The soybean (Glycine max L. Max) is one of the main crops grown in Brazil, the second largest world producer. Its evolution has been strongly supported by the development of technologies that allowed the acreage increase. Currently, the use of biotechnology tools aims to contribute to the development of new genotypes with characteristics that can continue increase the production. The identification of genes responsible for characters that can assist in developing varieties with improved seed quality may constitute something innovative. The seed tegument has an important function as for the seeds resistance to deterioration. In this sense, the partial characterization of the protein expression FLA 14 (Arabinogalactan), directly related to the cell wall synthesis in the seed tegument may contribute to the advancement of these studies. The aim of this work was to generate and validate primers capable of quantifying the gene expression of the FLA 14 protein in four soybean genotypes, two with black tegument (TP and IAC 222) and higher lignin content and two with yellow tegument (CD 202 and Potência) and lower lignin content. The plant tissue (tegument) was collected 50 days after the anthesis. In order to obtain the gene expression among genotypes, it was used the qRT-PCR technique, where it was tested four combinations of primers, constructed from sequence models of soybeans ESTs. With the validation results, the primers that were found within the standards were the FLA 14p1, FLA 14p2 and β-Actina. They were all used at the expression studies. The results of the relative expression showed that soybean genotypes with black tegument and higher lignin content express the FLA 14 protein quantitatively superior to the genotypes with yellow tegument and lower lignin content.
A soja (Glycine max L. Merr.) é uma das principais culturas produzidas no Brasil, sendo considerado o segundo maior produtor mundial. Sua evolução foi fortemente amparada pelo desenvolvimento de tecnologias que possibilitaram o aumento da área cultivada. Atualmente o emprego de ferramentas da biotecnologia busca contribuir para o desenvolvimento de novos genótipos com características que possam continuar incrementando a produção. A identificação de genes responsáveis por caracteres que possam auxiliar no desenvolvimento de variedades com melhor qualidade da semente, pode se constituir em algo inovador. O tegumento apresenta uma função importante quanto à resistência das sementes à deterioração. Nesse sentido, a caracterização parcial da expressão do gene da proteína FLA 14 (Arabinogalactano), relacionadas diretamente com a síntese da parede celular no tegumento da semente pode contribuir para o avanço dos estudos. O objetivo do trabalho foi gerar e validar primers capazes de quantificar a expressão gênica da proteína FLA 14 em quatro genótipos de soja, dois desses com tegumento preto (TP e IAC 222) e maior conteúdo de lignina, e outros dois com tegumento amarelo (CD 202 e Potência) e menor conteúdo de lignina. O tecido vegetal (tegumentos) foi coletado aos 50 dias após a antese. Para a análise da expressão gênica entre os genótipos, utilizou-se a técnica de qRT-PCR, onde foram testadas quatro combinações de primers construídos a partir de modelos de sequência de ESTs da soja. Com os resultados da validação, os primers que se encontraram dentro dos padrões foram os FLA 14p1, FLA 14p2 e β-Actina utilizando-se no estudo de expressão. Os resultados da expressão relativa permitiram concluir a expressão do gene da proteína FLA 14 é quantitativamente superior nos genótipos de soja com tegumento preto e conteúdo superior de lignina em relação aos genótipos com tegumento amarelo e menos teor de lignina.

Ce travail de thèse s'intéresse aux processus d'appropriation spatiale à travers l'étude des Cubano-américains et de leurs ancrages dans l'agglomération de Miami depuis la révolution cubaine. Dans une démarche entre géographies politique, culturelle et sociale, cette thèse cherche à démontrer comment la trajectoire de Miami est aujourd'hui indissociable de celle des Cubains qui ont gagné la ville. Minorité devenue majoritaire, les Cubano-américains ont bénéficié d'un contexte géopolitique particulier (la guerre froide) pour s'implanter et développer Miami. À différentes échelles, cette étude analyse les relations de pouvoir et les modalités de circulation qui font naître une multitude de territoires en perpétuelles renégociations dans cette ville en chantier. En abordant Miami comme un carrefour américain, cette thèse explore les processus en cours qui « produisent l'espace » de la ville mais font également de celle-ci le théâtre d'une « troisième Amérique » qui se forme et se transforme au gré de créolisations.

The examination and especially the celebration of image and style--of simulation--throughout recent cultural debate is incisively expressed through the framework of popular culture. Le but de cette thèse était d’identifier les mécanismes moléculaires responsables des propriétés particulières de la couche G et les propriétés mécaniques remarquables du bois de tension (BT). Trois acteurs moléculaires potentiels (des protéines à arabinogalactane avec domaine fasciclin-like (FLA), une protéine chitinase-like (CTL) et une β-galactosidase (BGAL)) ont été choisis et étudiés dans: analyse phylogénétique, analyses d'expression et caractérisation de peupliers transgéniques affectés dans l’expression de chacun de ces acteurs. La caractérisation fine de ce matériel a révélé que CTL2 et les FLA jouent un rôle dans la régulation de la cristallinité de la cellulose dans le BT. CTL2 apparaît également important dans l'organisation de la paroi cellulaire et des propriétés mécaniques des tiges. BGAL a été avant proposé pour une fonction dans modification de pectine RG-I potentiellement important pour des propriétés mécaniques de BT. Le bois de tension exhibe une activité BGAL plus élevée que dans le bois opposé. L’inhibition par RNAi de l’expression de BGAL7, spécifiquement exprimée dans le BT, n’est pas responsable à lui seul de la forte activité BGAL présente dans le BT. En contrepoint à l’étude menée sur le peuplier, nous avons également évalué la présence d’acteurs moléculaires potentiellement responsables des propriétés mécaniques du BT chez le simarouba qui développe dans leur BT des fibres ayant leurs sous-couches de la paroi intermédiaire entre la G et la S2. Des protéines à arabinogalactanes ainsi que des pectines du type RG-I sont présentes dans les fibres de BT de peuplier et de simarouba et pourraient avoir une fonction dans un mécanisme commun de génération des contraintes dans le BT. Finalement, un modèle est proposé sur le rôle présumé des différents acteurs moléculaires étudié dans la régulation des propriétés de la couche G et la génération des fortes contraintes du bois de tension
The aim of this thesis was to approach the underlying molecular mechanisms responsible for the particular properties of the G-layer and the outstanding mechanical properties of tension wood (TW). Accordingly, three potential molecular players (fasciclin-like arabinogalactan protein (FLA), chitinase-like protein (CTL) and β-galactosidase (BGAL)) were chosen and studied through a phylogenetic analysis, expression analyses and most importantly characterization of RNAi transgenic poplars. This multilevel characterization revealed that CTL2 and FLAs have function in the regulation of cellulose crystallinity in TW. CTL2 was also shown to be important both for the cell wall organization and stem mechanical properties. BGAL was studied in a light of the previously reported modifications of RG-I pectin, potentially important for the mechanical properties of TW. Study of BGAL revealed that the enzyme has higher activity in TW than in opposite wood. BGAL7, whose gene was expressed specifically in TW, does not seem to be responsible for the higher BGAL activity in TW. In comparison to poplar, we analyzed the occurrence of molecular players potentially responsible for TW mechanical properties in simarouba, a tropical species, which develops different TW fiber. Arabinogalactan proteins and RG-I pectin potentially targeted by BGAL were localized in TW fibers both in poplar and simarouba and therefore may be involved in a common mechanism of tensile stress generation in different TW types. A model was finally proposed to elucidate a potential function of the studied molecular players in the regulation of G-layer properties and tensile stress generation

Abstract:
This study attempted to locate some possible sources of oral Foreign Language Anxiety (FLA) among Japanese students in the United States. This study proposed that the following three factors were possible sources of FLA: 1) the subjects' traitlike anxiety, which is carried by individuals across all communication-bound contexts; 2) the subjects' self-perceived oral proficiency levels in English and 3) the subjects' gapsize (i.e., the distance between their self-perceived and their self-expected oral proficiency levels in English) . This research examined whether the above three independent variables and the dependent FLA variable were significantly correlated, and if so, which one had the strongest correlation with the FLA variable. Also, whether the subjects' biographical variables had a significant effect on their FLA levels was investigated. All the variables were quantified through a questionnaire. The subjects' FLA levels and traitlike anxiety levels were measured by a 10-item, Personal Report of Communication Apprehension inventory (PRCA, Mccroskey, 1978). The subjects' self-perceived oral proficiency levels were measured by asking the subjects to rate their self-perceived oral proficiency level from 1 (poor) to 5 (fluent). The gapsize was quantified by asking the subjects to rate it on a scale from 1 (minimal) to 5 (maximal). The statistical methodology used in obtaining the PRCA scores in this study differed from McCroskey's in its interpretation of Likert type scales. The scales were treated as interval data in McCroskey's study, while, in this study, they were interpreted as ordinal data. After hierarchically ordering the subjects' answers, non-parametric tests were performed on them. Overall, each of the three variables and the FLA variable were found to be significantly correlated at p < .01. The traitlike anxiety variable, the proficiency variable and the gapsize variable correlated at .46, -.45 and -.33, respectively. The participants' demographic variables (age, gender, status at school or year(s) of residence in English speaking places) did not have a significant effect on their FLA levels. A discussion of the results was provided, with references to previous studies.

The development and birth of the Orange County Regional History Center is perhaps unlike any other local history museum in the United States. Its story is worth telling because of its long gestation, the difficulties in bringing this center to life, and the goals of the people who made it possible. All of these elements are a vital part of the history of Orange County, Florida and should not be overlooked. In this light, this thesis will discuss more fully three topics in relation to the creation of the new History Center. First, it will look at the American museum field and its role as a community-building enterprise in American society. Second, it will examine the history of Central Florida, a region that sorely lacks a strong sense of community. Third, it will trace the transformation of the Orange County Historical Society and Museum from a small volunteer-run museum into the Orange County Regional History Center, the largest history museum in Central Florida. The ultimate goal of the History Center is to foster a sense of community in a region with little feeling of connectedness. This community building goal has evolved from both a renewed emphasis on community service in the museum field, and the desire of the History Center's leaders and staff to serve more effectively the Central Florida region. My thesis tells the story of the history and development of the Regional History Center, an institution dedicated to bringing the diverse community of Central Florida together for discussion, dialogue, and reflection. It also identifies some of the new roles and functions it must assume in the future and the new tasks that await it as it strives to become more useful and relevant to its community. In that way, and through that resource, the institution can help build the foundation for a more promising future for present and future residents of Orange County.
