Abstract:
ABSTRACT In budding yeast, diffusible mating pheromones initiate a signaling pathway that culminates in several responses, including cell cycle arrest. Only a handful of genes required for the interface between pheromone response and the cell cycle have been identified, among them FAR1 and FAR3; of these, only FAR1 has been extensively characterized. In an effort to learn about the mechanism by which Far3 acts, we used the two-hybrid method to identify interacting proteins. We identified five previously uncharacterized open reading frames, dubbed FAR7, FAR8, FAR9, FAR10, and FAR11, that cause a far3-like pheromone arrest defect when disrupted. Using two-hybrid and coimmunoprecipitation analysis, we found that all six Far proteins interact with each other. Moreover, velocity sedimentation experiments suggest that Far3 and Far7 to Far11 form a complex. The phenotype of a sextuple far3far7-far11 mutant is no more severe than any single mutant. Thus, FAR3 and FAR7 to FAR11 all participate in the same pathway leading to G1 arrest. These mutants initially arrest in response to pheromone but resume budding after 10 h. Under these conditions, wild-type cells fail to resume budding even after several days whereas far1 mutant cells resume budding within 1 h. We conclude that the FAR3-dependent arrest pathway is functionally distinct from that which employs FAR1.

It should be noted that, PMBOK 2004 will be used for definitions of project management and its framework for analyzing of project management risks and developing competency development program.

Abstract:
I samband med omorganiseringen av en av sina monteringslokaler, vill företaget Camfil Svenska AB se över lokalens tak. Verkstaden har i dagsläget ett väldigt lågt tak av lättbetong och förmodas ha dåliga isoleringsegenskaper som, i företagets ögon, skapar onödiga kostnader för uppvärmning respektive kylning. Ljudnivån vid drift skapar dessutom en bullrig arbetsatmosfär, som man gärna gör något åt. Fördelen med lättbetongen är dess höga brandklass, vilken företaget helst bibehåller. En offert för ombyggnad av taket till en konstruktion med ljuddämpande innertaksplattor, ett yttertak av korrugerad plåt och rejäl isolering är inhämtad från en byggfirma. Camfil vill dock inte utesluta alternativet tilläggsisolering, med invändig, ljuddämpande sprutisolering. Dessa två alternativ har jämförts med varandra och med den nuvarande takkonstruktionen, primärtmed avseende på hur väl de värmeisolerar. Även akustik, brandbeständighet och vilka möjligheter alternativen ger gällande andra nödvändiga installationer i lokalen är faktorer som undersökts. En utvärderingsmodell har tagits fram och använts på de olika alternativen. Modellen indikerar att man i detta fall inte kan räkna med tillräcklig återbetalning på något av investeringsalternativen. Kostnadsbesparingarna är alltså inte tillräckligt stora för att de ska vara ekonomiskt försvarbara. Det som kan sägas är att en klar förbättring i akustik ges av de båda förändringsalternativen. I alternativet för ombyggnad får man dessutom en utformning som förbättrar möjligheterna för andra verkstadsinstallationer. Med det ekonomiska perspektivet som det tyngsta, bör man emellertid utvärdera andra möjligheter att förbättra de icke uppfyllda faktorerna när inget av åtgärdsalternativen genomförs.
While reorganizing an assembly workshop, the company of Camfil Svenska AB wants to look over its ceiling. Today, the ceiling is built with lightweight concrete, which expects to perform poorly in thermal insulation and that leads to, as the company sees it, unnecessary high costs of heating and cooling. The level of noise during production hours is also a concern. In manners regarding firesafety the company does not want to lower the standard, as it is already high. A building company has made a prospect of costs and construction of a new roof. Their solution consists of an inner ceiling with sound absorbing board and an outer one with a thick layer of thermal insulation lying on corrugated iron. Camfil does not want to rule out the alternative to put an extralayer of insulation on the inside of the current roof either. A comparison between the two alternatives and with the current construction on how they perform as thermal insulators has been done. Also aspects as acoustics, fire-resistance, and use of the workshop have been taken in consideration. A model for evaluating the different scenarios was design and used on the different alternatives. The model shows that neither of the contemplated changes showed a good enough improvement to justify the costs that are related to enforce them. A huge improvement in acoustics was evident in both considered alternatives and better possibilities to use the workshop in the alternative with a newroof could be seen. Having the economical aspect as the strongest, other possibilities to improve characteristics that do not meet expectations with these alternatives should be investigated.

Ce travail comparatif étudie les ressemblances thématiques, symboliques et narratives entre plusieurs romans du Graal de la fin du XIIe siècle et du 1er quart du XIIIe siècle [le Conte du Graal de Chrétien de Troyes, le Parzival de Wolfram von Eschenbach, la Troisième Continuation du Conte du Graal de Manessier, le Haut Livre du Graal], et l’épopée persane du Livre des rois de Ferdowsi, achevée au début du XIe siècle. On étudie d’abord les ressemblances entre le Graal et le farr ou gloire royale iranienne : tous deux se caractérisent par la luminosité, constituent des sources d’abondance, ont une relation particulière avec les êtres, sont liés à la royauté. On étudie ensuite les destins parallèles du chevalier Perceval et du roi iranien Key Khosrow, en mettant en évidence les ressemblances entre les moments clés de leur existence : leur lignage, leur jeunesse orpheline loin du monde, leurs qualités spirituelles et royales, les épreuves qu’ils traversent, leur consécration, leur relation au sacré [au Graal pour le premier, au farr pour le second]
This comparative work examines the thematic, symbolic and narrative similarities between several texts of the Grail legend written between the late 12th century and the 1st quarter of the 13th century [the Conte du Graal of Chrétien de Troyes, Parzival of Wolfram von Eschenbach, the Troisième Continuation du Conte du Graal of Manessier, le Haut Livre du Graal], and the Book of Kings of Ferdowsi, a Persian epic completed at the beginning of the 11th century. A first part examines the similarities between the Holy Grail and the farr [the Iranian royal Glory]: both are characterized by light, are sources of abundance, have a special relationship with human beings, are linked to the kingship. A second part examines the destinies of the knight Perceval and of the Iranian King Key Khosrow, highlighting the similarities between the key moments of their lives: their lineage, their orphaned youth away from the world, their spiritual and royal qualities, their ordeals, their consecration, their relationship to the sacred [to the Grail for the former, to the farr for the latter]

Abstract:
This thesis examines the activities of the Statistical Society of London (SSL) and its contribution to early statistics-conceived as the science of humans in society-in Britain. The SSL as a collective entity played a crucial role in the formation of early statistics, as statisticians envisaged early statistics as a collaborative scientific project and prompted large-scale observation, which required cooperation among numerous statistical observers. The first three chapters discuss how the SSL shaped the concepts, practices, and institutions of statistical data production. The SSL demonstrated how the use of a hierarchical division of labour and blank form minimised observers' leeway to exercise individual observational skills and ensured uniformity in the production of statistical facts. This arrangement effectively depreciated first-hand observation in statistics and allowed statisticians to rely on the statistical facts collected by other people. It prompted the SSL to launch the Journal of the Statistical Society of London to serve as a virtual storage of observed facts where one could share their data for further aggregation and retrieve that of others for their analysis. The statisticians also engaged in contemporaneous discussion on the best mode of a statistical office with a view towards producing complete and internationally comparable statistical facts. The SSL's endorsement of the Belgian Central Statistical Commission model and the International Statistical Congress was intended to support the introduction of uniformity into statistical data at both the national and international levels. The last two chapters of this thesis discuss how the SSL's activities contributed to the historical formation of human sciences and the emergence of social scientists. Statisticians demanded the recognition of a scientific field which, independent from natural science, studied people as social beings and whose discourses moulded the treatment of the people they studied. The SSL's activities helped statisticians not only establish their scientific expertise but also develop their unique scientific ethos. Statisticians learnt not to trust their personal observations since individuals could see only a partial, and potentially distorted, picture of society. Instead, statisticians disciplined themselves to patiently wait for the accumulation of statistical facts and analyse data in their entirety because this was the only way, they believed, to truly understand the complex relationships people had with each other. The SSL's activities assisted statisticians' conception of statistical fact and produced a new kind of intellectual inquirer who patiently collected statistical facts as the basis of knowing and intervening in people's lives.

Abstract:
The new connection designs introduced by the drill pipe manufacturers (double shoulder connections, intelligent drill pipes or any new design for increased torque resistance) make the use of the Farr formula for calculating proper assembly torque more problematic. Additionally, severe drilling conditions like HPHT, directional drilling and extreme environments are affecting critical thread compound performance properties, which can make it impossible to attain the optimum makeup torque for the connection. The new drilling conditions are exposing the drill string components to higher loads and consequently reduce the safety margins. Since the makeup process is the only way to control and achieve the maximum tool joint loading capacity, it becomes important to understand the effect of friction on optimum makeup torque calculation. This paper presents a modified equation for optimum makeup torque calculation by using the latest research on thread compound frictional properties. Also, the thread compound lubrication mechanism will be explained. For the first time the thread turn load will be related to the optimum makeup torque. The paper will also explain why there is a need for a new API standard for determining thread compound frictional properties. The formula developed herein, based on experimental results, demonstrates that the newly introduced overall factor may differentiate between used and new connections.

Pressure vessel towers used in the petrochemical and chemical industry are designed to accommodate numbers of internals including trays and beds resulting in tall vertical structures. Transportation of tall towers from the fabrication shop to the construction site presents challenges that can result in high transportation costs or a logistically impossible task of moving the vessel. One of the solutions to this problem is to shorten the tower for transport by cutting part of the tower skirt and welding it in the field. Depending on the location, welding on site can be expensive, labour intensive and may cause problems in the quality of the weld and the tower being out of level. Using a flanged skirt connection will reduce the field labour spent on connecting the bottom part of the skirt to the rest of the vessel. The challenge that lies in front of designers is that the current codes and available literature do not give a specific design and calculation guidance for implementing such a solution. This paper looks at different analytical methods to be used for the design of a skirt splice. Methods provided by Jawad and Farr, the Canadian Institute of Steel Construction, the American Institute of Steel Construction, and the Peterson Method from the European Commission’s High-Strength Tower in Steel for Wind Turbines (HISTWIN) are analyzed. Based on this analysis, the most optimal and safe design and fabrication methodology for implementing a Flanged Skirt Connection is proposed.

Abstract:
Although peer prediction markets are widely used in crowdsourcing to aggregate information from agents, they often fail to reward the participating agents equitably. Honest agents can be wrongly penalized if randomly paired with dishonest ones. In this work, we introduce selective and cumulative fairness. We characterize a mechanism as fair if it satisfies both notions and present FaRM, a representative mechanism we designed. FaRM is a Nash incentive mechanism that focuses on information aggregation for spontaneous local activities which are accessible to a limited number of agents without assuming any prior knowledge of the event. All the agents in the vicinity observe the same information. FaRM uses (i) a report strength score to remove the risk of random pairing with dishonest reporters, (ii) a consistency score to measure an agent's history of accurate reports and distinguish valuable reports, (iii) a reliability score to estimate the probability of an agent to collude with nearby agents and prevents agents from getting swayed, and (iv) a location robustness score to filter agents who try to participate without being present in the considered setting. Together, report strength, consistency, and reliability represent a fair reward given to agents based on their reports.

Abstract:
Objectives: (1) Preparing panel data sets for both the United States and Israel that contain a rich set of farm attributes, such as size, specialization, and output composition, and farmers’ characteristics such as off-farm employment status, education, and family composition. (2) Developing an empirical framework for the joint analysis of all the endogenous variables of interest in a dynamic setting. (3) Estimating simultaneous equations of the endogenous variables using the panel data sets from both countries. (4) Analyzing, using the empirical results, the possible effects of economic policies and institutional changes on the dynamics of the farm sector. An added objective is analyzing structural changes in farm sectors in additional countries. Background: Farm sectors in developed countries, including the U.S. and Israel, have experienced a sharp decline in their size and importance during the second half of the 20th century. The overall trend is towards fewer and larger farms that rely less on family labor. These structural changes have been a reaction to changes in technology, in government policies, and in market conditions: decreasing terms of trade, increasing alternative opportunities, and urbanization pressures. As these factors continue to change, so does the structure of the agricultural sector. Conclusions: We have shown that all major dimensions of structural changes in agriculture are closely interlinked. These include farm efficiency, farm scale, farm scope (diversification), and off-farm labor. We have also shown that these conclusions hold and perhaps even become stronger whenever dynamic aspects of structural adjustments are explicitly modeled using longitudinal data. While the results vary somewhat in the different applications, several common features are observed for both the U.S. and Israel. First, the trend towards the concentration of farm production in a smaller number of larger farm enterprises is likely to continue. Second, at the micro level, increased farm size is negatively associated with increased off-farm labor, with the causality going both ways. Third, the increase in farm size is mostly achieved by diversifying farm production into additional activities (crops or livestock). All these imply that the farm sector converges towards a bi-modal farm distribution, with some farms becoming commercial while the remaining farm households either exit farming altogether or continue producing but rely heavily on off-farm income. Implications: The primary scientific implication of this project is that one should not analyze a specific farm attribute in isolation. We have shown that controlling for the joint determination of the various farm and household attributes is crucial for obtaining meaningful empirical results. The policy implications are to some extent general but could be different in the two countries. The general implication is that farm policy is an important determinant of structural changes in the farm sector. For the U.S., we have shown the different effects of coupled and decoupled (direct) farm payments on the various farm attributes, and also shown that it is important to take into account the joint farm-household decisions in order to conduct a meaningful policy analysis. Only this kind of analysis explains the indirect effect of direct farm payments on farm production decisions. For Israel, we concluded that farm policy (or lack of farm policy) has contributed to the fast structural changes we observed over the last 25 years. The sharp change of direction in farm policy that started in the early 1980s has accelerated structural changes that could have been smoother otherwise. These accelerated structural changes most likely lead to welfare losses in rural areas.
