This dissertation examines the recent emergence of popular recording artists who explicitly invoke the prophetic voices of their religious traditions in their music. In doing so, they situate their music within a pre-existing prophetic lineage. These musicians selfconsciously view their music as vehicle to spread a divine message, and they recognize that their lyrical voice can either become or echo a prophetic voice.

Visual arts education provides the knowledge, skills and attitudes necessary for students to become more responsive and responsible citizens who are aware of the interaction among the visual, aesthetic, economic and social aspects of their culture. A well-prepared visual arts educator is an essential contribution to the quality of student learning in this area of education. The purpose of this study was to suggest a series of guidelines and recommendations for the improvement of the art and art teacher education at Al-Fateh University in Al-Jamahiriya (Libya). Development of these guidelines and recommendations was based on the following steps: (1) Review and examination of developments and trends in Al-Jamahiriya's formal education system. These included secondary education, teacher preparation, and visual art education. (2) Examination of the present art education program at Al-Fateh University and its role in preparing qualified visual art teachers for Al-Jamahiriya's secondary schools. (3) Comparative study of selected factors pertaining to art education teacher preparation at Al-Fateh University and four exemplary American universities. (4) Library research regarding current thought and practices in the areas of visual art education and teacher preparation. A questionnaire was also developed and sent to the art education department at Al-Fateh University to help obtain descriptive data concerning the teacher education program. A panel of experts at the University of Arizona had identified four exemplary American university art education programs. These programs were examined as part of this study. Recommendations are made regarding selected improvements in the art and art teacher preparation programs at Al-Fateh University.

Abstract:
Higher education institutions (HEIs) around the world and in the developing countries in particular e.g. Libyan HEIs are facing challenges that increase the pressure on them. Some of these challenges are related to the remarkable changes in population growth and fast changes in the development of knowledge and technology. Also, providing adequate resources, maintaining quality, raising funding and strengthening the curriculum are other challenges that need to be faced by those institutions. This research is aimed to investigate issues enabling and affecting the quality of services provided by Libyan public universities using principles of Total Quality Management (TQM) philosophy as a framework: Al-Fateh University (AFU) as a case study. The research is qualitative in nature, employing a case study approach and using indepth semi-structured interviews (from different levels including senior leaders, faculty members, support staff, students, and main employers) as the main data collection tool within the two selected embedded case studies. Those embedded cases are Electric and Electronic Department (EED)-Faculty of Engineering-AFU and Social Service Department (SSD)-Literature Faculty-AFU. Documents are used in addition to interviews in order to fully understand issues enabling and affecting the quality of services provided by the two embedded case studies. Contribution to knowledge is evident by the study, which represents the first attempt to empirically investigate issues enabling and affecting the quality of services provided by Libyan public universities: AFU as a case study through two selected embedded cases EED and SSD. This research provides specific original findings which include the use of Arabic and English language in the same lecture, the concept of students' administration and its consequences, the speciality of leaders biases the understanding of the needs of subordinate staff, suspension of the students' performance regulations affected the quality of education programmes, and a unique situation was found to be the senior and junior staff programme that attempted to improve teaching through knowledge transfer. This research has reduced the gap in knowledge in Libyan HE context in specific and in Arabic HE context in general. Other implications for HEIs are also provided by this research.

This thesis aims to analyze Islamic rhetoric of the Palestine Liberation Organization (PLO) and its underlying objectives and reasons by examining policies, speeches and declarations of the PLO, Fateh and Yasser Arafat. It is widely accepted that the PLO functiones according to secular rules
perceives Israel, Palestine and the Israeli-Palestinian question on Palestinian nationalist and Third worldist grounds. However in spite of its secular functioning and goals, the PLO since its inception has referred to Islamic rhetoric on natinal struggle of Palestine&rsquo
s liberation. Henceforth it is argued that the PLO has chosen a pragmatist and instrumentalist language in referring to Islamic symbols and discourses. Islamic rhetoric of the PLO has aimed primarily to mobilize the Palestinians for national goals and to legitimize the PLO against rising power of political Islam headed by Hamas. Furthermore the thesis also demonstrates that the PLO&rsquo
s Islamic rhetoric was affected by Palestinian society&rsquo
s shift along Islamic lines and Islamic component of Palestinian nationalism and culture. The PLO seemed to intensify its resort to Islamic rhetoric with every crisis that decreased the PLO&rsquo
s authority and power. It is concluded that content of the PLO&rsquo
s pragmatic Islamic rhetoric was never related with making Islam as normative and legal basis of Palestinian society, rather it was related with mobilization and legitimating.

Cette faiblesse peut être attribuée à plusieurs facteurs dont : la prestation du professeur, les étudiants eux-mêmes, le contenu du programme, les méthodes d'enseignement ou à l'ensemble de ces facteurs. Le but de cette étude est donc d'évaluer le cours d'anglais de spécialité au sein de la faculté d'agriculture pour identifier les raisons qui causent l'inefficacité de cet enseignement. Ce processus d'évaluation couvre : la prestation des professeurs, les étudiants, le contenu du programme et les méthodes d'enseignement appliquées. The @Libyan University students at the faculties of agriculture at Al-Fateh University, who have passed a two years ESP course, remain unable to use English effectively in their academic study. This shortcoming may be attributed to more than one factor, including : teacher performance, the learners themselves, the designated syllabus, the methods of teaching or to all these factors. Therefore, the purpose of this study is to evaluate the ESP course (for the faculty of agriculture) by investigating the reasons behind the ineffectiveness of the ESP course. This evaluation process covers : the teachers performance, the learners, the designated syllabus and methods of teaching applied. This study is conducted on a sample of students and a sample of the English language teachers at the faculties of agriculture. The designated syllabus and the methodology applied in teaching are also investigated by means of class observations, and questionnaires addressing both teachers and students. The question of this thesis is whether the individual can resolve the problem of culture crisis in her own case. Culture crisis is a historical moment in which our culture leads us to expect a world drastically different from the one in which we find ourselves. This thesis will focus on the experience of Generation Y in the fall-out of the 2008 Recession. It will be argued that we need a Wittgensteinian view of language in order to account for the phenomenon of culture crisis. It will be suggested that our individual has to be a Nietzschean individual in order to resolve the problem of culture crisis in her own case. Potential incompatibilities between a Wittgensteinian view of language and the Nietzschean individual will be considered and rejected. It will be concluded that in order to resolve the problem of culture crisis in her own case the individual must change the way she lives.

War Memorials are often forgotten statues, right in the center of town, but still out of our sight. They do not tell you enough to understand them, neither are you interested in putting effort into getting to know and learn from them. This paper investigates how the web could be used to create a war memorial that is more alive, captivating and empathy awakening. There has been some virtual war memorials getting constructed since the web started to bloom, but we could not find a single one that had actually fully explored the potential of the web, and what it might have to offer for the creation of war memorials. Researching the web as a media, experience design, and information visualization, we find possibilities to mourn, commemorate and heal on virtual ground. Inspiring reflection and contemplation are another two purposes of The Common Fate Memorial. War memorial studies give us the background information needed, and ceremony mechanics are studied for further inspiration. Our findings are implemented in flash prototypes, which are user tested and evaluated.

Abstract:
“Foreknowledge, Fate and Freedom” is concerned with diagnosing and debunking a pervasive and prevalent folk intuition: that a foreknown future would be problematically, and freedom-hinderingly, fixed. In it, I discuss foreknowledge in and of itself, but also as a lens through which we can examine other intuitions and concepts: the apparent asymmetry of future and past; worries about fate and free will; notions of coincidence and likelihood; assumptions about God, time travel and ourselves. This thesis provides the first philosophical map of a region of conceptual space visited often by the folk and popular culture, and as a result ties together a host of disparate threads in the literature. I make three central claims: 1. The folk intuition is wrong in rejecting foreknowledge wholesale on the basis that it entails a problematically fixed future, and thereby undermines our freedom. 2. Foreknowledge gives rise to new problems, and sheds new light on old ones, but none of these are insurmountable. 3. The same paradoxes thought to plague backwards time travel can arise in foreknowledge cases, and can be defused in the same way. I conclude that foreknowledge is puzzling, but possible: it neither inevitably entails fatalism nor precludes free will. While its consequences may be strange, they are not sufficient to vindicate the folk intuition.

Die vorliegende Arbeit befasst sich mit Aspekten der frühen Entwicklung der Phoronida („Hufeisenwürmer“). An drei Arten wird der Furchungsprozess untersucht (Phoronis pallida, Phoronis muelleri, Phoronis vancouverensis). Dies erfolgt sowohl mithilfe der 4D-Mikroskopie als auch anhand von immunocytochemischen Markierungen der Mitosespindeln und konfokaler Laser-Scanning-Mikroskopie. Verschiedene morphologische Merkmale des Furchungsprozesses werden quantitativ erfasst und innerhalb sowie zwischen den Arten verglichen. Die Ergebnisse zeigen eine weitgehend übereinstimmende Furchung bei P. pallida und P. muelleri Embryonen: Ab dem dritten Zellzyklus teilen sich die Blastomeren meist schräg – und alternierend dextral und sinistral – zur animal-vegetativ Achse. Dieses Muster zeigt überraschende Übereinstimmungen mit dem Muster der Spiralfurchung. Dies kann als morphologische Unterstützung molekular-phylogenetischer Befunde einer Stellung der Phoronida innerhalb der Spiralia/Lophotrochozoa interpretiert werden. Die Furchung bei P. vancouverensis unterscheidet sich von der Furchung der anderen beiden Arten; sie weist jedoch auch Unterschiede zu einer Radiärfurchung auf. Generell zeigt die Furchung aller drei Arten einen gewissen Grad an Variabilität. Anhand von in-vivo Einzelzellmarkierungen untersucht die Studie darüber hinaus das Schicksal der Blastomeren früher P. pallida Embryonen bis zu späten Gastrulationsstadien. Diese Analysen zeigen, dass die ersten beiden Furchungsteilungen durch die spätere Achse Blastoporus-Apikalplatte, jedoch in keinem konstanten Orientierungsverhältnis zur Ebene der Bilateralsymmetrie der Gastrula verlaufen. Dies unterscheidet sich von der Situation, wie sie von spiralfurchenden Tieren bekannt ist. Die Unterschiede und die beobachtete Variabilität des Furchungsprozesses werden im Licht unterschiedlicher Mechanismen der Spezifizierung von Zellschicksalen und Körperachsen bei verschiedenen Taxa der Spiralia und den Phoronida diskutiert.
This study addresses aspects of the early development of Phoronida (“horseshoe worms”). The cleavage process is analyzed for three species (Phoronis pallida, Phoronis muelleri, Phoronis vancouverensis). These investigations are performed using 4D-microscopy as well as immunocytochemical stainings of the mitotic spindle apparatuses in combination with confocal laser-scanning microscopy. Different morphological features of the cleavage process are quantified and compared within as well as between the species. The results reveal a highly consistent cleavage of P. pallida and P. muelleri embryos: from the third cell cycle onward, the blastomeres divide mostly obliquely – and alternatingly dextral and sinistral – with respect to the animal-vegetal axis. This cleavage pattern shows surprising correspondences to the pattern of spiral cleavage. The finding can be interpreted as morphological support for recent molecule-based phylogenies, which indicate a position of Phoronida within the Spiralia/Lophotrochozoa clade. The cleavage of P. vancouverensis differs from the cleavage in the other two species; however, it also shows differences to a radial cleavage pattern. In all three species, the cleavage process also involves some degree of variability. Furthermore, the study traces the cell fates of early P. pallida embryos up to the state of late gastrulation, by the use of fluorescent in-vivo single cell markings. These analyses reveal that the first two cleavage divisions both pass through the later axis blastopore-apical plate of the gastrula, yet they do not pass in a constant relationship with respect to the later plane of bilateral symmetry. This differs from the situation known from spiral cleaving animals. The differences and the encountered variability of the cleavage process are discussed with respect to different mechanisms of the specification of cell fates and body axes in different taxa of the Spiralia and the Phoronida.

O presente trabalho tenta entender o roteiro do filme Contra a Parede (2004), de Fatih Akin, como texto dramático, buscando não apenas contribuir para os estudos do roteiro cinematográfico na área de Letras, mas também na área de Cinema. São enfoques distintos em suas especificidades, mas complementares. Entendido como texto dramático, o roteiro em questão apresenta características típicas da tragédia. Sendo assim, analisamos os personagens principais enquanto personagens dramáticos, o coro e a estrutura desse roteiro, a partir de cenas selecionadas. Interessa-nos em especial a questão da redenção dos protagonistas Cahit e Sibel. Para nos ajudar no substrato teórico, recorremos aos estudos de Aristóteles, Nietzsche, Bordwell, Jaspers e Romilly.
This paper aims to understand the script of the film Head-On (2004), by Fatih Akin, as a dramatic text, seeking not only to contribute to screenplay studies in the field of writing, but also in the field of cinema, for these are different yet complementary approaches. Understood as a dramatic text, the script in question presents features that are typical of the tragedy. Thus, we analyze the main characters as dramatic characters, the chorus and the structure of the script, from selected scenes. We are particularly interested in the matter of redemption of the main characters Cahit and Sibel. To help us in theoretical basis, we turn to the study of Aristotle, Nietzsche, Bordwell, Jaspers and Romilly.

Abstract

This essay is based on an investigation carried out with the help of the Corpus of Contemporary American English (COCA). Taking the system of transitivity as its theoretical base and using spoken and written discourse as its primary source, this study aspired to find out what kinds of actions Americans perceive Fate to perform, for example physical, mental or verbal, in order to control what happens to people. It also aimed to reveal what actions people are said to perform in their attempts to control Fate. Do Americans deem Fate capable of, for instance, “deciding”, “talking” or “conspiring” and do they say that people, for example, “challenge”, “defy” or “defeat” Fate? Furthermore, a comparison was made in terms of the actions performed by Fate and people between the different domains of discourse represented in the corpus: spoken, fiction, magazine, newspaper and academic.

Among other things, this investigation shows that in American discourse both Fate and people are perceived to resort to physical strategies rather than mental or verbal ones in their endeavor to control one another.

Nonylphenol is a widely used organic compound which has been reported to have potential risk to aquatic environment. According to the result of recent studies, it has been detected in many lakes in Stockholm, Sweden, which raised great concern. In this thesis, a dynamic fate model was adopted and modified from literature in order to study the distribution and concentration of nonylphenol in small lakes, guide the field sampling and provide information for corresponding decision making. Two lakes in Stockholm, Lake Trekanten and Lake Drevviken, were selected as case studies. Another model was included for comparison purpose. Based on the model result, the most important nonylphenol removal process in both lakes was the transformation in water. A sensitivity analysis showed that the model results were most sensitive to the process of nonylphenol water inflow. In terms of sediment concentration of nonylphenol, satisfactory agreements were obtained from the comparison between model results and field data. However, problems, such as the simultaneous handling of nonylphenol and nonylphenol ethoxylates, may cause uncertainties on the model performance. The result of the analysis about scenario load change and the seasonal variation showed that the sediment nonylphenol content is more stable to the seasonal change compare to nonylphenol water content, but the response times to load change of nonylphenol content in these two compartments are quite close and somewhat lower than the water residence time.

Abstract:
Synthetic musks are incorporated extensively in personal care products to improve their scent, increase their fragrance stability, and prolong their shelf-life. As a consequence, these persistent musks are being released at a considerable rate by wastewater treatment plants and are frequently detected in surface water, bottom sediment, air, and aquatic biota near urban areas. In addition to their hydrophobicity, two synthetic musks, Galaxolide (HHCB) and Tonalide (AHTN), were reported to cause endocrine disruption in fish species. Although most of the toxic effects in past studies were observed at high doses, HHCB and AHTN were shown to bioaccumulate very differently depending on the aquatic species tested. As bioaccumulation and fate of contaminants are important considerations when regulating persistent chemicals, an improved understanding of the bioaccumulation potential of these chemicals is needed. In this thesis, an assessment of the presence, bioconcentration and fate of HHCB and AHTH was provided in an area exposed to the effluent of the Gold Bar wastewater treatment plant in the North Saskatchewan River (Edmonton). HHCB and AHTN were quantified in fathead minnows (Pimephales promelas) exposed downstream of the effluent, with an adapted method for analysis of musks in fish, using PAHs as recovery standards. Method development and recoveries are summarized. Highest bioconcentration factors were 24,500 and 22,300 for HHCB and AHTN respectively, and were observed 1 km downstream of the outfall. Musks were found in most fathead minnows exposed at reference sites upstream as well as at the furthest site, 9.9 km from the outfall. Musk concentrations in water at these sites were used in the assessment of the fate of HHCB and AHTN by fugacity modeling with QWASI software. In order to assess fate, a contaminated portion of the North Saskatchewan River was divided in a series of compartments that were each treated as connected individual water bodies. HHCB and AHTN losses were mostly due to water advection and sedimentation fluxes. Model fit was assessed by comparing predicted to measured data.

Abstract:
The demonstration that simply by introducing four selected proteins it is possible to change mammalian somatic cells from one phenotype to another is providing important new opportunities in medicine. However, this approach has several limitations. In contrast to other methods of changing cell fate such as cloning and cell fusion it is very slow, very inefficient and it is necessary to have identified the key transcription factors. In both cloning and fusion the nucleus is exposed to the cytoplasm of the recipient cell and it is this that changes nuclear function. With the hope of creating the same effect, extracts from cells of the desired cell type have been introduced into candidate cells. This induced some changes in cell function, but did not change cells from one phenotype to another. The aim of this project was to improve methods for the introduction of extract when human skin cells were exposed to extracts of mouse pluripotent stem cells. During early studies, it was noticed that external materials could enter cells spontaneously at a specific stage of cell cycle, metaphase. When cells in metaphase were exposed to cell extracts pluripotent cell-like colonies were formed. These cells expressed markers of pluripotency such as SSEA4 and Tra-1-60 and could form embryoid bodies that would further differentiate to all three germ layers. Unfortunately this protocol was found to be unrepeatable. In subsequent studies the temperature of exposure to extract was raised from 37oC to 40oC with the aim of promoting the fluidity of the cell membrane and so enhancing uptake of extract. A new treatment regime was introduced to increase the proportion of cells exposed to extract while in metaphase. A new protease inhibitor was introduced in order to promote persistence of the extract within the cells. Also, a new TLR3 agonist was introduced to enhance chromosome modification. After these modifications were made pluripotent cell-like colonies formed within 14 days of treatment with extract and these colonies were positive for alkaline phosphatase live staining. Further research is required to complete the development of a routine procedure.

The present thesis considers the allusive and narrative function of fate and its associated concepts of providence, free will, necessity, and natural law in Paradise Lost. It argues that the narrative function of these concepts is shaped by Milton's allusions to classical epic, and assesses their impact on the Christian theology of the poem. It identifies unnoted allusions to well-known epic models (Homer, Vergil, Lucan), and examines how Lucretius' account of natural laws and post-Vergilian representations of epic aftermath influence Milton's own depiction of transgression and its aftermath in Paradise Lost. Chapter 1 considers Satan and other fallen angels' definition of fate as a materialist alternative for the personal rule of the Father. It traces several allusions to fate in cosmological and ethical settings, in Lucretius, Vergil, Lucan, and Statius, and analyses how these allusions interact with the Hesiodic mythical material in the opening books of Milton's epic. Chapter 2 focuses on a pattern of previously unnoted allusions to Lucretius' De Rerum Natura in the narrative of the Fall, culminating in Book 9. It argues that in his temptation of Eve, Milton's Satan subverts Lucretian teachings about the boundaries governing the physical universe as he persuades Eve to transgress her natural state in Eden. Chapter 3 discusses the appearance of the Father in an allusive epic council scene in Book 3. In the dialogue between Father and Son, I suggest, Milton evokes negotiations between the Homeric and Vergilian deities, depicting his God as surpassing his pagan epic counterparts who can only delay the fate of mortals, but not change them. Chapter 4 suggests that Milton's depiction of the aftermath of the Fall is indebted to post-Vergilian epic narratives of 'aftermath'. The final Books of Paradise Lost and the portrayal of Adam and Eve's moral freedom as they leave paradise, with providence their guide, should be read, I posit, against the backdrop of scenes and imagery from Lucan's Bellum Civile and Statius' Thebaid.

Abstract:
ESC/iPSC-derive somatic cells may be ideal for treating disorders caused by cellular deficiency or dysfunction. To form a lineage-specific cell population, ESCs/iPSCs undergo a multi-step process that recapitulates embryonic development. ESC/iPSC differentiation protocols are hampered by the limitation of our understanding in development. Zebrafish embryos are fertilized and developed externally, a feature facilitates the observation and manipulation of embryonic development. To explore the zebrafish as a system to study cell lineage determination, in this thesis, I 1) identified an ortholog of the key pluripotency regulator Nanog in zebrafish and examined its role in early cell fate determination; 2) developed a high-throughput image-based chemical screening system in zebrafish blastomere cell culture that is very similar to, but much faster than, ESC/iPSC differentiation screens. Specifically, in an effort to examine the role of Nanog in vivo, I identified a zebrafish Nanog ortholog, and found that its knockdown impaired endoderm formation. Genome-wide transcription analysis revealed that nanog-like morphants fail to develop the extra-embryonic yolk syncytial layer (YSL), which produces Nodal required for endoderm induction. I examined the genes that were regulated by Nanog-like, and identified the homeobox gene mxtx2, which is both necessary and sufficient for YSL induction. Chromatin immunoprecipitation assays and genetic studies indicated that Nanog-like directly activates mxtx2, which in turn specifies the YSL lineage by directly activating YSL genes. The study identifies a Nanog-like-Mxtx2-Nodal pathway and establishes a role for Nanog-like in regulating the formation of the extra-embryonic tissue required for endoderm induction. In the second part of the thesis, I developed a system that allows high-throughput image-based chemical screening using cultured zebrafish blastomere cells. To demonstrate its potential, this system is utilized to study skeletal muscle development. I screened 2,400 chemicals, finding 11 chemicals that block mature muscle cell differentiation and 17 chemicals that block skeletal muscle progenitor formation. The subsequent studies of these hits illustrate an RTK-PI3K-mTOR- GSK3 signaling cascade that is critical for skeletal muscle development. Preliminary data in mouse Skeletal Muscle Precursors (SMPs) suggest the pathway is conserved in murine adult muscle stem cells. This system, which can be modified for any cell lineage, promises to enhance our understanding of fundamental biology and to identify chemicals for cell-based therapies for many diseases.

Embryonic stem (ES) cells have a vast therapeutic potential given their pluripotency, or the ability to differentiate into tissues from all three germ layers. One of the ultimate goals of regenerative medicine is to isolate pluripotent stem cells from patients. Nuclear reprogramming offers the possibility of creating patient-specific cell lines, thus abrogating the need for immunosuppressants following cell transplantation therapy. It was recently reported that the forced expression of four transcription factors, Oct4, Sox2, c-Myc and Klf4 can induce a pluripotent state in somatic cells, without the need for embryo destruction. The work presented here aims to characterize reprogramming using defined factors and provide insight into the mechanisms governing this process. It also seeks to identify transient cues to induce reprogramming in somatic cells, alleviating the need for virally transduced transcription factors that hinder its eventual clinical use.

The thesis considers the eternal problem of fate and free-will as it applies in the Mahabharata, the longest epic in any literature. In the Indian tradition the problem is explored in terms of whether human beings have any control over their own destiny and the world around them; or whether they are under the control of powers and forces beyond them. Chapter 1 examines the Epic doctrine of karma, which in theory represents the ideal compromise solution for it accepts the importance of both determinism and free-will in the lot of the individual, viewed as an entity transmigrating through time. The chapter finds that it is not a solution which finds much favour with the pravrtti or 'this worldly' side of Hinduism, which predominates in the Mahabharata. From their standpoint, karma can all too easily seem just another term for the workings of inscrutable fate. Chapter 2 examines the Mahabharata's views on predestination, the idea that the destiny of the individual is controlled from the beyond by a god or gods. After establishing the structure and nature of the Epic cosmos, the chapter considers the part played by gods and goddesses in the affairs of mortals and finds that three great Gods alone, Brahma, Civa, and Visnu, can be considered to play a predetermining role in the triple-world, for they must for ever try to make good the deficiencies in their own creation. However, the degree of their predetermining intervention differs according to how they are conceived. In the theological sections the great Gods are conceived of as indeterminate essences that predetermine all actions; in the mythology and the legends, they are conceived of as thoroughly anthropomorphic beings who predetermine only the essential events in their desire to preserve the order of the triple-world. The freedom of mortals is thus restricted, instead of negated. Finally, the chapter examines the position of such personalised abstractions as the Placer, the Ordainer, and the Ruler, which play a very prominent part in the proceedings of the Mahabharata. Whether, in any particular context, they are considered to be an independent force, an epithet of a great God, or impersonal fate thinly veiled, they invariably play a much more thorough predetermining role in the affairs of the triple-world. Chapter 3 considers the Mahabharata's views on how the destiny of the individual is controlled from the beyond by the impersonal forces of fate and Time. After examining the difficulties of clearly distinguishing between personal and impersonal forces of control, the chapter considers the range of circumstances in which impersonal fate is cited as the causative force. It also emerges that fate is conceived in quite different ways. In some contexts it is inscrutable and purposeless, and in other contexts it assumes a moral dimension. Equally, in some contexts it is considered to be all-powerful and ineluctable, predetermining all events and actions; but elsewhere fate is a force that can be overcome or made favourable. Chapter 4 considers the Mahabharata 's views pertaining to human free-will. The chapter examines; various 'compromise' answers offered, in which outside forces of control and individual freedom all play a part; the considerable importance attached to human effort and exertion; the means by which individuals can control their own destiny (sacrifice, tapas, knowledge and devotion); and the problem of individual responsibility. Chapter 5 analyses the problem of fate and free-will in the most famous single component of the Epic, the Bhagavadgita. The chapter argues that efforts to find the Gita's answer to the problem are misguided for the Gita offers various answers. The chapter does suggest what was the the favoured view of the Gita's author, but argues that he effectively undermined this solution out of a desire to build up the position of his personal God, a vital component in the Gita's answer to the contemporary crisis in the orthodox tradition. The conclusion points to the great variety of solutions the Mahabharata offers to this eternal problem, but emphasises that despite the fatalistic image of Hinduism, the Mahabharata - for various important reasons - has as much. if not more, to say about the efficacy of human action and free-will.

The fungal disease Fusarium head blight affects cereal grains and can produce mycotoxins, like the water-soluble deoxynivalenol (DON). Wheat wet milling process begins with ground endosperm obtained by dry milling and ends with the separation of starch from gluten. Research was conducted on hard red spring wheat and durum wheat samples naturally contaminated with DON. The fate of DON in wheat dry milled fractionations (farina/semolina, shorts, and bran) during wet milling was investigated. Three wet milling processes were evaluated. DON levels were assessed by GC-ECD. Results showed that DON was present in all dry milled fractions. DON concentration in farina and semolina exceeded the safety threshold for human consumption. After wet milling farina and semolina, nearly all the DON was found in the water-soluble fraction, regardless the wet milling process. A negligible level of DON was found in the gluten extracted from HRSW with Martin wet milling process.

Muscle homeostasis and regeneration are complex cellular processes orchestrated by muscle stem cells and their interaction with their stem cell microenvironment. The fate of a muscle stem cell is influenced by different conditions such as muscle injury, cold stress, or disease. During extensive muscle repair and in the context of muscular dystrophy, we identified the critical function of the Epidermal Growth Factor Receptor (EGFR) in establishing cell polarity and in turn the efficient formation of myogenic progeny able to repair muscle. Using a novel drug screen, we identified the p53 protein to regulate muscle stem cell fate decision to repress the formation of brown adipose tissue as a means to regulate whole-body metabolism. To increase the impact of our research we also optimized protocols evaluating mouse satellite cell transplantation to delineate stem cell hierarchy and developed a new paradigm to model human muscle stem cell fate to better translate our findings into the clinical arena. These findings reveal the tunable nature of stem cell fate decisions and highlight the development of research tools to accelerate the translation of research findings to improve human health.

The dynamic (ac) Stark effect refers to the energy shifting of electronic states induced by an oscillating electric field. Conveniently, the magnitude of the ac Stark shift scales with the square of the electric field amplitude, i.e. with light intensity. Using this fundamental effect to reshape molecular potentials, and steer the course of chemical reactions, is known as dynamic Stark control. The aim of this study was to investigate the dynamic Stark effect on the photodissociation of molecular oxygen (O2) in the Schumann-Runge continuum, SRC (130–175 nm). Absorption in the SRC leads to dissociation via the so-called B state, yielding O(1D) + O(3P), or the J state, forming O(3P) + O(3P). Both of these dissociative excited states may be well-described in terms of mixed valence and Rydberg state character, in which each of the two states are strongly coupled to a Rydberg state of similar symmetry. Due to the mixed character of the B and J states, simulations predict that dynamic Stark shifting of the coupled Rydberg states leads to a dramatic change in dissociation channel branching ratio, as well as a red-shift of the absorption spectrum. This study aimed at experimentally testing this theoretical prediction. A 400-nm femtosecond laser pulse was employed as a combined pump and control field, simultaneously inducing a three-photon transition into the SRC and ac Stark shifting the potentials. A detection scheme to detect the changes in absorption of the B channel with pump pulse intensity was devised and implemented. The chosen detection scheme, in which emission at 762 nm from the O2(b−X) transition is measured, in principle monitors O(1D) from the B channel via an energy transfer reaction. The experimental results overall show consistency between simulations and experiment. The measured 762-nm emission exhibited a pump pulse intensity-dependence that likely reflects the dynamic Stark reshaping of the excited state potentials. However, saturation is clearly present in the data, complicating data interpretation. Furthermore, deviations between experiment and simulations are large at high pulse intensities, indicating that O(1D) is additionally generated by absorption into higher excited states. Finally, structured features that deviate from the simulations at low pulse intensities may possibly be assigned to vibrational resonances to high-lying Rydberg states by four-photon absorption.

Tillämpningen av denna fundamentala effekt i syfte att omforma molekylära potentialer, och därmed styra kemiska reaktioner, kallas för dynamisk Starkkontroll. I den valda metoden detekteras emission vid 762 nm från O2(b − X)-övergången, vilket i sin tur ger en mätning av O(1D) som genereras från B- kanalen via en energiöverföringsreaktion. De experimentella resultaten stämmer relativt väl överens med simuleringarna. Den uppmätta emissionen vid 762 nm uppvisar ett intensitetsberoende som i stora drag reflekterar ac Stark- skiftningen av potentialerna. Utöver detta finns dock ett stort bidrag från mättnad, vilket försvårar tolkningen av datan. Vi-dare avviker den experimentella datan betydligt vid höga pulsintensiteter, vilket sannolikt tyder på att O(1D) även genereras genom absorption till högre exciterade tillstånd. Slutligen ob-serveras mindre, men tydliga avvikelser vid låga pulsintensiteter. Dessa kan möjligen tillordnas vibrationsresonanser med högre Rydbergstillstånd genom fyrfoton-absorption. ​

Le complexe de liaison de la coiffe des ARN (CBC) joue un rôle essentiel dans leur maturation et déclenche une variété de réactions biochimiques, via son interaction avec différents partenaires. Deux complexes, CBC-ARS2-PHAX (CBCAP), et CBC-ARS2-ZC3H18-NEXT (CBCN), ont récemment été montré comme important pour cibler les ARN vers l'export (CBCAP) ou la dégradation (CBCN). Cependant, les mécanismes par lesquels la sélection se fait pour l'une voie ou l'autre reste mystérieuse. Ainsi, une question majeure qui reste à résoudre est de savoir quand et comment ces complexes sont recrutés sur les ARN. Dans ce travail, j'ai utilisé la procédure du iCLIP (Cross-Linking and Immuno-Precipitation), afin d'identifier les cibles de ces complexes sur l'ensemble du transcriptome humain. J'ai réalisé un iCLIP sur cinq composants de CBCAP et CBCN, et j'ai comparé les résultats à ceux obtenus avec RBM7, un composant de NEXT précédemment étudié par iCLIP. Mes résultats indiquent que: (i) CBP20, ARS2, PHAX et ZC3H18 se lient près de la coiffe des ARN, tandis que RBM7 et MTR4 se lient partout; (ii) CBP20, ARS2, PHAX et ZC3H18 s'associent à un large ensemble d'ARN transcrits par l'ARN polymérase II et montrent une faible sélectivité; (iii) la liaison de ces protéines varie avec l'état de maturation des ARN, avec le CBC enrichi sur les ARN matures, tout comme ARS2/PHAX/ZC3H18 et MTR4 (bien que dans une moindre mesure), tandis que RBM7 est préférentiellement lié sur les pre-mRNAs non épissés; (iv) une liaison différentielle de RBM7 et MTR4 sur les ARN, avec RBM7 enrichi sur les introns et les PROMPTs, et MTR4 plus présent sur les ARN mature. Bien que des expériences additionnelles soient requises, nous proposons que le CBCAP et le CBCN se lient à un même ensemble d'ARN, ce qui indique à la fois une compétition entre ZC3H18 et PHAX pour la liaison à ces ARN, et l'absence de voies de routage bien déterminées qui ciblerait les ARN vers l'une ou l'autre de ces protéines. Le devenir des ARN pourrait ainsi être déterminé par d'autres caractéristiques des ARN, ou encore par des protéines additionnelles. Ces facteurs pourraient s'allier aux protéines liées à la coiffe afin de favoriser la formation du CBCAP ou du CBCN. Dans le but d’identifier des facteurs additionnels, j'ai réalisé un screen d'interaction par spectrométrie de masse après purification de ARS2 ou CBP80. Ceci a été fait dans des conditions natives ou après un cross-link des complexes à la formaldéhyde, afin de stabiliser les interactions transitoires. Ceci a permis d'identifier de nouveaux partenaires de ARS2 et de CBP80, dont la majorité sont impliqués dans l'épissage des ARN. Des expériences additionnelles seront nécessaires pour valider ces interactions
The cap-binding complex (CBC) plays a pivotal role in post-transcriptional processing events and orchestrates a variety of metabolic pathways, through association with different interaction partners. Two CBC sub-complexes, the CBC-ARS2-PHAX (CBCAP) and the CBC-nuclear exosome targeting (NEXT) complex (CBCN), were recently shown to target capped RNA either toward export or degradation, but the mechanisms by which they can discriminate between different RNA families and route them toward different metabolic pathways still remain unclear. A major question to be answered is how and when the different CBC subcomplexes are recruited to the RNP. Here, we used an individual nucleotide-resolution UV cross-linking and immunoprecipitation (iCLIP) approach to identify the transcriptome-wide targets for 5 different components of the CBCAP and CBCN complexes, and compared results to the previously analysed NEXT-component RBM7. We report that: (i) CBP20, ARS2, PHAX and ZC3H18 bind close to the cap, while RBM7 and MTR4 bind throughout the mRNA body; (ii) CBP20, ARS2, PHAX and ZC3H18 associate with a broad set of RNA polymerase II (PolII)-derived RNAs and have only mild species preferences; (iii) binding varies with the RNA maturation stage, with the CBC being highly enriched on mature mRNA, ARS2/PHAX/ZC3H18/MTR4 less so, and RMB7 preferentially bound to pre-mRNAs; (iv) MTR4 and RBM7 show different specificities, with RBM7 being highly enriched on introns and promoter upstream transcripts (PROMPTs), while MTR4 is additionally present on mature RNAs. Although more experimental work is needed to fully support our model, we propose that CBCAP and CBCN bind overlapping sets of RNAs, indicating a competition between the proteins ZC3H18 and PHAX, and the lack of a strict RNA sorting mechanism. RNA fate may therefore be determined by additional RNA features and/or by other RNA-binding proteins, which may synergize with the cap and drive the formation of one specific CBC subcomplex instead of another. In an attempt to identify yet unknown factors that may interact with cap-bound CBCAP and CBCN, we performed a protein interaction screen leveraging affinity capture-mass spectrometry (ACMS), using ARS2 and CBP80 as bait proteins. As a complementary approach, we also employed a formaldehyde-based chemical cross-linking strategy, aimed at stabilizing weak/transient interactions. Although we failed to detect any transient interactions involving the CBC, we identified several potential CBC80 and ARS2 interactors, the majority of which are involved in pre-mRNA splicing. Additional quantitative experiments are required to validate our ACMS results and confirm the existence of such protein interactions in vivo

Abstract:
Colorectal cancer (CRC) arises in part from the dysregulation of cellular proliferation, associated with the canonical Wnt pathway, and differentiation, effected by the Notch signalling network. In this thesis, we develop a mathematical model of ordinary differential equations (ODEs) for the coupled interaction of the Notch and Wnt pathways in cells of the human intestinal epithelium. Our central aim is to understand the role of such crosstalk in the genesis and treatment of CRC. An embedding of this model in cells of a simulated colonic tissue enables computational exploration of the cell fate response to spatially inhomogeneous growth cues in the healthy intestinal epithelium. We also examine an alternative, rule-based model from the literature, which employs a simple binary approach to pathway activity, in which the Notch and Wnt pathways are constitutively on or off. Comparison of the two models demonstrates the substantial advantages of the equation-based paradigm, through its delivery of stable and robust cell fate patterning, and its versatility for exploring the multiscale consequences of a variety of subcellular phenomena. Extension of the ODE-based model to include mutant cells facilitates the study of Notch-mediated therapeutic approaches to CRC. We find a marked synergy between the application of γ-secretase inhibitors and Hath1 stabilisers in the treatment of early-stage intestinal polyps. This combined treatment is an efficient means of inducing mitotic arrest in the cell population of the intestinal epithelium through enforced conversion to a secretory phenotype and is highlighted as a viable route for further theoretical, experimental and clinical study.
