Full textThe factor VIII (FVIII) is a glucoprotein of the coagulation, being the cofactor of the activated factor IX (FIXa). Its metabolism depends on various limiting factors or enhancing its survey or function. The objective of this research’s work was to analyse different parameters that could influence the plasma FVIII level and its pro-coagulant function. It included 4 parts : 1) We showed that the known intra-cellular retention of FVIII was mainly due to its aggregation and degradation following both proteasomal and lysosomal pathways. 2) We analysed FVIII gene mutations responsible for hemophilia A in a large patients cohort. The bio-informatic analysis demonstrated its deleterious consequence. The influence of these mutations on the anti-FVIII antibodies occurrence was stratified, in association with ethnicity and familial antecedent of inhibitor. 3) The research of factors influencing FVIII levels in hemophilia A carriers showed : i) major determinants such as the presence of an additional genetic disease characterised by a FVIII deficiency, the factor Willebrad’s level and the non-random inactivation of the X chromosome; and ii) minor determinants : age, severity of hemophilia, the polymorphism D1241E of FVIII gene, and 5 new polymorphisms of LRP1 located in its binding site for FVIII.4) We analysed 8 recombinant FVIII with in vitro created mutations in its 1808-1818 region. Previous studies that analysed only the FVIII light chain, have shown that this region constituted the more affine binding site of FVIII for FIXa. We demonstrated here that the 1808-1818 region is not as essential as it was reported because within the entire molecule, its affinity decreases and mutations affecting it do alter mildly the FVIII activity

Full textTwo theories are to be found at the origin of this memoir. A fragment of the retinex theory by e. H. Land and the abney law about the interaction of two major wavelengths at the level of the luminosity and the field of transmittance. We shall work here on the relationship source surface. The first part of the memoir deals with the setting of a device of classifications made by individuals subjects on the one hand and on the other the systematic measures with spectrophometer of each one of the six sources and of each receiving surface (seven altogether, placed on a magnetic support which can be manipulated by each one of the forty subjects individuals). The setting up of this experimental device has taken about one year: and in the end, a method has been chosen for the reactions of the subjects as well as for the coherence of the results. One of the characteristics of the device deals with the mobility of the subject in front of the surfaces. In keeping with the answers obtained by the subjects a theoretical energetic classification has been set up from the measures of each sample and of each source on the spectrophotometer, integrating the efficiency luminancy coefficient in the photopic domain. The similitude of the results (93,27%) allows us to validate the abney law for the relationship between surface and support, as well as the classification method which has been used

Full textVon Willebrand factor (VWF) plays an essential role in hemostasis. It mediates platelet adhesion and aggregation after a vascular lesion. The present study is focused on the interaction of VWF with heparin. Heparin binding studies in 92 patients plasma with type 1 or 2 von Willebrand disease (VWD) showed that the mean value of heparin binding in type 2A on the one hand and 2M on the other hand, was significantly lower than in normal plasma, type 2 N, type 28 and type 1. Moreover, we characterised heparin-binding properties of mutated VWF in 24 patients plasma with type 2 VWD, and in 15 recombinant VWF (rVWF) with the corresponding mutations located in the A1 domain VWF. Four mutations, at position 509, 551, 552 and 611 lead to significantly decreased binding to heparin in bath plasma and rVWF. Structural analysis suggested how folding problems and destabilisation due to these mutations could induce reorganisation of surface regions involved in heparin binding. Next, we expressed fifteen mutated rVWF containing single or multiple mutations within these regions. Three variants (K643G/K644G, K642G/K643G/K644G/K645G and R571A/K572A/R573A/R578A/R579A/K585A) displayed significant reduction in heparin binding. Computational investigations showed that the most striking alterations of the electrostatic potential contours were seen for the same mutants. Taken together, these information allow us to propose the most likely position of heparin on the A1 domain surface, with VWF residues at position 571-572-573-578-579-585 and 642-643-644-645 acting in concert for optimal interaction with the polyanion. Full textThe main objective of this work was to study, using an in vivo approach, a number of proteins essential for haemostasis and coagulation, in pathophysiologicals conditions and by identifying potential determinants of their lifecycle. In the first part of the work, focused on the Von willebrand Factor (VWF)/ADAMTS13 couple, the critical role of VWF in the decrease of ADAMTS13 during sepsis has been highlighted. We also showed that VWF proteolysis by ADAMTS13 is not implicated in its clearance. In the second part of the thesis, the role of glycosylations on the function of both VWF and coagulation factor X (FX) has been studied in vivo. In FX, the major role of the two N-glycosylations present in the activation peptide of the molecule has been demonstrated. For VWF, we emphasized the fact that some O- glycosylations sites are involved in the maintenance of the protein plasma concentration while others are important for the functionality of the protein. This work, based on in vivo experimentations, underlines the importance of the relationship between VWF and ADAMTS13 during physiological processes like VWF clearance or pathophysological processes such as sepsis. This work also allowed us to understand the essential role of some glycosylations in the functionality of glycoproteins. Our approach, entirely using in vivo experiments, highlighted some unsuspected effects which pre-existing literature, based on in vitro experiments, did not indicate

Full textOccurrence of pro-coagulant factor VIII (FVIII) neutralizing antibodies in hemophilia A patients constitutes a major therapeutic failure. While the effector steps of the immune response, the nature of the involved immune cells and the antibodies properties have been well-reported, the early stages of the immune response are poorly described and constitute the topic of my PhD thesis. The first part of my thesis deals with the role of the microenvironment encountered by FVIII once administered in vivo, especially the role of complement system molecules. While complement system is a major actor of the innate immunity, it also participates to adaptive immune responses and can interact with the coagulation cascade. Though complement is involved in immune responses against pathogens, its contribution to immune responses against therapeutic proteins has never been reported so far. Therefore I have investigated the potential role of the complement system in the immunogenicity of therapeutic FVIII. The second part of my thesis focuses on the involvement of FVIII structure for its recognition by the immune system. It has been demonstrated that FVIII C1 domain plays a major role of its uptake by immune cells in vitro and its immunogenicity in vivo. Because of strong structural homologies between C1 and C2 domains, I investigated the potential role of FVIII C2 domain for its endocytosis by antigen presenting cells and the elicitation of the anti-FVIII immune response

Full textMorocco has invested in the tourism sector and placed it at the core of its development endeavors. The sector generates jobs and promotes a global cultural exchange. In fact, the tourism sector is nowadays the second largest contributor to national Moroccan GDP after the agriculture sector (12% of national GDP). Tourism in Morocco generated 57.5 billion dirhams and 500,000 direct jobs in 2013. Of course, tourism has evolved well in Morocco and visitor numbers have increased from 4,278,120 million in 2000 to 10,046 million in 2013. However, the share of foreign tourists represents only 53% of all visitors, and the share of Moroccans living abroad (MRE) represents 47%. This figure seems minimal compared to international visitors in the world, the brand image conveyed by the Moroccan state, and the assets available to the country. How is it that a country endowed with ancestral culture, a diversified landscape with 3600 km of coastline, a mild climate all year round, receives only 5,323,333 million foreign tourists? Tourism development goes hand in hand with economic and political development, social-cultural development, environmental development, consumption trends, and the international structure of tourism production. This thesis aims at answering this question using cross-research disciplines to identify the main obstacles hindering the development of tourism in Morocco, as well as the assets of the country

Full textMyanmar has gone through unprecedented changes since the year I997,when the country Jolned the Association of Southeast-Aslan Nations (ASEAN). Those changes have accelerated since the beginning of globalization processes and of economie liberalization, launched by the government in 2010. As a result, the territorial dynamics of the country are quickly evolving. Since the 19th Century, the Irrawaddy River has been considered both as the traditional artery of the country, the axis of settlement, the heart of population, of concentration of the economie activities of the country and as the main road. Beyond the perception of the territory by its inhabitants, I attempt to define the contemporary and meaningful role of the River. I will mesure the evolution of its role as an integration factor of the national territory and of the country in ASEAN. Although the role of the Irrawaddy Valley has decreased because of the rise of the terrestrial dynamics toward the East-especiallyas part of its integration into ASEAN -and towards the coastline and because of the development of the Valley of the Sittang River, some urban and industrial poles of the Irrawaddy actively take partin Burma development and in its regional integration. Except In Yangon and in Mandalay, this participation remains mostly in draft form. Despite the acceleration of the regionalization and of the globalization, the inhabitants, the urban ones in particular, reappropriate the river banks at a local scale. Full textFacing the dual challenge of poverty reduction and environment preservation, the non-extractive economic valorization of natural resources is likely to be one of the ways to achieve sustainable development, in the southern developing countries. Valuing the ecosystems enables to bypass a major constraint identified by economic theory, which is the lack of an efficient market revealing resources value. Ecotourism is one of the most well-known application fields of this strategy of resources valuation. By ratifying the Convention on Biological Diversity, developing countries agreed to implement the Convention guidelines for the ecotourism promotion through National strategies of biodiversity sustainable management. Thus, in many countries, ecotourism’s improvement is significant; and its results can be viewed in different ways. But, existing assessment methods do not always take into account the various and divergent interests issues. Full textAlthough considered one of the top priority sectors in the economic and social development of the country, the touristic activity remains less prosperous in morocco than in the rest of the mediterranean basin countries. The object of this thesis is not to present a comprehensive overview of tourism, nor the full range of connections that exist between touristic activity and the different social stratum, but rather to develop a line of investigation that these concerns tend to overlook. The goal here is to study the interrelations between transportation and tourism : the influence of transportation on the opening up of news touristic areas, the effect of transportation on attracting potential clients from a wider geographical zone, the restructuring of the moroccan touristic territory, and the new development of tourism that could result from the synergy between these factors. Along with accommodations, transportation constitutes an extremely important instrument in the integrated development of tourism in morocco, especially in a national tourism which could serve as a basis for the developing international tourism. The consequences of this new approach is a better economic integration, different marketing of tourist products and perhaps a new organization of the tourist areas that would be more compatible with their specific characteristics. Transportation is less and less independant of touristic activity with the increasing organizational co-ordination between carriers, tour-operators and the hotel business. However, if transportation is a necessary even indispensable condition for the development of tourism, it remains only one factor among many others in the economic dynamics of this development. Full textATF7 proteins, which are members of the b-Zip family of transcription factors, are able to bind ATF/CRE sequences (TGACGTCA) within different early adenoviral or cellular promoters. ATF7 can interact with the family of Jun/Fos oncoproteins and modulate their activities. To get an insight into the transactivation mechanism mediated by ATF7, we analyzed its relations with hsTAFs proteins, which are components of several multiproteic complexes like TFIID. Our results show that transactivation by ATF7 is specifically mediated by hsTAF12, mainly by its 20-kDa isoform. Moreover, ATF7 and hsTAF12 interact in vivo : the integrity of the ATF7 amino-terminal activation domain and of the hsTAF12 "histone-fold" domain is required for this interaction. We show that hsTAF12 mediated transactivation is specifically inhibited by hsTAF4 (its heterodimerization partner within TFIID), and not by hsTAF4b, a tissue-specific hsTAF4 homolog. Ubc9, a protein involved in the SUMOylation pathway, was found among the proteins interacting with ATF7 and modulating its activity. We have identified a consensus SUMOylation site within the N-terminal part of ATF7 and demonstrate that ATF7 is SUMOylated both by in vitro and in vivo experiments. Moreover, our results reveal that the intracellular localization of ATF7 is affected by its SUMOylation : ATF7 is found in the nucleus only in a de-SUMOylated form ; the cytoplasmic SUMOylation of ATF7 likely induces its sequestration at the level of the nuclear pore complexes (NPC). Using immunohistochemistry assays, we observe a colocalization between ATF7 and RanBP2, a protein of the NPC, which is an E3 ligase of the SUMOylation pathway. Accordingly, RanBP2 is able to stimulate ATF7 SUMOylation in vitro. This NPC targeting seems to influence ATF7 transactivation activity as an ATF7 mutant, whose SUMOylation is impaired, is more active than its SUMOylated counterpart. These results correlate with our chromatin-immunoprecipitation (ChIP) experiments, which show that the occupancy of a target promoter by ATF7 is highest in the presence of the unSUMOylated protein

Full textContrarily to a superficial vision, time is not neutral element in acquisitions because each actor tends to avail himself of it (entrepreneurs, managers, shareholders, bankers, experts and agents). Actually, the time-factor has three functions in acquisition operations: - it generates actions though it fixed the maturity of the goals that each one sets for himself and allows to measure the distance yet to be covered. - It reveals the tensions resulting from the disparate perspectives of the different decision-makers (time-discordance), and amplifies the corresponding conflicts. - It makes the search for solutions easier, as successful acquisitions are products of converging perspectives (time-concordance). In summary, the point of view defended in this thesis is that there is a cause and effect link between the time-factor and the result of acquisitions. In this respect, the recommendations listed in conclusion might increase the chances of success of such operations

Full textATAD2 is an evolutionarily conserved but poorly characterized factor that bears different types of func¬tional domains: an AAA ATPase domain and a bromodomain (BRD). ATAD2 is normally highly ex¬pressed in male germ cells and in embryonic stem cells (ESC), however the overexpression of this protein has been detected in a large variety of independent cancers. ATAD2 is proposed to act as a co-activator of androgen and estrogen receptors and in addition, this protein also seems to act as a co-factor for Myc oncogene and plays a role in the pRb/E2F pathway. Moreover, the overexpression of ATAD2 predicts poor prognosis in lung and breast cancers. All of these characteristics make ATAD2 a valuable prognosis biomarker and a promising therapeutic target in aggressive cancers.Herein, we show that hATAD2 binds to acetylated H4 tail through its BRD, and that its ATPase domain enables ATAD2 multimerization, affecting the ability of the BRD to bind acetylated lysine in cells. Additional investigations, including structural studies, show that ATAD2’s BRD is responsible for its specific interaction with acetylated lysine 5 of histone H4. We have also functionally analyzed the AAA ATPase domain and discovered elements that control its role in protein multimerization. In addition, we studied ATAD2 in ESC and in the H1299 lung cancer cell line, and demonstrated that this factor has crucial roles in cell proliferation in the absence of growth factors. Moreover, by using a combination of ChIP-seq, ChIP-proteomics and RNA-seq experiments in ESC, we found that ATAD2 is highly enriched in regions with high transcriptional activity and that it keeps chromatin accessible for chromatin templated factors. These data indicate that ATAD2 in its physiological context ensures a critical role in general chromatin-templated activities, such as transcription, by maintaining the accessibility of chromatin for transcription factors. Finally, in order to structurally characterize either ATAD2 or its homologue in Schizosaccharomyces pombe, ABOI, different fragments containing the AAA ATPase domain were produced in bacteria as well as in insect cells using baculovirus expression vectors. Conditions to produce soluble fragments were established and some of these fragments were purified. Nonetheless, solving the crystal structure of ATAD2 still requires further investigation
