Abstract:
In this article I explore the ways in which dogs and other companion species become family members and engage with the argument that this indicates the emergence of post-human families. Using empirical data from responses to a Mass Observation directive on Animals and Humans and in-depth interviews with people who share their homes with companion animals, I explore the ways in which humans and dogs live with each other and the ‘daily practices of kinship’ which constitute them as kin. I argue that practices of kinship blur the species barrier but that human-dog relations take place in the context of unequal power relations which are an inevitable consequence of dogs’ incorporation into families as dependents. I conclude that while it may be possible to identify post-human practices in multi-species households, they exist alongside practices which reinforce the human-animal boundary and that, given the unequal relations of entanglement within which humans and animals interact, attempts to identify empirically a post-human family seem problematic. What can be said, however, is that a post-human approach to kinship practices highlights the porousness of the category human and alerts us both to the deep connections between humans and other animals and to the profoundly unequal ways in which animals are incorporated into social relations with humans.

Abstract:
The lives of families who take care of people with disabilities are full of unexpected events, unwanted situations, and difficulties that accumulate at every step. Therefore, bearing in mind the purpose of this paper, I focused on determining those conditions which are a source of destabilization in the lives of families with intellectually disabled children. I attemptto reconstruct the sources of threats to the sense of subjectivity and the shaping of the identity of parents who have children with disabilities. The theoretical framework of the analysis is symbolic interactionism. The research material used in the presented article is composed of personal experiences of parents of disabled individuals, and unstructured interviews were performed with these people. Analysis of the research material was performed in accordance with the procedures of grounded theory methodology.

Abstract:
Finding the chromosomal location of human genes that heretofore have been defined solely by phenotypes, in particular clinical phenotypes that are transmitted in Mendelian fashion in families, is an early and often crucial step in the process of identifying the molecular basis of a disease. Recent progress in construction of chromosomal maps of genetically linked DNA markers has made almost the entire human genome accessible to linkage studies in families that are segregating genetic defects. Construction of linkage maps requires a panel of three-generation families for genotyping, a large number of polymorphic markers, and sophisticated computer programs for analysis of genotypic data. After a locus harboring a deleterious mutation has been identified by linkage to a mapped marker, a high-resolution map of the region can be constructed with new markers derived from cosmid libraries, to narrow the search for the gene in question. For example, this strategy has been pursued in the effort to characterize the gene responsible for familial adenomatous polyposis. When a target region has been narrowed to about 1 centiMorgan, corresponding to roughly a million base pairs in physical distance, other techniques of molecular biology can be brought to bear to isolate and clone the actual gene.Key words: genetic linkage, chromosome maps, DNA markers, chromosome 17, chromosome 10, genetic disease, familial adenomatous polyposis.

Abstract:
Social workers and other human services professionals helping families reintegrate after parental incarceration deal with multiple issues without a model of for facilitating family resilience. The purpose of this phenomenological research study was to explore the essence of the perceived role, activities, and practices of a sample of social workers and other human service professionals engaged in the use of family group conferences (FGCs). FGCs are also referred to as restorative justice, as they inform and assist human service professionals in developing clinical interventions and best practices to support reintegration, family preservation, and stabilization. The framework for this study was built around restorative justice theory, resiliency theory, and a larger social ecological theory and focused on the use of FGCs as a developing practice within family systems and the community. The primary research questions investigated the practitioners' experiences using FGCs. Data came from interviews of participants (15) drawn from professional associations and included their own case notes and reflections. The data was sorted and analyzed with the assistance of qualitative analysis software (Atlas.Ti7) to search for themes that may assist in identifying the phenomenon. The findings suggest that the FGC model should consist of a training curriculum, consistent practice, and dedicated and committed financial resources to support programs. This study impacts social change by informing human services professionals of current best practices and may provide a model of FGCs that will help implement services to families.

Partitioning an individual's phenotype into genetic and environmental components has been a major goal of genetics since the early 20th century. Formally, the proportion of phenotypic variance attributable to genetic variation in the population is known as heritability. Genome wide association studies have explained a modest percentage of variability of complex traits by genotyping common variants. Currently, there is great interest in what role rare variants play in explaining the missing heritability of complex traits. Advances of next generation sequencing and genomic enrichment technologies over the past several years have made it feasible to re-sequence large numbers of individuals, enabling the discovery of the full spectrum of genetic variation segregating in the human population, including rare variants. The four projects that comprise my dissertation all revolve around the discovery of rare variants from next generation sequencing datasets. In my first project, I analyzed data from the exon sequencing pilot of the 1000 Genomes Project, where I discovered variants from exome capture sequencing experiments in a worldwide sample of nearly 700 individuals. My results show that the allele frequency spectrum of the dataset has an excess of rare variants. My next project demonstrated the applicability of using whole-genome amplified DNA (WGA) in capture sequencing. WGA is a method that amplifies DNA from nanogram starting amounts of template. In two separate capture experiments I compared the concordance of call sets, both at the site and genotype level, of variant calls derived from WGA and genomic DNA. WGA derived calls have excellent concordance metrics, both at the site and genotypic level, suggesting that WGA DNA can be used in lieu of genomic DNA. The results of this study have ramifications for medical sequencing experiments, where DNA stocks are a finite quantity and re-collecting samples maybe too expensive or not possible. My third project kept its focus on capture sequencing, but in a different context. Here, I analyzed sequencing data from Mendelian exome study of non-sensorineural hearing loss (NSHL). A subset of 6 individuals (5 affected, 1 unaffected) from a family of European descent were whole exome sequenced in an attempt to uncover the causative mutation responsible for the loss of hearing phenotype in the family. Previous linkage analysis uncovered a linkage region on chr12, but no mutations in previous candidate genes were found, suggesting a novel mutation segregates in the family. Using a discrete filtering approach with a minor allele frequency cutoff, I uncovered a putative causative non-synonymous mutation in a gene that encodes a transmembrane protein. The variant perfectly segregates with the phenotype in the family and is enriched in frequency in an unrelated cohort of individuals. Finally, for my last project I implemented a variant calling method for family sequencing datasets, named Pgmsnp, which incorporates Mendelian relationships of family members using a Bayesian network inference algorithm. My method has similar detection sensitivities compared to other pedigree aware callers, and increases power of detection for non-founder individuals

Abstract:
Two closely related human Arls, Arl8a and Arl8b, were found to localise to lysosomes in mammalian cells. conventionally, membrane binding of Arf and Arl proteins is mediated by both an N-terminal myristoyl group and an N-terminal amphipathic helix that are inserted into the lipid bilayer upon activation of the GTPase. Arl8 GPTases lack myristolylation sites, and examination of the N-terminus of Arl8b revealed that it contains an acetyl group instead, and this acetylated methionine is necessary for its lysosomal location. Lysosomes of cells overexpressing Arl8b move more frequently, suggesting a role for Arl8a and Arl8b as positive regulators of lysosomal transport. Arl4a, Arl4c and Arl4d are very similar in sequence and were found to act in a pathway upstream of Arf6, Arf6 is a regulator of key processes at the plasma membrane, such as endocytosis, actin dynamics and cell adhesion. One of the major activators of Arf6 is the exchange factor ARNO (‘Arf nucleotide binding site opener’). In order to activate Arf6, ARNO must translocate from the cytoplasm onto the plasma membrane. ARNO was identified as an interaction partner of Arl4 GTPases in a yeast two hybrid screen, and Arl4 GTPases mediate membrane translocation of ARNO. Thus, Arl4 GTPases represent a novel signal transduction pathway upstream of Arf6. This study provides a characterisation of members of the human Arl family and suggests functions for Arl4 and Arl8 GTPases in membrane traffic related processes.

For most Caribbean countries, the COVID-19 pandemic will translate into the deepest single-year contraction of real GDP on record in 2020. With the exception of Guyana, countries have experienced deep recessions, severe increases in unemployment, and long-lasting damage to many corporate and household balance sheets. The social consequences of the crisis continue to mount, and despite governments best efforts to buffer the shock to families, enterprises, and domestic markets, there remains a dire need for continued and more broad-based stimulus to ensure that economic capital both human and other wise remains intact. Venezuela experimenta la recesión económica más severa y prolongada de su historia económica moderna. El país ha perdido tres cuartas partes de su Producto Interno Bruto, produciendo, como consecuencia, una degradación extrema de las condiciones materiales de vida de sus habitantes. En este contexto, no existe una evaluación apropiada de los efectos que esta profunda crisis económica ha tenido sobre los indicadores socioeconómicos de las clases medias en Venezuela. Este documento técnico hace uso de las fuentes de microdatos disponibles para analizar empíricamente el tamaño y la evolución reciente de la clase media venezolana a la luz de la reciente crisis económica. En líneas generales, se corrobora una caída abrupta y sistemática tanto en el tamaño de la clase media venezolana, así como del poder de consumo de quienes todavía forman parte de ella. En 2020 se verifica que aproximadamente 9 de cada 10 familias que era considerada de clase media a principios de la década pasada, ya no lo es. Además, el análisis dinámico de los datos muestra evidencia de que una porción muy importante de familias de clase media ha perdido su estatus sobretodo en los últimos años, tendencia comprobada por la evolución indicadores no monetarios, relacionados con los niveles multidimensionales de bienestar y seguridad económica de las familias de estratos medios. Venezuela llegó a ser el hogar de la más grande, sólida y próspera clase media de la región, hoy en día, la situación ha cambiado radicalmente: una parte importante de el acervo de capacidades y capital humano de la clase media ha alimentado la ola de emigración de venezolanos de los últimos años, mientras la otra parte, la poción que permaneció en Venezuela, sufre la erosión de sus condiciones en el marco de la depresión económica y la crisis humanitaria. En general, existe amplio consenso sobre el efecto positivo que tiene la clase media sobre el bienestar, el crecimiento económico, la equidad y la estabilidad de las economías, en este sentido, la rehabilitación de la clase media venezolana deberá formar parte de cualquier programa de recuperación futura.

To deliver infrastructure that sustain our communities, economy, and environment, we must innovate, modernize, and even revolutionize our approach to infrastructure development. Change takes courage, but as one starts down the path of innovation, what was once novel becomes more familiar, more established. The U.S. Army Corps of Engineers (USACE) is walking this path with our partners through the Engineering With Nature (EWN) Initiative, integrating human engineering with natural systems. The International Guidelines on Natural and Nature-Based Features for Flood Risk Management are the next step toward revolutionary infrastructure development—a set of real-world guidelines to help familiarize us with what was once novel. USACE and collaborators around the world have been building, learning, and documenting the best practices for constructing Natural and Nature-Based Features (NNBF) for decades. The consolidation of these lessons into a single guidance document gives decision-makers and practitioners a much-needed resource to pursue, consider, and apply NNBF for flood risk management while expanding value through infrastructure. Relationships and partnerships are vital ingredients for innovation and progress. The NNBF Guidelines was achieved because of the strong relationships in the nature-based engineering community. The magnitude and diversity of contributors to the NNBF Guidelines have resulted in a robust resource that provides value beyond a single agency, sector, or nation. Similarly, the work of incorporating NNBF into projects will require us to strengthen our relationships across organizations, mandates, and missions to achieve resilient communities. I hope you are inspired by the collaborative achievement of the NNBF Guidelines and will draw from this resource to develop innovative solutions to current and future flood risk management challenges. There is a lot we can achieve together along the path of revolutionary infrastructure development.

Abstract:
Un desafío clave de la política pública es cómo llevar a escala programas de primera infancia efectivos. Si la entrega de estos programas pudiera hacerse a través de la infraestructura de servicios públicos existentes, esto resultaría no solo escalable sino más costo-eficiente. Este documento es de los primeros en evaluar los impactos de corto plazo de un programa de visitas domiciliarias a escala integrado completamente con los servicios de atención primaria en salud (en este caso, la Estratégia Saúde da Família de Brasil). Nuestra estrategia de identificación explota la aleatorización original del programa Cresça Com Seu Filho en Fortaleza a nivel de agente comunitario de salud (ACS). En la estimación de intención de tratamiento (ITT, por sus siglas en inglés) encontramos un impacto positivo modesto en la calidad del entorno familiar (0.12 desvíos estándar, DE). Este resultado se alinea con el foco del currículo del programa en el cuidado sensible, receptivo y no punitivo de los niños. No encontramos evidencia de impacto en desarrollo infantil, muy probablemente debido a los desafíos con la intensidad y la fidelidad de las visitas. Nuestros resultados apuntan a la importancia de mejorar los sistemas de supervisión y acompañamiento del personal para lograr mejoras en la fidelidad de la intervención, así como en esquemas de motivación intrínseca del recurso humano que, en un contexto de mayor carga laboral, resulta central para el éxito de este tipo de intervenciones.

Abstract:
South Africa is a country with significant socio-economic development challenges, with the majority of South Africans having limited or non-existent access to basic infrastructure, services, housing and socio-economic opportunities etc. The urban housing backlog currently exceeds 2.4 million houses, with many families living in informal settlements. The Breaking New Grounds Policy, 2014 for the creation of sustainable human settlements, acknowledges the challenges facing human settlements, such as, decreasing human settlements grants allocation, increasing housing backlog, mushrooming of informal settlements and urbanisation. The White Paper on Science, Technology and Innovation (STI), 2019 notes that South Africa has not yet fully benefited from the potential of STI in addressing the socio-economic challenges and seeks to support the circular economy principles which entail a systematic change of moving to a zero or low waste resource-efficient society. Further to this, the Science and Technology Roadmap’s intention is to unlock the potential of South Africa’s human settlements for a decent standard of living through the smart uptake of science, technology and innovation. One such novel technology is the Three-Dimensional (3D) printing technology, which has produced numerous incredible structures around the world. 3D printing is a computer-controlled industrial manufacturing process which encompasses additive means of production to create 3D shapes. The effects of such a technology have a potential to change the world we live in and could subsequently pave the roadmap to improve on housing delivery and reduce the negative effects of conventional construction methods on the environment. To this end, the Academy of Science of South Africa (ASSAf), in partnership with the Department of Science and Innovation (DSI) and the University of Johannesburg (UJ) hosted the second virtual IID seminar titled: Exploring the Prospects of Using 3D Printing Technology in the South African Human Settlements, on 01 March 2021 to explore the potential use of 3D printing technology in human settlements. The webinar presented preliminary findings from a study conducted by UJ, addressing the following topics: 1. The viability of 3D printing technology 2. Cost comparison of 3D printed house to conventional construction 3. Preliminary perceptions on 3D printing of houses Speakers included: Dr Jennifer Mirembe (NDoHS), Dr Jeffrey Mahachi, Mr Refilwe Lediga, Mr Khululekani Ntakana and Dr Luxien Ariyan, all from UJ.
