Abstract:
In this article, we propose a creation order reconstruction method of deleted files for the FAT32 file system with Windows operating systems. Creation order of files is established using a correlation between storage locations of the files and their directory entry locations. This method can be utilized to derive the creation-time bound of files recovered without the creation-time information. In this article, we first examine the file allocation behavior of Windows FAT32 file system. Next, based on the examined behavior, we propose a novel method that finds the creation order of deleted files after being recovered without the creation-time information. Due to complex behaviors of Windows FAT32 file system, the method may find multiple creation orders although the actual creation order is unique. In experiments with a commercial device, we confirm that the actual creation order of each recovered file belongs to one of the creation orders found by the method.

Abstract:
In this paper, a new method using a Secure Digital Card in debugging an embedded system based on Android and its implementation are introduced. In debugging an embedded system, downloading image files to a RAM on a printed circuit board generally uses a net interface module and a server based on Trivial File Transfer Protocol (TFTP). Here a new method using an SD card is introduced. Compared with downloading by a TFTP server, the method is simple and inexpensive. In the paper, an SD card and its controller are introduced first. Then the designed circuit is presented. Third, two operation modes of the SD card, card identification and data transferring are analyzed in detail. Fourth, the structure of the FAT32 file system and the process for downloading image files into a RAM on board are shown.

It has been observed that there has been a great interest in computing experiments which has been useful on shared nothing computers and commodity machines. We need multiple systems running in parallel working closely together towards the same goal. Frequently it has been experienced and observed that the distributed execution engine named MapReduce handles the primary input-output workload for such clusters. There are numerous distributed file systems around viz. NTFS,ReFS,FAT,FAT32 in windows and linux, we studied them and implemented a few distributed file systems. It has been studied that distributed file systems (DFS) work very well on many small files but some do not generate expected output on large files. We implemented benchmark testing algorithms in each distributed files systems for small and large files, and the analysis is been put forward in this paper. Even we came across the various implementation issues of various DFS, they have also been mentioned in this paper.

Abstract:
Fatty acid transport protein 2 (FATP2) is highly expressed in the liver, small intestine, and kidney, where it functions in both the transport of exogenous long-chain fatty acids and the activation of very-long-chain fatty acids. Here, using a murine model, we investigated the phenotypic impacts of deleting FATP2, followed by a transcriptomic analysis using unbiased RNA-Seq to identify concomitant changes in the liver transcriptome. WT and FATP2-null (Fatp2−/−) mice (5 weeks) were maintained on a standard chow diet for 6 weeks. The Fatp2−/− mice had reduced weight gain, lowered serum triglyceride, and increased serum cholesterol levels and attenuated dietary fatty acid absorption. Transcriptomic analysis of the liver revealed 258 differentially expressed genes in male Fatp2−/− mice and a total of 91 in female Fatp2−/− mice. These genes mapped to the following gene ontology categories: fatty acid degradation, peroxisome biogenesis, fatty acid synthesis, and retinol and arachidonic acid metabolism. Targeted RT-quantitative PCR verified the altered expression of selected genes. Of note, most of the genes with increased expression were known to be regulated by peroxisome proliferator–activated receptor α (PPARα), suggesting that FATP2 activity is linked to a PPARα-specific proximal ligand. Targeted metabolomic experiments in the Fatp2−/− liver revealed increases of total C16:0, C16:1, and C18:1 fatty acids; increases in lipoxin A4 and prostaglandin J2; and a decrease in 20-hydroxyeicosatetraenoic acid. We conclude that the expression of FATP2 in the liver broadly affects the metabolic landscape through PPARα, indicating that FATP2 provides an important role in liver lipid metabolism through its transport or activation activities.

Abstract:
Neuronal shape and position are critical to the formation and function of neuronal circuits. Although neurons develop axons and dendrites cell-autonomously in vitro, how extracellular cues in vivo direct neurite specification and placement remains poorly understood. The role of atypical cadherin Fat3 in amacrine cell development illustrates how the same extracellular cue can guide both dendrite formation and migration. In the mouse retina, amacrine cells have a bipolar morphology during their migration. Upon reaching the nascent inner plexiform layer (IPL), they elaborate one neurite into the IPL and retract the other. Loss of Fat3 leads amacrine cells to develop an extra dendrite outside the IPL as well as errors in migration. We found Fat3 protein is concentrated at the IPL throughout amacrine cell development, suggesting Fat3 detects a directional signal. Here we investigated the signaling pathways upstream and downstream of Fat3 that mediate its role in amacrine cell development. In Drosophila, Fat’s ligand is Dachsous. Fat and Dachsous binding is modulated by the kinase Four-jointed. Our analysis of mutant retinas determined that Fat3 and vertebrate Four-jointed genetically interacted. However knockout studies of vertebrate Dachsous homologues suggested they are not relevant Fat3 ligands in the retina. Instead, analysis of retinas missing retinal ganglion cells suggested Fat3-mediated homophilic adhesion between amacrine cells may be important. Sparse loss of Fat3 from amacrine cells also led to extraneous neurites, suggesting Fat3 acts cell autonomously. Ex vivo live imaging revealed both migration and neurite dynamics were less directed in Fat3 mutant amacrine cells. We hypothesized Fat3 acts to target asymmetric localization of cytoskeletal regulators to the leading neurite. To identify downstream Fat3 effectors, we performed a pulldown assay using the Fat3 intracellular domain. We identified several cytoskeletal regulators as candidate binding partners. We focused on the actin regulators Ena/VASP and demonstrated a direct interaction with the Fat3 intracellular domain. In the retina, Ena/VASP localized with Fat3 to the IPL, and loss of Fat3 changed Ena/VASP distribution. Furthermore, forcing uniform membrane recruitment of Ena/VASP in developing amacrine cells phenocopied loss of Fat3. Together these results suggest Fat3 polarizes the activity of cytoskeletal effectors to help direct amacrine cell migration and dendrite placement.

Detta examensarbete reder ut arbetsprinciperna för olika typer av hårddiskskrivskydd; hårdvaruskrivskydd, mjukvaruskrivskydd, hybridskrivskydd och bygelskrivskydd. Slutsatsen av utredningen är att endast hårdvaruskrivskydd Detta examensarbete reder ut arbetsprinciperna för olika typer av hårddiskskrivskydd; hårdvaruskrivskydd, mjukvaruskrivskydd, hybridskrivskydd och bygelskrivskydd. Slutsatsen av utredningen är att endast hårdvaruskrivskydd bedöms ha tillräckligt pålitliga skyddsprinciper, vilket motiveras av dess oberoende från både hårdvara och operativsystem.

Vidare undersöks hårdvaruskrivskyddet Image MASSter(TM) Drive Lock från Intelligent Computer Solutions (ICS). Några egentliga slutsatser gick inte dra av kretskonstruktionen, bortsett från att den är uppbyggd kring en FPGA (Xilinx Spartan-II, XC2S15) med tillhörande PROM (XC17S15APC).

En egenutvecklad idé till autenticieringsmetod för hårddiskkloner föreslås som ett tillägg till arbetet. Principen bygger på att komplettera hårddiskklonen med unik information om hårddisk såväl kloningsomständigheter, vilka sammanflätas genom XOR-operation av komponenternas hashsummor.Autenticieringsmetoden kan vid sjösättning möjligen öka rättsäkerheten för både utredarna och den som står misstänkt vid en brottsutredning.

Arbetet är till stora delar utfört vid och på uppdrag av Statens kriminaltekniska laboratorium (SKL) i Linköping.

Abstract:
Tissue elongation is an important requirement for proper tissue morphogenesis during animal development. The Drosophila egg chamber is an excellent model to study the molecular processes underlying tissue elongation. An egg chamber is composed of germline cells that are enveloped by a somatic follicle epithelium. While the egg chamber matures, the drastic increase of the egg chamber’s volume is accompanied by a shape change from round to oval. Egg chamber elongation coincides with a circumferential alignment of F-actin filaments, microtubules, and fibrils of the extracellular matrix (ECM). Additionally, egg chambers rotate around their future long axis. It has been proposed that this rotation aligns F-actin filaments and ECM fibrils. The circumferentially aligned F-actin and ECM fibrils form a molecular corset that promotes egg chamber elongation. The atypical cadherin Fat2 is required for egg chamber rotation, the circumferential alignment of F-actin, microtubules, and ECM fibrils and for egg chamber elongation. However, the molecular mechanisms by which Fat2 influences egg chamber elongation remain unknown. In my thesis I performed a structure-function analysis of Fat2. I generated a Fat2 version that lacks the intracellular region and a second version, which lacks both intracellular region and the transmembrane domain and tested their ability to compensate for Fat2 functions in fat2-/- mutant egg chambers. My results reveal that the intracellular region is required for the microtubule alignment, and for egg chamber rotation. In contrast, the intracellular region is not required for F-actin and ECM alignment, and for egg chamber elongation. Hence, my findings for the first time demonstrate that egg chamber rotation is not required for F-actin and ECM fibril alignment and that egg chamber elongation can occur independently from egg chamber rotation. My work uncouples some of the parallel processes that take place during oogenesis and changes the view on the mechanisms that drive tissue elongation in this important model system
Das Strecken von Geweben ist ein wichtiger Prozess bei der Gestaltbildung während der Entwicklung von Organismen. Die Eikammer von Drosophila ist ein hervorragendes Modellsystem, um die Gewebestreckung zu untersuchen. Eine Eikammer besteht aus Keimbahnzellen und einem einschichtigen Follikelepithel, das die Keimbahn umschließt. Während die Eikammer heranwächst durchläuft sie eine drastische Gestaltveränderung von rund nach oval. Zeitgleich zur Streckung der Eikammer weist das Follikelepithel parallel angeordnete F-actin−Filamente, Mikrotubuli und Fasern der extrazellulären Matrix (ECM) auf, welche die Eikammer ringsum umlaufen. Zudem rotieren die Eikammern um ihre zukünftige Längsachse. Bisher nahm man an, die Rotation würde für die Ausrichtung der F-actin−Filamente, Microtubuli und ECM-Fasern gebraucht werden. Die Anordnung der F-actin−Filamente und ECM-Fasern bilden dann ein molekulares Korsett, das die Gewebestreckung fördert. Das atypische Cadherin Fat2 wird für die Rotation der Eikammern, die umlaufende Anordnung der F-actin–Filamente, Microtubuli und ECM-Fasern sowie für die Streckung der Eikammern benötigt. Die Mechanismen, mit denen Fat2 die Gewebestreckung beeinflusst, sind allerdings unbekannt. In meinem Projekt führte ich eine Struktur-Funktions-Analyse von Fat2 durch. Ich generierte eine Version von Fat2 mit einer Deletion der kompletten intrazellulären Region und eine zweite, die weder die intrazelluläre Region noch die Transmembran-Domäne besitzt und testete, ob diese Versionen die Funktionen von Fat2 in fat2-/- mutanten Eikammern kompensieren können. Meine Ergebnisse zeigen, dass die intrazelluläre Region für die Anordnung der Mikrotubuli und für die Rotation der Eikammern gebraucht wird. Die intrazelluläre Region wird jedoch weder für die Anordnung von F-actin–Filamenten und den ECM-Fasern noch für die Streckung der Eikammer benötigt. Meine Erkenntnisse zeigen erstmalig, dass die Streckung der Eikammern ohne Rotation stattfinden kann. Meine Arbeit entkoppelt damit mehrere parallel stattfindende Prozesse während der Entwicklung der Eikammer und eröffnet einen neuen Einblick in die Mechanismen der Gewebestreckung in diesem wichtigen Modellsystem

Abstract:
Two trials in 1991 and another two in 1992 were carried out on the broodstock conditioning of Manila clams, Tapes philippinarum. The main objective was to manipulate the lipid and polyunsaturated fatty acid content of Manila clam eggs by maintaining broodstock in different dietary regimes. The second objective was to assess dried algae as alternative diets for conditioning broodstock. Clams were brought into the laboratory from the natural environment early in the year, before gametogenesis had started. Supplements of cultured live (Dunaliella tertiolecta, Skeletonema costatum, Tetraselmis suecica and Isochrysis galbana) and dried algae (T. suecica) diets were fed to the broodstock, usually in a range of mixed diets, at rations equivalent to 3% or 6% of the initial dry meat weight of the broodstock in dry weight of algae per day. The microalgae differed in their long-chain polyunsaturated fatty acid content (PUFA). Unfed control clams received only the organic material which remained in the sea water after sand filtration. The nutritional value of these diets in relation to gametogenesis, fecundity, quality of eggs, and viability and growth of larvae were assessed. Dry T. suecica was the same food value as live T. suecica but Manila clams produced more eggs if supplements of live algae were added. The requirement for conditioning Manila clams (32 mm shell length) to spawn with live or dry T. suecica+S. costatum was 500 to 700 "day-degrees" (D°). With dry T. suecica on its own or mixed with I. galbana, S. costatum and D. tertiolecta it was 500 to 600 Do (44 mm shell length). In one trial clams spawned in the tanks (equivalent to 462 Do) before the first attempt to spawn them was made. Successful spawning was dependent on the quantity and quality of the algal diet during gametogenesis. With a 6% food ration, clams fed dry T. suecica+S. costatum or dry T. suecica+l. galbana produced the highest number of eggs (an average of 3.2 and 4.5 million eggs per female, respectively). The average fecundity was 83% lower when the diet was reduced to a 3% food ration. The dry meat weight, condition index and fecundity of fed broodstock were significantly higher than for unfed animals. The quantity of lipid in the eggs, usually between 4 and 9 ng egg-1, was similar whatever the broodstock diet. However, levels of the essential polyunsaturated fatty acids 20: 5w3 and 22: 6w3 in the eggs were low if the broodstock diet was deficient in these PUFAs. Even though diet manipulation caused changes in the fatty acid composition of the eggs, growth and survival of Manila clam larvae was not reduced in a hatchery situation.

Tissue and organ shapes arise from molecular activities that control and execute cellular processes, such as oriented cell divisions, cell shape changes or cell rearrangements. Abstract:
Thermal fatigue may occur downstream from mixing tee junctions where hot and cold water are turbulently mixed. Depending on the momentum of the incoming flows, different characteristic eddies are formed which create a local hot spot on the inner surface of the pipe. The hot spot have an unstable discrete boundary that spatially oscillates and this creates an oscillating multidimensional thermal load that induces high cycle thermal fatigue. To predict the thermal fatigue lifetime of a pipe subject to this kind of thermal load, transient FEM simulations with an ideal hot spot in two and three dimensions were conducted. The crack lengths induced by the frequency of the oscillating boundary was analysed and it was found that the most harmful frequency for a pipe is in the nondimensional frequency of 0.01 to 1.0. Additionally, the simulation method was validated against data publications of FAT3D experiments. The simulations have been compared to the current proposed Japanese design code for mixing tee junctions. The study shows that this multidimensional effect induces larger crack lengths than the current design code. The reason of this was analysed.

Thermal fatigue phenomena are long term deterioration mechanisms which become more and more important as the life-time of nuclear power plant increases. Some incidents as the incident of Civaux I and some works in thermal fatigue have disproved current methodologies and usual criteria to predict propagation of thermal fatigue cracks in nuclear power plants. This paper presents the results of the thermal fatigue tests, Fat3D, which are conducted on 304L austenitic stainless steel pipes. This experiment has been designed to study the problem of closure effect and fatigue crack growth under thermal fatigue conditions on quasi-structure specimens. The importance of the initiation and the propagation phases on a notched specimen and the evolution of the stress intensity factor according to the propagation are investigated as well. The use of different non destructive techniques to detect and follow crack propagation is also assessed. In parallel, a numerical interpretation is developed based on a material characterisation and using finite element analyses with the French Cast3M code. This combined experimental and numerical study enables to assess improvements of classical methods to accurately predict the crack growth propagation under thermal loads and to understand the influence of the main parameters concerning crack propagation in such components.

Abstract:
This paper covers work carried out by the French Atomic Energy Commission (CEA) to investigate on mechanisms leading to cracking of piping as a result of thermal loading existing in flow mixing zones. The main purpose of this work is to analyse, with a new experiment and its numerical interpretation, and to understand the mechanism of propagation of cracks in such components. To address this issue, a new specimen has been developed on the basis of the Fat3D experiment. This thermal fatigue test consists in heating a 304L steel pre-cracked tube while cyclically injecting ambient water onto its inner surface. The tube is regularly removed from the furnace for a crack characterisation. Finally, the crack growth is evaluated from the crack length differences between two stops. In parallel, a finite element analysis is developed using the finite element Cast3M code. A pipe with a semi-elliptical crack on its inner surface is modelled. A cyclic thermal loading is imposed on the tube. This loading is in agreement with experimental data. The crack propagates through the thickness. A prediction of the velocity of the crack is finally assessed using a Paris’ law type criteria. Finally, this combined experimental and numerical work on 304L austenitic stainless steel pipes will enable to improve existing methods to accurately predict the crack growth under cyclic thermal loadings in austenitic stainless steel pipe at the design stage.
