Abstract:
ABSTRACT Type 1 fimbriae have been suggested to play a role in the pathogenesis of extraintestinal Escherichia coli infection. Type 1 fimbriation in E. coli is phase variable and known to be dependent upon FimB and FimE, the two recombinases that invert the molecular switch fimS and control the expression of the downstream fim operon. Here we showed that HbiF, a novel site-specific recombinase, inverted fimS independently of FimB and FimE. HbiF-mediated fimS inversion appeared to be predominantly switching from “off” (termed OFF) to “on” (termed ON) orientation. This is different from the fimS inversion mediated by either FimB (bidirectional ON to OFF and OFF to ON) or FimE (unidirectional ON to OFF). Constitutive expression of the hbiF gene in E. coli resulted in a fimS-locked-ON phenotype, which resulted in the pathogenic E. coli K1 strain being incapable of inducing a high degree of bacteremia in neonatal rats. Discovery of HbiF-mediated OFF-to-ON fimS switching provides a new opportunity to develop a strategy for the prevention and therapy of extraintestinal E. coli infection including bacteremia and meningitis.

Abstract:

Abstract:

Abstract:
This article investigates the link between the Roman notion of fides and the contemporary notion of fiduciary duties. Etymologically, the word “fiduciary” derives from fides. The Roman fides was a very complex concept, blending religious, social, and legal valences. The religious and social fides entered Roman law in a substantive form, as bona fides, and as a standard of judgment, in the form of bonus vir. It is submitted that a close analogy can be drawn between bonus vir and the contemporary fiduciary standards.

Abstract:
The Protein Data Bank (PDB) was established at Brookhaven National Laboratories in 1971 as an archive for biological macromolecular crystal structures. In mid 2021, the database has almost 180,000 structures solved by X-ray crystallography, nuclear magnetic resonance, cryo-electron microscopy, and other methods. Many proteins have been studied under different conditions, including binding partners such as ligands, nucleic acids, or other proteins; mutations, and post-translational modifications, thus enabling extensive comparative structure-function studies. However, these studies are made more difficult because authors are allowed by the PDB to number the amino acids in each protein sequence in any manner they wish. This results in the same protein being numbered differently in the available PDB entries. For instance, some authors may include N-terminal signal peptides or the N-terminal methionine in the sequence numbering and others may not. In addition to the coordinates, there are many fields that contain structural and functional information regarding specific residues numbered according to the author. Here we provide a webserver and Python3 application that fixes the PDB sequence numbering problem by replacing the author numbering with numbering derived from the corresponding UniProt sequences. We obtain this correspondence from the SIFTS database from PDBe. The server and program can take a list of PDB entries or a list of UniProt identifiers (e.g., “P04637” or “P53_HUMAN”) and provide renumbered files in mmCIF format and the legacy PDB format for both asymmetric unit files and biological assembly files provided by PDBe.

Abstract:
The background in this study is the rise of cases of forest fires due to the element of deliberate humanity who deliberately burned the forest for personal interests to open new land in Kayuagung District, Ogan Komering Ilir Regency. The formulation of the problem in this study are 1) How is criminal law enforcement against the perpetrators of forest fires in Celikah Village, Kayuagung District, Ogan Komering Ilir Regency ?; and 2) What are the obstacles encountered in the enforcement of criminal law against the perpetrators of forest fires in Celikah Village, Kayuagung District, Ogan Komering Ilir Regency ?. The research method used is empirical research. Data sources used in this study consisted of primary data and secondary data. Based on the results of the study showed that 1) Criminal law enforcement against the perpetrators of forest fires in the village of Celikah, Kayuagung District, Ogan Komering Ilir District has gone through a penal effort that is repressive measures of Investigation because the perpetrators threatened with imprisonment in accordance with criminal theory in applying the principle of subsidiarity and in accordance with article 99 paragraph (1) Article 108 jo Article 69 letter h Law of the Republic of Indonesia No. 32 of 2009 concerning Environmental Protection and Management and regulates criminal sanctions alternatively, namely in the form of imprisonment or only fines and 2) Constraints faced by law enforcement agencies in carrying out criminal law enforcement against perpetrators of forest and land burning, namely first, at the level of investigation, constraints faced is the limited budget support for the management of smoke disasters, the tradition of the community opening land by burning, not yet optimized community empowerment to care about smoke disasters, and the limitations of environmental expert witnesses. Second, at the level of the public prosecutor and judge, the obstacles faced are that the police have never been involved at the level of investigation, the lack of certified public guides and judges (expertise) in the field of environment especially forest and land fires, users of tools that are not of Indonesian national standards.

Abstract:
Today, many wetlands have been converted into agricultural, industrial, and residential areas. This conversion of land functions creates new problems for the environment and human, one of which is land fires. The results of land fires can also cause other problems that results in losses in various fields, such as human health, the economy, and other fields. The emergence of smog as a result of land fires can cause respiratory problems, disruption of the transportation system, conflicts between neighboring countries, and the others. South Sumatra Province is one of the largest contributors to the haze as a result of land fires, especially its wetlands. Ogan Ilir Regency is one of the areas in South Sumatra Province which has extensive wetlands and fires frequently occur. Fires are caused by triggering factors, one of which is natural factors such as a prolonged dry season, availability of water supply, and intensity of rainfall. This study is an analytic descriptive study that aimed to provide an overview of environmental conditions on land, the majority of which are wetlands which experienced fires in Ogan Ilir Regency in 2019. The environmental conditions studied included soil and vegetation types. The data obtained will be displayed through tables and graphs, then interpreted and analyzed descriptively. The type of data used is secondary data in the form of a report by the Regional Disaster Management Agency Ogan Ilir Regency related to land fires in OI Regency and processed using the Geographic Information System (GIS) application. The results of the analysis show that the area of land fires that mostly occurred in North Indralaya District was 382,7 hectares with a total of 144 hotspots. The burnt area was dominated by peat soil (53%) and scrub vegetation (43%).

Abstract:
The use of English template files in workflows involving the multilanguage creation of subtitles from the same source audio assets, typically English, was one of the greatest innovations in the subtitling industry at the turn of the century. It streamlined processes, eliminated duplication of work, reduced direct costs, improved timelines and facilitated the quality control of large volumes of subtitle files whilst expanding the pool of available translators to complete the work. Template files set the basis for the globalisation of the subtitling industry and, almost two decades since their inception, they are still a topic of debate among language service providers and subtitlers. The present paper is a descriptive work, presenting a set of guidelines devised by the present author at the turn of the century, and used in practice for almost a decade in the production of multilingual subtitling work, enhanced and improved upon by the very subtitlers that worked with them. It aims at providing a reference point for debate to researchers in the subtitling field, so as to further the ongoing discussion on interlingual subtitling quality, practices and standards.
