Full textEsto permite verificar así la influencia de la necesidad de identificación de la ciudad con su frente costero y cómo las reformas urbanas aluden a un modelo de ciudad relacionado a un contexto más amplio, sin olvidar la formación de los distintos iconos urbanos que han ido determinando la imagen de esta fachada irresuelta de la ciudad, un aspecto que enlaza con uno de los temas suscitados hoy por la crítica arquitectónica: el de la construcción de la ciudad mediante edificios-imagen.
La tesis no propone únicamente seguir de nuevo las líneas de transformación de las infraestructuras portuarias y de la progresiva separación de la ciudad respecto a su fachada marítima-un proceso que ha sido "modelizado" por algunos autores desde los años setenta. El texto trata más bien indagar sobre, en una óptica de la larga duración, las tensiones presentes en el interior de esta dinámica, y utiliza para ello fuentes diversas, fuentes entre las cuales los distintos proyectos que intentan dar respuesta a estas tensiones se convierten en los documentos privilegiados.
El arco cronológico de la tesis abarca desde mediados del siglo XIX, cuando se aprueba el proyecto del Ensanche y el proyecto para la modernización del puerto de José Rafo, hasta mediados de los años sesenta del siglo XX cuando la formulación del llamado Plan de la Ribera es, al mismo tiempo consecuencia de la propia evolución de las infraestructuras portuarias y de todo el sistema de comunicaciones, y el anuncio del final de un largo ciclo de divorcio entre la ciudad y el mar.
En el mismo período de la modernización técnica del puerto, se percibe la emergencia de nuevas actitudes con respecto al mar, y a las actividades deportivas que propicia, pero también se registran otros procesos, como la formación del tendido ferroviario, las implantaciones industriales, o la extensión de un sistema de alcantarillado que comprometen seriamente los beneficios higiénicos y los atractivos potenciales que se le reconocen al frente marítimo como fuente de salud y de embellecimiento de la ciudad. Los distintos proyectos ponen en evidencia la constante aspiración a recuperar un contacto digno con el mar y comprueban al mismo tiempo los cambios de actitud y de referentes que van sucediendo a lo largo de los años.
The thesis proposes to give a vision of the long duration in the urban processes of reconversions involved in the redefinition of the marine front of harbor cities and to investigate the reasons for structural changes in the definition of Barcelona's marine facade, as a case study, to analyze the conflicts and the viscosity of the transformation processes.
In this manner, therefore, it allows us to verify the influence of the necessity of the identification of the city with its coastal facade, and how urban reforms allude to a model of a city related to an ampler context, without forgetting the formation of the different urban icons that have been determining the image of this unresolved facade of the city, an aspect that connects today with one of the subjects evoked by architectural critics: that of the construction of the city by means of buildings that are images/symbols.
The thesis does not only propose solely to again follow retrace the lines of transformation of harbor infrastructures and the progressive separation of the city with respect to their its marine facade--a facade process that has been "standardized" by some authors from the Seventies1970s. The text, rather, treats to investigates on, from the viewpoint of the long duration, the tensions that exist within this dynamic, and utilizes various sources, causing the different projects that try to solve these tensions to become privileged documents.
The time frame of the thesis is from the mid-nineteenth century, when the Ensanche project and Jose Rafo's project for the modernization of the port are approved, until the mid-sixties of the twentieth century, when the formulation of the so-called Shore Plan (Plan de la Ribera) is, at the same time, a consequence of the evolution of the harbor infrastructures and the entire system of communications themselves, and the announcement of the end of a long cycle of a divorce between the city and the sea.
In the same period of the technical modernization of the port, new attitudes with respect to the sea, and the sport activities that it causes, emerge. But other processes are also registered, such as the building of railways, industrial complexes, or the extension of a sewage system that seriously jeopardizes the hygienic benefits and the potential attractions that are attributed to the marine front as a source of health and beautification of the city. The various projects demonstrate the constant aspiration to recover a useful interaction with the sea, and at the same time verify the changes of attitudes and references that follow one another throughout the years.

Full textThe research aim was to investigate the experiences of people and families who are experiencing the early stages of dementia, in order to understand which specific helps could be provided to support people, with their particular needs, in this first and delicate phase of the disease. Considering its field of investigation and its object, this research has been particularly suited to the application of the participatory research methodology. The research in the entire research process has actively involved people who have a direct, personal or job-connected experience of the disease. Tools as a semi-structured interview and a diary has been used to investigate the point of view of family caregivers and their elderly relatives with cognitive impairment. The results obtained have been multiple, heterogeneous and allowed us to understand the experiences of the family caregivers and of the subjects who face the disease directly. These results have been divided by different areas and have been organized using concept maps. Six maps have been focusing on the point of view of the caregivers and four maps focusing on the experience of the elderly. The results allowed the researcher to understand different elements: what caring means for the caregivers and what to be assisted means for the elderly with cognitive impairment; the attitudes and emotions of both players about the disease; the role of the family and other significant figures in this situation; the relationship with the services and how families face this problem.

Full textThe lower third of the face exerts a strong influence on the aesthetics of individuals, and knowing the importance of its balance, and the orthodontist\'s contribution to the diagnosis of the facial profile, the present study had the purpose to test a method of correlation between a linear measure located from the glabella region with linear anteroposterior measurements on the lower third of the face (Triviño, 2012), and verify the predictability of anteroposterior proportions in the lower third of the face, from a reference line through a point in the glabella region. Anatomical tracings from lateral cephalometric radiographs of 120 individuals with balanced profile, leucoderm and class I were performed in order to establish cephalometric parameters to correlate anteroposterior references of the lower third of the face from the lines Sv-PRT and PRTv, obtained through the PRT point. The applicability of the method was performed in a group of 24 individuals with an unpleasant profile, in which it considered the predictability formulas that used the Sv-PRT line as an unsatisfactory reference to determine the anteroposterior proportions of the lower third of the face in some patients. The method that considered the PRTv line as a reference, using Pearson\'s statistical methods of correlation and multiple linear regression, presented statistically significant results for all reference values in females and males (PRTv-Ls, PRTv -Li and PRT-Pg\'). The Student t-test was used to verify that the measures were not statistically significant according to gender. The results of this study showed that it is coherent to consider the glabella region as an evaluation and diagnostic reference for the anteroposterior proportions of the lower third of the face.

O câncer de laringe ocorre em 25% dos carcinomas de cabeça e pescoço e compreende 2% de todas as doenças malignas. É comum o aparecimento de segundos tumores primários e, aproximadamente, 5% dos pacientes apresentam cânceres sincrônicos. Há várias evidências indicando que a população celular presente no fronte de invasão possui características moleculares diferentes das áreas tumorais superficiais, tornando esta região importante para avaliação prognóstica. Neste estudo foram investigadas por CGH cromossômico de alta resolução (HR-CGH) as alterações genômicas na área superficial e no fronte de invasão de carcinomas de laringe e selecionadas regiões específicas para serem avaliadas por outras metodologias para a confirmação dos resultados. O componente superficial e o fronte de invasão de 33 carcinomas de laringe fixados em formalina e em blocos de parafina foram avaliados por HR-CGH. Foram detectadas alterações comuns aos dois componentes assim como alterações exclusivas a cada um deles. Adicionalmente, foi investigada e confirmada a expressão aumentada da proteína ciclina D1 o gene CCND1 esta mapeada em 11q13) por análise de expressão em plataformas de microarranjos de tecidos contendo as areas do tumor e do fronte de invasão. Foi realizada também a análise de marcadores polimórficos de microssatélites mapeados em 3q e 18q em um grupo independente de 33 amostras (DNA tumoral e do sangue periférico) cujos resultados confirmaram as perdas encontradas nestas regiões cromossômicas. A expressão do gene CTTN (mapeada em 11q13) e de sua proteína foram avaliadas e revelaram que os altos níveis de expressão proteica foram correlacionados com invasão perineural nas células do fronte de invasão, sugerindo que esta área pode ser considerada como ferramenta prognóstica em carcinomas de laringe. Laryngeal cancer occurs in 25% of head and neck carcinomas and comprises 2% of all malignant diseases. The appearance of secondary tumors is common and approximately 5% of patients present concurrent cancers. Preliminary evidence indicates that the cell population present at the source of the invasion front possesses distinct molecular characteristics to surface tumor areas, making this an important region for prognostic evaluation. In this study, genomic alterations in superficial areas and the invasion front of laryngeal carcinomas were investigated by high resolution chromosomal comparative genomic hybridization (HR-CGH) and specific areas were selected for evaluation by other methodologies to confirm the results. The superficial and invasion front components of 33 laryngeal carcinomas fixed in formalin and embedded in paraffin blocks were analyzed by HR-CGH. Alterations common to both components and those exclusive to each area were detected. Additionally, increased ciclin D1 (gene is mapped at 11q13) protein expression was confirmed through immunohistochemistry analysis of tissue microarray platforms containing superficial tumor and invasion front tissues. Polymorphic microsatellites markers mapped at 3q and 18q was also performed on an independent group of 33 samples (tumor and peripheral blood DNA); the results confirmed the losses verified in these chromosomal regions. Expression of the gene CTTN (mapped at 11q13) and its protein were evaluated revealing that high levels of protein expression were correlated with perineural invasion in invasion front cells, suggesting that this area could be considered a prognostic tool in laryngeal carcinomas. The gains detected at 2q24 by HR-CGH were confirmed by FISH and transcript analysis by qRT-PCR. Pacientes com carcinoma espinocelular (CEC) de lábio apresentam prognóstico favorável em comparação aos pacientes com a lesão intraoral. O estadiamento clínico da lesão é a principal forma de avaliação do prognóstico do CEC oral, além dos sistemas de gradação histológica dos tumores. A análise do budding tumoral (BT) e do infiltrado inflamatório na região do fronte invasivo tumoral (FIT) podem configurar-se como fatores prognósticos complementares, já que para alguns tipos de câncer, entre eles o CEC intraoral, BT positivo, contagem elevada de neutrófilos no tumor, sangue ou ambos e razão neutrófilos/linfócitos (RNL) aumentada representam um pior prognóstico para os pacientes. O objetivo deste estudo foi avaliar a presença e a RNL no FIT, bem como a associação desses achados com o grau histológico de malignidade (GHM) dos tumores e o BT em uma amostra de CEC de lábio. Revisou-se os arquivos do Centro de Diagnóstico das Doenças da Boca da Faculdade de Odontologia da Universidade Federal de Pelotas, buscando-se lesões submetidas a biópsias excisionais de CEC em lábio inferior e classificando-as segundo um sistema de classificação do GHM. Realizou-se técnica imunoistoquímica com os anticorpos CD66b e CD3 para avaliação de neutrófilos e linfócitos, respectivamente. O anticorpo AE1/AE3 foi empregado para a classificação do BT. Neutrófilos CD66b+ e linfócitos CD3+ foram analisados semi-quantitativamente no FIT. De um total de 43 casos, 36 (83,7%) eram tumores bem diferenciados e 32 (74.4%) não apresentavam BT. Observou-se baixa infiltração de neutrófilos e alta de linfócitos (índices 0,8 e 2,0, respectivamente) na região do FIT, acarretando baixa RNL na mesma região. Portanto, a alta diferenciação tumoral, os poucos casos exibindo BT positivo e a baixa RNL no FIT podem estar associadas ao melhor prognóstico do CEC labial.
Patients with lip squamous cell carcinoma (LSCC) present favorable prognosis compared to those with intraoral lesion. Clinical staging of lesion is the main way of assessing the prognosis of oral squamous cell carcinoma (OSCC), followed by the histological grading systems. Analysis of tumor budding (TB) and the inflammatory infiltrate in the tumoral invasive front (TIF) can be configured as additional prognostic factors, since for some types of cancers, including intraoral SCC, findings as TB positive, high counting of neutrophils in tumor, blood or both and increased neutrophils/lymphocytes ratio (NLR) represent a worse prognosis for the patients. The aim of this study was to evaluate the presence and the NLR in the TIF, and the association of these findings with the histological malignancy grading (HMG) of tumors and the TB in a LSCC sample. The files from the Center of Diagnosis of Oral Diseases, Dental School, Federal University of Pelotas were reviewed searching specimens from excisional biopsies of lower LSCC (LLSCC) and classifying them according to a HMG system. Immunohistochemistry was performed with CD3 and CD66b antibodies for evaluation of neutrophils and lymphocytes, respectively. The antibody AE1/AE3 was used for the identification of TB. CD66b+ neutrophils and CD3+ lymphocytes were analyzed semi-quantitatively in the TIF. From a total of 43 cases, 36 (83.7%) represented well differentiated tumors, 32 (74.4%) showed none TB. There were low neutrophil and high lymphocyte infiltrations (indexes 0.8 and 2.0, respectively) in the TIF, leading to a low NLR in the same region. Therefore, the high differentiation grading, the few BT positive cases and the low NLR in the TIF may be associated with better prognosis in LSCC.

Full textThis study aims to explore existing paradoxes in the offer of food sustainable products and consequently consumers’ behaviours (coping strategies) (Lazarus and Folkman, 1984) at the time of this perception. For this purpose, the theoretical model of Mick and Fournier (1998), built initially for studying the paradoxes of modern technologies, were adapted to sustainable food consumption. At methodological level, a qualitative survey was conducted among 84 “ordinary” consumers in France and Italy. Later other 18 subjects, usually involved into sustainable purchases (participants of AMAP, GAS), were interviewed. The semi-structured interviews shed light on some differences between “ordinary” consumers of the two countries when they adopted coping strategies to go beyond perceived paradoxes. Thus we proposed three ordinary consumers’ profiles. On the contrary engaged individuals are like a more homogeny group going over national boundaries. They agree with paradoxes and sustainable practices. In addition, the research pointed out some divergent aspects connected with specific context, for instance market of sustainable food products, communication over this offer and social cultural characteristics. Finally, this work had three objectives: theoretical to verify the paradoxes listed and evocated consumers’ strategies by French and Italian consumers; methodological regarding the adaptation of the conceptual model “gears” of Mick et Fournier (1998) to the context of sustainable consumption; and then practical ones such as identification of barriers for developing sustainable consumption and specific possible ways of growth: more attention to environmental aspects in France (“filière nord/nord” of organic, fairly, local French peasants) and ethics products in Italy (products issued from social economies).

Full textThe aim of this work is to study the evolution of the British Imperial security strategy. The work investigates the period between the end of the 18th Century and the beginning of the 19th. In the light of the evolution and the international contest that created it, it is studied the strategy applied by London during the First World War and will be analysed the case of the British Army’s expedition in Italy after the battle of Caporetto. The thesis shows that the immobility of the western front led the British Government to seek new fronts in order to gain its strategic objectives. The study shows how Great Britain interpreted the Italian front such as a tool to take resources away from Germany. After Caporetto the British came to Italy to support the Italian Army, they tried to use it to exclude Austria-Hungary from the conflict and to close off Germany. The thesis proves that, by the British point of view, the Italian front was an example of expeditionary strategy, but it became an integrated part of the Imperial British strategy only when Italy isolated Germany and organised its invasion from the South, after the battle of Vittorio Veneto.

Full textThis analysis addresses multilevel forces present in the assesment of choice factors from a qualitative point of view and highlights similarities and differences between experts and ordinary people in decision-making behavior, relating to economic choices and evaluating actions in everyday life. Systematic reviews of the literature, Conjoint Analysis and a questionnaire with items proposing a series of decision-making scenarios were conducted. The results show that decision-makers with different degrees of economic expertise, through similar mental processes, reach to different conclusions. The subjects are distinguished by the importance they attribute to the same factors of the decision-making scenario, by categorizing, prototypiically or restrictively, options and by operating different strategies for using information. What seems distinguish an expert from an ordinary decision-maker is different cognitive skills and emotive heuristics used in way to organize knowledge

Full textInteraction between tumor cells and stroma plays a central role in cancer progression. Local growth and invasion of neoplasms depend on the controlled modification of components of the extracellular matrix. The invasive front has been considered the most aggressive part of a tumor whose cells have the greatest ability to invade surrounding tissue. Therefore, this is the most crucial area at the tumourhost interface. The aim of this study was to investigate the immunohistochemical expression and distribution of the basement membrane proteins laminin and collagen type IV, and the extracellular matrix proteins collagen type III, fibronectin and tenascin, in the invasive front region of lip SCC, the most frequent malignancy of the oral cavity, and compare the findings to the histological malignancy grading of the invasive front. Twenty cases of lip SCC, all showing solar elastosis, were studied. Histological grading of the invasive front was carried out according to the criteria proposed by Bryne et al. (1989), using 5?m sections stained with H&E. Immunohistochemistry reactions were performed in 3?m sections obtained from formalin-fixed, paraffin-embedded tissue, using the streptavidin-biotin-peroxidase method and appropriate antibodies. Of the 20 cases studied, 10 were graded welldifferentiated (WD), 7 moderately-differentiated (MD) and 3 poorly-differentiated (PD). Both laminin and collagen IV had similar results presenting frequent discontinuity. Almost every case showed at least some discontinuity in the invasive front. A few cases have showed a total loss of immunostaining in the area. It was not possible to determine a correlation between histological grade and degree of basement membrane discontinuity. Seventeen cases have shown total loss of collagen III expression in the invasive front. The three cases that presented positivity were represented by 2 WD and 1 MD SCC. Fibronectin was very abundant in all cases. It was present in a thick area starting from the basement membrane. Staining for tenascin was very variable among the cases and it was not correlated to the histological grading or intensity of inflammatory response. The results for laminin, collagen IV and III suggest invasive activity in the tumoral invasive front. Although the pattern of the proteins were altered in the invasive front of lip SCC in comparison to normal lining epithelium, these changes were not correlated to histological malignancy grade.

A área mais profunda e infiltrativa do CEC, conhecida como fronte de invasão, revela células neoplásicas malignas com menor grau de diferenciação comparadas à totalidade da lesão. Estudos recentes demonstram a influência do padrão do fronte de invasão tumoral no prognóstico de CEC de laringe e colo uterino. Contudo, não há estudos sobre o mesmo em relação a CEC cutâneos de cabeça e pescoço. Estudar as características clínicas, histopatológicas e perfil imuno-histoquímico do CEC de cabeça e pescoço e fronte de invasão tumoral, correlacionando-o com comportamento biológico. O estudo foi do tipo coorte retrospectivo, sendo selecionados os pacientes que tiveram diagnóstico de CEC de cabeça e pescoço confirmados pelo exame anatomopatológico no período de 1999 a 2009. A avaliação do fronte de invasão tumoral foi realizada empregando-se imuno-histoquímica para E-caderina, ki-67 e p53. Resultados: O tipo de fronte de invasão, bem como as expressões de E-caderina e ki-67, seja em sítio superficial ou profundo, não apresentaram associação com os critérios prognósticos histopatológicos. Os achados mostraram que a E-caderina, o p53 e o ki-67 foram mais expressos em tumores com tamanho ˃ 2cm. Objective: Investigate, on a comparative basis, the expression of the adhesion molecules E-cadherin (E-cad), β-catenin (β-cat) and the proliferation index (Ki-67) at the invasive tumor front (ITF) in Squamous Cell Carcinoma (SCC) and basaloid squamous cell carcinoma (BSCC). Material and Methods: Thirty-five SCC and 16 BSCC cases were evaluated by immunohistochemistry. Clinico-pathological data and survival data were evaluated and compared. Results: There was a low expression of E-cad in the cytoplasmic membrane (p = 0.50) as well as in the nucleus (p = 0.31) for both SCC and BSCC. A high expression of E-cad was seen in the cytoplasm for the SCC group (80%) when compared to the BSCC group (25%) (p<0.01). The expression of β-cat in the cytoplasmic membrane (p = 0.28) and in the cytoplasm (p = 0.44) was low in both SCC and BSCC groups. Both types of carcinoma presented low expressions of β-cat in the nucleus (p = 0.03). The Ki-67 expression was low irrespective of tumor variant. The high expression of E-cad in the cytoplasm was associated with T3/T4 tumors (p = 0.04) in the SCC group and there was no significant association of E-cad, β-cat, Ki-67 with the other clinical variables. In terms of disease-free survival and overall survival, there were no significant differences between SCC and BSCC. Conclusion: The E-cad-β-cat system was found to be dysregulated in both oral SCC and oral BSCC. The Ki-67 cell proliferation index was extremely low in the cases investigated and consequently had no prognostic value.
Objetivo: Investigar comparativamente a expressão das moléculas de adesão E-caderina (E-cad), β-catenina (β-cat) e o índice de proliferação (Ki-67) em Carcinoma Espinocelular (CEC) e Carcinoma escamoso basalóide (CEB). Material e Métodos: Nesse estudo, foram selecionados 35 casos de CEC e 16 casos de CEB e investigados por meio de técnica imuno-histoquímica. A associação das variáveis clinico-patológicas e dados de sobrevida foram avaliados. Resultados: Ambos os grupos tiveram baixa expressão de E-cad em membrana citoplasmática (p=0,50) e núcleo (p=0,31), essa associação não foi estatisticamente significante entre os grupos. Alta expressão de E-cad no citoplasma foi notada no grupo CEC (80%) quando comparado ao grupo CEB (25%), p<0,01. A expressão de β-cat em membrana citoplasmática (p=0,28) e citoplasma (p=0,44) foram baixos em CEC e CEB. Ambos os grupos tiveram baixa expressão de β-cat em núcleo, p=0,03. Não houve diferença estatisticamente significante quanto à expressão de Ki-67 entre os grupos. Alta expressão de E-cad em citoplasma foi associada a tumores T3/T4 (p=0,04) no grupo CEC. Pacientes estilistas apresentaram baixa expressão de β-cat em membrana citoplasmática (p=0,05). Não houve associação entre a expressão de E-cad, β-cat e Ki-67 com as demais variáveis clínicas dos grupos. A sobrevida livre de doença e a sobrevida global não foram estatisticamente significantes na associação entre os grupos CEC e CEB. Conclusão: O sistema E-cad-β-cat encontra-se desregulado tanto nos de CEC quanto nos de CEB de cavidade oral, levando as células epiteliais a perderem o seu fenótipo e promovendo a invasão e progressão tumoral. O índice de Ki-67 foi extremamente baixo não tendo valor prognóstico nos casos avaliados.

Resumo: Miofibroblastos são caracterizados pela expressão de niveis elevados de fatores de crescimento, proteases e proteinas da matriz extracelular, que podem influenciar a progressão tumoral. Recentes estudos encontraram miofibroblastos no estroma de varios tipos de canceres humanos, incluindo os de mama, rim, figado, bexiga, colon e prostata, sendo a presença dos miofibroblastos frequentemente associada a um pior prognostico. Os objetivos deste estudo foram avaliar a presença dos miofibroblastos em amostras de mucosa oral normal, leucoplasias com o diagnostico histologico de displasia e 2 grupos de carcinomas espinocelulares (CEC) orais e correlacionar a presença destas celulas com as caracteristicas cronico-patologicas dos tumores. O primeiro grupo foi formado por leses exclusivamente de lÃngua, enquanto que o segundo grupo foi composto por 38 CECs de várias localizações da cavidade oral. Adicionalmente, nós determinamos in vitro o efeito dos produtos da sÃntese das células de CEC oral sobre a transdiferenciação dos fibroblastos em miofibroblastos, bem como o efeito dos produtos de sÃntese dos miofibroblastos sobre a proliferação celular e atividade das metaloproteinases de matriz (MMP) nas linhagens celulares de CEC oral. A análise imunohistoquÃmica revelou a ausência dos miofibroblastos em amostras de mucosa oral normal e em lesões leucoplásicas com o diagnóstico histológico de displasia. Em contraste, miofibroblastos foram encontrados no estroma e/ou fronte invasivo de aproximadamente 60% das amostras de CEC oral. No primeiro grupo, a presença abundante dos miofibroblastos no estroma ou fronte invasivo do tumor correlacionou significantemente com o estádio clÃnico N, invasão linfática e vascular, presença de metástases histologicamente confirmadas em linfonodos e infiltração extra-capsular de metástases linfonodais. Presença abundante dos miofibroblastos foi também correlacionada com uma menor sobrevida global dos pacientes e com um maior potencial proliferativo das células tumorais. No segundo grupo de CECs orais, a presença abundante dos miofibroblastos correlacionou significantemente com o estádio clÃnico N, presença de células tumorais nas margens da peça cirúrgica e recorrência regional. Utilizando 4 linhagens celulares de CEC oral e 3 linhagens de fibroblastos de mucosa oral normal, nós demonstramos que as células tumorais induzem a transdiferenciação dos fibroblastos em miofibroblastos e que este evento é dependente da secreção de fator de crescimento transformante-β1 (TGF-β1) pelas células tumorais. Adicionalmente, nós demonstramos que os miofibroblastos secretam fatores que estimularam significantemente a proliferação celular e a produção de MMP-2 pelas linhagens celulares de CEC oral. Os resultados deste estudo sugerem que durante a progressão tumoral, as células neoplásicas secretam TGF-β1 que promove a transdiferenciação dos miofibroblastos, e por sua vez, miofibroblastos secretam fatores que induzem a proliferação celular e a produção de MMP-2 pelas células neoplásicas, favorecendo o crescimento e a invasão tumoral
Abstract: Myofibroblasts are characterized by the expression of elevated amounts of growth factors, proteases and extracellular matrix, which may influence the tumor progression. Recent studies have detected myofibroblasts in the stroma of several tumors, including those of breast, kidney, liver, bladder, colon and prostate, being the presence of those cells associated with a worse prognosis for the patient. The goals of this study were to evaluate the presence of myofibroblasts in samples of normal oral mucosa, pre-malignant leucoplakia, and 2 groups of squamous cell carcinomas (SCC), and to determine whether their presence is associated with clinicopathological features of the tumors. The first group was composed exclusively by tongue lesions, whereas the second group was formed by 38 SCCs of several sites of the oral cavity. We also investigated in vitro the mutual paracrine effects of tumor cells and myofibroblasts on fibroblast-myofibroblast transdifferentiation and tumor cell proliferation and production of matrix metalloproteinases (MMP). Immunohistochemical analysis showed the lack of myofibroblasts in the stroma of normal oral mucosa and pre-malignant oral leucoplakias. In contrast, ~60% of the SCCs contained myofibroblasts in the tumor stroma and/or deep invasive front of the tumor. In the first group, the abundant presence of myofibroblasts in the stroma or deep invasive front significantly correlated with N stage, vascular and lymphatic invasion, histopathologically confirmed lymph node metastasis, and extracapsular spread of lymph node metastasis. The abundant presence of myofibroblasts was also correlated with a shorter patient�s survival and the proliferative potential of the tumor cells. In the second group of oral SCCs, the abundant presence of myofibroblasts correlated significantly with N stage, presence of tumor cells at the surgical margins, and regional recurrence. Using 4 oral SCC cell lines and 3 primary oral normal fibroblasts (ONF) from buccal mucosa, we demonstrated that tumor cells induced transdifferentiation of ONFs to myofibroblasts via secretion of transforming growth factor-beta 1 (TGF-β1). Additionally, we demonstrated that transdifferentiated myofibroblasts secreted factors that significantly stimulated the cellular proliferation and production of MMP-2 by the oral SCC cell lines. The results of this study suggest that during tumor invasion SCC-derived TGF-β1 promote fibroblast to myofibroblast transdifferentiation, and in turn, myofibroblasts synthesize factors that induce cellular proliferation and production of MMP-2 by the tumor cells, favoring tumor growth and invasion

Full textObjective: To evaluate the preference of two designs of frontal labeling in industrialized products (octagon vs traffic light-GDA). Material and methods: Cross-sectional descriptive study in market users of Metropolitan Lima (Chorrillos, Breña, La Victoria and La Molina). We used a previously validated survey that consisted of 4 questions: “Which of the products do you find most healthy?", "Why did you choose this one?", "Which of the two labels do you find easiest to understand?", and "Which of the two labeled do you like the most?". The data are presented in percentages and median (interquartile range) and the differences between the groups were evaluated with Fisher's exact test (categorical data). The data was analyzed in the Stata v15 program. Results: 93 users were surveyed, the traffic light-GDA was chosen as the healthiest label (74.3%) and the one they liked the most (69.9%); however, the octagon was the one that seemed easier to understand (58.4%). The majority of users considered the traffic light-GDA as healthier because it "has more green on the label". Conclusions: Participants mentioned that the octagon was simpler and easier to read; the green symbols of the traffic light-GDA could influence that the product is perceived as healthier. Which could confuse the purpose of the nutritional warning, minimizing the impact of the red and/or yellow symbol by having them in less quantity than these.

Full textA regional three-dimensional parametric model of the Brazil Current (BC) was built with the goal of investigating whether or not the BC frontal meandering dominates the subinercial current variability in the Campos Basin. The BC cross-stream parametrization followed the formulation proposed by Schmidt et al. [2007]. The BC parameters were estimated from quasi-synoptic velocity profiling sampled during the Brazilian Navy cruise OCEANO SUDESTE II (OCSEII). The surface inshore thermal front (ITF),easily detected in sea surface temperature satellite images, is taken as the inshore border of the BC jet with negligible velocities. The ITF spatial pattern is then inferred by the maximum gradient method on a chosen date or period. With the ITF location, several BC cross-strem profiles are projected normally to it. The next step is to objectively interpolate the velocity field to obtain a three-dimensional BC. The spatial variability of the BC in this map is therefore solely due to the arching of the velocity/ITF front, and therefore, the velocity patterns are due to frontal meandering only. Meandering events were identified in the IFT time series, the model computed the BC frontal patterns for two separate events in the winter of 2007 and compared with PETROBRAS\' ADCP records mounted on four oil rigs. The comparison was done using weekly composites and averages to isolate mesoscale motions. It shows that there is a very favorable agreement between between the model and the open sea velocity measurements in both magnitude and direction. This reassures that the frontal meandering, or more specifically, the S~ao Tom´e Meander formation dominates the subinertial variability over the continental slope and the S~ao Paulo Plateau. The only exception is for the ADCP mounted on an oil rig on the continental shelf. The modeled and in situ observations disagree and indicate that other phenomena, such as shelf waves, should be accounted in the coastal ocean in addition to the BC meandering.

O dentista pertence a uma categoria que apresenta um alto índice de doenças osteomusculares associadas à sua prática, o que justifica o grande número de aposentadoria precoce. Ela traz consigo uma alta estatística de problemas localizados na coluna lombar, pescoço e ombros. Estes problemas estão associados com a postura estática, com a posição sentada adotada pela maioria dos dentistas e com a precisão visual e manual inerentes à tarefa que executam. Devido a esta necessidade de aproximação do campo operatório, o encosto do mocho odontológico deixa de cumprir seu principal papel de suporte para a coluna lombar. Tendo em vista este contexto, esta pesquisa objetivou desenvolver um novo conceito de mocho odontológico com apoio frontal (AF) a fim de verificar a influência deste na atividade elétrica dos músculos longuíssimos torácicos direito e esquerdo (LD e LE) e trapézios superiores direito e esquerdo (TD e TE) e na força aplicada nas partes do mocho. Para tanto foi projetado um AF para um mocho odontológico industrializado e o mesmo foi instrumentado com células de carga nas partes anterior e posterior do assento, no encosto e no AF. A partir disto foi verificada a distribuição da força no mocho odontológico simultaneamente à atividade elétrica dos músculos LD, LE, TD e TE através da eletromiografia (EMGS) durante um procedimento clínico realizado em um manequim odontológico. Participaram deste estudo 10 voluntários do sexo feminino, com idade média de 25,3 anos (±3,3), índice de massa corporal 22,3 kg/m² (± 2,4), sendo 9 destras e 1 sinistra. Durante o experimento as voluntárias realizaram o procedimento clínico restauração classe II de Black. Dentists belong to a category that presents a high index of osteomuscular disorders associated with the practice, which justifies the large number of professionals who retire early from the profession. It brings with it high statistics of problems in the lumbar spine, neck and shoulders. These problems are associated with the static posture of the seated position adopted by most dentists along with visual and manual precision inherent to the task they perform. Because of this need to be close to the operative field, the backrest of the dental stool fails to fulfill its main role: to support the lumbar spine. Given this context, the research aimed to develop a new concept of a dental stool with a front support (FS) in order to verify it’s influence on the electrical activity of the right and left longissimus thoracic muscles (RL and LL) and the right and left upper trapezius muscles (RT and LT), as well as the force applied to parts of the stool. In order to do so, an FS was designed for the dental stool and it was instrumented with load cells in the front and back of the seat, the backrest and the FS. From this, the distribution of force in the dental stool and the electrical activity of the RL, RT and LT muscles using electromyography (sEMG) were simultaneously verified during a medical procedure performed on a dental dummy. The study consisted of ten female volunteers, with an average age of 25.3 (±3.3), a body mass index of 22.3 kg/m² (±2.4), nine of whom were right-handed and one who was left-handed. During the experiment the subjects performed a Class II BLACK restorative procedure. The results pointed out a statistical difference (p<0.05) for the RT, LT and RL muscles when the FS was inserted. Full textThe aim of this thesis is to study two examples of propagation phenomena in heterogeneous reaction-diffusion equations.The purpose of the first part is to understand the effect of nonlocal exchanges between a line of fast diffusion and a two dimensional environment in which reaction-diffusion of KPP type occurs. The initial model was introduced in 2013 by Berestycki, Roquejoffre, and Rossi. In the first chapter we investigate how the nonlocal coupling between the line and the plane enhances the spreading in the direction of the line; we also investigate how different exchange functions may maximize or not the spreading speed.The second chapter is concerned with the singular limit of nonlocal exchanges that tend to Dirac masses. We show the convergence of the dynamics in a rather strong sense. In the third chapter we study the limit of long range exchanges with constant mass. It gives an infimum for the asymptotic speed of spreading for these models that still could be bigger than the usual KPP spreading speed.The second part of this thesis is concerned with entire solutions for heterogeneous bistable equations.We consider a two dimensional domain infinite in one direction, bounded in the other, that converges to a cylinder as x goes to minus infinity. We prove the existence of an entire solution in such a domain which is the bistable wave for t tends to minus infinity. It also lead us to investigate a one dimensional model with a non-homogeneous reaction term,for which we prove the same property

Full textIn coastal areas, when the summer stratification settles, the bottom and surface turbulent boundary layers can overlap and delineate thermal fronts. An example in the Iroise sea is the Ushant front that develops around areas of intense mixing (ZMP): the "Raz de Sein" and the area of Ouessant. Its extension is variable and exceeds the areas of mixed waters production. Both the extension of the front and its variability are sensitive to mechanisms of dispersion of homogenized waters. Those mechanisms of dispersion are studied using academic configurations where tidal mixing is represented as fixed areas where stratified waters are continuously homogenized. It is shown that baroclinic instability developing at the edge of the ZMPs represents the most efficient mechanism to disperse the homogenized waters. Any process that modifies the characteristics of baroclinic instability, or the auto-propagating efficiency of emerging structures, significantly impacts the dispersion rate and pattern. The sensitivity of dispersive processes to seasonal and local parameters is analyzed. Realistic configurations of the Iroise sea are then used to investigate the variability of thermal fronts when mixing processes are combined with dispersive mechanisms. Thermal gradients in the surface and subsurface layers are investigated considering extension patterns of the Ushant front during summer 2009. It is shown that thermal fronts are partly initiated by the presence of mixed waters produced in ZMPs and subducted in subsurface. Those anticyclonic structures thicken the subsurface layer, and can interact with surface turbulence, thus favoring the outcropping of cold to mild waters at the surface
