Abstract:
The latex of the common fig (Ficus carica) contains a mixture of at least five cysteine proteases commonly known as ficins (EC 3.4.22.3). Four of these proteases were purified to homogeneity and crystals were obtained in a variety of conditions. The four ficin (iso)forms appear in ten different crystal forms. All diffracted to better than 2.10 Å resolution and for each form at least one crystal form diffracted to 1.60 Å resolution or higher. Ficin (iso)forms B and C share a common crystal form, suggesting close sequence and structural similarity. The latter diffracted to a resolution of 1.20 Å and belonged to space groupP3121 orP3221, with unit-cell parametersa=b= 88.9,c= 55.9 Å.

Abstract:
The manufacture of cheese based on goat's milk is a traditional practice using fig tree latex. The objective of this paper is to study the possibility of substituting rennet with ficin as a vegetable coagulant milk and its characterization. In addition, two trials of manufacturing fresh and soft cheese using ficin were made. The results obtained show that the coagulant activity of ficin is optimal at pH = 5, at a temperature of 75℃ and at a CaCl2 concentration of 0.03 M with 2097.90 UR., 911.79 UR., and 192.30 UR respectively. The coagulant force of this enzyme is 1/21250.911 with a proteolytic activity estimated at 139 μg of tyrosine equivalent per ml of ficin extract. The fresh cheese is white with a creamy appearance and great solubility in the mouth. The ficin soft cheese has an organoleptic quality better than that of control cheese and a softer with a more viscous texture in comparison to that of rennet. The ficin soft cheese has a penetrometry of 2.5 mm in comparison with that of a rennet of 3.6 mm. Ficin can replace rennet in cheese making and deserves to be expanded in the manufacture of different cheeses.

Abstract:
Background and Objectives: Bromelain and ficin are aqueous extracts from fruits of Ananas comosus and Ficus carcia plants, used widely for medical applications. Angiotensin-converting enzyme 2 (ACE2) is a homolog of ACE, degrading Ang II to angiotensin 1-7 and decreasing the cellular concentration of Ang II. Materials and Methods: In this study, we investigated the ACE2-inhibitory, antiproliferative, and apoptosis-inducing effects of ficin and bromelain on caco-2 cells. Results: We found that bromelain and ficin significantly reduced the viability of human colon cancer cells with IC50 value concentrations of 8.8 and 4.2 mg/mL for bromelain after 24 and 48 h treatments, and 8.8 and 4.2 mg/mL for ficin after 24 and 48 h treatments, respectively. The apoptosis of the caco-2 cell line treated with bromelain was 81.04% and 56.70%, observed after 24 and 48 h. Total apoptotic proportions in caco-2 cells treated with ficin after 24 and 48 h were 83.7% and 73.0%. An amount of 1.6 mg/mL of bromelain and ficin treatments on caco-2 cells after 24 h revealed a higher decrease than that of other concentrations in the expression of ACE2 protein. Conclusions: In conclusion, bromelain and ficin can dose-dependently decrease the expression of ACE2 protein in caco-2 cells.

Abstract:
To investigate the effects of ficin on biofilm formation of conditionally cariogenic Streptococcus mutans (S. mutans). Biomass and metabolic activity of biofilm were assessed using crystal violet assay, colony-forming unit (CFU) counting, and MTT assay. Extracellular polysaccharide (EPS) synthesis was displayed by SEM imaging, bacteria/EPS staining, and anthrone method while acid production was revealed by lactic acid assay. Growth curve and live/dead bacterial staining were conducted to monitor bacterial growth state in both planktonic and biofilm form. Total protein and extracellular proteins of S. mutans biofilm were analyzed by protein/bacterial staining and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), severally. qRT-PCR was conducted to detect acid production, acid tolerance, and biofilm formation associated genes. Crystal violet assay, CFU counting, and MTT assay showed that the suppression effect of ficin on S. mutans biofilm formation was concentration dependent. 4 mg/mL ficin had significant inhibitory effect on S. mutans biofilm formation including biomass, metabolic activity, EPS synthesis, and lactic acid production ( p < 0.05 ). The growth curves from 0 mg/mL to 4 mg/mL ficin were aligned with each other. There was no significant difference among different ficin groups in terms of live/dead bacterial staining result ( p > 0.05 ). Protein/bacterial staining outcome indicated that ficin inhibit both total protein and biofilm formation during the biofilm development. There were more relatively small molecular weight protein bands in extracellular proteins of 4 mg/mL ficin group when compared with the control. Generally, ficin could inhibit biofilm formation and reduce cariogenic virulence of S. mutans effectively in vitro; thus, it could be a potential anticaries agent.

Abstract:
Chitosan derivatives – сarboxymethyl chitosan and N-(2-hydroxy)propyl-3-trimethyl ammonium chitosan with molecular weights of 200, 350, and 600 kDa have been synthesized. Complexes of ficin with chitosan and its named derivatives have been obtained. IR spectra of chitosan, carboxymethylchitosan, and N-(2-hydroxy)propyl-3-trimethyl ammonium chitosan and their complexes with ficin were recorded. The analysis of the spectra confirmed the formation of conjugates between the macromolecules of polysaccharides and ficin. The optimal ratio of protein content (0.7 mg/g of carrier) and specific activity (1590 units/mg of protein) was found during the complexation of ficin with the N-(2-hydroxy)propyl-3-trimethylammonium chitosan matrix with the molecular weight 350 kDa. The efficiency of ficin complexation (in terms of specific catalytic activity) with N-(2-hydroxy)propyl-3-trimethyl ammonium chitosan (350 kDa) exceeds that of chitosan (350 kDa) and carboxymethylchitosan (350 kDa) 2.4 and 9.8 times respectively. The types of interactions, first binding energies, amino acid composition of ficin surfaces, which interact with the carrier in the process of complexation, were studied by molecular docking. It has been established that bonds and interactions with chitosan and its derivatives are formed, among other things, with the participation of amino acid residues located near the ficin active site (Cys25 and His162), which explains the change in the proteolytic activity of the obtained complexes. Ficin complexes with N-(2-hydroxy)propyl-3-trimethyl ammonium chitosan are soluble in a wide pH range and, therefore, may be more promising than protease-chitosan complexes in the development of medical preparations and biocatalysts for the food, brewing, and leather industries.

Abstract:

Abstract:

Abstract:
The present work is devoted to research on the interaction between carboxymethyl cellulose sodium salt and its derivatives (graft copolymer of carboxymethyl cellulose sodium salt and N,N-dimethyl aminoethyl methacrylate) with cysteine protease (ficin). The interaction was studied by FTIR and by flexible molecular docking, which have shown the conjugates’ formation with both matrices. The proteolytic activity assay performed with azocasein demonstrated that the specific activities of all immobilized ficin samples are higher in comparison with those of the native enzyme. This is due to the modulation of the conformation of ficin globule and of the enzyme active site by weak physical interactions involving catalytically valuable amino acids. The results obtained can extend the practical use of ficin in biomedicine and biotechnology.

Abstract:
The publication in 1492 of the first Latin translation by Ficino of the Enneads and his monumental accompanying commentary marks the return of Plotinus in the West. Ficino's working manuscript, Par. gr. 1816 of the French Bibliothèque Nationale, offers us exceptional evidence concerning the major philosophical and historical milestones in the elaboration of the Plotinus latinus. The nearly 2,500 annotations written by Ficino in the margins of this manuscript, put at his disposition by Cosimo de' Medici, are here edited, translated and commented upon. This important, previously unpublished exegetical instrument illustrates the various facets of Ficino's work over several decades on Plotinus's very difficult text: through his corrections of the transmitted text – some of these interventions have found their way into contemporary editions though their origin has not always been recognised – in doctrinal confrontation with the profoundly original Platonism of Plotinus, the manuscript reveals a Ficino anxious to integrate the Enneads in his own vision – at once Christian and Platonic – of the concordia philosophorum, whilst at the same time conscious of the sneering and audacity hardly worth saving of the Plotinian text. This Ficinian reading of the Enneads found diverse echoes in Ficino's own work and thus contributed to philosophical debate in the Renaissance and beyond

Abstract:
Proteolytic enzymes from vegetal origin have been largely employed in several studies due to its capacity to cleave antibodies into specific fragments, which for some applications, could be more relevant than the immunoglobulin molecule itself. Beyond that, they could be used directley into a patient playing the role of a \"drug\" (There are examples of enzyme application in studies of several pathologies such as autoimmune, traumatic and neurological diseases), in the aid of clinical techniques for antibodies detection and as tools for serum production. Despite the fact that those enzymes have been of great importance and large applicability, its ease of access and ubiquity in nature, the protease mode of action is still not fully understood. Ficin (found in Fig trees) and Bromelain (found in pineapples) are, for example, enzymes that are being explored in an array of articles and demonstrate the capacity to digest antibodies. In this project, we propose to verify the activity of such enzymes from vegetal origins over the antibodies obtained from equines in the production of immunoreactive fragments of immunoglobulin, lowering the potential risk of animal origin diseases transmission. It is hoped that the fragments generated can contribute to studies of clinical level and the production of proper serum against toxins present in animal poisons. Plasma samples were obtained from horse immunization with a venon \"pool\" from Bothrops snakes at Instituto Butantan. These antibodies were purified using caprilic acid, digested by bromelain or ficin and purified again using chromatographic techniques. The results indicate the formation of an immunoglobulin fragment - F(ab)\'2, however, more experiments are needed to optimize the digestive process in order to improve the reaction yield.

Abstract:
This PhD dissertation is focused on astrology as a discipline during the Italian Renaissance and studies the case of the astrologer, philosopher, theologian and physician Marsilio Ficino (1422-1499). Through the example of Ficino, we study the survival of the ancient astrology derived from Greek and Roman practices. This dissertation proposes a genealogy of astrological humanism and studies the relationships amongst the political theology and the economic theology supported by Ficino. An archaeology of Renaissance humanism is followed by a study of the survival of the ancient model of make sexuality in the Medici culture and we propose a research about the sources of Ficino's astral magic and in order to nuance Frances A. Abstract:
Is it possible to censure music? This question may sound rather extravagant, and this is perhaps why music is often overlooked in studies on ecclesiastical censorship in the early modern period, for it would seem that its very essence is incompatible with any form of control. In fact, the huge transformation the Catholic Church had to face, between the sixteenth and the seventeenth century, had a big impact on the music scene. Unavoidably, music was involved in the disputes between Catholics and Protestants as essential liturgical element. The musicologists have mainly studied the Council of Trent action, which seemed to be animated by the desire to remove the secular textual and melodic components from the church repertory. Therefore, Rome became the main center of production for sacred and spiritual music used in celebrations, devotional practices and also in teaching catechism. The case of the Society of Jesus is exemplar: music represent a powerful means of education in college programs, and a strategic tool in the catechesis work. To the enhancement of spiritual genres corresponds as well the will of ‘suppress’ the profane repertories. This is well documented during the age of Counter Reformation, by the variety of cases of ‘‘travestimento’’ which invest canzonette and madrigals. This attitude was also proved by some of the measures the ecclesiastical censure adopted against the vocal production since the seventies of the sixteenth century. In ancient Greek musical theory, harmony was seen as being endowed with a natural virtue capable of altering the rational faculties of the listener’s soul, to the extent of depriving that person of his freedom. The many ethical implications of this classical axiom featured in early modern theological debates. My thesis tries to provide a first answer to these questions by studying the conciliar decrees, the documents of the Index Congregation, the treatises on music and the manuals on demonology. These sources reveal musical censorship did not only involve zealous inquisitors battling with some licentious musician, but also the language of worship and the circulation of prohibited knowledge, which included dangers in the form of sounds that went beyond erotic seduction

Abstract:
Peptones are protein hydrolysis products consisting of a mixture of free amino acids, oligopeptides, and other fragmented protein components easily digestible by microorganisms. This study aimed to investigate the effect of crude ficin enzyme on the characteristics of peptones derived from yellowfin tuna (Thunnus albacares) dark meat and their potential application in microbial growth. Crude ficin enzyme concentrations used to produce peptones from yellowfin tuna dark meat were 0%, 0.1%, 0.2%, 0.3%, and 0.4%. The results showed that using crude ficin enzyme impacted the characteristics of peptones from yellowfin tuna dark meat, including protein content, amino nitrogen, degree of hydrolysis, and optical density. The research findings revealed protein content of 16.20%-32.51%, amino nitrogen of 2.74%-6.53%, hydrolysis degree of 28.37%-39.16%, and optical density value of 0.342-0.574. The optimal concentration of crude ficin enzyme, based on the results obtained in this study, 0.2% crude ficin enzyme was used for producing peptone from yellowfin tuna (Thunnus albacares) dark meat. .

"MAPPING OF FUNCTIONAL DOMAINS OF HUMAN HIGH MOLECULAR WEIGHT (HMW) AND LOW MOLECULAR WEIGHT (LMW) KININOGENS BY USING MURINE MONOCLONAL ANTIBODIES (MAbs)." In XIth International Congress on Thrombosis and Haemostasis. Schattauer GmbH, 1987. http://dx.doi.org/10.1055/s-0038-1642849.

Full text
Abstract:
It has been widely known that HMW and LMW kininogens are the large potential sources of kinin in human blood, and that HMW kininogen also functions as a cofactor in the contact activation of blood coagulation. Recently, it has been demonstrated that the heavy chains of kininogens strongly inhibited a number of cysteine proteinases such as calpains, cathepsins, papain and ficin. We made an attempt at mapping of functional domains on the molecules of both kininogens by using MAbs.Thirty four MAbs raised against human HMW and LMW kininogens were screened by ELISA. By using HMW kininogen, kinin-free HMW kininogen, kinin and fragment 1.2 (fr 1.2)-free HMW kininogen, LMW kininogen, kinin-free LMW kininogen, heavy chain of LMW kininogen and light chains of both kininogens, the MAbs were characterized and.classified into four groups; [1] 20 MAbs reacted with heavy chain, a common region of HMW and LMW kininogens. These MAbs possessed the specificity for domain 1 (2 MAbs), domain 2 (2 MAbs), domain 3 (7 MAbs), and both domains 2 and 3 (7 MAbs) of the heavy chain; [2] 7 MAbs recognized the fr 1.2, a unique histidine-rich region; [3] 5 MAbs reacted with the light chain of HMW kininogen; [4] 2 MAbs recognized the light chain of LMW kininogen. Two MAbs, designated HKG H7 and H12, effectively inhibited the cysteine proteinase inhibitor activity of HMW and LMW kininogens and the others did not affect it. Further, the MAbs, which recognized the fr 1.2 or light chain of HMW kininogen, suppressed the clotting activity. Especially, 2 MAbs, named HKG L2 and L5, effectively suppressed the clotting activity of HMW kininogen. The former, which neutralized about 70% of the clotting activity, reacted specifically with fr 1.2 region of HMW kininogen, and the latter, which neutralized more than 90% of it, recognized the light chain of HMW kininogen. In the results of competition ELISA, fr 1.2 specific MAbs could be classified into 5 kinds of MAbs for recognition sites, and the light chain (HMW kininogen)-specific MAbs also could be classified into 3 kinds of MAbs. Further, 2 light chain (LMW kininogen)-specific MAbs were thought to recognize an identical antigenic site.

Abstract:
The issues of comfortable human existence in the Arctic region arise in the context of growing interest in the Arctic, active development of its resources, with geopolitical problems at once. The external visual environment is essential, which is lacking in color and detail. The vast expanses of snow, the absence of visible elements, and the colorless palette (from white to blue) form a monotonous, uncomfortable field of view. The mechanisms of visual perception do not work correctly and can lead to the development of stressful states in these conditions. The duration of work shifts can be a cause of chronic stress. We studied the features of eye-tracking and the autonomic nervous system's response in the visual perception of homogeneous and comfortable images. Images according to the classification of V. A. Filin, photos of shift complexes, a constant urban settlement in the Arctic region, created a visual environment of different comfort (homogeneous and comfortable). Activation of the sympathetic nervous system was registered when viewing an uncomfortable visual space. Significant changes in the static and dynamic parameters of eye-tracking were detected, confirming the development of a stress reaction in the body.
