Many species responsible for important damage to agriculture, forestry and stored products belong to the order Lepidoptera. Due to environmental and public health problems in addition to an incoming tolerance derived from the expanded application of pesticides, new alternatives have been developed in order to control these insect pests. Olfaction is crutial for Lepidoptera reproductive success, therefore the disruption of their perception system could be a good management tool. In this thesis, several aspects of the olfactory physiology in different pest species have been studied. One of these studies was the identification of the pheromone blend found in the Spanish strain of the species Spodoptera exigua, where an optimum formulation for its control was obtained using analytical, electrophysiological and behavioural techniques. This study, typical in insects chemical ecology, was complemented with a pioneering analysis of the olfactory proteins expressed in antennae of males and females in different Lepidoptera species. Thanks to different proteomic techniques used in this study such as 2DE, MALDI-TOF MS and MS/MS, a differential expression in an important olfactory group of proteins, like the OBPs, was observed in both sexes. Finally the combination of both kinds of analyses deciphered partially the type of chemical communication existing in the moth species Paysandisia archon, important pest of palm trees, where the females seem to have lost the hability to emit long-distance pheromones probably due to a high adaptation to diurnal life.

Full textDogs in groups (shelters, kennels, laboratory or research facilities) often face suboptimal living conditions that compromise their welfare. In this thesis, the effect of two natural anxiolytic products (a synthetic pheromone and a nutraceutical) on behavioural and physiological (concentration of cortisol in hair) indicators of welfare was studied. It was observed that the exposure to the synthetic pheromone reduced the presence of repetitive behaviours associated with chronic stress: pacing (P<0.0001), standing in rear legs (P= 0.02), as well as barking (P=0.014) and general vocalizations (P<0.0001). In addition, two behaviours related to positive emotions [social interaction (P=0.007) play with dog (P=0.03) or with toy (P<0.0001)] increased. The administration of the nutraceutical increased the frequency of locomotor behaviours: bouncing (P=0.016), jump (P<0.0001), standing (P<0.0045), standing in rear legs (P= 0.0002), sitting (P=0.015), exploring (P=0.0023) and hide (P=0.009) and decreased two oral behaviors associated with negative emotions [yawn (P=0.037) and lip licking (P=0.017)]. Hair cortisol concentration increased during the pheromone application (P<0.0068) and showed no significant changes due the nutraceutical administration P=0.927). The results of both studies show that these natural anxiolytics do have the ability to modify behavior in dogs that live in groups. The results also emphasize the importance of combining behavioural and physiological indicators in studies aimed at assessing animal welfare .

Full textFemale mice display an innate attraction towards male sexual pheromones, which in addition are reinforcing to them, as demonstrated by the induction of conditioned place preference rewarded by male-soiled bedding. Thus, male sexual pheromones provide an original and advantageous model to study reward mechanisms and neural basis. The thesis "Neurochemistry of pheromone-induced reward" aimed to explore what neurotransmitters control innate attraction and place preference acquisition conditioned by male pheromones, as well as the brain nuclei where they are acting. To do so, it was used a pharmachological approach and lesions of brain nuclei upon behavioural protocols involving pheromone-elicited behaviours. Additionally, the results from these experiments were compared with the role of studied neurotransmitters in sweet reward. First, studies involving catecholaminergic systems revealed that dopaminergic agonists, through D1 receptor, inhibit innate attraction towards male pheromones, whereas dopaminergic or noradrenergic antagonists do not affect pheromone reward. Similarly, opioidergic agonists abolish innate attraction towards male pheromones, but this behavoiour and place preference acquisition are not affected by opioidergic antagonists. Quite the opposite, opioidergic antagonists reduce, and agonists do not affect, preference for a sucrose solution in front of plain water. On the other hand, nitric oxide synthesis inhibition blocked innate attraction towards male pheromones, whereas serotonergic drugs use gave rise to inconclusive results. Finally, electrolytic lesions of the zone of the ventral striatum which receives direct projections from the vomeronasal cortex (centred at the medial islands of Calleja) abolished innate attraction as well. Taken together, the results of the thesis reveal that innate attraction behaviour that show female mice for male sexual pheromones depends on vomeronasal projections upon ventral striatum, which are inhibited by dopamine and opioids and facilitated by nitric oxide.

[EN] Mating disruption is the pest control method with better acceptation in market due to its results. This method is based on the releasing of the pheromone of the target insect into the environment with enough quantity to delay or to reduce their mating. Optimization-pheromone studies, which condition the use price of this technique a lot, are based on empirical essays about parameters such as the number of dispensers deployed and their distribution, pheromone initial content, etc. Airborne pheromone quantification is a more direct parameter that offers valuable information to do studies to optimize dispenser available use as to develop new dispensers with less pheromone emission. In this work, a methodology that allows the airborne pheromone quantification in real field conditions has been developed based on volatiles airborne sampling and their later analysis with gas chromatography techniques and mass spectrometry. Using this methodology, the airborne pheromone throughout the crop cycle in mating disruption treated fields against different agronomic important species, the European Grapevine Moth (Lobesia botrana Denis & Schiffermüller) and the Rice Stem Borer (Chilo suppressalis Walker), has been studied in vineyards and rice paddies treated with different types of dispensers. These studies have been useful to know how the pheromone is distributed along the time in relation to the dispensers' kinetics and which are the mechanisms involved in mating disruption in real conditions. Moreover, the climatic dependency of the passive dispensers in field conditions has also been studied as it will be related to the airborne pheromone quantity and it can affect the efficacy of the control method. The results show that the airborne pheromone concentration in mating disruption treated fields against L. botrana changes throughout the crop cycle. In addition, it is observed that canopy produces a buffer effect, so the presence of a developed foliage regulates the concentration of the airborne pheromone and allows a more homogeneous distribution. The correlation of emission rate with climatic parameters were useful to check that temperature and wind speed have an influence on dispensers' emission. On the other hand, with electrophysiological and behavioral bioassays, it was determined that higher airborne concentrations were needed to trigger non-competitive mechanisms in L. botrana males. In this way, it was concluded that competitive mechanisms are the ones acting in treated fields In rice paddies air samplings, airborne pheromone concentration data were used to study why an earlier dispensers' deployment achieved a better effect controlling the pest. Furthermore, it was observed a more direct relationship between airborne pheromone with dispensers' emission. This fact supports the hypothesis that thicker foliage, lesser in rice fields than in vineyards, can prevent pheromone from its washing by wind.

Full textMale mice soiled bedding contains non volatile sexual pheromones that are innately attractive to females. Male volatile signals are, in contrast, not attractive to females in their very first contact with them, but can acquire secondary attractive properties by its association with the non volatile and innately attractive signals. In this work we analysed whether male sexual pheromones are detected through the accessory olfactory system, and if the association between non-volatiles and volatile signals is also dependent on the integrity of vomeronasal system. To do so, we carried out a battery of experiments in a group of females with a lesion in their accessory olfactory bulb (AOB) comparing their behaviour with that from another group of animals with a sham lesion in their AOB. In addition, we also analysed if male sexual pheromones are rewarding to female mice, using a behavioural paradigm named place preference test. Finally, we studied which would be the neural system that mediates these reinforcing properties. To do so, we compared c-fos expression in different vomeronasal nuclei in animals exposed to male-soiled bedding and animals exposed to clean bedding. The results of our experiments show that male sexual pheromones are detected through the vomeronasal organ and that the acquisition of secondary attractive properties by male volatile signals depends on its association with non volatiles signals also detected through the vomeronasal system. Furthermore we have demonstrated that male sexual pheromones have reinforcing properties to female mice, ant that the neural system that mediates these reinforcing properties is not the classical dopaminergic tegmento-estriatal projection, but the projections from some regions of the posteromedial cortical nucleus of the amygdala and the amygdalo-hypocampal area to the most ventromedial Islands of Calleja. The results from these experiments show the existence of a new reinforcing stimulus in mice, the sexual pheromones. In addition, these pheromones are detected through the vomeronasal organ. Moreover, we have shown that there are more regions a part from the classical ones in ventral striatum involved in mediating reinforcing properties of natural stimuli.
