This dissertation examines the recent emergence of popular recording artists who explicitly invoke the prophetic voices of their religious traditions in their music. In doing so, they situate their music within a pre-existing prophetic lineage. These musicians selfconsciously view their music as vehicle to spread a divine message, and they recognize that their lyrical voice can either become or echo a prophetic voice.

Furthermore the thesis also demonstrates that the PLO&rsquo
s Islamic rhetoric was affected by Palestinian society&rsquo
s shift along Islamic lines and Islamic component of Palestinian nationalism and culture. The PLO seemed to intensify its resort to Islamic rhetoric with every crisis that decreased the PLO&rsquo
s authority and power. It is concluded that content of the PLO&rsquo
s pragmatic Islamic rhetoric was never related with making Islam as normative and legal basis of Palestinian society, rather it was related with mobilization and legitimating.

This shortcoming may be attributed to more than one factor, including : teacher performance, the learners themselves, the designated syllabus, the methods of teaching or to all these factors. Therefore, the purpose of this study is to evaluate the ESP course (for the faculty of agriculture) by investigating the reasons behind the ineffectiveness of the ESP course. This evaluation process covers : the teachers performance, the learners, the designated syllabus and methods of teaching applied. This study is conducted on a sample of students and a sample of the English language teachers at the faculties of agriculture. The designated syllabus and the methodology applied in teaching are also investigated by means of class observations, and questionnaires addressing both teachers and students. Full textWar Memorials are often forgotten statues, right in the center of town, but still out of our sight. They do not tell you enough to understand them, neither are you interested in putting effort into getting to know and learn from them. This paper investigates how the web could be used to create a war memorial that is more alive, captivating and empathy awakening. There has been some virtual war memorials getting constructed since the web started to bloom, but we could not find a single one that had actually fully explored the potential of the web, and what it might have to offer for the creation of war memorials. Researching the web as a media, experience design, and information visualization, we find possibilities to mourn, commemorate and heal on virtual ground. Inspiring reflection and contemplation are another two purposes of The Common Fate Memorial. War memorial studies give us the background information needed, and ceremony mechanics are studied for further inspiration. Our findings are implemented in flash prototypes, which are user tested and evaluated.

Full textThis study addresses aspects of the early development of Phoronida (“horseshoe worms”). The cleavage process is analyzed for three species (Phoronis pallida, Phoronis muelleri, Phoronis vancouverensis). These investigations are performed using 4D-microscopy as well as immunocytochemical stainings of the mitotic spindle apparatuses in combination with confocal laser-scanning microscopy. Different morphological features of the cleavage process are quantified and compared within as well as between the species. The results reveal a highly consistent cleavage of P. pallida and P. muelleri embryos: from the third cell cycle onward, the blastomeres divide mostly obliquely – and alternatingly dextral and sinistral – with respect to the animal-vegetal axis. This cleavage pattern shows surprising correspondences to the pattern of spiral cleavage. The finding can be interpreted as morphological support for recent molecule-based phylogenies, which indicate a position of Phoronida within the Spiralia/Lophotrochozoa clade. The cleavage of P. vancouverensis differs from the cleavage in the other two species; however, it also shows differences to a radial cleavage pattern. In all three species, the cleavage process also involves some degree of variability. Furthermore, the study traces the cell fates of early P. pallida embryos up to the state of late gastrulation, by the use of fluorescent in-vivo single cell markings. These analyses reveal that the first two cleavage divisions both pass through the later axis blastopore-apical plate of the gastrula, yet they do not pass in a constant relationship with respect to the later plane of bilateral symmetry. This differs from the situation known from spiral cleaving animals. The differences and the encountered variability of the cleavage process are discussed with respect to different mechanisms of the specification of cell fates and body axes in different taxa of the Spiralia and the Phoronida.

Understood as a dramatic text, the script in question presents features that are typical of the tragedy. Thus, we analyze the main characters as dramatic characters, the chorus and the structure of the script, from selected scenes. We are particularly interested in the matter of redemption of the main characters Cahit and Sibel. To help us in theoretical basis, we turn to the study of Aristotle, Nietzsche, Bordwell, Jaspers and Romilly.

This essay is based on an investigation carried out with the help of the Corpus of Contemporary American English (COCA). Taking the system of transitivity as its theoretical base and using spoken and written discourse as its primary source, this study aspired to find out what kinds of actions Americans perceive Fate to perform, for example physical, mental or verbal, in order to control what happens to people. It also aimed to reveal what actions people are said to perform in their attempts to control Fate. Do Americans deem Fate capable of, for instance, “deciding”, “talking” or “conspiring” and do they say that people, for example, “challenge”, “defy” or “defeat” Fate? Furthermore, a comparison was made in terms of the actions performed by Fate and people between the different domains of discourse represented in the corpus: spoken, fiction, magazine, newspaper and academic.
Among other things, this investigation shows that in American discourse both Fate and people are perceived to resort to physical strategies rather than mental or verbal ones in their endeavor to control one another.

Embryonic stem (ES) cells have a vast therapeutic potential given their pluripotency, or the ability to differentiate into tissues from all three germ layers. One of the ultimate goals of regenerative medicine is to isolate pluripotent stem cells from patients. Nuclear reprogramming offers the possibility of creating patient-specific cell lines, thus abrogating the need for immunosuppressants following cell transplantation therapy. It was recently reported that the forced expression of four transcription factors, Oct4, Sox2, c-Myc and Klf4 can induce a pluripotent state in somatic cells, without the need for embryo destruction. The work presented here aims to characterize reprogramming using defined factors and provide insight into the mechanisms governing this process. It also seeks to identify transient cues to induce reprogramming in somatic cells, alleviating the need for virally transduced transcription factors that hinder its eventual clinical use.

Full textDen dynamiska (ac) Starkeffekten beskriver energiskiftet för elektroniska tillstånd som induceras av ett oscillerande elektriskt fält. Storleken på detta skift ökar med kvadraten av den elektriska fältstyrkan, det vill säga med ljusintensitet. Tillämpningen av denna fundamentala effekt i syfte att omforma molekylära potentialer, och därmed styra kemiska reaktioner, kallas för dynamisk Starkkontroll. Syftet med denna studie var att undersöka hur den dynamiska Starkeffekten påverkar den fotoinducerade dissociationen av molekylärt syre (O2) inom Schumann-Runge kontinuumet, SRC (130–175 nm). Absorption i SRC resulterar i dissociation via det så kallade B-tillståndet, som bildar O (1D) + O(3P), eller via J-tillståndet, som leder till bildandet av O(3P) + O(3P). Båda dessa dissociativa tillstånd har en karaktär som kan beskrivas som en blandning av ett valenstillstånd och ett Rydbergstillstånd. Simuleringar antyder att, till följd av valens- och Rydbergskaraktären hos B och J-tillståndet, leder dynamisk Starkskiftning av de kopplade Rydbergstillstånden till en dramatisk ändring i det relativa utbytet för de två dissociationskanalerna, samt till ett röd- skift av absorptionsspektrumet. Denna studie hade som ändamål att experimentellt testa denna teoretiska förutsägelse. En femtosekundslaser vid 400 nm användes som kombinerat excitations- och kontrollfält, vilket parallellt inducerar en trefoton-övergång in i SRC och ac Starkskiftar potentialerna. En detektionsmetod som mäter variationer i absorptionen för B-kanalen som funktion av pulsintensitet designades och implementerades. I den valda metoden detekteras emission vid 762 nm från O2(b − X)-övergången, vilket i sin tur ger en mätning av O(1D) som genereras från B- kanalen via en energiöverföringsreaktion. De experimentella resultaten stämmer relativt väl överens med simuleringarna. Den uppmätta emissionen vid 762 nm uppvisar ett intensitetsberoende som i stora drag reflekterar ac Stark- skiftningen av potentialerna. Utöver detta finns dock ett stort bidrag från mättnad, vilket försvårar tolkningen av datan. Vi-dare avviker den experimentella datan betydligt vid höga pulsintensiteter, vilket sannolikt tyder på att O(1D) även genereras genom absorption till högre exciterade tillstånd. Slutligen ob-serveras mindre, men tydliga avvikelser vid låga pulsintensiteter. Dessa kan möjligen tillordnas vibrationsresonanser med högre Rydbergstillstånd genom fyrfoton-absorption.

Full textThe cap-binding complex (CBC) plays a pivotal role in post-transcriptional processing events and orchestrates a variety of metabolic pathways, through association with different interaction partners. Two CBC sub-complexes, the CBC-ARS2-PHAX (CBCAP) and the CBC-nuclear exosome targeting (NEXT) complex (CBCN), were recently shown to target capped RNA either toward export or degradation, but the mechanisms by which they can discriminate between different RNA families and route them toward different metabolic pathways still remain unclear. A major question to be answered is how and when the different CBC subcomplexes are recruited to the RNP. Here, we used an individual nucleotide-resolution UV cross-linking and immunoprecipitation (iCLIP) approach to identify the transcriptome-wide targets for 5 different components of the CBCAP and CBCN complexes, and compared results to the previously analysed NEXT-component RBM7. We report that: (i) CBP20, ARS2, PHAX and ZC3H18 bind close to the cap, while RBM7 and MTR4 bind throughout the mRNA body; (ii) CBP20, ARS2, PHAX and ZC3H18 associate with a broad set of RNA polymerase II (PolII)-derived RNAs and have only mild species preferences; (iii) binding varies with the RNA maturation stage, with the CBC being highly enriched on mature mRNA, ARS2/PHAX/ZC3H18/MTR4 less so, and RMB7 preferentially bound to pre-mRNAs; (iv) MTR4 and RBM7 show different specificities, with RBM7 being highly enriched on introns and promoter upstream transcripts (PROMPTs), while MTR4 is additionally present on mature RNAs. Although more experimental work is needed to fully support our model, we propose that CBCAP and CBCN bind overlapping sets of RNAs, indicating a competition between the proteins ZC3H18 and PHAX, and the lack of a strict RNA sorting mechanism. RNA fate may therefore be determined by additional RNA features and/or by other RNA-binding proteins, which may synergize with the cap and drive the formation of one specific CBC subcomplex instead of another. In an attempt to identify yet unknown factors that may interact with cap-bound CBCAP and CBCN, we performed a protein interaction screen leveraging affinity capture-mass spectrometry (ACMS), using ARS2 and CBP80 as bait proteins. As a complementary approach, we also employed a formaldehyde-based chemical cross-linking strategy, aimed at stabilizing weak/transient interactions. Although we failed to detect any transient interactions involving the CBC, we identified several potential CBC80 and ARS2 interactors, the majority of which are involved in pre-mRNA splicing. Additional quantitative experiments are required to validate our ACMS results and confirm the existence of such protein interactions in vivo
