Abstract:
Jaruk tigarun is a traditional fermented food from South Borneo made from tigarun flower that was fermented in water without salt addition. The objectives of the research were to isolate and identify the Lactic Acid Bacteria (LAB) from jaruk tigarun then to determine the DPPH radical scavenging activities of jaruk tigarun extracts. LAB was isolated from jaruk tigarun which was fermented for 7 days at room temperature. Media of MRS+CaCO3 were used to isolate the LAB while API 50 CHL kit was utilised to identify them. Fresh flowers and jaruk tigarun were also freeze-dryed, crushed and extracted using methanol, ethanol and ethyl acetate. The total phenolic and antioxidant activity of each extract were determine with Folin-Ciocalteu method and DPPH. Three isolates of LAB were isolated and identified as Lactobacillus plantarum. The fermentation was able to increase total phenolic of jaruk tigarun which was extracted with methanol, ethanol, and ethyl acetate (53.24 ± 0.73, 44.86 ± 0.90 and 23.95 ± 0.13 mg GAE/g extract, respectively). Similarly, the antioxidant activity of jaruk tigarun that were extracted with methanol, ethanol and ethyl acetate increased to 92.68 ± 0.02 %, 92.43 ± 0.11 %, and 42.94 ± 0.02 %. Methanolic extract of jaruk tigarun has the highest IC50 that was equal to 1.511 μg/mL. UV-Vis spectrum analysis and FT-IR were used to identify the compounds isolated from methanolic extract of jaruk tigarun resulting flavonoid as tentative identified compounds. ABSTRAKJaruk tigarun adalah salah satu makanan fermentasi tradisional dari Kalimantan Selatan yang dibuat dengan cara merendam bunga tigarun dalam air matang selama beberapa hari. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri asam laktat yang terlibat selama fermentasi jaruk serta mengetahui aktivitas penghambatan radikal DPPH pada jaruk tigarun. BAL diisolasi dari bunga tigarun yang difermentasi selama 7 hari pada suhu kamar sampai menjadi jaruk. Isolasi BAL menggunakan media MRS+CaCO3 dan diidentifikasi dengan API 50 CHL Kit. Bunga segar dan jaruk tigarun juga masing-masing dikeringbekukan, dihaluskan dan diekstraksi dengan metanol, etanol dan etil asetat. Total fenolik dan aktivitas antioksidan dari masing masing ekstrak ditentukan dengan metoda Folin Ciocalteu dan DPPH. Tiga isolat BAL berhasil diisolasi dan diidentifikasi sebagai Lactobacillus plantarum. Fermentasi berhasil meningkatkan total fenolik dari jaruk tigarun yang diekstrak dengan metanol, etanol dan etil asetat berturut-turut sebesar 53,24 ± 0,73, 44,86 ± 0,90 dan 23,95 ± 0,13 mg GAE/g ekstrak. Demikian pula halnya dengan aktivitas antioksidan dari jaruk yang diekstrak dengan metanol, etanol dan etil asetat meningkat menjadi 92,68 ± 0,02 %, 92,43 ± 0,11 %, dan 42,94 ± 0,02 %. Ekstrak metanol dari jaruk tigarun memiliki IC50 yang paling baik yaitu sebesar 1.511 μg/mL. Hasil isolasi dan identifikasi senyawa yang dimurnikan dari ekstrak metanol jaruk menggunakan analisis spektrum UV-Vis dan FT-IR diperoleh dugaan senyawa adalah golongan flavonoid.Kata kunci: DPPH; flavonoid; jaruk tigarun; L. Abstract:
Daun tempuyung merupakan salah satu tumbuhan obat tradisional Indonesia yang banyak dimanfaatkan untuk pengobatan batu ginjal, anti inflamasi, disentri, radang dan rematik. Tempuyung banyak mengandung senyawa flavonoid yang merupakan golongan fenol terbesar. Penelitian ini dimulai dengan pengumpulan dan persiapan sampel, skrining fitokimia, pembuatan ekstrak etanol, dan pengujian kandungan total fenol dengan metode kolorimetri pereaksi Folin-Ciocalteu menggunakan spektrofotometer UV-Vis dengan asam galat sebagai baku pembanding. Hasil skrining fitokimia menunjukkan daun tempuyung mengandung senyawa flavonoid. Penetapan kadar total fenol menggunakan metode spektrofotometri pereaksi Folin- Ciocalteu didapat nilai regresi y = 0,00077 x + 0,03667 dengan nilai r2 = 0,99757 sehingga didapat nilai rata – rata kadar total fenol 45,04 ± 0,46 mg GAE/g ekstrak. Tempuyung is one of traditional medical plant in Indonesia and used in kidney stone medication, anti-inflammatory, dysentery, inflammation, and rheumatism. Tempuyung contains substantial flavonoid which considers as the biggest phenol. This research started from the preparation and sample collection, phytochemical screening, ethanol extract preparation, and total phenol test using colorimetry method usingFolin-Ciocalteu reagent byUV-Vis spectrophotometer and gallic acid as the standard compound. The phytochemical screening indicated that tempuyung leaves contained flavonoid compound. The total phenol contentwas determined using colorimetry method of Folin-Ciocalteu reagent and resulted with regression value of y=0.00077 x+0.03667 and the value of r2=0.99757 and the average of total phenolcontent of 45.04 ± 0.46 mg GAE/g.

Kandungan total fenolik diukur dengan metode Folin-Ciocalteu sedangkan aktivitas antioksidan diukur dengan metode FTC (Ferric Thiocyanate) untuk mengetahui kemampuan penghambatan peroksida pada tahap pertama oksidasi lipid. Hasil penelitian menunjukkan bahwa ekstrak metanol memiliki kandungan total fenolik sebesar 99,67 mg/kg dan kemampuan dalam menghambat oksidasi asam linoleat sebesar 49,464%.This research has been conducted to measure the total phenolic compounds and the antioxidant activity of methanol extract of soyogik leaves (Saurauia bracteosa DC) as a reducer of free radicals linoleic acid. The total phenolic content was measured using folin-ciocalteu method, meanwhile the antioxidant activity was measured using FTC (Ferric Thiocyanate) method and to determine the peroxide inhibitory capability in the first stages of lipid peroxidation. The results showed that the methanol extract had a total phenolic content of 99, 67 mg/kg and peroxide inhibition of 49, 464%

Telah dilakukan penelitian mengenai analisis kandungan fitokimia dan aktivitas antioksidan dari ekstrak tanaman lire yang diekstraksi secara maserasi dengan menggunakan pelarut etanol. Analisis total kandungan fitokimia yaitu uji fenolik dengan menggunakan metode Folin-Ciocalteu dan uji flavonoid menggunakan metode AlCl3. Uji antioksidan menggunakan metode DPPH. Berdasarkan hasil pengujian kandungan fenolik dan flavonoid ekstrak etanol tanaman lire, menunjukkan bahwa kandungan fenolik lebih tinggi dibandingkan dengan kandungan flavonoid. Hasil pengujian aktivitas antioksidan menunjukkan nilai IC50 sebesar 108,673 mg/L pada ekstrak etanol tanaman lire.Research have been done to analyze the phytochemical compound and antioxidant activity from lire plant extract which was extracted based on maceration method using the ethanol solvent. The phythochemical compound analysing to determining phenolik using Folin-Ciocalteu method and AlCl3 method for flavonoid test. Antioxidant activity using the DPPH method. Based on the result, the phenolic and flavonoid compound from extract ethanol of lire plants, was showed that phenolic compound more higher than flavonoid compound. The result of this test was showed the value of IC50 is 108.673 mg/L from the lire plant extract.

Abstract:
The research objectives were to identify the secondary metabolite components, total phenolic content and determine the antioxidant activity of the ethanol extract of red betel leaf (Piper crocatum Ruiz & Pav.). The extraction process was carried out by materation using ethanol as a solvent. Determination of total phenolic content was carried out colorimetrically with Folin-Ciocalteu reagent measured at a maximum wavelength of 765 nm. Determination of antioxidant activity using the DPPH method measured by spectrophotometry at a maximum wavelength of 517 nm. The results of phytochemical screening of the ethanolic extract of red betel leaf contain secondary metabolites, including flavonoid, phenolic, tannin, alkaloids, steroids, and triterpenoids. The total phenolic content of the red betel leaf ethanol extract was 0.949±0.003 mg GAE/g d.w. and has antioxidant activity (IC50) 84,656 including strong category as an antioxidant. Telah dilakukan penelitian untuk menentukan rendemen santan dan total fenolik santan kelapa dalam pada berbagai tingkat kematangan. Santan kelapa dibuat dengan cara kelapa dikupas, dibuang kulit coklat yang menempel pada daging kelapa, selanjutnya daging kelapa diparut, ditimbang, diperas, dan disaring. Penentuan kandungan total fenol menggunakan metode Folin-Ciocalteu. Dari hasil pengujian menunjukkan rendemen santan kelapa dalam tua merupakan nilai yang paling tertinggi sebesar 28.57 g/mL dan total nilai fenolik yang tertinggi terdapat pada santan kelapa dalam tua sebesar 143.9 mg/kg.Research has been conducted to determine coconut content and total phenolic from coconut milk in various level of maturity. Coconut milk made by coconut peeled, discarded brown skin attached to coconut meat, then coconut meat grated, weighed, squeezed, and filtered. Determination of total phenol content using Folin-Ciocalteu method. The results showed that coconut milk content in the oldest was the highest value of 28.57 g / mL and the highest total phenolic value was found in coconut milk in old age of 143.9 mg / kg.

Tanaman kelengkeng (Dimoncarpus longan Lour) memiliki potensi sebagai sumber antioksidan alami. Senyawa metabolit sekunder yang berperan sebagai antioksidan tersebut diantaranya adalah fenol dan flavonoid. Penelitian ini bertujuan untuk menentukan kandungan fenol dan flavonoid total yang terkandung di dalam ekstrak daun kelengkeng (Dimoncarpus longan Lour). Sampel yang digunakan adalah daun kelengkeng segar dan kering yang diekstrasi dengam metode maserasi menggunakan pelarut etanol. Penetapan kandungan fenol total dilakukan menggunakan pereaksi Folin-Ciocalteu dan pereaksi AlCl3 untuk penetapan kandungan flavonoid total yang diukur menggunakan Spektrofotometer UV-Vis. Hasil penelitian menunjukkan bahwa ekstrak etanol daun segar dan daun kering mengandung senyawa metabolit sekunder fenol dan flavonoid. Kadar total fenol dari masing-masing ekstrak daun segar dan kering adalah sebesar 78,21 µg/mg dan 107,51 µg/mg ekstrak. Sedangkan untuk kadar flavonoid total masing-masing adalah 21,23 µg/mg untuk sampel daun segar dan 33,64 µg/mg untuk sampel daun kering. ABSTRACTCalamus rhizome contain phenolic as antimicrobial agent. Fermentation can increase thereleasing of phenolic content from plant cells. The aim of this research was to observe thecomparison of phenolic content and antimicrobial activity between fresh and fermentedcalamus rhizome extract. Phenolic content was determined by using Folin-Ciocalteu method,and antimicrobial test by agar well diffusion. Phenolic content of fresh and fermentedcalamus rhizome extract respectively are 97,272±0,525 and 223,553±3,542 mgGAE/gram.Inhibiton zone against Staphylococcus aureus are 4,04±0,05 and 8,24±0,58 mm, whileagainst Candida albicans are 14,90±0,57 and 18,16±1,47 mm. Kandungan fitokimia yang diuji adalah total fenolik dan total flavonoid. Total fenolik menggunakan metode Folin-Ciocalteu (Yadaf dan Agarwala, 2010), lalu untuk mengetahui total flavonoid menggunakan aluminium klorida (Sultana et al., 2011), dan pengujian aktifitas antioksidan menggunakan 1,1-difenil-2-pikrilhidrazil (DPPH) (Subedi et al., 2011). Hasil penelitian menunjukan kandungan fenolik total sebesar 26,75 mg/L dan kandungan flavonoidnya sebesar 12,56 mg/L,untuk pengujian aktifitas antioksidan didapatkan nilai IC50 sebesar 240,19 mg/L.Research have been done to determining the phytochemical compound and antioxidant activity of yantan (Blumea chinensis DC) leaves. The phytochemical compound which tested is phenolic total and flavonoid total. Phenolic total using Folin-Ciocalteu method (Yadaf dan Agarwala, 2010), then to determining the total flavonoid using aluminium chloride (Sultana et al., 2011), and test of the antioxidant activity using 1,1-difenil-2-pikrilhidrazil DPPH (Subedi et al., 2011).The result of the total phenolic content was showed 26,75 mg/L and the total flavonoid content is 12,56 mg/L, for antioxidant activity test obtained the value IC50 is 240,19 mg/L.

Metode ekstraksi dapat mempengaruhi komponen kimia dalam suatu ekstrak tanaman.Pada penelitian ini telah dilakukan uji penentuan kadar fenolat total dari daun piladang(Solenostemon scutellarioides (L.) Codd). yang diekstraksi dengan beberapa cara. Metodeekstraksi yang dilakukan antara lain cara tradisional (peremasan dan perebusan) dan ekstraksilaboratorium (sokletasi dan maserasi). Kadar fenolat total ekstrak daun piladang diperolehdengan metode Folin-Ciocalteu dengan menggunakan spektrofotometer UV-Visible. Asam galatdiukur pada panjang gelombang maksimum 758 nm. Kadar fenolat dinyatakan dalam mg setaraasam galat / gr ekstrak. Perolehan kadar fenolat tertinggi oleh sokletasi daun kering 376,5979mg/g, diikuti oleh maserasi daun kering 356,7619 mg/g, sokletasi daun segar 333,1509 mg/g,maserasi daun segar 293,3015 mg/g, rebusan daun segar 216,3534 mg/g, sari remasan 77,3158mg/g, rebusan daun kering 69,3957 mg/g.

This diploma thesis deals with the stability and basic properties of combined beverages and fruit concentrates. The theoretical part describes the production and use of fruit concentrates. Furthermore, the work deals with anthocyanic pigments and phenolic substances. The principle and instrumentation of liquid chromatography and electron paramagnetic resonance were described. In the next subchapter, the methods of determination of total anthocyanins and phenolic substances were described. The experimental part of the thesis described the determination of soluble solids, viscosity and antioxidant activity. The total content of phenolic compounds were determined by the Folin-Ciocalteua method and the total anthocyanins were determined by the pH-differential method. In the samples were determined cyanidine-3-glucoside and cyanidin-3-galactoside by liquid chromatography.

In questo elaborato è stato sviluppato un metodo elettrochimico per la determinazione dei polifenoli nelle diverse tipologie di vino mediante l’utilizzo dei transistor elettrochimici a base organica, in cui la parte organica è formata dal polimero PEDOT:PSS. Si è cercato di ottimizzare le condizioni di migliore riproducibilità del segnale, come la tecnica elettrochimica, il tipo di elettrolita e il suo pH. In seguito, il metodo elettrochimico viene confrontato con quelli convenzionali per la determinazione dei polifenoli, in particolare, quelli spettrofotometrici. Il numero consistente dei campioni di diversi tipi di vino ha permesso di stabilire se i metodi elettrochimici e spettrofotometrici sono correlabili tra loro e di quanto.

Abstract:
The discovery and dissemination of new knowledge are essential in food science. To advance our understanding of fruit chemistry, analytical methods were compared and applied. Polyphenols are secondary metabolites in fruits of particular importance in food science, as they contribute to the sensory attributes and health benefits of the products. Evaluation of common analytical methods for the quantification of polyphenols, including the Folin-Ciocalteu (F-C), Lowenthal permanganate (L-P), 4-dimethylaminocinnamaldehyde (DMAC) and the bovine serum albumin (BSA) precipitation methods, was conducted using analytical method validation procedures. The F-C method was not specific to polyphenols, and the L-P method had the widest working range but lacked accuracy. The DMAC method was the most specific to flavanols, and the BSA method was not suitable for quantification of smaller flavanols. Quantitative performance of these four methods was evaluated using a broad range of fruit-derived samples. Variation in quantitative results obtained using these four methods was explained by differences in polyphenol and matrix composition of these samples and differences in operating principles of the methods. The reactivity of individual polyphenol compounds (catechin, epicatechin, PC B2, PC pentamer, chlorogenic acid, phloretin, and quercetin) to the polyphenol and flavanol quantification results using Prussian blue (P-B), F-C, DMAC and BSA precipitation methods were also assessed and determined to differ by up to thirteen-fold, depending on the assay. Furthermore, the contribution and interactions of polyphenol compounds (catechin, PC B2, and chlorogenic acid) and potentially interfering compounds likely to be found in fruit and fruit products (ascorbic acid, glucose, and SO2) to the quantitative results of these methods were evaluated using a full factorial design. Significant interactions among polyphenol compounds, and among the interfering compounds were found. The standardized coefficient (β) for all factors and interactions of polyphenol compounds varied from 0.347 to 129, and from near 0 to -46.8 for all factors and interactions of interfering compounds. Our findings indicate that the choice of standards, polyphenol and matrix composition of the sample may cause disparity among the quantitative results of these methods. Amino acids in apple (Malus × domestica Borkh.) juice not only influence the quality of fermented cider through fermentation kinetics but also impact the flavor of the cider through yeast metabolism. Due to recent advances in analytical instrumentation, amino acids profiles in apple juice were determined much faster and more accurately than by previously applied methods. Twenty amino acids were quantified by UPLC-PDA in juices from 13 apple cultivars grown in Virginia. The relative amino acid profile was significantly different among the apple juices evaluated. The total amino acid concentration ranged from 18 mg/L in Blacktwig juice to 57 mg/L in Enterprise juice. L-Asparagine, L-aspartic acid and L-glutamine are the principal amino acids observed in most apple juices. These results will inform future research on yeast metabolism and nitrogen management during cider fermentation. To better disseminate knowledge gained through research to the next generation of food scientists, the effectiveness of new instructional technology—a cellphone-based personal response system—in food science education was evaluated. Students' academic performance was improved by the incorporation of this technology into lectures, and its use was well perceived by the students (easy to use and positively impacted their learning). This finding contributes to the scholarship of teaching and learning in food science by providing useful insight into the potential for application of such tools with improved student engagement and learning outcomes. Advances in food chemistry research will enable the development of value-added food products, and the pedagogical advancement in food science education will better convey new and existing knowledge to students, who will apply this knowledge to promote a safe and nutritious food supply that enhances human health and increases the value of specialty crops.

Abstract:
The composition of extracts isolated from black alder bark by ‘green’ microwave assisted water extraction in the temperature range of 70–150 ℃ was studied using the wet chemistry Folin-Ciocalteu method and Py-GC-MS/FID. The composition data were compared with those of the extracts obtained at the same temperature by accelerated solvent extraction (ASE) of bark. It was shown that microwave assisted extraction, compared with ASE, resulted in more significant transition of major cell wall components, including hemicelluloses and phenolics of lignin origination, into the solution. Depending on the microwave assisted extraction regimes, products with different portion of major cell wall components and secondary phenolic metabolites can be isolated that enlarge the possibilities of products valorisation. Thus, a significant promotion of secondary phenolic metabolites’ transition into extracts as a result of microwave extraction was observed at 70 ℃. At that time the relative portion of carbohydrates in extracts was increased at high temperature extraction, combining dynamic and isothermal microwave heating. Water extraction of black alder bark in a microwave extractor revealed 25–50% lower specific energy consumption and 1.8–2.6 times higher productivity in comparison with the conventional extraction, that is beneficial in view of the upscale and practical application of this innovative biomass processing.

Abstract:
Compared with other sources, potato can bring multiple nutritional benefits because it’s naturally low energy food (0.7 kcal), having high water, fiber and starch content. Even if the consummation of potatoes is in decline, it is still considered a source of valuable nutrition. Depending on the method of preparation, potatoes contain significant level of proteins and antioxidants and can offer considerable protection against cardiovascular diseases and cancer. Natural antioxidants are present under different forms in all plants, being the base source of these compounds for humans. The objective of this study was to determine the moisture content, crude protein, total antioxidant capacity and phenolic content of three assortments of potatoes (Solanum tuberosum) available on the Romanian local market (Timis County). The study was carried out on raw, unpeeled, boiled and roasted potatoes. The moisture content was determinate thermogravimetrically using Sartorius thermo balance, crude protein quantified by using a rapid colorimetric method, total antioxidant capacity determinate using CUPRAC method and total polyphenols content using Folin-Ciocalteu assay. The experimental results show that blue roasted potatoes present the highest content of crude protein, total antioxidant capacity and total polyphenols content and the lowest water content. The obtained data are used to create a graphical fingerprint of raw and processed potatoes in order to identify the best options to mix different potatoes assortments and to create innovative nutritious food products
