Our work falls within the field of high resolution spectroscopy of gas phase molecules in the far-, mid- and near-infrared ranges. Its guiding line are absorption spectral intensities, dealt with experimentally and theoretically. Initially, a method of simultaneously determining the cis and trans content, iodine value and saponification number of neat fats and oils using a heated transmission flow cell was developed. High resolution absorption spectra of the IF molecule in the X¹Σ⁺ ground state were observed with the Kitt Peak Fourier transform spectrometer in a F₂/I₂ flame. Iodine fluoride is the most unstable interhalogen compound. Accurate wavenumber measurements (±0.0002 cm⁻¹) were made for the 1-0, 2-1 and 2-0 bands and molecular constants were derived. The observation of a high resolution emission spectrum at 13 μm (750 cm⁻¹) is unusual. Seven bands (1-0 through 7-6) of the main isotopic form, ²⁸Si³²S, were observed along with three bands (1-0, 2-1 and 3-2) for each of the minor species, ²⁹Si³²S, ³⁰S

Abstract:
The application of infrared imaging employing focal plane array Fourier transform infrared (FPA-FTIR) instrumentation for the identification of bacteria was investigated. FPA-FTIR spectroscopy was shown to provide new opportunities for bacteria identification with unprecedented reliability and throughput by allowing 102--103 FTIR spectra to be acquired simultaneously from surface areas of 90 x 90 to 200 x 200 mum with a spatial resolution of &sim;6 mum. The combination of data redundancy and spatial resolution afforded by infrared imaging made it possible to acquire highly reproducible spectra

Bladder cancer is one of the most common urogenital cancers worldwide. Two techniques commonly used for bladder cancer diagnosis are urine cytology and cystoscopy. The application of Fourier transform infrared (FTIR) spectroscopy in the assessment of oil quality and stability and the determination of the degree and type (cis or trans) of unsaturation of fats and oils was investigated. FTIR spectroscopy was shown to provide a rapid means of monitoring changes in oils undergoing oxidation or subjected to thermal stress. Two new techniques for the study of biological molecules using vibrational spectroscopy have been investigated, and are presented in this thesis in two parts.<br>In Part I, the application of centrifugal vacuum techniques for sample preparation in FT-IR spectroscopy is demonstrated.
