Abstract:
Abstract Factor V (F.V) circulates as an inactive single chain procofactor. Only upon cleavage and activation with thrombin to F.Va does it function as a cofactor in the F.Xa-mediated conversion of prothrombin to thrombin. The conventional F.V coagulation assay measures the one-stage clotting activity, which most likely reflects the trace of F.V that is activated in the sample or becomes activated during the assay. Since F.V is activated by thrombin to F.Va, a second (two-stage) assay is crucial to fully assess the total F.V potential activity in the plasma sample. For this assay, normal human pooled plasma (diluted 100- to 500-fold) was treated with thrombin (1–5 NIH Units/ml, final concentration) for various times at 37°C to fully activate the F.V. The added thrombin was inhibited with a 2-fold molar excess of Phe-Pro-Arg chloromethylketone (PPACK) and the sample re-assayed. This second assay measures the total F.V activity of a sample. Maximal 10-fold activation of plasma F.V activity occurred with 2Units/ml thrombin after 1 to 2min at 37°C and this activity was stable for 30min prior to PPACK addition. Control experiments indicated that the levels of PPACK used did not affect the one-stage F.V activity of untreated plasma samples but were required to inhibit the added thrombin from clotting fibrinogen in the assay. Assay of timed plasma samples (n=48) from seven patients admitted to the Intensive Care Unit and subsequently diagnosed with disseminated intravascular coagulation (DIC), indicated that the total F.V activity, the specific activity (total F.V activity/antigen), and the activation quotient were decreased, on average, by 45%, 49%, and 31%, respectively, from that observed with normal human pooled plasma. The results indicate that the total amount of activatable F.V as well as its degree of activation decreases from normal as part of the pathogenesis of DIC in humans. Further, the decreased total F.V specific activity observed in DIC patient plasma samples indicates that the F.V that is present is less active per molecule than normal, possibly because of proteolytic inactivation. This total F.V clotting assay should provide new information about the biochemistry of F.V and its contribution to the pathogenesis of coagulation disorders.

In contrast, treatment of F.Va (600 nmol/L) with HNE (60 nmol/L) in the presence of Ca2+ resulted only in a time-dependent decrease in its cofactor activity. Under the conditions of these experiments, the maximum extent of F.V activation accomplished by incubation with HNE was approximately 65% to 70% of that observed with α-thrombin in presence of Ca2+. The extent of both the HNE-dependent enhancement in F.V cofactor activity and the HNE-dependent decrease in F.Va cofactor activity was not influenced by the addition of phosphatidylcholine/phosphatidylserine (PCPS) vesicles (50 μmol/L). The HNE-derived cleavage products of F.V, which correlated with increased cofactor activity, as demonstrated by SDS-PAGE under reducing conditions, were different from those generated using α-thrombin. Treatment of F.V (600 nmol/L) with HNE (2 nmol/L) in the presence of Ca2+ resulted in the production of three closely spaced doublets of: 99/97, 89/87, and 76/74 kD whose appearance over time correlated well with the increased cofactor activity as judged by densitometry. Treatment of F.Va (600 nmol/L) with HNE (60 nmol/L) in the presence of Ca2+ resulted in the cleavage of both the 96 kD heavy chain and the 74/72 kD light chain into products of: 56, 53, 35, 28, 22, and 12 kD. Although densitometry indicated that both the heavy and light chains of F.Va were hydrolyzed by HNE, cleavage of the 96 kD heavy chain was more extensive during the time period (10 to 30 minutes) of the greatest loss of F.Va cofactor activity. NH2 -terminal sequence analysis of F.V treated with HNE indicated cleavage at Ile819 and Ile1484 under conditions during which the procofactor expressed enhanced cofactor activity in the prothrombinase complex. NH2 -terminal sequence analysis of F.Va treated with HNE indicated cleavage at Ala341, Ile508, and Thr1767 under conditions, which the cofactor became inactivated, as measured by prothrombinase activity. Abstract:
Frans Vredenrijk Engelenburg (1863-1938) played a major role in the development and expansion of Afrikaans and the Afrikaans academic culture - especially in the northern part of South Africa. As a Dutch intellectual, lawyer and journalist in the nineteenth century South African Republic (Transvaal), he in particular played an important role as advisor and opinion maker from the 1890s onward. One of his biggest achievements was the key role that he played in the establishment of De Zuid Afrikaanse Akademie voor Taal, Letteren en Kunst in 1909. This study is the first part of a more comprehensive biographical project on the life of Engelenburg and the role he fulfilled in the history of the Akademie and South Africa until the thirties of the twentieth century. Since the 1600s the Engelenburg family has played a prominent role in the community where they lived. Aside from the high positions they had held for centuries before, they had in the fourth and again in the sixth generation married into noble families. This contributed to their important position in the community. Due to circumstances Engelenburg was not raised in the Engelenburg milieu. A family break in 1836 was the cause that Engelenburg's father, as a baby, was spirited away from this family milieu. Engelenburg received an extraordinarily good schooling. The solid intellectual foundation already laid then, to a large extent determined the course of his life. He was at the Stedelijk Gymnasium Arnhem when he met Marie Koopmans-De Wet (1834-1906), an aunt by marriage who lived in Cape Town, when on a visit to Europe. She was his soul mate and acted as a mentor and advisor to Engelenburg. The friendship strengthened with the years. He already at school had the desire to visit South Africa one day. His parents' divorce when he was still a student at the University of Leyden, steered his life in a very different direction than what he had foreseen for himself. The divorce was to a large extent the reason that, although he had studied law, he discarded the notion of a career in law after only a year. His decision to follow a career in journalism affected the rest of his life. The Transvaal War (1880-1881) meant that the Dutch developed an admiration for the Transvaalers for the determination and courage they displayed in their attempts to defeat the British army. President Paul Kruger's call shortly after the war that the Transvaal needed young Dutchmen further encouraged Engelenburg to come to South Africa. Previously Engelenburg had for a year worked for Fred Hogendorp at the Dagblad van Suidholland en s’Gravenhage in The Hague. Circumstances abruptly changed when Hogendorp suddenly became insane. During the same time, the owner of De Volksstem newspaper in Pretoria had committed suicide and Engelenburg seized the work opportunity. Within a matter of three months, he arrived in the Transvaal. Within a month after his arrival he was appointed chief editor of De Volksstem. He had studied the Transvaal situation thoroughly and by means of the newspaper and through tireless efforts, he contributed to improving the farming community’s cultural literacy. The education situation in the Transvaal enjoyed his constant attention. After the Anglo-Boer War (ABW) (1899-1902), he continued to work towards improving the education system in the Transvaal. He early on became involved in the Transvaal University College (later University of Pretoria). Before the ABW he did everything possible to promote the Dutch language to the Boer people. However, after the war he realised that Afrikaans had a rightful place, and he, in addition to Dutch, became a champion for the Afrikaans language. The battle between the proponents of Dutch and Afrikaans respectively, increased after the ABW. To achieve unity of action between the two groups, De Zuid Afrikaanse Akademie voor Taal, Letteren and Kunst was founded in 1909. Behind the scenes Engelenburg was one of the major driving forces to assist with the founding of the organisation. As a board member and later as chairman, he gave impetus to the Akademie. In 2009 the organisation celebrated its centenary. This is an important milestone, especially seen in the light of the current political climate in South Africa. The Akademie can now be regarded as a monument to Engelenburg as the fruit of his labour and perseverance during the first three decades of the Akademie’s existence.

Com o objectivo de conhecer alguns dos agentes infecciosos que afectam a população felina da Associação dos Amigos dos Animais Abandonados da Moita, e relacionar a sua eventual presença com as alterações clínicas apresentadas pelos animais em questão, nomeadamente a Gengivo-Estomatite Crónica Felina (GECF) e a Doença Respiratória Felina (DRF), foram avaliados neste estudo 50 felinos, incluídos aleatoriamente. A cada animal, e após exame clínico, foram colhidas as amostras de material biológico apropriado para realização de hemograma, detecção de anticorpos contra o Vírus da Imunodeficiência Felina (FIV) e de antigénio do Vírus da Leucemia Felina (FeLV), detecção de ácido nucleico de Herpesvírus Felino tipo 1 (FHV-1) e Calicivírus Felino (FCV), e pesquisa de Candida spp. O FIV e FeLV foram detectados por imunocromatografia de membrana, o FHV-1 por PCR em tempo real, o FCV por RT-PCR convencional e as leveduras isoladas em Sabouraud dextrose agar foram posteriormente identificadas quanto ao género e espécie. Constatou-se que, dos 50 animais, 22% eram positivos para FIV, 10% para FeLV, 62% para FHV-1 e 26% para FCV. Em relação às leveduras, foi detectada a presença de C. silvicola (n=3), C. valida (n=1) e C. parapsilosis (n=1). As associações estabelecidas entre a presença do vírus e determinada manifestação clínica que apresentaram significado estatístico foram: a presença de úlceras linguais em animais com FCV e co-infecção FCV / FIV; o desenvolvimento de estomatite nos animais infectados unicamente com FCV, FIV ou FeLV, e nas co-infecções FCV / FIV, FCV / FeLV, e FHV-1 / FeLV; a manifestação de gengivite de grau 2 ou 3 nos animais com FCV; e a ausência de úlceras linguais nos animais com infecção por FHV-1. O animal em que foi detectada a presença de C. parapsilosis apresentava gengivite ligeira, corrimento ocular e, a nível hematológico, leucocitose com linfocitose e monocitose. O animal infectado com C. valida exibia apenas estomatite ligeira e corrimento nasal, e os animais com C. silvicola revelaram gengivite ligeira, estomatite, úlceras linguais, corrimento ocular e nasal, e leucocitose com neutrofilia, linfocitose e monocitose. Nenhuma destas alterações revelou associação estatisticamente significativa com a presença da levedura. Para uma correcta avaliação da prevalência destes agentes infecciosos em populações errantes felinas e sua associação a GECF e DRF, deverão ser realizados estudos mais extensos quanto ao número de animais envolvidos. Por outro lado, considerando a escassa informação sobre a microbiota micológica da cavidade oral dos felinos, a detecção de Candida spp. contribuiu para um melhor conhecimento nesta área, sendo que a sua influência na etiologia da GECF e da DRF, deverá também ser estudada com maior profundidade.
ABSTRACT - Detection of Feline Immunodeficiency Virus, Feline Leukemia Virus, Feline Calicivirus, Feline Herpesvirus-1 and Candida spp. in stray cats and possible association with Feline Gingivostomatitis and Feline Respiratory Disease - The aim of this study was to analyze the viral and mycological microbiota of cats’ mouth and its relationship with the clinical symptoms, especially with Feline Gingivo-Stomatitis (FGS) and Feline Respiratory Disease (FRD), and infection with Feline Immunodeficiency Virus (FIV) and Feline Leukaemia Virus (FeLV), both able to induce immunodeficiency. The test group included 50 cats from the Associação dos Amigos dos Animais Abandonados da Moita, that were subjected to clinical examination with collection of biological samples for serological detection of Feline Leukemia Virus (FeLV) antigen and Feline Immunodeficiency Virus (FIV) antibodies, detection of Candida spp. and viral nucleic acid of Feline Herpesvirus (FHV-1) and Feline Calicivirus (FCV). FIV and FeLV were detected by immunochromatography membrane, FHV-1 by real time PCR, FCV by RT-PCR, and the yeasts by mycological culture and then identified to genus and species. Within the 50 cats sampled, 22% (11/50) were FIV seropositive, 10% (5/50) FeLV, 62% (31/50) FHV-1 and 26% (13/50) FCV. Regarding the yeasts, C. silvicola (n=3), C. valida (n=1) and C. parapsilosis (n=1), were detected. The associations between the agent detected and the clinical sign shown, that were statistically relevant were: the presence of tongue ulceration within FCV and FCV / FIV infected animals; the development of stomatitis in cats with FCV, FIV, FeLV, and FCV / FIV, FCV / FeLV, and FHV-1 / FeLV; moderate to severe gingivitis in FCV infected cats; and the absence of tongue ulceration in FHV-1 infected cats. The cat with C. parapsilosis showed mild gingivitis, ocular discharge, and hematological findings were leucocytosis with lymphocytosis and monocytosis. Mild gingivitis and nasal discharge were presented by the C. valida infected animal, while the C. silvicola infected animals showed mild gingivitis, stomatitis, tongue ulceration, ocular discharge, nasal, discharge, and leucocytosis with neutrophilia, lymphocytosis and monocytosis. Because of the small size of the sample, none of the clinical findings were statistically associated with yeasts presence. Once again, due to sampling issues, a more thorough analysis should be performed to correctly evaluate the true prevalence of these infectious agents in stray feline population. On the other hand, considering the lack of information regarding the mycological microbiota of feline oral cavity, the identification in our samples of yeasts included in the Candida genus, will certainly prove helpful. Furthermore, the role of C. valida, C. parapsilosis and C. silvicola in FGS and FRD should be deeply evaluated.

Tradisjonelle arbeidsformer i dagens byggebransje vier lite oppmerksomhet til forvaltning, drift og vedlkehold (FDV) av bygninger. BIM, bygningsinformasjonsmodellering, kan bidra til en ny digital arbeidsprosess, hvor FDV står i sentrum. Resultatet av en slik prosess er en modell, en database, hvor all informasjon om et bygg ligger lagret.LYDIA er FDV-programvare, utviklet og levert av Lydia AS. De ønsker å videreutvikle LYDIA i retning BIM, og se på hvordan sammenhengen BIM-LYDIA kan utføres i praksis. Lydia AS opplever i dag størst utfordringer ved import av informasjon til LYDIA-databasen. Kundene samler manuelt inn store mengder informasjon til databasen. Erfaringer viser at endel kunder strever med å forstå bruksnytten av LYDIA.Systemarkitekturen presentert i oppgaven løser Lydia AS sine forutsetninger for overgangen BIM-LYDIA. Funksjonaliteten i LYDIA er opprettholdt. buildingSMART sitt standardiserte filformat, IFC, er brukt som utgangspunkt for overføring av informasjon. Løsningen er realiserbar innen andre kvartal 2014. Modellen forenkler dagens importarbeid. Lydia AS sine kunder slipper manuell dataregistrering som øker risikoen for feil. Ved import direkte fra BIM forenkler man prosessen med å hente informasjon til LYDIA-databasen. Modellen bygger likevel ikke oppunder en ny samarbeidsprosess. Digitale arbeidsformer kan endre de tradisjonelle arbeidsformene i byggebransjen. Utgangspunktet for slike arbeidsprosesser er større tillit og mer fokus på samskaping. Lydia AS må selv tenke gjennom hvilken retning de ønsker å ta for fremtidige LYDIA.

Abstract In the year 2019 the vehicle workshops services returned to the Swedish Armed Forces. For six years they had been an integrated part of the Swedish Defence Materiel Administration and the business was set up in a corporate fashion. When returning to the Swedish Armed Forces the workshops will continue to use that corporate business model, and at the same time adjust to military setting and logic. Among other things, the workshops will be organized into military units, under the same conditions as all other parts of the Armed Forces. The purpose of this thesis is to examine and understand the meaning and consequences of the corporate and the military logic coexisting within the Vehicle Workshop Services units. A typology made specifically for this reason is used to depict the different configurations that occur when those two logic structures coexist. The study draws the conclusions that the dualistic relationship between the corporate and the military logic ends up in compromises that makes the goal and purpose of the organisation vague and the personnel involved experience a difference between how it is supposed to be functioning and how it actually does. Furthermore, the study states that the conditions for developing the Vehicle Workshop Services units into a strong and resilient organisation from a HR and symbolic perspective is less favourable. The Swedish Armed Forces need to address this actively to create a common understanding of the factors that are in play by all personnel involved in the matter. The study gives five hands on suggestions to mitigate those problems. Finally, some suggestions are given how to re-use the typology that was created for the study in other contexts.

Abstract:
After briefly examining the history of trade marks and establishing the theoretical framework of trade mark functions, an analysis from the perspective of semiotics is conducted, viewing the trade mark as a sign. This analysis serves as a tool for the suggestions and conclusions drawn in relation to the role of trade mark law today. The issues of trade mark registration, use and protection are examined, as main facets of the trade mark functions. An analysis of the legal provisions, legal doctrine and case law on trade mark registration and use in Greece, before and after the entry into force of the First Council Directive on the Harmonisation of trade mark laws in Europe 89/104/EEC is undertaken, so as to denote the effect of the European harmonisation on these issues. The Court of Justice case law is examined, in juxtaposition to the Greek case law on the respective implementing provisions. The similarities and differences are noted and analysed. Throughout, examples from a common law jurisdiction, namely the UK, are taken into account. The issues of risk of confusion and dilution are analytically examined at a national (Greek) and European level. In comparison to the Court of Justice jurisprudence, the national legal framework and case law of Greece on these issues is analysed. Examples from other European jurisdictions are taken into account. The theory of dilution in the context of the European trade mark framework is further analysed against developments in the USA. The controversial doctrine of trade mark dilution, introduced by Fr. I. Schechter, is of primary interest in the course of the thesis and suggestions are made for a new perspective of the theory. The controversial doctrine of Schechter is seen, in view of the above, under a new light.

FTV (Free-viewpoint Television) is visual media that transmits all ray information of a 3D space and enables immersive 3D viewing. The international standardization of FTV has been conducted in MPEG. The first phase of FTV is multiview video coding (MVC), and the second phase is 3D video (3DV). The third phase of FTV is MPEG-FTV, which targets revolutionized viewing of 3D scenes via super multiview, free navigation, and 360-degree 3D. After the success of exploration experiments and Call for Evidence, MPEG-FTV moved MPEG Immersive project (MPEG-I), where it is in charge of video part as MPEG-I Visual. MPEG-I will create standards for immersive audio-visual services.

Mg2+ anticoagulated PPP (20mM MgCl2) was applied at RT to a Zn2+ immobilised biscarboxymethylamino Sepharose 4B column and eluted with 20mM Tris, 20mM MgCl2, 2.5mM CaCl2 0.15M NaCl buffer pH 7.4. Following collection of the wash-through fractions, bound proteins were developed through application of linear (0 to 35mM) imidazole gradients.All fractions were screened for F.II, V, IX, X, vWF:Ag, Protein C, Fg, Fn and 2-macroglobulin by ELISA, VIII:Ag by IRMA, and VIII:C by both 1-stage and 2-stage bioassay methods. VIIIrAg (60-90% yield), vWF:Ag (100%), VIII:C 1-stage activity (35%), Fn (> 60%), Fg (> 80%), V (50%) and a2-macroglobulin (> 70%) were located only in the gradient fractions. Two distinct peaks demonstrated shortening of the 2-stage VIII:C assay:- one co-eluting with the 1-stage VIII:C activity and another major peak in the wash-through fractions where antigenic determinants of F.II, IX, X and part of the protein C were located and partially resolved from each other. Only F.II.-Ag co-eluted with the “ 2-stage VIII:C” activity. Similar observations were found in Mg2+ -anticoagulated severe Haemophilia A plasma and in citrated PPP developed with 20mM MgCl2, 20mM Tris buffer. A1(0H)3 treatment abolished the activity from citrate -, but not from Mg2+ -anticoagulated plasma.The relevant fractions showed no activities in F.V, VII, VIII:C, IX or X 1-stage bioassays. They did not clot Fg and did not contain detectable Xa or Ila as assessed by S-2222, S-2238 or S-2288. Following incubation with specific antisera against IgG, II, V, VII:Ag, X, protein C, a- and g- lipoproteins, and plasminogen only anti-II inhibited this "2-stage VIII:C" activity. 2-stage VIII:C assays depend upon Ca2+ -dependent generation of Xa. Since the activity in the wash-through fractions could not be ascribed to VIII, Xa, or Ila the results would indicate the presence of a hitherto undescribed Factor X activator activity.

The aim of this paper is to propose new algorithms for Field of Vision (FOV) computation which improve on existing work at high resolutions. FOV refers to the set of locations that are visible from a specific position in a scene of a computer game. We summarize existing algorithms for FOV computation, describe their limitations, and present new algorithms which aim to address these limitations. We first present an algorithm which makes use of spatial data structures in a way which is new for FOV calculation. We then present a novel technique which updates a previously calculated FOV, rather than recalculating an FOV from scratch. We compare our algorithms to existing FOV algorithms and show they provide substantial improvements to running time. Our algorithms provide the largest improvement over existing FOV algorithms at large grid sizes, thus allowing the possibility of the design of high resolution FOV-based video games.

These resources were selected with the intention to include practical recommendations on technology-enabled interventions which could support a proposed national distance learning system in Yemen.
