Full textThe general objective of this thesis is to extend the understanding of combustion instabilities by testing models, physical concepts and numerical procedures, and by providing new numerical post-processing tools to do so. Flame Transfert Function (FTF) : four methods for the determination of Flame Transfer Functions (FTFs) in LES have been tested. A new method based on the forcing of the flame by a filtered white noise is used to obtain the FTF at all frequencies with only one unsteady LES. Disturbance Energies and Stability Criteria in reacting Flows : two stability criteria in reacting flows are derived. The first one extends the Rayleigh criterion by taking into account the influence of the heat flux fluctuation. The second criterion is nonlinear and is based on the analysis of the budjet of a newly derived nonlinear disturbance energy during a controled thermo-acoustic instability

Full textModern pollutant regulation have led to a trend towards lean combustion systems which are prone to thermo-acoustic instabilities. The ability of Large Eddy Simulation (LES) to handle complex industrial heavy-duty gas turbines is evidenced during this thesis work. First, LES is applied to an academic single burner in order to validate the modeling against measurements performed at TU Berlin and against OpenFoam LES simulations done at Siemens. The coupling between acoustic and combustion is modeled with the Flame Transfer Function (FTF) approach and swirl number fluctuations are identified changing the FTF amplitude response of the flame. Then, an industrial gas turbine is analyzed for two different burner geometries and operating conditions. The FTF is only slightly influenced for the two operating points but slight modifications of the swirler geometry do modify the characteristics of the FTF showing that a simple model taking only into account the flight time is not appropriate and additional mechanisms are at play. Those mechanisms are identified being the inlet velocity, the swirl and the inlet mixture fraction fluctuations. The latter is caused by two mechanisms: 1) the pulsating injected fuel flow rate and 2) the fluctuating trajectory of the fuel jets. Although the diagonal swirler is designed to provide good mixing, effects of mixing heterogeneities at the combustion chamber inlet occur. Mixture perturbations phase with velocity (and hence with swirl) fluctuations and combine with them to lead to different FTF results. Another FTF approach linking heat release to inlet velocity and mixture fraction fluctuation (MISO model) shows further to be a good solution for complex systems. A nonlinear analysis shows that the forcing amplitude not only leads to a saturation of the flame but also to changes of the delay response. Flame saturation is only true for the global FTF and the gain increases locally with increasing forcing amplitude. Both, the linear and the nonlinear flames, are not compact: flame regions located right next to each other exhibited significant differences in delay meaning that at the same instant certain parts of the flame damp the excitation while others feed it

Full textThermoacoustic instabilities are spontaneously excited by a feedback loop between an oscillatory combustion process and one of the natural acoustic modes of the combustor. This phenomenon causes loss of performance and severe damages to the engine. However, aeronautical engines or gaz turbines manufacturers often observe this kind of instabilities during the experiments. In this work we propose a methodology dedicated to the computation of thermoacoustic instabilities usable in an industrial context. The coupling between acoustics and combustion is accounted for thanks to a specific Helmholtz equation including a reacting flow term. Using a finite element approach leads to an algebraic non-linear eigenvalue problem with N dimensions (N the number of nodes in the mesh). A point fix algorithm mixed with subspace iterative methods (Arnoldi implemented in ARPACK or Jacobi-Davidson) permits to solve this problem efficiently. Because aeronautical combustors usually present an annular shape with 10 to 20 injectors located all over the circumference, a multi-referenced (n-tau) flame transfer function is proposed. This thermoacoustic model shows amplified or damped, standing or spinning azimuthal modes depending on the parameter tau. This methodology is integrated in the calculation chain QUIET (AVBP-N3S-NOZZLE-AVSP) and it is used to study thermoacoustic instabilities of the Turbomeca ARDIDEN engine. The results are shown to be in good accordance with large eddy simulations of the whole combustor

Full textAn essential step of human immunodeficiency virus type 1 (HIV-1) reverse transcription is the first trand transfer that requires base pairing of the R region at the 3’- end of the genomic RNA with the complementary r region at the 3’-end of minus strand strong-stop DNA (ssDNA). HIV-1 nucleocapsid protein (NC) facilitates this annealing process. Determination of the ssDNA structure is needed to understand the molecular basis of NC-mediated genomic RNA-ssDNA annealing. For this purpose, we investigated ssDNA using structural probes (nucleases and potassium permanganate). This study is the first to determine the secondary structure of the fulllength HIV-1 ssDNA in the absence or presence of NC.Our probing data obtained in the absence of NC, suggest weak contacts between the u5 region of ssDNA and the U3 region the genomic RNA. The probing data and phylogenetic analysis support the folding of ssDNA into three stem-loop structures and the presence of four high-affinity binding sites for NC. Using the gel retardation assay, we analyzed the interaction of NC with each site. Taken together, our results support a model for the NC-mediated annealing process in which the preferential binding of NC to four sites triggers unfolding of the threedimensional structure of ssDNA, thus facilitating interaction of the r sequence of ssDNA with the R sequence of the genomic RNA. In addition, using gel retardation assays and ssDNA mutants, we show that the annealing of ssDNA to the 3’- end of the genomic RNA requires NC activity and does not rely on a single pathway (zipper intermediate or kissing complex)

Full textInfrared Focal Plane Arrays (IRFPAs) technological evelopment is currently evolving rapidly and can be applied to various fields (space/defence, astronomy..). The reduction of the pixel pitch poses a challenge in terms of characterization of the Modulation Transfer Function (MTF). MTF is related to the spatial response of the pixels. The optical projection of fine patterns then becomes a problem in the measurement due to the diffraction limit. The hyperspectral imagery applications and the diversity of pixel structures used in the industry also question the wavelength dependency of the MTF. In this thesis, we study an interferential method in order to best address these different issues of spatial response measurement. These gratings allow the 2D projection of high-resolution patterns that can be predicted by simulation. The detected pattern allows the restitution of the MTF. Since the IRFPAs considered here operate at cryogenic temperatures, a nitrogen-cooled bench is developed to measure the MTF with a reduced thermal background while having access to large aperture angles. A treatment is developed and tested on a near-infrared detector in order to estimate the propagation of noise through the method. This procedure also allowed to measure electronic diffusion effects between the pixels. In a second time, wavelength-dependent MTF measurements were performed on a mid-wavelength infrared detector within the cryogenic bench. This study revealed a spectral dependence of the MTF on specific pixel structures. Finally, during these experiments, residual errors about the prediction of the projected interferential patterns were highlighted. A feedback is then given, leading to the proposal of a new measurement procedure. The simulation of the propagation of the beam through the grating appears to be a key element of the method

Full textCombustion instabilities, induced by the resonant coupling of acoustics and combustion occur in many practical systems such as domestic boilers, gas turbine and rocket engines. They produce pressure and heat release fluctuations that in some extreme cases can provoke mechanical failure or catastrophic damage. These phenomena have been extensively studied in the past, and the basic driving and coupling mechanisms have already been identified. However, it is well known that most systems behave differently at cold start and in the permanent regime and the coupling between the temperature of the solid material and combustion instabilities still remains unclear. The aim of this thesis is to study this mechanism. This work presents an experimental investigation of combustion instabilities for a laminar premixed flame stabilized on a slot burner with controlled wall temperature. For certain operating conditions, the system exhibits a combustion instability locked on the Helmholtz mode of the burner. It is shown that this instability can be controlled and even suppressed by changing solely the temperature of the burner rim. A linear stability analysis is used to identify the parameters playing a role in the resonant coupling and retrieves the features observed experimentally. Detailed experimental studies of the different elementary processes involved in the thermo-acoustic coupling are used to evaluate the sensitivity of these parameters to the wall temperature. Finally a theoretical model of unsteady heat transfer from the flame root to the burner-rim and detailed experimental measurements permit to establish the physical mechanism for the temperature dependance on the flame response

Full textThe purpose of this research is to better understand the psychological workings of those youths that are called « unplaceable ». The term « unplaceable » is recurring in child welfare services. It defines those « troubled teenagers », « teenagers with behavioural issues », those « no one wants »… They are called by a variety of terms. The complex nature of this clinical case will lead to a review of the history and evolution of the institutions caring for children in France, so as to understand how they adapted to « troubled » children and teenagers. Through this issue, I will examine the psychological workings of those teenagers, linked to the lack of paternal function and the role of thirdness, a subject that seems central to better understand child care. The clinical case, studied throughout more than two years, is that of Paul and Laurent, two institutionalized youths that could be called “unplaceable”. This clinical case is made more complex by the diversity of concepts and terms used to define those teenagers. It implies a reflexion on the notions of transference and countertransference that will be present for the duration of the research work. The institutional stakes present in the care for those youths will be examined through a psychoanalytical angle, and sometimes other points of view – given that the notion of “unplaceability” is diverse. The notions of the absence of the father and maternal lack can shed some light on what makes Paul and Laurent “unplaceable”. But they will not be labelled thus indefinitely

Full textThe rate of pyrolysis of a fire-exposed solid fuel is a key parameter to evaluate the intensity of fire and its effects on surroundings. The present study is experimental, theoretical and numerical. It focuses on the determination of the steady-state pyrolysis rate of a vertical slab of PMMA. An original experimental set-up is thus developed along with a specific procedure in order to locate the regressing surface of the solid material. A theoretical approach which is based on the reactive laminar boundary layer theory (LBL) assumptions and a numerical two-dimensional CFD model are used. Experimental results show that a three-dimensional behavior appears for small slab widths. In the laminar part of the flow, near to the leading edge, experimental and numerical results reveal a decreasing power-law of the pyrolysis rate, in agreement with the LBL theory. Radiative and convective effects that are not taken into account by the theory are responsible for discrepancies on scaling laws. The assumptions of the LBL theory are discussed. It is found that for free flows the pyrolysis rate is inversely proportional to the normal gradient of the gas velocity at the fuel surface. A transient pyrolysis model based on the mass transfer number is proposed, avoiding the calculation of the thermal and flow fields of the gas phase

Full textThe characterization of an episome able to grow and live only in presence of XNA could be the key to reap part of GMO's benefits with limited environment or health risk. The aim of this work was to explore and implement in E. coli a vital reaction catalyzed by an RNA sequence in order to replace the RNA by XNA at a later stage.In this challenging task, different ribozymes activity were first studied by analytical chemistry technics. Unfortunately, none of the sequences selected using SELEX method demonstrated a catalytic activity with the formation of the expected product.A completely different strategy was set up thereafter to escape the same complications. This time, we chose the suppressor tRNA Cys T' structure and more specifically its variable loop as a scaffold to implement in E.coli a specific RNA sequence vital for the cell. The essential ThyA gene with a mutation at the codon 146 was used to complement the system and screen the tRNA suppressor activity. After several optimizations of the construction, we observed a correct insertion of the different A-Loops in the tRNA Cys T without altering its suppressor function. Finally, implementation of several random sequences was also performed by screening a library

Full textNon-premixed flame stabilization is still an important issue in combustion. Addressing this issue requires to evaluate the relative importance of aerodynamic, thermal and chemical phenomena involved in the stabilization mechanisms. This thesis develops to this end an experimental approach, with a particular focus on the influence of thermal boundary conditions, examining the role of heat transfer in a twofold analysis. At first, the attachment zone of a rim-stabilized jet-flame is investigated, by careful consideration of the coupling existing between the burner lip temperature and the flame attachment location relative to the burner. Several regions have been identified according to the flame leading edge behavior, which evolves from diffusive to propagative closer to the aerodynamic stability limits. Besides, by modifying the burner thermal properties, a change has been shown in the relative roles of thermal and chemical quenching of the flame by the burner wall, depending on the burner lip temperature. Secondly, the attention is directed to transitions between different combustion regimes, namely attached and lifted flames. In particular, beyond the stable state of an attached flame, its lifting process is investigated. Examining in which conditions destabilization of the flame occurs indeed appears to be an ideal opportunity to gain insight into the aerothermochemical coupling and equilibriums existing prior to lift-off. Thus, the lifting process has been shown to be modified by the reactant initial temperature, in close relationship with the change in occurrence of localized transitory extinction events of the reaction zone
