Full textThe paper introduces the new phenomena of dark tourism that has not ever been analyzed in Lithuanian academic literature. The notion of dark tourism and the variety of its forms is represented. This paper also analyzes the reasons of increasing popularity of dark tourism in postmodern society and the ethical points of dark tourism. This paper concentrates on assessment of Lithuania‘s dark heritage and explains the reasons, why this heritage is in such condition. The main focus is on Kaunas Fortress which is an outstanding example of dark heritage in Lithuania. The representation of dark heritage in Kaunas VII and Kaunas IX museums are compared and also the Lithuanian tourist‘s motivation of visiting the dark sites is analyzed.

Full textThis study examines professional sign language interpreters view on similarities and differences between interpreting on site and remote interpreting. The study also examines coordination and the understanding of the word by sign language interpreters. The material for this study consists of videorecorded interviews which have been analysed based on the study’s research questions. Sign language is a visual language and therefore has grammatical differences to spoken language, for example the signing space in front of the person signing. This study examines what parts of interpreting that change when it is performed remotely and therefore becomes two- instead of three-dimensional for the recipient. The study also investigates how the interpreting is affected when the interpreter users are not in the same physical room as the interpreter. The results show that remote interpreting limits the reference system as well as how bodily orientation is used as a resource in an interpreted conversation. The interpreter can turn towards an interpreter user during on-site interpreting in order to get their attention discreetly, however, in remote interpreting this body turn does not work in the same way, seeing as the screen is two dimensional and that the interpreter user is not in the same physical room as the interpreter.
Bose, Deepanwita. "Tat-independent lentivirus genomes for vaccination and host/pathogen interaction studies." Thesis, Université Grenoble Alpes (ComUE), 2017. http://www.theses.fr/2017GREAV009/document.
Full textOur lab has previously described the generation of a unique vaccine prototype against HIV-1/AIDS. It is a non-integrative DNA lentivector vaccine tested in pilot studies in animal models of HIV vaccine. The non-human primate study showed protection of all 6/6 macaques and immune response correlates were composed of a variety of effector (EM) and central memory (CM) T cells. More importantly, they also contained high proliferating antigen specific cells containing a type of stem cell-like memory T cells (TSCM). In this thesis the vaccine was enhanced further by switching the CXCR4 envelope of the vaccine to CCR5 tropic envelopes such as the clade B WARO obtained from a chronically infected patient and a series of three transmitted/founder (T/F) HIV Clade C strains from Zambia. To improve further the vaccine we developed new strategies to incorporate molecular adjuvants able to enhance and sustain the newly elicited immune responses.Since the human lentivirus HIV-1 has developed multiple complex strategies to persist, the focus of the next part of my thesis was to develop a tool to ease and better understand the underlying mechanisms of latency in infected memory CD4+ T cells. Latently-infected cells have non-expressed integrated viral DNA genomes. One of the main mechanisms of this latency is absence of Tat transactivation of the LTR promoter. The recent focus post development of efficient highly active antiviral therapy (HAART), is the cure of the reservoir of latently infected cells. One of the obstacles for this type of studies is the lack of proper primate lentivirus prototypes incapable of undergoing latency as extreme infection model in the evaluation. We hypothesized that a replication-competent SHIV genome driven by the Tat-independent constitutive-expression LTRs of CAEV will be a valuable tool for such studies. We designed chimeric CAEV LTRs bearing the attachment sequences of SIV at their extremities and used them to drive the complete genome of SHIV-KU2. The resulting construct is SHIV-YCC which is expected to generate virus that will not undergo latency due to absence of Tat. We found that cells transfected with SHIV-YCC genome produce SHIV proteins that are assembled into infectious particles released out of the cells. Virions are able to infect target CD4+ T cells both in primary PBMCs and cell lines. Passaged virus in macaques PBMCs increased virus replication and infectivity. SHIV-YCC is the first chimeric primate replication-competent lentivirus that constitutively expresses all viral proteins. This new model offers the possibility of studying the early events by which provirus undergoes latency particularly when the envelop gene will be replaced with that of the T/F CCR5 tropic HIV-1

Solving convex optimization problems has become extremely efficient and reliable after the recent development of polynomial-time algorithms and advancement in computing power. Geometric Programming (GP) and Signomial Programming (SP) has been proven successful in optimizing multidisciplinary designs due to exploiting the speed of convergence and the ability to model non-linear designs. However, an optimal solution of GPs and SPs can be sensitive to uncertainties in the parameters involved in the problem. In fact, robust optimization can incorporate the uncertainties in an optimization problem and solves for the worst-case scenario. Yet, robust geometric programs (RGPs) and robust signomial programs (RSPs) are not known to have a tractable formulation that current solvers can efficiently solve. In this thesis, approximate formulations of RGPs and RSPs are proposed. Recently, the curiosity regarding the deployment of GPs and SPs in model complex engineering systems has been growing. This awareness has motivated modeling the uncertainties that are fundamental to engineering design optimization. Consequently, RGPs and RSPs provide a framework for modeling and solving GPs and SPs while representing their ambiguities as belonging to an uncertainty set. The RGP methodologies presented here are based on reformulating the GP as a convex program and then robustifying it with methods from robust linear programming. The RSP methodology is based on solving sequential local RGP approximations. These new methodologies, along with previous ones from the literature, are used to robustify aircraft design problems, and the results of these different methodologies are compared and discussed.

Pagès, i. Pi Gemma. "Intrathecal administration of AAVrh10 coding for β‐glucuronidase corrects biochemical and histological hallmarks of mucopolysaccharidosis type VII mice and improves behavior and survival." Doctoral thesis, Universitat Autònoma de Barcelona, 2015. http://hdl.handle.net/10803/300733.
Full textMucopolysaccharidosis type VII (MPS VII) is an ultrarare monogenic lysosomal storage disease. It is caused by the lack of β-‐glucuronidase activity, a lysosomal enzyme involved in the degradation pathway of glycosaminoglycans. This inborn genetic alteration causes a dysfunction of the lysosomal system that entails abnormal lysosomal storage and disruption of cell homeostasis. The disease presents a range of clinical severity among patients, from death in utero to a life expectancy of up to 20 or 30 years for the milder forms. The severe form of MPS VII among cases with postnatal disease onset is characterized by hepatosplenomegaly, skeletal abnormalities, developmental delay and mental retardation, among other symptoms. Currently, the only treatments for MPS VII patients are interventions to alleviate the symptoms, but no curative treatment is available. Gene therapy is a promising therapeutic approach to find a cure for monogenic diseases. Among the available gene delivery vectors, adeno-‐associated viruses (AAVs) present several features that make them attractive for gene therapy strategies: they are able to transduce dividing and quiescent cells, they provide long term expression of the transgene, they are not able to autonomously replicate without a helper virus, and wild type AAV infections are not pathogenic. Different AAV serotypes have been used as gene therapy vectors in preclinical studies. Among them, AAV9 and AAVrh10 are the serotypes which show greater transduction capacity and a broader range of cell-‐type specific tropism, particularly in the central nervous system. The use of AAVrh10, a non-‐human serotype, may avoid the neutralization by anti-‐AAV immune factors present in human sera after natural AAV infections. However, cross-‐reactivity with antibodies raised against AAV2, the most common human AAV serotype, may still interfere in the therapeutic outcome. In this work, we propose a gene therapy strategy for MPS VII based on a single intrathecal injection of an AAVrh10 coding for the β-‐glucuronidase gene, tested in young adult MPS VII mice. We show that vector delivery to the CSF by lumbar puncture, a poorly invasive technique, allows the transduction of CNS structures using a lower vector dose than by intravenous delivery. In addition, the drainage of the vector from the CSF to the bloodstream results in transduction of somatic organs such as liver, thus providing a systemic β-‐glucuronidase source that achieves serum enzymatic activity comparable to wild type mice. The sustained recombinant enzyme expression by AAV-‐transduced cells, and the cross-‐correction provided by enzyme secretion, attains the correction of biochemical and histopathological hallmarks of the disease in CNS and somatic organs. This correction at the cellular level leads to a significant improvement of physical, cognitive and emotional characteristics of MPS VII mice and a doubling of the MPS VII mouse life span.

Full textPrior to acquisitions, potential investors must conduct a survey on the company of interest. This process is called Due Diligence and it aims to reduce the asymmetric information between sellers and investors. This essay describes how investors through Due Diligence canidentify and evaluate the information hidden within the asymmetric information. The study is conducted as a cross-sectional study consisting of personal interviews with three business brokers and seven actors who have participated in business transfers. The conclusions are that buyers’ behavior in general are consistent with the theoretical framework. All respondents recommend using Due Diligence, but the scope and knowledgeof the process differs. The factor that can be crucial for a successful or unsuccessful Due Diligence is the investors’ understanding of their own limitations and the ability to enlist thehelp from other specialists in the fields they cannot manage on their own. Consultants such as business brokers, accountants and lawyers are therefore of great importance for Due Diligence. There are some areas that the respondents choose to study with particular care during DueDiligence. The one area that is common to all respondents is the investigation of the data contained in Annual Reports. Other areas vary depending on the investors experiences and among others include the corporate culture, external relations, capacity utilization and environmental issues. Thanks to the extensive and overall good use of Due Diligence, it can be considered that thereis no general market failure in the corporate market, although some transfers are less successful.

To enable the fast and accurate cataloging of material samples, I designed a filtration device for selecting specific visible and near-infrared light wavelengths related to the red, green, and blue sensitivity peaks of a visible detection camera and the pixel response for a near-infrared camera. This filter device functions in conjunction with the Department of Architecture's Daylighting Laboratory goniophotometer to profile the complete reflection and transmission properties for sample building materials. The resulting data is used in computer simulations and material optimization. The goniophotometer uses two types of detection cameras, color and infrared, to measure the light that is transmitted or reflected off a sample of material. The spectral sensitivity variances of the cameras create inaccuracies in the resulting data when full-spectrum light is used. To remove these inaccuracies, the light is filtered into smaller sections of the full spectrum and the data is recombined by software, to remove these inaccuracies. The device to filter the light is the subject of this thesis. The final filter design uses a geneva drive to index wheels containing pairs of high-pass and low-pass filters into the light path between the light source and the test specimen.
(cont.) The device satisfies the design specifications dictated by the usability, function and spatial constraints. This design should prove to be very reliable and flexible through its continued use in studying building materials. As the project is advanced, future work includes installation of the control system and integration into the software used to coordinate the goniophotometer components.

Full textHIV antiretroviral therapeutic drug monitoring is currently based on total concentrations. Relationships between total concentration and efficacy have been reported in several studies. However, some virological failures are still not explained by total concentrations. Therefore, there is an interest to identify an alternative exposure marker in order to explain these virological failures. The unbound concentration is considered as the pharmacologically active form. The unbound form is able to diffuse into tissues and cross cell membranes to exert its pharmacological activity. Dolutegravir, darunavir and atazanavir are highly bound (over 85%) to plasma proteins. Thus, only a small amount of unbound antiretroviral is available to perform the pharmacodynamic activity. Therefore, it can be assume that a slight variability in the unbound concentration, not perceived by total concentrations measurement, may explain a difference in efficacy between patients with similar total concentrations. To date, few studies have explored the unbound form of antiretrovirals, either from a technical or pharmacokinetic point of view. Thus, the objectives of this study were designed to be achieved in four successive steps: (i) Validate a method to isolate the unbound from the bound form and to quantify accurately unbound and total concentrations (ii) Compare equilibrium dialysis and ultrafiltration techniques, and identify ultrafiltration conditions that ensure comparable results between the two methods, (iii) Investigate the properties of antiretroviral binding to plasma protein, (iv) Explore the relationships between unbound and total concentrations of DRV in HIV patients using a population pharmacokinetic method. First, we explored the pre-analytical processes that could interfere with antiretroviral protein binding with equilibrium dialysis. As a result, we observed that antiretrovirals binding to plasma protein were globaly not sensitive to pre-analytical conditions (Metsu et al Clinica Chimica Acta 2018). Then, we established the ultrafiltration conditions ensuring identical results to those achieved by equilibrium dialysis at 37°C. In this second part, we stressed that ultrafiltration conditions may considerably alter ultrafiltration results (Metsu et al submit in Scientific Report). We also explored the factors of variability on antiretroviral binding to plasma protein. In this study, we established that dolutegravir binding can be affected by other substances highly bound to albumin (Metsu et al IWCPAT 2017). Based on results from plasma protein concentrations and unbound concentrations results from published studies, we proposed dosing recommendations for atazanavir and darunavir in pregnant women. A population pharmacokinetic study was then conducted using paired unbound and total DRV concentrations measured in HIV patients' samples. In this study, high BMI was found to be a covariable of interest to explain unbound low exposure by increased intrinsic clearance. Moreover, lower dissociation constant results were observed from patients with high BMI (Metsu et al IWCPAT 2018). Then, based on Monte-Carlo simulations and data from the literature, we showed that patients with high BMI had lower unbound darunavir exposure in cerebrospinal fluid, compared to other patients

Full textIn this dissertation, we propose the expression and the modelling of the most important principles of privacy. We deduce the relevant privacy requirements that should be integrated in existing security policy models, such as RBAC models. We suggest the application of a unique model for both access control and privacy requirements. Thus, an access control model is to be enriched with new access constraints and parameters, namely the privacy contexts, which should implement the consent and the notification concepts. For this purpose, we introduce the privacy-aware Organisation role Based Access Control (OrBAC) model. The administration of this new model is significantly different from previous models. Three cases are identified. First, the privacy policy may be defined by the data collector but data owners have the possibility to set their preferences through a contracted Service Level Agreement (SLA). Second, the administration model allows legal organisations, for legal purposes, to impose their needs by bypassing user's preferences. Third, we present the case of a privacy policy which is negotiated between the data collector and the requestor based on user's preferences, defined in the SLA. Overall, our proposal is a distributed administration of privacy policies. Focusing on Location Based Services (LBSs), we finally propose a complete privacy framework ready to be deployed in information systems. We use the model prototype to adapt our solution to cellular networks when the requesters are the service providers. This prototype uses parlay gateways with web services. We also extend the set of Parlay X gateway standardised web services by proposing a dedicated privacy web service to enforce privacy protection

Full textBackground: Small enterprises have become increasingly important in the Swedish business world. Among other things, 60 percent of the contribution to the country’s economy comes from small businesses. To support future growth and development it is important for companies to venture onto the international markets. This is a big step for small enterprises with constrained capital and guidance on the internationalization process and choice of entry mode is needed. Research on these two factors are limited in relation to small technology-focused enterprises in the service sector, which is the most rapidly growing sector in the Swedish economy. Aim: The aim of the study is to analyze the internationalization process and the choice of entry mode for small and technology-focused enterprises in the Swedish service sector. Methodology: The study has a qualitative research strategy and a deductive research approach. It is based on semi-structured interviews with three small technology-focused enterprises in the Swedish service sector and can be classified as a comparative case study. Conclusion: The conclusion of this study is that the most essential tool, for small technology-focused enterprises in the Swedish service sector when going abroad, is a distinct network of contacts. The study also reveals that the technical aspect of these types of companies implies that collaboration is the least risk and capital-intensive choice of entry mode at the beginning of a foreign establishment, which challenges the current research on the topic.

understanding about resistance to management accounting change and also to survey reasons
for the resistance and how to cope with the resistance.
The study is a qualitative case study and semi-structured interviews have been used to
complement the study’s theoretical area, consisting of scientific articles.
The conclusion from the study is that resistance to management accounting change can be
understood and explained by the institutions of each organisation and its underlying structures. A
further conclusion is, with the structures in mind there are many reasons for resistance and also
many ways to cope with the resistance. A model has been produced in purpose to create
understanding and to survey the resistance to management accounting change.

A major benefit of Additive Manufacturing (AM) is a faster timeline from design to fabrication. As AM has matured to be able to create functional prototypes and end-use products, the ability to quickly fabricate physical hardware iterations without associated tooling costs and lead times is now possible. Software companies have embraced iterative-based product development processes (PDP) such as Agile. Iterative development has allowed for the validation of innovative and untried solutions, fueling the rapid speed of software development. However, within complex hardware industries, like automotive and aerospace, almost all companies instead follow a Waterfall or Phase-Gate PDP. Large capital costs, along with the aforementioned lengthy tooling and supplier lead times, make the control and predictability of a Phase-Gate process appealing. However, the trade-off is a process where the final content gets decided near the beginning of a multi-year timeline, often translating to product launches with soon-to-be stale technologies. Within the context of automotive, this thesis explores how leading edge technology could continue development in a parallel Agile process. Though the use of AM, the new technology could be integrated later into a Phase-Gate process with minimal schedule risk or cost. This process keeps the strict one-way review gates for the more stable components, while allowing greater flexibility for innovative features that could benefit from further iteration. I use Design Structure Matrix theory to simulate the performance and schedule of this proposed PDP. I then discuss the implications of this new PDP architecture and its benefits for complex hardware industries in general.

Full textOne of the main tasks of universities is to prepare students for an academic or professional career, and to make them responsible members of society by giving them the motivation and skills to create a better future. Education for Sustainable Development is a core feature of education in a university of the 21th century and in society. In a university, it has two aspects: firstly, to educate students and to help future academics to contribute to sustainable development, secondly, to contribute to sustainability and to spread education for sustainable development in the community. We consider examples of successful cooperation between the university and its stakeholders via projects that are relevant for education for sustainable development. We analyse the success factors and the educational and sustainability impacts of these projects.

Full textTitle: Midwives experiences of obtained support when screening for domestic violence Department: School of Health and Education, University of Skövde Course: Master Degree Project in Midwifery, 15 ECTS Author: Emma Johansson och Michelle Kiwalabye Parma Supervisor: Elisabeth Hertfelt Wahn and Stina Thorstensson Pages: 19 Month and year: August 2015 Keywords: midwives, experiences, support, screening for violence, domestic violence. Background: Domestic violence affects women in all countries and all social classes. Approximately 30 percent of women who have lived in a relationship have been exposed to physical and/or sexual violence by their partner. Unreported cases are presumably many. In Sweden no national violence-screening instrument exists, only recommendations. Research shows that midwives experience lack of time, knowledge and support when screening for domestic violence. Aim: To examine midwives experiences of obtained support when screening for domestic violence at an antenatal care unit. Method: This study had a qualitative approach. Seven midwives where interviewed. The interviews where analyzed based on a qualitative content analysis. Result: Support from colleagues strengthen midwives’ confidence in their ability to provide support to abused women. They described a well-functioning cooperation between other professions and agencies, but frustration that the help offered in theory, was not always available. The midwives requested more education and that it should be given continuously to improve and maintain competence. Recommendations and guidelines from the workplace and screening instruments were perceived as facilitating, lack of time was described as an aggravating factor. Conclusion: Screening for domestic violence is complex and requires the right competence and a well-functioning cooperation between colleagues, other agencies and professions. Antenatal care units need to give midwives the right conditions in order to promote health

Full textENGLISH ABSTRACT: When twentieth century music composed specifically for children is discussed, Sergei Prokofiev’s Peter and the Wolf and occasionally Benjamin Britten’s The Young Person’s Guide to the Orchestra are the two examples most often mentioned, as if they represent the sum total of works in this genre. It seems then that there is a general lack of awareness of the actual existence of a rich variety of twentieth century music for children. Consequently the conclusion seems justified that most music educators in South African primary schools do not include this music in their repertoire nor has it been specifically recommended for use in the 2005 or 2011 syllabi. The question that prompted the current study is whether this repertoire of contemporary children’s music can be exploited to a much larger extent in South African primary schools than has been the case up until now. In the educational system of 2005, where ‘subjects’ have been replaced by ‘learning areas’, the former subject, Class Music, has been integrated into a new learning area consisting of four components (music, drama, dance and visual arts). However, this more comprehensive learning area has been implemented without actually making provision for more time on the time table than was formerly allocated to Class Music. With the implementation of CAPS in the Foundation Phase (Gr R – 3) in 2011 and in the Intermediate Senior Phase (Gr 4 - 9) in 2013, extra time allocated proves to still be insufficient for continual, thorough teaching of music. Although strong concern had been expressed previously about the fragmentation of subject content and the low status of music education in South African schools, this concern was not really addressed in both the curricula of 2005 and 2011, and subject content within the components is still scaled down to compensate for lack of time. This has also had serious consequences for the training of teachers. Requirements of the curriculum are only given in the form of general guidelines and teachers with no musical education, who have to teach the musical component, have no clearly defined examples to go by. For that reason a catalogue is included in this study by which teachers can gain access to relevant examples of twentieth century music. In the main section of this study music for children is examined in its historical context, it is defined and categorised. Strategies by which composers attempt to find access to a child’s world are also examined. While it may be assumed that children are familiar with the tonal idiom of folk, sacred and popular songs, composers of twentieth century music see themselves challenged to introduce children to a modern musical idiom, not compromising accessibility and a child friendly approach. In addition to the general discussion of the extensive repertoire of twentieth century music for children, four examples are selected to examine more specifically how composers go about when composing such music. These are: Wir bauen eine Stadt (Paul Hindemith), l’Histoire de Babar (Francis Poulenc), Die Weihnachtsgeschichte (Carl Orff) and The little sweep (Benjamin Britten). (Die Weinachtsgeschichte was translated into the Griqua dialect by Prof Hans du Plessis especially for the purpose of this study). The educational potential of twentieth century music for children is vast. Even with minimal teaching time it can be an efficient and time saving medium of tuition that at the same time opens up an exciting contemporary sound world to the young learner.
The little sweep (Benjamin Britten).

Life cycle assessment (LCA) is one methodology for assessing a product's impact on the environment. LCA has grown in popularity recently as consumers and governments request more information concerning the environmental consequences of goods and services. In many cases, however, carrying out a complete LCA is prohibitively expensive, demanding large investments of time and money to collect and analyze data. This thesis aims to address the complexity of LCA by highlighting important product parameters, thereby guiding data collection. LCA streamlining is the process of reducing the necessary effort to produce acceptable analyses. Many methods of LCA streamlining are unfortunately vague and rely on engineering intuition. While they can be effective, the reduction in effort is often accompanied by a commensurate increase in the uncertainty of the results. One nascent streamlining method aims to reduce uncertainty by generating random simulations of the target product's environmental impact. In these random Monte Carlo simulations the product's attributes are varied, producing a range of impacts. Parameters that contribute significantly to the uncertainty of the overall impact are targeted for resolution. To resolve a parameter, data must be collected to more precisely define its value. This research project performs a streamlined LCA case study in collaboration with a diesel engine manufacturer. A specific engine is selected and a complex model of its production and manufacturing energy use is created. The model, consisting of 184 parameters, is then sampled randomly to determine key parameters for resolution. Parameters are resolved progressively and the resulting decrease in uncertainty is examined. The primary metric for evaluating model uncertainty is False Error Rate (FSR), defined here as the confusion between two engines that differ in energy use by 10%. Initially the FSR is 21%, dropping to 6.1% after 20 parameters are resolved, and stabilizing at 5.8% after 39 parameters are resolved. The case study illustrates that, if properly planned, a streamlined LCA can be performed that achieves desired resolution while vastly reducing the data collection burden.
