Abstract:
The aim of this paper regarding Pavol Strauss‘ Literary Work in the Central European Context is to introduce Pavol Strauss as a man and a writer, who was with his heart and soul a Central European. He had a strong bound to this territory spatially and temporally. At first, he was chased as a Jew, later on in the age of 30 he converted into Catholic. During the period of Stalinism, they considered him a second-class citizen, and he was wrongly called the clero-fascists however he was a believing Catholic. He was literally active in the second half of the 1930s, when he published two poetic collections in German. His poems in German were influenced by avant-garde which he became in touch with in Prague – its literary atmosphere defined Pavol Strauss’ literary experiments and further orientation. When speaking of Prague, also think of Franz Kafka, Franz Werfel and, of course, Reiner Maria Rilke. From the literary point of view, Pavol Strauss with his literary essays reached the highest level along with famous authors (R. Musil, K. Kraus, H. Broch). For his 82 years long life, Pavol Strauss cured many patients and left rare books of a unique philosophical dimension.

Abstract:
The Australian monospecific genus Palaeoscydmaenus Franz is revised based on the type material of P. australiensis Franz. The original placement of Palaeoscydmaenus in the tribe Scydmaenini is regarded as erroneous and evidence is provided to support a formal transfer of this genus to Cyrtoscydmini. Diagnostic characters of Scydmaenini and Cyrtoscydmini are discussed, Palaeoscydmaenus is redefined, and P. australiensis is redescribed.

Abstract:
In the first part discusses the contribution the question of so-called ‘progressive’ educational innovations and changes that can only work, however convincing, if they are based on a secure basis of legitimacy and argumentation. We must protest against that in education, ‘fashion’ adapted ideology to ideas, even fiction is highly stylized, which is used in its logical consequences rather the confusion in the structuring and specialized language, such as Franz Hofmann already sends a reminder. Knowledge from the history of science is the starting point of our deliberations. The second part focuses with the practical consequence, the relationship between teaching method and personal settings of the teachers and the associated impact on the learning environment. It addresses significantly shortcomings of current work, and tries to imagine about that ‘antitype’ of teaching in the form of a traditional, problematic conception of teaching, one of the current teacher ethos characterized teaching on the basis of historically grounded, and the times soon technical and professional competences.

"A new species of Plaumanniola Costa Lima in French Guiana (Coleoptera: Staphylinidae)." Zootaxa 4508, no. 3 (October 31, 2018): 431. http://dx.doi.org/10.11646/zootaxa.4508.3.6.

Full text
Abstract:
The genus Plaumanniola Costa Lima, 1962 was a subject of several taxonomical and morphological studies. It is one of very few scydmaenines originally placed not only in a different beetle family, but even a superfamily. The first species of Plaumanniola, P. sanctaecatharinae Costa Lima, 1962 was confused with myrmecophilous spider beetles and placed in a new subfamily Plaumanniolinae of Ptinidae. Transferred to Scydmaenidae by Lawrence & Reichardt (1966), it remained placed as a separate subfamily, and after merging the ant-like stone beetles with Staphylinidae, as a tribe Plaumanniolini of Scydmaenitae (Newton & Franz 1998; Grebennikov & Newton 2009). Franz (1990) redescribed P. sanctaecatharinae, but Jałoszyński (2013) found his redescription to have been based on a misidentified new species, which he described as P. regina Jałoszyński, 2013. The latter author illustrated previously unknown morphological structures, redefined Plaumanniola and discussed its possible close affinities with Glandulariini (Jałoszyński 2013). The unusually stout and flattened body, with especially flat and broad head (interpreted as possible adaptations towards myrmecophily, e.g., Lenko (1972)), differentiated Plaumanniolini from all other Scydmaeninae, but no other characters were found to support a separate position of this monogeneric tribe. Shortly later, a relatively slender P. simplicissima Jałoszyński, 2014 was described on the basis of material collected in Bolivia. Finally, a discovery made by Jałoszyński (2016) that Euconnus octosetosus Franz, 1994, was in fact a strikingly slender member of Plaumanniola, and that the aedeagus of Plaumanniola highly resembles that of Euconnus (Rhomboconnus Franz, 1986), lead to placing Plaumanniolini as a junior synonym of Glandulariini. It is possible that in future Plaumanniola will be reduced to a subgenus of Euconnus.

Abstract:
For orally administered drugs, control of the extent and rate of absorption depends on two important steps: solubility of the drug in physiological liquids and their permeability across biological membranes. Thus, the Biopharmaceutics Classification System (BCS) has been proposed as a tool for the development of new drugs, new formulations and aid in the biowaiver processes. However, another factor related to bioavailability that should be considered in biopharmaceutic studies is the metabolism. Thus, the Biopharmaceutics Drug Disposition Classification System (BDDCS) has been proposed for drug classification according to their solubility and metabolism characteristics, so it is possible to evaluate and predict the in vivo behavior of a compound. Metabolism has been extensively investigated, especially cytochrome P450 enzymes, which are also expressed in enterocytes. Besides, BDDCS provides support in evaluating the permeability mechanisms involved in the absorption processes, drug-drug interactions and drug-food interactions. Thus, the present study aimed to evaluate the mechanisms of permeability of antiretroviral drugs through the ex vivo (Franz cells) and in vitro (PAMPA, MDCK-MDR1 and microsomes) models considering aspects related to the intestinal metabolism and efflux of these drugs. Given the importance of the use of antiretroviral drugs in drug therapy against Acquired Immune Deficiency Syndrome (AIDS) and that these drugs are usually administered in a long-term way, understanding the mechanisms involved in the permeability is of a great importance, since they are not totally elucidated and no information is found in the literature. In addition, drugs as stavudine, lamivudine and zidovudine indicate variation in the permeability, which require further scientific investigation of absorptive processes. Thus, jejunum segments from rats were used to evaluate the intestinal permeability of these antiretroviral drugs, considering the evaluation of efflux by P-glycoprotein and intestinal metabolism by CYP3A. In a complementary manner, in vitro studies using parallel artificial membranes (PAMPA) and cell cultures MDCK-MDR1 were performed to aid in the elucidation of the permeability mechanisms of antiretroviral drugs. Also, the evaluation of the metabolism was carried out using microsomes to verify if such substances are substrates of CYP3A, and verify the impact of the intestinal metabolism in the absorption. The permeability results obtained in PAMPA were: 0.74±0.11x10-6 cm/s for stavudine, 0.25±0.12x10-6 cm/s for lamivudine and 1.14±0.25x10-6 cm/s for zidovudine. In ex vivo method using the intestinal segments in Franz cells, the results were: 1.56±0.32x10-5 cm/s for stavudine, 1.26±0.27x10-5 cm/s for lamivudine and 2.54±0.49x10-5 cm/s for zidovudine. Thus, based on the results obtained from these two methods, it is suggested that the antiretroviral drugs present other transport mechanism that is different from transcellular passive diffusion. For efflux studies, results obtained from experiments performed in Franz cells shown the increase of the permeability of the three antiretroviral drugs when the P-gp inhibitor was used: from 15.6x10-6 to 42,5x10-6 cm/s for stavudine, from 12.6x10-6 cm/s to 37.5x10-6 cm/s for lamivudine, and 25.4x10-6 to 56.6x10-6 cm/s for zidovudine. In MDCK-MDR1, the permeability results were used for obtaining ratio values between the directions B→A and A→B. The Papp values obtained with 33 inhibitor shown a ratio less than 1. For ratio B→A/A→B for each drug in experiments without inhibitor, the values obtained was equal or greater than 2, which shows the interaction between drug and transporter. Based on that, the ex vivo model using intestinal segments in Franz cells seems to be adequate for evaluation of efflux mechanism of antiretroviral drugs, which was confirmed by MDCK-MDR1 studies. Thus, the antiretroviral drugs presented interaction with P-gp. For metabolism studies in intestinal segments in Franz cells, a wide range of standard deviation was observed for the three antiretroviral drugs when the CYP3A inhibitor was used: from 15.6x10-6 cm/s to 23.5x10-6 cm/s for stavudine, from 12.6x10-6 cm/s to 27.3x10-6 cm/s for lamivudine, and from 25.4x10-6 cm/s to 40.5x10-6 cm/s for zidovudine. In experiments in microsomes, the results of metabolization in the absence and presence of CYP3A inhibitor were: from 16.56 to 19.79% for stavudine, from 14.56 to 15.55% for lamivudine and from 17.85 to 16.48% for zidovudine. Based on that, it is suggested the use of microsomes for metabolism evaluation, since the ex vivo method presented high variability between the results obtained. For each drug, no significative influence in pre-systemic metabolism related to CYP3A enzymes was observed, which indicates that the oral absorption of the drugs is not limited by these enzymes. The use of different methods in this work allowed to understand the mechanisms involved in the transport of antiretroviral drugs, which is of a great relevance in drug development and in the understanding of clinical events currently not clarified.

Abstract:
McClernand’s messages led to another round of Federal attacks on the afternoon of May 22. First, at 2 p. m., two brigades from Frank P. Blair’s Fifteenth Corps division, led by Giles A. Smith and Thomas Kilby Smith, combined with Thomas Ransom’s brigade of the Seventeenth Corps to attack the Confederate line between Stockade Redan and 3rd Louisiana Redan. They made it as close as twenty-five yards from the parapet before enemy fire stopped them. Then Joseph A. Mower’s brigade of James M. Tuttle’s Fifteenth Corps division attacked in column along Graveyard Road toward Stockade Redan. It faired about as well as Hugh Ewing’s brigade had done that morning, with only a handful of its members making it to the exterior slope of the redan. These afternoon attacks only served to prove that frontal assaults against this part of Samuel H. Lockett’s defence line, well manned by Confederates who were on the alert, were futile. These attacks completely failed to draw defenders from the Thirteenth Corps sector because the division commanders here had enough men to hold their works.

Abstract:

Waiho – (verb) (-ngia,-tia) to let be, leave alone, put, place, ignore.

SH6 Waiho Bailey Bridge is located just south of Franz Josef township in the South Island of New Zealand and is a critical connection for the West Coast. The Bailey bridge was first constructed in 1990 and has since been raised and extended three times due to significant aggradation of the riverbed. During a massive storm event on March 26, 2019 the northern abutment and northern- most pier were washed out leading to collapse of several spans of the bridge. The cost caused by the loss of the bridge was estimated to be in the order of $2-3M per day. Consequently, there was intense pressure on Waka Kotahi NZ Transport Agency to restore access across the river.

As Team Leader and Deputy Team Leader of the West Coast Bridge Management Contract, Jeremy Waldin and Ben Baty led the $6.5M emergency recovery managing an emergency response team which worked across multiple organisations to recover this 170m long bridge in just 18 days.

Abstract:
While the urban fabric has both three and two dimensional aspects, we rarely if ever experience urban form as a fully three-dimensional object nor as a two-dimensional ground plan. Rather, the urban fabric is experienced more in terms of places with a fractal dimension lying between two and three. Hence we can (re)interpret urban form elements from colonnades to streets as ‘fractal’ types. This in turn suggests the possibility for a ‘fractal’ typo-morphology, addressing elements that occupy a typological space ‘in between’ 3D architectural modelling and 2D urban plan analysis. While Moudon could note that aspects of type were ‘vague and flawed with ambiguity’ (1994), it seems that there is still room for clarity; Dovey’s recent review of type (2016) invites further development and integration of the concept of type what may still be a disparate and ambiguous territory. Meanwhile, fractal approaches to morphology have been traditionally more concerned with larger scale urban patterns (e.g. Frankhauser, 2004) or measurement of architectural elements (e.g. Ostwald et al, 2015) with a fractal dimension less than two. This paper explores the possibilities for establishing a ‘fractal typo-morphology’ that recognises the ‘2.x’ dimensional aspect of the urban fabric and its component types. The paper generates a solution-space of types, illustrated with empirical examples, and organizes these into a typology for onward use, so that ideas of type, form and fractal dimension can contribute more fully as ‘conceptual tools’ both for understanding the urban fabric and for use as building blocks for urban design. Abstract:
The activity of plasminogen activators (PA) is regulated in part by PA-inhibitors. Two distinct PAIs have been identified: PAI-1 and PAI-2. To be able to measure these proteins specifically we have purified PAI-1 and PAI-2, raised antisera and developed specific radioimmunoassays (RIAs), with detection limits in plasma of 10 and 15 ng/ml, respectively. In a population of over 350 individuals, PAI activity correlated well with PAI-1 :Ag (r=0.699), but not with PAI-2:Ag (r=0.070). In healthy individuals PAI-1 concentrations varied considerably (between 10 and 85 ng/ml, median 29 ng/ml). During venous occlusion (VO) applied in 40 healthy individuals, average PAI-1 in plasma increased fran 27 before, to 43 and 59 ng/ml after 10 and 20 min VO (p <0.001, Wilcoxon test). Overall fibrinolytic activity increased 15 fold, and t-PA:Ag levels eightfold. Increased PAI activity and PAI-1 antigen levels with respect to controls were found in patients with cardiovascular or thromboembolic disease, malignancies, hepatic insufficiency, after major trauma and in the postoperative period. This wide spectrum of pathologies with increased PAI-1 levels supports previous suggestions that PAI-1 behaves as an acute phase reactant. In patients undergoing extracorporeal circulation PAI activity and PAI-1 antigen were measured before and 1 h, 1 d and 7 d after the operation. Highest values of activity and antigen were observed 1 h postoperatively. These values remained elevated the day after the operation, but returned to preoperative levels within 7 days.PAI-2 antigen concentrations were at or below the detection limit in all controls and the majority of patients. Very high levels of PAI-2 (above 30 ng/ml) were only observed in pregnant women and in patients with hepatocellular carcinoma. The specific measurement of PAIs thus show increased levels in a variety of clinical conditions. The availability of specific RIAs permits to conduct prospective studies and to evaluate to what degree elevated PAI-1 and PAI-2 concentrations are correlated with the stage of the disease, its prognosis and the risk to develop thromboembolic complications.

Abstract:
Overview The adhesion-aggregation of platelets to a site of vessel wall injury is a quintessential blood flow phenomenon. Firstly, platelets are driven to the vicinity of the vessel wall by a form of convective diffusion in which red cells both mechanically augment the effective platelet diffusivity (Turitto et al., Ind. Eng. Chem. Fund. 11:216-223, 1972; Grabowski et al., Ind. Eng. Chem. Fund. 11:224-232, 1972) and enhance the near-wall piatelet concentration (Ti11es and Eckstein, Microvasc Res., In press, 1987). Secondly, red cells subjected to physiologic shear forces are capable of secreting sufficient adenine nucleotides to induce primary platelet aggregation without themselves undergoing frank lysis (Reimers et al, Blood 64:1200-1206, 1984). This "humoral" effect of erythrocytes is likely to contribute to primary hemostasis in a shear stress-dependent manner. Thirdly, endothelial cells are able to modulate platelet aggregation at a site of vessel injury by producing prostacyclin (and perhaps other antithrombotic substances) in a manner which increases with vessel shear rate (Grabowski et al, Blood 62:301a, 1983); production for a large range of arterial shear rates appears to be limited by plasma-borne substrate (arachidonate). This manner of production ensures a concentration of prostacyclin in the near-wall region which remains relatively independent of shear rate.Imaging primary hemostasis. In our work, epi-fluorescence videomicroscopy has allowed real time imaging of platelet adhesion-aggregation to a simulated vessel wall injury. The injury model is an endothelial cell monolayer (ECM) across which, prior to ECM exposure to flowing blood, a 6-0 sterile suture is drawn in a direction transverse to flow. Microinjuries result which measure 70 ± 15μm (Mean ± SD) in width. The fluorescent label is the TAB murine monoclonal antibody (courtesy of Dr. R.P. McEver) directed against human platelet GPIIB, together with a fluorescein-conjugated goat F(ab')2 against murine inmunoglobulin. The injured ECM's, grown to confluence on rectangular cover glasses precoated with microfibrillar collagen, comprise one wall of a flow chamber mounted on a vertical microscope stage. On microinjury sites and at shear rates of 100 to 700 sec-1, computer-enhanced video images show adherence, remodelling and growth of chains of platelet aggregates. Aligned with the flow direction, these chains have a spacing of approximately 30)im, a length similar to the average endothelial cell diameter. One may speculate that such chains provide a scaffold for wound healing insofar as they are likely rich in agents chemotactic for leukocytes and in platelet-derived growth factor.Modulatory role of endothelium. When the ECM's are pre treated with 1.0 mM FC lysine acetyl sal icy late (LA), aggregate length increases (P<0.001) up totwo-fold, outflow levels by RIA of serum thromboxane B2 increase (8 of 8 paired runs), and outflow levels of prostacyclin by RIA for 6-Keto PGFiot decrease (5 of 7 paired runs). The Table gives data for one of four similar experiments at 270 sec-1 and following five minutes of flow. These data imply that products of ECM which are inhibitable by aspirin modulate local adhesion-aggregation; their inhibition, as by vasculitis or drugs, may give rise to thrombotic states.Bleeding disorders. Aggregate length is reduced in von Willebrand's disease (4 patients), Hermansky-Pudlak syndrome (2 patients), and after 300 mg oral aspirin (Tablet 4 donors). The reduction in the first two, however, is greater (P<0.01) than that for oral aspirin. With oral aspirin, further, there is a paradoxic increase in the percent platelet coverage of the injury area. Summary. Rheology has profound effects on the rate, structure, and modulation of primary hemostasis. Many of these effects can be studied via real-time, epi-fluorescence videomicroscopy of platelet adhesion-aggregation to a site of injury to an endothelial cell monolayer exposed to flowing blood. The model described has application to the study of thrombotic and hemostatic disorders and unstable angina.
