BackgroundThe National Institute for Health and Care Excellence recommends the use of spirometry and measuring the fraction of exhaled nitric oxide (FeNO) as part of the diagnostic work-up for children with suspected asthma, and spirometry for asthma monitoring, across all care settings. However, the feasibility and acceptability of these tests within primary care are not known.AimTo investigate the feasibility, acceptability, training, and capacity requirements of performing spirometry and FeNO testing in children managed for asthma in UK primary care.Design and settingProspective observational study involving 10 general practices in the East Midlands, UK, and 612 children between 2016 and 2017.MethodTraining and support to perform spirometry and FeNO in children aged 5 to 16 years were provided to participating practices. Children on the practice’s asthma registers, and those with suspected asthma, were invited for a routine asthma review. Time for general practice staff to achieve competencies in performing and/or interpreting both tests, time to perform the tests, number of children able to perform the tests, and feedback on acceptability were recorded.ResultsA total of 27 general practice staff were trained in a mean time of 10.3 (standard deviation 2.7) hours. Usable spirometry and FeNO results were obtained in 575 (94%) and 472 (77%) children respectively. Spirometry is achievable in the majority of children aged ≥5 years, and FeNO in children aged ≥7 years. All of the staff and 97% of families surveyed provided positive feedback for the tests.ConclusionAfter training, general practice staff obtained quality spirometry and FeNO data from most children tested. Testing was acceptable to staff and families. The majority of general practice staff reported that spirometry helped them to manage children’s asthma better.

Aim and Objective: Flap endonuclease-1 (FEN1) plays a central role in DNA replication and DNA damage repair process. In mammals, FEN1 functional sites variation is related to cancer and chronic inflammation, and supports the role of FEN1 as a tumor suppressor. However, FEN1 is overexpressed in multiple types of cancer cells and is associated with drug resistance, supporting its role as an oncogene. Hence, it is vital to explore the multi-functions of FEN1 in normal cell metabolic process. This study was undertaken to examine how the gene expression profile changes when FEN1 is downregulated in 293T cells. Materials and Methods: Using the RNA sequencing and real-time PCR approaches, the transcript expression profile of FEN1 knockdown HEK293T cells have been detected for the next step evaluation, analyzation, and validation. Results: Our results confirmed that FEN1 is important for cell viability. We showed that when FEN1 downregulation led to the interruption of nucleic acids related metabolisms, cell cycle related metabolisms are significantly interrupted. FEN1 may also participate in non-coding RNA processing, ribosome RNA processing, transfer RNA processing, ribosome biogenesis, virus infection and cell morphogenesis. Conclusion: These findings provide insight into how FEN1 nuclease might regulate a wide variety of biological processes, and laid the foundation for understanding the role of other RAD2 family nucleases in cell growth and metabolism.

Abstract:
Flap endonuclease 1 (FEN1) is a member of the family of structure-specific endonucleases implicated in regulation of DNA damage response and DNA replication. So far, knowledge on the role of FEN1 during viral infections is limited. Previous publications indicated that poxviruses encode a conserved protein that acts in a manner similar to FEN1 to stimulate homologous recombination, double-strand break (DSB) repair and full-size genome formation. Only recently, cellular FEN1 has been identified as a key component for hepatitis B virus cccDNA formation. Here, we report on a novel functional interaction between Flap endonuclease 1 (FEN1) and the human cytomegalovirus (HCMV) immediate early protein 1 (IE1). Our results provide evidence that IE1 manipulates FEN1 in an unprecedented manner: we observed that direct IE1 binding does not only enhance FEN1 protein stability but also phosphorylation at serine 187. This correlates with nucleolar exclusion of FEN1 stimulating its DSB-generating gap endonuclease activity. Depletion of FEN1 and inhibition of its enzymatic activity during HCMV infection significantly reduced nascent viral DNA synthesis demonstrating a supportive role for efficient HCMV DNA replication. Furthermore, our results indicate that FEN1 is required for the formation of DSBs during HCMV infection suggesting that IE1 acts as viral activator of FEN1 in order to re-initiate stalled replication forks. In summary, we propose a novel mechanism of viral FEN1 activation to overcome replication fork barriers at difficult-to-replicate sites in viral genomes.

Abstract:
FENs (flap endonucleases) and related FEN-like enzymes [EXO-1 (exonuclease-1), GEN-1 (gap endonuclease 1) and XPG (xeroderma pigmentosum complementation group G)] are a family of bivalent-metal-ion-dependent nucleases that catalyse structure-specific hydrolysis of DNA duplex-containing nucleic acid structures during DNA replication, repair and recombination. In the case of FENs, the ability to catalyse reactions on a variety of substrates has been rationalized as a result of combined functional and structural studies. Analyses of FENs also exemplify controversies regarding the two-metal-ion mechanism. However, kinetic studies of T5FEN (bacteriophage T5 FEN) reveal that a two-metal-ion-like mechanism for chemical catalysis is plausible. Consideration of the metallobiochemistry and the positioning of substrate in metal-free structures has led to the proposal that the duplex termini of substrates are unpaired in the catalytically active form and that FENs and related enzymes may recognize breathing duplex termini within more complex structures. An outstanding issue in FEN catalysis is the role played by the intermediate (I) domain arch or clamp. It has been proposed that FENs thread the 5′-portion of their substrates through this arch, which is wide enough to accommodate single-stranded, but not double-stranded, DNA. However, FENs exhibit gap endonuclease activity acting upon substrates that have a region of 5′-duplex. Moreover, the action of other FEN family members such as GEN-1, proposed to target Holliday junctions without termini, appears incompatible with a threading mechanism. An alterative is that the I domain is used as a clamp. A future challenge is to clarify the role of this domain in FENs and related enzymes.

Two new South Asian catfish species of the family Erethistidae, Pseudolaguvia inornata and Pseudolaguvia muricata, are described from the Brahmaputra and Feni River drainages in Bangladesh. Pseudolaguvia inornata from the Feni River drainage can be distinguished from congeners in having a uniform coloration with a pale mid-dorsal stripe and brown submarginal stripes on each lobe of the caudal fin, while P muricata from the Brahmaputra River drainage can be distinguished from congeners by its elongate dorsal and pectoral-fin spines.

ABSTRACT Low-sulfate, acidic (approximately pH 4) fens in the Lehstenbach catchment in the Fichtelgebirge mountains in Germany are unusual habitats for sulfate-reducing prokaryotes (SRPs) that have been postulated to facilitate the retention of sulfur and protons in these ecosystems. Despite the low in situ availability of sulfate (concentration in the soil solution, 20 to 200 μM) and the acidic conditions (soil and soil solution pHs, approximately 4 and 5, respectively), the upper peat layers of the soils from two fens (Schlöppnerbrunnen I and II) of this catchment displayed significant sulfate-reducing capacities. 16S rRNA gene-based oligonucleotide microarray analyses revealed stable diversity patterns for recognized SRPs in the upper 30 cm of both fens. Members of the family “Syntrophobacteraceae” were detected in both fens, while signals specific for the genus Desulfomonile were observed only in soils from Schlöppnerbrunnen I. These results were confirmed and extended by comparative analyses of environmentally retrieved 16S rRNA and dissimilatory (bi)sulfite reductase (dsrAB) gene sequences; dsrAB sequences from Desulfobacca-like SRPs, which were not identified by microarray analysis, were obtained from both fens. Hypotheses concerning the ecophysiological role of these three SRP groups in the fens were formulated based on the known physiological properties of their cultured relatives. In addition to these recognized SRP lineages, six novel dsrAB types that were phylogenetically unrelated to all known SRPs were detected in the fens. These dsrAB sequences had no features indicative of pseudogenes and likely represent novel, deeply branching, sulfate- or sulfite-reducing prokaryotes that are specialized colonists of low-sulfate habitats.

Architecture can be viewed as a reflection of value placed on life. In colonial Hong Kong, a distinctive Gothic-style castle, Euston, was built by tycoon Eu Tong Sen (1877–1941) as his family's grand residence. Eu was a prominent figure in South China and Southeast Asia and remains a local legend decades after his death. Eu's castle, being built in 1928 and demolished in the 1980s, was and still is one of the most recognizable monuments in the region. Although Eu did not leave behind any autobiography or memoirs, the monumental castle can be regarded as a symbolic manifestation of his life story. The design of the castle is of mixed ancestry—it is a reconciliation of traditional Chinese design based on feng shui (Chinese geomancy) with European architectural elements. The fusion of East-West architectural building elements, as symbolized by the Eu castle, was a significant achievement symbolizing general social and cultural changes spanning more than a century.

Red Lake peatland, a vast mire complex in northern Minnesota, contains 21 Sphagnum species, 47 other mosses, and 17 liverwort taxa in its present flora, and several other species present only as fossils in the peat. Four broad vegetation types are recognized: (i) bogs, (ii) poor fens, (iii) forested rich fens, and (iv) rich-fen pools (flarks). These vegetation types are differentiated by water chemistry and bryophyte associations. Most of the major peat-forming moss species are common, circumboreally distributed taxa, and they are distinct in their ecology. Shifts in some of their habitat requirements, however, are evident among different geographic regions of the northern hemisphere. In bogs the major peat-formers are Sphagnum species, while on minerotrophic sites in Red Lake peatland they belong mainly to the family Amblystegiaceae.

This article examines Sarah Kofman's interpretation of Nietzsche in light of the claim that interpretation was for her both an articulation of her identity and a mode of deconstructing the very notion of identity. Faulkner argues that Kofman's work on Nietzsche can be understood as autobiographical, in that it served to mediate a relation to her self. Faulkner examines this relation with reference to Klein's model of the child's connection to its mother. By examining Kofman's later writings on Nietzsche alongside her autobiography, this article contends that Kofman's defense of anti-Semitism in Nietzsche serves to fend off her own ambivalence about being Jewish.

AbstractSix small peatlands, comprising three bogs and three fens, were surveyed for spiders in southern Ontario or southwestern Quebec. Collections from the Mer Bleue bog contained representatives of 108 species (54.6% of the total), those from the Crieff fen 97 (49.0%), those from the Wylde Lake bog 77 (38.9%), those from the Oliver bog 76 (38.4%), those from the Richmond fen 59 (29.8%), and those from the Poltimore fen 49 species (24.8% of the total). The family Erigonidae had 24.8% of the total 198 species, Lycosidae had 11.1%, Linyphiidae had 10.6%, and Salticidae had 9.1%, Web-building species outnumbered hunting species by 118 to 80. The majority of species in these peatlands are wide-ranging litter inhabitants, about one-third are tree, shrub, or herb inhabitants, and one-tenth were found only in or on the surface of the peat itself. Nine of the last-named group appear to be restricted to bogs, but none appear to be restricted to fens.

The hSuv3 (human Suv3) helicase has been shown to be a major player in mitochondrial RNA surveillance and decay, but its physiological role might go beyond this functional niche. hSuv3 has been found to interact with BLM (Bloom's syndrome protein) and WRN (Werner's syndrome protein), members of the RecQ helicase family involved in multiple DNA metabolic processes, and in protection and stabilization of the genome. In the present study, we have addressed the possible role of hSuv3 in genome maintenance by examining its potential association with key interaction partners of the RecQ helicases. By analysis of hSuv3 co-IP (co-immunoprecipitation) complexes, we identify two new interaction partners of hSuv3: the RPA (replication protein A) and FEN1 (flap endonuclease 1). Utilizing an in vitro biochemical assay we find that low amounts of RPA inhibit helicase activity of hSuv3 on a forked substrate. Another single-strand-binding protein, mtSSB (mitochondrial single-strand-binding protein), fails to affect hSuv3 activity, indicating that the functional interaction is specific for hSuv3 and RPA. Further in vitro studies demonstrate that the flap endonuclease activity of FEN1 is stimulated by hSuv3 independently of flap length. hSuv3 is generally thought to be a mitochondrial helicase, but the physical and functional interactions between hSuv3 and known RecQ helicase-associated proteins strengthen the hypothesis that hSuv3 may play a significant role in nuclear DNA metabolism as well.

RationaleThe relationship between clinical and biomarker characteristics of asthma and its severity in Africa is not well known.MethodsUsing the Expert Panel Report 3, we assessed for asthma severity and its relationship with key phenotypic characteristics in Uganda, Kenya and Ethiopia. The characteristics included adult onset asthma, family history of asthma, exposures (smoking and biomass), comorbidities (HIV, hypertension, obesity, tuberculosis (TB), rhinosinusitis, gastro-oesophageal disease (GERD) and biomarkers (fractional exhaled nitric oxide (FeNO), skin prick test (SPT) and blood eosinophils). We compared these characteristics on the basis of severity and fitted a multivariable logistic regression model to assess the independent association of these characteristics with asthma severity.ResultsA total of 1671 patients were enrolled, 70.7% women, with median age of 40 years. The prevalence of intermittent, mild persistent, moderate persistent and severe persistent asthma was 2.9%, 19.9%, 42.6% and 34.6%, respectively. Only 14% were on inhaled corticosteroids (ICS). Patients with severe persistent asthma had a higher rate of adult onset asthma, smoking, HIV, history of TB, FeNO and absolute eosinophil count but lower rates of GERD, rhinosinusitis and SPT positivity. In the multivariate model, Ethiopian site and a history of GERD remained associated with asthma severity.DiscussionThe majority of patients in this cohort presented with moderate to severe persistent asthma and the use of ICS was very low. Improving access to ICS and other inhaled therapies could greatly reduce asthma morbidity in Africa.

It is widely accepted that the conflict between women’s family obligations and professional jobs’ long hours lies at the heart of their stalled advancement. Yet research suggests that this “work–family narrative” is incomplete: men also experience it and nevertheless advance; moreover, organizations’ effort to mitigate it through flexible work policies has not improved women’s advancement prospects and often hurts them. Hence this presumed remedy has the perverse effect of perpetuating the problem. Drawing on a case study of a professional service firm, we develop a multilevel theory to explain why organizations are caught in this conundrum. We present data suggesting that the work–family explanation has become a “hegemonic narrative”—a pervasive, status-quo-preserving story that prevails despite countervailing evidence. We then advance systems-psychodynamic theory to show how organizations use this narrative and attendant policies and practices as an unconscious “social defense” to help employees fend off anxieties raised by a 24/7 work culture and to protect organizationally powerful groups—in our case, men and the firm’s leaders—and in so doing, sustain workplace inequality. Due to the social defense, two orthodoxies remain unchallenged—the necessity of long work hours and the inescapability of women’s stalled advancement. The result is that women’s thin representation at senior levels remains in place. We conclude by highlighting contributions to work–family, workplace inequality, and systems-psychodynamic theory.

Single fathers (i.e. men rearing their children without support from a ‘live-in’ adult companion) have been termed an ‘unnoticed group’ in Australian society (Bain, 1973).Apart from the author's own recently completed research (Wilson, 1988), only three previous Australian studies about single fathers have been undertaken. This evident lack of interest in single fathers needs to be understood within a broader context of traditional family research approaches, which denied the significance of the fathers' role in the socialisation and nurturance of children. Fathers have been termed the “forgotten contributors to child development’ (Lamb, 1975) since traditionally, social scientists have viewed fathers as aloof and distant from their own children, with little capacity to undertake core caretaking roles in families (Fein, 1978).

Abstract:
FENs (flap endonucleases) play essential roles in DNA replication, pivotally in the resolution of Okazaki fragments. In eubacteria, DNA PolI (polymerase I) contains a flap processing domain, the N-terminal 5′→3′ exonuclease. We present evidence of paralogous FEN-encoding genes present in many eubacteria. Two distinct classes of these independent FEN-encoding genes exist with four groups of eubacteria, being identified based on the number and type of FEN gene encoded. The respective proteins possess distinct motifs hallmarking their differentiation. Crucially, based on primary sequence and predicted secondary structural motifs, we reveal key differences at their active sites. These results are supported by biochemical characterization of two family members - ExoIX (exonuclease IX) from Escherichia coli and SaFEN (Staphylococcus aureus FEN). These proteins displayed marked differences in their ability to process a range of branched and linear DNA structures. On bifurcated substrates, SaFEN exhibited similar substrate specificity to previously characterized FENs. In quantitative exonuclease assays, SaFEN maintained a comparable activity with that reported for PolI. However, ExoIX showed no observable enzymatic activity. A threaded model is presented for SaFEN, demonstrating the probable interaction of this newly identified class of FEN with divalent metal ions and a branched DNA substrate. The results from the present study provide an intriguing model for the cellular role of these FEN sub-classes and illustrate the evolutionary importance of processing aberrant DNA, which has led to their maintenance alongside DNA PolI in many eubacteria.

Recently, a new class of histone methyltransferases that plays an indirect role in chromatin silencing by targeting a conserved lysine residue in the nucleosome core was described, namely the Dot1 (disruptor of telomeric silencing) family [Feng, Wang, Ng, Erdjument-Bromage, Tempst, Struhl and Zhang (2002) Curr. Biol. 12, 1052–1058; van Leeuwen, Gafken and Gottschling (2002) Cell (Cambridge, Mass.) 109, 745–756; Ng, Feng, Wang, Erdjument-Bromage, Tempst, Zhang and Struhl (2002) Genes Dev. 16, 1518–1527]. In the present study, we report the isolation, genomic organization and in vivo expression of a mouse Dot1 homologue (mDot1). Expressed sequence tag analysis identified five mDot1 mRNAs (mDot1a–mDot1e) derived from alternative splicing. mDot1a and mDot1b encode 1540 and 1114 amino acids respectively, whereas mDot1c–mDot1e are incomplete at the 5´-end. mDot1a is closest to its human counterpart (hDot1L), sharing 84% amino acid identity. mDot1b is truncated at its N- and C-termini and contains an internal deletion. The five mDot1 isoforms are encoded by 28 exons on chromosome 10qC1, with exons 24 and 28 further divided into two and four sections respectively. Alternative splicing occurs in exons 3, 4, 12, 24, 27 and 28. Northern-blot analysis with probes corresponding to the methyltransferase domain or the mDot1a-coding region detected 7.6 and 9.5 kb transcripts in multiple tissues, but only the 7.6 kb transcript was evident in mIMCD3-collecting duct cells. Transfection of mDot1a–EGFP constructs (where EGFP stands for enhanced green fluorescent protein) into human embryonic kidney (HEK)-293T or mIMCD3 cells increased the methylation of H3-K79 but not H3-K4, -K9 or -K36. Furthermore, DMSO induced mDot1 gene expression and methylation specifically at H3-K79 in mIMCD3 cells in a time- and dose-dependent manner. Collectively, these results add new members to the Dot1 family and show that mDot1 is involved in a DMSO-mediated signal-transduction pathway in collecting duct cells.

Whereas the physiological significance of microsomal fatty acid elongation is generally appreciated, its molecular nature is poorly understood. Here, we describe tissue-specific regulation of a novel mouse gene family encoding components implicated in the synthesis of very long chain fatty acids. The Ssc1 gene appears to be ubiquitously expressed, whereas Ssc2 and Cig30 show a restricted expression pattern. Their translation products are all integral membrane proteins with five putative transmembrane domains. By complementing the homologous yeast mutants, we found that Ssc1 could rescue normal sphingolipid synthesis in the sur4/elo3 mutant lacking the ability to synthesize cerotic acid (C26:0). Similarly, Cig30 reverted the phenotype of the fen1/elo2 mutant that has reduced levels of fatty acids in the C20–C24 range. Further, we show that Ssc1 mRNA levels were markedly decreased in the brains of myelin-deficient mouse mutants known to have very low fatty acid chain elongation activity. Conversely, the dramatic induction of Cig30 expression during brown fat recruitment coincided with elevated elongation activity. Our results strongly implicate this new mammalian gene family in tissue-specific synthesis of very long chain fatty acids and sphingolipids.

In all cases, the (4,4)-grid topology is formed by the bridging of octahedral iron(ii) sites in the equatorial plane by bis-pyridyl ligands of varying length, flexibility, and intermolecular interaction capacity. In particular, the six ligands n-(4-pyridyl)-isonicotinamide (pin), trans-1,2-bis(4′-pyridyl)ethane (tvp), 1,2-dibromo-1,2-bis(4′-pyridyl)ethane (dbbpe), bis(4-pyridyl)-1,2,4,5-tetrazine (bptz), 4,4′-bis(pyridyl)acetylene (bpac), and 1,4-bis(4-pyridylethynyl)benzene (bpeben) have been utilised. The seven new materials [Fe(NCS)2(pin)2]·2(MeCN) (pin-S), [Fe(tvp)2(NCS)2]·1/2(tvp)·(CH3CH2OH) (tvp-S), [Fe(dbbpe)2(NCS)2]·6(CH3CN) (dbbpe-S), [Fe(NCS)2(bptz)2]·2(CHCl3)·6(EtOH) (bptz-S), [Fe(NCSe)2(bptz)2]·4(CHCl3)·(EtOH)·(H2O) (bptz-Se), [Fe(NCS)2(bpac)2]·2(PrOH) (bpac-S), and [Fe(NCS)2(bpeben)2]·2(CHCl3) (bpeben-S) all form (4,4)-grids of varying size that are arranged in a parallel stacked topology. Despite being in the [FeN6] coordination environment known to be conducive to spin crossover, these materials all remain high-spin with thermal variation. These results are discussed in context with the large family of SCOFs that show varied spin crossover behaviours.

Accumulating evidence has suggested a role of the small GTPase Ras homolog gene family member A (RhoA) in DNA damage response (DDR) in addition to its traditional function of regulating cell morphology. In DDR, 2 key components of DNA repair, ataxia telangiectasia-mutated (ATM) and flap structure-specific endonuclease 1 (FEN1), along with intracellular reactive oxygen species (ROS) have been shown to regulate RhoA activation. In addition, Rho-specific guanine exchange factors (GEFs), neuroepithelial transforming gene 1 (Net1) and epithelial cell transforming sequence 2 (Ect2), have specific functions in DDR, and they also participate in Ras-related C3 botulinum toxin substrate 1 (Rac1)/RhoA interaction, a process which is largely unappreciated yet possibly of significance in DDR. Downstream of RhoA, current evidence has highlighted its role in mediating cell cycle arrest, which is an important step in DNA repair. Unraveling the mechanism by which RhoA modulates DDR may provide more insight into DDR itself and may aid in the future development of cancer therapies.

The Taiji Ruler is a traditional system of health-enhancing qigong that is attributed to the Song Dynasty Daoist recluse Chen Xiyi and was first taught publicly in the 1950s. The exercises may be practiced while holding a foot-long wooden object, the Ruler, or with a variety of training devices, such as a wooden or stone ball. Mr. Cohen began studying Taji Ruler with various teachers more than 30 years ago, but also bases his research on works in Chinese and English. Penelitian ini pada masyarakat Kecamatan Nunkolo, Kabupaten Timor Tengah Selatan di Desa Haumeni, Desa Putun dan Desa Nunkolo. Jumlah informen dalam penelitian ini 30 orang yang terdiri atas kepala Desa, tokoh masyarakat, pengrajin dan masyarakat yang mengetahui tentang pemanfaatan tumbuhan pewarna alami. Analisis data menggunakan analisis deskriptif. Dari hasil penelitian diperoleh 5 jenis tumbuhan pewarna alami yaitu rimpang dari famili Zingiberaceace yaitu Kunyit/Huki (Curcuma domestica L) menghasilkan warna kuning yang diolah dengan cara ditumbuk, warna merah dihasilkan dari kulit batang famili Rubiaceace yakni Mengkudu/Baok’ulu (Morinda citrifolia L) yang direbus, warna hijau dihasilkan dari famili Fabaceace yakni daun kacang Arbila/Koto (Phaseolus lunatus L) yang ditumbuk, warna hitam dihasilkan dari famili Rubiaceace yakni daun Tarum/Taum (Indigofera tinctoria L) yang direndam, dari famili Euphorbiaceae yang diambil adalah daun Pohon Buah Tinta/Meko (Phylantus reticulatus Poir) untuk warna biru diolah dengan direndam dan dicampuran tepung jagung, Selain itu terdapat tumbuhan yang digunakan sebagai bahan tambahan dalam pewarnaan yaitu daging buah kelapa/Noa (Cocos nucifera) dan kapur sirih. Tumbuhan yang digunakan dalam proses perendaman awal adalah Kemiri/Fenu (Aleurites mollucana L) dan Jagung/Pena (Zea Mays L). Tujuan penambahan kapur adalah sebagai penguat warna sehingga tidak mudah luntur. Tumbuhan yang berpotensi sebagai pewarna alami tenun ikat sebagian besar telah dibudidayakan. Tumbuhan yang dibudidayakan adalah Tarum, Kunyit dan kacang Arbila.

CD48, belonging to the CD2 family, participates in mast cell-stimulating cross-talk, facilitates the formation of the mast cell/eosinophil effector unit, and is expressed by eosinophils. <b><i>Objectives:</i></b> To assess the serum level of soluble form of CD48 (sCD48) in patients with IAR during and out of the pollen season and correlate with the disease severity and with eosinophil-related parameters. <b><i>Materials and Methods:</i></b> Sixty-three patients (female: 79%; mean age: 30.58) were included to the study. Forty-five patients were assessed during the pollen season and other 42 patients during out of the pollen season. Twenty-four patients (female: 37.50%; mean age: 27.90) were evaluated twice, during the pollen season and out of the pollen season. sCD48, ECP, eotaxin-1/CCL11 serum levels together with complete blood count, and fractional exhaled nitric oxide bronchial and nasal fraction (FeNO) were performed. The severity of symptoms was assessed using the Total Nasal Symptom Score (TNSS), and neutrophil-to-lymphocyte (NLR) and eosinophil-to-lymphocyte (ELR) ratios were calculated. <b><i>Results:</i></b> sCD48 serum level, FeNO nasal and bronchial fractions, and TNSS were significantly higher in the IAR group in the pollen season compared with out of the pollen season. Differences in ECP, eotaxin-1/CCL11 serum levels, and NLR and ELR were not significant between season and out of the season. No correlations were found between sCD48 and eosinophil-related parameters. <b><i>Conclusions:</i></b> sCD48 may be a biomarker to the exacerbation phase in patients with IAR. One can assume that CD48 participates in the pathogenesis of IAR.

Solute carrier family 11 member a1 gene (Slc11a1) previously known as natural resistance-associated macrophage protein 1 (Nramp1) is a gene of member of family of metal ion-transport protein. The cellular expression is restricted to phagocytic cells. Slc11a1 delivers bivalent metal cations from the cytosol into acidic endosomal and lysosomal compartments where Fenon and Haber-Weiss reaction generates toxic antimicrobial radicals for direct antimicrobial activity against harmful microorganisms. The present study was undertaken with the objective of analyzing Slc11a1 gene to gain insight into the evolutionary proximity and divergence as well as the polymorphism of this gene in ruminants and non-ruminants including the attendant effects of the genetic variants on the function of the Slc11a1 protein. Thirty Slc11a1 gene sequences of 6 mammalian species classified as ruminant (goat, sheep, cattle, Bubalus bubalis and Bubalus carabanensis) and non-ruminant (swine and horse) animals were investigated. The length of the Slc11a1 gene varied from 448-2,357. There was substantial genetic variation and polymorphism in the aligned sequences of Slc11a1 gene within and across species. Functional analysis of non-synonymous mutations in cattle revealed that twenty five of the amino acid substitutions at the peptide binding region could be beneficial, (E36G), (T52A), (N161S) and (V248I) were likely to be beneficial while only (Q312K) was harmful. In horse, two of the amino acid mutations were harmful, two were likely to be harmful, one was undecided, four were likely beneficial and the rest twenty were beneficial. The phylogenetic trees showed some form of proximity and differentiation in Slc11a1 sequences within and across species. The present information on the polymorphism of Slc11a1 gene might be exploited in the search for association with disease resistance in Nigerian livestock species.

The study was undertaken to examine the socioeconomic profile of buffalo farmers and to assess the marketing and consumers preference on buffen (buffalo meat) in the selected areas. Twelve districts namely: Mymensingh, Jamalpur, Moulovibazar, Bhola, Bagerhat, Feni, Potuakhali, Noakhali, Laxmipur, Chittagong, Tangail and Sirajgong were selected purposively. A total of 1400 buffalo farmers were interviewed following simple random sampling technique. Data were collected during June 2011 to April 2016 and analyzed data using SPSS software. Study revealed that the highest per cent of farmers were in age group 31-45 years indicating that farmers were mature enough to give more labour to their farming activities. On average, 88 per cent buffalo farmers were engaged purely in agriculture followed by business and service as primary occupation. The highest numbers of farmers were illiterate followed by primary education, SSC, HSC and Degree. About 49 per cent buffalo farmers had above 15 years of farming experience of rearing buffalo. Average farm size was estimated 0.95 hectare indicating small and medium category farm and average family size was calculated 6 persons per family which is higher than national average 4.9. Dependency ratio was also estimated to 0.94. The study showed that buffen contributes 7.16 per cent of total red meat production and 6.19 per cent of total meat production in Bangladesh and about 50 percent farmers reported that they did fattening before selling of buffalo. About 48 per cent consumers reported that they prefer buffen most among different kinds of meats. In view point of butcher, about 46 percent consumer preferred buffen than beef.

To estimate HIV-1 seroprevalence in the general population of Salvador, Bahia, Brazil, we conducted a cross-sectional survey of 3,437 residents from 1998 to 2000. Subjects were drawn from 30 sentinel areas representing a wide range of living conditions. Plasma samples were screened for HIV-1 antibodies by ELISA and confirmed by immunofluorescent assay. Subtype determination by HMA was performed after proviral DNA amplification. Phylogenetic analysis using parsimony was performed with the neighbor-joining method. Overall HIV-1 seroprevalence was 0.55% (19/3,446): 0.8% for men and 0.36% for women. Seroprevalence was higher in the 31-45-year age group (1%) and among persons with family income less than twice the minimum wage (0.78%) as compared to 0.33% for the higher income group. Syphilis was detected in 37% of HIV seropositive individuals. Phylogenetic inferences identified 10 samples as subtype B in the env region and 2 samples with Benv/Fgag/Fpol and Fenv/Bgag. Age > 30 years, male gender, and income < 2 times the minimum wage were identified as risk factors for HIV-1 infection. Extrapolating the proportion of seropositive individuals to Salvador, the number of HIV-1 infected individuals was estimated at 13,750.

AbstractVetulicolians are a group of exclusively Cambrian animals characterized by an anterior section with lateral pouches and a posterior section that appears segmented. The precise phylogenetic affinity of vetulicolians is debated because there is a lack of consensus regarding the interpretation of their anatomical features. Their disparate morphology might even cause one to question whether this is a monophyletic taxon. In total, there are 15 species grouped into three families included in vetulicolians. Here we focus on new specimens of Yuyuanozoon magnificissimi Chen, Feng, and Zhu in Chen et al., 2003, a species that was first described from only a single specimen from the Chengjiang Biota (Cambrian Series 2, Stage 3, Eoredlichia-Wutingaspis trilobite Biozone), Yunnan Province, China. The species is notable in being exceptionally large (up to 20 cm long). Morphological observations on the new specimens clarify the nature of the wide circular opening at the presumed anterior end of the animal and the cowl-shaped lateral openings within this anterior section. Taphonomic observations identify wrinkles in the anterior section and twists in the posterior segmented section. In particular, the shape of the anterior opening of Yuyuanozoon magnificissimi suggests significant differences from other vetulicolians. Taxonomic reappraisal of Y. magnificissimi indicates that it belongs within the family Didazoonidae, as that is presently defined.

This article is a case study of Huang Pao-tung (1921–2005), who was a polymer chemist and academician of the Chinese Academy of Sciences, and his wife, Feng Zhiliu (1921–2015), who was a polymer physicist. The couple studied in America and returned to China in the 1950s. Based on an analysis of first-hand data from the two scientists’ archives, diaries and memoirs, which recorded their economic lives after they returned, I found as follows: (1) their income after returning to China was about one-fifth of their income in the United States; (2) their income channel was narrow (there was no mechanism for wage increases, and their wages were unchanged for 25 years); (3) the main expenditure of their family was on food, and that proportion increased year by year; and (4) no taxes, low rents, free medical care and other benefits helped to reduce their cost of living in China. The importance of their profession as scientists and the government’s advocacy of scholars returning home brought them relatively good treatment, and their economic benefits and living standard were several times better than those of other ordinary social classes. However, this kind of preferential treatment was dependent on many other things, which caused them to lose independence and autonomy.

Two phylogenetically unrelated superoxide dismutase (SOD) families, i.e., CuZnSOD (copper and zinc SOD) and FeMn-CamSOD (iron, manganese, or cambialistic SOD), eliminate superoxide radicals in different locations within the plant cell. CuZnSOD are located within the cytosol and plastids, while the second family of SOD, which are considered to be of bacterial origin, are usually located within organelles, such as mitochondria. We have used the reactive oxygen species–producer methylviologen (MV) to study SOD isozymes in the indeterminate nodules on pea (Pisum sativum). MV caused severe effects on nodule physiology and structure and also resulted in an increase in SOD activity. Purification and N-terminal analysis identified CamSOD from the Rhizobium leguminosarum endosymbiont as one of the most active SOD in response to the oxidative stress. Fractionation of cell extracts and immunogold labeling confirmed that the CamSOD was present in both the bacteroids and the cytosol (including the nuclei, plastids, and mitochondria) of the N-fixing cells, and also within the uninfected cortical and interstitial cells. These findings, together with previous reports of the occurrence of FeSOD in determinate nodules, indicate that FeMnCamSOD have specific functions in legumes, some of which may be related to signaling between plant and bacterial symbionts, but the occurrence of one or more particular isozymes depends upon the nodule type.

Canine parvovirus (CPV) and canine influenza virus (CIV) are highly contagious virus infecting members of the canine family including dogs, coyotes, foxes and wolves. The research work was carried out to determine the prevalence of CPV and CIV in dogs (stray and pet dogs) in Dhaka, Mymensingh, Feni and Chittagong Districts by using Rapigen CPV and CIV Ag test kit. Fecal samples were collected from 50 dogs of different age and sex from different spots of Dhaka and Chittagong District. Nasal fluid samples were collected from 50 dogs of different age and sex from different spots. The samples were tested with Rapigen canine parvovirus Ag test kit and Rapigen canine influenza Ag test kit. Overall prevalence of CPV in pet dogs was 22% and in stray dogs was 30%. The prevalence of canine parvovirus in relation to age was gradually decreasing with higher age group and the prevalence was 28% in 1-6 months age, 16.66% in 7-12 months age group 11.11%o ver 18 months age group. The prevalence of canine parvovirus was significantly higher in male (24.13%) than that in female (19.04%). All samples showed negative test result for canine influenza virus. Prevalence of canine influenza virus infection were 00.00% in all age groups and sex. Overall prevalence of CIV in all pet and stray dogs were 00.00%.Asian J. Med. Biol. Res. On November 2, 1850, Jeremiah Root published a “Notice” in the Sacramento Transcript offering a reward for the arrest of his wife, Emeline, who had absconded with a younger man, twelve thousand dollars, and their two-year-old daughter, leaving Root and their five sons to fend for themselves at the roadhouse they ran along the American River. When Emeline and their daughter were found three months later on a bark in San Francisco preparing to leave California, Jeremiah met with her and the couple quickly reconciled. Charges were dropped against Emeline and her associates, and Jeremiah and the rest of their family joined her on the ship to travel east. The Transcript editorialized against the apparent tawdry nature of the affair, but a deeper inspection of the history of this forty-niner family reveals in intimate detail how Jeremiah and Emeline's personal struggles emerged from the incredible physical and spiritual turmoil experienced by early Mormon emigrants, who played a seminal role in Gold Rush–era California. Emeline and Jeremiah Root were early converts to Mormonism and arrived in California having survived a twelve-year odyssey that began in Kirtland, Ohio. They were expelled first from Kirtland and then from Nauvoo, Illinois, after the murder of their church leader, Joseph Smith. They persevered through starvation and malnutrition at Winter Quarters on the Missouri River while following Brigham Young to Salt Lake. They struggled with spiritual allegiances as the practice of polygamy and economic inequities became apparent among church leadership, and they ultimately defied Brigham Young by taking the physically demanding overland route from Salt Lake through the Forty-Mile Desert and over the Carson Pass to Gold Rush California in early 1849. Finally, they lived through a tumultuous year on the lower American River, surviving among unruly miners, deadly shootouts over property rights, and a rampant outbreak of cholera. These pressures erupted into a personal crisis when Emeline escaped, escorted by a family friend who was perhaps her lover, taking her only daughter and the family fortune with her. Emeline and Jeremiah's eventual reconciliation and the way Jeremiah ultimately lived out his life revealed them to be people of personal and spiritual integrity who, in this one incident, were overwhelmed by the struggles of the times. Their story illustrates the incredible resiliency of early California pioneers and integrates in vivid detail the physical, spiritual, and emotional challenges facing families in Gold Rush–era California.

National Commission on violence against women (2015) noted that violence against women showed a widespread pattern. The 2016 annual press release (Catahu) released that there were 321,752 of violence cases. The largest type of violence against women was violence in the personal sector. Violence in the form of rape was 72% (2,399 of cases), violence in the form of obscenity was 18% (601 of cases), and sexual abuse was 5% (166 of cases). The majority of personal violence victims (domestic / household) were women. The forms of domestic violence included physical violence, psychological violence, sexual violence and neglect of the household; while the main factor was the lack of communication and disharmony among family members. The impact in the short term was usually like a physical injury, disability, pregnancy, loss of work, and so forth; while the long-term effects were psychological disorders (psychiatric), loss of confidence, fend for oneself, trauma and appearance of fear to depression. The handling of domestic violence is one of the focuses of social workers to play a role in helping / assisting the recovery of victims. The social worker should be able to convince the victims to dare to express their problems, to give a sense of security, and comfort. Social workers in helping victims of domestic violence should have knowledge and alignment to the victims that the slightest violence is a form of crime against humanity. Victims of domestic violence must immediately obtain the protection, security assurance and social assistance. The efforts which can be done by social workers are counseling and family counseling. Those are done as a form of therapy so that victims do not feel the prolonged trauma and they can think calmly.

Roma and Gypsy women in Europe suffer from multiple deprivation (Council of the European Union 2011): Firstly, a large part of Roma and Gypsy people live in poverty. Secondly, their different cultural/ethnic traditions often lead to discrimination in school education. Thirdly, they also have disadvantages through the gender aspect, because the traditional Roma/Gypsy culture defines the place of women to be at home with the family and an educational career is not necessary for that (l. Forray, Hegedűs 2003; Durst 2015). That is why Roma and Gypsy women are often called the “minority of the minority” (Vincze 2010: 195). Despite of this multiple deprivation, Roma and Gypsy women are (not only in Hungary) more and more successful in the education system (Forray; Hegedűs 1991) and they increasingly take part in the political life as well (Bak., T.th 2008; K.cz. 2010). The research study focuses on Roma and Gypsy women who have come from a background of multiple deprivation but managed to achieve successful educational careers (defined by their university degree). To answer the research question “Which factors are regarded as beneficial for success in education from the perspective of Roma and Gypsy women?”, we chose the method of biographical narrative interviews. Additionally, we analysed statistical and empirical studies and used expert interviews as well. The aim of the research was to learn about the subjective theories of the interviewed women. The selection of the respondents was done through the snowball-system. The analysis of the interviews was based on the methodology of Fritz Schütze (Schütze 1983). The underclass theory of William Julius Wilson (Wilson, 1978; 1987) adapted by Iv.n Szel.nyi and J.nos Lad.nyi for the Hungarian situation (Lad.nyi; Szel.nyi, 2004) and the theory of Helmut Fend about the functions of the school in society (Fend, 1980; 2003) served as the theoretical framework for this study. For the interpretation we used the categorisation of factors for school success of Hungarian Roma and Gypsy women from Katalin Forray R. and Andr.s Hegedűs T. (Hegedűs, 1996; Forray, Hegedűs, 2003). The study shows that all interviewed women had a very strong learning motivation and were ready to have conflicts with the traditional female role model. Their parents and teachers played mostly a very positive role in this success, but the most important factor was their individual learning motivation.

Homer represents a new and diversified family of proteins that includes several isoforms, Homer 1, 2, and 3; some of these isoforms have been reported to be present in striated muscles. In this study, the presence of Homer isoforms 1a, 1b/c/d, 2b, and 3 was thoroughly investigated in rat skeletal muscles under resting conditions. Transition in Homer isoforms compositon was studied under experimental conditions of short-term and long-term adaptation, e.g., fatigue and regeneration, respectively. First, we show that Homer 1a was constitutively expressed and was transiently upregulated during regeneration. In C2C12 cell cultures, Homer 1a was also upregulated during formation of myotubes. No change of Homer 1a was observed in fatigue. Second, Homer 1b/c/d and Homer 2b were positively and linearly related to muscle mass change during regeneration, and third, Homer 3 was not detectable under resting conditions but was transiently expressed during regeneration although with a temporal pattern distinct from that of Homer 1a. Thus a switch in Homer isoforms is associated to muscle differentiation and regeneration. Homers may play a role not only in signal transduction of skeletal muscle, in particular regulation of Ca2+ release from sarcoplasmic reticulum (Ward CW, Feng W, Tu J, Pessah IN, Worley PF, and Schneider MF. Homer protein increases activation of Ca2+ sparks in permeabilized skeletal muscle. J Biol Chem 279: 5781–5787, 2004), but also in adaptation.

Background: We aimed to find out the age-related differences in risk profile among Bangladeshi adults suffering from stable angina.Methods: An observational study was conducted in three different districts in Bangladesh (Feni, Noakhali and Chandpur) through the medical camps during the period from 1st February 2018 to 31st July 2018. Data of 764 stable angina patients was analysed.Results: The male to female ratio in both groups was 4:1. The mean age of the study population in younger group was 34.0±5.2 years and in older group 57.0±6.3 years. Smokers were more in younger group (70.0% vs. 46.0%; p=0.032). Hypertension was less in the younger group (38.0% vs. 58.0%) (p=0.045). Presence of diabetes was higher in the older age group (34.0% vs. 4.0%) (p=0.001). The total cholesterol was higher in older group (182.9±33.1) vs. (171.1±24.8 mg/dl) (p=0.047). 68% of patients of older group and 38% of younger group had stenosis in left anterior descending artery (p=0.003). The involvement of left circumflex and right coronary artery in older age group were higher (56% and 66% respectively) than those in younger group (36% and 40% respectively) (p=0.045 and p=0.009). Patients had multiple risk factors like hypertension, family history of cardiovascular disease, smoking and ischemic heart disease (IHD).Conclusions: This study found that the incidence of IHD is very among the district-level diabetic patients of Bangladesh and this incidence increases with the rise of age.

New components and algorithms were added to represent the unique features of peatlands, such as their characteristic ground floor vegetation (mosses), the slow decomposition of carbon in the water-logged soils and the interaction between the water, energy, and carbon cycles. This paper presents the modifications introduced into the CLASS–CTEM modelling framework together with site-level evaluations of the model performance for simulated water, energy and carbon fluxes at eight different peatland sites. The simulated daily gross primary production (GPP) and ecosystem respiration are well correlated with observations, with values of the Pearson correlation coefficient higher than 0.8 and 0.75 respectively. The simulated mean annual net ecosystem production at the eight test sites is 87 g C m−2 yr−1, which is 22 g C m−2 yr−1 higher than the observed annual mean. The general peatland model compares well with other site-level and regional-level models for peatlands, and is able to represent bogs and fens under a range of climatic and geographical conditions.

Abstract “Cr-Zr-Ca armalcolite” is a mineral originally found in Apollo samples five decades ago. However, no structural information has been obtained for this mineral. In this study, we report a new occurrence of “Cr-Zr-Ca armalcolite” and its associated mineral assemblage in an Mg-suite lithic clast (Clast-20) from the brecciated lunar meteorite Northwest Africa 8182. In this lithic clast, plagioclase (An = 88–91), pyroxene (Mg#[Mg/(Mg+Fe)] = 0.87–0.91) and olivine (Mg# = 0.86–0.87) are the major rock-forming minerals. Armalcolite and “Cr-Zr-Ca armalcolite” are observed with other minor phases including ilmenite, chromite, rutile, fluorapatite, merrillite, monazite, FeNi metal, and Fe-sulfide. Based on 38 oxygen atoms, the chemical formula of “Cr-Zr-Ca armalcolite” is (Ca0.99Na0.01)Σ1.00(Ti14.22Fe2.06Cr2.01 Mg1.20Zr0.54Al0.49Ca0.21Y0.05Mn0.04Ce0.03Si0.03La0.01Nd0.01Dy0.01)Σ20.91O38. Electron backscatter diffraction (EBSD) results reveal that the “Cr-Zr-Ca armalcolite” has a loveringite R3 structure, differing from the armalcolite Bbmm structure. The estimated hexagonal cell parameters a and c of “Cr-Zr-Ca armalcolite” are 10.55 and 20.85 Å, respectively. These structural and compositional features indicate that “Cr-Zr-Ca armalcolite” is loveringite, not belonging to the armalcolite family. Comparison with “Cr-Zr-Ca armalcolite” and loveringite of other occurrences implies that loveringite might be an important carrier of rare earth elements in lunar Mg-suite rocks. The compositional features of plagioclase and mafic silicate minerals in Clast-20 differ from those in other Mg-suite lithic clasts from Apollo samples and lunar meteorites, indicating that Clast-20 represents a new example of diverse lunar Mg-suite lithic clasts.

Objective survey and laboratory analyzes that enabled determination of kidney functions (creatinine clearance, proteinuria, urine specific gravity and osmolality, fractional sodium excretion (FENa), the rate of tubular phosphate reabsorption (TRP), urine N-acetil-D-glycosaminidase and intestinal alkaline phosphatase) were done in all examined persons three times during the 6-month intervals. Results. At the first examination, hypertension was detected in 23 (52%) person, decreased creatinine clearance in two and proteinuria in 10 persons included in the study. In addition, proteinuria and tubular disorders were detected in 6, hypertension, proteinuria and/or tubular disorders in 9 persons. The analysis of the results obtained by three check-ups undertaken during one year showed that proteinuria and tubular disorders appeared intermittently in half of the examined endemic family members. All persons with detected renal disorders required further examination in order to establish accurate diagnosis of renal disease. Conclusion. Three check-ups performed at six-month intervals in the members of five endemic families detected various renal disorders including renal hypofunction. Regular systematic check-ups of endemic families could enable early detection of the disease and early initiation of measures for slowing down chronic renal disease progression.

Abstract:
The objective of this study was to evaluate the influence of flexible work arrangements strategy on job satisfaction among state corporations in Kenya. The greatest challenge facing organisations is whether their staffs are motivated to perform their duties and whether employees find their work interesting and meaningful. The ways jobs are designed has an important bearing on an employee’s job satisfaction. Understanding job design techniques and their influence on job satisfaction will lead organisations to keep the best employee pool for themselves and fend off competition. This study was based on several theories, namely: Job Characteristics model, Hertzberg's two-factor Hygiene theory, Clayton Alderfer’s ERG theory, equity theory, family-boarder theory, and boundary theory. The research design for this study was a cross-sectional descriptive design. This research sought to interrogate the extent to which the state corporations employ flexible work arrangements and its influence on job satisfaction and suggest possible areas of improvement. The target population was 187 state corporations. The unit of analysis was employees of the state corporations in Kenya. The research studied a sample size of 381 employees in 127 state corporations. The state corporations in Kenya were selected using stratified sampling, while employees were purposively selected. To obtain the data, semi-structured questionnaires were used in this research. Data collected were analysed using descriptive and inferential statistics with the aid of the SPSS package. The findings of this study show that a flexible work arrangement is a significant strategy in enhancing job satisfaction among state corporations in Kenya.
