatrical event. Thus, Gestalt theory will be used to elucidate the relationship between psychophysical perception and form. Besides, Ernani de Castro Malettas Polyphonic Performance Concept helps to explain and justify the presence of various voices in the making up of a theatrical phenomenon. Finally, Wassily Kandinskys Theory of Forms synthesizes those constituent elements of plastic and scenic arts. As a result of such analysis, a term is created, namely Corpoforma (bodilyformed). This not only helps distinguishing the dialogic artistic and theoretical aspects involved in such a discussion, but also encourage to reflection as well as to generate procedures that may lead to the understanding and a conscious use of form in the scenic body. It, consequently, will improve an organic organization involving form and expressive contents.
Ancorada no conceito de Atuação Polifônica, proposto por Ernani de Castro Maletta e que pressupõe a incorporação e apropriação consciente das diversas vozes que expressam e compõem o fenômeno teatral, a pesquisa teve como objetivo investigar, a partir das ArtesPlásticas, o elemento forma como um dos possíveis meios de desenvolver a sensibilidade visual e a consciência corporal do artista cênico. Forma entendida, aqui, como a estrutura essencial e interna, de construção de espaço e de matéria a qual permite a ressonância dasubjetividade pensamentos e sensações de seu criador, na configuração espacial externa de seus limites. Tanto o processo de organização visual quanto o processo artístico abordados na pesquisa realizada baseiam-se na percepção visual e, assim, a análiseestrutural da teoria da Gestalt colabora no esclarecimento das relações psicofísicas entre percepção e forma. A síntese da transposição dos elementos constitutivos das formas artísticas Plástica e Cênica apóia-se conceitualmente na Teoria das Formas de WassilyKandinsky. Cria-se o termo Corpoforma para distinguir o processo de investigação dialogal entre tais aspectos artísticos no intuito de apontar caminhos, suscitar reflexões e gerar procedimentos acerca da compreensão e uso consciente do elemento forma, literalmente incorporado em um corpo cênico, permitindo a organicidade estrutural entre forma e conteúdos expressivos.

L'animation expressive permet des styles de mouvements exagerés et artistiques comme l'étirement de parties du corps ou encore l'animation de créatures imaginaires comme un dragon. Créer ce genre d'animation nécessite des outils assez flexible afin de déformer les personnages en des poses quelconques, ainsi que de pouvoir contrôler l'animation à tout moment dans le temps. L'approche acutelle pour l'animation expressive est le keyframing: une approche manuelle avec laquelle les animateurs déforment leur personnage un moment spécifique dans le temps en cliquand et glissant la souris sur une partis spécifique du corps---un à la fois. Malgré le fait que cette approche soit flexible, il est difficile de créer des animations de qualité qui suivent les principes artistiques, puisque le keyframing permet seulement qu'un contrôle local spatiallement et temporellement. Lorsqu'ils dessinent des poses ou des mouvements, les artistes s'appuient sur différentes abstractions sous forme de croquis qui facillitent la réalisation de certain principes artistiques. Par example, certains animateurs dessinent des lignes d'action afin de créer une pose plus lisible et expressive. Afin de coordonner un mouvement, les animateurs vont souvent dessiner des abstractions de mouvement comme des demi-cercles pour des sauts, ou des boucles pour des pirouettes---leur permettant de pratiquer la coordination du mouvement. Malheureusement, ces outils artistiques ne font pas partis de l'ensemble d'outils de keyframing actuelle. Le fait que l'on ne puisse pas employer les même outils artistiques pour animater des personnages 3D a une forte conséquence: les outils d'animation 3D ne sont pas employés dans le processus créatif. Aujourd'hui, les animateurs créent sur du papier et utilisent le keyframing seulement à la fin pour réaliser leur animation. La raison pour laquelle nous n'avons pas ces outils artistiques (ligne d'action, abstractions de mouvement) en animation 3D, est parce qu'il manque une compréhension formelle de ceux-ci qui nous permettrais d'exprimer la forme du personnage---potentiellement au cours du temps---en fonction de la forme de ces croquis. Ainsi la contribution principale de cette thèse est une compréhension formelle et mathématique des abstractions de forme et de mouvement courrament employées par des artistes, ainsi qu'un ensemble d'algorithme qui permet l'utilisation de ces outils artistiques pour créer des animations expressives. C'est-à-dire que les outils développés dans cette thèse permettent d'étirer des parties du corps ainsi que d'animer des personnages de différentes morphologies. J'introduis aussi plusieurs extentions à ces outils. Par example, j'explore l'idée de sculpter du mouvement en permettant à l'artiste de dessigner plusieurs couches de mouvement une par dessus l'autre, de twister en 3D les croquis, ou encore d'animer un croquis ligne comme un élastique. Les contributions principales de cette thèse, aussi résumé ci-dessous: -La ligne d'action facilitant la création de poses expressives en dessinant directement le flow complet du personnage. -La courbe spatio-temporelle qui permet de spécifier un mouvement coordoné complet avec un seul geste (en dessinant une seule courbe), applicable à n'importe quel personnage 3D. -Un algorithme de matching rapide et robuste qui permet du ``squash and stretch''. -La ligne d'action élastique avec des attachements dynamiques à la ligne permettant d'animer un personnages à plusieurs jambes (bras) avec une seule ligne 2D animée
Free-form animation allows for exaggerated and artistic styles of motions such as stretching character limbs and animating imaginary creatures such as dragons. Creating these animations requires tools flexible enough to shape characters into arbitrary poses, and control motion at any instant in time. The current approach to free-form animation is keyframing: a manual task in which animators deform characters at individual instants in time by clicking-and-dragging individual body parts one at a time. While this approach is flexible, it is challenging to create quality animations that follow high-level artistic principles---as keyframing tools only provide localized control both spatially and temporally. When drawing poses and motions, artists rely on different sketch-based abstractions that help fulfill high-level aesthetic and artistic principles. For instance, animators will draw textit{lines of action} to create more readable and textit{expressive} poses. To coordinate movements, animators will sketch textit{motion abstractions} such as semi-circles and loops to coordinate a bouncing and rolling motions. Unfortunately, these drawing tools are not part of the free-form animation tool set today. The fact that we cannot use the same artistic tools for drawing when animating 3D characters has an important consequence: 3D animation tools are not involved in the creative process. Instead, animators create by first drawing on paper, and only later are 3D animation tools used to fulfill the pose or animation. The reason we do not have these artistic tools (the line of action, and motion abstractions) in the current animation tool set is because we lack a formal understanding relating the character's shape---possible over time---to the drawn abstraction's shape. Hence the main contribution of this thesis is a formal understanding of pose and motion abstractions (line of action and motion abstractions) together with a set of algorithms that allow using these tools in a free-form setting. As a result, the techniques described in this thesis allow exaggerated poses and movements that may include squash and stretch, and can be used with various character morphologies. These pose and animation drafting tools can be extended. For instance, an animator can sketch and compose different layers of motion on top of one another, add twist around strokes, or turning the strokes into elastic ribbons. The main contributions of this thesis are summarized as follows: -The line of action facilitating expressive posing by directly sketching the overall flow of the character's pose. -The space-time curve allowing to draft full coordinated movements with a single stroke---applicable to arbitrary characters. -A fast and robust skeletal line matching algorithm that supports squash-and-stretch. -Elastic lines of action with dynamically constrained bones for driving the motion of a multi-legged character with a single moving 2D line

Le rendu expressif a pour objectif de développer des algorithmes qui donnent la possibilité aux utilisateurs de créer des images artistiques. Il permet non-seulement de reproduire des styles traditionnels, mais surtout de communiquer un message spécifique avec un style qui lui correspond. Dans ce manuscrit, nous proposons de nouvelles solutions pour réintroduire la forme, souvent masquée dans les images réalistes. Nous montrons tout d'abord comment extraire les informations de surface pertinentes sur des objets 3D dynamiques, en nous basant sur les caractéristiques du système visuel humain, de sorte à obtenir des informations qui fournissent des niveaux de détail automatiques tout en prenant le point de vue en compte. Dans un deuxième temps, nous utilisons ces données extraites à la surface des objets 3D pour les intégrer en temps réel dans des styles variés, allant du rendu minimaliste noir et blanc au dessin au trait, en passant par des résultats réalistes
Expressive rendering aims at designing algorithms that give users the possibility to create artistic images. It allows to produce traditional styles, but also to convey a specific message with its corresponding style. In this thesis, we propose new solutions for enhancing shape, often hidden in realistic images. We first show how to extract relevant surface features on 3D dynamic scenes, taking the human visual system into account, in order to be able to control level-of-details. In a second step, we integrate this information in a variety of styles: minimalist black and white, realistic, or line-based renderings

Abstract:
The bloodstream form of Trypanosoma brucei differentiates into the stumpy form in the mammalian bloodstream, completing differentiation into the procyclic form on uptake by the tsetse fly. The underlying genetic events occurring during this differentiation process in pleomorphic cell lines were investigated through whole-genome microarray studies of key time points during differentiation from stumpy form cells to the procyclic form found in the insect midgut. The microarray was extensively validated and bioinformatic experiments conducted to detect motifs over represented in stumpy form or slender form cells. A positional-dependent motif was identified that was over represented in stumpy form cells, possibly representing a regulatory domain. The transcripts found to be enriched in stumpy form cells included a chloride channel, although RNAi directed against this gene showed no phenotype, suggesting the protein is redundant, as three other homologous proteins exist in the genome and showed similar mRNA profiles on the microarray. Stumpy form cells are G0 arrested and two proteins implicated in G0/G1 regulation in other organisms, Target of Rapamycin (Tor) and Cdh1, were investigated in T. brucei to determine whether these proteins were involved in differentiation. The result of depletion of either protein was rapid cell death in bloodstream form cells, although treatment with the drug rapamycin did not have any effect on the cells in contrast to other eukaryotes where this drug causes G1 arrest. A method for synchronisation of bloodstream form cells was also designed using a supravital dye and flow cytometry to allow investigation into cell cycle-dependent processes. This method was particularly suitable for harvesting populations enriched in G0/G1 stage cells, however differentiation of the isolated G0/G1 and G2/M populations did not show significantly different differentiation kinetics.

Abstract:
The transmission of Trypanosoma brucei, the causative agent of human African trypanosomiasis, depends upon the development in the bloodstream of 'stumpy forms' from non-transmissible 'slender forms'. In stumpy forms many mRNAs are downregulated and translation is generally repressed. However, a small subset of genes escape this repression and are upregulated, presumably as an adaptation for transmission. To understand the basic of this, regulatory sequences within the 3'UTR of a major stumpy-enriched transcript (an ESAG9 gene) have been characterised. This identified a signal responsible for gene silencing in slender forms and gene activation when cells develop to stumpy forms. An investigation was made of upstream open reading frames (uORFs) as a mechanism for the control of stumpy form gene expression. No evidence was found of uORF control, but one gene investigated was found to produce two transcripts through trans-splicing at different sites. These transcripts, which were found to exhibit some differential abundance between life-cycle stages, would generate a long and short form (from an internal ATG) of the encoded protein. Both are predicted to contain a UBA/TS-N (ubiquitin associated) domain, however, the longer form of the protein is also predicted to contain a transmembrane helix and cleavable signal peptide, suggesting a different localisation. However, ectopic expression of either protein form with a Ty epitope tag resulted in the same protein localisation. Additionally, the transcripts of two translational protein homologues, TbeIF4E4 and TbeIF6, were identified as upregulated in stumpy forms. Radiolabelled-methionine experiments and polysome analysis showed that overexpression or RNAi-mediated ablation of TbeIF6 resulted in a decrease in protein synthesis and decrease in translation. Unlike its archaeal homologue, TbeIF6 protein was not induced by coldshock treatment. Finally, to identify which transcripts escape translational repression in stumpy forms an analysis was made of polysome-associated transcripts by RNA-sequencing. This identified potentially interesting genes for further investigation, and showed that many procyclic-enriched transcripts were also enriched in stumpy form polysomeassociated RNA, confirming these cells as preadapted for transmission. Together, this work has characterised a 3’UTR regulatory element in a stumpy-enriched transcript, examined alternative trans-splicing of another transcript, investigated two translational protein homologues and identified transcripts that escape translational repression in the transmissible life-cycle stage of T. brucei.

L’expression « musicalité de la peinture », et tous ses usages dérivés se sont multipliés dans les discours depuis la fin du XIXe siècle. La vivacité de cette métaphore conflictuelle s’est peu à peu diluée en raison du phénomène d’auto-alimentation verbale et opérale dont elle est l’objet. La validation de l’usage d’une telle métaphore suppose donc sa revivification et sa relève par le concept. Elle peut en effet être pensée, par opposition à la musicité, comme part nécessaire (et non présence élargie) de la musique. Elle est alors comprise comme principe d’animation silencieuse et siège du sens et de l’expressivité musicale. Ainsi, en tant que liant et Forme structurante, elle est apte à s’incarner dans d’autres matériaux et notamment le matériau pictural. Elle est reconnaissable dans ce matériau insonore, du point de vue de la production, par l’esthétique de l’Achevé-inachevé qu’elle met en place, et du point de vue de la réception, par l’expérience esthétique spécifique qu’elle provoque, allant vers une compréhension émotive toujours réajustée et infinie de l’œuvre picturale « musicale »
The expression “musicality of the painting” and all its derivative uses have multiplied in the speeches since the end of the 19th century. The liveliness of this conflicting metaphor is gradually weakened because of the verbal and operative self-supplying phenomena to which it is the subject. The validation of the use of such a metaphor assumes its own revivification and its relief by the concept. It can indeed be considered, as opposed to the musicity, as a necessity (and not an extended presence) to music. It is thus understood as a silent moving principle and as a base for the meaning and the musical expressivity. Therefore, as binder and structuring Form, it is able to be embodied in other materials and in particular the pictorial material. The musicality is recognizable in this quiet material, in terms of production, thanks to the aesthetic of the Complete-incomplete that it sets up, and in terms of the reception, thanks to the specific aesthetic experience it triggers, reaching to an always readjusted, and infinite emotional comprehension of the “musical” pictorial work

A presente Dissertação de Mestrado pretende avaliar a eficácia da proteção do patrimônio cultural imaterial no Brasil por meio do registro previsto no Decreto nº 3.551/2000, que regulamentou em parte o artigo 216 da Constituição Federal de 1988, e foi concedido pelo Instituto do Patrimônio Histórico e Artístico Nacional (IPHAN). Para tanto, baseia-se em um estudo de caso envolvendo o gênero musical-coreográfico-poético e festivo denominado Fandango Caiçara, que é transmitido oralmente de geração a geração, no seio da comunidade tradicional caiçara localizada nos municípios de Iguape e Cananéia, no litoral sul do Estado de São Paulo, e nos municípios de Morretes, Paranaguá e Guaraqueçaba, no litoral norte do Estado do Paraná e cujo registro como Patrimônio Cultural do Brasil junto ao IPHAN ocorreu em 29 de novembro de 2012 no Livro das Formas de Expressão. Nossa análise também procura levar em consideração os aspectos socioambientais envolvidos, bem como os desafios que, em nossa opinião, devem ser enfrentados pela comunidade tradicional caiçara diante da obtenção do referido registro.
This Masters Dissertation intends to evaluate the effectiveness of the cultural heritage of immaterial natures protection in Brazil by means of the registration provided by Decree n. 3.551/2000, which was partially regulated by article 216 of the Federal Constitution of 1988 and was granted by the Institute of National Historical and Artistic Heritage (IPHAN). For that it is based on a case study involving the musical-choreographic-poetic and festive genre called Fandango Caiçara, which is orally transmitted from generation to generation within the caiçara traditional community located in the municipalities of Iguape and Cananéia, in the south coast of the State of São Paulo, and in the municipalities of Morretes, Paranaguá and Guaraqueçaba, in the north coast of the State of Paraná and whose registration as Cultural Heritage of Brazil before the IPHAN occured on November 29, 2012 in the Book of Forms of Expression. Our analysis also seeks to consider the socialenvironmental aspects involved as well as the challenges that, in our opinion, should be faced by the caiçara traditional community before the obtainment of said registration.
