Abstract:
Responses to immunization against somatotropin release inhibiting factor (SRIF) were compared in lambs injected with SRIF plus either Freund's complete adjuvant (FCA), muramyl dipeptide (MDP), Freund's incomplete adjuvant/muramyl dipeptide (FIA/MDP), DEAE-Dextran (DEAE), or Quill A. By comparison with control lambs, immunized lambs grew faster and produced more wool. In lambs immunized with SRIF plus MDP or FIA/MDP, depths of back-fat and of the m. longissirnus dorsi were increased. Anti-SRIF antibody titres were very variable between groups of immunized lambs, with titres being highest for lambs injected with SRIF plus FCA or DEAE. Adverse effects of immunization were observed in lambs given FCA, DEAE and Quill A. It appears from the present study, that MDP alone or in combination with FIA would be suitable for use in commercial vaccines to enhance productivity by immunization against SRIF.

Abstract:
Computer-controlled fluid-flow chemical analysis (CC-FCA) was investigated for the determination of trace amounts of toxic pollutants in the environment. For CC-FCA, automated chemical analysis systems were developed by using computer-controllable pumping and valve modules, and polytetrafluorethylene (PTFE) tubing and connectors. The systems demonstrated in this work were a flow injection-type system, a sequential injection-type system, a mini-column pretreatment system (Auto-Pret system), and an Auto-Pret hyphenated with flow injection analysis (FIA) system. Such systems were fully controlled by a computer program; the lab-made programs were written in Visual Basic. The systems can be hyphenated with some detectors, such as a spectrophotometric detector, an electrochemical detector, electrothermal-atomic absorption spectrometry (ET-AAS), a liquid electrode plasma-atomic emission spectrometry (LEP-AES) and inductively coupled plasma (ICP)-AES. Such systems were successfully applied to the determination of trace amounts of toxic pollutants in environmental water samples: they were heavy metal ions (Pb, Cd, Cr, etc.). In this paper, the author aims mainly at investigating the CC-FCA method for the determination of trace amounts of Cr(VI) in environmental water samples by spectrophotometry. The techniques used in this work were FIA, sequential injection analysis (SIA), and Auto-Pret/FIA, which were all computer-controllable. Limits of detection of Cr(VI) by FIA, SIA, and Auto-Pret/FIA were 8 × 10–9 mol/L (0.4 μg/L), 1.1 × 10–8 mol/L (0.6 μg/L), and 1.4 × 10–9 (0.07 μg/L), respectively. The methods were applied to the determination of Cr(VI) in river and drinking waters.

Abstract:
Non-radiometric immunoassays offer many advantages over radiometric assays, such as higher stability of kit compounds and absence of potential hazardous effects for users and environment. The comparison of cortisol measurements by fluoroimmunoassay (FIA) and fluorometric enzyme immunoassay (FEIA) with radioimmunoassay (RIA) in adrenal function evaluation of normal (n=50) and hypercortisolemic dogs (n=12) was proposed. Serum concentrations of cortisol were measured in basal conditions and 8 hours after dexamethasone (DEX) suppression (0.01mg/kg/IV). All our reference values were based on the 5th and 95th percentile. The values for basal cortisol of healthy dogs were 0.20 to 2.35mug/d<img SRC="http:/img/fbpe/cr/v32n2/a12img01.gif"> for FIA, 0.30 to 5.39mug/d<img SRC="http:/img/fbpe/cr/v32n2/a12img01.gif"> for FEIA, and 0.65 to 4.64mug/d<img SRC="http:/img/fbpe/cr/v32n2/a12img01.gif"> for RIA. In hypercortisolemic dogs, the values of cortisol (mean ± SD) in basal and post-DEX conditions were 2.71 + 0.41mug/d<img SRC="http:/img/fbpe/cr/v32n2/a12img01.gif"> and 1.73 + 1.15mug/d<img SRC="http:/img/fbpe/cr/v32n2/a12img01.gif"> for FIA, 7.05 + 2.85mug/d<img SRC="http:/img/fbpe/cr/v32n2/a12img01.gif"> and 4.93 + 2.26mug/d<img SRC="http:/img/fbpe/cr/v32n2/a12img01.gif"> for FEIA, and 4.80 + 1.43mug/d<img SRC="http:/img/fbpe/cr/v32n2/a12img01.gif"> and 3.52 + 1.08mug/d<img SRC="http:/img/fbpe/cr/v32n2/a12img01.gif"> for RIA. Statistically significant differences (p<0.05) between the normal and the hypercortisolemic groups (Kruskal-Wallis test) were observed in the three methods, and between basal and post-DEX values (Wilcoxon test) using RIA and FEIA methods but not with FIA. Cortisol determinations by FEIA and RIA methods at DEX suppression test showed 100% of sensitivity and specificity for the diagnosis of hyperadrenocorticism in dogs. The results demonstrate that serum cortisol concentrations measurements by FEIA is a suitable alternative to the traditional RIA method for adrenal function evaluation in dogs.

ABSTRACTBackgroundAnimal studies suggest prebiotics can increase iron absorption, but results from human studies are equivocal.ObjectivesIn iron-depleted women, before (baseline) and after daily consumption of galacto-oligosaccharides (GOS) for 4 wk, we sought to assess fractional iron absorption (FIA) from an iron supplement given with and without single doses of GOS in test meals or water.MethodsIn all women (n = 34; median serum ferritin concentration = 16.4 µg/L), FIA from doses of 14 mg iron labeled with stable isotopes was measured in the following conditions at baseline: 1) FIA from ferrous fumarate (FeFum) in water given with and without 15 g GOS; 2) FIA from FeFum in a test meal given with and without 15 g GOS; 3) FIA from ferrous sulfate (FeSO4) in a test meal given without 15 g GOS. All subjects then consumed ∼15 g GOS daily for 4 wk. Then the following conditions were tested: 4) FIA from FeFum in a test meal with and without 15 g GOS; and 5) FIA from FeSO4 in a test meal with 15 g GOS. FIA was measured as erythrocyte incorporation of stable isotopes.ResultsAt baseline, GOS significantly increased FIA from FeFum when given with water (+61%; P < 0.001) and the meal (+28%; P = 0.002). After 4 wk of GOS consumption, GOS again significantly increased FIA from FeFum in the meal (+29%; P = 0.044). However, compared with baseline, consumption of GOS for 4 wk did not significantly enhance absorption from FeFum in the meal given without GOS. FIA from FeSO4 given with GOS in a meal after 4 wk of GOS consumption was not significantly greater than FIA from FeSO4 in a meal without GOS at baseline.ConclusionsIn iron-depleted women, GOS given with FeFum increases FIA, but 4 wk of GOS consumption did not enhance this effect. The study was registered at clinicaltrials.gov as NCT03325270.

"Acute Consumption of Prebiotic Galacto-Oligosaccharides Increases Iron Absorption from Ferrous Fumarate, but not from Ferrous Sulfate and Ferric Pyrophosphate: Stable Iron Isotope Studies in Iron-Depleted Young Women." Abstract:
ABSTRACT Background Although acute consumption of high doses of prebiotic galacto-oligosaccharides (GOS) increases fractional iron absorption (FIA) from ferrous fumarate (FeFum), it is uncertain if low doses of GOS have this effect. Furthermore, whether GOS improve iron absorption from other commonly used iron compounds and whether ascorbic acid (AA) enhances the effect of GOS on iron absorption from FeFum is unclear. Objectives In iron-depleted women [serum ferritin (SF) &lt;30 μg/L], we assessed: 1) whether the acute enhancing effect of GOS on FeFum is dose dependent; 2) if GOS would affect FIA from ferrous sulfate (FeSO4) or ferric pyrophosphate (FePP); and 3) if AA and GOS given together enhance FIA from FeFum to a greater extent compared with GOS alone. Methods We recruited 46 women (mean age 22.0 y, mean BMI 21.3 kg/m2, median SF 17.1 μg/L), and measured FIA from 14 mg iron labeled with stable isotopes in the following conditions: 1) FIA from FeFum given with 3.5 g, 7 g GOS, and without GOS; 2) FIA from FeSO4 and FePP given with and without 15 g GOS; and 3) FIA from FeFum given with 7 g GOS with and without 93 mg AA. FIA was measured as erythrocyte incorporation of stable isotopes after 14 d. Comparisons were made using paired samples t-test or Wilcoxon rank sum test where appropriate. Results Giving 7 g of GOS significantly increased FIA from FeFum (+26%; P = 0.039), whereas 3.5 g GOS did not (P = 0.130). GOS did not significantly increase FIA from FeSO4 (P = 0.998) or FePP (P = 0.059). FIA from FeFum given with GOS and AA was significantly higher compared with FeFum given with GOS alone (+30%; P &lt;0.001). Conclusions In iron-depleted women, GOS does not increase FIA from FeSO4 or FePP, but it increases FIA from FeFum. Thus, a combination of FeFum and GOS may be a well-absorbed formula for iron supplements. The study was registered at clinicaltrials.gov as NCT03762148.

ABSTRACT This resume presents poli[AMOR]fia, a plan of immanence that draws the landscapes of love in education, created in a caosgrafia, which was born from a complex and multireferential posture, and was established at the meeting point between the “tinkering” of Lévi- Strauss, the expansion of Joe Kincheloe, and the geo-philosophy of Deleuze and Guattari. The composition of the landscapes, yet, was woven during meetings with Hannah Arendt and amor mundi; Zygmunt Bauman and contemporary liquidity; Luc Ferry and his new transhumanist revolution; Paulo Freire and his amorous epistemology; Humberto Maturana and the biology of love, in addition to Eros, Philia and Agapé platonic, myths, biblical writings, music, movies, tales, poems and other works of art. This plan, conceptually built, raises the question of the link between love and seemingly crystallized representations and ethical judgments, which tend to impose identities and profiles in teaching. Presenting preliminary landscapes sketched by teachers while they were expressing their views on love in relation to the profession on the interface of virtualization space, Facebook. It´s been traveling through the concepts of subjectivation, territory, molar and molecular segmentation, management, chart, decal and rhizome, to undertake an acentric tinkering, elaborate during methodological tests, deduced from a map guideline- sketch-choreography, the caosgrafia, which as been contemplating the crossing of heterogeneous references about love, which cohabit in contemporaneity and resonate in the field of education. The plan of poli[AMOR]fia expresses a process of pulsatile expansion and contraction, which occurs between flows of permanence and emergency flows and, which causes the creation of multiple forms of conception and expression of love. It is presented in textual and unorderly composites, which emerged from a chaotic writing, which was woven in the perspective of the indissociability of the sensible and the intelligible and, in that respect, from ratio-emotional tensions and intensities. It suggests, without indicating the means, the creation of a new amorous ethic for education, based on pluralistic relations and open to the processes of free subjectivation.

Abstract:
In this work it was developed an automatic procedure for the determination of selenium in ration for domestic animals digested in acidic medium. The automated method was based on the catalytic effect caused by selenium (IV) on the reaction of ammoniacal iron sulfate with EDTA in medium containing NaNO3. The analytical module was based on multicommuted flow process. Considering that the kinetic of reaction was slow, the analytical module was designed with six reaction coupled in parallel in order to allow that residence time of sample zone was increased up to 5 min without decrease of the analytical throughput. After definition of the system control variables, sample of animal ration mineralized in nitric/perchloric acid medium were processed. The results concerning to standard solution addition assays gave recoveries between the range of 95 % up to 109 %. Other profitable features such as a relative standard deviation of 2 % (n = 8) and an analytical throughput of 60 samples determination per hours were also achieved.

Abstract: The present work describes the development of ion-selective electrodes for calcium, potassium, sodium and chloride to be employed in flow injection analyses and its application for the simultaneous determination in samples, as well as, the development of solid contact ionselective electrodes with detection limit in the nanomolar range. The preparation of ion-selective electrodes for calcium, potassium, sodium and chloride was performed optimizing them in steady-state and then incorporated in simultaneous low injection analyses. The optimization of common parameters, such as: pH, buffer, injection volume, flow, etc., was performed looking for the simultaneous determination, using an array of electrodes in series. The proposed potentiometric FIA system was satisfactory for simultaneous determination of these ions in juice samples, and the results, when compared to the reference method, were statistically the same in the 95% of confidence level. In relation to the development of solid contact ion-selective electrodes with low detection limit, four different electrodes were developed for silver, lead, calcium and iodide, with excellent nernstian response and detection limit in the nanomolar range. The obtained detection limits were 2 nmol L for Ag, 1 nmol L for Pb, 4 nmol L for Ca e 10 nmol L for I. It was possible by using the conducting polymer, poly(3-octylthiophene) as an interface between the solid contact and polymeric membrane, methyl metacrylate decyl metacrylate copolymer. The electrodes presented short response time and good repetibility, being potentially useful for determination in trace levels

Abstract: Brazil is one of the world's largest consumers of pesticides. The national agency for health monitoring found, in 2008, that approximately 15% of samples had maximum residue limit (MRL) above the permitted by Law. When used properly pesticides help increase production. But when misused they can be extremely detrimental to the environment and to the population, which has led civil society to demand tighter control from environmental agencies and increased the demand for analytical data. In this work, we developed a scanning method for the determination of organophosphorus pesticides using a FIA procedure associated with an enzymatic biosensor based on acetylcholinesterase. The method is selective for inhibitors of this enzyme and is extremely simple, of low cost and very good. detection limits. For introduction into the FIA system, the solution containing the pesticide should be in aqueous solution but it is not necessary to perform any prior concentration of the analyte. For the pesticide methyl parathion the detection limit is the range of 10 mol L. The correlation coefficient is 0.9917 and the average relative standard deviation is 4.8% (n = 6). For the pesticide chlorpyrifos the detection limit is about 10 mol L, the correlation coefficient is 0.9979 and the average relative standard deviation is 2.3%. For the pesticide malathion the detection limit is in the range of 10 mol L, the correlation coefficient is 0.9902 and the average relative standard deviation is 5.8 (n = 3). For the pesticide dichlorvos the detection limit was about 10 mol L, the correlation coefficient was 0.9921 and the average standard deviation of 3.8 (n = 6)

Abstract:
Abstract Laser microchemical (LMC) technology has become an important element of the FIA and debug tool set by supplying key steps not well addressed by previous tools. In this paper we report the optimization of the LMC technology to solve key issues for flip chip FIA. Specific processes have been developed for localized thinning of flip chips, in order to enable access of conventional FIA tools. Additional applications include dramatic enhancement of focused ion beam (FIB) rework and 3-D micromachining for prototyping, in-situ trimming, and mastering of microelectromechanical systems (MEMS). Laser etching of silicon is with a high pressure chlorine assist and is l000X the rate of the fastest focused ion beam methods. In contrast to grinding methods, the process introduces no process stress or contamination and retains an average surface roughness of several hundred angstroms. Micronthickness metal lines are laid down in a one-step vapor phase deposition at 200 μm/s writing speed. Rapid deposition combined with the superior quality of the laser interconnect, translates into writing with a conductance per unit writing time of 1000 to 10,000 times the rate of a focused ion beam.
