Abstract:

This master´s thesis deals with calibration and determination of uncertainties of measurement for focus variation microscope Alicona Infinite Focus G4. Together with calibration, a measurement of chosen parameters with calculation of measurement uncertainties of a component was realised. The thesis also describes the whole calibration process and terminology used in calibration, as well as the calibrated instrument, it´s parameters and usage in practice. The measurements were realized on premises of Intemac Solutions s.r.o. Obtained data were than processed using MS Excel, Gwyddion and TalyMap software. At the end of the thesis, practical recommendations are formulated.

Побудовані математичні моделі та структурні схеми систем програмного управління з упередженням як для об'єктів з зосередженими параметрами, так і для об'єктів з розподіленими параметрами, які мають властивості лінійного об'єкту з самовирівнюванням. Моделі мають представлення у вигляді рекурентної формули, де враховується перехідні процеси у тепловому об'єкті за допомогою масиву коефіцієнтів відповідності. The mathematical model of the system of programmatic management is built for thermal objects which are characteristics of linear object with smoothing. A model knows as a recurrent formula, where transients are taken into account in a thermal object by the array of coefficients of accordance. A model is built for objects with a few sensors and a few sources of heat.

Abstract:

Bartonella spp. are emerging bacterial pathogens which continue to achieve an ever-increasing level of clinical importance and awareness. As Bartonella infections are common zoonoses in the U.S., the existence of Bartonella infection in Sweden, which had not been documented prior to the onset of our investigations, was of considerable interest. Initially, serological parameters of Bartonella infection were evaluated using a series of immunoblot analyses in an effort to elucidate the human humoral immunity induced by this pathogen. An immunodominant epitope of B. henselae was identified, designated Bh83, which was hypothesized to be a conserved antigen during Bartonella infection. Further, an in-depth characterization of the immunoglobulin (Ig) isotype and IgG subclass response revealed a primary role of IgA and IgG1 in the immune induction during Bartonella infection. As the presence and degree of exposure to Bartonella spp. in Sweden was previously unknown, several studies were initiated to evaluate the level of seroreactivity to Bartonella spp. antigen in various subgroups of the Swedish human and animal population using indirect fluorescent-antibody assay (IFA). A preliminary study of a select group of patients and blood donors represented the first documentation of Bartonella exposure in Sweden, as well as identified the first two cases of B. quintana-like infections in Sweden. To assess the endemic level of antibodies to Bartonella spp. in Sweden, a large sampling of blood donor sera collected nationwide were subsequently analyzed for reactivity to six Bartonella strains. The largest seroprevalence was to B. elizabethae, with relatively low baseline levels of reactivity to the other Bartonella antigens tested. Correlated risk factors for infection included working outdoors and contact with cats, among others. Further studies of sera obtained from elite orienteers and intravenous drug users in Sweden revealed a high level of exposure to Bartonella spp. in such cohorts, predominantly to B. elizabethae antigen. Consistent with the aforementioned findings of elevated titers to B. elizabethae in humans, a high seroprevalence was found in cats, a known reservoir for Bartonella spp., as well. With relatively little known of the pathogenicity and epidemiology of Bartonella infection, these studies provide insight into various aspects of the distribution, risk factors for infection, specific humoral immune response to infection, and the prevalence of Bartonella spp. in human and cat populations in Sweden and serve as a framework for future studies.

Abstract:

Additive manufacturing (AM) is the name given to the technology of building 3D parts by adding layer-by-layer of materials, including metals, plastics, concrete, etc. Of the different types of AM techniques, electron beam melting (EBM), as a powder bed fusion technology, has been used in this study. EBM is used to build parts by melting metallic powders by using a highly intense electron beam as the energy source. Compared to a conventional process, EBM offers enhanced efficiency for the production of customized and specific parts in aerospace, space, and medical fields. In addition, the EBM process is used to produce complex parts for which other technologies would be either expensive or difficult to apply. This thesis has been divided into three sections, starting from a wider window and proceeding to a smaller one. The first section reveals how the position-related parameters (distance between samples, height from build plate, and sample location on build plate) can affect the microstructural characteristics. It has been found that the gap between the samples and the height from the build plate can have significant effects on the defect content and niobium-rich phase fraction. In the second section, through a deeper investigation, the behavior of Alloy 718 during the EBM process as a function of different geometry-related parameters is examined by building single tracks adjacent to each other (track-by-track) andsingle-wall samples (single tracks on top of each other). In this section, the main focus is to understand the effect of successive thermal cycling on microstructural evolution. In the final section, the correlations between the main machine-related parameters (scanning speed, beam current, and focus offset) and the geometrical (melt pool width, track height, re-melted depth, and contact angle) and microstructural (grain structure, niobium-rich phase fraction, and primary dendrite arm spacing) characteristics of a single track of Alloy 718 have been investigated. It has been found that the most influential machine-related parameters are scanning speed and beam current, which have significant effects on the geometry and the microstructure of the single-melted tracks.

Abstract:

This thesis contributes to influence analysis in nonlinear regression and in particular the detection of influential observations. The focus is on a regression model with a known mean function, which is nonlinear in its parameters and where the function is chosen according to the knowledge about the process generating the data. The error term in the regression model is assumed to be additive. The main goal of this thesis is to work out diagnostic measures for assessing the influence of observations on various results from a nonlinear regression analysis. The obtained results comprise diagnostic tools for detecting observations that, individually or jointly with some other observations, are influential on the parameter estimates. Moreover, assessing conditional influence, i.e. the influence of an observation conditional on the deletion of another observation, is of interest. This can help to identify influential observations which could be missed due to complex relationships among the observations. Novelties of the proposed diagnostic tools include the possibility to assess influence of observations on a specific parameter estimate and to assess influence of multiple observations. A further emphasis of this thesis is on the observations' influence on the outcome of a hypothesis testing procedure based on Rao's score test. An innovative solution to the problem of visual identification of influential observations regarding the score test statistic obtained in this thesis is the so called added parameter plot. As a complement to the added parameter plot, new diagnostic measures are derived for assessing the influence of single and multiple observations on the score test statistic.

Abstract:

碩士 逢甲大學 機械與電腦輔助工程學系 107 Fresnel lens is one optical device. It has the same curvature of traditional lens. But it removes unnecessary parts of lens. It is often used in optical system for its less cost, lighter mass, and smaller focal length. In this work, effect geometric parameters of Fresnel lens on imaging and focusing is investigated and discussed. The trenches directions of Fresnel lenses would affect the focusing phenomena. Trenches outward would have better focusing effect and smaller optical aberration due to two refractions. However, trenches inward would have advantage of smaller focal length. It is found that draft angle has no significant effect on focusing and imaging. However, from simulation, the trenches direction should be considered in the design of the draft angle. For Fresnel lens design, draft angle of 5° for trenches outward and 2° for trenches inward have better performance. By observation of light trajectory, it is found that the number of rings has less effect on illumination. But it would induce optical aberration for ring number less than 15. The ring number had better be more than 15.

Relatório de Estágio do Mestrado Integrado em Ciências Farmacêuticas apresentado à Faculdade de Farmácia O presente documento é constituído pelo Relatório de Estágio em Farmácia Comunitária e pela Monografia intitulada "Desenvolvimento de comprimidos de dupla camada através duma estratégia de Qualidade Programada: foco nos parâmetros de fabrico"Sendo parte do plano curricular do último semestre do Mestrado Integrado em Ciências Farmacêuticas, o Estágio em Farmácia Comunitária decorreu na Farmácia Estádio, em Coimbra, entre janeiro e setembro de 2021 e é apresentado sob a forma de uma análise SWOT (Strengths, Weaknesses, Opportunities and Threats), onde é feita uma avaliação global das forças, fraquezas, oportunidades e ameaças sentidas ao longo destes meses de Estágio. Além disso, são apresentados 3 casos clínicos resolvidos durante este período e termina, em tom de conclusão, com uma análise pessoal do meu trajeto enquanto estagiário e da importância do Farmacêutico na comunidade. Relativamente à Monografia, as associações de fármacos em terapia combinada, onde se incluem os comprimidos de dupla camada, têm vindo a conquistar mercado e a confiança dos investidores nos últimos anos devido à sua capacidade de solucionar problemas como a falta de adesão à medicação por doentes que tomam um elevado número de comprimidos diariamente e a presença de interações entre princípios ativos. No entanto, o seu correto fabrico está dependente de um conjunto de parâmetros e processos que devem ser monitorizados de forma a obtermos a forma farmacêutica final com as melhores características físicas e químicas. A otimização da formulação e dos processos de fabrico durante etapas críticas da sua produção, como a compressão e a ejeção, vai evitar problemas como a delaminação, que são muito frequentes na produção deste tipo de forma farmacêutica.Este artigo de revisão, além de evidenciar as vantagens deste tipo de forma farmacêutica, aborda alguns dos mais importantes fatores que afetam o seu fabrico, focando-se na delaminação através duma estratégia de Qualidade programada. As propriedades dos materiais, a elasticidade e a recuperação plástica, a pré-compressão e compressão, a humidade e a forma dos punções são alguns dos parâmetros abordados neste artigo. Finalmente, é sumariamente explicada a metodologia da Qualidade Programada e abordada a sua aplicabilidade, com exemplos, na tecnologia dos comprimidos de dupla camada. This document comprises the Internship Report in Community Pharmacy and the Monograph entitled "Development of bilayer tablets by Quality by Design: Focus on Manufacturing parameters". As an integral part of the last semester of the Integrated Master´s in Pharmaceutical Sciences, the Internship in Community Pharmacy took place at Farmácia Estádio, in Coimbra, between January and September 2021 and is presented in the form of a SWOT analysis (Strengths, Weaknesses, Opportunities and Threats), where an overall assessment of the strengths, weaknesses, opportunities and threats felt during these months of Internship is made. In addition, 3 clinical cases solved during this period are presented and it ends with a personal analysis about my Internship and the importance of the Pharmacist in the community.Regarding the Monograph, Fixed-dose combinations, where bilayer tablets are included, have gained market and investor confidence in recent years due to their ability to solve problems. Examples of these problems are the lack of medication adherence by people who take a high number of pills daily and possible interactions between active pharmaceutical ingredients. However, bilayer tablets manufacture depends on a set of parameters and processes that must be monitored in order to obtain the final pharmaceutical form with the best physical and chemical characteristics. The optimization of formulation and manufacturing processes during critical stages of its production, such as compression and ejection, will avoid problems such as delamination which are very frequent in the production of this type of pharmaceutical form.This review article, in addition to the discrimination of bilayer tablets’ advantages, addresses some of the most important factors affecting their manufacture, with focus on delamination by using Quality by Design as a strategy. The role of material properties, elasticity and plastic recovery, pre-compression and main compression, moisture and punch shape on tablets properties is covered in this article. Finally, the Quality by Design methodology, more precisely its applicability is discussed with examples in double-layer tablet technology.
