In neuroscience one of the open problems is the creation of the alpha rhythm detected by the electroencephalogram (EEG). One hypothesis is that the alpha rhythm is created by the inhibitory neurons only. The mesoscopic approach to understand the brain is the most appropriate to mathematically modelize the EEG records of the human scalp. 碩士<br>國立清華大學<br>分子與細胞生物研究所<br>97<br>幽門螺旋菌為一重要的人類致病菌。自從被發現以來已經確認造成包括胃炎、十二指腸潰瘍甚至胃癌在內的胃部疾病有關。目前普遍的三合一療法是以兩種抗生素搭配一種質子幫浦抑制劑同時使用。然而過度的使用抗生素通常會增加幽門螺旋桿菌抗藥性菌株的發生而使得治療失敗，因此發展新型的抗生素用以治療幽門螺旋桿菌在未來將是一門重要的挑戰。shikimate路徑對於芳香族胺基酸的代謝十分重要，由於shikimate路徑的酵素廣泛存在於原核生物及植物細胞中，但在哺乳動物細胞此代謝路徑中不存在，因此常被認為是具有潛力開發成新一代抗生素的標靶酵素。Shikimate路徑中催化第二步關鍵反應的酵素dehydroquinate synthase (DHQS)能將3-deoxy-D-arabino-heptulosoe-7-phosphate (DAHP)催化生成3-dehydroquinate (DHQ)。幽門螺旋菌的DHQS的apo-form X光晶體結構為一open-form 構型，透過結構上的分析，顯示了當HpDHQS一旦與ligand結合後也有可能以close-form的構型存在。利用以結構為基礎的電腦對位虛擬篩選(docking virtual screening)技術，發展出具有良好抑制效果的抑制劑。但在虛擬篩選先導化合物(lead)的
