Full text
Abstract:
Abstract Increased genomic information available for young bulls has decreased age at time of semen collection. Factors effecting collection characteristics include collection method (electro ejaculate (EE), or artificial vagina (AV), and the number of ejaculates collected. The objective of this study was to determine the effect of managerial factors on collection characteristics. From 2008 to 2018, 11,642 individual ejaculates were analyzed by a single technician at the Kansas Artificial Breeding Service Unit. Bulls that were not receptive to the AV after 3 or 4 attempts, were subject to EE. Collection characteristics were evaluated using multiple regression models; fixed effects included collection method, sequence of ejaculates collected per day, and were evaluated for their impact on collection characteristics. Progressive motility before freezing was greater (P< 0.0001) for bulls collected with EE compared to AV. Ejaculate volume for EE collections was greater (P< 0.0001) than those collected with AV. Percent spermatozoa with secondary abnormalities was greater (P< 0.05) for bulls collected with EE compared to AV. Concentration of spermatozoa per mL was less (P< 0.0001) for bulls collected with an EE (514 x106) compared to AV (617 x106). Total number of straws frozen per ejaculate were less (P< 0.001) for bulls collected with EE (94) compared to AV (108). The number of ejaculates collected per day was significant for the percent of spermatozoa with secondary abnormalities (P< 0.001). As ejaculate number per day increased, the concentration of spermatozoa decreased (713, 580, 535, and 434 x 106 per ml respectively; P< 0.0001) and the number of straws frozen per ejaculate decreased (123, 107, 93, and 82 respectively; P < 0.0001). In conclusion, AV collections resulted in a higher number of straws frozen. The method of collection could cause a significant impact when collecting young high demand bulls.

Abstract:
The use of osmotic stress or desiccation in combination with 2% acetic acid reduced the numbers of Salmonella typhimurium or Listeria monocytogenes on beef tissue to a greater degree than the acid alone. Bacteria were osmotically stressed by NaCl, CaCl2, or sucrose solutions. Pre-washing with 20% NaCl followed by sanitizing with acetic acid reduced the numbers of S. typhimurium an additional 1.5-to 2-log cycles compared with acetic acid alone. Similar reductions were seen by allowing the tissue surfaces to dry before the application of the acid. Desiccation in combination with acetic acid was also effective in reducing the numbers of Enterobacteriaceae on inoculated beef carcasses.

Abstract:
Smartphone addiction is a growing social issue globally, especially among young consumers. Few studies have examined the role of smartphone addiction and student performance in Pakistan. However, how daily life disturbance, positive anticipation, withdrawal, cyberspace orientation, overuse, tolerance, and student academic performance has not been examined in the literature in Pakistan. The purpose of the study is to examine the effect of daily life disturbance, positive anticipation, withdrawal, cyberspace orientation, overuse, and tolerance on student academic performance. The study has adopted a self-administered survey-based approach. A total of 375 responses were gathered from the university students in Pakistan. The study has used SPSS 25.0 version to analyse the data and report the findings. The findings indicate that ‘overuse’ has the most indirect effect on students’ performance, followed by Cyberspace and oriented relationship, tolerance, positive anticipation, and daily life disturbance. The study demands a particular intervention by the students to self-regulate their behaviour. Parents and teachers play their role in controlling mobile addiction. Policymakers may introduce policy-related interventions to control the excessive use of mobile phones. The study lays down future directions and discusses the limitations towards the end.

Abstract:
The ruminant industry is crucial under Malaysia’s National Agrofood Policy (NAP) 2.0, which aims for a 50% Self-Sufficiency Level (SSL) by 2030 to bolster food security. Increasing the ruminant population to meet this goal could elevate greenhouse gas (GHG) emissions, especially methane (CH4) from enteric fermentation. Therefore, a targeted mitigation strategy is necessary, based on precise emission calculations. GHG emissions are determined by multiplying the total population by an emission factor (EF). For accurate estimates, breed-specific EFs are needed. This study developed enteric fermentation EFs for Malaysia’s main beef breed, Kedah-Kelantan (KK) crosses, using the IPCC 2006 tier-2 methodology. The study found that mature male and female KK crosses emit 52.5 and 39.7 kg CH4/head/year, respectively, with a pooled average of 46.1 kg CH4/head/year, differing from the previously developed EF from Brakmas cattle of 51.6 and 65.7 kg CH4/head/year for mature female and male, respectively, with a pooled average of 58.7 kg CH4/head/year and IPCC’s default EF of 47 kg CH4/head/year. Using these breed-specific EFs can lead to more accurate emission estimates and effective GHG mitigation strategies.

Abstract:
In this paper, plant straw densification technology has been studied. The corn straw was taken as raw material in the experiment. The influences of raw material moisture content, densification pressure and raw material size to the compact process have been studied. The results show that the 13%~18% moisture content of the corn straw is the best scope in compact process. When the raw materials are densified at a pressure of 60MPa, the products have smooth surface, high density. While the pressure continuously increases, the increase of density is not obvious. The raw material size 10~20mm are suitable for densification.

Mendelsohn, Robert. Three-Factor Model of Couples Therapy. The Rowman & Littlefield Publishing Group, 2017. https://doi.org/10.5040/9781666983524.

Full text
Abstract:
Couple psychotherapy extends the work of the psychotherapist to the patient’s most significant committed adult relationship, yet the therapy is difficult both conceptually and technically. One major reason for this difficulty is that in every couple’s treatment there is a confusing array of psychological defenses as well as regressive and nonregressive couple object relations-as distinct from the object relations that each individual member brings to the couple. Further, many of these processes are occurring outside consciousness and at the very same time. This book is an attempt to clarify all the confusing issues by presenting a three-factor model of couple psychotherapy within a psychodynamic framework. This model has been found to be very effective with many different kinds of couples. The book suggests that there are three powerful couple dynamics that shape every couple’s treatment: (A) the quality and quantity of the couple’s projective identifications; (B) the level of their “couple object relations”; and (C) the presence or absence of the defense of omnipotent control. These three variables are the most important factors in the therapy; they determine the success or failure of every therapy with every couple. These dynamics also determine quite a bit about how to conduct a couple therapy with regard to the therapist’s level of activity, tone, the way of sorting the material in his or her head, and even the kinds of interventions he/she chooses (whether or not, for example, the therapist will use certain resistance techniques). Understanding these three variables and how they interact is key to the success of the therapy.

Abstract:
The face of the employment interview has been gradually transforming over the past fifty years with the culmination of new research, theory, and practices. Now more than ever, researchers and human resource professionals are demanding interview formats that accurately and reliably predict a plethora of criteria in addition to the job candidate's skill set. No longer is the implementation of the traditional structured interview format sufficient for screening applicants. The effective interview is on its way to being transformed into a multifaceted instrument that aims to surpass the predictive precision of standardized selection tests. This article outlines the impetus of research, theory, and practice that spurred on the interview's transformation and the long-term payoff which employers are receiving from the changes. It gives a glimpse of where the employment interview is headed in the upcoming decade and the factors that make today's employment interview so effective.

Abstract:
This chapter begins with a discussion of recent vascular research that has unveiled the complex interaction between exposure to risk factors and pathological changes at the vessel wall. Risk factors such as smoking or hyperlipidaemia first cause a pre-morbid phenotype with reversible dysfunction of flow-mediated vasodilation, known as endothelial dysfunction (ED). If exposure to risk factor(s) does not cease, ED develops into the first morphological vascular changes that finally lead to atherosclerosis. Cholesterol crystals have been shown to lead to pro-inflammatory activation of macrophages. Progression from stable coronary plaques to the plaque rupture that underlies the acute coronary syndrome is discussed in detail. The chapter provides a basic up-to-date concept of the development and progression of atherosclerosis and highlights the stages where preventive measures may still be effective.

Abstract:
Effective cancer communication is crucial for both clinicians and patients, yet is often suboptimal. Health literacy—the ability to access, comprehend, evaluate, and communicate health information—is a latent factor that may contribute to ineffective medical interactions. Limited health literacy has been associated with significant negative health outcomes and higher medical costs. Given the compelling evidence that ethnically diverse populations are particularly vulnerable, we use a narrative case example—a hypothetical clinical meeting between an oncologist and a newly-diagnosed patient—to highlight how patient risk factors for low health literacy (e.g. age, language, distress, etc.) may be amplified by clinician and contextual factors (e.g. using medical jargon, complexity of patient educational materials, etc.). Finally, we re-imagine the same clinical encounter and illustrate how some simple strategies could ameliorate the effects of low health literacy and facilitate communication and patient decision-making.

Abstract:
Mozambique has achieved incipient but still fragile socio-economic development since 1975. The public financial management system has been reformed and improved, but its performance has weakened since 2013. Applying an institutional economics approach, we have identified the economic growth strategy, the separation of powers principle, and the degree of decentralization as key factors affecting the public financial management system. As the current strategy based on natural resources seems to be failing, we suggest an alternative balanced growth strategy in the context of an effective democratic political system. It would entail the identification of policy priorities required for fostering sustainable and inclusive development.

Abstract:
AbstractDesigning knowledge communication with external partners is a core activity of research networks. In science, such communication has been addressed only recently and is still considered as non-academic activity. Successful communication with practitioners, that is knowledge transfer, is a crucial factor for effective research. In the age of online communication, this requires special attention and skills, for example related to social media communication. Based on our own empirical results derived from interviews, the authors identify what factors affect the communication process and how the design of communication content may be influenced. To do so, successful examples of communication with external stakeholders are presented. For the theoretical basis, science communication, knowledge communication, knowledge management, and knowledge transfer were selected and consolidated. Although the findings stem from a research network specializing in sustainable land management, they can be transferred to other academic collaborations. Our results indicate that external communication is effective when knowledge has been transferred between academics and practitioners.

Abstract:
AbstractThis chapter uses a three-stage process of documentary analysis to illustrate how teaching effectiveness is assessed and studied in Spain. We begin by presenting the Spanish legal context, giving a historical overview of the most important education legislation. This is important, as there have been several reforms over recent years, and because of the decentralized model in Spain, which means that competencies and responsibilities are split between the Ministry of Education, Culture, and Sport and the regional administrations in the autonomous communities. The second part focuses on educational innovations and effective teaching behaviors resulting from policy changes and the traditional and dominant paradigms in the Spanish educational landscape. We use the six teaching effectiveness domains from the ICALT project as a reference: safe learning climate, efficient classroom management, clarity of instruction, activating teaching, teaching-learning strategies, and differentiation. The third part describes empirical research undertaken in three autonomous communities in Spain to assess teaching quality. We look at organizational, human, and curricular factors which can help in interpreting teaching standards and their impact on student engagement. Finally, the conclusions from the research are considered and discussed in terms of potential policy recommendations and practical decisions at both regional and national levels about teachers’ initial training, continued training, and professional development.

Abstract:
AbstractDifferentiated Instruction has been promoted as a model to create more inclusive classrooms by addressing individual learning needs and maximizing learning opportunities. Whilst differentiated instruction was originally interpreted as a set of teaching practices, theories now consider differentiated instruction rather a pedagogical model with philosophical and practical components than the simple act of differentiating. However, do teachers also consider differentiated instruction as a model of teaching? This chapter is based on a doctoral thesis that adopted differentiated instruction as an approach to establish effective teaching in inclusive classrooms. The first objective of the dissertation focused on how differentiated instruction is perceived by teachers and resulted in the DI-Quest model. This model, based on a validated questionnaire towards differentiated instruction, pinpoints different factors that explain differences in the adoption of differentiated instruction. The second objective focused on how differentiated instruction is implemented. This research consisted of four empirical studies using two samples of teachers and mixed method. The results of four empirical studies of this dissertation are discussed and put next to other studies and literature about differentiation. The conclusions highlight the importance of teachers’ philosophy when it comes to implementing differentiated instruction, the importance of perceiving and implementing differentiated instruction as a pedagogical model and the importance and complexity of professional development with regard to differentiated instruction.

Abstract Chapter 13 examines the important question of whether international environmental law is effective in solving environmental problems. It begins by outlining different meanings of effectiveness—distinguishing between legal effectiveness, behavioral effectiveness, and problem-solving effectiveness—and explaining the different ways in which the effectiveness of international environmental regimes can be measured. The chapter then discusses the extent to which international environmental regimes have been effective, suggesting that while progress is being made, the problem-solving effectiveness of international environmental law is still limited. The chapter end by discussing factors that help explain why some agreements have been more effective than others, distinguishing between exogenous factors that are outside the control of treaty negotiators (such as the nature of the problem) and endogenous factors that can be addressed through better treaty design.

Abstract This paper reviews the effect of temperature, H2S/CO2 ratio, time, velocity and inhibitor concentration on the corrosion of low carbon steel. Although considerable work has been reported in the literature on H2S and H2S/CO2 interaction with steel at ambient temperature and pressure, little work has been published under conditions found in deep hot sour gas wells. Laboratory data are presented and compared with relevant literature data showing the effects of the above variables.

Abstract:
ABSTRACT Chloride-Stress Corrosion Cracking (ClSCC) of austenitic stainless steels has been one of the biggest challenges in the refining industry and one of the main reasons where upgrading to stainless steel may not be the miracle solution for battling corrosion problems. Even with all that is known about this mechanism, the industry still faces failures, mainly because chlorides show up when they are not expected and accounted for, leading to economic or worse, catastrophic failures. Factors affecting ClSCC like pH, operating temperature and chloride ion concentration are considered in API 581 to designate a Severity Index. Other factors like the number of inspections, the effectiveness of the inspections and time since the last effective inspection etc. are considered to determine the Damage Factor. However, factors like presence of oxygen, effects of extreme pH and temperatures or stress relieving are not considered. A proposed Risk Assessment Tool using a new factor “susceptibility modifier” to API RP 581 Task Group was presented in November 2016 and has been incorporated in the newly released API 581 – Addendum 2 in October 2020.1 This paper will identify and document how these different factors affect the susceptibility of austenitic stainless steel to Chloride-Stress Corrosion cracking based on a review of currently available literature. A review of current industry best practices and a review of how the Oxygen content, the pH and application of stress relief affects Chloride-Stress Corrosion Cracking will be documented and presented.

Full text
Abstract:
Abstract The main theme of this paper is a concept: understanding and managing corrosion in nuclear power plants. The concept is not new--in various forms the concept has been applied throughout the development and maturing of nuclear technology. However, the concept has frequently not been well conceived and applied. Too often, understanding corrosion has been based on reaction rather than on anticipation. Regulatory and utility industry initiatives are creating a climate and framework for more effective application of the concept. This paper characterizes the framework and provides some illustrations of how the concept is being applied, drawing from work conducted under the Nuclear Plant Aging Research (NPAR) Program, sponsored by the Nuclear Regulatory Commission's (NRCs) Office of Research. Nuclear plants are becoming an increasingly important factor in the national electrical grid. Initiatives are currently underway to extend the operating licenses beyond the current 40-year period and to evaluate advanced reactor designs that feature higher safety factors. Corrosion has not caused a major nuclear accident, but numerous corrosion mechanisms have degraded nuclear systems and components. New corrosion phenomena continue to appear, and occasionally corrosion phenomena cause reactor shutdowns. Effective application of understanding and managing corrosion is important to safe and economic operation of the nuclear plants and also to public perception of a soundly operated technology.

Abstract:
Abstract Corrosion under insulation (CUI) has been a recognised issue in the North Sea for a number of years, resulting in considerable cost to operators in terms of safety and lost production implications. Several strategies and a variety of non-intrusive methods have been proposed and utilised with mixed success. The increasing number of mature assets operating beyond their original intended operating or design lives has necessitated the requirement for greater understanding in this area of corrosion management. This paper focuses on a series of case studies identifying the key contributory factors within each example and discusses the limitations of the first pass desktop approach. Experience in employing these differing techniques has verified that intimate knowledge of the system in question can greatly increase the probability of success and reduce the associated risks. This was found to include consideration of pipework wall thickness, degree of exposure and other less obvious factors. This practical approach employed in executing CUI strategies has produced greater success in identifying areas of concern. The inherent limitations of an abstract strategy are discussed, while highlighting some of the lessons learnt and the importance of applied local knowledge.

Abstract:
Abstract The "full ring" test has been recently developed as an intermediate type of test between small coupon tests and full scale testing under internal pressure in order to evaluate a linepipe for sour service conveniently. Factors of this test and their effects on test conditions or results have been Investigated in order to characterize the full ring test. The electrochemical hydrogen permeation measurement was effective to quantify the effects of environmental factors on test conditions. The hydrogen permeation coefficient (JxL) through a tested pipe was affected by not only pH and H2S content but also the galvanic corrosion and surface conditions. The metallurgical factors for hydrogen cracking were both nonmetallic inclusions in linepipe steels and welding defects in circumferential weldments. The JxL was also used to estimate a resistance exactly. The threshold value estimated from the full ring test results will show an excellent resistance in the actual environment.

Abstract:
Empirically measuring the Purcell factor, which represents the maximum enhancement to spontaneous emission in an optical cavity, is straightforward as the photoluminescence enhancement (PLE) can be determined directly by comparing the luminescence intensity with and without the cavity. However, predicting the Purcell factor have been proven to be tricky. In particular, the effective modal volume is difficult to analyze empirically, especially for non-Hermitian systems which includes open resonators and systems with dissipative materials [1].

Abstract:
In turbomachinery, annular seals are used to reduce leakage between regions of high and low pressure. Many configurations of annular seals have been developed and studied in the literature including plain, labyrinth, pocket-damper, honeycomb, and hole-pattern. In machines experiencing stability issues, honeycomb and hole-pattern type seals have been used to replace labyrinth seals. Bulk-flow models are typically used to predict the leakage and dynamic coefficients of hole-pattern seals, relying on empirically derived friction factor coefficients. Previous experimental studies have shown that, for hole-pattern seals, the leakage and stator friction factor are strongly influenced by hole-depth. However, this behavior is not a monotonic function of hole-depth, a fact that might reduce confidence in future bulk-flow model predictions if not properly accounted for. A recent numerical study has highlighted the role of vortex formation in the holes which has a strong influence on the flow in the clearance region. Depending on the shape of the vortex, the flow in the hole can act much like a pinch valve, reducing the effective clearance of the jet flow. In this paper, computational fluid dynamics simulations of several hole-pattern seal configurations have been performed to study the effect of hole-aspect ratio (depth versus diameter) on the leakage and friction factors. The Reynolds Averaged Navier Stokes (RANS) equations with k-ε turbulence model were solved using ANSYS CFX. It was found that the shape of the hole influences the vortex formation within the hole, effecting the jet flow in the clearance region and the seal leakage. The results show that the leakage is heavily dependent on the hole diameter in addition to the hole depth. The relationship between the friction factors and the geometry of the seal was found to be non-monotonic. It is therefore difficult to develop a friction factor model that will accurately encompass all configurations and it is recommended that friction factor data be interpolated from experimental or numerical results.

The pandemic has forced higher education to radically change the teaching format from face-to-face mode into virtual or hybrid format. Also, intensive courses, workshops and seminars have been conducted totally or partly online. These formats include lots of students` team work in order to succeed. Starting to work virtually with strangers might be difficult for students. Our paper presents how we developed and conducted two interdisciplinary team learning projects during the academic year 2021-2022 and the results we got. Our projects were implemented in the BSc degree programs of engineering, business administration and nursing at two Finnish UAS and at two foreign partner universities. After the courses we collected qualitative data from students´ learning diaries and other reflections and performed content analysis which revealed that there are supportive and hindering factors for virtual team work and they should be considered while designing the learning experiences.

Financial turmoil is becoming a fact of life in Latin America. The 1990s have been characterized by enormous volatility in the magnitude and cost of capital flows. The correlation of capital swings across disparate countries suggests that the quality of emerging market policies in addition to global factors have been the main actors in this drama. Therefore, the blame for financial turmoil has moved away from inappropriate domestic policies. Instead, the paradigm has shifted to one of determining which policies - domestic or international - are most effective in taming the destabilizing effects of inherently volatile capital flows.

Abstract:
Pedestrians are vulnerable road users since they are prone to more severe injuries in any vehicular collision. While innovative solutions promise improved pedestrian safety, a careful analysis of local conditions is required before selecting proper corrective measures. This research study had two focuses: (1) methodology to identify roads and areas in Indiana where the frequency and severity of pedestrian collisions are heightened above the acceptable level, and (2) selecting effective countermeasures to mitigate or eliminate safety-critical conditions. Two general methods of identifying specific pedestrian safety concerns were proposed: (1) area-wide analysis, and (2) road-focused analysis. A suitable tool, Safety Needs Analysis Program (SNAP), is currently under development by the research team and is likely the future method to implement an area-wide type of analysis. The following models have been developed to facilitate the road-focused analysis: (1) pedestrian crossing activity level to fill the gap in pedestrian traffic data, and (2) crash probability and severity models to estimate the risk of pedestrian crashes around urban intersections in Indiana. The pedestrian safety model was effectively utilized in screening and identifying high-risk urban intersection segments for safety audits and improvements. In addition, detailed guidance was provided for many potential pedestrian safety countermeasures with specific behavioral and road conditions that justify these countermeasures. Furthermore, a procedure was presented to predict the economic feasibility of the countermeasures based on crash reduction factors. The findings of this study should help expand the existing RoadHAT tool used by the Indiana Department of Transportation (INDOT) to emphasize and strengthen pedestrian safety considerations in the current tool.

Full text
Abstract:
This Evidence Check was commissioned by the NSW Ministry of Health, as part of a project to improve how preventive, sensitive health issues are raised in general practice. The review looked at what is known about discussing sensitive preventive health issues from both patients and GPs perspectives and approaches and factors that have been shown to be effective. The identified evidence was generally of moderate to high methodological quality. General behaviour change approaches that are applicable to this challenge include creating non-judgemental environments that normalise sensitive health issues; simulation training; and public campaigns that reduce stigma and challenge unhelpful cultural norms. Lack of time in consultations was identified as a challenging issue. Significant system-level change would be required to extend standard consultation times; focusing on optimising workflows may therefore be more feasible. Addressing GP patient–gender mismatch through diverse GP representation may also be feasible in larger practices. The key theme identified was the use of prompting, screening or other structured tools by GPs. Collectively, these approaches have two main features. First, they are a way of approaching sensitive health conversations less directly, for example by focusing on underlying risk factors for sensitive health conditions such as obesity and mental illness rather than addressing the issues directly. Second, through either risk-factor or more general question prompts, these approaches take the onus away from GPs and patients to come up with a way of asking the question using their own words.

The aim of this paper is to offer a brief analysis of the social investment funds for indigenous people that have been financed by the Inter-American Development Bank (IDB) and to consider how far they have been effective in meeting the needs of indigenous people. This paper identifies two types of targeting mechanisms: 1) Inclusive funds that do not distinguish indigenous people from other sectors of society based on ethnicity; and 2) Exclusive funds or components intended only for the indigenous or ethnic population. The author discusses the merits of both approaches and identifies four factors that determine their effectiveness in reaching targeting goals: 1) Geographic isolation; 2) Social exclusion based on racial of ethnic prejudice; 3) Social and cultural factors including language, structures of authority and economic values; 4) The capacity of the indigenous organizations.

Full text
Abstract:
Policymaking in Peru over the last 25 years has been largely dominated by the Executive, and has been influenced by a variety of structural and political factors as well as by the personal ambitions of presidents and the public perception of crisis. With few exceptions, neither the Congress nor the other branches and levels of government have played effective roles in defining the national policy agenda, promoting inter-temporal cooperation and providing checks and balances on executive power. Although this situation has been partially modified since 2001, it is not clear that the general pattern has changed. While certain arenas of decision-making have been reformed in recent years, in many spheres policymaking remains an arbitrary and unpredictable process, resulting in policies that are of low quality, poorly enforced and easily reversed. Although reforming aspects of the political and electoral systems could contribute to improving this outcome, the instability of the political regime per se has been a deterrent to longer-term institutional development.

Full text
Abstract:
Staphylococcus aureus is a major cause of mastitis in dairy cattle. The organism is able to adhere to and penetrate mammary epithelium, forming deep seated abscesses that result in chronic infections. This study was based on the observation that certain genotypes of S. aureus are isolated more frequently from field cases of bovine mastitis than others and the most prevalent genotypes of S. aureus have an increased ability to resist neutrophil phagocytosis and killing compared to the rare variants. It was hypothesized that these predominating genotypes differentially express virulence factors that allow them to overcome or suppress essential host defense mechanisms and successfully colonize mammary parenchyma. The overall objective of this study was to determine the mechanisms by which predominating S. aureus genotypes were able to resist mammary gland defense mechanisms. The following specific aims were accomplished to address the overall objectives of this project: 1. Analyze and compare cell surface and secreted protein profiles of common and rare S. aureus genotypes isolated from field cases of bovine mastitis. 2. Purify and sequence selectively synthesized proteins unique to the most prevalent genotypes of S. aureus . 3. Determine the in vitro effects of isolated proteins on essential host defense mechanisms. Results from each specific aim showed that these redominating genotypes differentially express factors that may allow them to overcome or suppress essential host defense mechanisms and successfully colonize mammary parenchyma. Using complementary approaches, both the US and Israeli teams identified differentially expressed S. aureus factors that were positively correlated with virulence as determined by the ability to modify host immune cell responses and increase disease pathogenesis. Several candidate virulence factors have ben identified at both the molecular (US team) and protein (Israeli team) levels. Components of the phosphotransferase system were shown to be differentially expressed in prevalent strains of S. aureus and to modify the growth potential of these strains in a milk microenvironment. Evidence provided by both the Israeli and US teams also demonstrated a potential role of Staphylococcal enterotoxins in the pathogenesis of mastitis. Certain enterotoxins were shown to directly affect neutrophil bactericidal activities which can profoundly affect the establishment of new intramammary infections. Other evidence suggests that S. aureus superantigens can suppress mammary defenses by enhancing lymphoid suppressor cell activity. Collectively, these data suggest that unique factors are associated with predominating S. aureus genotypes that can affect in vitro and in vivo virulence as related to the pathogenesis of bovine mastitis. The potential development of a subunit mastitis vaccine which incorporates only relevant antigenic determinants has not been investigated in depth. Experiments outlined in this proposal has identified putative virulence factors which contribute to the pathogenesis of S. aureus mastitis and which may be used to formulate an efficacious subunit mastitis vaccine. Results from these studies may lead to the development of new methods to prevent this costly disease, providing a viable alternative to less effective mastitis control procedures based on chemotherapy.
