FD-1 capable of using 4-fluoroaniline (4-FA) as the sole carbon and nitrogen source was investigated for its ability to utilize 4-FA isomers (2-FA, 3-FA) and homologs (2,4-DFA, 3,4-DFA, and 2,3,4-TFA). Both 4-FA and 3-FA could be mineralized as the sole carbon and nitrogen source by FD-1. 2-FA, 2,4-DFA, 3,4-DFA, and 2,3,4-TFA could not be degraded by FD-1, respectively, and were selected as secondary substrates for cometabolism with 500 mg/L 4-FA as growth substrate. A group of 4-[1-(2-deoxy-β-D-ribofuranosyl)]-derivatives of 5-fluoroaniline possessing a variety of aryl C-2 substituents (6a R = H, 6b R = F, 6c R = Me) were synthesized. Accordingly, a Heck-type coupling reaction of the 4-iodoaniline derivatives (13a–c) with the bis(tert-butyldimethylsilyl)glycal (11) in the presence of Pd(OAc)2 and Ph3As, followed by removal of the tert-butyldimethylsilyl protection groups using n-Bu4N+F-, yielded the corresponding 4-(β-D-glycero-pentofuran-3-ulos-1-yl)aniline derivatives (14a–c) having a C-3 C=O in the sugar ring. Reactions of fluorinated anilines with stoichiometric Ti(NMe2)4 in mesitylene (typically for 23 h at 120 °C) afforded moderate to high yields of the corresponding N,N-dimethyl-o-phenylenediamine derivatives resulting from defluoroamination of a fluorine atom ortho to the NH2 of the starting aniline. Reactivity increased with additional ring fluorination in general accordance with established regiochemical (activating and deactivating) trends. Within our experimental detection limit, the measured values are the same for both of the 35Cl and 37Cl isotopologues of 3C4FA.
